Despite the advances made over the last decade, infections caused by multi-drug resistant bacterial strains are increasingly important societal issues that need to be addressed. New approaches have already been developed in order to overcome this problem. Photodynamic antimicrobial chemotherapy (PACT) could provide an alternative to fight infectious bacteria. This approach has already inspired the development of innovative materials. Interesting results have been obtained against Gram-positive bacteria, but it also appeared that Gram-negative strains, especially Pseudomonas aeruginosa, were less sensitive to PACT. Enhanced efficacy against Gram-negative bacteria had been previously obtained with photosensitizers bound to antimicrobial peptides. In this work, we designed a photobactericidal organic material, CNCsc6-PMB, consisting of cellulose nanocrystals to which the photosensitizer chlorin-e6 and the antimicrobial polypeptide polymyxin B (PMB) were covalently attached. These modified nanocrystals were characterized by IR spectroscopy, zeta potential measurements and elemental analyses, after which antibacterial assays were carried out. Following light irradiation, CNCsc6-PMB demonstrated efficiency against Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and Gram-positive bacteria (Staphylococcus aureus and Staphylococcus epidermidis) by inhibition of bacterial growth. An amplifying effect of chlorin-e6 has been highlighted against these Gram-negative strains, based on membrane weakening and a potential docking effect from the polymyxin moiety. Such results confirmed the importance of using an antimicrobial peptide in order to broaden the spectrum of PACT.

Industrial processing of raspberries into juice and jam results in the production of with high content of lipophilic and hydrophilic phytochemicals. Usually considered as waste, raspberry pomace is occasionally cold-pressed to recover specialty oil. However, the resulting pomace press-cake (PPC) still contains 30% to 35% of lipophilic compounds, such as essential fatty acids, tocols, phytosterols, and ellagitannins initially present in pomace. In a perspective of sustainable development, we investigate an eco-friendly process using an aqueous enzyme-assisted extraction (AEAE) to simultaneously and effectively recover lipophilic compounds and polyphenols from the PPC. The performance of different combinations of carbohydrases and proteases was compared. After selecting the best enzymatic system, a definitive screening design involving six factors was then implemented to optimize the process. Under optimized conditions, 1.2 units of thermostable alkaline protease/100 g PPC, pH 9, 60 degrees C, and 2 hr hydrolysis, more than 38% of total PPC lipophilic content were recovered in the aqueous medium. The recovery of polyphenols and antioxidant activity was, respectively, 48% and 25% higher than obtained by extraction with methanol/acetone/water mixture. Such an AEAE extract might prove useful in food and nutraceutical applications. Practical Application Raspberry pomace, a food industrial by-product, is often considered as waste. However, it is a rich source of phytochemicals, such as tocols, polyphenols, and polyunsaturated fatty acids. To overcome the drawbacks of organic solvent use, an enzyme-assisted extraction process was developed as an eco-friendly alternative to recover these bioactive compounds. Definitive screening design experimental design was used to enhance polyphenols and lipophilics extraction yields while reducing process costs. This extract is an oil-in-water emulsion, with high content in antioxidant phytochemicals, which might have potential for use in nutraceutical applications. Therefore, this green process developed for the valorization of raspberry pomace is considered as a perspective of sustainable development.

International audience; We describe the synthesis, characterization and photophysical properties of two new polycarboxylic photosensitizers. Owing to their structural design, these two compounds show water solubilities larger than natural carboxylic photosensitizers (e.g., protoporphyrin IX, hematoporphyrin, etc.) and also good singlet oxygen quantum yields. These compounds were tested as photo-antimicrobial agents against Staphylococcus aureus and Bacillus cereus strains. Results reveal that their photocytotoxicities are strongly dependent on their amphiphilic character and more precisely the number and position of the carboxylic acid and mesityl substituents

International audience; Probiotics represent a potential strategy to influence the host's immune system thereby modulating immune response. Lipoteichoic Acid (LTA) is a major immune-stimulating component of Gram-positive cell envelopes. This amphiphilic polymer, anchored in the cytoplasmic membrane by means of its glycolipid component, typically consists of a poly (glycerol-phosphate) chain with d-alanine and/or glycosyl substitutions. LTA is known to stimulate macrophages in vitro, leading to secretion of inflammatory mediators such as Nitric Oxide (NO). This study investigates the structure-activity relationship of purified LTA from three probiotic Bacillus strains (Bacillus cereus CH, Bacillus subtilis CU1 and Bacillus clausii O/C). LTAs were extracted from bacterial cultures and purified. Chemical modification by means of hydrolysis at pH 8.5 was performed to remove d-alanine. The molecular structure of native and modified LTAs was determined by 1H NMR and GC-MS, and their inflammatory potential investigated by measuring NO production by RAW 264.7 macrophages. Structural analysis revealed several differences between the newly characterized LTAs, mainly relating to their d-alanylation rates and poly (glycerol-phosphate) chain length. We observed induction of NO production by LTAs from B. subtilis and B. clausii, whereas weaker NO production was observed with B. cereus. LTA dealanylation abrogated NO production independently of the glycolipid component, suggesting that immunomodulatory potential depends on d-alanine substitutions. d-alanine may control the spatial configuration of LTAs and their recognition by cell receptors. Knowledge of molecular mechanisms behind the immunomodulatory abilities of probiotics is essential to optimize their use.

Speeding is one of the main factors of car crash-risk, but it also contributes to increasing air-pollution. In two studies we attempted to lead drivers to abide by speed limits using "reducing air-pollution" as a new argument. We presented prevention messages that highlighted the role of speeding in increasing "crash-risk", "air-pollution", or both (Studies 1 and 2). The messages were also positively or negatively framed (Study 2). Given that women are more concerned with environmental issues than are men, we expected the following hypotheses to be validated for women. The message with the "air-pollution" argument was expected to be evaluated more positively than the "crash-risk" message (H1). The "air-pollution" and "crash-risk and air-pollution" messages were expected to be more effective than the "crash-risk" message on the behavioral intention to observe speed limits (H2a) and on the perceived efficacy of speed-limit observance in reducing air-pollution (H2b; Studies 1 and 2). Furthermore, positive framing was expected to be more effective than negative framing (H3), and presenting a message to be more effective than presenting no message (H4; Study 2). Broadly, our results argue in favor of our hypotheses. However in Study 2, the effects of message framing did not allow us to conclude that negative or positive framing was superior. All in all, messages with the "air-pollution" argument were more effective at leading drivers to observe speed limits. Thus, environmental protection may be a fruitful route to explore for increasing road safety.

Our first aim was to replicate in French rehabilitation training courses the results of Delhomme et al. [Delhomme, P., Kreel, V., and Ragot, I. The effect of the commitment to comply with speed limits during rehabilitation training courses for traffic regulation offenders in France. European Review of Applied Psychology, doi:10.1016/j.erap.2005.07.005 ] who found, from phone interviews, a positive effect of the commitment to comply with speed limits. Our second aim was to extend the previous study. In the present study, we used the same mixed Foot-in-the-door procedure and introduced one more experimental condition in which committed participants were additionally asked to specify the actions they would adopt in the next 6 months to keep their commitment. Six hundred and seventy-eight participants were assigned to one of the two experimental conditions, a comparison condition or a control condition. Only 38% of participants committed but we regained the positive effect of commitment on self-reported speed in the short term and for more than 5 months after the courses. In the committed group which specified actions, the effect appeared on more specific roads than in the committed group which did not specify actions. The actions were categorised according to whether they were exclusively cognitive or behavioural, or both. The committed participants who declared having complied with speed limits, compared to those who declared having broken them, before the first interview, reported during the course more of both (cognitive and behavioural actions). The interest of integrating commitment procedure in rehabilitation training courses together with the actions specified to keep one's commitment is discussed.

A mixed foot-in-the-door procedure was set up to lead offenders participating in French driver-rehabilitation training courses to commit to complying with speed limits. To improve this procedure, the reasons that led participants in such courses to commit or not were investigated in this preliminary study of the road safety research programs. The study participants were 89 men ( M= 34 yr., range = 18–64 yr., SD= 13) attending driver-rehabilitation courses. Among them. 55% committed. The commitment was mainly explained by the drivers' will to change their speeding behaviour and their refusal by the perceived positive consequences of speeding. Moreover, some participants stated that they would be unable to follow through with the commitment to comply strictly with speed limits. After talking to the participants, it became clear that complying with speed limits was understood in a more or less strict way. In subsequent studies, some of the reasons given were used to prompt group discussion before asking participants to commit, and the meaning of “compliance with speed limits” was better defined.
