Canola oil is an important source of oleic acid, in addition to being an accessible source for its use in the production of pigs’ diets. Oleic acid in turn is a type of unsaturated fatty acid that in pork is beneficial for human health. Therefore, this study was conducted to describe the fatty acid profile in brain and liver tissues from pigs supplemented with canola oil for 98 days, during the growth and finishing phases. For the analysis was used eighteen male pigs that had free access to feed and water throughout the experimental period. Dietary treatment consisted of corn-soybean meal growing-finishing diets supplemented with 3% fat from canola oil (CO). To obtain the fatty acid profile, the lipids from each tissue were cold extracted using the adapted method from Bligh and Dyer, methylated, and posteriorly injected in a gas chromatograph to obtain the fatty acid profile of the tissue. In both tissues there was a greater abundance of saturated fatty acids (stearic acid). The most abundant monounsaturated fatty acid was the oleic acid. Regarding polyunsaturated fatty acids, in the liver the most abundant was linoleic acid and in the brain docosahexaenoic acid. In summary, animals' diet influences the fatty acid profile in different tissues. Such modifications can increase unsaturated fatty acids concentration in relation to saturated, making pork healthier for human consumption.

Diets high in fat have been used in the production of pigs in the growing and finishing stages for the improvement of meat production and quality parameters. Therefore, this study was conducted to evaluate the effect of adding different oil sources to the diet of immunocastrated male pigs on meat quality characteristics. Ninety-six pigs were randomly allotted to one of four dietary treatments, with six replicate pens per treatment and four pigs per pen. Dietary treatments consisted of diets supplemented with 1.5% soybean oil (SO; control) or 3% oil from SO, canola oil (CO), or fish oil (FO). After 98 days on trial, 18 pigs per treatment (n = 72) were slaughtered and samples of the Longissimus dorsi muscle were taken for Warner-Bratzler shear force and color (L*, a*, and b*) measurements. Loins from pigs fed diets containing either 3% SO or CO had decreased Warner-Bratzler shear force (P = 0.04) than those from pigs fed the control diet. Dietary treatments had no effect on color components of fresh loin. These results indicate that feeding 3% SO or CO increased pork tenderness, without compromising color meat of immunocastrated male pigs.

Dietary l-arginine (ARG) supplementation has been studied as a nutritional strategy to improve reproductive performance of pregnant sows, since arginine is a conditionally essential amino acid. However, reports addressing the molecular mechanisms that mediate supplementation effects on embryos and fetuses development are still scarce. Therefore, we aimed to evaluate the effects of 1.0% ARG supplementation of commercial pregnant gilts on genes and proteins from energy metabolism and antioxidant defense pathways in embryos and fetuses. We also analyzed the global transcriptome profile of 25- and 35-day-old conceptuses. At Day 25, we observed a lower abundance of phospho-AMP-activated protein kinase (phospho-AMPK) protein and downregulation of oxidative phosphorylation system genes in ARG embryos. On the other hand, ARG fetuses showed greater expression of MLST8 and lower expression of MTOR genes, in addition to lower abundance of phospho-AMPK and phospho-mammalian target of rapamycin (phospho-mTOR) proteins. Transcriptome analysis at Day 35 did not present differentially expressed genes. For the antioxidant defense pathway, no differences were found between CON and ARG conceptuses, only trends. In general, supplementation of gilts with 1.0% ARG during early gestation affects energy sensitive pathways in 25- and 35-day conceptuses; however, no effects of supplementation were found on the antioxidative defense pathway in 25-day embryos.

The objective of this study was to evaluate the effects of dietary supplementation of purified lignin on the performance, relative organ weights, serum metabolites, and gene expression profiles of broiler chickens subjected to cyclic heat stress (HS). At 22 days old, 280 broilers were distributed in a completely randomized design with four treatments, ten repetitions, and seven birds per experimental unit. The birds were subjected to daily cyclic HS. A high temperature of 32±1 °C was maintained for 10 h/day (08:00–18:00 h), while a temperature of 22±1 °C was maintained for the remaining time. Treatments were a basal diet or basal diet with the addition of 5, 10, or 15 g of purified lignin/kg of diet. Data were analyzed using one-way ANOVA and means were compared by Tukey's test at 0.05 significance. There was no effect of lignin supplementation on performance, carcass yield, relative weights of the bursa, spleen, and liver, or serum levels of glucose, triglycerides, uric acid, malondialdehyde, triiodothyronine, or tetraiodothyronine. The abundance of mRNA of heat shock protein 70, nuclear factor-κB, glutathione peroxidase, and Cu,Zn-superoxide dismutase in the liver was similarly unaffected by treatments. Purified lignin supplementation does not improve performance or the antioxidant response of broiler chickens subjected to HS.

During gestation, nutrition is the main environmental factor that influences the intrauterine environment and, consequently, development of embryos and fetuses. In this context, l -arginine supplementation of pregnant sows has been shown to be efficient to improve females’ reproductive performance and conceptuses development. Among the products of arginine metabolism, nitric oxide and polyamines play important roles in placenta and pig conceptuses growth and development, through vascularization and blood flow regulation, cell proliferation, migration and differentiation, tissue formation, among other processes. In this review, we aimed to approach current knowledge about the nutrition influence on sow gestation, emphasizing how l -arginine supplementation may affect reproductive performance and conceptuses development and survival.

Simple Summary A complete and exploratory landscape of developmental biology during mammalian organogenesis is needed to improve livestock production and contribute with comparative developmental studies in other relative species, including humans. Since transcriptome analysis is suitable to understand systemic context and genomic interactions, we evaluated the main transcriptional changes during overall organogenesis comparing fetal and embryonic stages through RNA-seq analysis, using the pig as model. The differentially expressed genes between pig fetuses and embryos highlight the main biological events and pathways required for primary organs and body structure development at the embryonic stage, the biological events and pathways related to growth and specialization of these organs at the fetal stage, as well as the molecular mechanisms involved in embryonic to fetal transition. This exploratory transcriptional landscape contributes to improving our knowledge of the crucial biological events during normal prenatal development, which is relevant in prenatal programing studies as well as in livestock production. Abstract Since pre- and postnatal development are programmed during early prenatal life, studies addressing the complete transcriptional landscape during organogenesis are needed. Therefore, we aimed to disentangle differentially expressed (DE) genes between fetuses (at 35 days old) and embryos (at 25 days old) through RNA-sequencing analysis using the pig as model. In total, 1705 genes were DE, including the top DE IBSP, COL6A6, HBE1, HBZ, HBB, and NEUROD6 genes, which are associated with developmental transition from embryos to fetuses, such as ossification, skeletal muscle development, extracellular matrix organization, cardiovascular system, erythrocyte differentiation, and neuronal system. In pathway analysis, embryonic development highlighted those mainly related to morphogenic signaling and cell interactions, which are crucial for transcriptional control during the establishment of the main organs in early prenatal development, while pathways related to myogenesis, neuronal development, and cardiac and striated muscle contraction were enriched for fetal development, according to the greater complexity of organs and body structures at this developmental stage. Our findings provide an exploratory and informative transcriptional landscape of pig organogenesis, which might contribute to further studies addressing specific developmental events in pigs and in other mammals.

Sexual dimorphism is a relevant factor in animal science, since it can affect the gene expression of economically important traits. Eventually, the interest in the prenatal phase in a transcriptome study may not comprise the period of development in which male and female conceptuses are phenotypically divergent. Therefore, it would be interesting if sex differentiation could be performed using transcriptome data, with no need for extra techniques. In this study, the sex of pig conceptuses (embryos at 25 days-old and fetuses at 35 days-old) was determined by reads counts per million (CPM) of Y chromosome-linked genes that were discrepant among samples. Thus, ten genes were used: DDX3Y, KDM5D, ZFY, EIF2S3Y, EIF1AY, LOC110255320, LOC110257894, LOC396706, LOC100625207, and LOC110255257. Conceptuses that presented reads CPM sum for these genes (ΣCPMchrY) greater than 400 were classified as males and those with ΣCPMchrY below 2 were classified as females. It was demonstrated that the sex identification can be performed at early stages of pig development from RNA-sequencing analysis of genes mapped on Y chromosome. Additionally, these results reinforce that sex determination is a mechanism conserved across mammals, highlighting the importance of using pigs as an animal model to study sex determination during human prenatal development.

We aimed to evaluate apoptosis and myogenesis related genes expression in embryos and fetuses from two divergent genetic groups of pigs: Piau breed and a commercial line. Thirty females (15 Piau and 15 commercial line) were selected at 120 days of age. Estrous cycle was observed and on the third estrus females were considered sexually mature. Gilts were inseminated with semen from males of the respective breed. Three females from each breed were slaughtered at five different gestational ages: 15, 30, 45, 60 and 90 days. Whole embryos (15 and 30 d) and samples of longissimus dorsi muscle from fetuses (45, 60 and 90 d) were collected for RNA extraction. Expression of apoptosis and myogenesis related genes (BAX, BCL2, FGF4, IHH, HHIP, SHH, SOX2, WNT1 and WNT4) were evaluated by quantitative real time PCR. There was significant effect of interaction between breeds and gestational ages for all genes evaluated (P

l -arginine supplementation of sows has led to improvement of reproductive performance, but the mechanisms responsible for the positive effects of arginine during gestation on conceptuses survival and development are still poorly understood. Thus, we aimed to evaluate effects of 1.0% l -arginine supplementation (ARG) on phenotypic traits of commercial gilts, embryos and fetuses, concentration of gilts’ blood metabolites, expression of developmental and cellular apoptosis genes in conceptuses of 25 and 35 days. At 25 days, IGF1 gene was more expressed in embryos from ARG than in embryos from control gilts (CON) (P = 0.05). At this same gestational age, ARG embryos tended to be heavier compared to CON (P = 0.07) and ARG gilts showed a trend to have a greater arginine concentration in blood plasma (P = 0.06). However, at 35 days of gestation, arginine concentration in blood plasma of ARG gilts tended to be lower compared to CON (P = 0.06) and ARG fetuses showed smaller cephalic-caudal length (P = 0.05). These results indicate that duration of supplementation is determinant for arginine effects, not only on the females performance but also on the conceptuses, since supplementation upregulated IGF1 expression at 25 days, in addition to the reduction of cephalic-caudal length of 35-day fetuses.

In view of the importance of the genetic material of local breeds in the swine industry and the lack of information about reproductive performance of Piau females, two experiments were conducted to evaluate puberty and sexual maturity as well as the morphometry of embryos/fetuses, placenta and uterus during the first 90 days of gestation in Piau breed and Commercial line gilts. In experiment I, 37 Piau and 25 commercial line gilts were used. From the 120 days of age, detection of estrus was performed using mature boars from the first to third estrus of each gilt. Data regarding to age, body weight and estrus duration were recorded. After third estrus, females were slaughtered and ovaries were collected to determine ovulation rate. In experiment II, 36 Piau and 18 commercial line gilts were distributed into three groups according to the mating: Commercial, commercial line females x commercial line male; cross-mated, Piau females x commercial line male; and Piau, Piau females x Piau male. Gilts were slaughtered at 7, 15, 30, 45, 60 and 90 days of pregnancy. Piau females reached puberty and sexual maturity at the same age as commercial line females, but with lower weight; moreover, Piau group showed negative correlations of birth weight with puberty (−0.27) and sexual maturity (−0.29). Commercial gilts presented higher ovulation rate, weight and length of uterus, and length and thoracic circumference of fetuses. Nevertheless, number of fetuses was similar in all groups at 90 days of gestation suggesting that Piau females present higher survival rates of the conceptuses. The results showed differences between the genetic groups related to fetal and placental development, gestational losses, number of ovulations and uterine development. In addition, an intermediate status of fetal weight was observed in Piau/Commercial line crossbred conceptuses; thus, the selection of Piau females on reproductive traits to be mated with commercial line males would be an alternative to contribute to improvement of intramuscular fat content.

Comprehending mechanisms controlling corpus luteum (CL) angiogenesis and apoptosis in pregnant sows is essential to understand the physiological role of these processes in CL function, progesterone production and consequently in conceptus development and prenatal mortality. CL from 54 sows from two genetic groups, a commercial line (COM) and the local Piau breed (LPB), were obtained for gene expression (n = 3 COM; n = 6 LPB), histological and protein analysis (n = 3 COM; n = 3 LPB), divided in six gestational ages (seven, 15, 30, 45, 60 and 90 days). We observed differences between gestational ages in CL morphology, in which the average number of blood vessels/capillaries at 90-days was greater than at the seventh day by Tukey test. RT-qPCR analysis revealed that apoptotic genes (BAX, BCL2 and CASP3) were differentially expressed between genetic groups and gestational ages in each group. Angiogenesis genes also presented differences between genetic groups (ANGPT1) and gestational ages (MMP9, VEGFA and ANGPT1). No differences in protein abundance of steroidogenic enzymes (CYP11A1 and HSD3B1) were observed. Our findings indicate that despite the differences in gene expression, differences in corpus luteum vascularization were observed only across gestational ages, with no dissimilarities between genetic groups.

A raça local brasileira Piau é um suíno tipo banha a qual passou por um processo de seleção realizado pelo médico veterinário Antonio Teixeira Vianna em 1939, com o intuito de desenvolver um animal de dupla aptidão, em outras palavras que fosse adequado para a produção de carne e gordura. As principais características da raça são rusticidade, adaptabilidade e resistência a doenças. O status de conservação da raça é ameaçado e em 1998 a Granja de Melhoramento de Suínos da Universidade Federal de Viçosa (UFV, Viçosa, MG, Brazil) iniciou uma criação de suínos da raça Piau com o objetivo de conservá-la e utilizá-la em estudos genéticos. O objetivo do presente estudo é descrever o programa de conservação da raça Piau da UFV e avaliar uma série de parâmetros populacionais, a fim de gerar informações que possibilitem o monitoramento e a melhoria do programa. Para a análise da estrutura populacional foi utilizado arquivo de pedigree com 1349 indivíduos por meio do software ENDOG v. 4.8. O programa de conservação da raça possui boa estrutura com registro de dados zootécnicos e pedigree desde a introdução dos primeiros animais na granja de melhoramento de suínos, juntamente com o controle de acasalamentos, o qual é baseado no acasalamento de animais com menor coancestralidade e na manutenção das famílias fundadoras. A população de Piau avaliada possui número efetivo de fundadores e ancestrais iguais a 9 e 8, respectivamente. O coeficiente de endogamia médio, coeficiente de relação médio e tamanho efetivo da população são 6,55 %, 19,74 % e 18,59, respectivamente. De acordo com os parâmetros avaliados é recomendado aumentar o tamanho efetivo da população, por meio da introdução de animais de outra população, uma vez que o pequeno número de fundadores torna difícil o controle da endogamia.

MicroRNAs are a class of naturally occurring non-coding RNAs. Typically they are ∼22 nucleotides long and suppress translation of their targets genes. Several laboratories have attempted to identify miRNAs from pig muscle and the bioinformatics strategies using ESTs have proved to be successful for this aim. In this study we report an in silico identification of ncRNA in pig EST libraries focusing on novel pig miRNAs and further investigated the differential expression of pigs miRNAs (known and novel) by quantitative real-time PCR during pre- and postnatal stage from Commercial and local breed Piau pigs skeletal muscle tissue. We identified two miRNAs not yet described in pigs: hsa-miR-1207-5p and hsa-miR-665. Besides, we found 288 target genes for hsa-miR-1207-5p and 214 for hsa-miR-665; from them, four are muscle specific genes. Through expression analyses, differences were found between pre- and postnatal stages and genetics groups. The findings of miRNAs and their muscle-specific targets in pigs will be helpful for understanding the function and processing of this RNA class in the future. Besides, the miRNAs differentially expressed between Commercial and Piau breeds suggest that they can be used to uncover phenotypic differences across different genetic groups.

Summary Pig is an important animal for meat production; this is generally associated with characteristics determined prenatally during myogenesis. Expressed sequence tags (EST) can provide direct information on the transcriptome and indirect information on the relation between the genome and phenotype, giving information about differentially expressed genes (DEG). In this work, the identification and annotation of DEG from EST libraries of three pig breeds (Duroc, Large White and Local Breed Piau) were performed followed by real-time PCR analyses during pre- and postnatal stages (21, 40, 70 and 90 days of pregnancy and 107, 121 and 171 days postnatal) from commercial breed animals for analysis of genes expression levels. Therefore, 34 genes differentially expressed were identified, of which 21 grouped in a network related with muscle development. From this, the expression profile of 13 genes was measured, to confirm their relationship with myogenesis like ANKRD2, MYBPC1, NEB and MYL2. These genes showed a prenatal high expression in this study. Besides, novels candidates for muscle development (TP53 and DCTN1) were listed. These findings can contribute to better explaining gene function mechanism and are helpful in uncovering the pathways that mediate pre- and postnatal skeletal muscle development in vertebrates.
