The rumen simulation technique (RUSITEC) was used to investigate the effect of ergot alkaloids (EA) and a mycotoxin deactivating product (Biomin AA; MDP) on nutrient digestion, ruminal fermentation parameters, total gas, methane, and microbial nitrogen production. Ruminal fermentation vessels received a feedlot finishing diet of 90:10 concentrate:barley silage (DM basis). Using a randomized complete block design, treatments were assigned (n = 4 vessels/treatment) within two RUSITEC apparatuses in a 2 × 2 factorial arrangement. Treatments included: (1) control (CON) diet (no EA and no MDP); (2) CON diet + 1 g/d MDP; (3) CON diet + 20 mg/kg EA; and (4) CON diet + 20 mg/kg EA + 1 g/d MDP. The study was conducted over 14 d with 7 d of adaptation and 7 d of sample collection. Data were analyzed in SAS using PROC MIXED including fixed effects of EA, MDP, and the EA×MDP interaction. Random effects included RUSITEC apparatus and cow rumen inoculum (n = 4). Ergot alkaloids decreased dry matter (DMD) (P = 0.01; 87.9 vs. 87.2%) and organic matter disappearance (OMD) (P = 0.02; 88.8 vs. 88.4%). Inclusion of MDP increased OMD (P = 0.01; 88.3 vs. 88.9%). Neutral detergent fiber disappearance (NDFD) was improved with MDP; however, an EA×MDP interaction was observed with MDP increasing (P0.001) NDFD more with EA diet compared to CON. Acetate proportion decreased (P = 0.01) and isovalerate increased (P = 0.03) with EA. Consequently, acetate:propionate was reduced (P = 0.03) with EA. Inclusion of MDP increased total volatile fatty acid (VFA) production (P0.001), and proportions of acetate (P = 0.03) and propionate (P = 0.03), and decreased valerate (P0.001), isovalerate (P = 0.04), and caproate (P = 0.002). Treatments did not affect (P ≥ 0.17) ammonia, total gas, or methane production (mg/d or mg/g of organic matter fermented). The inclusion of MDP reduced (P0.001) microbial nitrogen (MN) production in the effluent and increased (P = 0.01) feed particle-bound MN. Consequently, total MN decreased (P = 0.001) with MDP. In all treatments, the dominant microbial phyla were Firmicutes, Bacteroidota, and Proteobacteria, and the major microbial genus was Prevotella. Inclusion of MDP further increased the abundance of Bacteroidota (P = 0.04) as it increased both Prevotella (P = 0.04) and Prevotellaceae_UCG-003 (P = 0.001). In conclusion, EA reduced OMD and acetate production due to impaired rumen function, these responses were successfully reversed by the addition of MDP.Ergot formed from a parasitic fungus (Claviceps purpurea) affects various types of grains (rye, wheat, or oats) and may contain several toxic ergot alkaloids (EA). Individual EA may impact the rumen microorganisms, and cattle feed intake, digestibility, health, and overall performance. A common method to alleviate toxicity in mycotoxin-contaminated feed is through the addition of mycotoxin binders (MDP); however, their efficacy against EA is unknown. To better understand the effect of EA in cattle, we performed an in vitro experiment to examine the impact of EA on the ruminal microbial populations and fermentation of a finishing feedlot diet using an artificial rumen (RUSITEC). Additionally, an MDP was added to test if it could reduce the detrimental effects of EA on rumen fermentation. MDP increased total volatile fatty acids (VFA) and reduced total microbial protein synthesis. Furthermore, EA reduced microbial diversity and the acetate:propionate ratio. Although EA reduced organic matter digestibility and acetate production, these negative effects were reversed by the addition of the MDP.

The objective of this study was to evaluate the effect of feeding pelleted or mash ergot contaminated grain continuously or intermittently on performance and blood parameters of backgrounding steers. Sixty black angus steers (300 ± 29.4 kg) were used in a complete randomized backgrounding (60% barley silage:40% concentrate DM basis) 84 d study. Steers were randomly assigned to 4 different treatments and housed individually. Treatments included: 1) control ration (CON; no added ergot), 2) continuous ergot mash (CEM; fed continuously at 2 ppm), 3) intermittent ergot mash (IEM; fed at 2 ppm on the first wk of each 21 d period and control diet for the remaining 2 wk), and 4) intermittent ergot pellet (IEP; fed at 2 ppm during the first wk of each 21 d period and control diet for the other 2 wk). Calves were weighed every 21 days and blood samples were collected on d 0, 42, and 84. No treatment effects (P>0.05) were observed for complete blood count. Steer DMI was reduced for all ergot diets compared to the CON diet (P< 0.01, 7.45 vs. 8.05 kg/d). Steer ADG was reduced for all ergot diets compared to the CON diet (P< 0.01). CEM fed steers had lower ADG (P< 0.01, 0.980 vs. 0.735 kg) and shrunk final weight (P< 0.01, 366 vs. 350 kg) than CON calves, but CEM and IEM were no different. CEM fed steers had lower Gain:Feed (P< 0.07, 0.130 vs. 0.142) than CON calves. Pelleting ergot contaminated grain did not reduce the impact of ergot alkaloids on any of the measured parameters. Results suggest that continuously or intermittently feeding an ergot contaminated TMR (2 ppm) can significantly reduce the growth performance of backgrounded feedlot steers but no effects were observed for blood parameters. Pelleting was not an effective method of reducing ergot toxicity.

We explored whether straw bedding at rest stations might affect latency and duration of lying down beyond the 8h rest required at rest stops during long distance transport. Animals arriving to commercially operated rest stops (n=75, 6/load, opportunistically selected) were rested in pens (15.5 × 9.5 m) that were either bedded (n=38, straw, 14 cm deep) or non-bedded (n=37). The lying activity of each animal was recorded every 10 min for 8 h. The independent variables recorded included: bedding treatment, mean animal weight/load (kg), and space allowance [k-value = (m2/animal) / (BW2/3)] in the trailer. Ordinary linear and mixed linear regression models were fitted to assess lying latency and duration, respectively. Bedding affected latency to lie down, but its effect depended on space allowance in the truck: among cattle transported with low space allowance (2.08 - 3.29 m2/ 300 kg animal), bedded cattle laid down sooner than non-bedded cattle (P< 0.001). Comparing only cattle in bedded pens, cattle laid down sooner when transported with low space allowance (2.08 - 3.29 m2/ 300 kg animal) compared with medium space allowance (>3.29 – 3.69 m2/ 300 kg animal; P=0.003). Bedding also affected lying duration, but the effect depended on mean animal weight; as mean animal weight of the load increased so did duration but the effect was greater among bedded animals (P=0.027). In summary, cattle transported at high stocking densities are most likely to benefit from bedding as are heavier animals.

The aim of this study was to assess the effects of conditioning, rest, and post-rest transport duration on welfare indicators of 6-7 mo old beef calves. A total of 328 weaned calves (237 ± 29.7 kg BW) were randomly assigned to a 2 × 2 × 2 factorial design: conditioning, conditioned (C) or non-conditioned (N); rest, 0 (R0) or 8 (R8) h, and post-rest transport, 4 (T4) or 15 (T15) h. Calves were sampled prior to and after 20h of transport, prior to and after the additional 4 or 15-h transport, and at 1, 2, 3, 5, 14, and 28 d after transport ended. Data were analyzed using the GLIMMIX procedure of SAS. Fixed effects were conditioning, transport and time nested within rest period, while random effects were animal and pen. For R0-T4 calves, the mean L-lactate concentrations were greater than R8-T4 calves on d 1 and 2 (p-values = 0.02) while, for R0 calves, mean ADG was greater than R8 calves 14 to 28 d after transport (p-values < 0.01). For R8-T4 calves 1 week after transport, mean ADG was greater and mean WBC counts were less than R8-T15 calves on d 5 (p-values < 0.01). For N calves, overall mean haptoglobin, creatine kinase, serum amyloid-A, and non-esterified fatty acids were greater than C calves (p-values ≤ 0.05). Overall, few and inconsistent differences were observed for rest, where rest improved L-lactate but affected ADG. Few differences were observed for transport, where shorter transport durations after the rest improved ADG and WBC counts. In general, N calves had greater physiological indicators of reduced welfare than C calves. Based on these findings, the best way to improve calf welfare during and after transport is to condition them prior to transport.

Lameness is a significant health issue in feedlot cattle, having major economic, labor, and welfare implications. Digital dermatitis (DD), a lesion of the plantar surface of the foot, and foot rot (FR), which affects the interdigital cleft, are common infectious causes of feedlot lameness with bacterial origins. These hoof lesions can occur alone or in combination in the same hoof (DD+FR). Over a one-year period (2019-2020), 208 hoof swabs were obtained from cattle with FR (n = 55), DD (n = 18), and DD+FR (n = 29), and 5 completely healthy cattle (controls) from three southern Alberta commercial feedlots. 16S rRNA gene sequencing was used to characterize the bacterial microbiota of DD, FR, and DD+FR lesions relative to corresponding healthy skin controls found within the same affected animals. DD and FR lesion microbiotas showed reduced diversity compared with corresponding control skin. Multiple Treponema (members of Spirochaetaceae Family) species were differentially abundant in DD lesions compared with contralateral control skin. FR lesions are characterized by an abundance of Fusobacterium necrophorum and Porphyromonas levii, compared with contralateral control skin. The microbiome of DD+FR lesions was similar to FR lesions, where F. necrophorum and P. levii were increased compared with contralateral control skin. Other differentially abundant taxa (Helcococcus, Succiniclasticum spp.) found in FR and DD+FR lesions had been previously associated with DD, suggesting there may be an overlap of bacteria that have a role in both types of hoof lesions. The findings support previous literature showing that FR and DD are polybacterial in nature. The identification of novel bacterial species of DD and FR lesions could contribute information for the development of more targeted and effective prevention and treatment strategies. A novel finding of the sequencing analysis for hooves with concurrent DD and FR infection was characterized.

Lameness is a significant health issue in Canadian feedlots resulting in substantial economic losses. However, the high frequency of misdiagnosis of lameness using traditional methods leads to ineffective treatment, suggesting a new diagnostic method is needed. Growing evidence indicates that microRNAs (miRNAs) can be used as biomarkers for identifying the animals’ physiological status and the diagnosis of certain diseases, but this approach has not been utilized in beef cattle. The objective of this study was to compare blood miRNA profiles between lame and healthy cattle to investigate the relationship between miRNA expression patterns and specific lameness phenotypes. Blood samples were collected from 156 feedlot cattle at 0, 1, 2 and 3 weeks after being diagnosed with either digital dermatitis (DD; n=62), toe tip necrosis syndrome (TTNS; n = 40), or footrot (FR; n = 40) and healthy controls (HC; n = 12) for miRNA libraries construction and sequencing. A total of 314 expressed miRNAs were identified in 89 blood samples collected at week 0 across all groups, with TTN having the largest number of expressed miRNAs (291, P < 0.01) compared to all other groups (HC=276, DD=281, FR=278). Although miRNA profiles did not differ among the lameness types, type-specific miRNAs were identified; 6 in DD, 10 in TTN, 5 in FR and 7 in HC cattle. In addition, 3, 6 and 7 DE miRNAs were detected in DD, TTNS and FR when compared with HC cattle. Most of the DE and group-specific miRNAs are related to inflammation and skin diseases. The DE miRNAs were different between week 0 and all other weeks, indicating miRNA profiles may differ over time and with disease progression and recovery. These findings provide an initial understanding of the relationship between the cattle blood miRNAome and lameness and suggest that miRNA expression holds promise in the discovery of novel biomarkers for identifying lameness.

The objective of this study was to determine if the quantity of concentrate provided in an automated milking system (AMS) affects dry matter intake (DMI), attendance to the AMS, milk and milk component yield, feeding behavior, cow activity, and ruminal fermentation of lactating dairy cows fed isocaloric diets. Eight ruminally cannulated primiparous Holstein cows were used in a replicated 4 × 4 Latin square design with 28-d periods. Cows were housed in a freestall facility with a guided-traffic (feed-first) flow barn design. Treatments included 0.5, 2.0, 3.5, or 5.0 kg/d of dry matter of pellet in the AMS with an equivalent reduction of the same pellet in the partial mixed ration (PMR). Days 21 to 24 of each treatment period were used for DMI, milking performance (visits, yield, and composition), behavior, and ruminal pH determination, and d 25 to 28 were used for ruminal short-chain fatty acid and ammonia concentrations as well as total-tract digestibility. As imposed, consumption of AMS pellet linearly increased, equating to 0.50, 2.00, 3.49, and 4.93 kg of dry matter/d for the 0.5, 2.0, 3.5, and 5.0 kg/d treatments, respectively. Correspondingly, the standard deviation in AMS pellet intake among days linearly increased from 0.06 to 0.85 kg of dry matter/d as the quantity of concentrate in the AMS increased from 0.5 to 5.0 kg. The PMR DMI decreased linearly with increasing AMS concentrate allocation, but total DMI (PMR + AMS) was not affected (25.3 kg/d). As the AMS concentrate allocation increased, the selection against particles retained on an 18-mm sieve linearly increased and selection against particles retained on the pan decreased. Milking frequency (3.22 milkings/d), milk yield (37.5 kg/d), milk fat yield (1.43 kg/d), and milk protein yield (1.22 kg/d) were not affected; however, milk urea nitrogen concentration decreased linearly with increasing AMS concentrate. Ruminal pH averaged 6.18 and was not affected by AMS concentrate. Total ruminal short-chain fatty acid concentration was greatest when 3.5 kg of concentrate was allocated in the AMS and ruminal ammonia decreased linearly with increasing AMS concentrate. Time spent lying, the number of lying bouts, and average bout duration were not affected by treatment. These data indicate that increasing the quantity of concentrate in the AMS increases daily variability in AMS concentrate intake while decreasing PMR intake without affecting voluntary visits to the AMS and milk or milk component yield. As such, under isocaloric dietary settings, increasing the supply of pellet in the AMS is not likely to affect voluntary visits to the AMS, milk and milk component yields, or ruminal fermentation.

Lameness in cattle is a health and welfare concern; however, limited information is available on risk factors and the relationship between lameness and common diseases like bovine respiratory disease (BRD). Therefore, the objectives of this study were to: 1) identify prevalence of lameness in feedlot cattle and related risk factors of cattle diagnosed as lame; and 2) determine associations between BRD occurrence and lameness. Feedlot cattle health records were available from 28 feedlots for 10 yr. The data set consisted of 663,838 cattle records, with 13.9% (92,156) diagnosed with a disease, including 32.3%, 46.0%, and 22.0% with lameness, BRD, and other diagnoses, respectively. Lameness was classified into four categories: foot rot (FR), joint infections (JI), lame with no visible swelling (LNVS), and injuries (INJ), with a prevalence of 74.5%, 16.1%, 6.1%, and 3.1%, respectively. Lameness was compared across cattle types (arrival date and weight) as well as age classification (calf vs. yearling), gender (steer vs. heifer), and season of placement in the feedlot (spring, summer, fall, and winter). Within the disease-diagnosed population, lameness represented 28.5% of treated fall-placed calves, 38.5% of winter-placed calves, and 40.8% of treated yearlings. Foot rot was the most common diagnosis with 74.5% of all lameness diagnoses, with winter- and fall-placed calves more likely to be diagnosed with FR compared to yearlings (OR: 1.19, 95% CI: 1.10–1.30 and OR: 1.46, 95% CI: 1.38–1.55, respectively). Joint infections were the second most common diagnosis (16.1%). Compared to yearlings, fall-placed calves had a higher odds (OR: 3.64, 95% CI: 3.12–4.24) for JI. Injuries and LNVS were the least common but again fall-placed calves had higher odds of this diagnosis compared to yearlings (OR: 2.26, 95% CI: 1.70–2.99 and OR: 9.10, 95% CI: 6.26–13.2, respectively). Gender was significantly different for JI as steers were less likely affected compared to heifers (OR: 0.687, 95% CI: 0.545–0.867), and more likely affected by LNVS (OR: 2.46, 95% CI: 1.57–3.84). Of all lameness-associated deaths, JI accounted for almost 50%. Finally, cattle diagnosed with BRD were subsequently more likely to be diagnosed with INJ, JI, or LNVS (P < 0.001 for all comparisons). In conclusion, animal type and gender were associated with type of lameness diagnoses, allowing feedlots to allocate resources to groups at highest risk and focus on early intervention strategies.

This study evaluated the effects of feeding ergot alkaloids (EA) at increasing levels (0, 0.75, 1.5, and 3.0 ppm) to feedlot beef cattle over backgrounding (BG) and finishing (FS) phases on growth performance and carcass quality. Steers (n=240; 280±32 kg) were blocked by weight and randomly allocated to 16 pens (4 pens/treatment; 15 steers/pen). Treatments included: no added EA (CTL); or EA added at 0.75 (EA075); 1.50 (EA150); and 3.0 ppm (EA300). Steers were fed barley-based BG diets containing 40% concentrate:60% silage (DM basis) for 84 d, then stepped up over 28 d to a 90% concentrate:10% silage (DM basis) FS diet for 119 d. Steers fed EA300 received CTL in FS after 77 days on feed (DOF). In the BG phase, a quadratic decrease (P=0.05) in ADG and a linear decrease in final BW (P=0.03) and total BW gain (P=0.02) were observed with increasing EA. In the FS phase, up to 77 DOF, there was a linear decrease in steers DMI (P=0.04), ADG (P=0.01), and total BW gain (P=0.01) with increasing levels of EA. Over the full FS phase (119 DOF), a quadratic response was observed for ADG (P=0.05), final BW (P=0.05), total BW gain (P=0.02) and carcass weight (P=0.05) with steers fed EA150 showing the lowest performance. Gain:feed was not affected (P>0.20) by EA in the BG or FS phase. Dressing percentage (P=0.02) showed a quadratic effect with EA300 being the lowest. Additionally, a linear increase in lean meat yield (P< 0.001) was observed with increasing EA. Including EA in the diet promoted a linear increase in rectal temperature (P< 0.01). In conclusion, increasing levels of EA in the diet reduced growth performance, but when EA was removed, steers showed compensatory weight gain.

Increased trailer motion, coupled with large accelerations and decelerations, has been associated with decreased carcass quality and increased stress indicators in cattle, sheep, and hogs. However, motion of livestock trailers has not been measured in North-American cattle semi-trailers over long distances (> 1000 km). The objectives of this study were to develop a practical method of measuring transport trailer accelerations, to describe the range of accelerations cattle are exposed to under North American conditions, and to conduct a preliminary analysis of trailer accelerations for each compartment and its effect on carcass bruising. The root mean square (RMS) of acceleration was measured at a sampling rate of 200 Hz in 3 orthogonal axes; x (vertical), y (front-to-rear), and z (lateral; side-to-side) by clamping an accelerometer to the cross beam below each of the five compartments of 8 trailers transporting a total of 330 animals (674 ± 33.3 kg BW) from an assembly yard to a processing facility. Journeys took place on separate days and ranged in duration from 13 to 15.7 h. The number and severity of bruises per carcass were determined prior to trimming for n = 290 carcasses and the number of bruises per carcass ranged between 0.38 and 12.75, whereas the bruising score per carcass ranged between 0.38 and 14.88. Mean number of bruises and severity of bruises (bruising scores were assigned according to size using a three-point scale: 1) ≤ 6.5 cm, 2) 6.5 to 12 cm, and 3) ≥ 12 cm and bruising severity was determined by applying the weighted score to each bruise according to bruise area) per carcass was 4.52 ± 2.43 (n) and 5.31 ± 2.84, respectively. Accelerations in commercial transport vehicles were found to range between 0.33 and 1.90 m/s2, whereas the mean RMS of acceleration for all trailers (n = 31 accelerometers) was 1.01 ± 0.32 m/s2, 0.72 ± 0.31 m/s2, and 0.97 ± 0.30 m/s2 for the x, y, and z axes, respectively. Horizontal acceleration was greatest in the nose, back, and doghouse compartments (P = 0.05), whereas lateral acceleration was greatest in the nose and back compartments (P = 0.08). Although the nose, back, and doghouse compartments had the highest RMS values for the lateral and horizontal axes, there were no significant relationships between bruising and acceleration. Replication of this research is required to further understand the relationships between trailer motion, carcass bruising, and overall animal welfare in cattle transported long distances.

Greater shrink (P < 0.01) was observed in C than N calves after the initial 20-h transport. The N calves had greater (P < 0.01) ADG than C calves between L1 and d 5, while C had greater (P < 0.01) ADG than N calves between 14 and 28 d. L-lactate concentrations and flight speed were greater (P ≤ 0.05) in C than N calves between L1 and d 5. The R8-T4 calves had greater (P < 0.01) ADG than R8-T15 calves between L1 and d 5. The R0-T4 calves had greater (P = 0.02) L-lactate concentrations than R0-T15 and R8-T4 calves on d 1. The R0 calves had greater (P < 0.01) ADG than R8 calves between 14 and 28 d. Preliminary results show physiological, behavioral, and performance differences across treatments, however, additional indicators are required to accurately assess the effect of conditioning, rest, and post-rest transport durations on calf welfare.

The epidemiology of hoof-related lameness (HRL) in western Canadian feedlots, with a focus on digital dermatitis (DD), was described and analyzed to help inform recommendations on lameness control and prevention in western Canadian feedlot cattle. The retrospective data in this study were accessed from 28 western Canadian feedlots that placed cattle in 2014–2018, inclusive. The total population for this study was 1,796,176 cattle, with an annual placement average of 12,830 cattle per feedlot. These data were accessed through iFHMS Consolidated Database, provided by Feedlot Health Management Services by TELUS Agriculture, and manipulated using Microsoft® Office Access 365 ProPlus and Microsoft® Office Excel 365 ProPlus. Epidemiological analyses determined that lameness accounts for 25.7% of all treatments in western Canadian feedlots. Of those treatments, 71.7% are localized to the hoof, corresponding to 18.6% of all treatments. The most common HRL diseases are infectious bovine pododermatitis [foot rot (FR)]; digital dermatitis (DD), also known as hairy-heel wart or strawberry foot rot; and toe-tip necrosis syndrome (TTNS). These diseases account for 89.6%, 7.9% and 2.4% of HRL, respectively. Between 2014 and 2018, HRL prevalence ranged between 1.93% and 3.09% of the population, with FR consistently having the highest prevalence and TTNS the lowest. HRL and DD were tested for their associations with several animal-level risk factors using © Ausvet 2021 Epitools software. The resultant crude, univariate odds ratio values, evaluated at 95% confidence, are summarized in Table 1. Based on this analysis, acquisition source has the largest influence on the odds of developing HRL and DD, followed by population size, and placement quarter. Using SAS® (Version 9.4, SAS Institute Inc, Cary, North Carolina) statistical software, these preliminary findings will be subjected to a multivariate statistical model, which will provide adjusted OR values and statistical significance for the data in this study.

Blood metabolites are used to assess a variety of animal conditions for veterinary diagnosis and research. Concentration of metabolites in blood can be measured using a commercially-available lab-based assay or in real-time using a handheld device developed to be more time- and cost-effective than the lab-based method. Lactate is a product of anaerobic glycolysis, used in animal research as an indicator of muscle fatigue. Therefore, it has been used as an indicator of cattle response to long distance transportation. The aim of this study was to assess the relationship of L-lactate concentrations measured using a Lactate Scout+ analyzer (Lactate Scout, EFK Diagnostics, Barleben, Germany) and a lactate assay colorimetric kit (Lactate Assay Kit, Cell Biolabs Inc., San Diego, CA). Blood samples were collected by venipuncture from 96 steers (245 ± 35.7 kg BW) prior to (L1) and after 36 h, and prior to and after an additional 4 h of road transportation, and on d 1, 2, 3, 5, 14, and 28 after transport. The Lactate Scout+ analyzer strip was dipped in blood at the time of sampling, while blood samples were collected into sodium fluoride tubes for use in colorimetric analysis. Pearson correlations were calculated to determine the relationship between the experimental methods for the quantification of L-lactate concentrations. The strengths and levels of statistical significance of the correlation varied over the observed time points, r = -0.03, P = 0.75 (L1) to r = 0.75, P = < 0.0001 (d 3). The correlation for the pooled data was weak but statistically significant (r = 0.33, P < 0.001). Based on the experimental results, the Lactate Scout+ analyzer is not a suitable alternative to a lab-based assay for measuring L-lactate in transported cattle, due to variability across sampling time points and weak correlation with the traditional enzymatic method.

Lameness is a significant economic and welfare issue in feedlot production. Effective treatment depends on diagnosing the cause of lameness accurately. There is increasing evidence that circulating microRNAs may serve as useful biomarkers for diagnosing disease. However, their association with lameness in feedlot cattle is currently unknown. The objective of this study was to investigate whether blood miRNA profiles can be associated with a specific lameness phenotype. Blood samples were collected via venepuncture (into Tempus™ Blood RNA Tubes) from 39 feedlot cattle diagnosed with digital dermatitis (DD; n = 2), toe tip necrosis syndrome (TTNS; n = 5), footrot (FR; n = 26) and healthy controls (HC; n = 6) for miRNA profiling using RNA sequencing. Samples were obtained at the time each animal was pulled from their pens for medical treatment. Total RNA was extracted from blood samples and subjected to small RNA libraries construction and RNA-sequencing. The sequence data analysis was performed using a web-based tool, sRNAtoolbox. A total of 596 miRNAs were identified across 39 blood samples with the expression of 444, 437, 465 and 575 miRNAs detected in the HC, DD, TTNS and FR groups, respectively. In addition, group-specific miRNAs were identified with 9 in the DD group, 45 in the TTN group, 28 in the FR group and 2 in the HC group. Moreover, 41, 8 and 36 differential expressed miRNAs (DE miRNAs) were detected in DD, TTNS and FR groups, respectively, when compared to the HC group using DeSeq2. These data suggest that miRNA profiles may differ according to lameness diagnosis in beef cattle. However, further investigation incorporating functional analyses of miRNA, increased sample size and the physiological profile of the animals are required to better understand the relationship between blood miRNA profiles and lameness in feedlot cattle.

The aim of this study was to assess the effect of an 8h rest stop after 36 h of road transport on physiological and behavioral indicators of welfare in 7–8-mo-old conditioned and non-conditioned beef calves, sourced from either a ranch or an auction market. Three hundred and twenty weaned calves (245 ± 35.7 kg BW) were randomly assigned to a 2 × 2 × 2 factorial design: conditioning, conditioned (C) or non-conditioned (N); source, ranch direct (R) or auction market (A); and rest, 0 (R0) or 8 (R8) h. After resting, animals were transported for an additional 4 h. A subset of 12 animals/treatment were sampled for non-esterified fatty acids (NEFA), serum amyloid-A (SAA) and creatine kinase (CK) concentrations prior to the first loading (L1); after 36 h of transport; prior to and after the additional 4 h of transport, on the day of arrival (0); and 1, 2, 3, 5, 14, and 28 d after the 4h transport. Daily standing % and dry matter intake (DMI) were assessed for 3 d after transport. Data were analyzed using the GLIMMIX procedure of SAS. Fixed effects included conditioning, source, and time (nested in rest), while random effects were animal and pen. At L1, mean concentrations of NEFA, SAA and CK were greater (P < 0.05) for N-R0 than C-R0 and for N-R8 than C-R8. The N and R groups had greater (P < 0.05) standing % than the C and A groups, respectively. On d 0 and 1, the C-R0 and C-R8 groups had greater (P < 0.05) DMI than the N-R0 and N-R8 groups, respectively. Overall, physiological and behavioral indicators of welfare were greater in conditioned than non-conditioned calves, while fewer differences were observed between ranch and auction market calves, as well as rested or unrested calves.

One hundred and thirty-one 2 mo. old pasture housed Angus cross bull calves were evaluated for 62 d over two years (Year 1: n = 69, 134.1 ± 20.37 kg BW; Year 2: n = 62, 118.1 ± 15.49 kg BW) to determine 1) the effects of a subcutaneous (s.c) injection of meloxicam on indicators of long term pain after castration and 2) the potential use of cow-calf proximity and home range as indicators of pain. Calves were randomly assigned to treatments using a 3 × 2 factorial design including castration - sham (CT; n = 47), band (BA; n = 46) or knife (KN; n = 38) castration and medication – s.c. meloxicam (M; n = 66) or s.c. lactated ringers solution (NM; n = 65). Measurements included performance, scrotal temperature, swelling (WS) and healing (WH) scores, and pain sensitivity, collected on d -1, 6, 13, 20, 34, 48, and 62 post-castration. Suckling, lying, standing and walking duration, and head-turning, lesion-licking, foot-stamping and tail-flick frequencies were collected immediately following and up to 2-d after castration. Cow-calf proximity and home range were obtained from d 0 to 2 and from d 14 to 16. With the exception of suckling, no medication (P > 0.05) effects were found. Greater (P < 0.05) pain sensitivity was observed in KN from d 6 to 34 and on d 62, and in BA from d 6 to 62 compared to CT calves. Knife calves showed an earlier (d 20) absence of inflammatory responses (WS; P < 0.05) than BA (d 34) and overall, KN calves had greater (P < 0.05) standing, walking, and head turning than BA and CT. Knife and BA had greater (P < 0.05) foot stamping than CT for the first 2 h post-castration, but KN exhibited greater (P < 0.05) frequencies between 9 and 11 h (d 0) compared to BA and CT, and had greater (P < 0.05) tail flicks from d 0 to 2 than CT. Banded calves were closer to their dams on d 15 while KN calves and their dams had a reduced home range on d 0 than CT cow-calf pairs. Although meloxicam did not reduce indicators of pain (with exception of suckling behavior), our results suggest that knife castration causes greater acute pain, while band castration resulted in greater chronic-pain. Consumers are increasingly concerned about the welfare of farm animals, especially during transportation. The Canadian transport regulations state that weaned cattle require an 8 h rest after 36 h of transport. There are, however, only a few studies that assess the effect of rest on the welfare of beef cattle. The aim of this study was to assess the effect of conditioning, source and rest on indicators of welfare in 7-8-month-old beef calves during long-distance transport. Treatments consisted of a 2 × 2 × 2 factorial design where the main factors were conditioning: conditioned (C; n = 160) or non-conditioned (N; n = 160); source: auction market (A; n = 160) or ranch direct (R; n = 160); and rest: 0 h (0 h; n = 160) or 8 h (8 h; n = 160). Means of non-esterified fatty acids (NEFA), serum amyloid-A, haptoglobin, creatine kinase (CK), and percentage of time standing from N calves were greater than C calves (all p ≤ 0.05). Means of percentage of time standing and CK of R calves were greater than A calves (both p ≤ 0.05). The mean of NEFA of 0 h calves was greater than the 8 h calves (p < 0.01), while the percentage of time standing of 0 h calves was less than 8 h calves (p < 0.01). Statistically significant differences between means of NEFA and standing percentage, were observed between 0 h and 8h calves. Few and inconsistent indicators of reduced welfare were observed between auction market and ranch direct calves, while non-conditioning was associated with greater physiological and behavioural indicators of reduced welfare. Based on these results, conditioning should be implemented as a management practice to improve the welfare of transported calves.

This study was conducted to evaluate the effects of the forage-to-concentrate ratio of the partial mixed ration (PMR) and the quantity of concentrate offered in an automated milking system (AMS), in a feed-first guided-flow barn, on the behavior and performance of dairy cows. Eight ruminally cannulated multiparous Holstein cows were used in a replicated 4 × 4 Latin square balanced for carry-over effects. Treatments were arranged in a 2 × 2 factorial consisting of a PMR that contained (dry matter basis) either a low (54:46; L-FOR) or a high (64:36; H-FOR) forage-to-concentrate ratio and AMS concentrate provision to achieve low (2 kg/d; L-AMS) or high (6 kg/d; H-AMS) intake. Each period consisted of 28 d with 6 d for dietary transition, 13 d for adaptation, and 9 d of collection. The first 4 d of data and sample collection were used to evaluate behavioral data (milking frequency, feeding behavior, and standing and lying behavior) and ruminal pH. Subsequently, a sampling device removal day was provided, and the last 4 d were used to evaluate ruminal fermentation and apparent total-tract digestibility. All 9 d were used for milk yield measurement, and the 8 d were used for dry matter intake measurement. Cows fed the H-AMS consumed 3.5 kg/d less PMR while consuming 4.2 kg/d more AMS concentrate, but total dry matter intake (PMR+AMS) was not affected by treatments averaging 27.3 kg/d. Although cows fed H-AMS had greater concentrate intake, they also had greater variability for AMS concentrate intake among days (0.85 vs. 0.25 kg/d, respectively). The number of PMR meals and PMR eating behavior were not affected by the PMR or AMS treatments. Feeding H-AMS did not affect milking frequency averaging 3.63 milkings/d, but tended to increase milk yield by 1.25 kg/d relative to L-AMS. Likewise, cows fed the L-FOR tended to have greater milk yield relative to H-FOR (39.3 vs 37.9 kg/d, respectively), but had greater holding area time. Minimum ruminal pH tended to be lower for cows fed L-FOR compared with cows fed H-FOR but was not affected by the AMS concentrate treatment. When fed the L-FOR, feeding the H-AMS increased total short-chain fatty acid concentration in the rumen relative to cows fed L-AMS, whereas the response for H-FOR was not affected by the AMS concentrate. These data suggest that feeding H-AMS may improve milk yield, but also increases the day-to-day variability in AMS concentrate consumption. Feeding a L-FOR PMR may increase milk yield without affecting variability in AMS concentrate consumption; however, it may reduce ruminal pH and increase the time spent in the holding area compared with cows fed a H-FOR PMR.

The aim of this study was to evaluate the effect of a single dose of subcutaneous (s.c.) meloxicam administered at the time of knife and band castration on inflammatory response and wound healing over 56-d post-castration. Seventy-two Angus crossbred calves (47.3 ± 6.70 kg of body weight [BW] and 7 to 8 d of age) were randomly assigned according to a 3 × 2 factorial design assessing castration method: sham (CT), band (BA), and knife (KN) castration, and pain mitigation: non-medicated (NM) and medicated calves (M) injected s.c. with meloxicam (0.5 mg/kg of BW). Calf BW, rectal temperature, swelling ("0": no swelling; "4": swelling needing intervention) and healing score ("1" to "5" with "5" being completely healed), scrotal circumference, and maximum scrotal temperature were measured on d -1, immediately before castration (d 0), and weekly thereafter over a 56-d period. Blood samples for haptoglobin (Hp), serum amyloid-A (SAA), and complete blood cell count were collected according to the same schedule. Hair samples were collected on d -1, 28, and 56 to determine cortisol concentrations. Standing and lying behaviors were measured using accelerometers that were placed on the calves on d -1 until d 35, and visual observations of behaviors related to pain were recorded once a week for 35 d. Knife-castrated calves achieved swelling scores of "3" and "2" between d 7 and 14, which was sooner (Z 0.10) were observed among treatments for hair cortisol on d -1 and 28, but on d 56, hair cortisol concentrations in BA-NM calves were greater (P > 0.05) than for CT-NM, BA-M, KN-NM, and KN-M, and tended to be greater (P = 0.08) than for CT-M calves. Lying duration tended (P = 0.10) to be greater and suckling behavior tended (P = 0.08) to be lower in NM than M calves. A single s.c. injection of meloxicam did not reduce long-term inflammatory responses or improve wound healing; however, it may be useful in reducing pain and stress in band castrated calves as evidenced by reduced hair cortisol concentrations up to 56 d post-castration.

In Western Canada, approximately half of the calves produced are castrated before 1 wk of age. Therefore, it is important to identify effective analgesic drugs to mitigate pain associated with castration and consequently improve animal welfare. The aim of this study was to assess the efficacy of a single s.c. dose of meloxicam at mitigating pain associated with knife and band castration in 1-wk-old calves. Seventy-two Angus crossbred bull calves (47.3 ± 6.70 kg of body weight (BW), 1 wk old) were used in a 3 × 2 factorial design where main factors included castration method–sham (CT), band (BA) or knife (KN) castration– and medication–lactate ringer (NM) or 0.5 mg/kg BW of meloxicam (M). Measurements included different physiological and behavioral parameters. Samples were collected on day −1, immediately before castration (T0); and 60, 90, and 120 min and 1, 2, 3, and 7 d after castration except for visual analog scale (VAS) which was collected at the time of castration. The salivary cortisol concentrations were greater (P = 0.04) in KN and BA calves than CT calves 60 min after castration, while 90 min after castration BA had greater concentrations than CT calves. Substance P concentrations were greater (P = 0.04) in NM calves than M calves on d 3 and 7 after castration. The serum amyloid-A (SAA) concentrations were greater (P = 0.05) in KN calves than BA and CT calves on days 0, 2, and 3, while BA calves had greater SAA concentrations on day 7 than KN and CT calves. The visual analog scores were greater (P < 0.01) in KN calves than BA, and in BA compared to CT calves. The KN calves tail flicked more (P < 0.01) than BA and CT calves, and NM calves tail flicked more (P = 0.03) than M calves. No castration or medication effect (P > 0.10) was observed for stride length, walking, standing, lying ventral, eating, foot stamping, head turning, lying and standing percentage, performance, platelets, or body temperature. Overall, knife castrated calves exhibited a greater acute pain response than band castrated calves. Meloxicam was able to reduce substance P concentrations, white blood cell counts and number of tail flicks after castration, suggesting that the drug was able to mitigate acute pain to some extent. However, meloxicam did not have an effect on the other physiological and behavioral parameters assessed.

In Canada, cattle must be unloaded, fed, watered, and rested after 36 h of transport; however, little is known about what constitutes appropriate rest station conditions. The objective of this study was to determine the relationship between providing straw bedding (14-cm deep) and trip, load, and commercial rest station characteristics, on lying behaviour. Truckloads (n = 13) were split; half the animals were assigned to either bedded (n = 452 cattle) or non-bedded (n = 470 cattle) pens. Trip characteristics [time in motion (TIM), duration of stops en route (DUR_STOPS)], load characteristics (sex: heifers, steers, both; and load weight), and rest station characteristics [ambient temperature at unloading; rest pen space allowance (k_PEN); time in resting pen (TIP)] were recorded. Once unloaded we counted the number of cattle lying/pen, every 10 min for 8 h. A mixed logistic regression model with random intercepts for load and truck compartment was fitted to examine associations between the proportion of animals lying and the independent variables (i.e., treatment, trip, load, and rest station characteristics). Odds of lying increased with load weight (i.e., cattle weight class, P = 0.02) and with DUR_STOPS (P < 0.03). There was an interaction (P < 0.01) between treatment and TIM: as TIM increased, the odds of observing cattle lying showed a notable increase for cattle rested in bedded pens, whereas for those rested in non-bedded pens, the odds showed little change as TIM increased. An interaction (P < 0.01) was also found between treatment and TIP: early in the observation period, the odds of cattle lying were greater in bedded pens. Both groups showed an increase in the probability of lying over time, plateauing at similar levels, near the end of the 8-h observation period. In conclusion, providing straw bedding at rest stations influenced cattle’s motivation to lie, particularly following longer transport durations.

A rumen simulation technique (RUSITEC) investigated effects of ergot alkaloids (E) and a mycotoxin deactivating product (Biomin® AA; B) on nutrient digestion, rumen fermentation, total gas and methane emissions. Fermenters received a finishing diet of 90:10 concentrate:barley silage. Using a randomized complete block design, treatments were assigned (4 vessels/treatment) within two RUSITECs in a 2 × 2 factorial. Treatments included: 1) control diet, 2) control + 1 g B, 3) control + 20 ppm E, and 4) control + 20 ppm E + 1 g B. The study had a 14-d experimental period, with 7-d adaptation and 7-d sample collection. Data were analyzed in SAS using PROC mixed including fixed effects of E, B, and E×B interaction. Random effects included RUSITEC apparatus and cow rumen inoculum (n = 4). Ergot decreased dry matter disappearance (DMD) (P < 0.05; 87.9 vs. 87.2%) and organic matter disappearance (OMD) (P < 0.05; 88.8 vs. 88.4%). Adding B increased OMD (P < 0.05; 88.3 vs. 88.9%) and neutral detergent fiber disappearance (NDFD) (P < 0.01); however, an E×B interaction was observed for NDFD (P < 0.01) with B promoting greater increases with E. Ergot decreased acetate proportions (P < 0.01) and increased isovalerate (P < 0.05). Consequently, acetate:propionate was reduced (P < 0.05) with E. Inclusion of B increased total volatile fatty acids (P < 0.01), and proportions of acetate (P < 0.05) and propionate (P < 0.05), and decreased valerate (P < 0.01), isovalerate (P < 0.01), and caproate (P < 0.01). Treatments did not affect (P ≥ 0.17) ammonia-N, total gas, or methane production (mg/d or mg/g of OM fermented). In conclusion, E reduced OMD and acetate production, but these responses were reversed by the addition of B.

Lameness is an important health issue in feedlot cattle; however, there is a paucity of information regarding its economic impact. Decision tree models are excellent tools for assessing costs of disease such as the net return (net return = benefit – cost). Models were developed using expert opinion, literature and retrospective feedlot data provided by Vet-Agri Health Services (VAHS, Airdrie, Alberta, Canada) collected from 2005 to 2015 on individually treated cattle (n = 30,940) from 28 feedlots. The objective was to estimate net return of various lameness diagnoses and impacts of cattle type, season of treatment, and extreme high and low cattle prices. Cattle were diagnosed as lame according to the following categories: foot rot, foot rot in heavy cattle (BW > 363 kg at treatment), injury, lame with no visible swelling, and joint infection. Records consisted of arrival and treatment weight, cost of treatment, and cattle deaths. Records included cattle types classified as: fall calves (heifer and steer), winter calves (heifer and steer) and yearling cattle (heifer and steer). Lastly, variables ADG, days on feed (DOF), and Season (spring, summer, fall, and winter) were created. Models estimated net return using cattle slaughter prices for healthy cattle that reached a slaughter weight of 635 kg and for three possible outcomes for each diagnosis after final treatment: cattle that recovered after treatment and reached a slaughter weight of 635 kg; cattle that were removed before they reached slaughter weight; or cattle that died. Compared to undiagnosed cattle with 1.36 kg/d ADG, cattle diagnosed with foot rot and foot rot heavy cattle had the highest ADG until first treatment (1.14 and 1.57 kg/d, respectively) and differed significantly (P < 0.05) compared to cattle diagnosed with injuries (0.87 kg/d), lame with no visible swelling (0.64 kg/d), and joint infections (0.53 kg/d). Yearling steers had the most positive returns compared to all other cattle types. Cattle with lighter arrival weight had lower ADG and increased economic losses after treatment compared to heavier weighted cattle on arrival. Based on average slaughter prices over a 10-yr period for healthy cattle, return was $690. Return after final treatment for cattle with foot rot was $568, foot rot in heavy cattle was $695, and injury was $259. However, joint infections and lame with no visible swelling had negative returns of –$286 and –$701, respectively.

The newly revised Canadian Codes of Practice for the management of beef cattle requires that as of 2018, calves older than 6 mo of age be castrated using pain control. Castration is a husbandry procedure commonly done without pain control, and there is a lack of agreement on an effective pain mitigation strategy specific to castration. The aim of this study was to identify the optimal time of administration of meloxicam prior to castration. Thirty-four Angus and Angus crossbred bull calves (282 ± 28.0 kg BW) were randomly assigned to 1 of 3 treatments receiving a single s.c. injection of meloxicam (0.5 mg/kg BW): 6 h (6H; = 11), 3 h (3H; = 12), or immediately (0H; = 11) before knife castration. Measurements included visual analog scale (VAS), head movement (HM), accelerometer movement (AM) and strain gauge exertion force (EF) on the squeeze chute, stride length (SL), lying and standing behavior, salivary cortisol (SC), haptoglobin, serum amyloid A (SAA), substance P (SP), and scrotal temperature (ST). Samples were collected on d -7, -5, -2, -1, and immediately before castration (T0) and 30, 60, 120, and 240 min and 1, 2, 5, 7, 14, 21, and 28 d after castration, except for VAS, AM, EF, and HM, which were obtained at the time of castration. A time × treatment effect ( = 0.01) was observed for SP, where 0H had lower concentrations than 3H and 6H calves 1 d after castration, whereas 3H calves tended to have greater levels than 6H calves 5 d after castration. Mean ST was greater ( 0.10) were observed for the number of peaks and area for AM and EF, VAS, HM, SC, or haptoglobin. On the basis of these results, the optimal time to administer s.c. meloxicam in 7- to 8-mo-old knife-castrated calves is immediately before castration (0H), as evidenced by fewer indicators of pain and inflammation compared to 3H and 6H calves.

Three experiments were conducted to evaluate the effects of band and knife castration on behavioral and physiological indicators of chronic pain in beef calves at 3 different ages (36 calves/age group): 1 wk of age (Exp. 1, 4 ± 1.1 d of age, 43 ± 1.1 kg BW), 2 mo of age (Exp. 2, 63 ± 2.3 d of age, 92 ± 1.7 kg BW), and 4 mo of age (Exp. 3, 125 ± 4.6 d of age, 160 ± 3.4 kg BW). In each experiment calves were randomly assigned to either sham (CT), band (BA) or knife (KN) castration. Experiments 1, 2, and 3 ended when the testicles of banded calves had sloughed off (68, 49, and 42 d, respectively). Animal BW and rectal temperature were recorded weekly over the experimental period. Salivary cortisol, substance P, haptoglobin, scrotal area temperature using infrared thermography, visual evaluation of swelling (5-point scale), and gait stride length were collected on d -1 and immediately before castration and weekly thereafter until the end of the study. Hair samples were collected 1 d prior to and 28 d after castration and at the end of the study for cortisol concentration. Standing and lying behaviors were recorded over a 28-d period immediately after castration. No differences ( > 0.10) were observed in salivary cortisol, substance P, haptoglobin, or hair cortisol among castration methods for any of the 3 ages. No changes in behavior were observed in calves castrated at 1 wk or 2 mo of age. In 4 mo-old-calves, BA spent less time lying ( < 0.01) than CT and KN calves. Also, the average duration of lying time for BA calves was greater ( < 0.05) than for CT calves. Both, 1-wk- and 2-mo-old calves had inflammation in the scrotal area lasting 7 d after KN castration, whereas inflammation was observed for up to 14 d in 4-mo-old calves. Swelling in BA calves lasted for 21 to 28 d in the 2 younger groups of calves, whereas in 4-mo-old calves swelling was observed until d 35 postcastration. Knife- and band-castrated calves did not exhibit indicators of chronic pain or distress when the procedures were performed in calves younger than 2 mo of age. Therefore, pain mitigation should be used when castrating to improve animal welfare, especially when castrations are performed in calves older than 2 mo of age independent of the method of castration.

Infectious and non-infectious feet lesions (FL) in livestock have been associated with reduced welfare, production, carcass and meat quality. Currently, the incidence of FL in sheep and its relationship with carcass quality has not been documented in Canada. The objectives of this study were to determine 1) the incidence and type of lesions in Alberta sheep at slaughter and 2) the relationship between FL and carcass quality. A total of 4,487 sheep carcasses were assessed for FL at a slaughter plant in Alberta, Canada between October 2017 and March 2019. Approximately 300 sheep were evaluated monthly by two experienced observers who recorded common infectious (IN) and non-infectious (NIN) lesions. In addition, carcass information including hot carcass weight (HCW), and back fat thickness (BFT) were obtained from the slaughter plant records. Data were analyzed by multivariable regression models using SAS PROC GLIMMIX. Overall, FL incidence was 9.4% (7.0% having one lesion and 2.4% having more than one lesion). Infectious lesions accounted for 45.7% of all lesions [footrot (37.1%), interdigital dermatitis (7.3%), and contagious ecthyma (1.3%)], while NIN lesions accounted for 54.3% [overgrown horn (21.9%), injury (13.6%), laminitis (10.2%), and other lesions (8.6%)]. Sheep with IN had lower BFT (13.5 ± 1.32 cm, P = 0.0002) and HCW (51.5 ± 1.89 kg, P = 0.0189) than NIN (15.8 ± 1.51 cm and 54.7 ± 1.97 kg, respectively). For every 1 cm decrease in BFT and 1 kg decrease in HCW the odds of IN increased by 0.89 (P < 0.0001) and 0.98 (P = 0.0229), respectively. Based on our findings, foot lesions are a significant issue for the Canadian sheep industry and infectious lesions may have greater detrimental effect on carcass quality than non-infectious lesions. Further studies are necessary to understand the relationship of carcass quality and feet lesion.

Biomarkers are commonly used to assess pain and analgesic drug efficacy in livestock. However, the diagnostic sensitivity and specificity of these biomarkers for different pain conditions over time have not been described. Receiver operating characteristic (ROC) curves are graphical plots that illustrate the diagnostic ability of a test as its discrimination threshold is varied. The objective of this analysis was to use area under the curve (AUC) values derived from ROC analysis to assess the predictive value of pain biomarkers at specific timepoints. The biomarkers included in the analysis were blood cortisol, salivary cortisol, hair cortisol, infrared thermography (IRT), mechanical nociceptive threshold (MNT), substance P, and outcomes from a pressure/force measurement system and visual analog scale. A total sample size of 7,992 biomarker outcomes were collected from 6 pain studies involving pain associated with castration, dehorning, lameness, and surgery were included in the analysis. Each study consisted of three treatments; pain, no pain, and analgesia. All statistics were performed using statistical software (JMP Pro 14.0, SAS Institute, Inc., Cary, NC). Results comparing analgesia verses pain yielded good diagnostic accuracy (AUC > 0.7; 95% CI: 0.40 to 0.99) for blood cortisol (timepoints 1.5, 2, and 6 hours); IRT (timepoints 6, 8, 12, and 72 hours); and MNT (timepoints 6, 25, and 49 hours). These results indicate that ROC analysis can be a useful indicator of the predictive value of pain biomarkers and certain timepoints seem to yield good diagnostic accuracy while many do not.

Ovariectomy (spaying) using the trans-vaginal dropped ovary technique (DOT) is performed to prevent pregnancy in cull female beef cattle that are not retained for breeding stock in areas practicing extensive grazing management. There are no reports describing analgesia for this surgical procedure. The objective of this study was to measure behavioral and physiological responses to determine whether an analgesic protocol of BXK [butorphanol (0.01 mg/kg), xylazine (0.02 mg/kg), and ketamine (0.04 mg/kg)] injected intramuscularly (i.m.) before spaying could mitigate procedural and immediate postsurgical pain, and whether oral meloxicam (1 mg/kg) administered at the time of spaying could mitigate postsurgical inflammatory pain. Forty-four red Angus and Angus crossbred yearling heifers (322 ± 27.0 kg BW) were randomly allocated to 1 of 3 groups: PALP (control; palpated but not spayed; n = 14), SPAY (spayed with no analgesia; n = 15), and BXKM (spayed with analgesia; n = 15). Behavioral measurements included visual analog scale (VAS) score, flight speed (FS), stride length (SL), and gait score (GS), as well as activity (lying, standing) and feeding behavior. Physiological measurements included salivary cortisol (SC), haptoglobin (Hp), serum amyloid A (SAA), substance P (SP), complete blood count (CBC), and rectal temperature (RT). Saliva and blood samples were collected, and RT, FS, SL, and GS were measured on day −1, day 0 (time of palpation/spaying), and hours 1, 2, 4, and days 1, 2, 4, and 7 after palpation/spaying. The BXKM heifers had lower SC concentrations than SPAY heifers at 1 h (P = 0.01) and 2 h (P = 0.004). Heifers treated with BXKM had Hp concentrations lower than SPAY heifers at 2 d (P = 0.01), 4 d (P < 0.001), and 7 d (P = 0.008), and lower Hp concentrations than PALP heifers at 4 d (P < 0.001). Concentrations of SAA were greater (P = 0.04) in BXKM heifers than in PALP heifers at 1 h and lower in PALP heifers than in BXKM heifers (P = 0.02) and SPAY heifers (P = 0.05) at 1 d. Heifers in the BXKM group had higher RT than PALP and SPAY heifers at 1 h (P < 0.001) and 2 h (P = 0.004). Results suggest that DOT ovariectomy is acutely stressful and painful and administration of BXK before spaying and meloxicam at the time of spaying mitigated the procedural and postsurgical stress, pain, and inflammation.

Infrared thermography (IRT) has been used as a non-invasive tool to detect inflammatory processes associated with disease in livestock. The aim of this study was to evaluate IRT as a tool to compare healthy and footrot (FR) affected hooves in feedlot lambs with varying degrees of lameness over two seasons. A total of 106 lame lambs with footrot from a feedlot in Alberta were individually categorized according to a 3-point locomotion scale [1 = mild (n = 7), 2 = moderate (n = 46) and 3 = severe lameness (n = 53)] during the summer (n = 39) and fall (n = 68) of 2018. All lambs were physically examined once by two experienced observers to determine if the lamb had footrot. IRT images of the interdigital space were used to obtain the maximum hoof temperature (MHT) of both FR affected as well as healthy (CT) hooves within the same animal. Generalized linear mixed models (SAS PROC GLIMMIX) were performed separately for each season and diagnosis and included locomotion score as a fixed effect and ambient temperature as a co-variate. Predicted means were compared using the limits at 95% of confidence. Overall, greater MHT (P < 0.05) were observed for FR affected compared to unaffected hooves for lambs categorized as moderately and severely lame, within each season. However, no differences (P > 0.05) in MHT were observed for lambs categorized as mildly lame, likely due to the small number of lambs having a locomotion score of 1. Under the conditions of this study, thermal images can be effectively used as a tool to distinguish footrot affected hooves in feedlot lambs with moderate and severe lameness. Further studies should be conducted with more lambs with a locomotion score of 1 to assess the relationship between mild lameness, IRT, and footrot diagnosis.

Forthcoming revisions to Canadian Transport regulations indicate un-weaned and weaned calves can be transported a maximum of 12 and 36 h, respectively, before an 8 h rest is required. The aim of this study was to assess the effect of rest duration after 12 and 36 h of transport on physiological and behavioral indicators of welfare in 7–8 mo old beef calves. Three hundred and twenty weaned calves (258 ± 23.9 kg BW) were randomly assigned to a 2 × 4 factorial design: 12 and 36 h of transport; and 0 (R0), 4 (R4), 8 (R8), and 12 (R12) h of rest. After the resting period animals were transported for an additional 4 h. A subset of 12 animals/treatment were sampled for non-esterified fatty acids (NEFA), haptoglobin and lactate concentrations prior to, and after the first and the 4 h transport, and 7 h, 2 and 28 d after the 4 h transport. Standing and lying behavior was assessed for 14 d after transport. Data was analyzed using the GLIMMIX procedure of SAS, where transport, and time nested within rest period were fixed effects and animal was a random effect. NEFA concentrations were greater (P < 0.01) in 12-R4 than 12-R8 and 12-R12 calves, while 36-R0 calves had greater (P ≤ 0.05) NEFA concentrations than 36-R4, 36-R8 and 36-R12 calves after the 4 h transport. Haptoglobin concentrations were greater (P < 0.01) in 36 than 12 h calves. No differences (P > 0.10) were observed for lactate. The day after transport, 36-R8 calves spent more (P < 0.01) time lying than 12-R8 calves. Overall, physiological indicators were greater in calves transported for 36 than 12 h, while no differences were observed between rest stops with the exception of NEFA, where overall concentrations were greater after shorter than longer rest periods.

Agri-food supply chains in North America have become remarkably efficient, supplying an unprecedented variety of items at the lowest possible cost. However, the initial stages of the COVID-19 pandemic and the near-total temporary loss of the foodservice distribution channel, exposed a vulnerability that many found surprising. Instead of continued shortages, however, the agri-food sector has since moved back to near normal conditions with prices and production levels similar to those typically observed in years prior to the pandemic. Ironically, the specialization in most food supply chains designed for “just-in-time” delivery to specific customers with no reserve capacity, which led to the initial disruptions, may have also been responsible for its rapid rebound. A common theme in assessing the impacts across the six commodities examined is the growing importance of understanding the whole supply chain. Over the longer term, a continuation of the pandemic could push the supply chain toward greater consolidation of firms and diversification of products given the increasing option value of maintaining flexibility. Other structural changes will be felt through input markets, most notably labour, as the trend toward greater automation will continue to accelerate as a response to meeting concerns about a consistent supply of healthy and productive workers. The economic fall out from the pandemic may lead to greater concentration in the sector as some firms are not able to survive the downturn and changes in consumer food buying behaviour, including movement toward online shopping and enhanced demand for attributes associated with resiliency, such as local. On the other hand, online shopping may provide opportunities for small producers and processors to shorten supply chains and reach customers directly. In the long term, COVID-19 impacts on global commerce and developing country production are more uncertain and could influence poverty reduction. While COVID-19's impacts on North American agriculture should have minimal effect on the Sustainable Development Goals (SDGs) through food prices, the ongoing global trends in trade and agribusiness accelerated by the pandemic are relevant for achievement of the SDGs., Highlights • Initial stages of the COVID-19 pandemic exposed a vulnerability in agri-food sector supply chains. • Most sectors have since moved back to near normal conditions. • Importance of understanding the whole supply chain is growing. • The trend toward greater automation will continue to accelerate.

Forty newly weaned beef calves (260 ± 32.6 kg) were transported 15 h in a livestock trailer (7.3 by 2.1 m) on 2 separate hauls 1 wk apart (20 calves/haul) to evaluate the effect of rest stop duration on indicators of calf welfare. Immediately following the 15-h journey, 15 calves/haul were randomly unloaded at a feedlot and randomly assigned to 1 of 3 rest stop treatments; calves without resting time (5 calves/haul) remained on the trailer and were used as the control group. Treatments included 0- (Control [CON]), 5- (RS5), 10- (RS10), or 15-h (RS15) rest periods in pens containing ad libitum access to water and long-stem hay. Following each rest period, calves were reloaded onto the same trailer and taken on another 5-h journey, before they were unloaded at the same feedlot, for a total transport event lasting 20 h. Control calves did not have access to feed or water until the end of the 20-h transit event. Behavioral measurements included bunk attendance (min/d) and standing and lying duration (min/d) recorded for 5 h after the 20-h transport event. Physiological measurements included saliva and hair cortisol, complete blood cell count, serum NEFA, haptoglobin, and substance P concentrations. All physiological measurements as well as BW were taken immediately prior to initial loading, at arrival at the feedlot after the 20-h event, and 48 h after the transport to the same feedlot. Hair cortisol was collected prior to the initial loading and 25 d after transportation. No differences ( = 0.78) in BW loss were observed among treatments after transportation. Standing time was greater ( < 0.001) in CON calves compared with RS5, RS10, and RS15 calves. Salivary cortisol was greater ( < 0.01) in CON and RS15 calves than in RS5 and RS10 calves at the end of the 20-h journey. Serum NEFA concentration was greater ( = 0.03) in RS5 and RS10 calves at arrival compared with CON and RS15 calves, but those differences were no longer observed ( = 0.49) 48 h after transportation. Concentration of substance P did not differ ( = 0.18) between treatments, and haptoglobin concentration tended to be greater ( = 0.07) in CON calves compared with the other treatments 48 h after arrival. Hair cortisol tended ( = 0.10) to be lower in RS5 calves compared with the other treatments. The results of this study indicate that rest stop periods ≥ 10 h did not prevent short- and long-term stress after transport in weaned calves.

The implementation of on-farm pain mitigation strategies is dependent on feasibility and importance to producers. Currently, there is a lack of information regarding adoption of management practices associated with pain in cattle within the Canadian beef industry. The objective of this mixed methods study was to describe pain-associated practices implemented on farm and producer perceptions toward pain mitigation strategies. A questionnaire about calving management and calf processing was delivered to 109 cow-calf producers in western Canada. In addition, 15 respondents were purposively selected based on questionnaire responses to participate in individual semistructured, on-farm interviews. The prevalence of pain mitigation strategies used for dystocia and cesarean section by respondents were 46 and 100%, respectively. The majority of operations reported castrating and dehorning calves before 3 mo of age (95 and 89%, respectively). The majority of operations did not use pain mitigation strategies for castration and dehorning (90 and 85%, respectively). Branding was practiced by 57% of respondents, 4% of which used pain mitigation. Thematic content analysis revealed that producers' perception of pain were influenced by what they referred to as "common sense," relatability to cattle, visual evidence of pain, and age of the animal. Factors that influenced participant rationale for the implementation of pain mitigation practices included access to information and resources, age of the animal, benefit to the operation, cost and logistics, market demands, and personal conscience. Overall, management practices were generally in compliance with published Canadian guidelines. Results of this study may provide direction for future policy making, research, and extension efforts to encourage the adoption of pain mitigation strategies.

The goal of this beef checkoff sponsored symposium was to provide clarity regarding cattle well-being, research, the current state of the industry, and the future of cattle transportation in North America. In today’s marketing programs, feeder calves might be transported as many as 6 times. Loading conditions, time in transit, weather conditions, commingling, segregation of different sexes and weight classes into separate trailer compartments, driver experience, animal nutrition, health status, and physical condition are all factors to be considered when transporting cattle. Transportation related live weight loss (shrink) is not only attributed to loss of gut fill but also tissue loss, which can exceed 60% of total BW loss. High ambient temperature and rough handling of cattle significantly increase the amount of shrink incurred by cattle. If following best management practices could result in a 1% decrease in shrink among the feeder cattle shipped at least once in the United States, the economic benefit would exceed $325 million dollars. The beef industry needs to create a cattle transporter quality assurance program that incorporates sound research data, development of robust guidelines for animal handling, education of cattle transportation employees, and implementation of an online database specifically for individuals who transport cattle.

The objective of this experiment was to investigate the effect of method and timing of weaning on the behavior and DMI of calves throughout weaning, transportation, and a simulated receiving period. Cow-calf pairs (n = 30) housed in a common paddock were randomly assigned to 1 of 3 treatments: preweaning by abrupt removal of the dam 5 d before transportation (AW-P), 2-stage weaning using nose flaps applied 5 d before transportation (2-STG), or abrupt weaning on the day of transport (AW-T). The experiment included 5 d of baseline measurement (BLN), a 5-d weaning period (WN), 4.5 h of transportation (TRANS), and a simulated receiving period encompassing two 1-wk measurement periods (REC1 and REC2, respectively). During the REC1 and REC2, calves were housed by treatment (n = 3). Pedometers were used to measure lying time and steps from BLN to REC1. Lameness in sheep caused by footrot (FR) is a significant health, welfare, and economic concern worldwide. To date, no studies have documented the incidence of FR or associated risk factors in feedlot lambs in Alberta. The objectives of this study were to determine 1) FR incidence and 2) animal, managerial and environmental risk factors associated with FR in one Southern Alberta, lamb feedlot. Assessments were conducted biweekly (average of 10 pens per visit) by 2 experienced observers. A total of 73,150 lambs were assessed between October 2017 and March 2019. All lame lambs were scored according to a 3-point mobility scale (1 = mild, 2 = moderate, and 3 = severe lameness) and physically examined to diagnose the cause of lameness. Risk factors associated with FR were documented and included gender, days on feed (DOF), diet composition, and season. Multivariable regression models (SAS PROC GLIMMIX) were used to determine significant risk factors. A total of 473 lambs were identified as lame, 107 of which were diagnosed with FR (incidence of 22.6%). Footrot affected lambs had greater mobility scores (≥ 2; P < 0.0001) than all other lame diagnoses. Footrot was 4.40 and 0.10 times more likely (P < 0.0001) in female and wether than ram lambs, and 0.60 and 0.23 times more likely (P < 0.0001) in fall and summer than winter and spring seasons. The odds of being diagnosed with FR increased for each additional DOF and each unit increase of barley in the diet (P = 0.0268), while the odds decreased (P = 0.0016) for each additional unit of supplement in the diet. Based on our findings, footrot is an issue to lambs in Alberta, and further studies are still necessary to understand the risk factors associated with potential strategy for mitigating FR in feedlot lambs.

Oral meloxicam is labelled for reducing pain and inflammation associated with castration in cattle in Canada, however, subcutaneous meloxicam is only labelled for pain associated with dis-budding and abdominal surgery. The aim of this project was to determine the pharmacokinetic profile of oral (PO; 1.0 mg/kg BW) and subcutaneous meloxicam (SC; 0.5 mg/kg BW), and to assess the effect of meloxicam on physiological and behavioural indicators of pain associated with knife castration in 7–8 month old calves. Twenty-three Angus crossbred beef calves (328 ± 4.4 kg BW) were randomly assigned to two treatments: PO n = 12 or SC n = 11 administration of meloxicam immediately before knife castration. Physiological parameters included salivary and hair cortisol, substance P, haptoglobin, serum amyloid-A, weight, complete blood count, scrotal and rectal temperature. Behavioural parameters included standing and lying behaviour, pen behaviour and feeding behaviour. Data were analyzed using PROC GLIMMIX (SAS), with repeated measures using mixed procedures including treatment as a fixed effect and animal and pen as a random effect. The pharmacokinetic profile of the drug including area under the curve, volume of distribution and clearance was greater (P < 0.05) in PO than SC calves. After surgery, substance P concentrations, white blood cell counts (WBC), weight and lying duration were greater (P < 0.05) in PO than SC calves, while scrotal circumference was lower (P < 0.05) in PO calves than SC calves. One hundred and seventy-four Angus bull calves (248 ± 27.1 kg of body weight (BW), 6-mo-old) were used in a 71 d study to assess the efficacy of the combination of flunixin meglumine and lidocaine in mitigating pain associated with band and knife castration. The experiment consisted of a 3 × 2 factorial design that included castration method -sham (C), band (B) or knife (K); and medication – lidocaine (scrotal ring block 30 mL, 2% HCl lidocaine) and flunixin meglumine (single s.c. dose of 2.2 mg/kg BW) (M), or saline solution (NM). Animals were weighed on d 0 and weekly until d 71 (final BW) post-castration to obtain ADG. Physiological indicators included salivary cortisol collected on d 0 (30, 60, 120 and 240 min), d 2, 8, and weekly until d 48 post-castration; scrotal and eye temperature assessed on d 1, 2, 6, 8, and weekly until d 36 post-castration; fecal samples for E. coli collected on d 0, 2, 6, 8, and 22 post-castration. In North America, consumer expectations regarding welfare conscience routine management practices in farm animal has grown steadily over the last 20 years, with the most rapid growth occurring in the last 5 years. Increased pressure from retailers and packers, who are responding to consumer concerns and demands for humanely raised beef, has heightened the need for producers to adopt welfare conscience management practices. Common on-farm management practices include transportation, handling, weaning, castration, spaying, dehorning and branding as well as identification and treatment of ill and injured animals and when required, euthanasia. Most of these practices (or lack of them) have been shown to cause stress and or pain in cattle to some degree, increasing the possibility of animal morbidity and mortality. The stress and pain can be further exacerbated if the practices are performed with disregard for animal welfare, resulting in even greater potential for reduced physical and mental well-being and productivity. For example, cull cows and young calves transported for more than 30 h experience greater shrink and mortality and are at greater risk of becoming non-ambulatory and lame than the same types of cattle transported for shorter durations. In the case of castration, calves administered an anesthetic and an analgesic prior to castration experience fewer behavioral and physiological indicators of pain compared to calves not provided pain control. This talk will examine the relationship between sustainable and profitable beef production, welfare friendly management practices and consumer acceptance. In addition, the role of on-farm welfare audits in achieving good animal welfare and meeting consumer expectations will be discussed.

The objective of this study was to evaluate the effect of a single subcutaneous injection of meloxicam on scrotal healing, inflammatory response, and behaviour in castrated or castrated and branded beef calves for 42 d post procedure. Seventy-two 2-mo-old Angus crossbred bull calves were used to assess the effects of procedure (Trt): (1) sham control, (2) knife castration, and (3) knife castration and hot-iron branding; and pain mitigation (Med): (1) nonmedicated and (2) medicated with meloxicam according to a 3 × 2 factorial design. Body weight, scrotal circumference (SC), and healing scores were collected weekly until day 42. Blood samples were collected weekly until day 42 to assess haptoglobin, serum amyloid-A, and complete blood cell count. Hair was collected on day −1 and day 42 to assess cortisol concentrations. Lying and standing behaviour were recorded for 42 d, whereas pain-related behaviours were recorded on days 7, 15, 22, 29, and 34 post procedure. Two groups of cattle were used to develop (model data set: 384 heifers, 228 ± 22.7 kg BW, monitored over a 225-d feeding period) and to validate (naive data set: 384 heifers, 322 ± 34.7 kg BW, monitored over a 142-d feeding period) the use of feeding behavior pattern recognition techniques to predict morbidity in newly arrived feedlot cattle. In the model data set, cattle were defined as morbid (MO) if they were removed from their pen to be treated due to visual observation of clinical signs of bovine respiratory disease and healthy (HL) if they remained within their pen and lacked lung lesions at slaughter. Individual feeding behavior parameters collected with a GrowSafe automated feeding behavior monitoring system were reduced via principal component analysis to 5 components that captured 99% of the variability in the data set. Combinations of clustering and cluster classification strategies applied to those components, along with pattern recognition techniques over different time windows, produced a total of 105 models from which precision, negative predictive value, sensitivity, specificity, and accuracy were calculated by comparing its predictions with the actual health status of individual cattle as determined by visual assessment. When the models with the best specificity (models 79 and 87), sensitivity (models 33 and 66), and accuracy (models 3 and 14) in the model data set were used in a naive data set, models 79 and 87 were not able to predict any MO heifers (0%), with all animals (100%) being predicted as HL. Model 33 predicted 58.3% of the HL and 66.7% of the MO heifers, with MO heifers identified 3.1 ± 1.64 d earlier than by visual observation. Model 66 predicted 50.0% of the HL and 75.0% of the MO heifers, with MO heifers predicted 3.1 ± 1.76 d earlier than by visual observation. Model 3 predicted 100% of the HL and 50.0% of the MO cattle, with MO cattle predicted 1 d earlier than by visual observation. Model 14 predicted 83.3% of the HL and 58.3% of the MO cattle, with MO cattle detected 2.4 ± 1.99 d earlier than visual observation. The application of pattern recognition algorithms to feeding behavior has potential value in identifying MO cattle in advance of overt physical signs of morbidity. Work on an integrated system that would automatically process data collected from automated feed bunk monitoring systems is still required, however, for this method to have value to the commercial feedlot industry as a practical means of identifying MO cattle in real time.

The current study evaluated 17 loads of cull beef cows transported in Canadian winter conditions to assess in-transit temperature and humidity, evaluation of events during loading and unloading, and animal condition and bruising. Regardless of the use of boards to block ventilation holes in trailers, temperatures were higher within trailers than at ambient locations during both travel and stationary periods (P < 0.01). Boarding was associated with smaller differences in trailer temperature, compared with ambient conditions, while the trailer was traveling at highway speeds versus when trailers were stationary (P < 0.01). Moisture levels within trailers were not different from ambient conditions when loads using boarding were traveling (P < 0.01), whereas loads without boarding had a larger difference (P < 0.01). The moisture within trailers relative to ambient conditions increased when trailers were stationary compared with traveling when boarding was used (P < 0.01). The majority of cattle transported were in good body condition (97.4% within BCS of 2 to 3.5) and had calm temperaments (96.7%). Although all comparisons were made, only the doghouse compartment had an increased risk of severe bruising compared with all other compartments (odds ratio [95% confidence interval]: 3.0 [1.6–5.5], 3.7 [2.1–6.4], 2.2 [1.3–3.7] and 3.8 [1.5–9.6] in comparison with the back, belly, deck, and nose compartments, respectively; P < 0.05). Increasing the duration of waiting to unload 30 min relative to a 1 h duration increased the odds of severe bruising by 1.18 times (95% confidence interval: 1.09–1.29; P < 0.01). Scoring systems that have been developed for auditing unloading of cattle had limited variation across loads at both loading and unloading. Pretransport assessment of animal condition using the American Meat Institute’s compromised animal score was the only scoring system that was consistent with posttransport scores. We inferred from the temperature and humidity data in the current study that under commercial conditions, boarding may increase ventilation within trailers during travel and decrease ventilation during stationary periods. The current study provides the first indication that issues in Canadian cull cow transport may be related to pretransport animal condition and management of unloading.
