Honey bees are threatened by many fungal, parasitic and bacterial diseases. This study was aimed at identifying filamentous fungi colonizing the intestinal region of dead adult honey bees and determining resultant mycotoxin activity and its potential adverse implications for bee and human health by the PCR and UHPLC-Orbitrap- HRMS methods. For this purpose, dead bees were collected from the ground in front of 95 hives displaying mass mortality in the Hatay, Iğdır and Bingöl provinces, and 22 filamentous and 6 yeast-like fungi were isolated and identified from the intestinal region of these bees. Of the filamentous fungal isolates, 8 were identified as Aspergillus spp., 3 as Fusarium spp., 2 as Alternaria spp. 5 as Penicillium spp., 4 as Mucor spp., and 2 as Rhizopus spp.. The PCR analysis of the filamentous fungi using primers targeting the aflatoxin and ochratoxin A genes revealed the presence of aflatoxin in only 1 out of the 22 samples. Aflatoxin and ochratoxin were not detected in any of the other samples. The UHPLC-Orbitrap-HRMS method revealed the presence of aflatoxin B1 in 17, both B1 and B2 in 6, B1, B2 and G1 in 5, and aflatoxin G2 in 1 of the filamentous fungal samples. The results obtained in the present study suggest that filamentous fungi may produce mycotoxins in the intestinal region of honey bees, and thereby, honey bees may distribute mycotoxins into the environment and cause indirect adverse effects on human and animal health.

The aim of this study is to investigate the effect of essential oil mixture supplemented in drinking water on antioxidant capacity and intestinal health in geese. One hundred eight chicks (which were 3 days old) were randomly allocated to 3 groups and each group was allocated to 6 subgroups. Research groups have been as follows: C (Control; without supplementation); E1 (0.4 ml/L essential oil mixture supplementation) and E2 (0.8 ml/L essential oil mixture supplementation). The duration of the experiment was 13 weeks. In the first 4 weeks of the trial, the animals were fed for the chick period. In the last 9 weeks of the trial, geese were fed in the pasture under the conditions of Kars province. In the 4th week and at the end of the experiment, GSH exhibited a linear response (P=0.008 and P=0.004, respectively). However, MDA, GSH, SOD, GPx, CAT, nitric oxide, ceruloplasmin, albumin, total protein and globulin were not affected. At the end of the experiment, acetic acid, butyric acid, isocaproic acid and total short-chain fatty acid concentrations were linearly affected with the gradued level of essential oil mixture. There were no significant differences in propionic acid, isobutyric acid, valeric acid, isovaleric acid, caproic acid and BCFA concentrations. In conclusion, water containing essential oil mixture in geese can be used to improve antioxidant capacity and intestinal health.

The way in which content producers in the media industry do business has changed because of the constraints caused by the pandemic while the media consumption habits of consumers have also changed. Magazines, whose number of prints and advertising revenues decrease with each passing year, are one of the media branches most affected by this process. Although it is predicted that there are different crises for magazines in a digitalized world, there are important opportunities for digital magazine publishing, which analyzes the demands and habits of consumers. In the study, the situation of the magazine industry before and after the pandemic, the crises and opportunities in front of the media were evaluated with an argumentative method.

The aim of this study was to detect the presence of aflatoxin M 1 (AFM1) in samples of raw milk (n=38), UHT milk (n=12), white pickled cheese (n=50), and yogurt (n=50) collected from the Sanliurfa city markets and locally produced dairy products by ELISA. The mean contamination rates were 56.74±40.32, 43.1±23.19, 103.2±29.13, and 55.28±12.68ng/kg, respectively, for raw milk, UHT milk, white pickled cheese, and yogurt. According to the data, 21 (55%) raw milk, 3 (25%) UHT milk, 10 (20%) white pickled cheese, and 10 (20%) yogurt samples were contaminated with AFM1 over the acceptable levels (≥50ng/kg), ranging from 0.82 to 130.89ng/kg. None of the white pickled cheese samples contained AFM1 levels above the Turkish legal limit (250ng/kg). Consequently, the AFM1 contamination levels determined in this study in white pickled cheese were not considered to pose a serious public health hazard. However, the AFM1 levels in raw and UHT milk and yogurt samples indicate an increased human health risk in Turkey related to high aflatoxin levels. Therefore, milk and dairy products have to be monitored by the Turkish public health authorities continuously to detect AFM1 contamination.

A total of 28 Wistar albino male rats were used. Four study groups were randomly formed with seven animals in each. The groups were treated for 21 days with distilled water (control group), with water containing 100 ppm fluoride (fluoride group), with resveratrol (12.5 mg/kg i.p., resveratrol group), or with 100 ppm fluoride + 12.5 mg/kg resveratrol i.p. (fluoride + resveratrol group). At the end of the trial, blood samples were collected by cardiac puncture and tissue samples were taken simultaneously. The total antioxidant and oxidant status in plasma and tissues as well as plasma 8-hydroxydeoxyguanosine levels were measured. Histopathological analyses of rat liver and brain tissues were performed in all groups to identify any changes. In the fluoride group, the total oxidant levels increased in plasma, liver and brain and total antioxidant levels decreased, as did the plasma 8-hydroxy-deoxyguanosine levels. These changes were prevented by co-administration of resveratrol. In addition, fluoride-associated severe histopathological changes in brain and liver tissues were not observed in the fluoride + resveratrol group. 19. Effects of reduced glutathione on nitric oxide level, total antioxidant and oxidant capacity and adenosine deaminase activity

The aim of this study was to determine the effect of exogenous GSH, an important antioxidant containing thiol group, on total antioxidant capacity (TAC) and total oxidant capacity (TOC), adenosine deaminase (ADA), a significant indicator of cellular immunity, and nitric oxide (NO) concentrations in rabbits. Sixteen healthy New Zealand rabbits were divided into 2 groups as control and GSH-treated group. Animals in control group received single intraperitoneal (ip) administration of 0.9% saline. Rabbits in GSH-treated group received reduced L-glutathione (10 mg/kg) (ip). Blood samples were taken from the marginal ear vein 0, 3, 6 and 12 hours after injection. Plasma TAC and NO levels were not statistically different between control and GSH group at 0 and 12 hours. Plasma TAC level was found to be significantly higher (p0.05) while NO level was found to be significantly lower (p0.05) in GSH treated group when compared to control group at 3 and 6 hours. Plasma TOC and ADA activity were not statistically different between control and GSH group during experiment. In conclusion, exogenous GSH resulted in an alteration of TAC and NO but not TOC and ADA, so exogenous GSH may be a valuable enhancer of the antioxidant system.

Bu çalışmada, etçi piliçlerde ağızdan kullanılan üç farklı enrofloksasin müstahzarının biyoeşdeğerliği incelendi.Çalışmada 30 gün boyunca ilaç/kirletici madde kalıntısı içermeyen yemle beslenmiş 40 etçi erkek piliç (Ross 308 ırkı) kullanıldı. Her grupta 10 hayvan olmak üzere 4 deneme grubuna ayrıldı. Enrofloksasin Dİ çözeltisi Grup 1'e, Referans İlaç Grup 2'ye, Test 1 İlaç Grup 3'e ve Test 2 İlaç Grup 4'e verildi. Grup 2, 3 ve 4'e uygulama, ağızdan kursak içi yolla (10 mg/kg c.a dozunda) sonda ile yapıldı. Grup 1'dekilerden ilaç verilmeden önce (0,0. dk) ve ilaç verildikten sonra 5. dk'da; diğer gruplarda ilaç verildikten sonra 0,25., 0,5., 1., 2., 4., 8., 12., 18., 24., 36. ve 48. saatlerde steril, heparinli tüplere 1,0-2,0 ml kan numuneleri alındı. Kanların plazmaları ayrıldı.Plazmada enrofloksasin ve siprofloksasin özütlemesini takiben, ölçümleri Yüksek Basınçlı Sıvı Kromotoğrafisi (YBSK) yöntemi ile yapıldı. Özütleme aşamasında internal standart olarak danofloksasin etkin maddesi kullanıldı.Enrofloksasin etkin maddesinin Dİ verilmesini takiben çizilen plazma yoğunluğu-zaman eğrisinden ilacın vücutta 2-bölmeli dışarıya açık modele göre dağıldığı anlaşıldı. Farmakokinetik hesaplamalar buna göre yapıldı.Yöntemin duyarlılığı, tekrarlanabilirliği ile ilgili testler yapıldı. Buna göre, YBSK'da enrofloksasin 11,9-12,8 dakikalarda, siprofloksasin 8,4-9,2 dakikalarda, danofloksasinin 10,4-11,2 dakikalarda pik verdiği belirlendi. Yöntemin duyarlılığı enrofloksasin için 0,01 µg/ml, metaboliti siprofloksasin için 0,04 µg/ml; geriye kazanç enrofloksasin için %75-90, metaboliti siprofloksasin için %55-70 olarak tespit edildi.Referans ve Test İlaçlarının biyoeşdeğerliği incelendi. Biyoeşdeğerliğin belirlenmesinde Test 1 ve Test 2 İlaçlarının EAA ve Ydoruk değerlerinin Referans İlaca bölünmesi ile elde edilen değerlerin (EAA için sırasıyla 0,96 ve 0,97; Ydoruk için sırasıyla 0,92 ve 0,97) kabul edilebilir sınırlar içerisinde (0,80-1,25) olduğu belirlendi.Çalışmada elde edilen veriler üç ilacın birbiriyle eşdeğer olduğunu gösterdi. In this study, three different orally administered enrofloxacin preperations were investigated for their bioequivalance.40 male broilers (Ross 308 strain) were fed by drug-contaminant free feed for 30 days and then divided into four groups, each containing 10 broilers. Enrofloxacin IV solution was administered to Group 1 by intravenously; whereas Reference drug to Group 2, Test 1 drug to Group 3 and Test 2 drug to Group 4 were administered orally (10 mg/kg bw) by intragastric catheters. Around 1,0-2,0 ml of blood samples were taken to sterile heparinized tubes, from Group 1 before (0,0. min) and after (5 min) the drug administration; and the rest of the groups in 0,25., 0,5., 1., 2., 4., 8., 12., 18., 24., 36. ve 48. hours.After the seperation of plasma, enrofloxacin and ciprofloxacin extraction were carried out by High Pressure Liquid Chromatography (HPLC). Danofloxacin was used as internal standart for the extraction phase.It was found that enrofloxacin followed two-compartmental open distribution model according to the results by plasma concentration-time curve after IV administration of enrofloxacin. Pharmacokinetic calculation are made with reference to this.The sensitivity and repeatability tests were carried out in HPLC; where enrofloxacin showed its peak at 11,9-12,8 min, ciprofloxacin at 8,4-9,2 min and danofloxacin at 10,4-11,2 min. Sensitivity was 0,01 µg/ml for enrofloxacin and 0,04 µg/ml for its metabolite ciprofloxacin; recovery value was 5-90% for enrofloxacin and 55-70% for its metabolite ciprofloxacin.The bioequivalance of the reference and the test drugs were figured out. Test 1 and Test 2 drugs showed the results for their bioequivalance in acceptable limits (0,80-1,25) which were calculated by dividing the AUC and Cmax values by the reference drugs (for AUC 0,96 and 0,97; for Cmax 0,92 and 0,97 respectively).The results indicate that three drugs are bioequivalent with each other and to conclude; therefore, these drugs can be used in replacable means.Key words: Bioequivalance, broiler, enrofloxacin, ciprofloxacin, HPLC. 64

Tedavisi sadece organ ve doku nakli ile mümkün olan hastalıklardan ıstırap çeken ve organ nakli bekleyen hastaların sayısı her geçen gün artmaktadır. Organ nakli bugün hâlâ birçok hastalık için yegâne tedavi metodu olarak görülmekte ve uygulanmaktadır. Tüm dünyada organ bağışı ihtiyacı karşılamaktan uzak olmakla Türkiye' de durum gelişmiş ülkelerin çok gerisindedir ve bu ülkelerde organ naklinin çoğunluğu kadavradan karşılanmakta iken ülkemizde canlıdan karşılanmaktadır.Organ bağışında tüm çabalara rağmen istenilen seviyeye ulaşılamamıştır. Organ yetersizliği beraberinde organ ticaretini getirmektedir. Organ nakline ilişkin temel yasa 2238 sayılı yasadır. Yasa 03.06.1979 tarihinde yürürlüğe girmiştir. Tarih ve düzenleme biçimiyle yasa gelişmiş ülkelerden ileri olmakla birlikte kabul ettiği genişletilmiş rıza yöntemi bu gün için başarılı bir yöntem olup olmadığı tartışılmaktır. İspanya' nın kabul ettiği itiraz yöntemi ile İran' da uygulanmakta olan ödül yöntemi ülkemiz için tartışılmaktadır.Bu çalışmada; Sağlık Bakanlığı Organ ve Doku Nakli Dairesi Başkanlığında bulunan veriler incelenmiş, ortalama organ naklinde organ nakli bekleyen hastaların sayısı, nakil edilecek organ türü, nakilden sonra takip edilen hastalarda ölüm sayısı, kadavradan organ naklinde Avrupa standardı altında kalmamızın sebepleri, Türkiye' de organ ticaretinin nedenleri, donör bulmakta karşılaşılan sıkıntılar irdelenmiştir.Organ bağışında temel eksiklik bilgisizlikten kaynaklanmaktır. Eğitim ile bilgi eksikliği giderilebilecek ve istenilen seviyeye ulaşılabilecektir. Ayrıca medya ve Diyanet İşleri Başkanlığı ile topluma mal olmuş kişilerin organ bağışını sürekli gündemde tutması ile bağış sayısında artış olacaktır. The number of people who are suffering from an illness that can only be cured via transplantation of a tissue or an organ, and awaiting their turn for organ transplantation has been increasing day by day. Organ transplantation is still been accepted and implemented as the unique tratment method for many illnesses. The organ donation rate is far from satisfaction of the necessity all around the world, even the situation in Turkey is worse than the developed countries. Whereas cadavers are mostly used in those countries, living organisms are used for organ transplantations in Turkey.Despite all the efforts, the desired level has not been reached in organ donation. The deficiency in organ donation brings about the problem of organ trade. The main regulation on transplantation is The Law no: 2238 which has come into effect on 03.06.1979. With respect to the date and structure the Law is far beyond the situation in developed countries, however the success of extended consent method that it covered is still on debate. The adoption of ?the rejection method of Spain? and ?the reward method of Iran? is under discussion for Turkey.In this study the data gathered from Organ and Tissue Department of Ministry of Health has been examined and the number of patients awaiting their turn for transplantation, the type of organ, the number of people that died after transplantation, the reasons why Turkey is below standards in the transplantation from cadaver, the reasons of organ trade in Turkey and the difficulties in finding donors have been discussed.The main difficulty faced in organ donation stems from illiteracy. Training and education will overcome the problem and help to reach the desired level. Besides, with the continuous efforts of media, Department of Religious Affairs and well- known people on the issue, an increase will happen in organ donation. 106

The aim of this study was to evaluate the protective efficacy of L-carnitine administration in the prevention of toxic effects of experimental chronic aflatoxicosis. A total of 80 and eight weeks old Japanese quail (Coturnix coturnix japonica) were used in this experiment. They were divided into one control and three treatment group each containing 20 quails square group II L-carnitine 200 mg/l via drinking water, group III aflatoxin (AF) 60 mu g/kg feed, group IV AF (60 mu g/kg feed) plus L-carnitine (200 mg/l via drinking water)D. The experimental period lasted 60 days. Control group was fed with unsupplemented basal diet and water.

In this study prepared as a master thesis the information about irregularities instructural systems of buildings has been given. A2 type irregularities, which takeplace in the irregular building definition, have been detailed in specification forStructural to be Built in Disaster Areas. In the researches made on frame and shearwalls and frames building systems, it has been examined how to place opening offloor on the story, the change of ratio opening floor in the building and whetherstructural systems is frame or shear walls and frame will have effect on cross section.The scope and the main idea of this study, which consist of 8 parts, are explained indetail in the firs part. In the second part, the effect of earthquake on buildings ispresented. In the third part, regular and irregular structures and their commonbehavior is presented. In the fourth part, earthquake regulations and the definitions ofstructural system irregularities are given. In the fifth part, calculation methods givenin the National Earthquake Code are classified and definition concerned calculationmethods are presented. In the sixth part, which is numeric examples section,examples were analyzed according to Method of Equivalent Static Load by usingcomputer program Sap 2000. In the last part, which is the conclusion part, the resultsobtained from all the analyses carried out and all the works done in this study areexplained and summarized.

The aim of this study was to detect the presence of aflatoxin M1 (AFM1) in samples of raw milk (n = 38), UHT milk (n = 12), white pickled cheese (n = 50), and yogurt (n = 50) collected from the Şanlıurfa city markets and locally produced dairy products by ELISA. The mean contamination rates were 56.74 ± 40.32, 43.1 ± 23.19, 103.2 ± 29.13, and 55.28 ± 12.68 ng/ kg, respectively, for raw milk, UHT milk, white pickled cheese, and yogurt. According to the data, 21 (55%) raw milk, 3 (25%) UHT milk, 10 (20%) white pickled cheese, and 10 (20%) yogurt samples were contaminated with AFM1 over the acceptable levels (≥50 ng/kg), ranging from 0.82 to 130.89 ng/kg. None of the white pickled cheese samples contained AFM1 levels above the Turkish legal limit (250 ng/kg). Consequently, the AFM1 contamination levels determined in this study in white pickled cheese were not considered to pose a serious public health hazard. However, the AFM1 levels in raw and UHT milk and yogurt samples indicate an increased human health risk in Turkey related to high aflatoxin levels. Therefore, milk and dairy products have to be monitored by the Turkish public health authorities continuously to detect AFM1 contamination. [ABSTRACT FROM AUTHOR]

The aim of this study was to determine the effect of exogenous GSH, an important antioxidant containing thiol group, on total antioxidant capacity (TAC) and total oxidant capacity (TOC), adenosine deaminase (ADA), a significant indicator of cellular immunity, and nitric oxide (NO) concentrations in rabbits. Sixteen healthy New Zealand rabbits were divided into 2 groups as control and GSH-treated group. Animals in control group received single intraperitoneal (ip) administration of 0.9% saline. Rabbits in GSH-treated group received reduced L-glutathione (10 mg/kg) (ip). Blood samples were taken from the marginal ear vein 0, 3, 6 and 12 hours after injection. Plasma TAC and NO levels were not statistically different between control and GSH group at 0 and 12 hours. Plasma TAC level was found to be significantly higher (p<0.05) while NO level was found to be significantly lower (p<0.05) in GSH treated group when compared to control group at 3 and 6 hours. Plasma TOC and ADA activity were not statistically different between control and GSH group during experiment. In conclusion, exogenous GSH resulted in an alteration of TAC and NO but not TOC and ADA, so exogenous GSH may be a valuable enhancer of the antioxidant system. [ABSTRACT FROM AUTHOR]
