Abstract Great concern has long been raised about nitrate leaching in cropland due to its possible environmental side effects in ground water contamination. Here we employed two common techniques to measure nitrate leaching in tea plantation soils in subtropical China. Using drainage lysimeter as a reference method, the adaptability of estimating drainage and nitrate leaching by combining the water balance equation with the suction cup technique was investigated. Results showed that the final cumulative leachate volume for the calculated and measured method was 721.43 mm and 729.92 mm respectively during the study period. However, nitrate concentration exerted great influence in the estimation of nitrate leaching from the suction cup-based method. The cumulative nitrate leaching loss from the lysimeter and suction cup-based method was 47.45 kg ha−1 and 43.58 kg ha−1 under lysimeter nitrate concentrations ranging from 7 mg L−1 to 13 mg L−1, 156.28 kg ha−1 and 79.95 kg ha−1 under lysimeter nitrate concentrations exceeding 13 mg L−1. Therefore, the suction cup-based method could be an alternative way of monitoring nitrate leaching loss within a range of 7–13 mg L−1 of nitrate concentrations in leachate. Besides, lower results occurred in suction cup samplers due to lack of representative samples which mainly leached via preferential flow when in strong leaching events. Thus, it is advisable to increase sampling frequency under such special conditions. The results of this experiment can serve as a reference and guidance for the application of ceramic cups in monitoring nitrogen and other nutrient-ion leaching in tea plantation soils.

Epilepsy-associated cognitive disorder (ECD), a prevalent comorbidity in epilepsy patients, has so far uncharacterized etiological origins. Our prior work revealed that lysyl oxidase (Lox) acted as a novel contributor of ferroptosis, a recently discovered cell death mode in the regulation of brain function. However, the role of Lox-mediated ferroptosis in ECD remains unknown. ECD mouse model was established 2 months later following a single injection of kainic acid (KA) for. After chronic treatment with KA, mice were treated with different doses (30 mg/kg, 100 mg/kg and 300 mg/kg) of Lox inhibitor BAPN. Additionally, hippocampal-specific Lox knockout mice was also constructed and employed to validate the role of Lox in ECD. Cognitive functions were assessed using novel object recognition test (NOR) and Morris water maze test (MWM). Protein expression of phosphorylated cAMP-response element binding (CREB), a well-known molecular marker for evaluation of cognitive performance, was also detected by Western blot. The protein distribution of Lox was analyzed by immunofluorescence. In KA-induced ECD mouse model, ferroptosis process was activated according to upregulation of 4-HNE protein and a previously discovered ferroptosis in our group, namely, Lox was remarkably increased. Pharmacological inhibition of Lox by BAPN at the dose of 100 mg/kg significantly increased the discrimination index following NOR test and decreased escape latency as well as augmented passing times within 60 s following MWM test in ECD mouse model. Additionally, deficiency of Lox in hippocampus also led to pronounced improvement of deficits in ECD model. These findings indicate that the ferroptosis regulatory factor, Lox, is activated in ECD. Ablation of Lox by either pharmacological intervention or genetic manipulation ameliorates the impairment in ECD mouse model, which suggest that Lox serves as a promising therapeutic target for treating ECD in clinic.

The positive effects of afforestation in karst rocky desertification areas are often limited by soil nitrogen (N) availability. However, how plant N limitation evolves in response to prolonged afforestation as well as the mechanisms underlying plant N limitation and soil N availability are poorly understood. In this study, plant leaves and soil samples were collected from cropland, Dodonaea viscosa plantations 10, 20 and 40 years after their establishment on abandoned cropland in a karst rocky desertification area. The natural succession of shrubland served as the control. Foliar N/phosphorus (P) stoichiometry was used to evaluate plant N limitation, and the N concentration and δ15N value of foliar and soil were measured to evaluate the changes in soil N availability. Soil N transformation rates were also determined, using 15N tracing technique to elucidate the mechanism underlying soil inorganic N availability. The plant community-level foliar N/P ratio (13.5) was lower than 14 following 10-y afforestation, indicating that plant growth is predominantly N limited. With the prolonged afforestation, the plant community-level foliar N/P ratio increased to 14.0–15.5, suggesting decreased plant N limitation. This could be attributed to increased soil N availability confirmed by the raises in the N concentration and δ15N of plant community-level foliar and soil δ15N with prolonged afforestation. Compared to cropland, D. viscosa afforestation increased the soil inorganic N supply capacity, by stimulating organic N mineralization, heterotrophic nitrification and the release of adsorbed ammonium, more obviously with prolonged afforestation. The rates of mineralization, heterotrophic nitrification and the release of adsorbed ammonium correlated significantly and positively with the plant community-level foliar N concentration, N/P ratio, and δ15N, indicating that a larger soil inorganic N supply alleviates plant N limitation. This effect can be attributed to the increases in organic matter content, bacterial and fungal abundances and soil structure improvement following afforestation, all of which facilitate mineralization and heterotrophic nitrification. However, plant N limitation after 40 years of afforestation still exists, suggesting that monoculture afforestation may need to be adapted to maximize soil N availability and to alleviate plant N limitation in karst rocky desertification areas.

The excessive application of nitrogen in tea plantations leads to severe soil acidification and N2O emission boosting. To promote sustainable agriculture, nanoparticles (NPs) have emerged as alternative fertilizers, but their effects on soil nitrification and greenhouse gas emissions in tea plantations remain unclear. In this study, the effects of NP type (ZnO-NPs and Fe2O3-NPs) and dose (0, 1, 10, and 100 mg·kg−1) on soil N2O emissions were investigated via a lab incubation trial. Soil pH, ammonium, and nitrate changes were also monitored during the incubation period. The abundance of functional genes related to nitrification and denitrification processes was analyzed as well. The results showed that ZnO-NPs led to a decrease in N2O emissions. The reduction effect was stronger with increasing dose and resulted in a 33% reduction at an addition rate of 100 mg·kg−1. The cumulative N2O emissions had significantly positive correlations with NH4+-N and NO3−-N. ZnO-NP addition showed a significantly negative effect on Ammonia-Oxidizing Archaea (AOA) but a positive effect on Ammonia-Oxidizing Bacteria (AOB) gene abundance. In contrast, Fe2O3-NPs showed an insignificant impact on N2O emissions and soil N content, as well as nitrification–denitrification gene abundance, regardless of different doses. These results imply that the application of ZnO-NPs may inhibit nitrification through the retarding of AOA activity. This study provided us with a potential practice to reduce N2O emissions in tea plantations by applying ZnO-NPs, but the efficiency of this reduction needs further examination under ambient conditions before field application.

Organic-based fertilizers have been ratified to be effective in ameliorating tea growth and the fertility of soil. However, the effect of integrated fertilization on tea growth and quality and the chemical properties of the soil in tea gardens are unclear. To address this, from 2020 to 2021, five different treatments were carried out in the greenhouse of the Tea Research Institute, Hangzhou, CAAS, including CK (control), NPK (chemical fertilizers), RC (rapeseed cake), NPK+B (chemical fertilizer + biochar), and NPK+RC, to investigate the effects of different fertilizations on soil chemistry and tea growth and quality. The results indicated that NPK+B and NPK+RC significantly improved the different amino acid and catechin concentrations in the young shoots, stems, and roots of the tea compared to the CK. The plant growth parameters, e.g., the plant height, no. of leaves, mid-stem girth, and fresh weights of stems and leaves, were significantly increased with integrated fertilization (NPK+B and NPK+RC) compared to the CK and solo organic and inorganic fertilizers. The chlorophyll contents (Chl a, Chl b, and Chl a+b) were generally higher with NPK+RC than with the CK (37%, 35%, and 36%), RC (14%, 26%, and 18%), and NPK (9%, 13%, and 11%) treatments. Integrated fertilization buffered the acidic soil of the tea garden and decreased the soil C:N ratio. NPK+RC also significantly increased the soil’s total C (31% and 16%), N (43% and 31%), P (65% and 40%), available P (31% and 58%), K (70% and 25%), nitrate (504% and 188%), and ammonium (267% and 146%) concentrations compared to the CK and RC. The soil macro- (Mg and Ca) and micronutrients (Mn, Fe, Zn, and Cu) were significantly improved by the RC (100% and 72%) (49%, 161%, 112%, and 40%) and NPK+RC (88% and 48%) (47%, 75%, 45%, and 14%) compared to the CK. The chlorophyll contents and soil macro- and micronutrients were all significantly positively correlated with tea quality (amino acids and catechin contents) and growth. These results indicated that integrated fertilization improved the soil nutrient status, which is associated with the improvement of tea growth and quality. Thus, integrated nutrient management is a feasible tool for improving tea growth, quality, and low nutrient levels in the soil.

Nitrogen is one of the most important nutrients for tea plants, as it contributes significantly to tea yield and serves as the component of amino acids, which in turn affects the quality of tea produced. To achieve higher yields, excessive amounts of N fertilizers mainly in the form of urea have been applied in tea plantations where N fertilizer is prone to convert to nitrate and be lost by leaching in the acid soils. This usually results in elevated costs and environmental pollution. A comprehensive understanding of N metabolism in tea plants and the underlying mechanisms is necessary to identify the key regulators, characterize the functional phenotypes, and finally improve nitrogen use efficiency (NUE). Tea plants absorb and utilize ammonium as the preferred N source, thus a large amount of nitrate remains activated in soils. The improvement of nitrate utilization by tea plants is going to be an alternative aspect for NUE with great potentiality. In the process of N assimilation, nitrate is reduced to ammonium and subsequently derived to the GS-GOGAT pathway, involving the participation of nitrate reductase (NR), nitrite reductase (NiR), glutamine synthetase (GS), glutamate synthase (GOGAT), and glutamate dehydrogenase (GDH). Additionally, theanine, a unique amino acid responsible for umami taste, is biosynthesized by the catalysis of theanine synthetase (TS). In this review, we summarize what is known about the regulation and functioning of the enzymes and transporters implicated in N acquisition and metabolism in tea plants and the current methods for assessing NUE in this species. The challenges and prospects to expand our knowledge on N metabolism and related molecular mechanisms in tea plants which could be a model for woody perennial plant used for vegetative harvest are also discussed to provide the theoretical basis for future research to assess NUE traits more precisely among the vast germplasm resources, thus achieving NUE improvement.

Abstract Background Small-cell carcinoma (SMCC) of hypopharynx is an extreme rare entity in the head and neck region. It is commonly arising from pulmonary in origin. We report a rare case of small-cell carcinoma of hypopharynx that caused upper airway obstruction in an elderly gentleman. Case presentation A 67-year-old man is presented to otorhinolaryngology clinic with a 4-month history of worsening shortness of breath and stridor. He was heavy tobacco smoker for 60 pack-years. Flexible nasopharyngolaryngoscopy revealed a large mass at the right pyriform fossa extending superiorly and obstructing the laryngeal inlet. He underwent tracheostomy, examination under general anesthesia with tissue biopsy. Histology from tissue biopsy revealed small-cell carcinoma. The positron emission tomography-CT showed exophytic mass from right pyriform sinus with regional and lung pleural metastasis as well. There was the absence of formal staging system for SMCC due to its limited and rare incidence. He was treated with combination of carboplatin and etoposide, similar to the regime for cell carcinoma of lung. The tumor regressed loco-regionally, and his pleural metastasis had shrunk to sub-centimeter in size. Conclusions To date, there is very limited data available on head and neck small cell carcinomas to guide treatment recommendations as well as staging system. Paucity of clinical data and outcome about this rare malignancy makes this report a valuable piece of experience to be shared together with existing literature.

Abstract Background Epithelial–mesenchymal transition (EMT) is one of the mechanisms of airway remodeling in chronic asthma. Interleukin (IL)-24 has been implicated in the promotion of tissue fibrosis, and increased IL-24 levels have been observed in the nasal secretions and sputum of asthmatic patients. However, the role of IL-24 in asthmatic airway remodeling, especially in EMT, remains largely unknown. We aimed to explore the effect and mechanism of IL-24 on EMT and to verify whether IL-37 could alleviate IL-24-induced EMT in chronic asthma. Methods BEAS-2B cells were exposed to IL-24, and cell migration was assessed by wound healing and Transwell assays. The expression of EMT-related biomarkers (E-cadherin, vimentin, and α-SMA) was evaluated after the cells were stimulated with IL-24 with or without IL-37. A murine asthma model was established by intranasal administration of house dust mite (HDM) extracts for 5 weeks, and the effects of IL-24 and IL-37 on EMT and airway remodeling were investigated by intranasal administration of si-IL-24 and rhIL-37. Results We observed that IL-24 significantly enhanced the migration of BEAS-2B cells in vitro. IL-24 promoted the expression of the EMT biomarkers vimentin and α-SMA via the STAT3 and ERK1/2 pathways. In addition, we found that IL-37 partially reversed IL-24-induced EMT in BEAS-2B cells by blocking the ERK1/2 and STAT3 pathways. Similarly, the in vivo results showed that IL-24 was overexpressed in the airway epithelium of an HDM-induced chronic asthma model, and IL-24 silencing or IL-37 treatment could reverse EMT biomarker expression. Conclusions Overall, these findings indicated that IL-37 mitigated HDM-induced airway remodeling by inhibiting IL-24-mediated EMT via the ERK1/2 and STAT3 pathways, thereby providing experimental evidence for IL-24 as a novel therapeutic target and IL-37 as a promising agent for treating severe asthma.

In order to find out the differences in tea quality characteristics from different regions and the reasons for their formation, 32 biochemical components, 22 mineral elements, and 3 stable isotope ratios were determined in 93 white leaf tea (WLT) samples. The findings revealed that significant differences in the content of free amino acids, catechins, caffeine, several mineral elements, and stable isotope ratios were observed among the WLT samples of different origins. Furthermore, a Pearson correlation analysis showed significant correlations of 15 chemical composition qualities and 14 mineral elements with soil factors. The δ13C and δ18O values were significantly correlated with meteorological factors. Classification models based on multiple chemical signatures facilitated successful differentiation of WLT of different origins, achieving a high prediction accuracy of over 90%. This study comprehensively identified distinct quality characteristics of WLT from different regions in China and proved an effective method in classifying WLT samples from the main production areas.
