Zingiberaceae family of plants are ethnomedicinally well known. One of the less‐studied endemic species from this family, Curcuma neilgherrensis, and its leaves have been used as a folklore medicine by tribes of Western Ghats of India since ancient times. The aim of the study is to investigate in vitro antioxidant and antiproliferative potentials of organic solvent extracts prepared from C. neilgherrensis leaves as well as phytochemical profiling of the selected extract capable of apoptotic induction in cancer cells. In vitro antioxidant activity was evaluated by the estimation of total phenolics and flavanoid contents, DPPH, ABTS radical scavenging, and lipid peroxidation inhibition assays. Cytotoxic/antiproliferative potential was assessed employing human cancer cell lines (K562, HCT116, MDA‐MB‐231, and HL60) and normal lymphocyte cultures by MTT and trypan blue dye exclusion assays. Fluorescence microscopy, flow cytometry, DNA fragmentation, and caspase activity assays were utilized to study apoptosis induction by the extract in HCT116 cells. HR‐ESI‐MS/MS was used to identify chemical constituents present in the extract. C. neilgherrensis leaves extracts were found to contain varying amounts of total phenolics and flavanoid contents as well as antioxidant activity, with the methanolic extract (CNLM) displaying maximal activity. Higher antiproliferative potential was shown by the chloroform extract (CNLC) for a 48 h treatment period, against colorectal carcinoma HCT116 cells (IC50‐8.3 ± 0.8 μg/mL). CNLC treatment caused cytomorphological alternations, phosphatidylserine externalization, reduction in mitochondrial membrane potential, increase in intracellular ROS, sub‐G1 phase cell population, and DNA fragmentation, as well as caspase 3 and 9 activities, confirming apoptosis induction in HCT116 cells. Phytochemical profiling of the CNLC extract by HR‐ESI‐MS/MS enabled identification of bioactive compounds, previously reported with anticancer potential such as rutin, tamarixetin, embelin, ginkgolide B, traumatic acid, and rosmarinic acid. To our knowledge, this is the first report demonstrating in vitro antioxidant and antiproliferative potentials of C. neilgherrensis leaf extracts. The results presented reveal that C. neilgherrensis leaves are indeed a potent bioresource for chemotherapeutic candidate drugs for future use. [ABSTRACT FROM AUTHOR]

This cross‐sectional study investigated nonconventional therapies for COVID‐19 in Trinidad, emphasizing the need for documentation supporting future pharmaceutical research. The survey, conducted from June 20 to July 19, 2022, garnered responses from 57 participants aged 18 and above, with 82.46% vaccinated. The majority (81%) utilized both conventional and nonconventional therapies, revealing insights for potential alternatives to traditional treatments. Conventional treatments, including antibiotics, Ivermectin, anti‐inflammatories, analgesics, bronchodilators, and cough/flu syrups, were frequently reported. Nonconventional therapies encompassed vitamins, minerals, supplements, and various plant and animal products. When participants used conventional therapies, either alone or in combination with nonconventional ones, 13.21% reported side effects. These included severe thirst, headache, nausea, drowsiness, and one case of weight gain. Conversely, those exclusively using nonconventional treatments reported no side effects. Encouragingly, nonconventional therapies demonstrated promising effects in managing COVID‐19, emphasizing the need for meticulous selection, research, and development of their bioactive compounds as potential alternatives to conventional therapies. Simple Summary: Lung cancer is the deadliest of all cancer types and often develops resistance to chemotherapy, necessitating the search for new therapeutic options. In this context, the current study aimed to demonstrate the application of mahanine (MH), a plant-derived phytochemical from the curry leaf, as a potential treatment for drug-sensitive and drug-resistant lung cancer, while also proposing a potential mechanism of action. The results of this study establish MH as a potential treatment option for both drug-sensitive and -resistant lung cancer. Furthermore, this study may serve as a foundation for research into specific oncogene regulation and the potential use of MH as an adjuvant therapy alongside other chemotherapeutics for lung cancer patients. Background/Objectives: Lung cancer is one of the deadliest cancers, and drug resistance complicates its treatment. Mahanine (MH), an alkaloid from Murraya koenigii has been known for its anti-cancer properties. However, its effectiveness and mechanisms in treating non-small cell lung cancer (NSCLC) remain largely unexplored. The present study aimed to investigate MH's effect on drug-sensitive and drug-resistant NSCLC and its potential mechanism of action. Methods: We isolated MH from M. koenigii leaves and the purity (99%) was confirmed by HPLC, LC-MS and NMR. The antiproliferative activity of MH was determined using MTT and colony formation assays against drug-sensitive (A549 and H1299) and Taxol-resistant lung cancer cells (A549-TR). Western blot analysis was performed to determine MH's effects on various molecular targets. Anti-tumor activity of MH was determined against lung tumors developed in female NOD Scid mice injected with A549-Fluc bioluminescent cells (1.5 × 106) intrathoracically. Results: MH dose-dependently reduced the proliferation of all lung cancer cells (A549, H1299 and A549-TR), with IC50 values of 7.5, 5, and 10 µM, respectively. Mechanistically, MH arrested cell growth in the G0/G1 and G2/M phases of the cell cycle by inhibiting cyclin-dependent kinase 4/6 (CDK4/6) and cell division control 2 (CDC2) and induced apoptosis through the downregulation of B-cell leukemia/lymphoma 2 (BCL2) and B-cell lymphoma-extra large (BCL-XL). The apoptotic induction capacity of MH can also be attributed to its ability to inhibit pro-oncogenic markers, including mesenchymal–epithelial transition factor receptor (MET), phosphorylated protein kinase B (p-AKT), phosphorylated mammalian target of rapamycin (p-mTOR), survivin, rat sarcoma viral oncogene (RAS), myelocytomatosis oncogene (cMYC), and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) levels. In vivo, MH (25 mg/kg b. wt.) significantly (p < 0.001) inhibited the growth of A549 lung cancer orthotopic xenografts in NOD Scid mice by 70%. Exosomes are unique biogenic nanocarriers of endocytic origin that are generated from most of the cells and found in biofluids like milk, plasma, saliva, and urine. Bovine milk represents the largest and an economic source for the production of exosomes. In recent past, the utility of the milk exosomes as drug carriers is intensified. Exosomes are emerging for delivery of both small and large therapeutics due to their biocompatibility. In this article, we highlighted the various exosomal isolation techniques, physicochemical properties, their biodistribution, and utility of milk exosomes in delivering the small drug molecules and siRNA to combat cancer.
