Rice straw is an important feedstock for second-generation biorefineries, the majority being conventional white rice. This research investigated the potential of the straw of black rice Chakhao Poireiton, a rice variety of increasing importance, for biorefining. Pre-treatment of black rice straw was carried out in a pressurised microwave reactor and free sugar release was measured following digestion with Cellic® CTec3. This was compared to white rice straw. Pre-treatment (100 − 200° C for 5 min) did not drastically impact the sugar composition but brought about an enhanced release of glucose after enzymatic hydrolysis from 25 to 50% (black rice) and from 26 to 55% (white rice). For xylose digestibility, the increase was around seven-fold for black rice (8 to 57%) and five-fold for white rice (13 to 64%). This improvement in digestibility of the straw samples could have come from modification in the lignocellulose structural features making the polysaccharides more susceptible to hydrolysis. Black rice could be a suitable feedstock for a combined biorefinery–food and nutraceuticals from grain (much higher than white rice) and the straw for fermentable sugars comparable to white rice straw for biofuels.

Indian black rice (Chakhao Poireiton) is a pigmented variety, rich in anthocyanins and other phytonutrients. With growing interest in the use of local raw materials in brewing, it was of interest to develop protocols for malting and brewing with Chakhao Poireiton to see whether the antioxidant capacity of anthocyanins could be delivered into finished beer. Protocols for brewing with 100% malted rice were developed and the performance of Indian black rice compared with that of an Italian white rice cultivar suited to brewing. The apparent fermentabilities of rice worts were 69.5% (black) and 67.3% (white), yielding beers of 3.28 and 3.19% ABV respectively. Black rice worts were deficient in free amino nitrogen (83.5 mg/L relative to 137 mg/L for white rice) and would need nitrogen supplementation to avoid issues with fermentation, e.g. elevated diacetyl. Black rice beer had an orange‐red hue as a result of extraction of anthocyanin pigments (2.84 mg/L). The oxidative stability of 100% rice beers was measured using electron spin resonance spectroscopy and both samples were found to be unusually stable. Interestingly, when rice beers were blended with a control barley malt derived lager in varying proportions (10, 25, 50%), the oxidative stability was improved, relative to the control lager, particularly so in the case of black rice beer, which contained an antioxidant capacity over and above that of the white rice beer. Future studies are required to determine whether the noted oxidative stability of 100% rice malt beers results in a more flavour‐stable beer. © 2019 The Institute of Brewing & Distilling

Significance Organ formation is an essential process in plants and animals, driven by cell division and cell identity establishment. Root branching where lateral roots form along the primary root axis increases the root system and aids the capture of water and nutrients. We have discovered that tight control of the cell width is necessary to coordinate asymmetric cell divisions in cells that give rise to a new lateral root organ. Although biomechanical processes have been shown to play a role in plant organogenesis, including lateral root formation, our data give mechanistic insights into cell size control during lateral root initiation., In plants, postembryonic formation of new organs helps shape the adult organism. This requires the tight regulation of when and where a new organ is formed and a coordination of the underlying cell divisions. To build a root system, new lateral roots are continuously developing, and this process requires the tight coordination of asymmetric cell division in adjacent pericycle cells. We identified EXPANSIN A1 (EXPA1) as a cell wall modifying enzyme controlling the divisions marking lateral root initiation. Loss of EXPA1 leads to defects in the first asymmetric pericycle cell divisions and the radial swelling of the pericycle during auxin-driven lateral root formation. We conclude that a localized radial expansion of adjacent pericycle cells is required to position the asymmetric cell divisions and generate a core of small daughter cells, which is a prerequisite for lateral root organogenesis.

Chakhao, the Geographical Indication (GI) tagged aromatic black rice of Manipur, India, is a rich source of bioactive compounds such as anthocyanins. However, there is limited work available on Chakhao and research has focused on its grain. No reports on the anthocyanin, antioxidant properties or in vitro bioactivities of its straw are available. Anthocyanins content of its different fractions displayed a positive correlation with their antioxidant potentials (r = 0.90). In this study, microwave-assisted sub-critical water extraction, at 90 °C for 5 min, from the straw gave an anthocyanins extraction efficiency of 85.8%. Furthermore, this microwave extract displayed higher antioxidant activity than an equivalent conventional methanol extract. In vitro studies on the microwave extracts of both straw and bran showed no apparent cytotoxicity on Jurkat cells and dose-dependent inhibition of colony growth in colorectal cancer cells. Black rice bran and straw are valuable by-products of the food industry that are rich in phytochemicals. This study reports the extraction of anthocyanins from black rice straw and presents evidence that the straw, in addition to the bran, contains important bioactive compounds; the extracts of which could further be explored as a natural antioxidant and/or functional ingredients.

Lignocellulosic feedstocks for biorefinery are likely to be seasonal and the supply of feedstock to cellulosic biorefineries remains a challenge. One way to overcome this is by utilizing a mixed feedstock which facilitates the maintenance of a year-round feedstock supply. This study investigated the impact of mixing of three industrially relevant cellulosic feedstocks—wheat straw, willow and Miscanthus using two major performance indicators—sugar yield and fermentation inhibitor production. A microwave hydrothermal pre-treatment regime of 200 °C for 5 min was applied to each feedstock individually and to 1:1 (w/w) mixes and the predicted sugar yield in the mixes was compared to the observed values. All the mixes resulted in improved sugar yields with willow + Miscanthus (15.4%, p = 0.015) and wheat + willow (13.6%, p = 0.010) showing a statistically significant improvement. Saccharification kinetics, inhibitor production, impact on yeast metabolic activity and growth were compared and no adverse impacts of mixing were observed. The use of mixed feedstocks in a hot water based commercial production of biofuels is unlikely to have any adverse effects on productivity and may indeed prove beneficial.
