11. Post Traumatic Lesion absence of -Dystroglycan-Immunopositivity in Brain Vessels Coincides with the Glial Reaction and the Immunoreactivity of Vascular Laminin

Abstract

Following brain lesions, the gliovascular basal lamina undergoes destruction and the gliovascular connections ‘decouple’. Laminin receptors, as dystroglycan, are essential in these processes. The present study compares the immunoreactivities of -dystroglycan, glial fibrillary acidic protein (GFAP), and laminin following stab wounds in adult rats. In intact brain the vessels were immunopositive to -dystroglycan, whereas the laminin of their basal lamina proved to be unavailable to immunoreactions. Following stab wound, however, the adjacent vessels lost their immunopositvity to -dystroglycan, whereas immunopositivity to laminin became detectable in them. In an advanced stage of glial reaction the territory of GFAP immunopositive reactive astrocytes coincided with the area where vessels lost their immunopositivity to -dystroglycan. When glial reaction regressed, the -dystroglycan immunopositivity re-appeared, and laminin immunopositivity became undetectable again. Post-lesional disappearance of vascular -dystroglycan immunostaining was described earlier, and was attributed to the cleavage of -dystroglycan by matrix metalloproteinases as a mechanism of the decoupling of the gliovascular connections. Our results, which were obtained in a different type of lesion support that the loss of vascular -dystroglycan immunopositivity is a general phenomenon following cerebral lesions, and an indirect marker of gliovascular decoupling. For the first time coincidences were presented between vascular immunonegativity to -dystroglycan, glial reaction and detectability of laminin. Manifestation of laminin immunoreactivity also indicates gliovascular decoupling. Coincidence between glial reaction and lack of vascular -dystroglycan suggests mutual enhancement between them. The observations may have clinico-pathologic importance since similar investigations may help to follow the progression and regression of post-lesion processes.

12. Comparative Studies on Fresh and Postmortem Isolated Synaptosomes: ATPase Activities and Ouabain Binding

Abstract

In earlier studies we have established that rat cortical synaptosomes isolated after a 2-h postmortem storage period showed functional performance and morphological characteristics similar to those isolated immediately after decapitation. The present paper is an account of further functional studies involving the activity of Na-K- and Mg-dependent ATPase and the binding of ouabain in postmortem synaptosomes. These functions were found to survive without major alteration even in the 6-h postmortem fractions. On the other hand the 6-h postmortem fractions showed a very slow oxygen consumption unaffected by DNP and veratrine. In both control and postmortem synaptosomes the actual activity of Na-K-ATPase as calculated from the ouabain-sensitive component of oxygen consumption was consistently less than the potential activity of the enzyme revealed if determination was carried out on the basis of phosphate splitting.

13. Histological alterations in the fetal mouse cerebellum after neutron irradiation: A light and electron microscopic study

Abstract

Histological effects of in utero neutron irradiation (0.5 Gy) were investigated in fetal mouse cerebellum. Irradiation was performed on the 18th embryonic day. Histological alterations were examined at 2, 6 and 12 h after irradiation. The effect was most pronounced in the EGL, 6h after irradiation. In some cells (about 24%) karyopycnosis, destruction of cytoplasmic organelles and shrinkage were observed. Remnants of destroyed cells-the pycnotic nuclei with some cytoplasmic debris around them-were rapidly phagocytized by the surrounding cells. In addition to glial elements, the EGL germinal cells also showed phagocytic activity. Dense particles appearing light-microscopically as pycnotic nuclei, proved to be mostly remnants of destroyed, phagocytized cells under electron microscope.

16. Hero Shtetls: Jewish Armed Self-Defense From the Pale to Palestine, 1917-1970

Abstract

Relying on a research in more than two dozen archives in Russia, Ukraine, Belarus, Israel, and the United States, my dissertation traces the history of Jewish paramilitarism from the waning days of the Russian Empire to early Mandatory Palestine. By examining a heretofore neglected aspect of Jewish reactions to violence, my dissertation aims to contribute to the historiography of the pogroms, and of Jewish power, militarism, and violence. Situating self-defense vis-à-vis early Soviet Jewish policies and longue durée developments in Russian Jewish politics, I argue that Jewish self-defense developed in unison with the political violence of the period in question, and served as a tool for carving out an autonomous Jewish space during the Civil War. Finally, I detect how self-defense activists transplanted their experiences to Palestine. In the wake of the February 1917 revolution the Russian Army began to disintegrate along ethnic lines. Zionist and other Jewish soldiers formed military organizations, culminating in the establishment of Union of Jewish Soldiers (UJS) in the fall, which strove to safeguard the national and civic rights of Jewish soldiers. In Ukraine, the nationalization of the army and the increasingly hostile authorities led to the dispersal of the UJS in January 1918, but its activists played a role in establishing self-defense across Ukraine. The UJS in Russia remained in existence for over a year, cooperating with Soviet Jewish authorities in providing aid to Jewish2soldiers, and setting up self-defense units. However, the internationalist ethos of the Soviet state, coupled with the encroachment of Soviet Jewish authorities on Jewish institutions rendered its existence untenable. The deadliest pogroms erupted in 1919, claiming tens of thousands of victims, propelling Jews throughout Ukraine and Belarus to take to arms, with the first large and longevous unit established in Odessa. By early 1921, a network of a nearly 15,000-strong Jewish armed force uniting 50 localities operated in Central Ukraine, the area hit hardest by pogroms. Jewish units drafted local Jews, levied taxes on inhabitants, guarded settlements, and operated against insurgent bands, essentially fulfilling quasi-state functions. Soviet military and civilian authorities, struggling to reestablish state power in the rebellious countryside, cooperated with, relied on, and legalized Jewish paramilitaries, as Jews were seen as more trustworthy than non-Jewish locals on account of sharing enemies with the Soviet state. While Jewish units were trained, armed, and coopted by Soviet authorities throughout 1920-1923, they also evoked the ire of Soviet Jewish, and of some state and party authorities, anxious to gain influence among the local (Jewish) populace, and accusing self-defense groups of Zionist leanings. I untangle the complex web of negotiations conducted on the local and national level about the legalization and maintenance of Jewish self-defense units, particularly with an eye to how local Jewish communities and self-defense groups straddled the divide between their own existential fears and the professed values and institutional interests of Soviet military and civilian authorities. The last chapter examines the migration of activists, ideas, and memories of self-defense to Mandatory Palestine. In particular, it focuses on the “Odessa Group,” a loose network of former Odessa self-defense members active in the Haganah and the Irgun. I detect concepts about paramilitarism originating in Civil War Russia and unearth examples when recollections of former self-defense members were publicized in pre-state Palestine, posing a direct challenge to the Zionist concept of a defenseless Diaspora Jewry.

Abstract

Present study demonstrates a novel localization of beta-dystroglycan in rat brain. Beside the meningeal surface and the cerebral vessels where beta-dystroglycan immunopositivity has been described, now immunopositive solid bodies were found at the basis of ependymocytes. Since they proved to be round in either coronal, sagittal, and horizontal section, revealing their globular shape, they received the term 'globules.' Double immunofluorescent labeling of GFAP (glial fibrillary acidic protein) and beta-dystroglycan revealed that 'globules' are within the 'cordon' of subventricular astrocytes. The 'globules' were found throughout the ventricular system, except the ventral part and floor of the third ventricle. The latter one comprised the median eminence and extended caudally to the inframamillary recess. The area where the 'globules' were missing, corresponded to that where ependymocytes were replaced by GFAP-immunopositive tanycytes. Utrophin and alpha-dystrobrevin were co-localized with dystroglycan, whereas alpha1-syntrophin was detected along the borders of the ependymal cells. Comparing our results to former data, the 'globules' seem to be the anchoring places of the 'fractons' formed by laminin [Mercier et al. (2003) J Comp Neurol 455:324-340], which interconnect the vascular basal lamina and the ependymal layer. The ependymal localization of the 'globules' was proved by pre-embedding electron microscopic immunohistochemistry. The 'globules' proved to be a labyrinthine structure, which seemed to be identical with the 'basal membrane labyrinth' described by Leonhardt [Leonhardt (1970) Z Zellforsch Mikrosk Anat 105:395-404]. The description of the beta-dystroglycan-immunopositive 'globules' contributes to the better understanding of the structure of the subventricular zone where neurogenesis can occur during adulthood and provides an important link to the meningeo-glial network., ((c) 2008 Wiley-Liss, Inc.)
