Bu çalışmada hidroksiapatitin sahip olduğu biyouyumluluk ve biyoaktivite özelliklerini kaybetmeden mekanik ve biyouyum özellikleri yüksek, biyoseramik kompozit sentezlenmesi amaçlanmıştır. Bu çalışma iki etaptan oluşmaktadır. 1. Etap: Klinoptilolit (Cp), alümina (Al2O3) katkılı bovine hidroksiapatit (BHA) kompozitleri üretmek. Bu kompozitlerin fiziksel, mekanik ve mikroyapısal özellikleri incelenerek en yüksek mekanik özelliği olan kompoziti belirlemektir.2. Etap: En yüksek mekanik özelliği tespit edilen kompozitin biyouyum ve elektriksel özelliklerinin incelenmesidir. İlk etapta dana kemiğinden elde edilen hidroksiapatite (BHA) ağ. % (5–15) arasında değişen oranlarda Cp ve Al2O3 katılarak elde edilen karışımlardan BS 7253 standardına uygun peletler üretilmiştir. Bu peletler 1000oC, 1100oC, 1200oC ve 1300oC olmak üzere dört farklı sıcaklıkta 4 saat süre ile sinterlenerek biyoseramik kompozitler elde edilmiştir. Üretilen kompozitlerin fiziksel, mekanik, mikroyapısal özelliklerin incelenmesinden 1300oC sıcaklıkta sinterlenen, ağ. % 15 Cp ve ağ. %15 Al2O3 bulunan kompozitin (yoğunluk 2.337 g/cm3, basma dayanımı 105.6 MPa, sertlik 305 HV) en yüksek mekanik dayanıma sahip olduğu tespit edilmiştir. İkinci etap çalışmaya bu kompozit ve karşılaştırmanın yapılabilmesi için katkı oranı olarak en az olan ağ. % 5 Cp ve ağ. %5 Al2O3 kompoziti de dahil edilmiştir. Ayrıca sonuçların literatür ile karşılaştırılabilmesi için ikinci etap çalışmaya saf BHA, saf ticari hidroksiapatit (CSHA) ve saf Al2O3 da dahil edilmiştir. Seçilen bu beş saf ve karışım tozlardan peletler üretilerek 1300oC sıcaklıkta 4 saat süre ile sinterlenmiştir. Peletlerden birer adedi elektriksel ölçüm için ayrılırken geri kalan peletler biyouyum incelemesinde kullanılmıştır. Bu kapsamda çalışmada gerçek hücreler (osteosarcoma, SAOS-2) kullanılarak sitotoksisite testi (MTT) ve peletlerin üzerine hücre ekimi yapılarak SEM görüntüleri alınmıştır. Sitotoksisite sonuçları kontrol grubu ile karşılaştırıldığında saf Al2O3'nın hücre çoğalmasını desteklemediği görülmüştür. Diğer tüm numuneler, kontrol gurubu ile karşılaştırıldığında daha yüksek hücre çoğalması sağlamıştır. En yüksek oran ağ. % 15 Cp ve ağ. %15 Al2O3 bulunan kompozitte elde edilmiştir. Bu kompozitte %15 Al2O3 bulunmasına rağmen BHA ve CSHA numunelerinden daha yüksek hücre çoğalmasının gerçekleşmiş olması Cp'nin kemik hücrelerine desteğini göstermektedir. Bu durum literatür ile de desteklenmektedir.SEM görüntülerinde CSHA numunesinde hücre yayılımı görülmektedir. Ağ. % 15 Cp ve ağ. %15 Al2O3 bulunan kompozit numunesinde ise hücrelerin yayılımı CSHA numunesindeki kadar açık görülmemesi, kompozitin poroz yapısı nedeni ile kompozitin içine doğru yayılımı düşündürmektedir. Numunelerin elektriksel özellikleri 0.1Hz -10MHz aralığında incelenmiştir. Ölçüm sonuçlarına göre BHA, CSHA ve Al2O3'nın kapasite değerleri, kayıp faktörü ve dielektrik katsayısı küçük frekans değerlerinde yüksek iken, frekans arttıkça logaritmik olarak azalmaktadır. Kompozitlerde 0.1Hz'de saf malzemelere göre daha düşük değerden başlayıp benzer bir logaritmik azalma gözlenmektedir. Numuneler arasında bu özelliklerde en düşük değer, ağ. % 5 Cp ve ağ. %5 Al2O3 olan kompozitte elde edilmiştir. Numunelerin 1MHz'e kadar iletkenlik değeri 10-11 S/m'nin üzerinde iken 5MHz sonrası hızlı bir artış görülmüştür. Kompozitler, saf malzemelere göre daha yalıtkan bir özellik göstermişlerdir. Literatürde HA'nın Cp ve Al2O3 ile ikili veya üçlü kompozitinin elektriksel özelliklerine dair çalışma bulunamadığı için bu yönde bir karşılaştırma yapılamamıştır. Sonuç olarak hidroksiapatit esaslı biyoseramik malzemelerin karakterizasyonu kapsamında yaptığımız çalışmada en yüksek mekanik özellikler ağ. % 15 Cp ve ağ. %15 Al2O3 bulunan kompozitte elde edilmiş olup biyouyum çalışmaları bu kompozitin kemik hücresi olan osteosarcoma (SAOS-2) ile yüksek uyum gösterdiği için medikal uygulamalarda uygun biyoseramik malzeme olarak önerilebilir. In order to compare these results with the literature, pure BHA, pure CSHA, pure Al2O3 was also included in the second stage study. From each group pellets were produced from these five selected pure and mixed powders and sintered at 1300° C for 4 hours. The one sample in each group was separated for electrical measurement and the remaining samples were used for the biocompatibility study. The cytotoxicity test (MTT) was performed using real cells (osteosarcoma, SAOS-2) and cell proliferation was performed on pellets and SEM images were obtained.Compared with the control group it is seen that cytotoxicity results did not support cell proliferation of pure Al2O3. The results of all other samples were higher than the control group, allowing for cell proliferation. The highest ratio was obtained for 15 wt.% Cp and 15 wt.% Al2O3 containing composite. The fact that higher cell proliferation than BHA and CSHA samples was achieved the presence of 15 wt.% Al2O3' in the body indicates that Cp supports cell bone cells. This situation is also supported by previous studies. According to SEM images, cell proliferation is clearly visible for the CSHA specimens. In the case of composite with 15 wt.% by. Cp and 15 wt.% Al2O3, the spread of cells is not as high as seen in the case of CSHA. This situation clearly suggest that the spreading of cell may move to porosity area in the samples. Therefore, it is not clearly seen the cells on the surface of samples. 115

V. ÖZET Bu tez çalışmasında atmosfer basıncında çalışan bir azot laser yapılması amaçlanmıştır. Laser ışımasının temel prensibi olan uyarılmış ışımayı gerçekleştirebilmek için azot laserin elektrotlarına çok kısa sürede çok yüksek gerilim (dVg/dt = lkV/ns) uygulandı. Azot laser 337.1nm'de pulsif ışıma yapar. Bu çalışma sırasında laser ışıma teknolojisini ve ışıma karakteristiklerini de inceledik. Bir başka amaç diğer laser çeşitleri hakkında da bilgilenmek istediğimizden birinci bölümde (GİRİŞ) laser ışıması, karekteri, çalışma şartlarını ve kullanım alanlarının anlatıldı. Aynı bölümde Azot laserin özellikleri ve çalışma şartlan anlatıldı. İkinci bölümde (MATERYEL ve METOD); azot laserin optik, elektrik, elektronik ve mekaniksel elemanların açıklandı. Bölüm 2.5'te bu elemanların nasıl birleştirilip bağlantılarının yapılacağı gösterildi. Üçüncü bölümde (BULGULAR) azot laser çalışmamızdan elde ettiğimiz laser ışıması incelendi. Dördüncü bölümde ( TARTIŞMA ve SONUÇ); elde edilen azot laser ışıması, özellikleri ve geliştirilmesi gereken yönleri ile açıklandı. EKLER bölümünde bu çalışmada kullanılan bazı teknik terimler örneklerle açıklandı. 67 SUMMARY In this study we intended to building a NITROGEN LASER, which works at atmospheric pressure. We produced very high voltage in very short time between electrodes (dVg/dt = lkV/ns) of nitrogen laser for success stimulated emission of fundamental principle of laser action. Nitrogen laser makes impulsive radiation in 337. lnm. During this study, we also examined laser emission technology and lasing characteristics. Another aim is to get technical background to build other laser types. In the first part (INTRODUCTION); we explained laser radiation characteristics, working condition of laser types and used fields. Under this chapter we discuss also nitrogen laser and examined characteristics. In the second chapter (MATERIAL and METHOD); we explained elements of the nitrogen laser system (optics, electric, electronics and mechanical parts). In chapter 2.5 we showed how can be done installation and connections of these elements. In chapter three (ARGUMANTS) we explained laser radiation, that we get from our nitrogen laser. In chapter four (DISCUSSION and CONCLUSION); we made clear nitrogen laser radiation, characteristics and base directions for improve these study. WOS: 000379683500001, PubMed ID: 27388324, Background: We evaluated the Bovine hydroxyapatite (BHA) structure. BHA powder was admixed with 5 and 10 wt% natural pumice (NP). Compression strength, Vickers micro hardness, Fourier transform infrared spectroscopy, scanning electron microscopy (SEM) and X-ray diffraction studies were performed on the final NP-BHA composite products. The cells proliferation was investigated by MTT assay and SEM. Furthermore, the antimicrobial activity of NP-BHA samples was interrogated. Results: Variances in the sintering temperature (for 5 wt% NP composites) between 1000 and 1300 degrees C, reveal about 700 % increase in the microhardness (similar to 100 and 775 HV, respectively). Composites prepared at 1300 degrees C demonstrate the greatest compression strength with comparable result for 5 wt% NP content (87 MPa), which are significantly better than those for 10 wt% and those that do not include any NP (below 60 MPa, respectively). Conclusion: The results suggested the optimal parameters for the preparation of NP-BHA composites with increased mechanical properties and biocompatibility. Changes in micro-hardness and compression strength can be tailored by the tuning the NP concentration and sintering temperature. Natural calcium phosphates derived from fish wastes are a promising material for biomedical application. However, their sintered ceramics are not fully characterized in terms of mechanical and biological properties. In this study, natural calcium phosphate was synthesized through a thermal calcination process from salmon fish bone wastes. The salmon-derived calcium phosphates (sCaP) were sintered at different temperatures to obtain natural calcium phosphate bioceramics and then were investigated in terms of their microstructure, mechanical properties and biocompatibility. In particular, this work is concerned with the effects of grain size on the relative density and microhardness of the sCaP bioceramics. Ca/P ratio of the sintered sCaP ranged from 1.73 to 1.52 when the sintering temperature was raised from 1000 to 1300 &deg, C. The crystal phase of all the sCaP bioceramics obtained was biphasic and composed of hydroxyapatite (HA) and tricalcium phosphate (TCP). The density and microhardness of the sCaP bioceramics increased in the temperature interval 1000&ndash, 1100 &deg, C, while at temperatures higher than 1100 &deg, C, these properties were not significantly altered. The highest compressive strength of 116 MPa was recorded for the samples sintered at 1100 &deg, C. In vitro biocompatibility was also examined in the behavior of osteosarcoma (Saos-2) cells, indicating that the sCaP bioceramics had no cytotoxicity effect. Salmon-derived biphasic calcium phosphates (BCP) have the potential to contribute to the development of bone substituted materials.
