Abstract

A comparison of radio-frequency (rf) glow discharges at 13.56 megahertz between dielectric electrodes in an Argon environment with discharges between aluminum electrodes reveals that the discharge amplitude in dielectric electrodes is less than that for metal electrodes. The rf glow discharges are determined by spatiotemporal optical emission spectroscopy. The total excitation rate is different for the two types of electrodes and is revealed as peaks in the anodic and cathodic cycle of the electrodes.

Abstract

Background: Pathologic subjective halitosis is known as a halitosis complaint without objective confirmation of halitosis by others or by halitometer measurements; it has been reported to be associated with social anxiety disorder. Olfactory reference syndrome is a preoccupation with the false belief that one emits a foul and offensive body odor. Generally, patients with olfactory reference syndrome are concerned with multiple body parts. However, the mouth is known to be the most common source of body odor for those with olfactory reference syndrome, which could imply that the two conditions share similar features. Therefore, we investigated potential causal relationships among pathologic subjective halitosis, olfactory reference syndrome, social anxiety, and preoccupations with body part odors. Methods: A total of 1360 female students (mean age 19.6 ± 1.1 years) answered a self-administered questionnaire regarding pathologic subjective halitosis, olfactory reference syndrome, social anxiety, and preoccupation with odors of body parts such as mouth, body, armpits, and feet. The scale for pathologic subjective halitosis followed that developed by Tsunoda et al.; participants were divided into three groups based on their scores (i.e., levels of pathologic subjective halitosis). A Bayesian network was used to analyze causal relationships between pathologic subjective halitosis, olfactory reference syndrome, social anxiety, and preoccupations with body part odors. Results: We found statistically significant differences in the results for olfactory reference syndrome and social anxiety among the various levels of pathologic subjective halitosis (P < 0.001). Residual analyses indicated that students with severe levels of pathologic subjective halitosis showed greater preoccupations with mouth and body odors (P < 0.05). Abstract

To investigate the role of reactive oxygen species (ROS) generation in the pathogenesis of adriamycin (ADR) nephropathy, chemiluminescence was measured using isolated glomeruli from ADR rats. The results showed a marked increase of chemiluminescence which paralleled the increase of albuminuria. A superoxide dismutase (SOD) mimic, which enters easily into the cells was administred and a significant decrease of proteinuria was noted. Addition of SOD in vitro inhibited the enhanced chemiluminescence in rats administered with ADR. A slight glomerular cell increase, but no recruitment of leucocytes and/or monocytes/marcrophages, was observed. We therefore conclude that ADR-induced nephropathy is mainly due to ROS generation of intrinsic glomerular cells.

Abstract

IL-1 receptor antagonist (IL-1Ra)-deficient mice spontaneously develop several inflammatory diseases, resembling rheumatoid arthritis, aortitis, and psoriasis in humans. As adoptive T cell transplantation could induce arthritis and aortitis in recipient mice, it was suggested that an autoimmune process is involved in the development of diseases. In contrast, as dermatitis developed in scid/scid-IL-1Ra-deficient mice and could not be induced by T cell transfer, a T cell-independent mechanism was suggested. The expression of proinflammatory cytokines was augmented at the inflammatory sites. The development of arthritis and aortitis was significantly suppressed by the deficiency of TNFα or IL-17. The development of dermatitis was also inhibited by the deficiency of TNFα. These observations suggest that TNFα and IL-17 play a crucial role in the development of autoimmunity downstream of IL-1 signaling, and excess IL-1 signaling-induced TNFα also induces skin inflammation in a T cell-independent manner. Abstract

It is unknown whether adipokines derived from adipose tissues modulate endoplasmic reticulum (ER) stress induced in obesity. Here, we show that visceral adipose tissue-derived serine protease inhibitor (vaspin) binds to cell-surface 78-kDa glucose-regulated protein (GRP78), which is recruited from ER to plasma membrane under ER stress. Vaspin transgenic mice were protected from diet-induced obesity, glucose intolerance, and hepatic steatosis, while vaspin-deficient mice developed glucose intolerance associated with upregulation of ER stress markers. With tandem affinity tag purification using HepG2 cells, we identified GRP78 as an interacting molecule. The complex formation of vaspin, GRP78, and murine tumor cell DnaJ-like protein 1 (MTJ-1) (DnaJ homolog, subfamily C, member 1) on plasma membrane was confirmed by cell-surface labeling with biotin and immunoprecipitation in liver tissues and H-4-II-E-C3 cells. The addition of recombinant human vaspin in the cultured H-4-II-E-C3 cells also increased the phosphorylation of Akt and AMP-activated protein kinase (AMPK) in a dose-dependent manner, and anti-GRP78 antibodies completely abrogated the vaspin-induced upregulation of pAkt and pAMPK. Vaspin is a novel ligand for cell-surface GRP78/MTJ-1 complex, and its subsequent signals exert beneficial effects on ER stress-induced metabolic dysfunctions. [ABSTRACT FROM AUTHOR]

Abstract

We have succeeded in transplanting a human osteogenic sarcoma of the mandible into athymic mice. The transplanted tumor showed marked chondrogenesis and mineralization. Recently, a cell line (USAC) with phenotypes of chondrocyte has been established from the transplanted tumor. USAC cells were stellate or spindle-shaped in sparse culture, but polygonal or spherical at sub-confluency to confluency. In long-term culture, the cells were condensed and calcified nodules were formed. Production of types I, II and X collagen were detected by immunohistochemical staining and Western blot analysis. Type I collagen was strongly expressed in the stellate or spindle-shaped cells. Although type II collagen was usually present in all cells during culture, it was strongly stained in polygonal cells at confluency. Type X collagen was seen in large polygonal cells around calcified nodules. Marked [35S]-sulfate uptake and metachromasia were seen at the confluent stage and in the nodule. The cells around the nodules were positive for alkaline phosphatase, and the center of the nodules was stained with alizarin red. The potentiality of cartilage formation was confirmed by in vivo experiments using a diffusion chamber in athymic mice. These observations indicate that USAC cells maintain characteristics of chondrocyte progenitor cells and thus may serve as a useful model to study the sequential events of chondrogenesis and the process of morbid endochondral calcification. This experiment also demonstrated that transplantation of tumor tissue into athymic mice is a convenient strategy for establishment of a cell line. [ABSTRACT FROM AUTHOR]

Abstract

Although the morphological features of osteosarcoma have been widely investigated, there is little information about its biochemistry. In this study, materials obtained from human calcified chondroblastic osteosarcoma were analyzed by microanalytical techniques revealing both morphological and biochemical characteristics at the same tissue site. Morphologically, tumor cells in the surface area showed atypical and mitotic figures. Hypertrophic chondroblastic cells appeared and increased in density closer to the calcified region. Almost all cells in the calcified region were hypertrophic. Biochemical analysis revealed that alkaline phosphatase activity was steeply elevated and inorganic phosphate gradually increased towards the calcified region. The spread of the calcified region was coincident with the contents of acid-soluble phosphate and calcium of the tumor. These results were similar to those found in endochondral calcification of epiphyseal growth cartilage. [ABSTRACT FROM AUTHOR]

Abstract

Five new tetrahydrofuran lignans (1−5), accompanied by four known compounds, were isolated from the ethyl acetate extract of Peperomia dindygulensis. Structures were elucidated mainly using 1D NMR, 2D NMR, and mass spectroscopic studies. The relative configurations of 1−5 were determined by NOE correlations. Several of the compounds showed weak growth inhibitory activity against three cell lines (WI-38, VA-13, and HepG2). Compound 5 exhibited stronger MDR (multidrug resistance) reversal activity than verapamil at 2.5 μg/mL in a cellular calcein accumulation assay. Compounds 4 and 5 showed weak inhibitory activity against induction of the intercellular adhesion molecule-1 (ICAM-1) in anti-inflammatory activity experiments.

Abstract

Treatment of the triruthenium pentahydrido complex {Cp*Ru(μ-H)}3(μ3-H)2 (1; Cp* = η5-C5Me5) with cycloalkene resulted in exclusive formation of the μ3-cycloalkyne complex {Cp*Ru(μ-H)}3(μ3-η2:η2(⊥)-CnH2n-4) (2a; n = 5, 2b; n = 6) as a result of vinylic C−H bond cleavage. VT NMR studies revealed fluxional behavior of the perpendicularly coordinated cycloalkyne ligand. Reaction of 2b with carbon monoxide afforded the 48-electron complex (Cp*Ru)3(μ-CO)(μ3-CO)(μ-H)(μ3-η2(∥)-C6H8) (3b) with a parallel-coordinated alkyne ligand.

Abstract

Reaction of the cationic η2-alkyne iron complex, [Fp(η2-Ph-C&tbd1;CPh)]BF4 (1) [Fp = (η5-C5H5)Fe(CO)2], with various O-, N-, and C-nucleophiles afforded products containing the Ph−C&dbd;C−Ph linkage. i.e., cis- (3) and trans-alkenyl complexes (4), Fp−C(Ph)&dbd;C(Ph)−Nu, and metallacycles, Cp(CO)Fe−C(Ph)&dbd;C(Ph)−C(&dbd;O)−Nu [coordinated at &dbd;O (5) or Nu (6)]. Crystallographic determination of the configuration of the C&dbd;C moiety in 3 and 4 and analysis of the product distribution revealed that basic nucleophiles bearing alkyl substituents produced the cis-alkenyl complex (3) and/or the metallacycles (5, 6), whereas less basic nucleophiles bearing aryl substituents afforded the trans-alkenyl complex (4). Correlation between the product distribution and pKb values of the nucleophiles leads to the conclusions that (1) products 3, 5, and 6 arise from acyl intermediates formed by initial nucleophilic addition of basic nucleophiles at the CO ligand, (2) trans-alkenyl complexes 4 result from addition of less basic nucleophiles to the η2-alkyne ligand from the exo-side, and (3) the equilibrium of the CO addition governed by the basicity of the nucleophile is the key step of the diversity of the stereochemistry of the alkenyl products. Reactions of 2-hexyne complex, [Fp(η2-Et−C&tbd1;C−Et)]BF4 (2), were also examined.
