Abstract Lightweight thin-walled parts are widely used in the aviation and aerospace industries, and with the further increase in the complexity of their features, the traditional manufacturing process can no longer fully meet the high requirements of industrial component manufacturing. In this work, thin-walled parts are processed by Laser based powder bed fusion of metals (PBF-LB/M), and the effects of process parameters on residual stress, hardness, mechanical properties and microstructure of thin-walled parts are systematically investigated. The simulation results show that the maximum equivalent residual stresses are distributed in the combination of the solid and the substrate, and the minimum equivalent residual stresses are mainly distributed in the top two ends and the middle part of the solid, and the stress distribution is symmetrical. In addition, the maximum equivalent residual stress increases with the increase of laser power, and decreases with the increase of scanning spacing or scanning speed. The experimental results show that with the increase of laser energy density, the tensile strength and yield strength of thin-walled parts show a tendency of increasing first and then decreasing. Finally, high-quality thin-walled parts were successfully fabricated by the optimized process parameters, and their tensile and yield strengths were increased by 6.1% and 15.9%, respectively.

Abstract African swine fever (ASF) is an acute and fatal hemorrhagic disease in domestic pigs and wild boars caused by African swine fever virus (ASFV) that currently threatens the pig industry worldwide. Since the 2018 ASF outbreak in China, ASFV has evolved and caused diverse clinical manifestations, such as chronic and asymptomatic infections. Therefore, it is important to understand the molecular mechanisms underlying ASFV attenuation in the field. Here, we isolated three ASFVs from one diseased and two asymptomatic pigs by using primary porcine alveolar macrophages (PAMs) from both domestic pigs and Bama minipigs. The three ASFVs exhibited similar phenotypes in cell culture, including cytopathic effects (CPEs), hemadsorptions (HADs), viral protein expressions and growth curves. Genome sequencing revealed that all three ASFVs were genotype II strains. Genomic comparisons suggested that the disruption of the viral genes MGF360 and MGF110, rather than EP402R and EP153R, is likely involved in the potential attenuation of ASFV via the upregulation of innate immune responses.

Globally, anthropogenic disturbance and climate change caused a rapid decline of submerged macrophytes in lake ecosystems. Potamogeton crispus (P. crispus), a species that germinates in winter, explosively expanded throughout many Chinese lakes, yet the underlying mechanism remained unclear. Here, this study examined the long-term changes in the distribution patterns of P. crispus in Lake Gaoyou by combining remote sensing images and hydrometeorological data from 1984 to 2022 and water quality data from 2009 to 2022. It aims to unravel the relationships between the distribution patterns of P. crispus and hydrometeorological and water quality factors. The results showed that the area of P. crispus in Lake Gaoyou showed a slight increase from 1984 to 2009, a marked increase from 2010 to 2019, followed by a decline after 2020. Spatially, P. crispus was primarily distributed in the western and northern parts of Lake Gaoyou, with less distribution in the central and southeastern parts of the lake. Wind speed (WS), temperature (Temp), water level (WL), ammonia nitrogen (NH3-N), and Secchi depth (SD) were identified as the key factors regulating the variation in the P. crispus area in Lake Gaoyou. We found that the P. crispus area showed an increasing trend with increasing Temp, WL, and SD and decreasing WS and NH3-N. The influence of environmental factors on the area of P. crispus in Lake Gaoyou varied among seasons. The results indicated that hydrometeorology (WS, Temp, and WL) may override water quality (NH3-N and SD) in driving the succession of P. crispus distribution. The findings of this study offer valuable insights into the recent widespread expansion of P. crispus in shallow lakes across Eastern China.

The intestinal tract plays an important role in digestion and absorption, nutrient metabolism, and immune anti-stress. At the same time, the intestine is also the major site for pathogens and various harmful substances to invade. Probiotics, as living microbial agents, improve animal health by maintaining the microecological balance of the host's intestinal flora. To explore the protective effect of Clostridium butyricum (CB) on the gut, we investigated the changes in digestion, antioxidant, and gut microbiota in juvenile largemouth bass. The fish were fed with diets containing 0 (control, CB0), 4.3×108 (CB1), 7.5×108 (CB2), 1.5×109 (CB3), and 3.2×109 (CB4) CFU/kg Clostridium butyricum for 56 days. After 56 days of dietary supplementation with 4.3×108 and 3.2×109 CFU/kg of CB, significant growth promotion and feed utilization improvement were observed in largemouth bass (P < 0.05). The activities of digestive enzymes (trypsin, protease and amylase) and intestinal functional indicators (sodium-potassium ATP, γ-GT and CK) were significantly increased in the intestine of largemouth bass (P < 0.05). The contents of intestinal short-chain fatty acids (acetic acid, propionic acid, isobutyric acid, N-butyric acid and isovaleric acid) in largemouth bass were significantly increased (P < 0.05). CB significantly increased the activities of antioxidant enzymes (T-AOC, CAT, POD, GSH-Px and SOD) and decreased the content of MDA in the intestine of largemouth bass (P < 0.05). It was also observed that CB significantly increased the activities of immune-related markers lysozyme and alkaline phosphatase (P < 0.05). High-throughput sequencing of the 16 S rRNA gene in largemouth bass revealed a significant increase in gut microbial richness. KEGG pathway analysis showed that CB had significant effects on intestinal microbiota functions, such as nutrient metabolism and energy conversion. The results showed that CB could stimulate intestine to produce the organic product butyric acid, which was customized in the gut to enhance digestive performance, promote nutrient metabolism, and protect microbial homeostasis in the intestinal tract of largemouth bass. These results provide new insights into the protective effects of CB on the intestinal tract of aquatic animals.

Spinal cord injury (SCI) is a type of central nervous system (CNS) injury in which ferroptosis is becoming a promising target for treatment. Alpha-tocopherol (Vitamin E, Vit E) is a compound with anti-ferroptosis activity. The mechanism of alpha-tocopherol in regulating ferroptosis after SCI has not been deeply studied. In this study, rats with SCI were treated by Alpha-tocopherol based on bioinformatic analysis and molecular docking prediction. Behavioral tests and histological findings showed that Alpha-tocopherol promoted neural function recovery and tissue repairment in rats with SCI. Subsequently, regulatory effects of Alpha-tocopherol on Alox15 and ferroptosis were detected and then localized by immunofluorescence. In vitro, alpha-tocopherol improved the ROS accumulation, iron overload, lipid peroxidation and mitochondrial dysfunction. The effects of Alpha-tocopherol on the expression of Alox15, Ptgs2 and 4Hne were validated in vitro. Finally, the inhibitory effects of Alpha-tocopherol on Alox15 and ferroptosis were weakened by the mutation of 87th residue of Alox15. In summary, alpha-tocopherol could alleviate SCI-induced ferroptosis by downregulating Alox15 to promote neural function recovery in rats with SCI. Findings in this study could help further our understanding on SCI-induced ferroptosis and provide a novel insight for treating SCI.

ABSTRACTTigecycline and colistin were referred to as the “last resort” antibiotics in defending against carbapenem-resistant, Gram-negative bacterial infections, and are currently widely used in clinical treatment. However, the emergence and prevalence of plasmid-mediated tet(X4) and mcr-1 genes pose a serious threat to the therapeutic application of tigecycline and colistin, respectively. In this research, a tigecycline- and colistin-resistant bacteria resensitization system was developed based on efficient and specific DNA damage caused by Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)-Associated Protein 9 (Cas9) nucleases. A conjugation method was used to deliver the resensitization system, which harbors two single-guide RNAs targeting tet(X4) and mcr-1 genes and constitutively expressed Cas9. The conjugation efficiency was nearly 100% after conjugation condition optimization in vitro, and the resensitivity efficiency for clinical isolates was over 90%. In addition, when performing resensitization in vivo, the resistance marker was replaced with a glutamate-based, chromosomal, plasmid-balanced lethal system to prevent the introduction of additional resistance genes in clinical settings, making this strategy a therapeutic approach to combat the in vivo spread of antibiotic resistance genes (ARGs) among bacterial pathogens. As a proof of concept, this resensitive system can significantly decrease the counts of tigecycline- and colistin-resistant bacteria to 1% in vivo. Our study demonstrates the efficacy and adaptability of CRISPR-Cas systems as powerful and programmable antimicrobials in resensitizing tet(X4)- and mcr-1-mediated, tigecycline- and colistin-resistant strains, and opens up new pathways for the development of CRISPR-based tools for selective bacterial pathogen elimination and precise microbiome composition change.IMPORTANCEThe emergence of plasmid-encoded tet(X4) and mcr-1 isolated from human and animal sources has affected the treatment of tigecycline and colistin, and has posed a significant threat to public health. Tigecycline and colistin are considered as the “last line of defense” for the treatment of multidrug-resistant (MDR) Gram-negative bacterial infections, so there is an urgent need to find a method that can resensitize tet(X4)-mediated tigecycline-resistant and mcr-1-mediated colistin-resistant bacteria. In this study, we developed a glutamate-based, chromosomal, plasmid-balanced lethal conjugative CRISPR/Cas9 system, which can simultaneously resensitize tet(X4)-mediated tigecycline-resistant and mcr-1-mediated colistin-resistant Escherichia coli. The counts of tigecycline- and colistin-resistant bacteria decreased to 1% in vivo after the resensitization system was administered. This study opens up new pathways for the development of CRISPR-based tools for selective bacterial pathogen elimination and precise microbiome composition change.

Abstract Mass spectrometry-based proteomics plays a critical role in current biological and clinical research. Technical issues like data integration, missing value imputation, batch effect correction and the exploration of inter-connections amongst these technical issues, can produce errors but are not well studied. Although proteomic technologies have improved significantly in recent years, this alone cannot resolve these issues. What is needed are better algorithms and data processing knowledge. But to obtain these, we need appropriate proteomics datasets for exploration, investigation, and benchmarking. To meet this need, we developed MultiPro (Multi-purpose Proteome Resource), a resource comprising four comprehensive large-scale proteomics datasets with deliberate batch effects using the latest parallel accumulation-serial fragmentation in both Data-Dependent Acquisition (DDA) and Data Independent Acquisition (DIA) modes. Each dataset contains a balanced two-class design based on well-characterized and widely studied cell lines (A549 vs K562 or HCC1806 vs HS578T) with 48 or 36 biological and technical replicates altogether, allowing for investigation of a multitude of technical issues. These datasets allow for investigation of inter-connections between class and batch factors, or to develop approaches to compare and integrate data from DDA and DIA platforms.

Abstract Background Genetic variants of outer dynein arm docking complex subunit 2 (ODAD2) have been reported to be closely associated with primary ciliary dyskinesia and colorectal cancer in previous studies, but the association of genetic variants of ODAD2 with hepatocellular carcinoma (HCC) has not been reported. Methods We enrolled 80 healthy subjects and 468 Guangxi hepatitis B virus (HBV)-related HCC patients in this study. A case–control study method was used to explore the association of different ODAD2-rs7893462 genotypes with hepatocarcinogenesis. A comprehensive survival analysis was used to explore the association of rs7893462 with the prognosis of HBV-related HCC in Guangxi. Results Through a case–control study, we observed that patients carrying the G allele of rs7893462 had a markedly increased susceptibility to hepatocarcinogenesis (odds ratio = 1.712, 95% confidence interval = 1.032–2.839, P = 0.037). We found that there were significant prognosis differences among three different genotypes of rs7893462. Nomogram analysis suggested that the contribution of rs7893462 polymorphisms to the prognosis of HBV-related HCC was second only to the BCLC stage. Stratified survival analysis suggested that the AG genotype of rs7893462 was an independent prognostic risk factor for HBV-related HCC. Joint effect survival analysis also observed that the AG genotype of rs7893462 combined with clinical parameters could significantly identify HBV-related HCC patients with different prognostic outcomes more accurately, and the AG genotype was also observed to be independent of clinical factors in HBV-related HCC survival. Conclusion The ODAD2-rs7893462 polymorphisms can be used as an independent prognostic indicator of HBV-related HCC overall survival and are significantly associated with susceptibility to hepatocarcinogenesis.

A large language model refers to a deep learning model characterized by extensive parameters and pretraining on a large-scale corpus, utilized for processing natural language text and generating high-quality text output. The increasing deployment of large language models has brought significant attention to their associated privacy and security issues. Recent experiments have demonstrated that training data can be extracted from these models due to their memory effect. Initially, research on large language model training data extraction focused primarily on non-targeted methods. However, following the introduction of targeted training data extraction by Carlini et al., prefix-based extraction methods to generate suffixes have garnered considerable interest, although current extraction precision remains low. This paper focuses on the targeted extraction of training data, employing various methods to enhance the precision and speed of the extraction process. Building on the work of Yu et al., we conduct a comprehensive analysis of the impact of different suffix generation methods on the precision of suffix generation. Additionally, we examine the quality and diversity of text generated by various suffix generation strategies. The study also applies membership inference attacks based on neighborhood comparison to the extraction of training data in large language models, conducting thorough evaluations and comparisons. The effectiveness of membership inference attacks in extracting training data from large language models is assessed, and the performance of different membership inference attacks is compared. Hyperparameter tuning is performed on multiple parameters to enhance the extraction of training data. Experimental results indicate that the proposed method significantly improves extraction precision compared to previous approaches.

【Objective】In order to provide reference for the application of condensed tannins in Micropterus salmoides feed, this study was conducted to assess the effects of short-term feeding condensed tannins on growth performance and liver health of large-sized M. salmoides.【Method】A total of 600 fish with an initial weight of 510 (±10.4) g were selected and randomly divided into 4 groups, with 3 replicates per group and 50 fish per replicates. Fish in the control group were fed with basic diet (G1), while fish in the experimental groups were fed with basic diet supplemented with 0.1% (G2), 0.2% (G3) and 0.4% (G4) of condensed tannins, respectively. The experimental period was 30 d.【Result】Compared with G1, the feed coefficient of fish in G4 was significantly increased by 17.44% (P < 0.05), while the survival rate was significantly decreased by 15.79% (P < 0.05); The abdominal fat rate of fish in G2, G3, and G4 were significantly decreased by 12.03%, 16.18%, and 13.69% respectively (P < 0.05). There was no significant difference in feed intake, weight gain rate, specific growth rate among all groups; The contents of serum glucose, total cholesterol, and triglycerides of fish in G4 were significantly decreased by 49.73%, 28.46% and 35.71%, respectively (P < 0.05), while the contents of serum low-density lipoprotein cholesterol of fish in G3 and G4 were significantly decreased by 38.71% and 51.61% respectively (P < 0.05). There was no significant difference in the contents of albumin, globulin, high-density lipoprotein cholesterol and the activities of alanine transaminase and glutamic transaminase among all groups; With the increase of dietary condensed tannins, the "fatty liver" status of large-sized M. salmoides was improved, the vacuolation and inflammatory infiltration of liver tissue were also weakened, and the liver lipid content and lipid droplet area ratio were significantly reduced.【Conclusion】Dietary supplemented with 0.1% and 0.2% of condensed tannins had no significant effects on the survival rate and feed coefficient of large-sized M. salmoides. However, it could promote the metabolism of glucose and lipid, reduce liver fat and abdominal fat, and improve liver health of fish. If the evaluation is based on liver health, the appropriate concentration of condensed tannins in the diet of M. salmoides is 0.2%.

The hydraulic characteristics of the lock chamber section in a bottom outlet are complex. Particularly, cavitation is a prominent problem when the flow in the tunnel transitions from pressurized to unpressurized conditions. As a countermeasure, sidewall contractions are often installed at the end of the pressurized section. In this paper, with numerical simulations and vacuum tank experiments, we analyzed the design parameters of several prototype projects and studied the flow properties and cavitation characteristics of the transition section. The four main factors that influence cavitation risks are the contraction ratio of the transition section, the top press slope, the length of the transition section, and the shape of the orifice. The study of a real-world project demonstrates that by implementing a severe constriction degree, the top press slope can be slowed down, and the pressurized transition section can be extended, thereby reducing cavitation risks and improving flow patterns. These research results provide a reference for the optimal design of hydraulic structures. In the realm of sustainable city development, evaluating the spatial vitality of urban green spaces (UGS) has become increasingly pivotal for assessing public space quality. This study delves into the spatial heterogeneity of park vitality across diverse urban landscapes at a city scale, addressing limitations inherent in conventional approaches to understanding the dynamics of park vitality. Leveraging geotagged check-in data from 65 parks in the study case of Changsha City, a quantitative analysis was undertaken to assess spatial vitality. The investigation incorporated data concerning internal and external factors influencing park vitality, employing the Multi-scale Geographically Weighted Regression (MGWR) model to dissect nuanced spatial heterogeneity. The research uncovers notable spatial discrepancies in factors influencing park vitality across diverse urban areas, emphasizing the reliance on adjacent residential communities and internal commercial amenities provision. These dependencies correspond with economic development differences among urban locales, revealing distinct geographic trends. This study has a novel perspective and methodology for investigating urban park vitality, providing significant insights for urban green space planning and management. It emphasizes the necessity of acknowledging spatial diversity in urban park planning and design by incorporating the distinct socio-economic characteristics of each urban zone, which is crucial for both urban planners and policymakers.

BackgroundLiver metastasis is one of the most common causes of death in patients with colorectal cancer. Therefore, improving the treatment effect of liver metastatic carcinoma of colorectal cancer is also one of the effective ways to improve the survival time of patients with colorectal cancer. The main treatment method for liver metastasis of colorectal cancer is preoperative neoadjuvant chemotherapy through intravenous administration. However, no one has reported a conversion therapy approach for the treatment of colorectal cancer liver metastases patients through arterial infusion chemotherapy combined with targeted agents and PD-1 monoclonal antibody. This case reports a conversion therapy method of liver metastases of colorectal cancer by hepatic arterial infusion chemotherapy (HAIC), sintilimab injection combined with lenvatinib to achieve radical resection of liver metastatic carcinoma after treatment.Case presentationThe patient was a 69-year-old man who had previously undergone laparoscopic left hemicolectomy for descending colorectal cancer and multiple interventional and surgical treatments for hepatocellular carcinoma. During this treatment, the patient underwent radiological and serological tests, and primary liver cancer was considered at the initial diagnosis stage. Therefore, this liver malignant tumor lesion was treated according to the primary liver cancer treatment protocol before surgical resection. Therefore, the patient received HAIC combined with sintilimab injection and lenvatinib. After three treatment cycles, radiological examination showed no obvious tumor activity, alpha-fetoprotein (AFP) decreased to normal, protein induced by vitamin K absence or antagonist II (PIVKA II) decreased significantly, and the curative effect was evaluated as complete remission. Subsequently, we performed surgical resection of this liver lesion. The pathological response of left lobe tumor was partial remission (PR). Background: Neurodegenerative retinal diseases such as retinitis pigmentosa are serious disorders that may cause irreversible visual impairment. Ferroptosis is a novel type of programmed cell death, and the involvement of ferroptosis in retinal degeneration is still unclear. This study aimed to investigate the related ferroptosis genes in a mice model of retinal degeneration induced by light damage. Methods: A public dataset of GSE10528 deriving from the Gene Expression Omnibus database was analyzed to identify the differentially expressed genes (DEGs). Gene set enrichment analysis between light damage and control group was conducted. The differentially expressed ferroptosis-related genes (DE-FRGs) were subsequently identified by intersecting the DEGs with a ferroptosis genes dataset retrieved from the FerrDb database. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) were further performed using the DE-FRGs. A protein-protein interaction (PPI) network was constructed to identify hub ferroptosis-related genes (HFRGs). The microRNAs (miRNAs)-HFRGs, transcription factors (TFs)-HFRGs networks as well as target drugs potentially interacting with HFRGs were analyzed utilizing bioinformatics algorithms. Results: A total of 932 DEGs were identified between the light damage and control group. Among these, 25 genes were associated with ferroptosis. GO and KEGG analyses revealed that these DE-FRGs were mainly enriched in apoptotic signaling pathway, response to oxidative stress and autophagy, ferroptosis, necroptosis and cytosolic DNA-sensing pathway. Through PPI network analysis, six hub ferroptosis-related genes (Jun, Stat3, Hmox1, Atf3, Hspa5 and Ripk1) were ultimately identified. All of them were upregulated in light damage retinas, as verified by the GSE146176 dataset. Bioinformatics analyses predicated that 116 miRNAs, 23 TFs and several potential therapeutic compounds might interact with the identified HFRGs. Conclusion: Our study may provide novel potential biomarkers, therapeutic targets and new insights into the ferroptosis landscape in retinal neurodegenerative diseases.

ABSTRACT Integrative conjugative elements (ICEs) are important mobile elements that are associated with the dissemination of antibiotic resistance genes (ARGs) in Proteus. Recent studies demonstrated that the tigecycline resistance gene cluster tmexCD-toprJ has emerged in ICEs of Proteus. However, the prevalence of tmexCD-toprJ positive Proteus from animal sources is unclear. To cover the gap, a total of 762 Proteus spp. were isolated from animal source from six provinces of China to identify the tmexCD-toprJ positive isolates. Eight tmexCD-toprJ positive isolates were identified, with the tmexCD-toprJ positive rate of 1.05% against all Proteus spp. and 2.79% against ICE-bearing Proteus spp. The tmexCD-toprJ gene cluster in these Proteus spp. were tmexCD3-toprJ1b and all of them were carried by ICEs. Genetic structure analysis showed that tmexCD3-toprJ1b-bearing ICEs were complicated and plastic, but the tmexCD3-toprJ1b was specifically integrated into variable region III (VRIII) of ICEs with the help of integrases. Furthermore, we found that the umuC gene, which is presented in VRIII of ICEs and other genetic structures of many other bacterial genomes, was a hotspot for the integration of tmexCD-toprJ-bearing potential mobile elements. In summary, our results suggested that Proteus were important reservoirs for tmexCD-toprJ due to the high prevalence of ICEs. Therefore, continuous surveillance of ICEs-associated ARGs in Proteus is necessary and significant for controlling their future transmission. IMPORTANCE The emergence and spread of tmexCD-toprJ have greatly weakened the function of tigecycline. Although studies have demonstrated the significance of Proteus as carriers for tmexCD-toprJ, the epidemic mechanism and characteristics of tmexCD-toprJ in Proteus remain unclear. Herein, we deciphered that the umuC gene in VRIII of SXT/R391 ICEs was a hotspot for the integration of tmexCD3-toprJ1b-bearing mobile genetic elements by genomic analysis. The mobilization and dissemination of tmexCD3-toprJ1b in Proteus were mediated by highly prevalent ICEs. Furthermore, the co-occurrence of tmexCD3-toprJ1b-bearing ICEs with other chromosomally encoded multidrug resistance gene islands warned that the chromosomes of Proteus are significant reservoirs of ARGs. Overall, our results provide significant insights for the prevention and control of tmexCD3-toprJ1b in Proteus.

In multi-stage machining-assembly production, collaborative scheduling for multiple production lines can effectively improve the execution efficiency of production planning and increase the effective output of the production system. In this paper, a production scheduling mathematical model was constructed for the collaborative scheduling problem of machining-assembly multi-production lines with kitting constraints, with the optimization objectives of minimizing assembly completion time and tardiness time. For the scheduling model, the product assembly process is constrained by the machining sequence of the jobs on the machining lines. Only by collaborating on the production scheduling schemes of the machine line and the assembly line as a whole can the output efficiency of the product on the assembly line be improved. An improved hybrid multi-objective optimization algorithm named SMOEA/D is designed to solve this scheduling model. The algorithm uses adaptive parents’ selection and mutation rate strategies and integrates the Tabu search strategy for the search process in the solution space when the solution of the sub-problem has not been improved after specified search generations, to improve the local search ability and search accuracy of MOEA/D algorithm. To verify the performance of the SMOEA/D algorithm in solving machining-assembly collaborative scheduling problems in production systems with different resource configurations and scales, two sets of numerical experiments were designed, corresponding to situations where the number of operations on each production line is equal or unequal. The running results of the proposed algorithm were compared with three other well-known multi-objective algorithms. The comparison results indicate that the SMOEA/D algorithm is effective and superior for solving such problems.

The effects of selenium supplementation in Tachysurus fulvidraco (T. fulvidraco) on low-temperature stress are not known. In this study, 280 healthy T. fulvidraco were divided into two groups, the G0 group (a control group) and the T0 group (a selenium treatment group on a 0.22 mg/kg diet), for a 6-week feeding time. Then, low-temperature stress (water temperature dropped from 26 to 13 °C, with a rate of 1 °C/h) was administered after that. The feeding results showed that selenium increased the percent weight gain (PWG), specific growth rate (SGR), and survival rate (SR) of T. fulvidraco and decreased the feed conversion rate (FCR), but these differences were not significant (p > 0.05). Under low temperatures, selenium still has no significant effects on antioxidant indexes such as glutathione peroxidase (GSH-Px) activity, superoxide dismutase (SOD) activity, and malondialdehyde (MDA) content in serum (p > 0.05). However, metabolomic analysis revealed that selenium caused changes in lipids and lipid-like molles, organic acids and their derivatives, and fatty acyls. Choline, linoleic acid, and glycerophospholipid metabolism pathways; d-arginine and d-ornithine metabolism; valine, leucine, and isoleucine degradation; and biosynthesis pathways, as well as pyrimidine metabolism pathways, were activated to produce these metabolites to combat against this stress. In addition, selenium increased the diversity of intestinal microbes in T. fulvidraco and decreased the relative abundance of Plesiomonas. However, the combined analysis showed the intestinal microbe changes did not affect metabolite production. In summary, selenium activated lipid, carbohydrate, and amino acid metabolism for energy substance provision, reduced the oxidation and production of other harmful substances, and increased the intestinal microbe diversity of T. fulvidraco to improve resistance to low-temperature stress.

The fourth mobile sulfonamide resistance gene sul4 has been discovered in many metagenomic datasets. However, there is no reports of it in cultured bacteria. In this study, a sul4 positive clinical Salmonella enterica SC2020597 was obtained by conventional Salmonella isolation methods and characterized by species identification and antimicrobial susceptibility testing. Meanwhile, the genomic DNA was sequenced using both long-read and short-read methods. Following that, the complete genome was analyzed by bioinformatic methods. The sul4 gene in S. enterica SC2020597 differed from the sul4 identified in metagenomic data by one amino acid and could confer full resistance to sulfamethoxazole. Genetic location analysis showed that the sul4 in SC2020597 was carried by a complex chromosomally integrated hybrid plasmid. ISCR20-like was strongly associated with the mobilization of sul4 by core genetic context analysis. To the best of our knowledge, this is the first report of the emergence of sul4 in clinically cultured S. enterica. More important, the sul4 has the potential to spread to other bacteria with the help of mobile elements.

Salmonella is one of the most important zoonotic pathogens and a major cause of foodborne illnesses, posing a serious global public health hazard. The emergence of plasmid-mediated mcr genes in Salmonella has greatly reduced the clinical choice of salmonellosis treatment. The aim of this study was to investigate the plasmid characteristics of mcr-positive Salmonella identified from patients in Sichuan, China during 2014 to 2017 by whole genomes sequencing. In this study, a total of 12 mcr-positive isolates (1.15%, ; mcr-1, n=10; mcr-3, n=2) were identified from 1046 Salmonella isolates using PCR. Further characterization of these isolates was performed through antimicrobial susceptibility testing, conjugation assays, whole genome sequencing, and bioinformatics analysis. The mcr-1 gene in these isolates were carried by three types of typical mcr-1-bearing plasmids widely distributed in Enterobacteriaceae (IncX4, IncI2 and IncHI2). Of note, two mcr-1-harboring IncHI2 plasmids were integrated into chromosomes by insertion sequences. Two mcr-3-bearing plasmids were IncC and IncFIB broad-host-range plasmids respectively. Genetic context analysis found that mcr-1 was mainly located in Tn6330 or truncated Tn6300, and mcr-3 shared a common genetic structure tnpA-mcr-3-dgkA-ISKpn40. Overall, we found that mcr gene in clinical Salmonella were commonly carried by broad-host plasmids and have potential to transfer into other bacteria by these plasmids. Continuous surveillance of MDR Salmonella in humans and investigation the underlying transmission mechanisms of ARGs are vital to curb the current severe AMR concern.

Federated Learning (FL) is a new computing paradigm in privacy-preserving Machine Learning (ML), where the ML model is trained in a decentralized manner by the clients, preventing the server from directly accessing privacy-sensitive data from the clients. Unfortunately, recent advances have shown potential risks for user-level privacy breaches under the cross-silo FL framework. In this paper, we propose addressing the issue by using a three-plane framework to secure the cross-silo FL, taking advantage of the Local Differential Privacy (LDP) mechanism. The key insight here is that LDP can provide strong data privacy protection while still retaining user data statistics to preserve its high utility. Experimental results on three real-world datasets demonstrate the effectiveness of our framework.

Objective To investigate the current situation concerning volume control of red blood cells in additive solution produced by blood service in Chongqing, and to lay a foundation for promoting the homogenization of preparation process of red blood cells in additive solution. Methods A questionnaire was designed to investigate the factors related to the preparation of red blood cells in additive solution. The questionnaire was sent by Chongqing Association of Blood Transfusion via E-mail to 18 blood services in the city, and the collected data was sorted, revised and analyzed by research team. Results A total of 18 blood services(including 1 blood center + 1 sub-center, 6 central blood stations and 11 central blood banks) returned the questionnaires. The results showed that there were differences among blood services across Chongqing, regarding the centrifugal parameters during preparation, the operation mode and monitoring situation of the capacity control during preparation, and the formulation of the capacity standard of red blood cells in additive solution etc. Conclusion The preparation process of red blood cells in additive solution, produced by Chongqing blood services, should be further standardized, and the capacity control method of this product in Chongqing should be gradually unified to achieve regional homogeneity and to ensure blood safety.

Chinese seabass (Lateolabrax maculatus) is a popular carnivorous fish with delicious taste. Although the feed value of condensed tannins has been well documented for L. maculatus, information about the effects of hydrolyzable tannins (HTs) on the growth and health of L. maculatus is limited. This study was conducted to assess the effects of dietary HTs on growth performance, body composition, intestinal digestive enzyme activities, serum metabolites, antioxidant and immune response, and intestine and liver morphology of L. maculatus. A total of 640 fish were randomly divided into four groups with four replicates per group and 40 fish per replicate. Four diets were prepared to contain 0 (G0), 1 (G1), 2 (G2), and 4 (G4) g/kg of HTs. Fish were fed to apparent satiation twice a day during the 56-day feeding trial. Results showed that the final body weight, weight gain rate, specific growth rate, and feed intake were linearly decreased (p< 0.001) as dietary HTs increased. All fish had similar (p > 0.05) whole body compositions. Fish fed G2 and G4 had lower (p< 0.05) intestinal trypsin and lipase activities than those fed G0 and G1, whereas G4 had higher (p< 0.05) aspartate aminotransferase and alanine aminotransferase activities than G0. Serum total antioxidant capacity and lysozyme were linearly decreased (p< 0.01), but the malondialdehyde concentration was linearly increased (p< 0.01) as dietary HTs increased. Intestinal villi in G2 and G4 showed increased deformation, and the vacuolation of liver cells began to appear in G1 and was aggravated as dietary HTs increased. This study showed that HTs should be used with caution due to their growth-inhibiting effect, and the dietary HT level for L. maculatus is recommended to be less than 1 g/kg.

Non-small cell lung cancer (NSCLC) is a prominent etiology of cancer-related mortality. The heterogeneous nature of this disease impedes its accurate diagnosis and efficacious treatment. Consequently, constant advancements in research are imperative in order to comprehend its intricate nature. In addition to currently available therapies, the utilization of nanotechnology presents an opportunity to enhance the clinical outcomes of NSCLC patients. Notably, the burgeoning knowledge of the interaction between the immune system and cancer itself paves the way for developing novel, emerging immunotherapies for treating NSCLC in the early stages of the disease. It is believed that with the novel engineering avenues of nanomedicine, there is a possibility to overcome the inherent limitations derived from conventional and emerging treatments, such as off-site drug cytotoxicity, drug resistance, and administration methods. Combining nanotechnology with the convergence points of current therapies could open up new avenues for meeting the unmet needs of NSCLC treatment.

Cyclin-dependent kinases (CDKs) are a large family of proteins exerting different regulatory functions in eukaryotic cells, including control over the cell cycle and gene transcription. CDK1 controls the cell cycle and promotes cell proliferation by regulating the initiation and transition of the G2/M phase. Similar to mammals, CDK1 has also been characterized in arthropods; however, its functions and physiological importance in crustaceans remain unknown. Our pervious transcriptomic data indicated that CDK1 was a candidate cold-stress response gene in the Pacific white shrimp (Litopenaeus vannamei). Therefore, in the present study, the full-length sequence of CDK1 (LvCDK1) was obtained from L. vannamei containing a conserved PSTAIRE sequence and ATP-binding region. The expression of LvCDK1 is tissue-specific, with relatively higher in the hepatopancreas. The in situ hybridization (ISH) signals for LvCDK1 were present in the distal zone of hepatopancreas and mainly located in the cytoplasm and nucleus. The expression of LvCDK1 in hepatopancreatic was sensitive to environmental temperature. Furthermore, under cold stress, knockdown of LvCDK1 led to increased mortality in shrimp. Our study provides the first evidence of LvCDK1 responding to cold stress in shrimps. This mechanism for modifying the transcriptional roles of LvCDK1 may show new light on the molecular regulation of crustaceans in response to cold stress.

Objective To provide reference for formulating relatively unified quality control strategies and meeting the requirements of homogenization construction of blood banks across Chongqing area by retrospectively analyzing sampling results of different blood components during the past two years in all levels of blood banks in Chongqing area. Methods The key quality data of blood components prepared by 6 blood banks in Chongqing were analyzed retrospectively. At present, the fault diagnosis methods of lithium battery pole rolling mill mostly rely on manual experience and the self-test function of mature control devices such as frequency converters and lack the ability of intelligent fault diagnosis for the whole equipment and the ability to evaluate the health state of the equipment during operation. To improve the intellectual health diagnosis ability of lithium battery pole double rolling mill equipment, starting from the structure and technology of lithium battery pole double rolling equipment, this paper analyzes its common fault types. It summarizes the shortcomings and common fault types of existing equipment. Then, we introduce fuzzy reasoning into the fault diagnosis method based on Expert Systems and establish the FEFDM of lithium battery pole double rolling equipment. Finally, we introduce the concept of health degree, effectively connect BP neural network and health degree through the fuzzy set, and establish an equipment operation health state evaluation method based on an improved BP Neural Network, which realizes the evaluation ability of the health state of double roller equipment. In addition, we use Extended Kalman Filtering (EKF) to clean the “dirty data” and filter out the Gaussian white noise from the signal. The health diagnosis method proposed in this paper can meet the ability to accurately locate and diagnose the fault of lithium battery pole double roller equipment and evaluate the health state of equipment operation and maintain the equipment in advance.

Tension control is very important for the intelligent control system of rapier looms because stable tension guarantees a tight fabric structure, moderate elasticity and good forming. To solve the problems of low-tension measurement accuracy of the existing rapier loom, the complex structure of the tension control strategy and algorithm, and the high research and development cost, this paper proposes new research from the perspective of high-precision and nonlinear processing of tension detection signals. The median filtering and limiting filtering algorithms are integrated to solve the uncertainty problem caused by the disturbance and fluctuation of the tension signal, and an excellent sample dataset is obtained. The attenuation factor and the number of learning times are introduced to design and adjust the learning rate of the back propagation neural network algorithm. In addition, the overfitting problem of the backpropagation neural network model in the current research process is solved. The experimental simulation results show that the tension detection fluctuation range of this method is 0.8 kg, and the tension detection error is within 0.1%. On the basis of the existing tension control algorithm, the tension detection accuracy is improved, which presents a new research perspective for the wide application of high-precision tension control strategies in rapier looms. It is of great importance to improve the production efficiency and fabric quality of rapier looms.

Trend prediction based on sensor data is an important topic in the thickness control system of lithium battery electrode mills. As the number of sensors increases, we can measure and store more and more data. The characteristics of nonlinearity, uncertainty, and time-variability in the lithium battery electrode thickness control system. The increase of control system complexity and data volume does not effectively improve the prediction performance. This paper proposes a physical-data fusion modeling prediction method based on a multiform coupling model and Bayesian LSTM (Bayesian Long Short-Term Memory) to achieve dynamic prediction of lithium battery electrode thickness, overcome data irrelevance and sensor noise, ensure the consistency of electrode thickness, and improve the operational efficiency of battery electrode production: Firstly, we establish the underlying physical model of the roll to further obtain the specific parameters affecting the thickness control and overcome the data irrelevance and sensor noise; secondly, we use Bayesian method to obtain the characteristics of the weight distribution of the sub-prediction network and construct the Bayesian LSTM predictor. An MLP (Multilayer Perceptron) is used as the fusion layer to fuse the results of different sub-predictions to improve the robustness of the nonlinear control system prediction model and solve the problems of slow approximation speed and ease to fall into local minimization of traditional neural networks. Finally, the advantages of the deep learning model are analyzed in terms of data feature self-extraction and model generalization generalizability. Compared with other neural network models, Bayesian LSTM has better generalizability for small sample data. The results show that the predictor can effectively model the large measurement data of the thickness control system of lithium battery electrode mills and improve the prediction performance.

In eutrophic lakes, even if external loading is controlled, internal nutrient loading delays the recovery of lake eutrophication. When the input of external pollutants is reduced, the dissolved oxygen environment at the sediment interface improves in a season without algal blooms. As an important part of lake ecosystems, macroinvertebrates are sensitive to hypoxia caused by eutrophication; however, how this change affects macroinvertebrates is still unknown. In this study, we analysed the monitoring data of northern Lake Taihu from 2007 to 2019. After 2007, the external loading of Lake Taihu was relatively stable, but eutrophication began to intensify after 2013, and the nutrients in the sediments also began to decline, which was related to the efficient use of nutrients by algal blooms. The community structure and population density of macroinvertebrates showed different responses in different stages. In particular, the density of oligochaetes and the Shannon–Wiener index showed significant differences in their response to different stages, and their sensitivity to eutrophication was significantly reduced. Under eutrophication conditions dominated by internal loading, frequent hypoxia occurs at the sediment interface only when an algal bloom erupts. When there is no bloom, the probability of sediment hypoxia is significantly reduced under the disturbance of wind. Our results indicate that the current method for evaluating lake eutrophication based on oligochaetes and the Shannon–Wiener diversity index may lose its sensitivity.

The prevalence of plasmid-mediated tigecycline resistance gene tet(X4) is presenting an increasing trend. Once tet(X4)-bearing plasmids are captured by multidrug-resistant bacteria, such as blaNDM and mcr-coharboring bacteria, it will promote bacteria to develop an ultra-broad resistance spectrum, limiting clinical treatment options. However, little is known about the destiny of such bacteria or how they will evolve in the future. Herein, we constructed a multidrug-resistant bacteria coharboring tet(X4), blaNDM-5, and mcr-1 by introducing a tet(X4)-bearing plasmid into a blaNDM-5 and mcr-1 positive E. coli strain. Subsequently, the stability of tet(X4) and the plasmid was measured after being evolved under tigecycline or antibiotic-free circumstance. Interestingly, we observed both tet(X4)-bearing plasmids in tigecycline treated strains and non-tigecycline treated strains were stable, which might be jointly affected by the increased conjugation frequency and the structural alterations of the tet(X4)-positive plasmid. However, the stability of tet(X4) gene showed different scenarios in the two types of evolved strains. The tet(X4) gene in non-tigecycline treated strains was stable whereas the tet(X4) gene was discarded rapidly in tigecycline treated strains. Accordingly, we found the expression levels of tet(X4) gene in tigecycline-treated strains were several times higher than in non-tigecycline treated strains and ancestral strains, which might in turn impose a stronger burden on the host bacteria. SNPs analysis revealed that a myriad of mutations occurred in genes involving in conjugation transfer, and the missense mutation of marR gene in chromosome of tigecycline treated strains might account for the completely different stability of tet(X4)-bearing plasmid and tet(X4) gene. Collectively, these findings shed a light on the possibility of the emergence of multidrug resistant bacteria due to the transmission of tet(X4)-bearing plasmid, and highlighted that the antibiotic residues may be critical to the development of such bacteria.

A 100-d feeding trial was conducted to evaluate the effects of replacing soybean meal (SBM) and rapeseed meal (RSM) with faba bean meal (FBM) on growth performance and muscle quality of tilapia. Abstract Smart city has obtained increasing attention from both academic and industry which has the potential to improve human living standards. A smart city consists of a great number of smart devices which are generating large amounts of data and emerging applications all the time. However, the computing capacity of smart devices are limited. Fortunately, the emergence of MEC can solve the above problem. However, the resources of edge servers in MEC are limited and the capabilities of edge servers are heterogeneous. It is important to improve the average resource utilization of all edge servers and keep load balancing of edge server cluster simultaneously. On the other hand, quite a few numbers of applications are delay-sensitive, it is necessary to ensure the security of these applications. In this paper, we consider joint optimization of mobile device and edge server in MEC-enabled smart city, improving the overall performance of the system. Technically, a new multi-objective computation offloading method is implemented to reduce time consumption, energy consumption, and keep load balancing of edge servers, as well as increase average resource utilization of edge servers while meeting the deadline constraint of delay-sensitive applications. Sufficient experiments have been conducted to prove the effectiveness and superiority of our proposed method in different scenarios.

Soybean meal is one of the major components of aquatic animal diets, whereas little information is available about the evaluation of soybean meal growth suppression mode of action. A 42-day feeding trial was performed to assess the effects of dietary soybean meal on growth performance, apparent digestibility, intestinal digestive enzyme activity, and muscle growth–related gene expression of Litopenaeus vannamei. A total of 600 shrimp were randomly distributed into 20 tanks with 30 shrimp per tank and four tanks per group. The soybean meal was added to the diets at the rate of 20% (T20), 28% (T28), 35% (T35), 42% (T42), and 50% (T50), respectively. Shrimp were fed with apparent satiation three times daily. Results indicated that the final body weight, weight gain rate, specific growth rate, feed intake, intestinesomatic index, dressed weight percentage, and the apparent digestibility coefficients of dry matter, crude protein, crude lipid, and ash were linearly decreased (p < 0.05), but feed coefficient was linearly increased (p < 0.05) as dietary soybean meal increased from 20% to 50%. The intestinal trypsin and amylase activities were decreased (p < 0.05) as dietary soybean meal increased from 20% to 50%, and reached significance at the level of 35%, 42%, and 50%. Shrimp fed with T20 had higher (p < 0.05) intestinal lipase activity than those fed with other diets. The mRNA relative expression of growth hormone, myogenic regulatory factor 5, and target of rapamycin was downregulated (p < 0.05) as dietary soybean meal increased from 20% to 50%. To conclude, dietary soybean meal exceeded 28% significantly inhibited growth performance of L. vannamei, mainly due to the negative impact of soybean meal on digestion and feed utilization and also the inhibition on the muscle growth and related gene expressions.

The experimental trial was conducted to assess the effects of dietary pyridoxine on growth performance, physiological metabolics, intestinal enzymatic activities and antioxidant properties of Pelteobagrus fulvidraco. Six isonitrogenous and isolipidic diets (420 g/kg protein and 70 g/kg lipid) were formulated to contain 3.69 (control), 5.45, 7.31,11.17, 19.43 and 33.94 mg/kg pyridoxine. ABSTRACT In clinical practice, carbapenems and tigecycline are considered significant options for treating infections caused by multidrug-resistant Klebsiella spp. The continual evolution of resistance mechanisms to carbapenems and tigecycline is shattering the present condition. Meanwhile, convergence of the two resistance mechanisms in a single strain has been reported repeatedly, posing a significant threat to public health and safety. In this study, two carbapenem- and tigecycline-resistant Klebsiella species were obtained from patients and investigated using antimicrobial susceptibility testing, conjugation assay, whole-genome sequencing, and bioinformatics analysis. In Klebsiella variicola FK2020ZBJ35, an untransferable multidrug IncFIB(Mar)/IncHI1B-like plasmid carrying tmexCD2-toprJ2, blaIMP-4, and blaNDM-1 was discovered, as was a similar plasmid carrying tmexCD1-toprJ1 and blaIMP-4 in Klebsiella quasipneumoniae 2019SCSN059. Genetic context analysis found that two distinct tmexCD-toprJ variants were detected in comparable mobile units with genetic array int-int-hp-hp-tnfxB-tmexCD-toprJ and integrated into separate genetic locations. blaIMP-4 and blaNDM-1 were carried by an integron In1377 and a truncated Tn3000, respectively. These findings revealed that the carbapenem and tigecycline resistance genes carried by the two strains were located on mobile elements and might potentially transmit horizontally to additional strains. Furthermore, our findings showed that IncFIB(Mar)/IncHI1B-like plasmids represent a significant reservoir of essential resistance genes that warrants continued monitoring. IMPORTANCE Tigecycline is an essential antibiotic that is used to treat infections caused by carbapenem-resistant Klebsiella pneumoniae (CRKP). The emergence of high-level tigecycline-resistant CRKP poses a serious hazard to human health. This work screened two tigecycline-resistant CRKP strains from clinical patients and found a type of plasmid that encoded carbapenemase and TmexCD-ToprJ in Klebsiella. Importantly, one plasmid cocarried tmexCD-toprJ, blaNDM-1, and blaIMP-4, hinting that this plasmid could be a critical vector for superbug development. Furthermore, we discovered that the carbapenem and tigecycline resistance genes are located in mobile units by genetic structure analysis. Our research tracks the formation of clinically super-resistant Gram-negative bacteria.

A 63-day feeding trial with 640 juvenile L. maculatus was conducted to assess the effects of dietary CT on serum metabolites, antioxidant and immune response, liver histomorphology and glycometabolism enzyme activities of fish. Four diets were formulated to contain 0 (T1), 1 (T2), 2 (T3), and 2 g/kg of CT plus 4 g/kg of polyethylene glycol (PEG) (T4). PEG specifically binds with CT to neutralize CT activity. Fish were randomly distributed into 16 tanks (4 tanks per group and 40 fish per tank) and were fed to apparent satiation twice daily. Results indicated that fish fed T2 and T3 had lower (P0.05) between T1 and T4. In summary, dietary CT up to 2 g/kg reduced serum lipid and glucose concentrations, enhanced liver antioxidant and immune response, improved glucose utilization but of L. maculatus. CT induced liver injury of L. maculatus which provided a caution for its application in aquaculture.

ABSTRACT Emergence of pathogens harboring multiple resistance genes incurs great concerns. Cooccurrence of mobile resistance genes conferring resistance to tigecycline, colistin, and carbapenems in Escherichia coli has not been investigated. This study aimed to characterize three E. coli isolates coharboring tet(X4), mcr-1, and blaNDM-5. Isolates coharboring tet(X4), mcr-1, and blaNDM-5 were identified and characterized by PCR, Sanger sequencing, antimicrobial susceptibility testing, conjugation assays, Illumina sequencing, nanopore sequencing, and bioinformatic analysis. Three E. coli isolates carrying tet(X4), mcr-1, and blaNDM-5 were identified from pigeons in China. They were resistant to almost all antimicrobials except enrofloxacin. tet(X4) and blaNDM-5 could be conjugated into E. coli C600, but mcr-1 was nontransferable in three isolates. Three isolates belonged to sequence type 6775 (ST6775), and clonal dissemination of isolates carrying tet(X4), mcr-1, and blaNDM-5 existed in the pigeon farm. Genetic analysis revealed that mcr-1 mediated by the Tn6330 was located on the chromosome, tet(X4) was located on the IncFII plasmid, and blaNDM-5 was located on the IncX3 plasmid. We first characterized the E. coli isolates carrying tet(X4), mcr-1, and blaNDM-5 simultaneously. Relevant measures should be taken to decrease the prevalence of pathogens carrying tet(X4), mcr-1, and blaNDM-5. IMPORTANCE Tigecycline and colistin are regarded as vital antimicrobials to treat multidrug-resistant (MDR) bacterial infections, such as that caused by carbapenemase-producing Enterobacteriaceae (CPE). Cooccurrence of mobile resistance genes conferring resistance to last-resort antimicrobials in E. coli remains unknown. Here, we characterized E. coli strains coharboring tet(X4), mcr-1, and blaNDM-5 phenotypically and genetically. Resistance genes tet(X4), mcr-1, and blaNDM-5 were located on transposons or plasmids that were mobile genetic elements related to the capture, accumulation, and dissemination of such important resistance genes. The emergence of E. coli isolates carrying tet(X4), mcr-1, and blaNDM-5 highlights the importance of monitoring the coexistence of novel mobile resistance genes in different settings with a One Health approach. Risk of transmission of such MDR pathogens from animals to humans should be evaluated comprehensively.

Mobile edge computing is a new paradigm which provides cloud computing capabilities at the edge of pervasive radio access networks in close proximity to users. The problem of edge server selection in mobile edge environment in terms of user’s overhead is investigated in this paper. Due to the limited resources of edge server, we firstly study the task completion probability of edge servers. Secondly, we formally model the problem of edge server selection in terms of time latency and energy consumption. More especially, the computation overhead method for completing the task in cases of both service migration and non-migration is investigated. Then, a new optimized edge server selection algorithm, called combined Genetic algorithm and simulated Annealing algorithm for edge Server Selection (GASS) is designed. Finally, a series of experiments on a real-word data-trace are conducted to evaluate the performance of GASS. The results show that GASS can effectively minimize the overhead of the user and outperform traditional heuristic algorithms.

Snail (Sna) plays a pivotal role in epithelia-mesenchymal transition and cancer metastasis, yet its functions in normal tissue development remain elusive. Here, using Drosophila as a model organism, we identified Sna as an essential regulator of Hippo signalling-mediated cell proliferation and tissue growth. First, Sna is necessary and sufficient for impaired Hippo signalling-induced cell proliferation and tissue overgrowth. Second, Sna is necessary and sufficient for the expression of Hippo pathway target genes. Third, genetic epistasis data indicate Sna acts downstream of Yki in the Hippo signalling. Finally, Sna is physiologically required for tissue growth in normal development. Mechanistically, Yki activates the transcription of sna, whose protein product binds to Scalloped (Sd) and promotes Sd-dependent cell proliferation. Thus, this study uncovered a previously unknown physiological function of Sna in normal tissue development and revealed the underlying mechanism by which Sna modulates Hippo signalling-mediated cell proliferation and tissue growth.

Advancement of novel sequencing technologies facilitates modern life science and medicine unprecedentedly. Exploring complete genome sequences of bacteria by long-read sequencing technology is significant for microbial genomics research. However, third-generation long-read sequencing technologies are available with limited choices, which generate technological barrier to scientific research. Recently, a novel QitanTech nanopore long-read sequencing technology has emerged in China, but the potential application and performance were unexplored. Herein, we comprehensively evaluated the feasibility of the emerging sequencing technology in assembling complete genomes of MDR pathogens. The results showed that 500 Mbp QitanTech nanopore sequencing data could be generated within 8 h in one flow cell with the standard library preparation method. The mean read length, longest read length, and mean read-level accuracy of QitanTech sequencing data were 6,041 bp, 57,037 bp, and 81.50% (LAST)/81.40% (Minimap2), respectively. Two routine assembly strategies including long-read assembly and hybrid assembly enable the achievement of complete bacterial genomes. The accuracy of assembled draft bacterial genomes with QitanTech long-read data could be improved up to 99.9% dramatically by polishing using accurate short-read data. Furthermore, the assembled bacterial genomes cover accurate structures of complex resistance plasmids harboring critical resistance genes such as tet(X), tmexCD-toprJ, and blaVIM–2, even the complex fusion MDR plasmid generated from homologous recombination. In conclusion, QitanTech nanopore sequencing, as a nanopore long-read sequencing technology launched in China, could be a good option for investigation of complex bacterial genomes. More potential applications based on this novel platform warrant investigations.

Low-salinity tolerance and growth are crucial economic traits of shrimp in areas with low-salinity water. In the current study, an F1 full-sib family of Pacific white shrimp (Litopenaeus vannamei) was obtained by the “semidirectional cross” method, and 2 parents and 146 progenies were genotyped using double-digest restriction site-associated DNA sequencing technology (ddRAD-Seq). A total of 3335 high-quality markers were identified and used to construct a genetic linkage map. This genetic map comprised 48 linkage groups (LGs), and the total map length spanned 3954.462 centimorgan (cM) with an average marker interval of 1.186 cM. Three quantitative trait loci (QTLs) for low-salinity tolerance and 3 QTLs for growth (body weight) were detected under the condition of restricted logarithmic odds (LOD) score ≥ 2.50. With the assistance of the genome of L. vannamei and relevant annotations, 3 potential growth-related genes (sob, TJP1 and ZBTB24) and 3 low-salinity response candidate genes (Rnf168, CNOT2 and RPB3) were identified from the genomic scaffolds. Moreover, the qPCR results showed that the expression levels of the 3 candidate genes were significantly affected by low-salinity stress, indicating that they possibly play an important role in low-salinity stress. Here, we present the first report of mapping of low-salinity tolerance QTLs in the genetic mapping of L. vannamei, which will be beneficial for the selection of shrimp breeding markers and might help promote the study of the L. vannamei genome.

Dietary inclusion of aflatoxin B1 (AFB1) has been reported to induce intestinal damage and dysregulate intestinal microbiota of shrimp, but limited information is available in fish. A 56-day feeding trial was conducted to assess the effects of dietary AFB1 on growth performance, serum metabolites, intestinal tight junction structure and bacterial microbiomes of Lateolabrax maculatus. Four diets were formulated to contain 0 (G0), 0.1 (G0.1), 0.5 (G0.5) and 1.0 (G1.0) mg/kg of AFB1, and each diet was randomly assigned to three groups of 40 fish per tank. Fish were fed to apparent satiation twice daily. Results indicated that increased dietary AFB1 concentration from 0 to 1.0 mg/kg decreased (P 0.05) alpha diversity indexes of ace, chao, shannon and simpson in the intestinal microbiota of fish. In conclusion, inclusion of AFB1 up to 1.0 mg/kg in diets decreased growth performance, induced liver and intestinal injury, and altered intestinal microbiota composition without impacting the bacterial diversity of Lateolabrax maculatus.

In order to further solve the problems in promoting the classification of media content in colleges and universities, the effective analysis and understanding of multimedia data content can be better realized based on the characteristics of multimedia data in colleges and universities, combining with the characteristics of rich information, large differences in performance, and large amount of large-scale data. This essay mainly introduces the technology of university media content detection and classification based on information fusion algorithm and focuses on the application of university multimedia content detection, analysis, and understanding, to explore the image discrimination auxiliary attribute feature learning and content association prediction and classification. A benchmark model for media content detection and classification is constructed. Through the model test, it is found that the F1 value of the model is more than 70%, the check rate is more than 80%, and the recall rate is more than 50%. On this basis, a content detection system based on campus network is constructed.
