DNA guanine (G)-quadruplexes (G4s) are unique secondary structures formed by two or more stacked G-tetrads in G-rich DNA sequences. These structures have been found to play a crucial role in highly transcribed genes, especially in cancer-related oncogenes, making them attractive targets for cancer therapeutics. Significantly, targeting oncogene promoter G4 structures has emerged as a promising strategy to address the challenge of undruggable and drug-resistant proteins, such as MYC, BCL2, KRAS, and EGFR. Natural products have long been an important source of drug discovery, particularly in the fields of cancer and infectious diseases. Noteworthy progress has recently been made in the discovery of naturally occurring DNA G4-targeting drugs. Numerous DNA G4s, such as MYC-G4, BCL2-G4, KRAS-G4, PDGFR-β-G4, VEGF-G4, and telomeric-G4, have been identified as potential targets of natural products, including berberine, telomestatin, quindoline, sanguinarine, isaindigotone, and many others. Herein, we summarize and evaluate recent advancements in natural and nature-derived DNA G4 binders, focusing on understanding the structural recognition of DNA G4s by small molecules derived from nature. We also discuss the challenges and opportunities associated with developing drugs that target DNA G4s.

Abstract This study assesses the midsummer precipitation prediction over eastern China by the dynamic downscaling method. Based on the Climate Forecast System version 2 of the National Centers for Environmental Prediction and the Weather Research and Forecasting Model, the prediction performance of global and regional models on the July precipitation over eastern China is further analyzed by hindcast experiments from 1982 to 2010 and prediction experiments from 2011 to 2021. The results suggest that the regional model forced by the global model can noticeably improve the prediction skill for precipitation in eastern China, especially in the region from the South of North China to the Yangtze River Basin, referred as the northern China in this paper. In addition, we perform a diagnostic analysis of the reason for the improvement of the model prediction skill. The results indicate that the high resolution of the regional model and the refinement of physical process parameterizations contribute to improving the simulation ability for the East Asian atmospheric circulation pattern, heat flux, especially for the meridional teleconnection pattern in East Asia and the sensible heat flux in the northern China, thus further improving precipitation prediction.

Abstract In recent years, with the development of ship electric propulsion system, High Temperature Superconducting (HTS) motors have gained attention as a promising potential resolution due to their high-power density and the progression of cryogenic refrigeration technology. We present an electromagnetic characteristics numerical analysis of 40 MW, 120 rpm, HTS synchronous motor which is a semi-superconducting motor: in fact, it has a superconducting rotor composed of YBCO material and a conventional stator. Combining electric resistivity variation of rotor copper shielding sleeve in low temperature environment and YBCO coil critical current, the electromagnetic behavior of the 40 MW HTS synchronous motor is analyzed with non-ferromagnetic stator support structure. To assess the effects of varying stator winding currents and thickness on eddy current losses of the copper shielding sleeve, calculations were done, and a preliminary discussion was initiated. This study has certain reference significance for electromagnetic design and optimization of multi-ten MW HTS motors, especially used in ship electric propulsion system.

Abstract Gastric cancer (GC) remains the third leading cause of cancer-related mortality in the world, and ninety-five percent of GC are stomach adenocarcinomas (STAD). The active ingredients of Croci Stigma, such as Isorhamnetin, Crocin, Crocetin and Kaempferol, all have antitumor activity. However, their chemical and pharmacological profiles remain to be elusive. In this study, network pharmacology was used to characterize the action mechanism of Croci Stigma. All compounds were obtained from the traditional Chinese medicine systems pharmacology (TCMSP) database, and active ingredients were selected by their oral bioavailability and drug-likeness index. The targets of Croci Stigma active ingredients were obtained from the traditional Chinese medicine integrated database (TCMID), whereas the related genes of STAD were obtained from DisGeNET platform. Cytoscape was used to undertake visual analyses of the Drug Ingredients–Gene Symbols–Disease (I–G–D) network, and 2 core genes including MAPK14, ERBB3 were obtained, which are the predicted targets of isorhamnetin (IH) and quercetin, respectively. Data analysis from TCGA platform showed that MAPK14 and ERBB3 all upregulated in STAD patients, but only the effect of MAPK14 expression on STAD patients’ survival was significant. Molecular docking showed that IH might affect the function of MAPK14 protein, and then the underlying action mechanisms of IH on STAD were experimentally validated using human gastric cancer cell line, HGC-27 cells. The results showed that IH can inhibit cell proliferation, migration, clonal formation, and arrest cell cycle, but promote the apoptosis of HGC-27 cells. qRT-PCR data demonstrated that IH downregulated the MAPK14 mRNA expression and EMT related genes. WB results showed that IH regulates MAPK/mTOR signaling pathway. These findings suggest that IH has the therapeutic potential for the treatment of STAD.

The G-quadruplex formed in KRAS oncogene promoter (KRAS-G4) is a transcriptional modulator and amenable to small molecule targeting. Herein, the authors report the NMR solution structures of a bulge-containing KRAS-G4 that bound to two small molecules. The study provides molecular details of ligand interactions with KRAS-G4 and contributes insight into the design of specific KRAS-G4-interactive drugs.

The thermoelectric transport properties of four kinds of DNA bases sandwiched between two zigzag-edge graphene nanoribbon electrodes are investigated by first-principles calculation. The modulation of quantum interference, molecular length, and molecular torsion on thermoelectric transport properties are studied. It is found that the thermopower and efficiency can be obviously enhanced in the guanine and electrode coupling geometry that manifests a quantum interference effect. A decrease of thermopower with a number of DNA bases has been found, while the efficiencies have a different trend with the increase of the number of different molecules. In addition, the thermopower and efficiency also can be considerably enhanced by the twisting angle between the molecule and graphene electrodes.

Summary: Incorporating graphite/graphene into a Mg alloy matrix is a promising approach for developing lightweight heat dissipation materials. However, carbon material is inherently incompatible with Mg because of their distinctly different surface characteristics, resulting in the challenge of composite fabricating and interface controlling. Herein, a new strategy of in situ interfacial modification was proposed to achieve excellent thermal conductivity and mechanical properties in graphite/Mg composites. A super-nano CaCO3 interfacial layer was reported in this paper. The detailed interfacial structure, reaction thermodynamics and kinetics, and interface strengthening mechanisms were analyzed and discussed. Several preferential epitaxial relationships of the Mg/CaCO3 interface were revealed, which are conducive to minimize the interfacial energy, stabilize and strengthen the interface. Moreover, strong ionic bond of graphite/CaCO3 interface was demonstrated. The strong chemical interface bonding of graphite-Mg via in situ interface modification facilitates both the interfacial cohesion and interfacial thermal conduction, which endows the graphite/Mg composites with superior strength–thermal conductivity synergy.
