14. Matrix-Assisted laser desorption ionization mass spectrometry imaging by freeze-spot deposition of the matrix

Abstract

Imaging mass spectrometry has emerged as a powerful metabolite measurement approach to capture the spatial dimension of metabolite distribution in a biological sample. In matrix-assisted laser desorption ionization–mass spectrometry imaging (MALDI-MSI), deposition of the chemical-matrix onto the sample serves to simultaneously extract biomolecules to the sample surface and concurrently render the sample amenable to MALDI. However, matrix application may mobilize sample metabolites and will dictate the efficiency of matrix crystallization, together limiting the lateral resolution which may be optimally achieved by MSI. Here, we describe a matrix application technique, herein referred to as the “freeze-spot” method, conceived as a low-cost preparative approach requiring minimal amounts of chemical matrix while maintaining the spatial dimension of sample metabolites for MALDI-MSI. Matrix deposition was achieved by pipette spot application of the matrix-solubilized within a solvent solution with a freezing point above that of a chilled sample stage to which the sample section is mounted. The matrix solution freezes on contact with the sample and the solvent is removed by sublimation, leaving a fine crystalline matrix on the sample surface. Freeze-spotting is quick to perform, found particularly useful for MALDI-MSI of small sample sections, and well suited to efficient and cost-effective method development pipelines, while capable of maintaining the lateral resolution required by MSI.

19. The blood supply of the midbrain in man and monkey

Abstract

A review of the literature indicated that the blood supply of the midbrain had been studied but little in detail. An investigation was therefore carried out on human and monkey material. Of the techniques tried, coloured gelatin, radio-opaque, and especially Indian ink injections proved most informative. 1. The generally accepted sources of blood supply were confirmed. Accepted data on the origin, variations, number and size of vessels have been presented in schematic form, restricting description of the various named vessels involved to relevant detail. 2. Superficial vascular anastomoses on the midbrain have been confirmed. Midline surface anastomoses over the tectum have been shown to exist. The extent of midbrain supply by different arteries as observed by surface inspection and in microscopic sections has been described and illustrated. 4. Differences from and agreements with previous descriptions, and the originality of certain observations,have been pointed out. 5.An absence of arteriovenous anastomoses in the midbrain pia mater has been confirmed. 6. Entirely original observations on the intramesencephalic veins have been presented. Collecting veins of the midbrain have been described. Incidence of variations affecting the basal and lateral mesencephalic veins have been presented in schematic form. 8. Anastomoses of the dorsal mesencephalic and mesencephalic-cerebellar veins have been described. Through these anastomoses the possibility of a short circuiting of the internal or great cerebral veins to the superior petrosal sinus has been suggested. 9. Capillary density studies on representative tracts and nuclei of the midbrain (and some neighbouring structures) were carried out by a dot graticule method. Data on their relative vascularities have been tabulated. 10. The results have been discussed and conclusions drawn.

Abstract

This article contains highlights and a selection of the scientific advances from the European Respiratory Society's Clinical Assembly (Assembly 1 and its six respective groups) that were presented at the 2017 European Respiratory Society International Congress in Milan, Italy. The most relevant topics from each of the groups will be discussed, covering a wide range of areas including clinical problems, rehabilitation and chronic care, thoracic imaging, interventional pulmonology, diffuse and parenchymal lung diseases, and general practice and primary care. In this comprehensive review, the newest research and actual data as well as award-winning abstracts and highlight sessions will be discussed.

29. A market-based programme to improve housing in the mountains of northern Pakistan: Addressing seismic vulnerability

Abstract

The Building and Construction Improvement Programme (BACIP) has been working with the high mountain communities of the Gilgit-Baltistan region of Pakistan since 1997. Alongside its contribution to the general built environment and housing improvement of the area, the programme is engaged in the development and promotion of solutions for making the buildings seismically resistant. Gilgit-Baltistan falls in a high seismic zone and the earthquake of 2005 caused the death of nearly 90,000 people in the neighbouring state of Azad Kashmir and the Khyber Pakhtunkhwa province of Pakistan, which share long borders with Gilgit-Baltistan. BACIP believes that investment in making communities safer will minimize the chance of loss of life and assets and reduce the cost of reconstruction. BACIP works with local communities in a participatory manner to improve the local housing by improving safety and comfort without changing the local culture and way of living. For the sustainability of its approach, it has made efforts to make its solutions part of the local market so that entrepreneurs and artisans are available to manufacture, sell or construct these solutions. A number of profitable enterprises have been established. Alongside handson training and demonstration, BACIP uses media such as radio for the promotion of its solutions and awareness of communities. In December 2013 with the support of the Building and Social Housing Foundation (BSHF), BACIP revisited a number of houses that were constructed using seismic-resistant technologies and it was found that the solutions applied to these houses had greatly contributed to the safety and comfort of the users. 100 per cent of the houses were in use and were occupied by the original owners.

Abstract

Although federal support for medical education comes from several sources, only 1 targets generalist education--Title VII of the Public Health Service Act. With governmental streamlining and downsizing, the federal investment in medical education should be evaluated. We tested the relationship between 2 Title VII authorities and presence of a medical school generalist training infrastructure, and the relationship between this infrastructure and generalist production. Based on our definitions for receipt of Title VII support, generalist infrastructure, and generalist production, we found that, for private schools, sustained receipt of Title VII funds directed for undergraduate medical education is positively associated with presence of family medicine departments, which is positively associated with higher rates of generalist production. Establishment and maintenance of family medicine departments in private schools and their generalist production are positively associated with Title VII support. Title VII support in public schools, the major generalist producers, has less of a unique measurable impact on generalist production.
