Abstract Investigating microorganisms in metal-enriched environments holds the potential to revolutionize the sustainable recovery of critical metals such as lanthanides (Ln3+). We observe Hyphomicrobium spp. as part of a Fe2+/Mn2+-oxidizing consortia native to the ferruginous bottom waters of a Ln3+-enriched lake in Czechia. Notably, one species shows similarities to recently discovered bacteria expressing proteins with picomolar Ln3+ affinity. This finding was substantiated by developing an in-silico ionic competition model and recombinant expression of a homolog protein (Hm-LanM) from Hyphomicrobium methylovorum. Biochemical assays validate Hm-LanM preference for lighter Ln3+ ions (from lanthanum to gadolinium). This is comparable to established prototypes. Bioinformatics analyses further uncover additional H. methylovorum metabolic biomolecules in genomic proximity to Hm-LanM analogously dependent on Ln3+, including an outer membrane receptor that binds Ln3+-chelating siderophores. These combined observations underscore the remarkable strategy of Hyphomicrobium spp. for thriving in relatively Ln3+ enriched zones of metal-polluted environments.

Abstract The Neoproterozoic oxygenation event is a milestone in Earth’s history, yet the redox structure and elemental cycling of the Neoproterozoic ocean remain debated. Here, we investigated iron speciation, molybdenum, and nitrogen isotopes in black shales and manganese carbonates from the upper Ediacaran Doushantuo Formation, South China, to examine the links between redox state, manganese mineralization, and molybdenum cycling. In both lithologies, high pyritic iron ratios (> 0.8) indicate a localized euxinic zone, while estimated seawater molybdenum (1.9‰) and sediment nitrogen isotopic compositions (4.19 ± 1.96‰) resemble modern values, suggesting oxygenated surface waters over euxinic depths. Negative molybdenum isotope in manganese(II) carbonates points to manganese(IV) oxide reduction, acting as a molybdenum shuttle from oxygenated surface to euxinic deep waters. Periodic euxinic contractions drove manganese(II) oxidation and mineralization, shaping molybdenum cycling. This study highlights essential manganese(II) oxidation for manganese carbonate mineralization and offers new insights into molybdenum geochemistry and ancient ocean oxygenation events.

Study region: A 706 km2 watershed located in a heavily industrialized region near Fox Creek, Alberta, Canada. Study focus: Petroleum exploration has increasingly disturbed boreal ecosystems in North America. Seismic lines are one of the major footprints of the petroleum industry: they form cleared linear corridors in forests and their regeneration is generally poor. For this study, field measurements of soil, vegetation, water and snow were conducted on 5 paired (seismic line and adjacent undisturbed area) sites located in lowland and upland ecosites, and a 1-D physically based hydrologic model, the Simultaneous Heat and Water Model (SHAW), was used to investigate the impacts of seismic lines on the water budget over a year. New hydrological insights for the region: Field measurements indicate that parameters, including soil water content and temperature, vary according to a complex combination of factors and are highly site-specific between seismic lines and undisturbed areas. Hydrological modeling of the two ecosites (lowland and upland) showed up to a 33 % reduction in evapotranspiration and a decrease in percolation (leading to none) below the root zone on seismic lines. Considering that there are 2300 km of seismic lines in the study area and that 36 % of the forest has been cleared for industrial activities, these results suggest that the water budget is impacted by anthropogenic lineaments. Climate change will likely intensify these impacts.

Many species exhibit social living which offers ecological advantages such as increased foraging opportunities, more efficient locomotion and reduced predation risk. Additionally, exposure to multiple individuals of the same species can decrease an individual’s stress and metabolic demand, termed social buffering. If disruption to an animal’s social structure occurs and prevents social buffering, an elevated metabolic rate and thus ventilation frequency and gill permeability are likely. A potential consequence of this physiological response could be the increased accumulation of toxicants. The objective of this study was to investigate whether inducing social stress in marine threespine stickleback (Gasterosteus aculeatus) through social isolation during a sublethal water-borne copper (Cu) exposure would affect Cu uptake and whether that would translate to differences in behaviour and biochemical functioning. We hypothesized that isolating threespine stickleback during a Cu exposure would increase Cu uptake and sublethal toxicity compared to a grouped exposure. Wild-caught fish were exposed to control, low Cu or high Cu conditions (0 – 150 µg/L of Cu, nominally), either in isolation or groups of six for 96 h. Isolated stickleback travelled three times less distance, took six times longer to consume food and exhibited moderately increased gill sodium-potassium ATPase activity than group exposed fish, with no effect of Cu. Isolated stickleback also demonstrated significantly higher Cu levels in their gill and liver tissue compared to the group exposed fish. However, this Cu distribution was also present within the control fish, which had not been exposed to Cu, suggesting that the social context affects endogenous Cu distribution under stressful conditions. Our results illustrate the differences in physiology and behaviour that can arise when social contexts are manipulated and stress the importance of considering sociality when conducting toxicity tests with social organisms.

ABSTRACT Marine biofilms were newly revealed as a giant microbial diversity pool for global oceans. However, the cyanobacterial diversity in marine biofilms within the upper seawater column and its ecological and evolutionary implications remains undetermined. Here, we reconstructed a full picture of modern marine cyanobacteria habitats by re-analyzing 9.3 terabyte metagenomic data sets and 2,648 metagenome-assembled genomes (MAGs). The abundances of cyanobacteria lineages exclusively detected in marine biofilms were up to ninefold higher than those in seawater at similar sample size. Analyses revealed that cyanobacteria in marine biofilms are specialists with strong geographical and environmental constraints on their genome and functional adaption, which is in stark contrast to the generalistic features of seawater-derived cyanobacteria. Molecular dating suggests that the important diversifications in biofilm-forming cyanobacteria appear to coincide with the Great Oxidation Event (GOE), “boring billion” middle Proterozoic, and the Neoproterozoic Oxidation Event (NOE). These new insights suggest that marine biofilms are large and important cyanobacterial factories for the global oceans.IMPORTANCECyanobacteria, highly diverse microbial organisms, play a crucial role in Earth’s oxygenation and biogeochemical cycling. However, their connection to these processes remains unclear, partly due to incomplete surveys of oceanic niches. Our study uncovered significant cyanobacterial diversity in marine biofilms, showing distinct niche differentiation compared to seawater counterparts. These patterns reflect three key stages of marine cyanobacterial diversification, coinciding with major geological events in the Earth's history.

Abstract Temporal community stability, here defined as temporal mean divided by temporal SD, plays an important role in predicting certain ecosystem services. However, temporal stability can change with invasion, with greater abundances of invasive species potentially having greater impacts on native community stability. The exact consequences of invasion for temporal stability are unclear and, in part, depend on the particular metric of stability measured. In rangeland ecosystems, predicable forage is important for livestock production but can be threatened by invasion. Therefore, using an observational field study conducted over three years in Wyoming, we assessed which metrics of plant community stability were altered by invasion and whether those effects were mediated by two environmental variables (light and soil moisture). Bromus arvensis and Bromus tectorum are two invasive annual weeds found across US rangelands, including the northern mixed‐grass prairies of Wyoming. We established plots along natural invasion blocks of B. arvensis and B. tectorum abundance and collected plant species composition data over three growing seasons. We tested associations between seven different metrics of plant community stability and invasion by B. arvensis and B. tectorum. We found that species turnover increases with invasion by both species, while stability of forb (both brome species), C4 grass (B. arvensis only), and C3 grass (B. tectorum only) cover decreases with invasion. All metrics of stability associated with invasion supported the hypothesis of a destabilizing effect of invasion on the native plant community. Further, we found that light and soil moisture did mediate some associations between stability and invasion. Overall, our results align with previous work suggesting that invasive annual bromes can lead to decreased native plant stability, which has important implications for forage production and, thus, food security.

Abstract Marine sedimentary rocks deposited across the Neoproterozoic Cryogenian Snowball interval, ~720-635 million years ago, suggest that post-Snowball fertilization of shallow continental margin seawater with phosphorus accelerated marine primary productivity, ocean-atmosphere oxygenation, and ultimately the rise of animals. However, the mechanisms that sourced and delivered bioavailable phosphate from land to the ocean are not fully understood. Here we demonstrate a causal relationship between clay mineral production by the melting Sturtian Snowball ice sheets and a short-lived increase in seawater phosphate bioavailability by at least 20-fold and oxygenation of an immediate post-Sturtian Snowball ocean margin. Bulk primary sediment inputs and inferred dissolved seawater phosphate dynamics point to a relatively low marine phosphate inventory that limited marine primary productivity and seawater oxygenation before the Sturtian glaciation, and again in the later stages of the succeeding interglacial greenhouse interval.

Recent research has hypothesized that hydrogen peroxide (H2O2) may have emerged from abiotic geochemical processes during the Archean eon (4.0–2.5 Ga), stimulating the evolution of an enzymatic antioxidant system in early life. This eventually led to the evolution of cyanobacteria, and in turn, the accumulation of oxygen on Earth. In the latest issue of Redox Biology, Koppenol and Sies (vol. 29, no. 103012, 2024) argued against this hypothesis and suggested instead that early organisms would not have been exposed to H2O2 due to its short half-life in the ferruginous oceans of the Archean. We find these arguments to be factually incomplete because they do not consider that freshwater or some coastal marine environments during the Archean could indeed have led to H2O2 generation and accumulation. In these environments, abiotic oxidants could have interacted with early life, thus steering its evolutionary course.

Abstract Microbial anaerobic oxidation of methane coupled to the reduction of Mn(IV)-oxides, typically MnO2, derived from continental weathering and riverine transport has been proposed as a globally important sink of methane. However, the potential role of hydrothermal Mn(IV) oxide-rich sediments as a methane sink, and the mechanisms of underpinning methane oxidation at high-temperature hydrothermal fields remain poorly understood. Here, we report the occurrence of almost pure rhodochrosite with extremely negative δ13CPDB values (as low as –76.4‰) in direct association with hausmannite formed through hydrothermal activity in the Late Triassic Heqing Mn deposit in Southern China. Based on detailed petrography and geochemistry, this rhodochrosite is interpreted as the result of the thermochemical oxidation of methane by hausmannite during early diagenesis. Given high hydrothermal Mn2+ and CH4 release coupled to low sulfate concentration observed in the Archean oceans, we propose that hydrothermal Mn(IV) oxides may have been effective methane sinks in Earth’s ancient oceans.

A new aim for grassland management is to increase soil organic carbon (SOC) and to offset CO2 emissions by companies. This practice of carbon ranching may be informed by grazing-induced shifts in plant biomass and diversity which may foretell changes in SOC. Unfortunately, little is known about how grazing-induced shifts in plant properties correspond with shifts in SOC stocks. To help fill this gap, we used data from a field experiment to test whether above-ground plant properties (i.e. biomass, species richness) act as leading indicators of grazing-induced SOC accrual in the Northern Great Plains. The 5-yr bovine grazing experiment had a randomized complete block design and pre-treatment data. Moderate summer grazing (control) is widely used in the Northern Great Plains, and treatments that may alter grassland vegetation and SOC included: severe summer grazing, moderate fall grazing, and severe fall grazing. Severe fall and summer grazing increased SOC but had no effect on plant species richness and biomass relative to controls. Fall moderate grazing increased above-ground plant biomass but had no effect on SOC relative to controls. Changes to grazing practices can affect SOC without measurably affecting plant properties and can affect plant properties without measurably affecting SOC. While two drivers of SOC are plant carbon inputs and microbial respiration, our study indicates that grazing-induced change in above-ground vegetation is not predictive of change in SOC.

Abstract One of the paradoxes of the Archean rock record is how evidence for oxidative continental weathering is plausible when the atmosphere lacked oxygen prior to the Great Oxidation Event ca. 2.5 billion years ago. We detected reactive oxygen species (ROS) production at various silicate-water interfaces in a simulated Archean atmosphere and calculated homolysis rates of silicon–oxygen bonds in these minerals. We find that ROS can be produced while silicates were eroded and transported by streams to depositional basins. The magnitude of ROS production increases from mafic- to felsic-silicates. During early continent formation, e.g., the Kaapvaal and Pilbara basins in the Paleoarchean, the emplacement of granitoids and their subsequent exposure to physical weathering might have annually provided at least 1.73 × 108 mol O2 before the biological O2 production. Importantly, this process could have oxidatively mobilized redox-sensitive elements to seawater, increasing the availability of bioessential nutrient elements for the Archean biosphere.

The authors discovered that proinflammatory senescent myeloid cells may recruit peripheral immune cells in the aged mouse brain. Their findings implicate senescent cell clearance as a strategy to counter aged brain inflammation and cognitive decline.

Summary: Ecological engineering of soil formation in tailings is an emerging technology toward sustainable rehabilitation of iron (Fe) ore tailings landscapes worldwide, which requires the formation of well-organized and stable soil aggregates in finely textured tailings. Here, we demonstrate an approach using microbial and rhizosphere processes to progressively drive aggregate formation and development in Fe ore tailings. The aggregates were initially formed through the agglomeration of mineral particles by organic cements derived from microbial decomposition of exogenous organic matter. The aggregate stability was consolidated by colloidal nanosized Fe(III)-Si minerals formed during Fe-bearing primary mineral weathering driven by rhizosphere biogeochemical processes of pioneer plants. From these findings, we proposed a conceptual model for progressive aggregate structure development in the tailings with Fe(III)-Si rich cements as core nuclei. This renewable resource dependent eco-engineering approach opens a sustainable pathway to achieve resilient tailings rehabilitation without resorting to excavating natural soil resources.

Abstract Background The medical case vignette has long been used in medical student education and frequently includes demographic variables such as race, ethnicity and gender. However, inclusion of demographic variables without context may reinforce assumptions and biases. Yet, the absence of race, sexual orientation, and social determinants of health may reinforce a hidden curriculum that reflects cultural blindness. This replication study compared proportions of race, ethnicity, and gender with University of Minnesota (UMN) findings. This study sought to determine if there has been progress in the representation of demographic characteristics in case vignettes. Methods: University of North Carolina (UNC) case vignettes from 2015–2016 were analyzed and compared to UMN case vignettes from 1996–1998. Data included mentions of race, ethnicity, gender and social determinants of health. Results: In the 278 UNC vignettes, white race was noted in 19.7% of cases, black race was in 7.9% cases, and 76.6% of cases were unspecified. In the 983 UMN vignettes, white race was recorded in 2.85% cases, and black race in 0.41% cases. The institutions were significantly different in the proportion of their cases depicting race (0.20; 95% CI (0.15, 0.25)). Males were represented in the majority of vignettes. Discussion: Comparing case vignettes results from two medical schools suggests that reporting explicit demographic diversity was not significantly different. The findings illustrate that sex was the demographic characteristic consistently described, where males were over-represented. Based on these findings, greater cultural diversity as it intersects with social determinants of health is needed in medical student education.

Highlights The surface functionality of pyrolyzed biomass-derived nanoparticles (PBNPs), described here as those derived from bulk biochar and activated carbon, differs from parent material. The literature on PBNPs derived from biochar is growing, unlike that of activated carbon derived PBNPs. There is a need for better understanding of co-transport of contaminants, photochemistry, and toxicity of PBNPs.

Abstract Background Lignocellulosic conversion residue (LCR) is the material remaining after deconstructed lignocellulosic biomass is subjected to microbial fermentation and treated to remove the biofuel. Technoeconomic analyses of biofuel refineries have shown that further microbial processing of this LCR into other bioproducts may help offset the costs of biofuel generation. Identifying organisms able to metabolize LCR is an important first step for harnessing the full chemical and economic potential of this material. In this study, we investigated the aerobic LCR utilization capabilities of 71 Streptomyces and 163 yeast species that could be engineered to produce valuable bioproducts. The LCR utilization by these individual microbes was compared to that of an aerobic mixed microbial consortium derived from a wastewater treatment plant as representative of a consortium with the highest potential for degrading the LCR components and a source of genetic material for future engineering efforts. Results We analyzed several batches of a model LCR by chemical oxygen demand (COD) and chromatography-based assays and determined that the major components of LCR were oligomeric and monomeric sugars and other organic compounds. Many of the Streptomyces and yeast species tested were able to grow in LCR, with some individual microbes capable of utilizing over 40% of the soluble COD. For comparison, the maximum total soluble COD utilized by the mixed microbial consortium was about 70%. This represents an upper limit on how much of the LCR could be valorized by engineered Streptomyces or yeasts into bioproducts. To investigate the utilization of specific components in LCR and have a defined media for future experiments, we developed a synthetic conversion residue (SynCR) to mimic our model LCR and used it to show lignocellulose-derived inhibitors (LDIs) had little effect on the ability of the Streptomyces species to metabolize SynCR. Conclusions We found that LCR is rich in carbon sources for microbial utilization and has vitamins, minerals, amino acids and other trace metabolites necessary to support growth. Testing diverse collections of Streptomyces and yeast species confirmed that these microorganisms were capable of growth on LCR and revealed a phylogenetic correlation between those able to best utilize LCR. Identification and quantification of the components of LCR enabled us to develop a synthetic LCR (SynCR) that will be a useful tool for examining how individual components of LCR contribute to microbial growth and as a substrate for future engineering efforts to use these microorganisms to generate valuable bioproducts.

Abstract Earth’s earliest fossils of complex macroscopic life are recorded in Ediacaran-aged siliciclastic deposits as exceptionally well-preserved three-dimensional casts and molds, known as “Ediacara-style” preservation. Ediacara-style fossil assemblages commonly include both macrofossils of the enigmatic Ediacara Biota and associated textural impressions attributed to microbial matgrounds that were integral to the ecology of Ediacara communities. Here, we use an experimental approach to interrogate to what extent the presence of mat-forming microorganisms was likewise critical to the Ediacara-style fossilization of these soft-bodied organisms. We find evidence that biofilms can play an instrumental role in fostering fossilization. Rapid silica precipitation associated with macroorganism tissues is enhanced in the presence of mat- and biofilm-forming microorganisms. These results indicate that the occurrence of microbial mats and biofilms may have strongly shaped the preservational window for Ediacara-style fossils associated with early diagenetic silica cements, and therefore influenced the distribution and palaeoecological interpretation of the Ediacara Biota fossil record.

Hydrogen peroxide (H2O2) has been proposed as an electron donor for photosynthesis before water, however, the amount of H2O2 available on early Earth was thought to be limited. Here the authors propose a new abiotic pathway wherein abrasion of quartz surfaces would have provided enough H2O2.

How early photosynthesizers managed oxidative stress remains relatively unresolved. Analyses of enzymes dealing with reactive oxygen species traces the evolutionary history of superoxide dismutases and finds evidence of CuZnSOD in the ancestor of all cyanobacteria, dating back to the Archaean.

Expanded phosphorus availability possibly triggered a marine bioproduction boom after 2.3 billion years ago, but its delivery mechanisms remain unclear. Here we propose a kaolinite shuttle which efficiently adsorbs phosphorus in continental weathering settings and releases it under marine conditions.

Abstract Plant‐soil feedbacks (PSFs) have been shown to strongly affect plant performance under controlled conditions, and PSFs are thought to have far reaching consequences for plant population dynamics and the structuring of plant communities. However, thus far the relationship between PSF and plant species abundance in the field is not consistent. Here, we synthesize PSF experiments from tropical forests to semiarid grasslands, and test for a positive relationship between plant abundance in the field and PSFs estimated from controlled bioassays. We meta‐analyzed results from 22 PSF experiments and found an overall positive correlation (0.12 ≤ r¯ ≤ 0.32) between plant abundance in the field and PSFs across plant functional types (herbaceous and woody plants) but also variation by plant functional type. Thus, our analysis provides quantitative support that plant abundance has a general albeit weak positive relationship with PSFs across ecosystems. Overall, our results suggest that harmful soil biota tend to accumulate around and disproportionately impact species that are rare. However, data for the herbaceous species, which are most common in the literature, had no significant abundance‐PSFs relationship. Therefore, we conclude that further work is needed within and across biomes, succession stages and plant types, both under controlled and field conditions, while separating PSF effects from other drivers (e.g., herbivory, competition, disturbance) of plant abundance to tease apart the role of soil biota in causing patterns of plant rarity versus commonness.

Abstract Background Kolb’s Cycle of Learning Theory acts as a foundational framework for the evolution of knowledge gained by learners throughout their education. Through Kolb’s cycle of experiential learning, one’s preferred way of learning could impact academic achievement in the pre-clinical years of medical education. Methods The medical student classes of 2020 and 2021 at a public university in the southeastern U.S. were invited to complete Kolb’s Learning Style Inventory (LSI). For those participants completing the LSI, examination results for their pre-clinical blocks were obtained and matched to the LSI results. Examination scores (locally-developed examinations and customized National Board of Medical Examiners (NBME) final examinations) were compared by LSI classification for each examination using Kruskal-Wallis Test. Results Out of 360 possible participants, 314 (87.2%) completed the Learning Style Inventory. Convergers and Assimilators made up 84.1% [Convergers (n = 177, 56.4%), Assimilators (n = 87, 27.7%)]. Accommodators (n = 25, 7.9%) and Divergers (n = 25, 7.9%) made up the remaining sample. Accomodators’ scores were significantly lower on locally-developed examinations in Principles of Medicine, Hematology, and Gastrointestinal System. The only NBME examination that demonstrated a significant difference across learning styles was from the Cardiovascular block. Conclusions Upon reviewing Kolb’s LSI, our study indicated that performance on the customized NBME examinations minimized the variance in performance compared to locally-developed examinations. The lack of variance across learning styles for all but one NBME final examination appears to provide a more equitable assessment strategy.

Abstract Little research has assessed how the timing and intensity of grazing might affect plant biomass, available nutrients, soil extracellular enzyme activity (EEA), and how climate change might influence these responses. We tested the effect of two spring precipitation schemes (70, 100% of ambient), two mowing intensities (moderate, severe), and two mowing season treatments (June, October) on plant and soil properties. We detected an interactive effect of precipitation, mowing intensity, and mowing season on plant biomass. When plots were mowed at a moderate intensity, water reductions had irregular effects on plant biomass depending on the mowing season. Plant biomass was also 11% greater in plots mowed at moderate than at severe intensities. Most soil nutrients were unaffected by treatments except for calcium. Soil EEA was unaffected by treatments; however, the activity of a phosphorus (P)‐acquisition enzyme was more than four times greater than the activity of nitrogen (N)‐ and carbon‐acquisition enzymes. A substantial amount (adjusted R2 = .51) of plot‐to‐plot variation in plant biomass was explained by three soil properties especially an N‐acquisition enzyme and to a lesser degree by plant available P and soil pH. The grassland had a high degree of natural buffering capacity as most soil properties were resistant to shifts in 6‐yr spring precipitation and 5‐yr simulated grazing intensity and season. Grassland plant biomass varied by treatments and was seemingly limited by biogeochemical constraints especially the prevalent need to mobilize P and a secondary need to acquire N as plant biomass increased.

The transition from smectite to illite requires potassium incorporation, yet the role of microbes in facilitating K+ uptake remains debated, especially during the early Earth. Here, the authors suggest that the Paleoproterozoic microbial mats extracted potassium from sea water and induced localized illitization during early low-temperature diagenesis.

Background: There is an ongoing call for leadership development in academic health care and medical students desire more training in this area. Although many schools offer combined MD/MBA programs or leadership training in targeted areas, these programs do not often align with medical school leadership competencies and are limited in reaching a large number of students. Methods: The Leadership Initiative (LI) was a program created by a partnership between a School of Medicine (SOM) and Business School with a learning model that emphasized the progression from principles to practice, and the competencies of self-awareness, communication, and collaboration/teamwork. Through offerings across a medical school curriculum, the LI introduced leadership principles and provided an opportunity to apply them in an interactive activity or simulation. We utilized the existing SOM evaluation platform to collect data on program outcomes that included satisfaction, fidelity to the learning model, and impact. Results: From 2017 to 2020, over 70% of first-year medical students participated in LI course offerings while a smaller percentage of fourth-year students engaged in the curriculum. Most students had no prior awareness of LI course material and were equivocal about their ability to apply lessons learned to their medical school experience. Students reported that the LI offerings provided opportunities to practice the skills and competencies of self-awareness, communication, and collaboration/teamwork. Discussion: Adding new activities to an already crowded medical curriculum was the greatest logistical challenge. The LI was successful in introducing leadership principles but faced obstacles in having participants apply and practice these principles. Most students reported that the LI offerings were aligned with the foundational competencies.

The unique cellular organization and transparent function of the ocular lens depend on the continuous differentiation of immature epithelial cells on the lens anterior surface into mature elongated fiber cells within the lens core. A ubiquitous event during lens differentiation is the complete elimination of organelles required for mature lens fiber cell structure and transparency. Distinct pathways have been identified to mediate the elimination of non-nuclear organelles and nuclei. Recently, we reported the discovery of a unique structure in developing fiber cells of the chick embryo lens, called the Nuclear Excisosome, that is intractably associated with degrading nuclei during lens fiber cell differentiation. In the chick lens, the Nuclear Excisosome is derived from projections of adjacent cells contacting the nuclear envelope during nuclear elimination. Here, we demonstrate that, in contrast to the avian model, Nuclear Excisosomes in a primate model, Galago (bush baby) monkeys, are derived through the recruitment of mitochondria to form unique linear assemblies that define a novel primate Nuclear Excisosome. Four lenses from three monkeys aged 2-5 years were fixed in formalin, followed by paraformaldehyde, then processed for Airyscan confocal microscopy or transmission electron microscopy. For confocal imaging, fluorescent dyes labelled membranes, carbohydrate in the extracellular space, filamentous actin and nuclei. Fiber cells from Galago lenses typically displayed prominent linear structures within the cytoplasm with a distinctive cross-section of four membranes and lengths up to 30 μm. The outer membranes of these linear structures were observed to attach to the outer nuclear envelope membrane to initiate degradation near the organelle-free zone. The origin of these unique structures was mitochondria in the equatorial epithelium (not from plasma membranes of adjacent cells as in the chick embryo model). Early changes in mitochondria appeared to be the collapse of the cristae and modification of one side of the mitochondrial outer membrane to promote accumulation of protein in a dense cluster. As a mitochondrion surrounded the dense protein cluster, an outer mitochondrial membrane enclosed the protein to form a core and another outer mitochondrial membrane formed the outermost layer. The paired membranes of irregular texture between the inner core membrane and the outer limiting membrane appeared to be derived from modified mitochondrial cristae. Several mitochondria were involved in the formation and maturation of these unique complexes that apparently migrated around the fulcrum into the cytoplasm of nascent fiber cells where they were stabilized until the nuclear degradation was initiated. Thus, unlike in the chick embryo, the Galago lenses degraded nuclear envelopes with a Nuclear Excisosome derived from multiple mitochondria in the epithelium that formed novel linear assemblies in developing fiber cells. These findings suggest that recruitment of distinct structures is required for Nuclear Excisosome formation in different species.

It has a multifactorial etiology. It had been thought nocturia was a nonspecific symptom of lower urinary system dysfunction, but it has been determined that many diseases, related to different organ systems, might be reasons for this nonspecific symptom. Along with the importance of systemic diseases that cause nocturia, the symptom itself has adverse effects on patients' health and quality of life. There are several studies reporting a direct relationship between nocturia and depression, cognitive dysfunction, mood disturbances, falls, and fractures. For this reason, it is important to treat nocturia both to increase quality of life and to decrease related complications. Treatment opportunities have been under investigation for 20 years. Most of the studies in the literature have reported the results of single-drug medication on nocturia, which may be insufficient for a situation that has such a multifactorial etiology. Soil aggregate stability data are often predicted to be positively associated with measures of plant productivity, rangeland health, and ecosystem functioning. Here we revisit the hypothesis that soil aggregate stability is positively associated with plant productivity. We measured local (plot-to-plot) variation in grassland community composition, plant (aboveground) biomass, root biomass, % water-stable soil aggregates, and topography. After accounting for spatial autocorrelation, we observed a negative association between % water-stable soil aggregates (0.25-1 and 1-2 mm size classes of macroaggregates) and dominant graminoid biomass, and negative associations between the % water-stable aggregates and the root biomass of a dominant sedge (Carex filifolia). However, variation in total root biomass (0-10 or 0-30 cm depths) was either negatively or not appreciably associated with soil aggregate stabilities. Overall, regression slope coefficients were consistently negative thereby indicating the general absence of a positive association between measures of plant productivity and soil aggregate stability for the study area. The predicted positive association between factors was likely confounded by variation in plant species composition. Specifically, sampling spanned a local gradient in plant community composition which was likely driven by niche partitioning along a subtle gradient in elevation. Our results suggest an apparent trade-off between some measures of plant biomass production and soil aggregate stability, both known to affect the land's capacity to resist erosion. These findings further highlight the uncertainty of plant biomass-soil stability associations.

An unresolved issue in structural biology is how the encapsulated lens removes membranous organelles to carry out its role as a transparent optical element. In this ultrastructural study, we establish a mechanism for nuclear elimination in the developing chick lens during the formation of the organelle-free zone. Day 12-15 chick embryo lenses were examined by high-resolution confocal light microscopy and thin section transmission electron microscopy (TEM) following fixation in 10% formalin and 4% paraformaldehyde, and then processing for confocal or TEM as described previously. Examination of developing fiber cells revealed normal nuclei with dispersed chromatin and clear nucleoli typical of cells in active ribosome production to support protein synthesis. Early signs of nuclear degradation were observed about 300 μm from the lens capsule in Day 15 lenses where the nuclei display irregular nuclear stain and prominent indentations that sometimes contained a previously undescribed macromolecular aggregate attached to the nuclear envelope. We have termed this novel structure the nuclear excisosome. This complex by confocal is closely adherent to the nuclear envelope and by TEM appears to degrade the outer leaflet of the nuclear envelope, then the inner leaflet up to 500 μm depth. The images suggest that the nuclear excisosome separates nuclear membrane proteins from lipids, which then form multilamellar assemblies that stain intensely in confocal and in TEM have 5 nm spacing consistent with pure lipid bilayers. The denuded nucleoplasm then degrades by condensation and loss of structure in the range 600 to 700 μm depth producing pyknotic nuclear remnants. None of these stages display any classic autophagic vesicles or lysosomes associated with nuclei. Uniquely, the origin of the nuclear excisosome is from filopodial-like projections of adjacent lens fiber cells that initially contact, and then appear to fuse with the outer nuclear membrane. These filopodial-like projections appear to be initiated with a clathrin-like coat and driven by an internal actin network. In summary, a specialized cellular organelle, the nuclear excisosome, generated in part by adjacent fiber cells degrades nuclei during fiber cell differentiation and maturation.

Senescent cells accumulate in fat with aging. We previously found genetic clearance of senescent cells from progeroid INK-ATTAC mice prevents lipodystrophy. Here we show that primary human senescent fat progenitors secrete activin A and directly inhibit adipogenesis in non-senescent progenitors. Blocking activin A partially restored lipid accumulation and expression of key adipogenic markers in differentiating progenitors exposed to senescent cells. Mouse fat tissue activin A increased with aging. Clearing senescent cells from 18-month-old naturally-aged INK-ATTAC mice reduced circulating activin A, blunted fat loss, and enhanced adipogenic transcription factor expression within 3 weeks. JAK inhibitor suppressed senescent cell activin A production and blunted senescent cell-mediated inhibition of adipogenesis. Eight weeks-treatment with ruxolitinib, an FDA-approved JAK1/2 inhibitor, reduced circulating activin A, preserved fat mass, reduced lipotoxicity, and increased insulin sensitivity in 22-month-old mice. Our study indicates targeting senescent cells or their products may alleviate age-related dysfunction of progenitors, adipose tissue, and metabolism.

Background: Curricular redundancy can be a significant problem for any educational curriculum. Redundancy can be both desirable and undesirable, but differentiating the two can be quite challenging. Further, pinpointing undesirable redundancy and quantifying it so as to produce an estimate of inefficiency is even more difficult. Purpose: The purpose of this research is to describe a student-led strategy for evaluating redundancy in a highly integrated medical school curriculum. It is our hope that the methodology presented here will serve as a useful evaluation model for persons attempting similar work in various educational arenas. Setting: A highly-integrated medical school at a large public university. Intervention: This research did not require an intervention. Research Design: We identified two advanced medical students and asked them to identify redundant material across the first two years of the medical school curriculum. The students had to operationalize ‘redundancy’, develop an evaluation plan/framework, and evaluate the extent to which undesirable redundancy was prevalent in the current curriculum. Data Collection and Analysis: Students reviewed course syllabi, notes, and materials and documented the amount of redundant material they found in the curriculum. Findings: A total of approximately 167 hours, or 8.35 weeks, could be eliminated from the curriculum; the vast majority of the redundancy occurred as a result of small group activities.

Many tree species have seedling recruitment patterns suggesting that they are affected by non-competitive distance-dependent sources of mortality. We conducted an experiment, with landscape-level replication, to identify cases of negative distance-dependent effects and whether variation in these effects corresponded with tree recruitment patterns in the southern Appalachian Mountains region. Specifically, soil was collected from 14 sites and used as inocula in a 62 day growth chamber experiment determining whether tree seedling growth was less when interacting with soil from conspecific (like) than heterospecific (other) tree species. Tests were performed on six tree species. Three of the tree species had been previously described as having greater recruitment around conspecifics (i.e. facilitator species group) compared to the other half (i.e. inhibitor species group). We were then able to determine whether variation in negative distance-dependent effects corresponded with recruitment patterns in the field. Across the six species, none were negatively affected by soil inocula from conspecific relative to heterospecific sources. Most species (four of six) were unaffected by soil source. Two species (Prunus serotina and Tsuga canadensis) had enhanced growth in pots inoculated with soil from conspecific trees vs. heterospecifics. Species varied in their susceptibility to soil pathogens, but trends across all species revealed that species classified as inhibitors were not more negatively affected by conspecific than heterospecific soil inocula or more susceptible to pathogenic effects than facilitators. Although plant-soil biota interactions may be important for individual species and sites, it may be difficult to scale these interactions over space or levels of ecological organization. Generalizing the importance of plant-soil feedbacks or other factors across regional scales may be especially problematic for hyperdiverse temperate forests where interactions may be spatially variable.
