Abstract

Autophagy, an intracellular degradation process, has emerged as a crucial innate immune response against various plant pathogens, including viruses. Tomato spotted wilt orthotospovirus (TSWV) is a highly destructive plant pathogen that infects over 1000 plant species and poses a significant threat to global food security. However, the role of autophagy in defence against the TSWV pathogen, and whether the virus counteracts this defence, remains unknown. In this study, we report that autophagy plays an important role in antiviral defence against TSWV infection; however, this autophagy‐mediated defence is counteracted by the viral effector NSs. Transcriptome profiling revealed the up‐regulation of autophagy‐related genes (ATGs) upon TSWV infection. Blocking autophagy induction by chemical treatment or knockout/down of ATG5/ATG7 significantly enhanced TSWV accumulation. Notably, the TSWV nucleocapsid (N) protein, a major component of the viral replication unit, strongly induced autophagy. However, the TSWV nonstructural protein NSs was able to effectively suppress N‐induced autophagy in a dose‐dependent manner. Further investigation revealed that NSs inhibited ATG6‐mediated autophagy induction. These findings provide new insights into the defence role of autophagy against TSWV, a representative segmented negative‐strand RNA virus, as well as the tospoviral pathogen counterdefence mechanism. Abstract

Myzus persicae and Brevicoryne brassicae are the main aphid pests and vectors of viruses that attack many crops, including oilseed rape, the most commonly grown dicotyledonous crop in the Czech Republic. In this study, we analysed the flight activity of both aphids from five suction traps over 20 years (2004–2023). We focussed on (i) the influence of the ban on seed dressing with neonicotinoids on aphid catches, (ii) the influence of the average temperature on flight activity, and (iii) the number of males/females caught in the suction traps. We compared the data on aphid flight activity at the coldest and warmest sites and found that migration starts about 10 days earlier at the warmest site than at the coldest site and that aphid populations are more numerous here. The number of catches in suction traps was observed for both aphids after the ban on neonicotinoids was significantly increased. For M. persicae, it was about 9 times higher, while for B. brassicae, it was about 1.3 times higher. In addition, we observed a significant decrease in the number of males in M. persicae with a simultaneous increase in the number of females (in autumn), which could be an indication of the formation of anholocyclic populations. Overall, our results showed a significantly high abundance of both aphid species depending on the observed locations, which seems to be related to the ban of neonicotinoids. Therefore, an effective alternative is needed to improve the control of aphid vectors that can ensure the stability of crops against aphid-transmitted virus diseases. [ABSTRACT FROM AUTHOR]

Abstract

Wheat streak mosaic virus (WSMV) causes wheat streak mosaic, a disease of cereals and grasses that threatens wheat production worldwide. It is a monopartite, positive-sense, single-stranded RNA virus and the type member of the genus Tritimovirus in the family Potyviridae. The only known vector is the wheat curl mite (WCM, Aceria tosichella), recently identified as a species complex of biotypes differing in virus transmission. Low rates of seed transmission have been reported. Infected plants are stunted and have a yellow mosaic of parallel discontinuous streaks on the leaves. In the autumn, WCMs move from WSMV-infected volunteer wheat and other grass hosts to newly emerged wheat and transmit the virus which survives the winter within the plant, and the mites survive as eggs, larvae, nymphs or adults in the crown and leaf sheaths. In the spring/summer, the mites move from the maturing wheat crop to volunteer wheat and other grass hosts and transmit WSMV, and onto newly emerged wheat in the fall to which they transmit the virus, completing the disease cycle. WSMV detection is by enzyme-linked immunosorbent assay (ELISA), reverse transcription-polymerase chain reaction (RT-PCR) or quantitative RT-PCR (RT-qPCR). Three types of WSMV are recognized: A (Mexico), B (Europe, Russia, Asia) and D (USA, Argentina, Brazil, Australia, Turkey, Canada). Resistance genes Wsm1, Wsm2 and Wsm3 have been identified. The most effective, Wsm2, has been introduced into several wheat cultivars. Mitigation of losses caused by WSMV will require enhanced knowledge of the biology of WCM biotypes and WSMV, new or improved virus detection techniques, the development of resistance through traditional and molecular breeding, and the adaptation of cultural management tactics to account for climate change.

Abstract

The two contrasting cultivars, the very susceptible Ludwig and moderately susceptible Svitava, previously evaluated in field trials to determine their resistance levels to Wheat dwarf virus (WDV), were analysed by four different test methods to allow for the comparison of the methods and to consider whether the resistance level of cv. Svitava can be an effective component of wheat protection from the virus. The differences in the proportion of the diseased plants were observed after the inoculation by the viruliferous leafhoppers. The differences in the cultivar resistance levels were evaluated by comparing the biomass production after the infection. The amount of viral DNA was determined using quantitative polymerase chain reaction (PCR). Concurrently, the cultivar response to the virus infection after the controlled inoculation by the leafhoppers [Psammotettix alienus (Dahlbom, 1850)] under field conditions was further evaluated. The infected plants of cv. Svitava produced five times more dry matter in the vegetative growth stage than cv. Ludwig. The quantitative PCR analysis confirmed the resistance in cv. Svitava. The plant inoculation under a standardised infection pressure showed that the probability of the WDV infection after the inoculation feeding is smaller in the cv. Svitava plants. The comparison of the results from the different test methods shows that the field resistance tests are necessary to obtain a proper assessment of the possibilities for the cultivars' utilisation in the crop protection. 43. Two mutations in the truncated Rep gene RBR domain delayed the Wheat dwarf virusinfection in transgenic barley plants

Abstract

Wheat dwarf virus(WDV), an important cereal pathogen, is closely related to Maize streak virus(MSV), a model virus of the Mastrevirusgenus. Based on its similarity to known MSV resistance strategies, a truncated part of the WDV replication-associated (RepA) gene (WDVRepA215) and the WDV RepA gene with a mutated retinoblastoma-related protein (RBR) interaction domain (WDVRepA215RBRmut) were cloned into the pIPKb002 expression vector and transformed into immature embryos of spring barley cv. Golden Promise plants through Agrobacterium-mediated transformation. A detailed study of T1-generation plants infected by leafhoppers (Psammotettix alienus) fed on infection sources of variable strength was performed over a 5-week period encompassing the initial stages of virus infection. A DNA WDV TaqMan qPCR assay normalized using the DNA puroindoline-b SYBR Green qPCR assay for samples on a per week basis revealed an approximately 2-week delay in WDVRepA215RBRmutplants to WDVRepA215 plants before significant increases in the WDV viral levels occurred. Both WDVRepA215 and WDVRepA215RBRmutplants showed similar levels of transgenic transcripts over the screened period; however, the transgenic plants also showed increased numbers of infected plants compared to the control plants.
