Abstract Bio- and phytoremediation, being encouraging terms implying the use of biological systems for cleansing purposes, have risen a worthy venture toward environmental restoration in discouraging scenarios, such as the augmentation of indestructible heavy metals. Hyperaccumulating plants and heavy metal resistant microbes own mechanisms embedded in their metabolism, proteins, and genes that confer them with “super characteristics” allowing them to assimilate heavy metals in order to amend polluted soils, and when combined in a symbiotic system, these super features could complement each other and be enhanced to overpower the exposure to toxic environments. Though xenobiotic pollution has been an object of concern for decades and physicochemical procedures are commonly carried out to offset this purpose, a “live” remediation is rather chosen and looked upon for promising results. A variety of benefits have been registered from symbiotic relationships, including plants teaming up with microbes to cope down with non-biodegradable elements such as heavy metals; but a carefully maneuvered interaction might signify a greater insight toward the application of bioremediation systems. These manipulations could consist of genetic engineering and/or additional supplementation of molecules and microbes. In the present study, a contemporary connection between plants and microbes involving their controlled management is summarized in a visionary display.

The increasing need for metals leads to contaminated post-mining landscapes. At the same time, the contamination with organic, recalcitrant contamination increases. This poses a problem of reuse of large areas, often co-contaminated with both, metals, and organic pollutants. For the remediation of areas contaminated with multiple contaminants and combining many stress factors, technical solutions including groundwater treatment, where necessary, have been devised. However, this is applied to highly contaminated, small sites. The reuse of larger, co-contaminated landscapes remains a major challenge. Mycoremediation with fungi offers a good option for such areas. Fungi cope particularly well with heterogeneous conditions due to their adaptability and their large hyphal network. This chapter summarizes the advantages of basidiomycetes with a focus on wood rot fungi in terms of their ability to tolerate metals, radionuclides, and organic contaminants such as polycyclic aromatic hydrocarbons. It also shows how these fungi can reduce toxicity of contaminants to other organisms including plants to allow for restored land-use. The processes based on diverse molecular mechanisms are introduced and their use for mycoremediation is discussed. [ABSTRACT FROM AUTHOR]

Weathering of ancient organic matter contributes significantly to biogeochemical carbon cycles over geological times. The principle role of microorganisms in this process is well recognized. However, information is lacking on the contribution of individual groups of microorganisms and on the effect of labile carbon sources to the degradation process. Therefore, we investigated the contribution of fungi, Gram-positive and Gram-negative bacteria in the degradation process using a column experiment. Investigations were performed on low metamorphic black slates. All columns contained freshly crushed, sieved (0.63-2mm), not autoclaved black slates. Two columns were inoculated with the lignite-degrading fungus Schizophyllum commune and received a culture medium containing 13C labeled glucose, two columns received only this culture medium and two control columns received only water.The total mass balance was calculated from all carbon added to the slate and the CO2 and DOC losses. Phospholipid fatty acids (PLFA) were extracted to investigate microbial communities. We used both the compound specific 14C and 13C signal of the PLFA to quantify carbon uptake from black slates and the glucose of the culture medium, respectively.The total carbon loss in these columns exceeded the amount of added carbon by approximately 60%, indicating that black slate carbon has been used. PLFA associated with Gram-positive bacteria dominated the indigenous community and took up 22% of carbon from black slate carbon, whereas PLFA of Gram-negative bacteria used only 8% of carbon from the slates. PLFA of Gram-negative bacteria and fungi were both mostly activated by the glucose addition. The added Schizophyllum did not establish well in the columns and was overgrown by the indigenous microbial community. Our results suggest that especially Gram-positive bacteria are able to live on and degrade black slate material. They also benefit from easy degradable carbon from the nutrient broth. In natural environments priming due to root exudates might consequently enhance weathering. © 2011 Elsevier Ltd.

The wood degrading fungus Schizophyllum commune is a model system for mushroom development. Here, we describe the 38.5 Mb assembled genome of this basidiomycete and application of whole genome expression analysis to study the 13,210 predicted genes. Comparative analyses of the S. commune genome revealed unique wood degrading machinery and mating type loci with the highest number of reported genes. Gene expression analyses revealed that one third of the 471 identified transcription factor genes were differentially expressed during sexual development. Two of these transcription factor genes were deleted. Inactivation of fst4 resulted in the inability to form mushrooms, whereas inactivation of fst3 resulted in more but smaller mushrooms than wild-type. These data illustrate that mechanisms underlying mushroom formation can be dissected using S. commune as a model. This will impact commercial production of mushrooms and the industrial use of these fruiting bodies to produce enzymes and pharmaceuticals
