The microbial communities that characterize modem hydrothermal ecosystems serve as modern analogs to those thought to have dominated early environments on Earth and possibly Mars. The importance of such hydrothermal systems as targets in exploring for an early biosphere on Mars is well established. Such work provides an important basis for the analysis of Martian samples associated with such environments. The surviving molecular structure and isotopic signature of diagnostic lipid biomarkers found as chemical fossils can provide a link between modern bacterially dominated ecosystems and their ancient counterparts. We are interested in the processes involved in the deposition and/or degradation of organic material in moderately thermal, silicifying microbial mats, particularly as this relates to the potential for preservation of some biomarker components known to be more highly resistant to microbial degradation. Several excellent biomarker molecules are associated with the cyanobacteria that dominate these mats, particularly the 2-methylbacteriohopanepolyols (2-MeBHP). These compounds are ubiquitous on Earth and are not easily degraded in nature, a fact documented by their detection in ancient Earth rocks dating back as far as 2,700 Ma.

As the prospect grows for a Mars sample return mission early in the next millennium, it will be important to ensure that appropriate planetary protection (PP) controls are incorporated into the mission design and implementation from the start. The need for these PP controls is firmly based on scientific considerations and backed by a number of national and international agreements and guidelines aimed at preventing harmful cross contamination of planets and extraterrestrial bodies. The historical precedent for the use of PP measures on both unmanned and manned missions traces from post-Sputnik missions to the present, with periodic modifications as new information was obtained. In consideration of the anticipated attention to PP questions by both the scientific/technical community and the public, this paper presents a comprehensive review of the major issues and problems surrounding PP for a Mars Sample Return (MSR) mission, including an analysis of arguments that have been raised for and against the imposition of PP measures. Also discussed are the history and foundations for PP policies and requirements; important research areas needing attention prior to defining detailed PP requirements for a MSR mission; and legal and public awareness issues that must be considered with mission planning.

The purpose of the workshop was to re-evaluate the existing guidelines for the probability of growth of terrestrial organisms on Mars that were established by the National Academy of Sciences following the Viking Mission. A panel of specialists in microbiology and allied fields reviewed this issue in the light of current information about the physical and chemical environments expected on Mars. Their deliberations resulted in the virtually unanimous conclusion that the existing Pg guidelines were either appropriate or that the values for Pg should be further reduced. Individual assessments of this problem by each of the participants, together with those of additional invited experts are included.

An attempt is made to describe the scope of scientific areas that comprise the current field of exobiology in the United States. From investigations of astrophysical phenomena that deal with the birth of stars and planetary systems to questions of molecular biology involving phylogenetic relationships among organisms, from attempts to simulate the synthesis of biological precursor molecules in the chemistry laboratory to making measurements of the organic constituents of Titan's atmosphere, these researches all converge toward a common objective--answering the question of how life came about in the universe.

Five general categories of experimentation on the moon are reviewed which would provide important scientific information about chemical evolution and the origin and early evolution of life. Attention is given to: (1) chemical analyses of samples for products of prebiotic evolution, (2) analysis of crater records and understanding the implications for the origin and evolution of life on earth, (3) exposure of microbes and organics to the space environment, (4) collection of cosmic dust particles and analysis of volatiles and organics, and (5) observations of planetary environments and the interstellar medium to determine the nature and extent of chemical evolution.

A brief review of the purposes and the results from the Viking Biology experiments is presented, in the expectation that the lessons learned from this mission will be useful in planning future approaches to the biological exploration of Mars. Since so little was then known about potential micro-environments on Mars, three different experiments were included in the Viking mission, each one based on different assumptions about what Martian organisms might be like. In addition to the Viking Biology Instrument (VBI), important corollary information was obtained from the Viking lander imaging system and from the molecular analysis experiments that were conducted using the gas chromatograph-mass spectrometer (GCMS) instrument. No biological objects were noted by the lander imaging instrument. The GCMS did not detect any organic compounds. A description of the tests conducted by the Gas Exchange Experiment, the Labeled Release experiment, and the Pyrolytic Release experiment is given. Results are discussed. Taken as a whole, the Viking data yielded no unequivocal evidence for a Martian biota at either landing site. The results also revealed the presence of one or more reactive oxidants in the surface material and these need to be further characterized, as does the range of micro-environments, before embarking upon future searches for extant life on Mars.

Mars has been an object of interest for the better part of this century. To a biologist, Mars assumes special importance because many aspects of the theory of chemical evolution for the origin of life can be tested there. The central idea of this theory is that life on a suitable planet arises through a process in which the so-called biogenic elements combine to form increasingly more complex molecules under the influence of naturally-occurring energy sources ultimately resulting in the formation of replicating organic molecules. The biogenic elements are present on Mars today. Furthermore, the available evidence also strongly suggests that Mars may have had an early history similar to that of the Earth, including a period in which large amounts of liquid water once flowed on its surface and a denser atmosphere and higher global temperatures prevailed. This is important since many lines of evidence indicate that living organisms were already present on the Earth within the first billion years after its formation at a time when the environment on Mars may have closely resembled that of Earth. Our current knowledge of the state of chemical evolution on Mars can best be described as paradoxical. Most of what we have learned has come from experiments performed on the Viking landers. The combination of planned investigations covered a broad range of techniques to detect signs of chemical evolution. The most surprising data from all of these was the absence of any detectable quantities of organic compounds at the two landing sites. On the other hand, the Viking experiments did indicate that the Martian surface samples contained unidentified strong oxidant(s) that could account for their absence.

This report is the work of a panel of eight US scientists who surveyed and assessed Soviet research in the spare sciences. All of the panelists were very familiar with Soviet research through their knowledge of the published scientific literature and personal contacts with Soviet and other foreign colleagues. In addition, all of the panelists reviewed considerable additional open literature--scientific, and popular, including news releases. The specific disciplines of Soviet space science research examined in detail for the report were: solar-terrestrial research, lunar and planetary research, space astronomy and astrophysics, and, life sciences. The Soviet Union has in the past carried out an ambitious program in lunar exploration and, more recently, in studies of the inner planets, Mars and especially Venus. The Soviets have provided scientific data about the latter planet which has been crucial for studies of the planet's evolution. Future programs envision an encounter with Halley's Comet, in March 1986, and missions to Mars and asteroids. The Soviet programs in the life sciences and solar-terrestrial research have been long-lasting and systematically pursued. Much of the ground-based and space-based research in these two disciplines appears to be motivated by the requirement to establish long-term human habitation in near-Earth space. The Soviet contributions to new discoveries and understanding in observational space astronomy and astrophysics have been few. This is in significant contrast to the very excellent theoretical work contributed by Soviet scientists in this discipline.

A procedure for the purification of a stable acetyl-coenzyme A synthetase (ACS) from aerobic cells of Saccharomyces cerevisiae is presented. The steps include differential centrifugation, solubilization of the bound enzyme from the crude mitochondrial fraction, ammonium sulfate fractionation, crystallization to constant specific activity from ammonium sulfate solutions followed by Bio-Gel A-1.5 m column chromatography. The resulting enzyme preparation is homogeneous as judged by chromatography on Bio-Gel columns, QAE-Sephadex A-50 anion exchange columns, analytical ultracentrifugal studies, and polyacrylamide gel electrophoresis. Sedimentation velocity runs revealed a single symmetric peak with an s(sub (20,w)) value of 10.6. The molecular weight of the native enzyme, as determined by gel filtration and analytical ultracentrifugation, is 250,000 +/- 500. In polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate, the molecular weight of the single polypeptide chain is 83,000 +/- 500. The purified enzyme is inhibited by palmityl-coenzyme A with a Hill interaction coefficient, n, of 2.88. These studies indicate that the ACS of aerobic Saccharomyces cerevisiae is composed of three subunits of identical or nearly identical size.

During the purification of the "aerobic" acetyl-CoA synthetase (ACS) of Saccharomyces cerevisiae, strain LK2Gl2, it was noted that stronge at 4 C resulted in the loss of enzyme activity within 24 hr. Similar losses were observed during column chromatography. Addition of boiled extracts from either aerobic or anerobic cells completely prevents this. The stabilizing factor (SF) in these extracts is non-dialyzable and organic in nature. SF is excluded on G-25 and G-50 Sephadex columns and is slightly retarded on G-75 columns. On G-100 columns, SF elutes as a peak exactly coincident with that of cytochrome c, indicating a molecular weight of 13,000. SF activity was not destroyed by Pronase treatment, was adsorbed onto Norite, and absorbed in the UV with a single maximum at 260 nm. The action of SF could be replaced by a number of nucleotides. At 0.01 M, the order of effectiveness was: ATP>ADP>AMP>GTP>CTP>/=UTP>XTP. Even at 2 x 10(exp -4) M, ATP and ADP, but not AMP, cyclic AMP, adenosine or adenine, were effective in stabilizing this ACS. The mechanism of stabilization by ATP and AMP appears to be the same, since AMP competitively inhibited the ACS with respect to ATP in in vitro assays, while ADP gave a mixed type of inhibition, thus indicating a different mechanism. ACS from nonaerobic cells is also unstable in the absence of SF but, unlike aerobic ACS, is not affected by ATP or other nucleotides.
