In this study, using the concept of "perezhivanie" as an analytical tool, we were trying out new ways to investigate L2 identity taking the peripheral educational context into account. We examined how two graduates from non-academic high schools perceived their peripheral school situation through their "perezhivanie," and described how this situation affected the individual trajectories of L2 identity development as well. When two students immersed themselves in marginalized classroom contexts, they have experienced academic stigma in the context of classroom community. Moreover, they all experienced emotional conflicts related to English learning. Besides, they struggled over deficit remedial L2 identity, entailing identity tensions. As they attempted to reconcile the contradictions between themselves and their circumstances, their "perezhivanie" made their social situation of development differently. Depending on how the contradiction was being emotionally experienced through the prism of each student's "perezhivanie," the same contradiction had different meanings, led to different reactions, and had differing impacts on their L2 identity.

Background: Transient receptor potential vanilloid 1 (TRPV1) is associated with skin sensitivity and mainly activated by capsaicin and heat. Interestingly, troxerutin can inhibit TRPV1 activation. However, its efficacy in reducing skin sensitivity remains undetermined. Aims: We evaluated the efficacy of troxerutin in alleviating skin sensitivity using clinical tests and in vitro experiments. Methods: For the in vitro experiment, HaCaT keratinocytes were pretreated with different concentrations of troxerutin, followed by incubation with 50 μM capsaicin for 1, 24, or 48 h. The gene and protein expressions of four inflammatory cytokines involved in skin irritation were determined. Among 35 Korean women with sensitive skin recruited for the clinical trial, 13 were involved in assessing the immediate soothing effects of 0.1% and 0.0095% troxerutin following capsaicin irritation, whereas 22 participated in evaluating the preventive soothing effect of 10% and 1% troxerutin over 4 weeks against capsaicin‐ and heat‐induced irritation. We evaluated the soothing rate using skin redness, visual analog scale, and high temperature sensitive index as evaluation indices. Results: Troxerutin inhibited the mRNA and protein expressions of cytokines in capsaicin‐treated keratinocytes. In the clinical study, 0.1% and 0.0095% troxerutin promptly alleviated capsaicin‐induced skin redness, whereas 10% troxerutin notably decreased both the visual analog scale and high temperature sensitive index for capsaicin‐ and heat‐related irritation. However, 1% troxerutin was only effective in reducing the visual analog scale in response to capsaicin irritation. Conclusions: Troxerutin can inhibit TRPV1 activation in clinical and in vitro tests. Purpose: The study explored the enhanced skin moisturizing capabilities and moisture retention effects achieved by forming a polyion complex using sulfated glycosaminoglycan (GAG), specifically chondroitin sulfate (CS), and amino acids (AA) such as glutamine (Q) and arginine (R). The overall hydration effect of this CS‐AA complex was examined. Methods: After analyzing the CS‐AA polyion complex structure using spectroscopic methods, the ex vivo moisture retention ability was assessed under dry conditions using porcine skin samples. Additionally, the efficacy of the CS‐AA polyion complex in reducing transepidermal water loss (TEWL) and improving skin hydration was evaluated on human subjects using a digital evaporimeter and a corneometer, respectively. Results: Validating a systematic reduction in particle size, the following order was observed: CS > CS/AA simple mixture > CS‐AA complex based on dynamic light scattering (DLS) and transmission electron microscopy (TEM) analysis. Furthermore, observations revealed that the CS‐AA complex exhibits negligible surface charge. Additionally, Fourier‐transform infrared spectroscopy (FT‐IR) analysis demonstrated a distinct peak shift in the complex, confirming the successful formation of the CS‐AA complex. Subsequently, the water‐holding effect through porcine skin was assessed, revealing a notable improvement in moisture retention (weight loss) for the CS‐Q complex: 40.6% (1 h), 20.5% (2 h), and 18.7% (4 h) compared to glycerin. Similarly, the CS‐R complex demonstrated enhancements of 50.2% (1 h), 37.5% (2 h), and 33% (4 h) compared to glycerin. Furthermore, TEWL improvement efficacy on human skin demonstrated approximately 25% improvement for both the CS‐Q complex and CS‐R complex, surpassing the modest 12.5% and 18% improvements witnessed with water and glycerin applications, respectively. Finally, employing a corneometer, hydration changes in the skin were monitored over 4 weeks. Although CS alone exhibited nominal alterations, the CS‐Q complex and CS‐R complex showed a significant increase in moisture levels after 4 weeks of application. Conclusion: In this study, polyion complexes were successfully formed between CS, a sulfated GAG, and AA. Comparisons with glycerin, a well‐known moisturizing agent, confirmed that the CS‐AA complex exhibits superior moisturizing effects in various aspects. Neonatal cholestasis is a potentially life-threatening condition requiring prompt diagnosis. Mutations in several different genes can cause progressive familial intrahepatic cholestasis, but known genes cannot account for all familial cases. Here we report four individuals from two unrelated families with neonatal cholestasis and mutations in NR1H4, which encodes the farnesoid X receptor (FXR), a bile acid-activated nuclear hormone receptor that regulates bile acid metabolism. Clinical features of severe, persistent NR1H4-related cholestasis include neonatal onset with rapid progression to end-stage liver disease, vitamin K-independent coagulopathy, low-to-normal serum gamma-glutamyl transferase activity, elevated serum alpha-fetoprotein and undetectable liver bile salt export pump (ABCB11) expression. Our findings demonstrate a pivotal function for FXR in bile acid homeostasis and liver protection.

Background/Aim: Frondoside A is a sea cucumber extract which is well known for its anti‐inflammatory and immunomodulatory properties. The purpose of this study was to evaluate the effect of Frondoside A application in the alveolar socket on inflammatory responses after delayed replantation in rat teeth. Materials and Methods: Human periodontal ligament cells were cultured and exposed to Frondoside A. Cell‐counting kit‐8 assay was performed to evaluate the cell viability and nitric oxide assay was performed to assess the anti‐inflammatory effect of Frondoside A. Molars were extracted from 32 Sprague–Dawley rats and randomly divided into control and Frondoside A groups. After 30 min of extra‐oral dry time, molars were replanted. In the Frondoside A group, Frondoside A solution was applied in the alveolar socket before replantation. The animals were sacrificed after 28 days and histologically and immunohistochemically evaluated. Results: 0.5 μM Frondoside A showed higher cellular viability at 6 h and lower production of nitric oxide compared with other Frondoside A solutions (p <.05). The Frondoside A group demonstrated lower inflammatory resorption scores in both middle 1/3 and apical 1/3 of root compared to the control group (p <.05). The Frondoside A group showed lower levels of expression in both cathepsin K and CD45 compared with the control group (p <.05). Conclusions: Within the limits of this study, intra‐alveolar delivery of Frondoside A alleviates inflammatory root resorption in delayed replantation of rat teeth. [ABSTRACT FROM AUTHOR]

Background/Aim: Tooth avulsion and delayed replantation may cause inflammatory responses and root resorption of the tooth. The aim of this study is to investigate the effect of a doxycycline-loaded nitric oxide-releasing nanomatrix (DN) gel on the delayed replantation of avulsed rat teeth, with a focus on assessing the gel's potential to promote regeneration and inhibit complications associated with the replantation process. Materials and Methods: Twenty-four right maxillary first molars from male Sprague-Dawley rats were atraumatically extracted using sterile extraction forceps. The molars were dried for 1 h at room temperature (approximately 23 °C) and divided into four groups according to the root conditioning methods after extra-alveolar 60-min drying: Group 1, no root conditioning treatment prior to replantation; Group 2, soaking in 2% NaF solution for 5 min before replantation; Group 3, 5-min soaking in NO gel and injection of the gel into the alveolar socket; Group 4, 5-min soaking in DN gel and injection of the gel into the alveolar socket before replantation. The animals were euthanized four weeks after the operation and the specimens were evaluated histologically. Results: The use of NO gel alone showed better anti-inflammatory and periodontal effects than the control group, but it did not show a significant effect compared to the group using NaF. When using NO gel loaded with doxycycline, it showed a significant anti-inflammatory effect compared to the control group and showed a similar inhibitory effect to the group using NaF. Conclusions: Within the limits of this study, in delayed replantation situations, the control of inflammatory resorption and replacement resorption is an important factor for achieving a better prognosis of replanted teeth.
