Magnesium is well known as a biodegradable biomaterial that has been reported to promote bone remodeling in several studies; however, the underlying biological mechanism remains unclear. In the present study, the role of magnesium ions in the migration of U-2 OS cells, which are osteoblast-like cell lines, was investigated. Magnesium treatment did not significantly alter the global transcriptome of U-2 OS cells, but increased the protein expression level of SNAI2, an epithelial-mesenchymal transition (EMT) marker. In addition, it was confirmed that the junctional site localization of Zona-occludens 1 (ZO-1), a representative tight junction protein, was destroyed by magnesium treatment; furthermore, it was determined that cytoplasmic localization increased, and alkaline phosphatase (ALP) activity increased. The obtained results on the mechanism by which magnesium is involved in osteoblast migration, which is important for fracture healing, will contribute to the understanding of the bone-formation process in patients with osteoporosis and musculoskeletal injury.

Power and dependence in economic exchange shape industry structure. When a focal industry faces powerful suppliers or buyers, this can reduce industry rents. The authors argue that these dynamics also affect job quality by reducing the economic surplus available to be shared with workers. Drawing on ideas from power-dependency theory, this article explains industry earnings and job quality differences by examining inter-industry exchange patterns. The authors build on Ronald Burt’s seminal analysis of structural constraint in economic exchange using industry input-output tables. They calculate market constraint measures for recent years in the United States and link these with CPS data on wages and benefits. Analyses reveal that workers in more buyer-constrained industries (dependence on powerful buyers) experience lower wages and benefits. Findings also show that market constraint reduces the economic surplus available for union bargaining. Theory and results suggest that market concentration reduces suppliers’ economic rents, harming job quality.

This study aims to provide an improved understanding of the local-level spatiotemporal evolution of COVID-19 spread across capital regions of South Korea during the second and third waves of the pandemic (August 2020~June 2021). To explain transmission, we rely upon the local safety level indices along with latent influences from the spatial alignment of municipalities and their serial (temporal) correlation. Utilizing a flexible hierarchical Bayesian model as an analytic operational framework, we exploit the modified BYM (BYM2) model with the Penalized Complexity (PC) priors to account for latent effects (unobserved heterogeneity). The outcome reveals that a municipality with higher population density is likely to have an elevated infection risk, whereas one with good preparedness for infectious disease tends to have a reduction in risk. Furthermore, we identify that including spatial and temporal correlations into the modeling framework significantly improves the performance and explanatory power, justifying our adoption of latent effects. Based on these findings, we present the dynamic evolution of COVID-19 across the Seoul Capital Area (SCA), which helps us verify unique patterns of disease spread as well as regions of elevated risk for further policy intervention and for supporting informed decision making for responding to infectious diseases.

OBJECTIVES: The purpose of this study was to enhance the understanding of the local-level spatiotemporal dynamics of COVID-19 transmission in the Greater Seoul Area (GSA), Korea, after its initial outbreak in January 2020.METHODS: Using the weekly aggregates of coronavirus disease 2019 (COVID-19) cases of 77 municipalities in the GSA, we examined the relative risks of COVID-19 infection across local districts over 50 consecutive weeks in 2020. To this end, we employed a spatiotemporal generalized linear mixed model under the hierarchical Bayesian framework. This allowed us to empirically examine the random effects of spatial alignments, temporal autocorrelation, and spatiotemporal interaction, along with fixed effects. Specifically, we utilized the conditional autoregressive and the weakly informative penalized complexity priors for hyperparameters of the random effects.RESULTS: Spatiotemporal interaction dominated the overall variability of random influences, followed by spatial correlation, whereas the temporal correlation appeared to be small. Considering these findings, we present dynamic changes in the spread of COVID-19 across local municipalities in the GSA as well as regions at elevated risk for further policy intervention.CONCLUSIONS: The outcomes of this study can contribute to advancing our understanding of the local-level COVID-19 spread dynamics within densely populated regions in Korea throughout 2020 from a different perspective, and will contribute to the development of regional safety planning against infectious diseases.

With a recent surge of the new severe acute respiratory syndrome-coronavirus 2 (SARS-Cov-2, COVID-19) in South Korea, this study attempts to investigate the effects of environmental conditions such as air pollutants (PM2.5) and meteorological covariate (Temperature) on COVID-19 transmission in Seoul. To account for unobserved heterogeneity in the daily confirmed cases of COVID-19 across 25 contiguous districts within Seoul, we adopt a full Bayesian hierarchical approach for the generalized linear mixed models. A formal statistical analysis suggests that there exists a positive correlation between a 7-day lagged effect of PM2.5 concentration and the number of confirmed COVID-19 cases, which implies an elevated risk of the infectious disease. Conversely, temperature has shown a negative correlation with the number of COVID-19 cases, leading to reduction in relative risks. In addition, we clarify that the random fluctuation in the relative risks of COVID-19 mainly originates from temporal aspects, whereas no significant evidence of variability in relative risks is observed in terms of spatial alignment of the 25 districts. Nevertheless, this study provides empirical evidence using model-based formal assessments regarding COVID-19 infection risks in 25 districts of Seoul from a different perspective.

Along with the rapid demographic change, there has been increased attention to the risk of vehicle crashes relative to older drivers. Due to senior involvement and their physical vulnerability, it is crucial to develop models that accurately predict the severity of senior-involved crashes. However, the challenge is how to cope with an imbalanced severity class distribution and the ordered nature of crash severities, as these can complicate the classification of the severity of crashes. In that regard, this study investigates the influence of implementing ordinal nature and handling imbalanced class distribution on the prediction performance. Using vehicle crash data in Ohio, U.S., as an example, the eight machine learning classifiers (logistic and ordered logistic regressions and random forest and ordered random forest with or without handling imbalanced classes) are suggested and then compared with their respective performances. The analysis outcomes show that balancing strategy enhances performance in predicting severe crashes. In contrast, the effects of implementing ordinal nature vary across models. Specifically, the ordered random forest classifier without balancing appears to be superior in terms of overall prediction accuracy, and the ordered random forest with balancing outperforms others in predicting severer crashes.

(1) Background: Dental calculus works as a niche wherein pathogenic bacteria proliferate in the oral cavity. Previous studies revealed the anticalculus activity of pyrophosphates, however there was no clinical study that evaluated microbiome changes associated with calculus inhibition. Therefore, the aim of this randomized clinical trial was to evaluate the calculus inhibition of pyrophosphate-containing toothpaste and its effect on oral microbiome changes. (2) Methods: Eighty subjects with a calculus index &ge, 2 on the lingual of the mandibular anterior tooth were randomly allocated to the test group that pyrophosphate-containing toothpaste was given to or the placebo control group. Full mouth debridement and standardized tooth brushing instruction were given before the allocation. Plaque index, gingival index, calculus index, probing depth, and bleeding on probing were measured at the baseline, and at 4, 8 and 12 weeks. Genomic DNA was extracted from the plaque samples collected at the baseline and at 12 weeks, and 16S ribosomal RNA gene amplicon sequencing was applied for microbiome analysis. (3) Results: None of the clinical parameters showed significant differences by visits or groups, except the plaque index of the test group, which reduced significantly between 4 and 12 weeks. A significant difference of microbiome between the baseline and 12 weeks was observed in the test group. Between baseline and 12 weeks, the proportion of Spirochetes decreased in the control group, and the proportions of Proteobacteria, Fusobacteria and Spirochetes in the phylum level and the proportions of Haemophilus, Fusobacterium and Capnocytophaga in the genus level decreased in the test group. In the test group, as plaque index decreased, Streptococcus increased, and Fusobacterium and Haemophilus parainfluenza decreased. (4) Conclusion: The use of pyrophosphate-containing toothpaste effectively inhibited the dysbiosis of the oral microbiome and the proliferation of pathogenic species in periodontal disease. Clinically, plaque formation in the pyrophosphate-containing toothpaste group was effectively decreased, however there was no significant change in calculus deposition.

Chronic hypoperfusion is a key contributor to cognitive decline and neurodegenerative conditions, but the cellular mechanisms remain ill-defined. Using a multidisciplinary approach, we sought to elucidate chronic hypoperfusion-evoked functional changes at the neurovascular unit. We used bilateral common carotid artery stenosis (BCAS), a well-established model of vascular cognitive impairment, combined with an ex vivo preparation that allows pressurization of parenchymal arterioles in a brain slice. Our results demonstrate that mild (~ 30%), chronic hypoperfusion significantly altered the functional integrity of the cortical neurovascular unit. Although pial cerebral perfusion recovered over time, parenchymal arterioles progressively lost tone, exhibiting significant reductions by day 28 post-surgery. We provide supportive evidence for reduced adenosine 1 receptor-mediated vasoconstriction as a potential mechanism in the adaptive response underlying the reduced baseline tone in parenchymal arterioles. In addition, we show that in response to the neuromodulator adenosine, the action potential frequency of cortical pyramidal neurons was significantly reduced in all groups. However, a significant decrease in adenosine-induced hyperpolarization was observed in BCAS 14 days. At the microvascular level, constriction-induced inhibition of pyramidal neurons was significantly compromised in BCAS mice. Collectively, these results suggest that BCAS uncouples vessel-to-neuron communication—vasculo-neuronal coupling—a potential early event in cognitive decline. The intracellular delivery of functional proteins in their native forms into cells is a theme of paramount importance in research owing to their diverse biological applications. Porous inorganic nanoparticles are emerging as efficient nanocarriers for the delivery of small molecules and drugs. To expand the range of cargos from small molecules to large native functional proteins, cubic mesoporous silica nanoparticles (cMSNs) with a Pm3n pore symmetry with an average particle dimension of 180 nm were prepared. The as-prepared cMSNs were subsequently etched with a methanolic solution of Ca(NO3)2 to expand their mesopores and simultaneously remove the template. The original mesopores with a pore dimension of 2.41 nm partially collapsed and combined into ultra-large mesopores with an average pore diameter of 13.89 nm without perturbing the original cubic symmetry of the remaining mesopores. The maximum pore dimension was around 60 nm. Various techniques including powder X-ray diffraction, transmission electron microscopy, and electron tomography identified the unique three-dimensional structure of pore-enlarged cMSNs (Ca-cMSNs). Moreover, their surfaces were functionalized with a guanidinium-rich cell-penetrating R8-azido-peptide (p-azidophenylalanine-GSGSGGRRRRRRRR) through the click reaction. The intracellular delivery of functional proteins such as Cre recombinase into human TE671(LoxP-LacZ) cells was realized by using R8-Ca-cMSNs as native protein delivery synthetic nanocarriers. The delivery efficiency when using the R8-Ca-cMSNs significantly enhanced compared to that when using Ca-cMSNs without surface-bound cell-penetrating peptides.

Hypertension is an important contributor to cognitive decline but the underlying mechanisms are unknown. Although much focus has been placed on the effect of hypertension on vascular function, less is understood of its effects on non-vascular cells. Because astrocytes and parenchymal arterioles (PA) form a functional unit (neurovascular unit), we tested the hypothesis that hypertension-induced changes in PA tone concomitantly increases astrocyte Ca(2+). We used cortical brain slices from eight week old mice to measure myogenic responses from pressurized and perfused PA. Chronic hypertension was induced in mice by 28-day angiotensin II (Ang II) infusion; PA resting tone and myogenic responses increased significantly. In addition, chronic hypertension significantly increased spontaneous Ca(2+) events within astrocyte microdomains (MD). Likewise, a significant increase in astrocyte Ca(2+) was observed during PA myogenic responses supporting enhanced vessel-to-astrocyte signaling. The transient potential receptor vanilloid 4 (TRPV4) channel, expressed in astrocyte processes in contact with blood vessels, namely endfeet, respond to hemodynamic stimuli such as increased pressure/flow. Supporting a role for TRPV4 channels in aberrant astrocyte Ca(2+) dynamics in hypertension, cortical astrocytes from hypertensive mice showed augmented TRPV4 channel expression, currents and Ca(2+) responses to the selective channel agonist GSK1016790A. In addition, pharmacological TRPV4 channel blockade or genetic deletion abrogated enhanced hypertension-induced increases in PA tone. Together, these data suggest chronic hypertension increases PA tone and Ca(2+) events within astrocytes MD. We conclude that aberrant Ca(2+) events in astrocyte constitute an early event towards the progression of cognitive decline.

An analysis method to examine the column shortening and shore safety of high-rise buildings from the construction period through the service life is developed. The effects of both the construction sequence and the inelastic behavior of concrete are considered in the analysis in three dimensions. The examination of safety during construction and the column shortening predictions are also discussed. Through a comparison of the analysis results and the measured data, it was found that the analysis results generally simulated the trends in the measured data well in all cases. Finally, the effects of the construction method on column shortening and shore safety were examined.

Six different types of Au nanostructures with rough surfaces were readily prepared through the redox reactions between Au precursor, AuCl4−, and furfuryl derivatives without extra metal surface capping ligands, in deionized water at room temperature. Furfuryl alcohol (FA) or furfurylamine (FFA) was used as a sole reducing agent for the reduction of Au precursor. Both FA and FFA effectively polymerized during the redox reactions to form polyfuran polymers. These polymers are thought to act as surface capping ligands during the formation of Au nanostructures. Experiments were conducted with three different concentrations of each furfuryl derivative. Interestingly, Au particles prepared from the reaction with varying concentration of FA or FFA showed large differences in size, and revealed that the higher the ratios of [FA]/[AuCl4−] or [FFA]/[AuCl4−], the smaller the size of Au particles. The size of Au particles was in the range of 1 μm to under 30 nm. Among these samples, two nanostructured Au particles, AuFA-4 and AuFFA-1, deposited on a Si wafer by a simple drop-casting method, were revealed as highly active surface-enhanced Raman scattering (SERS) substrates for the detection of methylene blue (MB) and crystal violet (CV). High SERS enhancement factors (EFs) of 106 ∼ 108 for MB and CV were observed. Small size Au nanoparticles (AuFFA-2 and AuFFA-4) were also found to be very active for the catalytic hydrogenation of 4-nitrophenol to 4-aminophenol in the presence of NaBH4 at room temperature. AuFFA-2 could be recycled eight times, without losing its activity.

An electromagnetic suspension system-type (EMS-type) magnetically levitated vehicle (Maglev) maintains the airgap between the guiderail and the electromagnet by controlling the electric current through the levitation controller and runs with the help of a linear induction motor. ECOBEE, an EMS-type Maglev, is designed for the purpose of urban transit and must run on a curved guideway with a small radius of curvature. However, while the EMS-type Maglev controls the vertical airgap using the levitation force of the electromagnet, in the case of the lateral direction, a guidance force, which is determined by the levitation force through the intensity of lateral displacement, passively controls the lateral displacement. However, when an excessive lateral displacement is triggered by a disturbance that is greater than the guidance force, an antiaberration skid comes into contact with the rail. Therefore, the Maglev running on a curve with a small radius should maintain the airgap without touching the rail when turning directions on a curved rail, while the vibrations of the vehicle caused by external factors, such as entering the circular curve from the transition curve, or a crosswind, can affect the levitation stability. Therefore, in order to secure the levitation stability of the electromagnet while also improving the curving performance of the vehicle, inserting a lateral damper between the cabin and the bogie has been suggested. In the present study, by using an integrated model of the Maglev system developed by multibody dynamics, the influence of a crosswind as the train runs along a curve and the effect of the lateral damper were analyzed.

Continuous cerebral blood flow is essential for neuronal survival, but whether vascular tone influences resting neuronal function is not known. Using a multidisciplinary approach in both rat and mice brain slices, we determined whether flow/pressure-evoked increases or decreases in parenchymal arteriole vascular tone, which result in arteriole constriction and dilation, respectively, altered resting cortical pyramidal neuron activity. We present evidence for intercellular communication in the brain involving a flow of information from vessel to astrocyte to neuron, a direction opposite to that of classic neurovascular coupling and referred to here as vasculo-neuronal coupling (VNC). Flow/pressure increases within parenchymal arterioles increased vascular tone and simultaneously decreased resting pyramidal neuron firing activity. On the other hand, flow/pressure decreases evoke parenchymal arteriole dilation and increased resting pyramidal neuron firing activity. InGLAST-CreERT2;R26-lsl-GCaMP3mice, we demonstrate that increased parenchymal arteriole tone significantly increased intracellular calcium in perivascular astrocyte processes, the onset of astrocyte calcium changes preceded the inhibition of cortical pyramidal neuronal firing activity. During increases in parenchymal arteriole tone, the pyramidal neuron response was unaffected by blockers of nitric oxide, GABAA, glutamate, or ecto-ATPase. However, VNC was abrogated by TRPV4 channel, GABAB, as well as an adenosine A1receptor blocker. Differently to pyramidal neuron responses, increases in flow/pressure within parenchymal arterioles increased the firing activity of a subtype of interneuron. Together, these data suggest that VNC is a complex constitutive active process that enables neurons to efficiently adjust their resting activity according to brain perfusion levels, thus safeguarding cellular homeostasis by preventing mismatches between energy supply and demand.SIGNIFICANCE STATEMENTWe present evidence for vessel-to-neuron communication in the brain slice defined here as vasculo-neuronal coupling. We showed that, in response to increases in parenchymal arteriole tone, astrocyte intracellular Ca2+increased and cortical neuronal activity decreased. On the other hand, decreasing parenchymal arteriole tone increased resting cortical pyramidal neuron activity. Vasculo-neuronal coupling was partly mediated by TRPV4 channels as genetic ablation, or pharmacological blockade impaired increased flow/pressure-evoked neuronal inhibition. Increased flow/pressure-evoked neuronal inhibition was blocked in the presence of adenosine A1 receptor and GABABreceptor blockade. Results provide evidence for the concept of vasculo-neuronal coupling and highlight the importance of understanding the interplay between basal CBF and resting neuronal activity.

In the literature related to maglev vehicles, most studies have primarily discussed the trade-off between a levitation system and guideway smoothness, such as deflection and surface roughness, and the optimization of a levitation controller. A dynamic model integrated with a kinematic vehicle, electromagnetic suspension system, guideway profile, and levitation controller model has been widely used for this purpose. However, in addition to guideway smoothness and stiffness, the normal force of a Linear induction motor (LIM) that is installed in the same bogie frame and drives the maglev vehicle can be regarded as a disturbance element affecting a levitation system. In addition, a linear model of an electromagnetic force is usually used in linear approximations around the nominal equilibrium point, in spite of its nonlinear characteristics. In this study, to analyze the interaction between electromagnetic and normal forces of the LIM, a dynamic model coupled with the normal force calculated from finite element analysis was developed. In addition, to comparatively analyze levitation stability by considering the linear and nonlinear characteristics of electromagnets, the non-linear electromagnetic forces measured in the experiment are coupled to the dynamic model. From the simulation results, it was confirmed that although a slight increase in current was observed with the LIM model when compared to without the LIM model, it produced no adverse effect on levitation stability. In simulation, considering the non-linear characteristics of an electromagnet, it was confirmed that the nominal current of the linear model converged to a higher value than the designed nominal current when the vehicle was standing still, whereas the nominal current of the non-linear was similar to the designed one.

In an attractive-type high-speed magnetic levitation (maglev) train, separate electromagnets are used for lateral guidance. For the guidance system, a U-shaped electromagnet that uses transverse flux has been designed and analyzed. Electromagnets counterbalance centrifugal forces during curving as well as under all operating conditions so that the clearances between on-board magnets and the guideway surface can be maintained within an allowable range. Because of the rigidity of the bogie, coupling effects exist among the magnets that are controlled independently. This study incorporates bogie yaw motion into control law for adjusting the voltages applied to magnets to provide constant clearance control. The effects of bogie yaw motion on guidance control performance are investigated experimentally using a full-scale vehicle.

In this paper, we aimed to develop a pulsed electromagnetic field (PEMF) system and to investigate its effects on diverse cell types. To ensure uniform magnetic flux density in the space for cell culture, the PEMF system was designed with an efficient solenoid using an analytical design and finite-element analysis. We investigated its effects on diverse cell types by using cell proliferation assay and flow cytometry analysis. The results of this paper show promise in furthering the PEMF-related biomedical research and its applications in translational studies.

The brain possesses two intricate mechanisms that fulfill its continuous metabolic needs: cerebral autoregulation, which ensures constant cerebral blood flow over a wide range of arterial pressures and functional hyperemia, which ensures rapid delivery of oxygen and glucose to active neurons. Over the past decade, a number of important studies have identified astrocytes as key intermediaries in neurovascular coupling (NVC), the mechanism by which active neurons signal blood vessels to change their diameter. Activity-dependent increases in astrocytic Ca(2+) activity are thought to contribute to the release of vasoactive substances that facilitate arteriole vasodilation. A number of vasoactive signals have been identified and their role on vessel caliber assessed both in vitro and in vivo. In this review, we discuss mechanisms implicating astrocytes in NVC-mediated vascular responses, limitations encountered as a result of the challenges in maintaining all the constituents of the neurovascular unit intact and deliberate current controversial findings disputing a main role for astrocytes in NVC. Finally, we briefly discuss the potential role of pericytes and microglia in NVC-mediated processes.

N-doped microporous carbon nanospheres (MCNs) are prepared directly from the as-prepared mesoporous silica nanospheres (MSNs) with a disordered pore structure by a new, simple and direct carbonization method. The as-prepared MSNs containing cetylpyridinium bromide (CTPB) template are effective for direct carbonization. Neither MSN with a two-dimensional hexagonal pore symmetry nor MSN with cetyltrimethylammonium bromide (CTAB) template is effective for this new method. Thus, this method is very cost-effective and environmentally benign. Furthermore, N-doping into the carbon matrix naturally occurrs during the carbonization process, because of the presence of pyridinium N atoms in the CTPB template. The N-doped MCN material is a good sorbent material for CO2 adsorption owing to the Lewis basic sites of N atoms as well as good microporosity. The low surface coverage isosteirc heat of CO2 adsorption (Qst) is calculated by the Clausius–Clapeyron equation using the data obtained at 273 and 298 K, and the calculated value of Qst is 58.4 kJ mol−1. This value is relatively very high compared to the other N-doped porous carbon materials. Both the Lewis basic sites of N atoms and uniform microporosity in the N-doped MCNs are responsible for this high value.

Rbfox3, an RNA-binding fox protein, binds to the antibody to pan-neuronal marker, neuronal nuclei (NeuN). Rbfox3 is expressed in neural tissues across a wide range of species including mammals, birds, and amphibians. However, the molecular identity of Rbfox3 in the zebrafish is largely unknown. In this study, we cloned two zebrafish Rbfox3 genes, Rbfox3a and Rbfox3b. We also cloned the Rbfox3-d31 isoform, which excludes a 93-nucleotide alternative exon within the RNA-recognition motif in both, Rbfox3a and Rbfox3b. Multiple protein sequence alignment revealed that the amino acid sequence for residues 1-20 of the zebrafish Rbfox3, which is the epitope region of NeuN antibody, was different from that of other species. Therefore, NeuN antibody lost its function as a neuronal marker antibody in zebrafish. Reverse transcriptase-polymerase chain reaction showed that both Rbfox3-d31 transcripts were abundant in the early blastula stage, after which they dramatically reduced, suggesting that these isoforms exist mainly as maternal transcripts. In contrast, full-length Rbfox3 transcripts were detected from the 24 h post-fertilization embryo, expression was also maintained at a constant level. Furthermore, full-length Rbfox3-expressing cells were located within the central nervous system during later stages of the zebrafish embryo. Our study provides insight into the molecular structure of zebrafish Rbfox3 as a step towards genetic association studies investigating the developmental role of Rbfox3.

Normally, Maglev (magnetic levitation) vehicles run on elevated guideways. The elevated guideway must satisfy various load conditions of the vehicle, and has to be designed to ensure ride quality, while ensuring that the levitation stability of the vehicle is not affected by the deflection of the guideway. However, because the elevated guideways of Maglev vehicles in South Korea and other countries fabricated so far have been based on over-conservative design criteria, the size of the structures has increased. Further, from the cost perspective, they are unfavourable when compared with other light rail transits such as monorail, rubber wheel, and steel wheel automatic guided transit. Therefore, a slender guideway that does have an adverse effect on the levitation stability of the vehicle is required through optimisation of design criteria.
