The intensification of urbanization has resulted in a significant increase in the generation of engineering waste sludge. Traditional treatment methods on engineering waste sludge often fail to achieve desired dewatering targets. Electroosmosis has garnered considerable attention for its substantial potential in soft ground treatment and sludge consolidation. Consequently, both domestic and international experts have studied rapid dewatering and reducing initial moisture content. However, there is still an incomplete understanding regarding the efficiency of electroosmosis. A series of experiments were performed under different potentials with Hangzhou engineering waste sludge, using ferrum and graphite electrodes. Electro-osmotic efficiency was analyzed in terms of total energy consumption, effective energy consumption, and electrode loss. The relationship between the effective energy coefficient and effective potential was explored based on the experimental results and theoretical models. It was found that total and effective energy consumptions increase with the potential. The rates of increase are larger with higher potentials. Total and effective electro-osmotic efficiency decreases with the increasing potentials. Electrode materials possess little influence on the electro-osmotic efficiency, but have indirect impact through effective potential due to the anode material loss. A linear relationship was obtained between the effective energy coefficient and effective potential. Low voltage is a disadvantage to the electro-osmotic effect, while high voltage is a disadvantage to electro-osmotic efficiency. Therefore, both the electro-osmotic effect and efficiency should be considered when determining the optimal voltage value in practice. The experimental results on the significant reduction of the energy consumption of electroosmosis can provide a scientific basis for the further promotion and application of electroosmotic technology in practical engineering projects.

Abstract Hepatocellular carcinoma (HCC) mainly depends on liver fibrosis/cirrhosis, which is regulated by tumor cells and the tumor microenvironment (TME), and is a crucial factor in tumor progression. This study aimed to identify abnormally expressed miR‐500a‐3p in the hepatitis‐cirrhosis‐HCC pathway and explored the roles of miR‐500a‐3p in HCC progression. A clinical cohort of patients with HCC is studied retrospectively. Subsequently, the role of miR‐500a‐3p transported by HCC exosomes in hepatic stellate cell (HSC) activation, hepatoma growth and invasion, and immune cell differentiation is determined by in vitro and in vivo experiments. In clinical tissues, miR‐500a‐3p is significantly enriched in HCC and cirrhosis tissues, and co‐expression of the immune marker CD4 or PD‐L1 significantly correlates with low survival rates in patients. Extracellular miR‐500a‐3p is taken up by HSC and PBMC, which promotes the secretion of the cytokines TGF‐β1 and IL‐10, increases PD‐L1 expression in HSC, and stabilizes PD‐1 expression in PBMC to affect the TME. Moreover, miR‐500a‐3p is associated with CD4+ T‐cell exhaustion and Treg differentiation and is significantly associated with increased tumorigenicity in in situ mouse HCC models. Mechanistically, HCC‐derived exosomal miR‐500a‐3p directly influences SOCS2 to regulate the JAK3/STAT5A/STAT5B signaling pathway. MiR‐500a‐3p promotes the growth and migration of HCC through the SOCS2/JAK3/STAT5A/STAT5B axis.

Abstract Background Rheumatoid arthritis (RA) is a prevalent inflammatory autoimmune disease characterised by persistent inflammation and joint damage with elevated levels of reactive oxygen species (ROS). Current treatment modalities for RA have significant limitations, including poor bioavailability, severe side effects, and inadequate targeting of inflamed joints. Herein, we synthesised cerium/manganese oxide nanoparticles (NPs) as efficient drug carriers with antioxidant and catalytic-like functions that can eliminate ROS to facilitate the polarization of macrophages phenotype from M1 to M2 and alleviate inflammation. Methotrexate (MTX), a first-line RA medication, was loaded into the NPs, which were further modified with bovine serum albumin (BSA) and integrated into dissolving hyaluronic acid-based microneedles (MNs) for transdermal delivery. Result This innovative approach significantly enhanced drug delivery efficiency, reduced RA inflammation, and successfully modulated macrophage polarization toward an anti-inflammatory phenotype. Conclusion This research not only presents a promising drug delivery strategy for RA but also contributes broadly to the field of immune disease treatment by offering an advanced approach for macrophage phenotypic reprogramming.

Objective: In this study, we aimed to determine whether proly4-hydroxylase-III (P4HA3) could be used as a biomarker for the diagnosis of colorectal cancer (CRC) as well as for determining prognosis. Methods: We used The Cancer Genome Atlas (TCGA) database to analyze P4HA3 expression in CRC and further investigated the association between P4HA3 and clinicopathological parameters, immune infiltration, and prognosis of patients with CRC. Enrichment analysis was conducted to investigate the potential biological role of P4HA3 in CRC. To verify the results of TCGA analysis, we performed immunohistochemical staining of 180 clinical CRC tissue samples to probe into the relationship of P4HA3 expression with lymphocyte infiltration and immune checkpoints expression. Results: The expression of P4HA3 was significantly higher in CRC tissues and associated with a higher degree of malignancy and poorer prognosis in CRC. The results of enrichment analysis indicated that P4HA3 may be associated with the epithelial-mesenchymal transition process and the immune response. Immunohistochemical staining results showed that high P4HA3 expression was associated with high infiltration levels of CD8+ and Foxp3+ TILs and high PD-1/PD- L1 expression. Lastly, patients with CRC co-expressing P4HA3 and PD-1 had a significantly worse prognosis. Conclusion: High expression of P4HA3 is associated with adverse clinical features and immune cell infiltration in CRC, and has the potential to serve as a biomarker for predicting CRC prognosis.

Simultaneous vibration isolation and energy harvesting (SVIEH) are becoming increasingly attractive in suspension systems. However, there is a conflict between low natural frequency and large loading capacity. In addition, the energy harvesting performance is always limited by maintaining the vibration isolation performance in most existing studies. Therefore, how to enhance vibration energy harvesting while satisfying vibration isolation is still a challenge. In this paper, a pair of magnetic rings is used to generate quasi-zero stiffness (QZS) for dealing with the conflict between large loading capacity and low natural frequency, and a Halbach magnetic ring is presented to enhance the energy harvesting performance. Based on them, a novel low-frequency QZS-based SVIEH (QZS-SVIEH) structure is proposed. Firstly, the structural condition of generating QZS is derived. Next the electro-mechanical coupling equations of the QZS-SVIEH system are built and solved by using the harmonic balance method (HBM), which is also verified by the Runge-Kutta method. Then the displacement transmissibility and output power are defined as the two performance indicators for vibration isolation and energy harvesting, respectively. The effects of electrical and mechanical parameters on the two performance indicators are studied in detail. Finally, the prototype of the proposed QZS-SVIEH structure is constructed for experimental testing. The experimental results show that the QZS-SVIEH can effectively suppress vibrations with frequencies above 7.2Hz and generate the peak power of 0.8W under the excitation of acceleration 1.5g and frequency 10Hz.

Abstract Background Infertility affects approximately 10–15% of reproductive-age men worldwide, and genetic causes play a role in one-third of cases. As a Bin-Amphiphysin-Rvs (BAR) domain protein, protein interacting with C-kinase 1 (PICK1) deficiency could lead to impairment of acrosome maturation. However, its effects on auxiliary germ cells such as Sertoli cells are unknown. Purpose The present work was aimed to use multi-omics analysis to research the effects of PICK1 deficiency on Sertoli cells and to identify effective biomarkers to distinguish fertile males from infertile males caused by PICK1 deficiency. Methods Whole-exome sequencing (WES) was performed on 20 infertility patients with oligozoospermia to identify pathogenic PICK1 mutations. Multi-omics analysis of a PICK1 knockout (KO) mouse model was utilized to identify pathogenic mechanism. Animal and cell function experiments of Sertoli cell-specific PICK1 KO mouse were performed to verify the functional impairment of Sertoli cells. Results Two loss-of-function deletion mutations c.358delA and c.364delA in PICK1 resulting in transcription loss of BAR functional domain were identified in infertility patients with a specific decrease in serum inhibin B, indicating functional impairment of Sertoli cells. Multi-omics analysis of PICK1 KO mouse illustrated that targeted genes of differentially expressed microRNAs and mRNAs are significantly enriched in the negative regulatory role in the vesicle trafficking pathway, while metabolomics analysis showed that the metabolism of amino acids, lipids, cofactors, vitamins, and endocrine factors changed. The phenotype of PICK1 KO mouse showed a reduction in testis volume, a decreased number of mature spermatozoa and impaired secretory function of Sertoli cells. In vitro experiments confirmed that the expression of growth factors secreted by Sertoli cells in PICK1 conditional KO mouse such as Bone morphogenetic protein 4 (BMP4) and Fibroblast growth factor 2 (FGF2) were decreased. Conclusions Our study attributed male infertility caused by PICK1 deficiency to impaired vesicle-related secretory function of Sertoli cells and identified a variety of significant candidate biomarkers for male infertility induced by PICK1 deficiency.

Wireless traffic prediction is essential to developing intelligent communication networks that facilitate efficient resource allocation. Along this line, decentralized wireless traffic prediction under the paradigm of federated learning is becoming increasingly significant. Compared to traditional centralized learning, federated learning satisfies network operators’ requirements for sensitive data protection and reduces the consumption of network resources. In this paper, we propose a novel communication-efficient federated learning framework, named FedCE, by developing a gradient compression scheme and an adaptive aggregation strategy for wireless traffic prediction. FedCE achieves gradient compression through top-K sparsification and can largely relieve the communication burdens between local clients and the central server, making it communication-efficient. An adaptive aggregation strategy is designed by quantifying the different contributions of local models to the global model, making FedCE aware of spatial dependencies among various local clients. We validate the effectiveness of FedCE on two real-world datasets. The results demonstrate that FedCE can improve prediction accuracy by approximately 27% with only 20% of communications in the baseline method.

Gastric cancer (GC) ranks as the third most prevalent malignancy and a leading cause of cancer-related mortality worldwide. However, the majority of patients with GC are diagnosed at an advanced stage, highlighting the urgent need for effective perioperative and postoperative chemotherapy to prevent relapse and metastasis. The current treatment strategies have limited overall efficacy because of intrinsic or acquired drug resistance. Recent evidence suggests that dysregulated long non-coding RNAs (lncRNAs) play a significant role in mediating drug resistance in GC. Therefore, there is an imperative to explore novel molecular mechanisms underlying drug resistance in order to overcome this challenging issue. With advancements in deep transcriptome sequencing technology, lncRNAs—once considered transcriptional noise—have garnered widespread attention as potential regulators of carcinogenesis, including tumor cell proliferation, metastasis, and sensitivity to chemo- or radiotherapy through multiple regulatory mechanisms. In light of these findings, we aim to review the mechanisms by which lncRNAs contribute to drug therapy resistance in GC with the goal of providing new insights and breakthroughs toward overcoming this formidable obstacle.

Abstract Glioma treatment remains a challenge owing to unsatisfactory targeted chemotherapy, where the blood–brain barrier (BBB) hinders the efficient uptake of therapeutics into the brain. Vasculogenic mimicry (VM) formed by invasive glioma cells negatively affects the treatment of glioma. Herein, we developed a targeted biomimetic drug delivery system comprising a doxorubicin-loaded platelet membrane-coated nanogel (DOX@PNGs). The nanogels provide great redox/pH dual responsiveness, while the platelet membrane (PLTM) promotes stability and circulation time. In vitro cellular uptake and in vivo imaging experiments demonstrated that the DOX@PNGs delivery system could penetrate the BBB, target gliomas, and destruct VM. DOX@PNGs increased drug penetration and prolonged mouse survival time during the treatment of orthotopic gliomas. These results indicate this biomimetic drug delivery system to be promising for glioma treatment and may be clinically translated in the future.

Purpose: The function played by cartilage intermediate layer protein 2 (CILP2) between colorectal cancer (CRC) progression and immune response remains unclear, especially with respect to immune cell infiltration and checkpoints. Materials and Methods: We examined CILP2 expression in The Cancer Genome Atlas (TCGA) COAD-READ cohort and analyzed its relationship with clinicopathological features, mutations, survival, and immunity. Gene ontology, Kyoto Encyclopedia of Genes and Genomes pathway analysis, and gene set enrichment analyses (GSEA) were performed to determine CILP2 related pathways. To further investigate the results of TCGA analysis, validation was performed using CRC cell lines, fresh pathological tissues, and a CRC tissue microarray (TMA). Results: In both TCGA and TMA cohorts, CILP2 expression was increased in CRC tissues and was associated with patient T stage (T3 and T4), N stage (N1), pathological stage (III and IV), and overall survival. Immune cell infiltration and checkpoint analysis revealed that CILP2 expression is highly correlated with multiple immune marker genes, including PD-1. In addition, results of enrichment analysis indicated that CILP2 related genes was mainly enriched in extracellular matrix related functions. Conclusion: Elevated CILP2 expression is associated with adverse CRC clinical features and immune cells, it has potential as a biomarker detrimental to CRC survival.

Objective: Glioblastoma is one of the most common intracranial malignant tumors with an unfavorable prognosis, and iron metabolism as well as ferroptosis are implicated in the pathogenesis of glioblastoma. The present study aims to decipher the role and mechanisms of tripartite motif-containing protein 7 (TRIM7) in ferroptosis and glioblastoma progression. Methods: Stable TRIM7-deficient or overexpressing human glioblastoma cells were generated with lentiviral vectors, and cell survival, lipid peroxidation and iron metabolism were evaluated. Immunoprecipitation, protein degradation and ubiquitination assays were performed to demonstrate the regulation of TRIM7 on its candidate proteins. Results: TRIM7 expression was elevated in human glioblastoma cells and tissues. TRIM7 silence suppressed growth and induced death, while TRIM7 overexpression facilitated growth and inhibited death of human glioblastoma cells. Meanwhile, TRIM7-silenced cells exhibited increased iron accumulation, lipid peroxidation and ferroptosis, which were significantly reduced by TRIM7 overexpression. Mechanistically, TRIM7 directly bound to and ubiquitinated nuclear receptor coactivator 4 (NCOA4) using K48-linked chains, thereby reducing NCOA4-mediated ferritinophagy and ferroptosis of human glioblastoma cells. Moreover, we found that TRIM7 deletion sensitized human glioblastoma cells to temozolomide therapy. Conclusion: We for the first time demonstrate that TRIM7 modulates NCOA4-mediated ferritinophagy and ferroptosis in glioblastoma cells, and our findings provide a novel insight into the progression and treatment for human glioblastoma.

Pancreatic adenocarcinoma (PAAD) is one of the most common malignant tumors with poor prognosis worldwide. Mounting evidence suggests that the expression of lncRNAs and the infiltration of immune cells have prognostic value for patients with PAAD. We used Gene Expression Omnibus (GEO) database and identified six genes (COL1A2, ITGA2, ITGB6, LAMA3, LAMB3, and LAMC2) that could affect the survival rate of pancreatic adenocarcinoma patients. Based on a series of in silico analyses for reverse prediction of target genes associated with the prognosis of PAAD, a ceRNA network of mRNA (COL1A2, ITGA2, LAMA3, LAMB3, and LAMC2)–microRNA (miR-15a-5p)–long non-coding RNA (LINC00511, LINC01578, PVT1, and TNFRSF14-AS1) was constructed. We used the algorithm “CIBERSORT” to assess the proportion of immune cells and found three overall survival (OS)–associated immune cells (monocytes, M1 macrophages, and resting mast cell). Moreover, the OS-associated gene level was significantly positively associated with immune checkpoint expression and biomarkers of immune cells. In summary, our results clarified that ncRNA-mediated upregulation of OS-associated genes and tumor-infiltration immune cells (monocytes, M1 macrophages M1, and resting mast cell resting) correlated with poor prognosis in PAAD.

Ischemic stroke remains the leading cause of death and disability, while the main mechanisms of dominant neurological damage in stroke contain excitotoxicity, oxidative stress, and inflammation. The clinical application of many neuroprotective agents is limited mainly due to their inability to cross the blood-brain barrier (BBB), short half-life and low bioavailability. These disadvantages can be better eliminated/reduced by nanoparticle as the carrier of these drugs. This review expounded the currently hot researched nanomedicines from the perspective of the mechanism of ischemic stroke. In addition, this review describes the bionic nanomedicine delivery strategies containing cells, cell membrane vesicles and exosomes that can effectively avoid the risk of clearance by the reticuloendothelial system. The potential challenges and application prospect for clinical translation of these delivery platforms were also discussed.

A partial pre-rusted wire beam electrode (WBE) was designed to study the influence of the rust layer on rebar corrosion in the carbonated simulated concrete pore solution (SCPS). The results show that the passive film generated on the pre-rusted steel area is more fragile than that formed on the fine polished steel area in carbonaceous media. Nevertheless, the pitting corrosion resulting from the presence of chloride ions still tends to occur on the fine polished steel surface due to the local acidification process being hindered by the rust layer. The rust layer could play a more important role than the passive film in inhibiting the initiation of chloride-induced corrosion on rebar. The expansion path of the corrosion product would be blocked by the rust layer, leading to the pit propagating in the fine polished region. Furthermore, the growth of pitting corrosion is greatly accelerated due to the catalytic cathodic reaction of the rust layer.

On the Chongqing’s straddle-type monorail train in China, serious abnormal wear of running wheel tires occurs in its routine use, leading to a series of safety, environmental, and economic concerns. To analyze the causes of abnormal wear, a dynamic model of straddle-type monorail vehicle with full consideration to the lateral behavior of running wheel tire was established using Lagrange equations and verified by Zhong’s model created using SIMPACK. Based on the boundary conditions provided in the dynamic model and the friction characteristics of tire and pavement, a finite element model of the running wheel tire was built and validated with vertical, lateral, and longitudinal stiffness tests of corresponding tires. The wear situations of each running wheel tire under extreme turning condition were described in detail with the contact behavior and an assessment was done based on the friction work and the friction work skewness value. Besides, the influence of slip angle, roll angle, and their combined effect on the abnormal wear of running wheel tire was analyzed. This research proposes that, reasonable matching of the slip angle and the roll angle is of engineering significance and it can reduce uneven wear of the running wheel tire.

In this paper, a new kind of carbon steel (CS) and stainless steel (SS) galvanic sensor system was developed for the study of rebar corrosion in different pore solution conditions. Through the special design of the CS and SS electronic coupons, the electronic resistance (ER) method and zero resistance ammeter (ZRA) technique were used simultaneously for the measurement of both the galvanic current and the corrosion depth. The corrosion processes in different solution conditions were also studied by linear polarization resistance (LPR) and the measurements of polarization curves. The test result shows that the galvanic current noise can provide detailed information of the corrosion processes. When localized corrosion occurs, the corrosion rate measured by the ER method is lower than the real corrosion rate. However, the value measured by the LPR method is higher than the real corrosion rate. The galvanic current and the corrosion current measured by the LPR method shows linear correlation in chloride-containing saturated Ca(OH)2 solution. The relationship between the corrosion current differences measured by the CS electronic coupons and the galvanic current between the CS and SS electronic coupons can also be used to evaluate the localized corrosion in reinforced concrete.

Abstract The Yadong trout (Salmo trutta), a species endemic to the Yatung River in Tibet, China, was classified as a second-class protected species in the 20th century. Now, it is considered one of the most important fishery resources in China. In this study, we assembled a near-complete genome of the S. trutta, integrating PacBio HiFi, Hi-C, and ONT sequencing technologies. The genome assembly spans 2.49 Gb, with 96.87% of the sequence anchored onto 40 chromosomes. In this assembly, a total of 12 chromosomes were assembled to a gap-free level, with 8 of them reaching the telomere-to-telomere level. The completeness of this assembly was assessed at 99.50% by BUSCO, containing approximately 63.24% repetitive sequences, and predicted to encode 41,782 protein-coding genes. This is the first near-complete genome assembly of the S. trutta, providing an essential resource for molecular breeding and germplasm conservation of this important species.

Abstract Lymphocyte receptors independently evolved in both jawed and jawless vertebrates with similar adaptive immune responses. However, the diversity of functional subtypes and molecular architecture in jawless vertebrate lymphocytes, comparable to jawed species, is not well defined. Here, we profile the gills, intestines, and blood of the lamprey, Lampetra morii, with single-cell RNA sequencing, using a full-length transcriptome as a reference. Our findings reveal higher tissue-specific heterogeneity among T-like cells in contrast to B-like cells. Notably, we identify a unique T-like cell subtype expressing a homolog of the nonlymphoid hematopoietic growth factor receptor, MPL-like (MPL-L). These MPL-L+ T-like cells exhibit features distinct from T cells of jawed vertebrates, particularly in their elevated expression of hematopoietic genes. We further discovered that MPL-L+ VLRA+ T-like cells are widely present in the typhlosole, gill, liver, kidney, and skin of lamprey and they proliferate in response to both a T cell mitogen and recombinant human thrombopoietin. These findings provide new insights into the adaptive immune response in jawless vertebrates, shedding new light on the evolution of adaptive immunity.

Abstract Elucidating cellular architecture and cell-type evolution across species is central to understanding immune system function and susceptibility to disease. Adaptive immunity is a shared trait of the common ancestor of cartilaginous and bony fishes. However, evolutionary features of lymphocytes in these two jawed vertebrates remain unclear. Here, we present a single-cell RNA sequencing atlas of immune cells from cartilaginous (white-spotted bamboo shark) and bony (zebrafish and Chinese tongue sole) fishes. Cross-species comparisons show that the same cell types across different species exhibit similar transcriptional profiles. In the bamboo shark, we identify a phagocytic B cell population expressing several pattern recognition receptors, as well as a T cell sub-cluster co-expressing both T and B cell markers. In contrast to a division by function in the bony fishes, we show close linkage and poor functional specialization among lymphocytes in the cartilaginous fish. Our cross-species single-cell comparison presents a resource for uncovering the origin and evolution of the gnathostome immune system.
