Internal tandem duplication is the most common form of mutation in FMS-like tyrosine kinase 3 (FLT3) in different haematological malignancies, highlighting in acute myeloid leukaemia (AML) and is associated with increased risk of relapse and reduced overall survival. A major breakthrough in the treatment of FLT3-mutated AML has been achieved through the use of highly selective FLT3 tyrosine kinase inhibitors, both in monotherapy and in combination with standard intensive cytotoxic chemotherapy. The desire to improve the outcomes of patients with AML, including those with relapse and refractory disease, has led to attempts to use non-standard therapeutic options. Enhancement of the antileukemic effects of the second-generation FLT3 inhibitor Gilteritinib may be achieved through synergy with the hypomethylating agent 5-azacytidine and the selective Bcl-2 inhibitor Venetoclax. Thus, targeted triple therapy is a promising option in the treatment of patients with FLT3-mutated AML. This study sought to evaluate the effectiveness of “triple therapy” regimen in 4 patients with relapsed/refractory FLT3 mutated AML. We found that the use of this combination showed rapid response with good safety and frequently allowed subsequent transplant and achieve durable clinical benefit.

Abstract Developments in missile proliferation are analyzed, including the main drivers, trends, and actors. The increasing complexity of the global missile proliferation landscape is shown. They key areas of technological development are identified, as well as several types of missile proliferators, to substantiate the importance of missile proliferation in the context of international security. Existing and prospective mechanisms are discussed with respect to arms control, nonproliferation, and multilateral risk reduction measures. It is stressed that the existing mechanisms are aimed primarily at reducing the risks associated with creating delivery vehicles for weapons of mass destruction whereas the greatest importance is now attached to the potential use of conventionally armed missiles. It is proposed to concentrate efforts on “soft arms control” in order to lay a foundation for a gradual increase in controllability in the absence of real prospects for verifiable and intrusive multilateral measures.

Dysfunction of genes that control mitosis and are responsible for the correct segregation of sister chromatids in anaphase is often accompanied by aneuploidy, which is frequently detected in leukemia. One of the components of the kinetochore complex, namely, the AF15q14/KNL1/CASC5 protein, is an important factor ensuring the correct binding of the pericentromeric region of chromosomes with the spindle microtubules. As shown recently, in some leukemias, the gene of this protein can be involved in the generation of the chromosomal translocation t(11;15)(q23;q14) or a variant of the chimeric MLL-AF15Q14 oncogene, which serves as a biomarker of poor prognosis. Despite the implication of mRNA of the CASC5 gene in oncogenesis of solid tumors, expression of this gene in hematopoietic neoplasms has not been studied. We analyzed expression levels of the CASC5 gene and the nearest regulatory genes, including WT1, APOBEC3A (A3A), and N-MYC. A pronounced decrease in CASC5 expression in bone marrow cells of primary leukemia patients compared with healthy donors was found. The effectiveness of the use of “direct” sowing in the cultivation of grain crops in the north-east of the Non-Chernozem zone of the Russian Federation has been revealed. Field experiments were carried out in 2019-2021 on medium loamy low-humus gray forest soils with a high content of mobile phosphorus and exchangeable potassium. The weather conditions during the years of the experiments differed both in terms of heat supply and the amount of precipitation. The objects of the study were: winter wheat of the Moskovskaya 39 variety, spring wheat – Arhat, malting barley – Elf, fodder barley – Pamyaty Rodinoy, corn for silage and grain hybrid NK Gitago. “Direct” sowing was carried out using the Primera DMC (Amazone) sowing complex with the simultaneous application of mineral fertilizers for the maximum planned yield of each crop. On average, the maximum (3.43 t/ha) grain yield when using “direct” sowing was formed when cultivating corn, and the minimum (1.86 t/ha) – malting barley. In the production of these crops, the highest (2.42) and the lowest (1.85) energy efficiency coefficient, respectively, was also revealed. An analysis of economic indicators testified to the expediency of using “direct” sowing in the cultivation of winter wheat, which was reflected in obtaining the maximum (73.3%) level of profitability. The minimum (31.5%) level of profitability was noted in the cultivation of malting barley, which is explained by the high degree of material and labor costs and its relatively low yield. The profitability of the production of other types of crop products was 1.18-1.94 times lower. In general, the agronomic, energy and economic assessment showed bright prospects for the cultivation of winter wheat and maize for grain using the “direct” seeding method after potatoes.

The features of improving self-assembled films based on different-sized gold particles were investigated in this work. A study of the SERS signal was carried out to compare SPF samples consisting of gold nanoparticles of different sizes. For this purpose, colloids with different particle sizes were synthesized. These are spherical particles 20 nm and 40 nm in diameter and lamellar particles with an approximate size of 60x40x15 nm. Optimization of the concentration of the self-assembly agent of the reaction mixture in the preparation of the self-assembled plasmon film (SPF) has been carried out. The concentration of the self-collecting agent in the organic phase, at which the maximum values of the SERS enhancement of the signal were observed, is 10−5 M. The showing the reproducibility of the SERS signal for samples with the maximum enhancing of the SERS signal value is close for the all the studied particle sizes consisted SPF.

At present, cytogenetic and PCR-based techniques are generally used in the diagnosis of leukemia. Due to high accuracy, specificity, and sensitivity of PCR assays, they have the advantage over traditional cytogenetic tools. As a rule, the classical real time PCR is carried out in a 25-μL reaction mixture. It requires a large volume of each reagent and takes a long time to finish the test. The molecular genetic assay presented here is microchip-based real-time PCR that is optimized for simultaneous analysis of 15 oncogene mutations and one housekeeping gene abl. Moreover, this diagnostic tool requires a minimal amount of cDNA and PCR reagents (10-fold less) and a short time (2-fold less) for quantitative estimation of more than five copies of gene-target. Thus, microchip-based PCR analysis can improve the detection of oncogene mutations in leukemia patients and may be used for both early diagnostics and long-term monitoring of leukemia.

Genetic diagnostics is widely used for detection of risk factors of hereditary thrombophilias caused by molecular defects in the coagulation system. The hereditary thrombophilias are frequently associated with higher incidences of point mutations in hemostasis (F2 20210G>A, F5 1691G>A) and folate metabolism (MTHFR 677C>T, MTHFR 1298A>C) genes. Combinations of gene abnormalities in F2 and/or MTHFR with Leiden mutation (F5 1691G>A) significantly increase risk of thrombosis. Thus, simultaneous analysis of allele polymorphism of these genes is of clinical importance. This study has demonstrated high efficiency of microchip-based multiplex real time PCR for analysis of allele specific polymorphism in hemostasis and folate metabolism genes. Using this test it is possible to analyze polymorphism of the three genes (four point mutations) in a short time; it requires a minimal quantity of DNA template and PCR reagents including DNA polymerase, and thus can be recommended for clinical laboratory diagnostics.

The paper considers an alternative to traditional methods of tooth machining for worm, spiroid and bevel gearwheels – by means of an assembled multi-tooth running-in cutter head with hard-alloy inserts. Within the cutting process the cutter head is rotated and fed along its axis concurrently with the rotation of the workpiece, thus implementing the generating worm with a continuous or variable pitch. It is preferable to make the multi-thread generating worm with regard to increasing the production efficiency and valid control of the level of contact localization. The design scheme of setting parameters is considered on the basis of providing the conjugacy at two design points assigned at opposite tooth flanks. The design scheme of the generated tooth geometry is also discussed. Design examples are given to show the possibility of providing the necessary degree of contact localization. The method is implemented at common and simply set metal cutting machine tools – universal gear hobbing machines and machining centres. As compared to traditional methods, the proposed method provides the effective control of tooth geometry, reduction of time and cost of production preparation, especially in the absence of the specialized equipment, staff and corresponding techniques at the production site.

In present research the comparative analysis of donor chimerism (DC) using different tests was performed to improve the diagnostic tool in patients with malignant hematological disorders after allo-HSCT. The RBC antigen typing, identification of ABO blood type and quantitative analysis of InDel-, STR-, Y-polymorphisms were carried out for detection of DC. In addition, the expression of well-known oncogenes and CD-markers for monitoring MRD was evaluated to predict relapse and clinical outcome. According to our research, the analysis of InDel polymorphism using AlleleSEQR-PCR is more sensitive test for estimation of DC as compared with other assays. Moreover, the sensitivity of AlleleSEQR-PCR may be increased after isolation of the CD34 cell population in bone marrow. Nevertheless, observation of high levels in DC (³95%) in some leukemia patients (ALL, Ph+, bcr-abl/p190+) during first 6 months after HSCT cannot exclude the possibility of relapse. Thus, the combined monitoring of both DC (InDel) and MRD (oncogenes, WT1 and CD-markers) is a more advisable and useful test in managing hematologic malignancies and predicting relapse risk after allo-HSCT.S tsel'iu uluchsheniia diagnostiki u onkogematologicheskikh patsientov v posttransplantatsionnyĭ period provodili kompleksnoe issledovanie biomarkerov-misheneĭ donorskogo khimerizma (DKh) s ispol'zovaniem neskol'kikh testov. The purpose of the study is to develop basic approaches to digital models of cross-cutting production of frame and casing parts in order to forecast stress-strain behavior and change of a part form at all production stages. The proposed method covers stage-by-stage modeling of part production including machining, shaping (levelling) and face hardening using mechanical characteristics of a material, technological residual stress and initial shape. This approach in combination with equipment under control will allow optimizing machining conditions and achieving the required accuracy of parts and maximum capacity without pilot-scale works. The use of developed technique will make it possible to increase the production efficiency of parts due to time minimization of finishing operations and cost reduction for preproduction for the launch of new products.

Обработка винтовых поверхностей резьб и червяков широко распространена на машиностроительных предприятиях. Получение профиля обусловливается требованиями к локализации первоначального прилегания сопрягаемых поверхностей (обычно в центральной части высоты витка) и подразумевает применение инструментов управления геометрией профиля при нарезании витков. И если в конструкциях традиционных резьбошлифовальных и червячношлифовальных станков имеются механизмы точной настройки углов и формы профиля формируемых витков, то при лезвийной обработке витков их профиль традиционно жестко связан с профилем инструмента, что делает коррекцию геометрии затруднительной. Рассмотрены возможности такой коррекции при лезвийной обработке инструментами, оснащенными твердосплавными неперетачиваемыми пластинами, что обеспечивает и требуемую геометрию витков, и высокую производительность обработки. В частности, рассмотрены схемы обработки на широко распространенных токарно-винторезных станках с ЧПУ круглой (скругленной) и прямолинейной режущими кромками сборного резца, а также сборной торцовой резцовой головкой, что предполагает сравнительно простую реализацию на большинстве машиностроительных предприятий. В качестве инструментов управления параметров предлагаются линейные смещения инструмента поперек и вдоль оси центров станка (для схем обработки прямой режущей кромкой и торцовой резцовой головкой) и параметры траектории подачи инструмента (для схемы с круглой (или скругленной) пластиной). Приведены численные примеры, показывающие возможности и эффективность управления геометрией профиля, и результаты нарезания опытных образцов червяков. Определены основные требования к монтажу твердосплавных пластин, связанные с обеспечением точности позиционирования, прочностью и жесткостью крепления пластин. Применение предложенных приемов в ряде случаев позволит отказаться от дорогостоящего шлифования витков, используя современные возможности токарно-винторезных станков с ЧПУ с обеспечением высокой точности и высокой производительности обработки деталей с винтовыми поверхностями. Перспективы методов связаны с оптимизацией управляющих параметров и применением их для обработки термоупрочненных деталей с винтовыми поверхностями.

Leukemia is a clonal proliferative disorder of a multipotent hematopoietic stem cell that leads to abnormal cell growth and/or differentiation. The hallmark of the disease is the presence of oncogene expression in bone marrow or peripheral blood resulting from chromosome translocations. The development of leukemia and the progression of the disease are multistage processes implicated in a series of molecular changes preceeded chromosomal instability and aneuploidy. The most likely of the above-mentioned changes include the following: the appearance of additional chromosome translocations, the activation of other oncogenes not expressed previously, the loss of tumor suppressor genes, abnormal centrosome duplication, and the dysfunction of genes that coordinate accurate chromosome alignment and chromosome segregation during mitosis. The last two molecular events controlled by mitotic spindle checkpoint genes play a role in leukemogenesis and are probably involved in apoptosis.

Although c-Abl and D40 proteins are located predominantly in the nucleus, they are involved in different cellular processes. c-Abl is a tyrosine-kinase that takes part in protein phosphorylation at tyrosine. Recently, D40 has been identified as a component of the outer kinetochore complex. Despite the functional differences between c-Abl and D40 proteins, they do have some similarities. First, high expression levels of c-Abl and D40 were observed not only in proliferating somatic cells, such as tumors, but also in healthy human testis. The increased expression levels of c-Abl and D40 protein in spermatocytes and acrosome of spermatids indicate their role in meiosis and spermatogenesis. Second, both proteins interact with specific regions of chromatin and are involved in the regulation of cell growth and division. Third, ABL and D40 (AF15q14) genes are involved in chromosomal translocations that subsequently form chimeric oncoproteins BCR-ABL, TEL-ABL, and MLL-AF15q14 in human leukemia. Lastly, both proteins interact with the tumor suppressor pRb protein and, consequently, can lead to the regulation of cell proliferation. The possible regulatory pathways that are controlled by c-Abl and D40 proteins are described in detail.

Жесткая конкуренция со стороны ведущих зарубежных производителей побуждает искать новые решения, казалось бы, в досконально изученной теме улучшения технико-экономических характеристик опор скольжения. Одним из лучших решений, применяемых в низкоскоростных тяжелонагруженных опорах, де-факто стало применение металлофторопластового листа (с нанесением порошковой бронзы, инфильтрованной фторопластом). В статье рассмотрены главные вопросы разработки альтернативного материала указанных опор путем создания на поверхности стального листа прочного антифрикционного состава на основе баббита, дисульфида молибдена и графита. В частности, рассмотрены вопросы подбора материала, лазерной обработки, сравнительного экспериментального исследования и отработки серийного производства опор скольжения, практически не уступающих по антифрикционным свойствам и долговечности лучшим зарубежным образцам, но отличающихся резко сниженной стоимостью.

Рассмотрен вариант практической реализации метода предварительной обработки витков червяка на токарном станке с ЧПУ с применением многорезцового режущего инструмента чашечного типа, оснащенного стандартными сменными твердосплавными пластинами. Дается описание схемы модернизации токарного станка и конструкции стандартной чистовой двусторонней торцовой головки для нарезания колес с круговым зубом. Сформулированы основные требования к модификации токарного станка с ЧПУ и зуборезной резцовой головки. Представлены главные зависимости для расчета параметров формообразования и методы расчета параметров станочных наладок. Также отражаются результаты предварительного нарезания витков червяка данным методом.

The methods for extraction of nucleic acids from the fungi and clinical materials containing various microorganisms have currently modified and make it possible to extract DNA with its highest yield and to minimize its destruction. In particular, this concerns mycelial fungi that have a rigid cell wall. Addition of glass microbeads to the lysed sample, followed by mixture stirring, gives rise to undamaged DNA from fungi and their spores. Extracted DNA is suitable for the identification of micromycetes by qualitative or quantitative polymerase chain reaction (PCR), real-time PCR-based hybridization, and LightCycler PCR and for the polymorphism analysis of the spacer regions of ribosomal genes by the sequencing technique. A combination of quantitative PCR and sequencing permits co-determination of the number of gene target copies in 9 different micromycetes in the clinical material being examined, which is of great importance for the diagnosis and treatment of patients with higher immunosuppression and, of those after transplantation of organs and tissues in particular.

Chronic myeloid leukemia (CML) is a hemopoietic condition caused by chromosomal translocation t(9;22)(q34;q11) or bcr-abl fusion gene. The predominant variants of bcr-abl oncogene rearrangement are b3a2 and b2a2. The present study evaluated the efficacy of interferon-a therapy of CML patients and molecular prognostic factors. Cytogenetic response and complete hematological remission were more frequent in CML b3a2 treatment with interferon-a. Moreover, after therapy, chronic phase lasted in that group (p = 0.026) much longer. Overall survival in the group was significantly longer, too (p = 0.046).

The investigation deals with a simplified modification or molecular-genetic detection of translocation t(9;22) using a combination of reverse transcription and polymerase chain reactions (RT-PCR). Unlike the available protocols, analysis is carried out using one enzyme--TET-Z polymerase--(instead of two) which has both revertase and DNA-polymerase activities. The present modification is highly sensitive, less time-consuming and cheaper. The method has proved useful for both diagnosing t(9;22) translocation and diagnosing and monitoring minimal residual disease remaining after marrow transplantation.

The yield of UV-induced DNA double-strand breaks was studied for white blood cells ("light" fraction) derived from peripheral blood, and from patients with lymphomas, chronic lymphoid leukemia (CLL), and chronic myeloid leukemia (CML). The method employed was constant-field electrophoresis of plug-embedded DNA in agarose gel. Characteristic dose-response curves were obtained for various cell populations. Lymphoid cells, both from healthy subjects and CLL patients, revealed less damage to DNA under UV-irradiation, whereas CML cells were much more affected. Possible interpretation of these results includes species-specific differences in UV-induced DNA damage, as well as sufficient DNA crosslinking, thus interfering with DNA dsbs detection in irradiated cells.

A neural expert network system for diagnosis and prognosis of cardiovascular diseases was developed. Computer experiments using the system revealed that the current medical practice of recommending hypocholesteric diet, avoidance of pernicious habits, limitation in consumption of coffee and alcohol, sliming, and reduced physical and mental load to all cardiologic patients is not universally correct. It is demonstrated that some of these recommendations can even prove harmful to some patients. The diagnostic system suggested in this work allows such atypical patients to be recognized and personal recommendations for such patients to be compiled.
