1. Enucleated L929 mouse fibroblasts support invasion and multiplication of Shigella flexneri 5a

Abstract

Invasive bacteria can induce their own uptake and specify their intracellular localization; hence it is commonly assumed that proximate modulation of host cell transcription is not required for infection. However, bacteria can also modulate, directly or indirectly, the transcription of many host cell genes, whose role in the infection may be difficult to determine by global gene expression. Is the host cell nucleus proximately required for intracellular infection and, if so, for which pathogens and at what stages of infection? Enucleated cells were previously infected with Toxoplasma gondii, Chlamydia psittaci, C. trachomatis, or Rickettsia prowazekii. We enucleated L929 mouse fibroblasts by centrifugation in the presence of cytochalasin B, and compared the infection with Shigella flexneri M90T 5a of nucleated and enucleated cells. Percent infection and bacterial loads were estimated with a gentamicin suppression assay in cultures fixed and stained at different times after infection. Enucleation reduced by about half the percent of infected cells, a finding that may reflect the reduced endocytic ability of L929 cytoplasts. However, average numbers of bacteria and frequency distributions of bacterial numbers per cell at different times were similar in enucleated and nucleated cells. Bacteria with actin-rich tails were detected in both cytoplasts and nucleated cells. Lastly, cytoplasts were similarly infected 2 and 24 h after enucleation, suggesting that short-lived mRNAs were not involved in the infection. Productive S. flexneri infection could thus take place in cells unable to modulate gene transcription, RNA processing, or nucleus-dependent signaling cascades.

Abstract

The RNA exosome is a multisubunit protein complex involved in RNA surveillance of all classes of RNA, and is essential for pre-rRNA processing. The exosome is conserved throughout evolution, present in archaea and eukaryotes from yeast to humans, where it localizes to the nucleus and cytoplasm. The catalytically active subunit Rrp44/Dis3 of the exosome in budding yeast (Saccharomyces cerevisiae) is considered a protein present in these two subcellular compartments, and here we report that it not only localizes mainly to the nucleus, but is concentrated in the nucleolus, where the early pre-rRNA processing reactions take place. Moreover, we show by confocal microscopy analysis that the core exosome subunits Rrp41 and Rrp43 also localize largely to the nucleus and strongly accumulate in the nucleolus. These results shown here shed additional light on the localization of the yeast exosome and have implications regarding the main function of this RNase complex, which seems to be primarily in early pre-rRNA processing and surveillance.

Abstract

This study aimed to partially purify and characterise the bacteriocin produced by Leuconostoc mesenteroides A11 and to evaluate the bacteriocin production in whey at different concentrations and temperatures (30 and 25 °C). The partially purified bacteriocin was obtained by chromatography and analysed by electrophoresis. Thus, the molecular weight of the bacteriocin was estimated by electrophoresis, ranging from 3.5 to 8.5 kDa. Leu conostoc mesenteroides A11 produced bacteriocins in all concentrations of whey and temperatures evaluated, indicating that it would be feasible to employ low whey concentrations and room temperature for the production of bacteriocins, thereby reducing processing costs.

23. Recrystallization behaviour of IF steel at the interface of Al junction

Abstract

The recrystallization behavior of an IF steel was investigated at the interface with aluminum junction, using cold-rolled IF steel sheets with high recrystallization temperature. In the region surrounded by the tongue-like η phase, recrystallization of the steel was fast at first glance, and the equiaxed structure was formed, but actually, it had a subgrain structure. The η phase grew preferentially to the c axis, and the growth of the η-Fe2Al5 phase distorted the surrounding iron due to the difference in the molar volume; compressive deformation was expected in the a-axis and b-axis directions, and tensile deformation was expected in the c-axis. The growth of η phase obviously changed the recrystallization behavior of the IF steel. The development of γ-fiber crystalline texture (/ND), which was a typical recrystallization texture of IF steel, was not observed in the region surrounded by η phase.

Abstract

Background The use of biological disease modifying anti-rheumatic drugs (bDMARDs) or synthetic DMARDs for the treatment of rheumatoid arthritis (RA) has improved the management of RA and prevented patients with RA from joint destruction. However, there are some cases in which it is difficult to continue the treatment with these medicines due to insufficient therapeutic effect or complications. In Japan, 8 of bDMARDs and 2 of synthetic DMARDs can be used for the treatment of RA. Among them, infliximab (INF) and etanercept (ETN) were applied for RA treatment in 2003 and 2005, respectively, and have been utilised for more than 10 years. Although adalimumab (ADA) and tocilizumab (TCZ) were applied in 2008, RA treatment by ADA and TCZ had been conducted by clinical trials since 2003, and they have also been utilised for more than 10 years. Objectives In this study, we evaluated the retention rate of these 4 bDMARDs (INF, ETN, ADA, and TCZ) for over 10 years. Methods RA patients who started the treatment with INF, ETN, ADA, or TCZ before 2007 in Matsubara mayflower hospital and were observed for over 10 years were included in this study. INF was used in the treatment for 111 patients with RA, and 95 out of them were observed for over 10 years (INF group). 95 out of 119 RA patients treated with ETN (ETN group), 24 out of 27 RA patients treated with ADA (ADA group), and 21 out of 25 RA patients treated with TCZ (TCZ group) were included in this study. Kaplan-Meier analysis was applied to determine the differences in the retention rate of these bDMARDs. Results Kaplan-Meier analysis showed no statistically significant difference in the retention rate of these bDMARDs. The retention rate of INF group and TCZ group decreased with time and was 10.5% and 33.3% at the final survey in November 2017, respectively. That of ETN group and ADA group plateaued at 9 years after the start of treatment and was 22.1% and 8.33% at the final survey, respectively. Conclusions In this study, there was no statistically significant difference in the retention rate of these bDMARDs. The retention rate of over 10 years was the highest in TCZ group and the lowest in ADA group. The rate of INF group and TCZ group continued to decrease, and that of ETN group and ADA group was almost constant after 9 years from the start of treatment. Disclosure of Interest None declared

Abstract

This paper discusses a multiscale stochastic analysis of a laminated composite plate consisting of unidirectional fiber reinforced composite laminae. In particular, influence of a microscopic random variation of the elastic properties of component materials on mechanical properties of the laminated plate is investigated. Laminated composites are widely used in civil engineering, and therefore multiscale stochastic analysis of laminated composites should be performed for reliability evaluation of a composite civil structure. This study deals with the stochastic response of a laminated composite plate against the microscopic random variation in addition to a random variation of fiber orientation in each lamina, and stochastic properties of the mechanical responses of the laminated plate is investigated. Halpin-Tsai formula and the homogenization theory-based finite element analysis are employed for estimation of effective elastic properties of lamina, and the classical laminate theory is employed for analysis of a laminated plate. The Monte-Carlo simulation and the first-order second moment method with sensitivity analysis are employed for the stochastic analysis. From the numerical results, importance of the multiscale stochastic analysis for reliability evaluation of a laminated composite structure and applicability of the sensitivity-based approach are discussed.

Abstract

BACKGROUND AND PURPOSE: Branch atheromatous disease is a stroke subtype considered a risk factor for early neurologic deterioration. Meanwhile, crossed cerebellar diaschisis is thought to be influenced by the degree and location of supratentorial perfusion abnormalities and is associated with the clinical outcome in the case of an ischemic stroke. In this article, our aim was to clarify the utility of using a whole-brain 3D pulsed continuous arterial spin-labeling method as an imaging biomarker for predicting neurologic severity in branch atheromatous disease. MATERIALS AND METHODS: Twenty-three patients with branch atheromatous disease in the lenticulostriate artery territory were enrolled. All patients underwent MR imaging, including DWI, 3D-TOF-MRA, and 3D-arterial spin-labeling. We measured the asymmetry index of CBF in the affected area (branch atheromatous disease), the asymmetry index of the contralateral cerebellar hemisphere (crossed cerebellar diaschisis), and the DWI infarct volume in the lenticulostriate artery territory. We also compared each parameter with the initial NIHSS score with the Pearson correlation coefficient. RESULTS: Among the 23 patients, we found no correlation between NIHSS score and the asymmetry index of CBF in the affected area (branch atheromatous disease) (r = −0.027, P = .724), whereas the asymmetry index of the contralateral cerebellar hemisphere (crossed cerebellar diaschisis) and DWI infarct volumes were significantly correlated with NIHSS score (r = 0.515, P = .012; r = 0.664, P = .001, respectively). CONCLUSIONS: In patients with branch atheromatous disease, 3D-arterial spin-labeling can detect crossed cerebellar diaschisis, which is correlated with the degree of neurologic severity.

Abstract

The exosome is a conserved multiprotein complex essential for RNA processing and degradation. The nuclear exosome is a key factor for pre-rRNA processing through the activity of its catalytic subunits, Rrp6 and Rrp44. In Saccharomyces cerevisiae, Rrp6 is exclusively nuclear and has been shown to interact with exosome cofactors. With the aim of analyzing proteins associated with the nuclear exosome, in this work, we purified the complex with Rrp6-TAP, identified the co-purified proteins by mass spectrometry, and found karyopherins to be one of the major groups of proteins enriched in the samples. By investigating the biological importance of these protein interactions, we identified Srp1, Kap95, and Sxm1 as the most important karyopherins for Rrp6 nuclear import and the nuclear localization signals recognized by them. Based on the results shown here, we propose a model of multiple pathways for the transport of Rrp6 to the nucleus.

Abstract

The Am and Bm phenotypes are characterized by weak expression of the A or B antigens, respectively, by red blood cells with a normal expression by the saliva of secretors. Deletion of the regulatory element in the first intron of the ABO gene and disruption of the GATA motif in the element were found to be responsible. In this study, we identified a novel mutation within the GATA motif (G>C substitution at position c.28 + 5830) in the regulatory element of the A allele that might diminish transcription activity causing the generation of the Am B phenotype.

Abstract

Background The aim of this study was to clarify the outcomes of lung resection for lung cancer in patients with cardiac disease, especially coronary artery disease, in a large-scale multi-institutional cohort. Methods We retrospectively analyzed the data on 1,254 patients who underwent major lung resection for lung cancer and had been diagnosed with coronary stenosis, atrial fibrillation, or both, in 58 institutions in Japan between January 2009 and December 2011. The primary outcome was 90-day postoperative mortality or in-hospital death. Results Among the 1,254 patients, 902 (71.9%) and 452 patients (36.0%) were preoperatively diagnosed with coronary stenosis and atrial fibrillation, respectively, and 951 patients (75.8%) received antiplatelet therapy. Among the patients with coronary stents (n = 532; 42.4%), 204 (16.3%) received drug-eluting stents. The 90-mortality or in-hospital death rate was 2.6% (n = 32), including stent thrombosis (n = 1), thromboembolic events without stent thrombosis (n = 2), and bleeding events (n = 2). In the multivariate analyses, blood transfusion, history of cerebrovascular disease, amount of bleeding, and history of congestive heart failure were associated with a higher independent risk of 90-day mortality or in-hospital death (odds ratio, 9.400, 3.574, 2.827, and 2.945, respectively). Preoperative discontinuation of antiplatelet therapy was not associated with an independent risk of 90-day mortality or in-hospital death on univariate analysis. Conclusions Major lung resection for lung cancer in patients with coronary artery disease is feasible. Our study suggests that discontinuation of antiplatelet therapy may not increase postoperative complications in patients with coronary artery disease.

Abstract

High-strength medium-manganese steel sheets recrystallized in the alpha phase after cold-rolling have a good press formability due to the //ND texture. If the A3 temperature and the alpha-to-gamma transformation temperature are decreased, the addition of manganese affects the recrystallization and the formability. In this work, the annealing texture evolution of medium-manganese steel was investigated, carbon-free Fe-Mn sheets being used for preliminary research, and some temperatures around the T0 temperature, at which ferrite and austenite have the same free energy, were selected for annealing. It was confirmed that the To temperature is a key factor to control annealing texture, i.e., although evolving with progress of recovery and partial recrystallization at annealing temperatures below the T0, the gamma fiber ( //ND) texture disappears at temperatures above the T0. The texture memory effect during alpha-gamma-alpha transformation was not confirmed in this binary system.

Abstract

Introduction: Streptococcus gordonii interacts with the salivary pellicle on the tooth surface and plays an important role in dental biofilm formation. Reports show that the analog Ssp peptide (A11K; alanine to lysine at position 11 in the arranged sequence, 1DYQAKLAAYQAEL13) of SspA and SspB of S. gordonii increased binding to the salivary agglutinin (gp-340/DMBT1) peptide (scavenger receptor cysteine-rich domain 2: SRCRP2). To determine the role of lysine in the binding of the Ssp(A11K) peptide to SRCRP2, we investigated whether an additional substitution by lysine influenced the binding of Ssp(A11K) peptide to SRCRP2 using a BIAcore biosensor assay. Methods: Six analogs of the Ssp peptide with positive charges in surface positions on the structure were synthesized using substitution at various positions. Results: The binding activity of analog Ssp(A4K–A11K) peptide was significantly higher than the other Ssp analogs. The binding activity rose under low ionic strength conditions. The distance between positively charged amino acids in the Ssp(A4K–A11K) peptide between 4K and 11K was 1.24 ± 0.02 nm and was close to the distance (1.19 ± 0.00 nm) between Q and E, presenting a negative charged area, on SRCRP2 using chemical computing graphic analysis. The molecular angle connecting 1D–11K–4K in the Ssp(A4K–A11K) peptide secondary structure was smaller than the other peptide angles (1D–11K–XK). The Ssp(A4K–A11K) peptide showed higher inhibiting activity for Streptococcus mutans binding to saliva-coated hydroxyapatite than the (A11K) peptide. Conclusion: The positioning of lysine is important for binding between Ssp peptide and SRCRP2, and the inhibiting effect on S. mutans binding to the tooth surface.

Abstract

Bacterial infection is a major cause of periapical periodontitis. Eradication of these microorganisms from apical lesions is essential to the success of endodontic treatment. The aim of this study was to clarify the molecular interaction between Fusobacterium nucleatum, Porphyromonas gingivalis and other microorganisms associated with periapical periodontitis.Microorganisms isolated from periapical lesions were inoculated into type-I collagen-coated polystyrene microtiter plates and maintained at 37 degrees C under anaerobic conditions for 2 days, after which, the quantity of organized biofilm on the plates was evaluated by crystal violet staining. Growth enhancement via soluble factor was evaluated by separated coculture using a 0.4-mum membrane filter.F. nucleatum exhibited strong adherence to type-I collagen-coated polystyrene microplates. Biofilm formation by F. nucleatum was significantly enhanced by P. gingivalis. It was complemented by compartmentalized coculture with P. gingivalis. Enhancement of biofilm formation by P. gingivalis was only slightly reduced by inactivation of its autoinducer-2-producing gene luxS.The results suggest that P. gingivalis enhances biofilm formation by F. nucleatum by releasing diffusible signaling molecules other than autoinducer-2.

Abstract

Immunization involving a DNA vaccine prime followed by an adenovirus type 5 (Ad5) boost elicited a protective immune response against SHIV challenge in monkeys. However, the hepatocellular tropism of Ad5 limits the safety of this viral vector. This study examines the safety and immunogenicity of a replication-defective chimeric Ad5 vector with the Ad35 fiber (Ad5/35) in BALB/c mice and rhesus monkeys. This novel Ad5/35 vector showed minimal hepatotoxicity after intramuscular administration with the novel Ad5/35 vector. In addition, an Ad5/35 vector expressing HIV Env gp160 protein (Ad5/35-HIV) generated strong HIV-specific immune responses in both animal models. Priming with a DNA vaccine followed by Ad5/35-HIV boosting yielded protection against a gp160-expressing vaccinia virus challenge in BALB/c mice. The Ad5/35-HIV vector was significantly less susceptible to the pre-existing Ad5 immunity than a comparable Ad5 vector. These findings indicate that an Ad5/35 vector-based HIV vaccine may be of considerable value for clinical use.

The Na-related luminescence, which is the dominant scintillation of CsI:Na, is activated by heat treatment and associated with formation of NaI nanoparticles. It is found that the Na-related luminescence, which is remarkably degraded in atmosphere, the so-called hygroscopic property, is reversibly recovered in vacuum, while the luminescence due to the self-trapped exciton of CsI hardly decreases in ambient conditions. This finding indicates that the hygroscopic property of CsI:Na results from degradation of NaI nanoparticles embedded in the CsI:Na thin film and that the recovery in vacuum is due to recrystallization. This is a novel proposal to explain the high hygroscopicity of CsI:Na. Furthermore, it is demonstrated from the temperature dependence of the luminescence properties that the energy transfer from the host material of CsI to the NaI nanoparticles is mediated through thermally activated hopping of the self-trapped exciton of CsI.

Abstract

The nonlinear evolution equation for the fluctuation of a terrace edge with anisotropy in step stiffness during step flow growth is derived in the two-sided model where adatoms can be incorporated into the step from the upper terrace as well as from the lower terrace. It is shown by reductive perturbation method based on the linear stability analysis that (1) up to e 3 order in the smallness parameter of instability the nonlinear evolution equation reduces to the closed Kuramoto–Sivashinsky (KS) equation with extra coefficients compared with the one-sided model and (2) the nonlinear evolution equation up to e 5 order is also derived in order to take into account the anisotropy in step stiffness in the lowest order. Abstract

Objective Our purpose was to evaluate the safety, efficacy and procedural difficulties of intraoperative autologous blood transfusion during laparoscopic surgery for ectopic pregnancy. Design A retrospective study was performed. Methods The Cell Saver, an intraoperative autologous blood salvage device, was used in laparoscopic operations on 13 patients with ectopic pregnancy, who had haemoperitoneum but good physical status without severe anaemia and hypovolaemic shock. Results Homologous blood transfusion was not needed in any patient. The mean blood loss was 327±178 ml and the mean volume of reinfused processed blood was 288±92 ml. The haemoglobin level of reinfused processed blood was proportional to the blood loss. Neither adverse reactions to norprocedural difficulties of autotransfusion were observed. Conclusions When laparoscopic surgery is performed for ectopic pregnancy in patients with haemoperitoneum, intraoperative autologous transfusion employing the Cell Saver is considered to be a potentially very useful technique.

Abstract

To describe the residual stress-induced interfacial debonding and stress–strain behavior of unidirectional brittle fiber/brittle matrix composite with weak interface, a simulation method, based on the modified shear lag analysis which can take residual stresses into account and Monte Carlo procedure, was presented, and was applied to a model composite similar to polymer-derived SiC/ceramic glass system. Main results are summarized as follows. (1) It was shown concretely that the broken component-induced debonding is encouraged and discouraged when the corresponding component has tensile and compressive residual stresses, respectively. (2) The feature of the damage- (breakage of fiber, breakage of matrix and interfacial debonding)-accumulation process and its influence on the stress–strain curve could be realized in the computer; the damages are accumulated intermittently, resulting in a serrated stress–strain curve of composites. In such a damage accumulation process, the existence of residual stresses change the order and location of occurrence of damages and therefore the stress–strain curve and strength of the composite. (3) The dependence of the composite strength on the matrix strength in the presence of residual stresses was clarified; the composite strength increases significantly with increasing matrix strength when the fiber and matrix have compressive and tensile residual stresses along the fiber axis, respectively, but only marginally when they have reversed residual stresses.
