1. Effect of Si content on the microstructure and mechanical properties of 9Cr-ferritic/martensitic steels

Abstract

9Cr-F/M−xSi (x = 0–1.0 wt%) steels were fabricated through vacuum induction melting technique and processed by hot forging, hot rolling, normalizing and tempering, subsequently. Their microstructure and mechanical properties were characterized using OM, SEM, TEM, Vickers hardness tester and tensile tester. The steel has a typical ferrite/martensitic structure, with M23C6 and MX phases precipitated at the martensite lath boundary or in the lath. With Si content increasing, the average ultimate tensile strength (UTS) and hardness of 9Cr-F/M−xSi increase simultaneously from 678 MPa and 235 HV for 9Cr-F/M−0Si steel to 788 MPa and 265 HV for 9Cr-F/M−1.0Si steel, respectively, while the total elongation kept almost constant at 22.5%. The main strengthening mechanism is the solid solution strengthening due to silicon addition and the change in the carbide precipitation behavior caused no remarkable hardening contribution. These results can provide a reference for the composition design of structural materials for nuclear reactors.

Abstract

Semiconducting single-walled carbon nanotubes (SWCNT) containing covalent defects are a promising class of optoelectronic materials, demonstrating single photon emission and long-lived spins. The defect introduces new optical transitions due to both symmetry breaking induced band splitting and introduction of in-gap electronic states. We investigate sulfur-oxide containing compounds as a new class of optically active dopants on (6,5) SWCNT. The SWCNT is exposed to sodium dithionite with the resulting compound displaying a red-shifted and bright photoluminescence peak that is characteristic of these covalent defects. Density functional theory calculations are then performed on the adsorbed compounds that may arise (S, SO, SO2 and SO3). These calculations indicate that the two smallest molecules strongly bind to the SWCNT with binding energies of ~ 1.5-1.8 eV and 0.56 eV for S and SO, respectively. Moreover, these adsorbates introduce in-gap electronic states into the bandstructure of the tube consistent with the measured red-shift of (0.1-0.3) eV. Abstract

Background & aims: Genome-wide association studies in primary biliary cholangitis (PBC) have failed to find X chromosome (chrX) variants associated with the disease. Here, we specifically explore the chrX contribution to PBC, a sexually dimorphic complex autoimmune disease. Methods: We performed a chrX-wide association study, including genotype data from 5 genome-wide association studies (from Italy, United Kingdom, Canada, China, and Japan; 5244 case patients and 11,875 control individuals). Results: Single-marker association analyses found approximately 100 loci displaying P < 5 × 10-4, with the most significant being a signal within the OTUD5 gene (rs3027490; P = 4.80 × 10-6; odds ratio [OR], 1.39; 95% confidence interval [CI], 1.028-1.88; Japanese cohort). Although the transethnic meta-analysis evidenced only a suggestive signal (rs2239452, mapping within the PIM2 gene; OR, 1.17; 95% CI, 1.09-1.26; P = 9.93 × 10-8), the population-specific meta-analysis showed a genome-wide significant locus in East Asian individuals pointing to the same region (rs7059064, mapping within the GRIPAP1 gene; P = 6.2 × 10-9; OR, 1.33; 95% CI, 1.21-1.46). Indeed, rs7059064 tags a unique linkage disequilibrium block including 7 genes: TIMM17B, PQBP1, PIM2, SLC35A2, OTUD5, KCND1, and GRIPAP1, as well as a superenhancer (GH0XJ048933 within OTUD5) targeting all these genes. GH0XJ048933 is also predicted to target FOXP3, the main T-regulatory cell lineage specification factor. Consistently, OTUD5 and FOXP3 RNA levels were up-regulated in PBC case patients (1.75- and 1.64-fold, respectively). Conclusions: This work represents the first comprehensive study, to our knowledge, of the chrX contribution to the genetics of an autoimmune liver disease and shows a novel PBC-related genome-wide significant locus.

Abstract

The aim of this study was to analyze the microstructural changes of two dissimilar Ti-6Al-4V/Ti17 and Ti-6Al-4V/BT9 joints welded via electron beam welding (EBW), and establish the relationship between microstructure and properties. The results showed that the occurrence of phase transformation was mainly dependent on the peak heating temperature during EBW, and despite the same welding parameters the microstructures of two dissimilar joints were different except in the heat-affected zone (HAZ) on the Ti-6Al-4V side. Both dissimilar joints exhibited a higher strength compared with Ti-6Al-4V base metal (BM), and the strength of Ti-6Al-4V/Ti17 joint was slightly higher than that of Ti-6Al-4V/BT9 joint due to the presence of finer martensite. Fatigue life of dissimilar joints was almost the same as that of Ti-6Al-4V BM within the experimental scatter. Fatigue failure of both dissimilar joints occurred in the HAZ, and fatigue crack basically initiated from the specimen surface.

9. Long non-coding RNA LINC00152 is up-regulated in ovarian cancer tissues and regulates proliferation and cell cycle of SKOV3 cells

Abstract

To characterize functions of long non-coding RNA (lncRNA) in the progression of epithelial ovarian cancer.Epithelial ovarian cancer tissues and matching normal tissues were collected from two individual patients for RNA microarray analysis. Besides, twenty-two ovarian cancer samples and ten healthy ovarian epithelial tissues were collected for Reverse Transcription-quantitative Polymerase Chain Reaction (RT-qPCR). Microarray assay suggested that a list of cancer relating mRNAs and lncRNAs were upregulated. The identified lncRNAs were validated via RT-qPCR, which led to the identification of long intergenic non-protein coding RNA 152 (LINC00152). To determine the function of LINC00152 in ovarian cancer, we knocked down the expression of LINC00152 in epithelial ovarian cancer cell line SKOV3 with small interference RNAs (siRNAs). The effects of LIN00152 on the proliferation and cell cycle were determined by comparing the cell viability of SKOV3 cells with LIN00152 knockdown and the control cells with negative siRNA. The cell viability was assessed using Cell Counting Kit-8 (CCK-8) and flow cytometry assay. RNA microarray assay was used again in control and LINC00152 knockdown SKOV3 cells to identify downstream signaling pathways.Fourteen ovarian cancer relating lncRNAs were identified by RNA microarray assay. Up-regulation of LINC00152 was validated via RT-qPCR. A higher expression of LINC00152 in late cancer stage (III-IV) compared to the early stage tumors was also demonstrated. Inhibition of LINC00152 in SKOV3 cells inhibited cell proliferation and induced cell cycle arrest that involved prolonged G1 phase and shortened S phase. The microarray assay data of SKOV3 cells suggested that Cyclin-Dependent Kinase Inhibitor 1C (CDKN1C) was a potential downstream target of LINC00152.LINC00152 is upregulated in epithelial ovarian cancer tissues comparing to normal tissues. Knockdown of LINC00152 expression inhibits cell proliferation and induces cell cycle arrest. LINC00152 possibly interacts with Tumor Necrosis Factor (TNF) signaling pathway. CDKN1C is a potential downstream target of LINC00152.

Abstract

The cells of the innate immune system continually undergo surveillance of the extracellular milieu to serve as the first line of defense against invading microbial pathogens. As a result, these innate immune cells have evolved receptors to sense microbial motifs and danger signals. The inflammasome is an important component of the innate immune system’s “arsenal” against invading microbes such as viruses and bacteria and acts as a homeostatic checkpoint to regulate inflammation. Inflammasomes are cytosolic multi-protein complexes that activate caspase 1 and 11, which result in the production of pro-inflammatory interleukin (IL)-1β and IL-18, and cell death. Several studies have uncovered the mechanisms of inflammasome activation in response to various stimuli ranging from environmental stimulants to microbial components. This mini-review summarizes the current developments and updates in the mechanism of action of inflammasomes.

Enhanced glow discharge plasma immersion ion implantation and deposition (EGD-PIII&D) has been successfully used for depositing diamond-like carbon (DLC) on the inner surface of the quartz glass tube with an inner diameter as small as 0.9 mm and aspect ratios of over 100. And the uniformity is a key problem for the DLC films deposited on the inner surface. By changing the C2H2 flow rate and negative voltage, the effects of the gas flow rate and voltage on the uniformity of the DLC films deposited on the inner surface of the quartz glass tube are experimentally examined. The electron-neutral collision frequency, νe, is used to qualitatively analyze the mechanism of uniformizing DLC films. The results reveal that uniform DLC films may be obtained by balancing both the gas flow rate and the negative voltage.

Abstract

We investigated the effects of combined targeted gene therapy on recurrence and metastasis after liver cancer resection in nude mice. Twenty BALB/C mice were randomly divided into control and treatment groups with 10 mice in each group and a male/female ratio of 1:1. Luciferase gene-labeled human primary hepatic carcinoma cell line MHCC97-H was then used to prepare a carcinoma model. An optical in vivo imaging technique (OIIT) was used 10 days later to detect the distribution of tumor cells, followed by partial liver resection and gene therapy. In the treatment group, 100 mL phosphate-buffered saline (PBS) containing 1 x 1012 rAAV/AFP/IL-24 gene viral vectors was injected into liver sections and peritumoral posterior peritoneal tissues; in the control group, the same amount of PBS containing 1 x 1012 empty viral vectors was injected at the same sites. OIIT was then used to detect the in vivo tumor metastasis 21 days later. Luciferase gene-labeled human primary hepatic carcinoma cell line MHCC97-H successfully infected 20 nude mice, and OIIT showed that the two groups exhibited metastasis after local tumor resection, but there were more tumor cells in the control group (P < 0.05). rAAV/AFP/IL-24 gene therapy can inhibit recurrence after liver cancer resection.

Hepatitis C virus (HCV) is one of the major causes of liver inflammation. The aim of this study was to investigate the associations of T-cell immunoglobulin and mucin domain-3 (Tim-3) polymorphisms and the alternate reading frame protein (F protein) with the outcomes of HCV infection. Three single-nucleotide polymorphisms (SNPs; rs10053538, rs12186731, and rs13170556) of Tim-3 were genotyped in this study, which included 203 healthy controls, 558 hepatitis C anti-F-positive patients, and 163 hepatitis C anti-F-negative patients. Abstract

Hypoxia-inducible factor-1α (HIF-1α) is a transcription factor that has a central role in the regulation of tumour metabolism under hypoxic conditions. HIF-1α stimulates glycolytic energy production and promotes tumour growth. Sirtuins are NAD(+)-dependent protein deacetylases that regulate cellular metabolism in response to stress; however, their involvement in the hypoxic response remains unclear. In this study, it is shown that SIRT2-mediated deacetylation of HIF-1α regulates its stability in tumour cells. SIRT2 overexpression destabilized HIF-1α under hypoxic conditions, whereas HIF-1α protein levels were high in SIRT2-deficient cells. SIRT2 directly interacted with HIF-1α and deacetylated Lys709 of HIF-1α. Deacetylation of HIF-1α by SIRT2 resulted in increased binding affinity for prolyl hydroxylase 2, a key regulator of HIF-1α stability, and increased HIF-1α hydroxylation and ubiquitination. Moreover, a pharmacological agent that increased the intracellular NAD(+)/NADH ratio led to the degradation of HIF-1α by increasing SIRT2-mediated deacetylation and subsequent hydroxylation. These findings suggest that SIRT2-mediated HIF-1α deacetylation is critical for the destablization of HIF-1α and the hypoxic response of tumour cells.

In-situ Mossbauer studies for dehydrogenation of ethylbenzene to styrene has been carried out. The results show that the formation of iron oxide is essentially Fe3O4 and there is no KFeO2 under the reaction condition. As a promoter, the existence of potassium is in favour of the electron exchange between Fe2+ and Fe3+ ions.

Abstract

Induction of gene expression has great potential in the treatment of many human diseases. Peptide nucleic acid (PNA) as a novel DNA-binding reagent provides an ideal system to induce gene-specific expression. In our recent studies, we have demonstrated that PNA bound to double-stranded DNA targets and, therefore, generated single-stranded D-loops and induced transcription of target genes both in vitro and in vivo. Most importantly, we have demonstrated that treatment of cultured human cells with PNAs led to expression of an endogenous target gene. Therefore, the study of the molecular mechanism of PNA binding-induced gene expression will have great implications for the gene therapy of many human diseases. In the current study, we have investigated the PNA length requirement for PNA binding-induced transcription initiation. Using a series of PNAs with different lengths, we have determined that PNAs with lengths of 16 approximately 18 nt induce very high levels of transcription in a HeLa nuclear extract in vitro transcription system. Transfection of the PNA-bound GFP reporter gene plasmid into human normal fibroblast (NF) cells led to a similar result. Gel-mobility shift assays revealed very strong binding affinities of these PNAs. DNA footprinting analysis further demonstrated the specificity of PNAs binding to the targets. These results lead to important understanding of the molecular mechanism of transcription initiation and highly valuable information in PNA design, especially for PNA binding-induced, gene-specific expression.

Abstract

The hybrid wheat line, 93784 is a stable line selected from progenies of Leymus molli Trin. Hara x common wheat, which shows a very high level of resistance to yellow rust. The results of GISH using genomic DNA of Leymus mollis as probe indicated that the 93784 was a wheat-Leymus mollis small segment translocation line, and the translocated alien chromosome fragment were located on a pair of short arm terminals of what chromosomes. Using this translocation line, we established F2 segregation population and identified its yellow rust resistance in adult stage. Genetic analysis suggested that the yellow rust resistance of 93784 was controlled by a single gene. The gene was located on the translocated chromosome segments of Leymus mollis. We called it YrLm. By AFLP analysis with Taq I (T1-T4)/Pst I (P1-P6) primes in individuals of F2 segregation population and their susceptive and resistant parents, we screened a molecular marker that linked to yellow rust resistance gene YrLm. We also cloned and sequenced the molecular marker. The length of the marker is 205 bp, named P1T(3)205.

The chromosome constitution on H96276-2, which is a stable line derived from a sterile wheat-Agropyron intermedium partial amphiploid Zhong 3 cross to common wheat Hui 75, was analyzed by sequential GISH and FISH. The results of GISH using genomic DNA of Agropyron intermedium as probe showed that H96276-2 possessed 20 pairs of wheat chromosomes and one pair of translocation chromosomes of wheat and Ag. intermedium. The translocated alien chromosome fragments located on the terminals of one pair of wheat chromosomes. Furthermore, reprobing of repeat sequence pSc119 was used to identify the wheat chromosomes involved in the translocation. It was determined that the alien fragments located in the terminals of the short arms of 2B chromosomes of wheat.

Abstract

Over 90% of human cervical carcinoma is associated with high risk human papillomavirus (HPV), and a number of biological effects that could contribute to cancer suppression by ω3-polyunsaturated fatty acids (ω3-PUFAs) have been reported. However, the anti-cancer effect of ω3-PUFAs on cervical cancer has been not known yet. In this study, we report inhibitory mechanisms of ω3-PUFAs on cell growth and invasion in cervical cancer. DHA inhibited the cell growth in a dose- and time-dependent manner. In flow cytometry analysis, cell cycle of DHA-treated HeLa cells was arrested in G2/M phase and SubG1 cells also increased. Moreover, apoptotic cell death was confirmed by TUNEL assay, the induction of PARP cleavage and down-regulation of Bcl-2. The invasiveness of cells was significantly inhibited by DHA treatment in vitro transwell assay as well. The MMP-9 and MMP-2 promoter activities were decreased after DHA treatment. Cox-2 and VEGF promoter activities were also inhibited by DHA. Furthermore, DHA decreased the levels of reporter activity of NF-κB, which is transcription factor that regulates MMPs, Cox-2 and VEGF expression. In in vivo experiments, when human papillomavirus type 16 (HPV16)-transformed mouse TC-1 cells were injected into the tail vein of Fat1 mice (Fat1 transgenic mice express a Caenorhabditis elegans ω3-desaturase converting ω6- to ω3-PUFAs endogenously.) and WT (wild type) mice, lung metastasis of TC-1 cells was dramatically inhibited in Fat1 transgenic mice compared to WT mice. Taken together, these findings provide evidence that ω3-PUFAs may inhibit metastasis as well as cell growth and invasion through suppression of MMPs/COX-2/VEGF expression by inhibition of NF-κB expression in cervical cancer cells, indicating that the utilization of ω3-PUFAs may represent a potential effective therapy for the chemoprevention and treatment of human cervical cancer. Abstract

Introduction: Reactive oxygen species (ROS) produced by docosahexaenoic acid (DHA) have an important function in cancer cell death. However, the exact mechanism of ROS production, after DHA stimulation, is not clearly understood. Here, we determined that elevated levels of ROS generated by mitochondrial respiration is directly associated with DHA-induced cervical cancer cell death. Material and Method: The effects of DHA on cell proliferation and cell cycle were examined by MTT assay and FACS. DHA-induced apoptosis was analyzed using the TUNEL assay, caspase activity assay, and western blot. Dihydroethidium (DHE) was used for reactive oxygen species (ROS) measurement in cytosol. MitoSox was performed for ROS measurement in mitochondria. Results and Discussion: The levels of caspase 3 activity, TUNELpositive staining cells and Sub-G1 portion were markedly increased in DHA-treated cancer cells, suggesting that apoptosis is responsible for the DHA-induced cervical cancer cell death. Furthermore, DHA was able to induce both mitochondrial complex I substrateand complex II substratesupported mitochondrial ROS production in isolated mitochondria from rodent liver. Meanwhile, a reduction in oxygen consumption rate and an increase in mitochondrial ROS production as measured by MitoSOX, were also observed in DHA-treated cancer cells, indicating that DHA can directly act on mitochondrial respiration and enhance ROS generation. The role of DHA-induced mitochondrial ROS production in apoptosis was further identified by the findings that DHA reduced the mitochondrial membrane potential, resulting in cardiolipin oxidation and cytochrome c release from mitochondria, and that N-acetylcysteine, an antioxidant almost completely blocked these processes as well as ROS production occurred in mitochondria and remarkably reversed the apoptotic cell death triggered by DHA. Conclusion: From the results presented here, we conclude that mitochondria actively participate in the DHA-induced apoptotic cell death by the generation of mitochondrial ROS. Abstract

Recent molecular and clinical studies have shown that invasion and metastasis may occur very early in tumour development, thus emphasizing the potential importance of spesific and sensitive detection of circulating tumour cells (CTC). With the ability of refined technologies the identification of CTCs from peripheral blood is emerging as a useful tool for the detection of malignancy, monitoring disease progression and measuring response to therapy. The goal of this study was to identify optimal marker or marker combinations for detection of CTCs in the gastrointestinal malignancies using RT-PCR. Materials and Methods: To detect the presence of CTCs, we analyzed Cytokeratin 19 (CK19), Cytokeratin 20 (CK20) and Mucin 1 (MUC1) mRNA in the peripheral blood of 31 patients with gastrointestinal (gastric, stomach and colorectal) carcinoma and 30 healthy individuals. Results: In RT-PCR analysis of the peripheral blood, 77.4% (24/31), 58.06% (18/31) and 3.22% (1/31) of cancer patients were positive for MUC1, CK20 and CK19 mRNA respectively. The sensivity and specificity for any one of mRNA detected in peripheral blood is 83.3% and 66.6% respectively, with an acurracy of 59%. Conclusions: Our study suggest that MUC1 and CK20 mRNAs in the peripheral blood could be useful molecular markers for gastrointestinal tumours. Combination of these two tumour-spesific mRNA markers would increase the detection rate and may be clinically helpful in predicting the tumour presence and colorectal cancer metastasis.

Abstract

Experiments demonstrated that the cell differentiation of HL-60 cells induced by low concentrations of daidzein (S86019), BC-4 (active principle of Boswellia carterii Birdw) and Ara-C was not impressive. However, when they were used in combination 80% of HL-60 cells exhibited NBT reduction and 82% of the cells showed phagocytosis after four days exposure to daidzein and BC-4. When HL-60 cells were exposed to daidzein and Ara-c, 70% of the cells exhibited NBT reduction and phagocytosis. Flow cytometry indicated that the majority of the cells were blocked at G1 phase under the induction of combination of daidzein with BC-4 or Ara-C.

Abstract

Angiogenesis, the process of new blood vessel formation, is essential for tumor growth and metastasis. Although the protective role of ω3-PUFAs on cancer progression has been reported, the anti-angiogenic mechanisms remain to be further understood. The aim of this study is to investigate the anti-angiogenic mechanisms of ω3-PUFAs in pancreatic cancer. In angiogenesis protein array chip assay, only angiogenic ELR+ CXC chemokines expression, which activate endothelial cell migration and neutrophils recruitment, was significantly reduced by DHA treatment in SW1990 human pancreatic cancer cells. The reduced expression of ELR+ CXC chemokine was confirmed by RT-PCR. Chemotactic migration of HUVEC was significantly reduced in DHA-treated SW1990 conditioned medium (CM) compared to control CM. DHA also inhibited VEGF reporter activity in SW1990 cells. VEGF-induced endothelial cell proliferation and migration were markedly inhibited by DHA treatment. MMP-2 and MMP-9 promoter activities were reduced by DHA treatment, and ultimately attenuated the invasiveness of SW1990 cells as revealed by matrigel droplet assay. Moreover, NF-κB reporter activity, which is closely correlated with the expression of CXC chemokine, VEGF and MMPs, was reduced by DHA treatment. When mouse pancreatic cancer PANC02 cells were implanted into Fat-1 transgenic mice, tumor size and volume were dramatically reduced in Fat-1 mice compared to control mice. In immunohistochemical analysis of tumors of Fat-1 mice, apoptosis (TUNEL positive cells) was markedly increased and microvessel density (CD31 intensity) was significantly reduced in Fat-1 mice compared to control mice. In addition, VEGF-induced neovascularization to a subcutaneous matrigel plug was significantly inhibited in Fat-1 mice compared with control mice. Our finding suggests that ω3-PUFAs can suppress tumor angiogenesis through inhibition of ELR+ chemokines expression and VEGF signal in pancreatic cancer. Thus ω3-PUFAs supplementation may beneficial for chemoprevention and treatment of pancreatic cancer by anti-angiogenic effect. Abstract

Context Glucose-insulin-potassium (GIK) infusion is a widely applicable, low-cost therapy that has been postulated to improve mortality in patients with acute ST-segment elevation myocardial infarction (STEMI). Given the potential global importance of GIK infusion, a large, adequately powered randomized trial is required to determine the effect of GIK on mortality in patients with STEMI. Objective To determine the effect of high-dose GIK infusion on mortality in patients with STEMI. Design, setting, and participants Randomized controlled trial conducted in 470 centers worldwide among 20,201 patients with STEMI who presented within 12 hours of symptom onset. The mean age of patients was 58.6 years, and evidence-based therapies were commonly used. Intervention Patients were randomly assigned to receive GIK intravenous infusion for 24 hours plus usual care (n = 10,091) or to receive usual care alone (controls; n = 10,110). Main outcome measures Mortality, cardiac arrest, cardiogenic shock, and reinfarction at 30 days after randomization. Results At 30 days, 976 control patients (9.7%) and 1004 GIK infusion patients (10.0%) died (hazard ratio [HR], 1.03; 95% confidence interval [CI], 0.95-1.13; P = .45). There were no significant differences in the rates of cardiac arrest (1.5% [151/10 107] in control and 1.4% [139/10,088] in GIK infusion; HR, 0.93; 95% CI, 0.74-1.17; P = .51), cardiogenic shock (6.3% [640/10 107] vs 6.6% [667/10 088]; HR, 1.05; 95% CI, 0.94-1.17; P = .38), or reinfarction (2.4% [246/10,107] vs 2.3% [236/10,088]; HR, 0.98; 95% CI, 0.82-1.17; P = .81). The rates of heart failure at 7 days after randomization were also similar between the groups (16.9% [1711/10,107] vs 17.1% [1721/10,088]; HR, 1.01; 95% CI, 0.95-1.08; P = .72). The lack of benefit of GIK infusion on mortality was consistent in prespecified subgroups, including in those with and without diabetes, in those presenting with and without heart failure, in those presenting early and later after symptom onset, and in those receiving and not receiving reperfusion therapy (thrombolysis or primary percutaneous coronary intervention). Conclusion In this large, international randomized trial, high-dose GIK infusion had a neutral effect on mortality, cardiac arrest, and cardiogenic shock in patients with acute STEMI.

Abstract

Five wheat-triticale doubled haploid (DH) lines- M08, V209, DH220-14-2, DH696-3-4 and M16-derived from anther culture of F1s resulting from crosses involving hexaploid or octoploid triticale × hexaploid wheat, were characterized by cytological and biochemical markers. Cytological evidence from genomic in situ hybridization and C-banding indicated that DH lines M08 and V209 (2n = 42) each contained a pair of 1BL/1RS translocation chromosomes. DH220-14-2 (2n = 42) was also a translocated line with two pairs of chromosomes containing small fragments of rye. One of the translocation fragments carried the Sec-1 R gene originating from the satellite region of 1 RS; the origin of the other one remains unknown. DH696-3-4 (2n = 42) contained a 3D(3R) substitution. In M16 (2n = 44), three pairs of rye chromosomes, 3R, 4R and 6R, were present, 4R as an addition and 3D(3R) and 6D(6R) as substitutions. Biochemical, isozyme and storage protein markers confirmed the cytological conclusions. The advantages of transferring alien chromosomes or chromosome fragments into wheat and creating alien aneuploid lines by anther culture of hybrid F1s are discussed. [ABSTRACT FROM AUTHOR]

Abstract

PGM1 subtypes of 275 Chinese living in the Guangzhou area were determined using isoelectric focusing. The allele frequencies of PGM1 were 1+ = 0.6036, 1- = 0.1018, 2+ = 0.2200, and 2- = 0.0673. A rare variant, named PGM1*8, was found with a gene frequency of 0.0073. The phenotype distribution was consistent with Hardy-Weinberg equilibrium.

Abstract

Background: Anti-p200 pemphigoid is a rare autoimmune subepidermal blistering disease. Although the phenomenon of epitope spreading has been reported to be common in anti-p200 pemphigoid, the association between its clinical and immunoserological features has yet to be elucidated., Objectives: Our aim was to compare the clinical and immunoserological characteristics of anti-p200 pemphigoid patients with and without epitope spreading., Methods: We performed a retrospective cohort study encompassing 30 patients with anti-p200 pemphigoid between January 2015 and December 2022. The clinical and immunoserological characteristics of anti-p200 pemphigoid were analyzed using combined immunoserological assays., Results: Epitope spreading was observed in 11 of 30 patients (36.7%) with anti-p200 pemphigoid. Compared with patients in the non-epitope spreading group, patients in the epitope spreading group showed more heterogeneous clinical presentations (P = 0.018), a higher proportion of mucosal involvement (P = 0.003), higher Bullous Pemphigoid Disease Area Index (BPDAI) scores for skin erosions/blisters (P = 0.018), mucosal erosions/blisters (P = 0.001), activity (P = 0.017) and total scores (P = 0.022), and required a higher initial dose of prednisone for disease control (P = 0.040)., Conclusions: This study supported the idea that anti-p200 pemphigoid was prone to epitope spreading. Anti-p200 pemphigoid patients with epitope spreading are more likely to present heterogeneous clinical phenotypes, frequent mucosal involvement, and a more severe and recalcitrant disease course., (© 2024 the International Society of Dermatology.)

Abstract

Obesity is a major public health concern and burden on individuals and healthcare systems. Due to the challenges and limitations of lifestyle adjustments, it is advisable to consider pharmacological treatment for people affected by obesity. However, the side effects and limited efficacy of available drugs make the obesity drug market far from sufficient. Drug repurposing involves identifying new applications for existing drugs and offers some advantages over traditional drug development approaches including lower costs and shorter development timelines. This review aims to provide an overview of drug repurposing for anti-obesity medications, including the rationale for repurposing, the challenges and approaches, and the potential drugs that are being investigated for repurposing. Through advanced computational techniques, researchers can unlock the potential of repurposed drugs to tackle the global obesity epidemic. Abstract

Backgrounds: Considering the crucial role of military personnel's mental health in the armed forces, society, and the nation, the present study aims to describe the prevalence and influential factors of depression, anxiety, insomnia, and post-traumatic stress disorder (PTSD) among Chinese military personnel during the COVID-19 pandemic., Methods: The present study conducted an epidemiological investigation of mental health among Chinese military personnel between October 25th and November 10th, 2022. Data was collected from military units in thirty provinces across the country. Participants completed a self-reported questionnaire about their demographic characteristics and four validated scales (PHQ-9, GAD-7, ISI-7 and PCL-5), assessing the symptoms of depression, anxiety, insomnia, and PTSD., Results: A total of 17,423 military personnel participated in the study. The prevalence of depression, anxiety, insomnia, and PTSD were 2.69%, 0.99%, 2.90% and 1.61%, respectively. Certain demographic groups were identified as having a higher prevalence, including older individuals, women, those with longer military service, higher education levels, divorced or widowed individuals, only child, those with one child, and those from urban areas., Conclusions: The prevalence of mental health problems among Chinese military personnel has remained at a relatively low level, but our data still suggest that a significant number of individuals in the Chinese military population are affected by mental health issues. 46. Dupilumab, corticosteroids and their combination for the treatment of bullous pemphigoid.

Abstract

Background: Conventional systemic corticosteroid therapy for bullous pemphigoid (BP) has been challenged due to severe adverse events. Dupilumab has emerged as an alternative therapeutical option of BP patients., Objectives: To evaluate the efficacy of dupilumab monotherapy and the combination with medium/low-dose corticosteroids for BP treatment., Methods: Thirteen, twenty-four and thirty-two BP patients treated with Dupilumab monotherapy (Dupi group), dupilumab combined with corticosteroids (Dupi + CS group), and corticosteroid monotherapy (CS group), respectively, were retrospectively analyzed for various clinical and laboratory parameters., Results: In the Dupi group, the total Bullous Pemphigoid Disease Area Index (BPDAI) Total, Erosion/Blister, Urticaria/Erythema and Itching NRS scores were all reduced significantly after 2-4 weeks of treatment, but the BPDAI Mucosal Score was not changed significantly at the end of the overextended time of treatment. All the above clinical parameters and many laboratory parameters (including the serum anti-BP180 autoantibodies [IgG] level, blood eosinophil count, and percentage) were significantly reduced in both Dupi + CS and CS groups after treatment, but no statistical differences were found in the reduction rates of these parameters between the two groups. However, the Dupi + CS group had less baseline dose and cumulative dosage of prednisone at the time of disease control, and fewer adverse effects were reported than the CS group., Study Limitations: The retrospective design and small clinical sample size of the Dupi group., Conclusions: For BP patients, dupilumab monotherapy based on the treatment of atopic dermatitis can significantly improve skin lesions and pruritus symptoms but may be ineffective for oral mucosal lesions. Abstract

Development of functional recovery therapies is critical to reduce the global impact of stroke as the leading cause of long-term disability. Our previous studies found that acute-phase protein orosomucoid (ORM) could provide an up to 6 h therapeutic time window to reduce infarct volume in acute ischemic stroke by improving endothelial function. However, its role in neurons and functional recovery post-stroke remains largely unknown. Here, we showed that exogenous ORM administration with initial injection at 0.5 h (early) or 12 h (delayed) post-MCAO daily for consecutive 7 days significantly decreased infarct area, improved motor and cognitive functional recovery, and promoted mitochondrial biogenesis after MCAO. While neuron-specific knockout of ORM2, a dominant subtype of ORM in the brain, produced opposite effects which could be rescued by exogenous ORM. In vitro, exogenous ORM protected SH-SY5Y cells from OGD-induced damage and promoted mitochondrial biogenesis, while endogenous ORM2 deficiency worsened these processes. Mechanistically, inactivation of CCR5 or AMPK eliminated the protective effects of ORM on neuronal damage and mitochondrial biogenesis. Abstract

In this paper, the adsorption of gatifloxacin (GAT) by three types of polystyrene nano-plastics (PSNPs), including 400 nm polystyrene (PS), amino-modified PS (PS-NH 2 ), and carboxyl-modified PS (PS-COOH) was studied and the adsorption mechanism were assessed. Experimental findings revealed that the equilibrium adsorption capacity of PSNPs to GAT followed the order PS-NH 2  > PS-COOH > PS. The adsorption was regulated by both physical and chemical mechanisms, with intra-particle and external diffusion jointly controlling the adsorption rate. The adsorption process was heterogeneous, spontaneous, and entropy-driven. Sodium chloride (NaCl), alginic acid, copper ions (Cu 2+ ), and zinc ions (Zn 2+ ) inhibited adsorption, with Cu 2+ and Zn 2+ having the strongest effect on PS-NH 2 . Theoretical computations indicated that π-π and electrostatic interactions dominated PS adsorption of GAT, while PS-COOH and PS-NH 2 adsorbed GAT through electrostatic interactions, hydrogen bonds, and van der Waals (vdW) forces. The surface electrostatic potential of PS-COOH and PS-NH 2 was considerably higher than that of PS, with the maximum vdW penetration distance of GAT-PS-NH 2 being 1.20 Å. Abstract

As the hybrid between cattle and yak, cattleyak is a typical male sterile mammal, and the underlying mechanism for its spermatogenic arrest is still unclear. In this study, the coding region of cattleyak TAF4B gene was cloned by RT-PCR and analysed by bioinformatics. To investigate the effects of TAF4B on cellular proliferation and differentiation, an expression vector was generated and introduced into undifferentiated spermatogonia (UDSPG) of cattleyak. The results showed that the protein encoded by TAF4B did not contain the signal peptide sequence. The expression level of TAF4B in UDSPG of cattleyak was lower than that in yak, while the overexpression of TAF4B in cattleyak promoted the proliferation activity of cattleyak UDSPG. Meanwhile, the expression of proliferation and meiosis-related genes was increased but the differentiation-related genes were decreased. Therefore, the aberrant expression of TAF4B in cattleyak UDSPG possibly impaired its proliferation and differentiation equilibrium and decreased its growth potentiality, thereby reducing the quantity of UDSPG and affecting spermatogenesis. Abstract

Antibiotics are a widely distributed and effective antibacterial agents. Human medical treatment and livestock aquaculture are major sources of antibiotics in aquatic ecosystems, potentially damaging the biofilms that are the foundation of stream food webs. In this study, we conducted antibiotic biotransformation experiments using biofilms cultured in streams upstream and downstream of a wastewater treatment plant outlet to distinguish different fate processes of antibiotics in biofilms. It was found that stream biofilms have biotransformation potential mainly for specific sulfonamide antibiotics. Flavobacterium and Dyadobacter were identified to be associated with biofilm biotransformation of antibiotics by 16S rDNA sequencing. Besides, microorganisms released from treated wastewater integrated into downstream biofilm communities, thereby enhancing the biotransformation potential of downstream biofilm communities compared to upstream biofilm.
