Due to ongoing climate change, methane (CH4) emissions from vegetated wetlands are projected to increase during the 21st century, challenging climate mitigation efforts aimed at limiting global warming. However, despite reports of rising emission trends, a comprehensive evaluation and attribution of recent changes remains limited. Here we assessed global wetland CH4 emissions from 2000–2020 based on an ensemble of 16 process-based wetland models. Our results estimated global average wetland CH4 emissions at 158 ± 24 (mean ± 1σ) Tg CH4 yr−1 over a total annual average wetland area of 8.0 ± 2.0×106 km2 for the period 2010–2020, with an average increase of 6–7 Tg CH4 yr−1 in 2010–2019 compared to the average for 2000–2009. The increases in the four latitudinal bands of 90–30° S, 30° S–30° N, 30–60° N, and 60–90° N were 0.1–0.2, 3.6–3.7, 1.8–2.4, and 0.6–0.8 Tg CH4 yr−1, respectively, over the 2 decades. The modeled CH4 sensitivities to temperature show reasonable consistency with eddy-covariance-based measurements from 34 sites. Rising temperature was the primary driver of the increase, while precipitation and rising atmospheric CO2 concentrations played secondary roles with high levels of uncertainty. These modeled results suggest that climate change is driving increased wetland CH4 emissions and that direct and sustained measurements are needed to monitor developments.

Background and purpose: As preparation for a national randomized study comparing proton radiotherapy to photon radiotherapy, DAHANCA 35, we performed a non-randomized pilot study to investigate patient selection, logistics, planning, and treatment delivery. With the present study, as a comprehensive safety analysis, we want to compare toxicity during and up to two months after therapy to a historically matched group of patients treated with photon radiotherapy. Materials and methods: 62 patients treated with protons were matched to 124 patients who received photon treatment outside a protocol. Available data were retrieved from the DAHANCA database. Patients were matched on treatment centre, concurrent chemotherapy, tumour site, stage, p16 status for oropharynx cancers. Selection of patients for proton therapy was based on comparative treatment plans with a NTCP reduction for dysphagia and xerostomia at six months. Results: Baseline characteristics between groups were well balanced, except for the type of drug used concurrently; more photon patients received Carboplatin (21.2 % vs 5.8 %, p = 0.01). Salt marshes play an important role in the global carbon cycle due to the large amount of organic carbon stored in their soils. Soil organic carbon formation in these coastal wetland ecosystems is strongly controlled by the plant primary production and initial decomposition rates of plant belowground biomass and litter. This study used a field warming experiment to investigate the response of belowground litter breakdown to rising temperature (+1.5 and +3.0 ∘C) across whole-soil profiles (0–60 cm soil depth) and the entire intertidal flooding gradient ranging from the pioneer zone via the low marsh to high marsh. We used standardized plant materials, following the Tea Bag Index approach, to assess the initial decomposition rate (k) and the stabilization factor (S) of labile organic matter inputs to the soil system. While k describes the initial pace at which labile (= hydrolyzable) organic matter decomposes, S describes the part of the labile fraction that does not decompose during deployment in the soil system and stabilizes due to biochemical transformation. We show that warming strongly increased k consistently throughout the entire soil profile and across the entire flooding gradient, suggesting that warming effects on the initial decomposition rate of labile plant materials are independent of the soil aeration (i.e., redox) status. By contrast, negative effects on litter stabilization were less consistent. Specifically, warming effects on S were restricted to the aerated topsoil in the frequently flooded pioneer zone, while the soil depth to which stabilization responded increased across the marsh elevation gradient via the low to high marsh. These findings suggest that reducing soil conditions can suppress the response of belowground litter stabilization to rising temperature. In conclusion, our study demonstrates marked differences in the response of initial decomposition rate vs. stabilization of labile plant litter to rising temperature in salt marshes. We argue that these differences are strongly mediated by the soil redox status along flooding and soil-depth gradients.

Lycopsids represent a distinct lineage of vascular plants with a long evolutionary history including numerous extant and extinct species that started out as tiny herbaceous plants and later went on to grow into forests with tree-like structures. They enriched the soil carbon pool through newly developed root-like structures and promoted soil microbial activity by providing organic matter. These plants enhanced soil carbon dioxide (CO2) via root respiration and also modified soil hydrology. These effects potentially promoted the dissolution of silicate minerals, thus intensifying silicate weathering. The weathering of silicate rocks is considered one of the most significant geochemical regulators of atmospheric CO2 over a long (hundreds of thousands to millions of years) timescale. The motivation for this study is to achieve an increased understanding of the realized impacts of vascular plants, represented by modern relatives of the most basal plants with vascular tissues and shallow root systems, on silicate weathering and past climate. To this end, it is necessary to quantify physiological characteristics, spatial distribution, carbon balance, and the hydrological impacts of early lycopsids. These properties, however, cannot be easily derived from proxies such as fossil records. Hence, as a first step, a process-based model is developed to estimate net carbon uptake by these organisms at the local scale. The model includes key features such as the distribution of biomass above and below ground, along with a plausible root distribution in the soil affecting water uptake by plants. The stomatal regulation of water loss and its immediate implications for photosynthesis are considered. Moreover, root respiration plays a crucial role in the model by affecting soil carbon dioxide and weathering rates. The model features ranges of key physiological traits of lycopsids to predict the emerging characteristics of the Lycopsida class community under any given climate by implicitly simulating the process of selection. In this way, extinct plant communities can also be represented. In addition to physiological properties, the model also simulates weathering rates using a simple limit-based approach and estimates the biotic enhancement of weathering by these plants. We run the Lycopsid model, called LYCOm, at seven sites encompassing various climate zones under today's climatic conditions. LYCOm can simulate realistic properties of lycopsid communities at the respective locations and estimates values of net primary production (NPP) ranging from 126 to 245 g carbon m−2 yr−1. Our limit-based weathering model predicts a mean chemical weathering rate ranging from 5.3 to 45.1 cm ka−1 of rock with lycopsids varying between different sites, as opposed to 0.6–8.3 cm ka−1 of rock without these plants, thereby highlighting the potential importance of such vegetation at the local scale for enhancing chemical weathering. Our modeling study establishes a basis for assessing the biotic enhancement of weathering by lycopsids at the global scale and also for the geological past. Although our method is associated with limitations and uncertainties, it represents a novel, complementary approach towards estimating the impacts of lycopsids on biogeochemistry and climate.

Abstract Aim The aim was to assess safety and feasibility of Hybrid High-Intensity Interval Training (HIIT) using Functional Electrical Stimulation (FES) leg cycling and arm ski ergometer in people with Spinal Cord Injuries (SCI). Method Eight outpatients (mean age 42.8 years; 7 men) with stable SCI paraplegia (mean 14.5 years since injury) participated in hybrid HIIT (90% peak watts; 4 × 4–min intervals), three times a week (over 8 weeks). Primary outcomes were Adverse Events (AE), participant acceptability, shoulder pain, training intensity (% peak watts), and attendance. Secondary outcomes were effect on peak oxygen uptake (VO2peak) during FES hybrid poling, mean watts, self-reported leisure time physical activity, quality of life, and fatigue. Results No serious AE occurred; acceptability with the training modality was high, while shoulder pain increased by 9% (SD 95.2). During training, 50% of the participants reached > 90% peak watts during the intervals, three with the legs (FES cycle) and one with the arms (Ski-Erg). Overall, mean training intensity (% peak watts) was 92% (SD 18.9) for legs and 82% (SD 10.3) for arms. Proportion of fulfilled training minutes was 82% (range 36–100%); one participant dropped out after 6 weeks due to back pain. Mean VO2peak increased by 17% (SD 17.5). Participants reported increased leisure time physical activity and health-related quality of life, besides reduced fatigue. Conclusion Hybrid HIIT was safe for people with SCI paraplegia. The majority of the criteria for feasibility were met with acceptable attendance rate, limited drop out, participants enjoyed training, and increased VO2peak and mean watts. However, the intensity of 90% peak watts was reached by < 60% of the participants despite high RPE ratings during training. The method of measuring and calculating intensity needs to be studied further before a study using this HIIT protocol is undertaken. Climate change can strongly alter soil microbial functioning via plant–microbe interactions, often with important consequences for ecosystem carbon and nutrient cycling. Given the high degree of intraspecific trait variability in plants, it has been hypothesized that genetic shifts within plant species yield a large potential to control the response of plant–microbe interactions to climate change. Here we examined if sea-level rise and plant genotype interact to affect soil microbial communities in an experimental coastal wetland system, using two known genotypes of the dominant salt-marsh grass Elymus athericus characterized by differences in their sensitivity to flooding stress – i.e., a tolerant genotype from low-marsh environments and an intolerant genotype from high-marsh environments. Plants were exposed to a large range of flooding frequencies in a factorial mesocosm experiment, and soil microbial activity parameters (exo-enzyme activity and litter breakdown) and microbial community structure were assessed. Plant genotype mediated the effect of flooding on soil microbial community structure and determined the presence of flooding effects on exo-enzyme activities and belowground litter breakdown. Larger variability in microbial community structure, enzyme activities, and litter breakdown in soils planted with the intolerant plant genotype supported our general hypothesis that effects of climate change on soil microbial activity and community structure can depend on plant intraspecific genetic variation. In conclusion, our data suggest that adaptive genetic variation in plants could suppress or facilitate the effects of sea-level rise on soil microbial communities. If this finding applies more generally to coastal wetlands, it yields important implications for our understanding of ecosystem–climate feedbacks in the coastal zone.

Seasonal and interannual variations in global wetland area are a strong driver of fluctuations in global methane (CH4) emissions. Current maps of global wetland extent vary in their wetland definition, causing substantial disagreement between and large uncertainty in estimates of wetland methane emissions. To reconcile these differences for large-scale wetland CH4 modeling, we developed the global Wetland Area and Dynamics for Methane Modeling (WAD2M) version 1.0 dataset at a ∼ 25 km resolution at the Equator (0.25∘) at a monthly time step for 2000–2018. WAD2M combines a time series of surface inundation based on active and passive microwave remote sensing at a coarse resolution with six static datasets that discriminate inland waters, agriculture, shoreline, and non-inundated wetlands. We excluded all permanent water bodies (e.g., lakes, ponds, rivers, and reservoirs), coastal wetlands (e.g., mangroves and sea grasses), and rice paddies to only represent spatiotemporal patterns of inundated and non-inundated vegetated wetlands. Globally, WAD2M estimates the long-term maximum wetland area at 13.0×106 km2 (13.0 Mkm2), which can be divided into three categories: mean annual minimum of inundated and non-inundated wetlands at 3.5 Mkm2, seasonally inundated wetlands at 4.0 Mkm2 (mean annual maximum minus mean annual minimum), and intermittently inundated wetlands at 5.5 Mkm2 (long-term maximum minus mean annual maximum). WAD2M shows good spatial agreements with independent wetland inventories for major wetland complexes, i.e., the Amazon Basin lowlands and West Siberian lowlands, with Cohen's kappa coefficient of 0.54 and 0.70 respectively among multiple wetland products. By evaluating the temporal variation in WAD2M against modeled prognostic inundation (i.e., TOPMODEL) and satellite observations of inundation and soil moisture, we show that it adequately represents interannual variation as well as the effect of El Niño–Southern Oscillation on global wetland extent. This wetland extent dataset will improve estimates of wetland CH4 fluxes for global-scale land surface modeling. Introduction: The aim was to characterise patterns and predictability of aeration changes in the ipsilateral maxillary sinus during intensity-modulated radiotherapy (IMRT) for sinonasal cancer (SNC), and in a sample evaluate the dosimetric effects of aeration changes for both photon and proton therapy. Materials and methods: The study included patients treated with IMRT for SNC in a single institution in 2009–2017. The volume of air in the ipsilateral maxillary sinus was recorded in 1578 daily cone beam computer tomography (CBCT) from 53 patients. Patterns of changing air volumes were categorised as ‘stable’, increasing’, ‘decreasing’, or ‘erratic’. For the prediction analysis, categorisation was performed based both on the entire treatment course and the first five fractions (F1–5). Photon and proton therapy plans were generated for four patients, the one from each category with the largest aeration variation. Synthetic CT images were generated for each CBCT and all plans were recalculated on the daily synthetic CTs. Results: The absolute volume of air varied considerably during the treatment course, ranging from 0 to 25.9 cm3. Changes within a single participant varied in the range of 0–18.7 cm3. In the categorisation of patterns, most patients had increasing aeration of the sinus. Generally, patterns of aeration could not be predicted from F1–5. Patients categorised as increasing in F1–5 had the best prediction, with 78% predicted correctly as increasing for the entire treatment course. The numeric correlation coefficients for target coverage and air volume were low for 3/4 scenarios (photons 0.03–0.23, protons 0.26–0.48). No straightforward correlation between the dosimetric effect and the volume changes could be detected in the sample test of four patients for neither photon nor proton therapy. Conclusion: The variation of aeration was large and unpredictable. No clear dosimetric consequences of the aeration variation were evident for neither IMRT nor proton therapy for the patients investigated.

The persistence of tidal wetland ecosystems like salt marshes is threatened by human interventions and climate change. In particular, the threat of accelerated sea level rise (SLR) has increasingly gained the attention of the scientific community recently. However, studies investigating the effect of SLR on plants and vertical marsh accretion are usually restricted to the species or community level and do not consider phenotypic plasticity or genetic diversity. To investigate the response of genotypes within the same salt-marsh species to SLR, we used two known genotypes of Elymus athericus (Link) Kerguélen (low-marsh and high-marsh genotypes). In a factorial marsh organ experiment we exposed both genotypes to different flooding frequencies and quantified plant growth parameters. With increasing flooding frequency, the low-marsh genotype showed higher aboveground biomass production compared to the high-marsh genotype. Additionally, the low-marsh genotype generally formed longer rhizomes, shoots and leaves, regardless of flooding frequency. Belowground biomass of both genotypes decreased with increasing flooding frequency. We conclude that the low-marsh genotype is better adapted to higher flooding frequencies through its ability to allocate resources from below- to aboveground biomass. Given the strong control of plant biomass production on salt-marsh accretion, we argue that these findings yield important implications for our understanding of ecosystem resilience to SLR as well as plant species distribution in salt marshes.

Purpose: The aim of the study was to evaluate neurocognitive late effects, structural alterations and associations between cognitive impairment and radiation doses as well as cerebral tissue damage after radiotherapy for sinonasal cancer. Furthermore, the aim was to report quality of life (QoL) and self-reported cognitive capacity. Materials and methods: Recurrence-free patients previously treated with intensity-modulated radiotherapy with a curative intent were eligible for the study. Study examinations comprised comprehensive neurocognitive testing, MRI of the brain, and self-reported outcomes. Results: A total of 27 patients were included. Median age was 67 years (range 47–83). The majority of test outcomes were below normative values in any degree, and 37% of the participants had clinically significant neurocognitive impairment when compared with normative data. Correlations between absorbed doses to specific substructures of the brain and neurocognitive outcomes were present for Wechsler’s Adult Intelligence Scale-digit span and Controlled Oral Word Association Test-S. Structural MRI revealed macroscopic abnormalities in three patients; infarction (n = 1), diffuse white matter intensities (n = 2) and necrosis (n = 1). In the analysis of atrophy of cerebral tissue, no correlations were present with neither radiation dose to cerebral substructures nor neurocognitive impairment. The global QoL of the cohort was 75. The most affected outcomes were ‘fatigue’, ‘insomnia’, and ‘drowsiness’. A total of 59% of participants reported significantly impaired quality of sleep. Self-reported cognitive function revealed that ‘memory’ was the most affected cognitive domain. For the domains of ‘memory’ and ‘language’, self-reported functioning was associated with objectively measured neurocognitive outcomes. Conclusion: Cerebral toxicity after radiotherapy for sinonasal cancer was substantial. Clinically significant cognitive impairment was present in more than one third of the participants, and several dose–response associations were present. Furthermore, the presence of macroscopic radiation sequelae indicated considerable impact of radiotherapy on brain tissue.

Tidal wetlands, such as tidal marshes and mangroves, are hotspots for carbon sequestration. The preservation of organic matter (OM) is a critical process by which tidal wetlands exert influence over the global carbon cycle and at the same time gain elevation to keep pace with sea-level rise (SLR). The present study assessed the effects of temperature and relative sea level on the decomposition rate and stabilization of OM in tidal wetlands worldwide, utilizing commercially available standardized litter. While effects on decomposition rate per se were minor, we show strong negative effects of temperature and relative sea level on stabilization, as based on the fraction of labile, rapidly hydrolyzable OM that becomes stabilized during deployment. Across study sites, OM stabilization was 29 % lower in low, more frequently flooded vs. high, less frequently flooded zones. Stabilization declined by ∼ 75 % over the studied temperature gradient from 10.9 to 28.5 °C. Additionally, data from the Plum Island long-term ecological research site in Massachusetts, USA, show a pronounced reduction in OM stabilization by > 70 % in response to simulated coastal eutrophication, confirming the potentially high sensitivity of OM stabilization to global change. We therefore provide evidence that rising temperature, accelerated SLR, and coastal eutrophication may decrease the future capacity of tidal wetlands to sequester carbon by affecting the initial transformations of recent OM inputs to soil OM.

Introduction: Pharyngeal and laryngeal cancers are highly curable; however survivors are at high risk for long-term dysphagia after radiation. To address lack of access to preventive care in community settings, we developed a responsive web-based application to help patients adhere to preventive swallowing exercises and cope with radiation side effects. We conducted an interim study analysis to determine website usage characteristics and to examine the effect size for future trials. Methods: Pharyngeal and laryngeal cancer patients were recruited for enrollment by speech language pathologists before primary radiation and introduced to the interactive website. The program (English and Spanish) features tracking logs for preventive exercises, instructional videos, patient stories and search features. Patients' self-reported swallowing function was assessed with the MD Anderson Dysphagia Inventory (MDADI) at baseline and at 6 months. Adherence to preventive exercises was assessed during the 10 week intervention. Number of unique website visits, total duration of website exposure, and rankings of the most popular webpages were calculated. Preliminary regression models were run using adherence and MDADI as outcomes. Results: Of the 160 enrolled, 96 had 10-week adherence data and 61 had 6-month MDADI data. The average age was 63 (SD = 12.26), 49.4% were from rural counties, 44% had a high school education or lower, and 42% reported annual income of $30,000 or less. The average number of visits was 5.49 (SD = 9.96) and the average total time spent with the website was 41.09 min (SD =88.48). Preliminary analyses indicated that number of unique visits to the website was independently associated with increased adherence to preventive exercises (p = .001–.008). Conclusion: Our website showed significant effects in promoting adherence to swallowing exercises. However, our return visit rate showed that the platform needs improvement in navigability and usability for this older population undergoing challenging treatment in community settings with low resources. Synthetic biology dictates the data-driven engineering of biocatalysis, cellular functions, and organism behavior. Integral to synthetic biology is the aspiration to efficiently find, access, interoperate, and reuse high-quality data on genotype-phenotype relationships of native and engineered biosystems under FAIR principles, and from this facilitate forward-engineering strategies. However, biology is complex at the regulatory level, and noisy at the operational level, thus necessitating systematic and diligent data handling at all levels of the design, build, and test phases in order to maximize learning in the iterative design-build-test-learn engineering cycle. To enable user-friendly simulation, organization, and guidance for the engineering of biosystems, we have developed an open-source python-based computer-aided design and analysis platform operating under a literate programming user-interface hosted on Github. The platform is called teemi and is fully compliant with FAIR principles. In this study we apply teemi for i) designing and simulating bioengineering, ii) integrating and analyzing multivariate datasets, and iii) machine-learning for predictive engineering of metabolic pathway designs for production of a key precursor to medicinal alkaloids in yeast. The teemi platform is publicly available at PyPi and GitHub.

Misfolding of the prion protein (PrP) is the key step in the transmission of spongiform pathologies in humans and several animals. Although PrP is highly conserved in mammals, a few changes in the sequence of endogenous PrP are proposed to confer protection to dogs, which were highly exposed to prion during the mad-cow epidemics. D159 is a unique amino acid found in PrP from dogs and other canines that was shown to alter surface charge, but its functional relevance has never been tested in vivo. Here, we show in transgenic Drosophila that introducing the N159D substitution on mouse PrP decreases its turnover. Additionally, mouse PrP-N159D demonstrates no toxicity and accumulates no pathogenic conformations, suggesting that a single D159 substitution is sufficient to prevent PrP conformational change and pathogenesis. Understanding the mechanisms mediating the protective activity of D159 is likely to lessen the burden of prion diseases in humans and domestic animals.

Cold atomic ensembles have been considered suitable platforms to realize quantum memories, but their scalability is limited by the cooling apparatuses. Here, the authors show how room temperature atomic microcells can be used for discrete variable ensemble-based quantum information processing.
