One of the issues faced by future fusion devices will be high divertor target heat loads. Alternative divertors can promote detachment, flux expansion and dissipation mechanisms to mitigate these heat loads. They have been investigated in several devices including TCV and DIII-D, and will be investigated on MAST-U. To evaluate their effectiveness, accurate target heat flux and power balance measurements are required in these machines. Infrared (IR) thermography is a widely used technique to determine the target heat flux, but is susceptible to surface effects and emissivity in carbon-walled machines. In this work, the effect of plasma exposure on graphite is assessed to understand what may happen in MAST-U. A sample of fine grain graphite, as used on MAST-U, is exposed to 30min plasma exposures, with density ne=6 × 1018m−3 and temperature Te=0.08eV as measured by Thomson scattering. During these pulses, the temperature is measured by a medium wave IR camera and is seen to decrease by ≈70 °C over the course of 3h of plasma exposure. Pyrometer measurements suggest that the IR camera data is affected by a change in the surface emissivity. Profilometry confirms erosion of graphite at the tile centre to a depth of ≈100µm, and a larger region of deposition further out, amounting to ≈40µm of material.

We present results of the design, implementation and testing of a Bayesian multi-diagnostic inference system which combines various divertor diagnostics to infer the 2D fields of electron temperature T e , density n e and deuterium neutral density n 0 in the divertor. The system was tested using synthetic diagnostic measurements derived from SOLPS-ITER fluid code predictions of the MAST-U Super-X divertor which include appropriate added noise. Two SOLPS-ITER simulations in different states of detachment, taken from a scan of the nitrogen seeding rate, were used as test-cases. Taken across both test-cases, the median absolute fractional errors in the inferred electron temperature and density estimates were 10.3% and 10.1% respectively. Differences between the inferred fields and the test-cases were well explained by solution uncertainty estimates derived from posterior sampling. This work represents a step toward a larger goal of obtaining a quantitative, 2D description of the divertor plasma state directly from experimental data, which could be used to gain better understanding of divertor physics phenomena.

The mega amp spherical tokamak (MAST) was a low aspect ratio device (R/a = 0.85/0.65 ~ 1.3) with similar poloidal cross-section to other medium-size tokamaks. The physics programme concentrates on addressing key physics issues for the operation of ITER, design of DEMO and future spherical tokamaks by utilising high resolution diagnostic measurements closely coupled with theory and modelling to significantly advance our understanding. An empirical scaling of the energy confinement time that favours higher power, lower collisionality devices is consistent with gyrokinetic modelling of electron scale turbulence. Measurements of ion scale turbulence with beam emission spectroscopy and gyrokinetic modelling in up-down symmetric plasmas find that the symmetry of the turbulence is broken by flow shear. Near the non-linear stability threshold, flow shear tilts the density fluctuation correlation function and skews the fluctuation amplitude distribution. Results from fast particle physics studies include the observation that sawteeth are found to redistribute passing and trapped fast particles injected from neutral beam injectors in equal measure, suggesting that resonances between the m = 1 perturbation and the fast ion orbits may be playing a dominant role in the fast ion transport. Measured D–D fusion products from a neutron camera and a charged fusion product detector are 40% lower than predictions from TRANSP/NUBEAM, highlighting possible deficiencies in the guiding centre approximation. Modelling of fast ion losses in the presence of resonant magnetic perturbations (RMPs) can reproduce trends observed in experiments when the plasma response and charge-exchange losses are accounted for. Measurements with a neutral particle analyser during merging-compression start-up indicate the acceleration of ions and electrons. Transport at the plasma edge has been improved through reciprocating probe measurements that have characterised a geodesic acoustic mode at the edge of an ohmic L-mode plasma and particle-in-cell modelling has improved the interpretation of plasma potential estimates from ball-pen probes. The application of RMPs leads to a reduction in particle confinement in L-mode and H-mode and an increase in the core ionization source. The ejection of secondary filaments following type-I ELMs correlates with interactions with surfaces near the X-point. Simulations of the interaction between pairs of filaments in the scrape-off layer suggest this results in modest changes to their velocity, and in most cases can be treated as moving independently. A stochastic model of scrape-off layer profile formation based on the superposition of non-interacting filaments is in good agreement with measured time-average profiles. Transport in the divertor has been improved through fast camera imaging, indicating the presence of a quiescent region devoid of filament near the X-point, extending from the separatrix to ψ n ~ 1.02. Simulations of turbulent transport in the divertor show that the angle between the divertor leg on the curvature vector strongly influences transport into the private flux region via the interchange mechanism. Coherence imaging measurements show counter-streaming flows of impurities due to gas puffing increasing the pressure on field lines where the gas is ionised. MAST Upgrade is based on the original MAST device, with substantially improved capabilities to operate with a Super-X divertor to test extended divertor leg concepts. SOLPS-ITER modelling predicts the detachment threshold will be reduced by more than a factor of 2, in terms of upstream density, in the Super-X compared with a conventional configuration and that the radiation front movement is passively stabilised before it reaches the X-point. 1D fluid modelling reveals the key role of momentum and power loss mechanisms in governing detachment onset and evolution. Analytic modelling indicates that long legs placed at large major radius, or equivalently low at the target compared with the X-point are more amenable to external control. With MAST Upgrade experiments expected in 2019, a thorough characterisation of the sources of the intrinsic error field has been carried out and a mitigation strategy developed.

New results from MAST are presented that focus on validating models in order to extrapolate to future devices. Measurements during start-up experiments have shown how the bulk ion temperature rise scales with the square of the reconnecting field. During the current ramp up models are not able to correctly predict the current diffusion. Experiments have been performed looking at edge and core turbulence. At the edge detailed studies have revealed how filament characteristic are responsible for determining the near and far SOL density profiles. In the core the intrinsic rotation and electron scale turbulence have been measured. The role that the fast ion gradient has on redistributing fast ions through fishbone modes has led to a redesign of the neutral beam injector on MAST Upgrade. In H-mode the turbulence at the pedestal top has been shown to be consistent with being due to electron temperature gradient modes. A reconnection process appears to occur during ELMs and the number of filaments released determines the power profile at the divertor. Resonant magnetic perturbations can mitigate ELMs provided the edge peeling response is maximised and the core kink response minimised. The mitigation of intrinsic error fields with toroidal mode number n>1 has been shown to be important for plasma performance., 34 pages, 10 figures. Disruptions pose a significant challenge in future devices where the increased stored energy can lead to unacceptably large transient heat loads on plasma facing components (PFCs). One means of mitigating disruptions is that of massive gas injection (MGI), which produces a radiative collapse of the plasma discharge through the injection of impurity gases. The MAST disruption mitigation system is capable of injecting up to 1.95 bar litres into the MAST vacuum vessel over a timescale of 1–2 ms, corresponding to a particle inventory of 5 × 10 22 , around 100 times the plasma particle inventory. High speed infrared thermography, offering full divertor coverage, has shown a 60–70% reduction in divertor power loads during mitigation. A combination of high temporal (0.2 ms) and spatial resolution (1 cm) Thomson scattering and soft X-ray camera array data show evidence for a cooling front associated with the inward propagation of the injected impurities.

Hydrophilic chelating polymers have been prepared by the reaction of organic compounds containing both a reactive halogen and a chelating function, with cellulose and, more especially, polyvinyl alcohol or their alkali metal derivatives. Suitable organic compounds used to illustrate the general reaction included: 5-chloromethyl-8-hydroxyquinoline, 5-chloro-8-hydroxyquinoline, ethyl γ-chloroacetoacetate and chloromethyl-kojic acid. Subsequent reaction of the kojic acid-polyvinyl alcohol polymer with diazotised o-arsanilic acid to give a polymer containing the azo-phenylarsonic acid group indicates further scope for these materials. Preliminary tests have shown that the polymers remove metal ions from aqueous solutions. Degradation occurred, however, when the polymers were regenerated with acids to elute adsorbed metal ions.

Photon cleaning by means of a flash-lamp was used for in-situ detritiation of the inner wall tiles of the JET divertor in May 2004. Additional trials were also performed ex-situ in October 2004 on divertor base tiles. Early work confirmed that for pulse energies between 150 J and 300 J some deposited material was removed. To increase the amount of material removed during photon cleaning, further experiments with higher pulse energies (500 J) were performed and are reported here. Analysis of cross sections confirmed a removal rate of 0.04 μm/pulse, removing ∼80 μm from 200 μm thick deposits over a treatment area of 15 × 10 −4 m 2 . During the photon cleaning tests at least 12% of the tritium inventory for the tile was removed. It was also shown that deuterium was desorbed from a depth ∼7 μm beyond the depth of material removed.

The use of carbon-based materials for first wall components in tokamaks results in the formation of hydrocarbon deposits on divertor components that could lead to a high level of tritium retention in future fusion devices. Experiments at UMIST have demonstrated that photonic cleaning using high power Xenon flashlamp sources is an efficient method for removing such films and represents a good candidate technology for international thermonuclear experimental reactor (ITER) operations. Studies have shown that effective film removal occurs at a fluence threshold of between 1.9 and 2.5 J/cm2. The by-products of the cleaning process, both particulates and gases, have been characterised using particle sizing spectrometry and quadrupole mass spectrometry respectively. It is found that hydrogen, methane, acetylene, ethylene, ethane and carbon dioxide are the principal gaseous products produced during the cleaning process, which also produces a significant fraction of particulates in the size range 2–20 μm.

We report on studies of detached plasma operation in the UMIST linear system (ULS). The ULS, designed to study a range of edge physics issues relevant to tokamak divertors, is capable of producing plasmas with electron densities and temperatures in the range 10 17 –10 19 m −3 and 2–15 eV, respectively. Previous studies of the interaction between the hydrogen plasma and low-pressure hydrogen gas have identified a regime where molecular activated recombination (MAR) processes dominate plasma losses. Here we report on studies in which the upstream plasma parameters are varied such that three-body and radiative electron–ion recombination (EIR) dominates. Initial modelling of the recombination region is undertaken using a simplified version of the one-dimensional electron energy and continuity equations. We determine the factors that govern the threshold between MAR and EIR dominated detached regimes in terms of upstream plasma parameters and compare this with predictions based on the competing effects of electron cooling and recombination.

The Mega Ampère Spherical Tokamak (MAST) programme is strongly focused on addressing key physics issues in preparation for operation of ITER as well as providing solutions for DEMO design choices. In this regard, MAST has provided key results in understanding and optimizing H-mode confinement, operating with smaller edge localized modes (ELMs), predicting and handling plasma exhaust and tailoring auxiliary current drive. In all cases, the high-resolution diagnostic capability on MAST is complemented by sophisticated numerical modelling to facilitate a deeper understanding. Mitigation of ELMs with resonant magnetic perturbations (RMPs) with toroidal mode number n RMP = 2, 3, 4, 6 has been demonstrated: at high and low collisionality; for the first ELM following the transition to high confinement operation; during the current ramp-up; and with rotating n RMP = 3 RMPs. n RMP = 4, 6 fields cause less rotation braking whilst the power to access H-mode is less with n RMP = 4 than n RMP = 3, 6. Refuelling with gas or pellets gives plasmas with mitigated ELMs and reduced peak heat flux at the same time as achieving good confinement. A synergy exists between pellet fuelling and RMPs, since mitigated ELMs remove fewer particles. Inter-ELM instabilities observed with Doppler backscattering are consistent with gyrokinetic simulations of micro-tearing modes in the pedestal. Meanwhile, ELM precursors have been strikingly observed with beam emission spectroscopy (BES) measurements. A scan in beta at the L–H transition shows that pedestal height scales strongly with core pressure. Gyro-Bohm normalized turbulent ion heat flux (as estimated from the BES data) is observed to decrease with increasing tilt of the turbulent eddies. Fast ion redistribution by energetic particle modes depends on density, and access to a quiescent domain with ‘classical’ fast ion transport is found above a critical density. Highly efficient electron Bernstein wave current drive (1 A W−1) has been achieved in solenoid-free start-up. A new proton detector has characterized escaping fusion products. Langmuir probes and a high-speed camera suggest filaments play a role in particle transport in the private flux region whilst coherence imaging has measured scrape-off layer (SOL) flows. BOUT++ simulations show that fluxes due to filaments are strongly dependent on resistivity and magnetic geometry of the SOL, with higher radial fluxes at higher resistivity. Finally, MAST Upgrade is due to begin operation in 2016 to support ITER preparation and importantly to operate with a Super-X divertor to test extended leg concepts for particle and power exhaust.

We present the pedestal structure, as determined from the high-resolution Thomson scattering measurements, for a database of low and high triangularity (????0.22?0.39) 2.5?MA, type I ELMy H-mode JET plasmas after the installation of the new ITER-like wall (JET-ILW). The database explores the effect of increasing deuterium fuelling and nitrogen seeding with a view to explain the observed changes in performance (edge and global). The low triangularity JET-ILW plasmas show no significant change in performance and pedestal structure with increasing gas dosing. These results are in good agreement with EPED1 predictions. At high triangularity, for pure deuterium fuelled JET-ILW plasmas, there is a 20?30% reduction in global performance and pressure pedestal height in comparison to JET-C plasmas. This reduction in performance is primarily due to a degradation of the temperature pedestal height. The global performance and pressure pedestal height of JET-ILW plasmas can be partially recovered to that of JET-C plasmas with additional nitrogen seeding (Giroud et al 2013 Nucl. Fusion 53 113025). This observed improvement in performance is predominately due to a significant increase in density pedestal height as well as a small increase in the temperature pedestal height. A key result with increasing deuterium fuelling for JET-ILW plasmas is there is no improvement in pressure pedestal height however the pedestal still widens which is inconsistent with the scaling. Furthermore, a key result with increasing nitrogen seeding is the pressure pedestal widening is due to an increase in the temperature pedestal width whilst the density pedestal shows no clear trend. The comparison of EPED1 predictions with the measurements at high triangularity is complex as, for example, for pure deuterium fuelled plasmas there is very good agreement for the pedestal height but not the width. In addition, current EPED1 runs under-predict the pedestal height and width at high nitrogen seeding for JET-ILW plasmas however further work is required to determine the significance of these deviations. Understanding these deviations is essential as provides an insight to the physical mechanisms governing the pedestal structure and edge performance.

Physical activity has been associated with decreased risk for developing breast cancer yet to date, the mechanism remains unknown. The purpose of this investigation was to evaluate the effects of moderate exercise training on the normal mammary gland in an attempt to identify alterations or differences that might be associated with tumour inhibition. A total of 170 female Sprague-Dawley rats were randomized to baseline (n = 10), exercise (EX; n = 80), or sham-exercise groups (SHAM; n = 80). Treadmill training (20-25 m min -1 , 15% grade, 30 min day -1 , 5 days week -1 ) was started at 28 days of age (DOA). Animals were killed at 28, 42, 56, 70 and 84 DOA. Mammary glands were evaluated by histology and immunohistochemistry. Terminal end buds (TEB), structures susceptible to carcinogenesis, were counted. Sexual maturation, estradiol and progesterone, and organ and muscle weights were also evaluated. No differences in growth, sexual maturation, or steroid hormones were observed in response to training. No difference in the number of TEBs was observed at any timepoint between EX and SHAM. Proliferation was significantly increased at 56 DOA and tended to be increased at 42 and 70 DOA in the EX animals whereas cell death was significantly increased at 70 DOA and tended to be increased at 84 DOA in the EX animals. These data suggest no difference in the number of carcinogen-susceptible structures as a result of moderate exercise. The changes in cell proliferation and apoptosis with exercise training suggest altered cell turnover that will necessitate future study particularly with relevance to carcinogenesis.

We report on further studies of current drive in the SPHEX spheromak device when operated as a spherical tokamak. Previous work has demonstrated that the current amplification factor, defined as the ratio of the toroidal plasma current to the driving gun current, increased linearly with the applied toroidal field. Results are presented from a more detailed analysis of this scaling for a fixed gun current of 60 kA, utilizing direct internal measurements of current density and equilibrium modelling of internal magnetic fields. The data indicate that the increase in the current amplification factor is largely the result of changes in the wavelength of the helical distortion of the open flux (identified with the global n = 1 mode) connected to the helicity injector: the increase in toroidal current on closed flux surfaces is much more modest. We discuss changes in current profiles at differing applied toroidal fields and demonstrate the operating regime over which helicity-injected current drive is effective in SPHEX. The implications of this work for proposed helicity injection schemes in the next generation of spherical tokamaks are discussed.

We describe an experiment, the UMIST Linear System (ULS), in which a hydrogen plasma stream, guided by a longitudinal magnetic field, is injected through a diaphragm containing an orifice into a separately-pumped target chamber in which the neutral hydrogen pressure can be raised to a maximum of 8 mTorr. The stream is about 6 mm in diameter, has an electron temperature of up to 15 eV and an ion flux of 3×1018 s-1; it is supersonic with Mach number up to M≈3. We have studied both the passage of the stream through the orifice and the interaction of the supersonic plasma with neutral hydrogen in the target chamber. We find that transmission is incomplete even when the orifice diameter is five times that of the plasma; we attribute this to the presence of ion trajectories which extend well outside the visible plasma and are intercepted by the diaphragm. In the target chamber, the stream does not broaden, but the ion flux decreases approximately exponentially with distance, with a scale length of the order of the mean free path for momentum transfer in ion-neutral collisions, and much less than that expected for other processes, such as charge exchange or electron-ion recombination. Elastic collisions alone cannot decrease the flux, but would lead to a large accumulation of slow ions in thermal equilibrium with the neutral gas, which must be limited by some other loss process: collisional diffusion and electron-ion recombination are too slow, leading to a density approaching 1020 m-3. The observed density is of the order of 1018 m-3, requiring a process with a rate of 10-100 times faster. Calculated rates for molecular-activated recombination (MAR) of the slow ions are of this order, and the predicted density agrees with our observations to order of magnitude.

Electrical currents in the scrape-off layer (SOL) of MAST are modelled using an interpretive Onion-Skin Model (OSM) constrained with experimental data from MAST diagnostics. The model was extended to include the effects of the magnetic mirror force, which has a strong influence on the particle and momentum balance in spherical tokamaks, such as MAST [1] . These modifications serve to more accurately model the parallel electric fields present in the MAST SOL, which can alter plasma dynamics via the E × B drift. Simulations show that the electrical current at the divertor targets is predominantly thermoelectric, whereas Pfirsch–Schluter currents have a greater contribution to the total current in the bulk of the SOL plasma.

Steady-state current drive by helicity injection has been demonstrated in the SPHEX spheromak. In order to understand the processes by which plasma toroidal current is driven on the closed flux, it is necessary first to diagnose the internal magnetic field and current structure. A method is described which fits force-free equilibria to magnetic field data from a linear insertable probe. It is shown that the overall field configuration and associated global parameters can be well fitted, though the detailed shape of the internal current profile is sensitive to small errors in the data and the choice of fitting function. The current drive process is closely associated with fluctuations having toroidal mode number n = 1. We investigate in detail shots with Ti gettering, when the n = 1 mode is episodic, showing that the current drive mechanism is switched on and off for periods during a single shot, corresponding to the presence or absence of the mode. This provides definitive evidence that the n = 1 mode is responsible for the current drive.

A neutral particle analyser (NPA) and high-resolution spectrometer have been used to make ion energy and rotation measurements during the driven and decaying phases of SPHEX, the UMIST spheromak. During the driven phase NPA spectra reveal the presence of a small (

In this paper we present in-situ satellite data, theory and laboratory validation that show how small scale collisionless shocks and mini-magnetospheres can form on the electron inertial scale length. The resulting retardation and deflection of the solar wind ions could be responsible for the unusual "lunar swirl" patterns seen on the surface of the Moon., 5 pages, 5 figures

Saccharomyces cerevisiae was used as a model system to characterize the covalent attachment of palmitoleic acid to proteins. Chemically synthesized cis-[9,10-3H]hexadecenoic acid (palmitoleic acid) was used to demonstrate the attachment of this lipid species to at least six proteins (m = 122, 58, 45, 41, 31, and 17 kDa). The majority of the labeled proteins are distinct from those labeled with [3H]palmitic acid (16:0). Based on the lability of the bond in the presence of methanolic KOH or hydroxylamine (pH 8), we propose that [3H] palmitoleic acid is attached to proteins via a thioester linkage. The identity of the palmitoleic acid was established by C-18 reverse phase high performance thin layer chromatography and argentation thin layer chromatography analysis after the fatty acid was liberated from the proteins by either transesterification or saponification. Incorporation of [3H]palmitoleic acid into proteins was only slightly inhibited (relative to [3H] myristic acid) by the presence of cycloheximide, indicating that the attachment of [3H]palmitoleic acid occurs post-translationally. This report is the first description of multiprotein acylation by a long chain unsaturated fatty acid.

The START (Small Tight Aspect Ratio Tokamak) experiment became operational in January 1991 at AEA Fusion Culham Laboratory. It is the only tokamak producing hot plasmas at aspect ratios as low as A=R/a approximately 1.3, and is providing valuable insight into the physical processes (such as toroidicity and trapping) involved at low aspect ratio. Three topics are discussed: equilibrium properties, including evidence of neo-classical and bootstrap effects; energy confinement studies; and MHD properties, including possible explanations for the apparent absence of the major disruption at low A.

A Thomson scattering diagnostic designed to measure both edge and core physics has been implemented on MAST. The system uses eight Nd:YAG lasers, each with a repetition rate of 30 Hz. The relative and absolute timing of the lasers may be set arbitrarily to produce fast bursts of measurements to suit the time evolution of the physics being studied. The scattered light is collected at F/6 by a 100 kg six element lens system with an aperture stop of 290 mm. The collected light is then transferred to 130 polychromators by 130 independent fiber bundles. The data acquisition and processing are based on a distributed computer system of dual core processors embedded in 26 chassis. Each chassis is standalone and performs data acquisition and processing for five polychromators. This system allows data to be available quickly after the MAST shot and has potential for real-time operations.

We describe results from the gun-injected spheromak device SPHEX, which show that the power required to sustain the plasma is initially deposited in a column, about 8 cm in radius, along the geometric axis of the device, and is transmitted from the column to the remainder of the plasma by a radially propagating oscillation at about 20 kHz. These results are relevant to the process of relaxation in spheromak systems

A new infrared Thomson scattering system has been designed for the MAST tokamak. The system will measure at 120 spatial points with approximately 10 mm resolution across the plasma. Eight 30 Hz 1.6 J Nd:YAG lasers will be combined to produce a sampling rate of 240 Hz. The lasers will follow separate parallel beam paths to the MAST vessel. Scattered light will be collected at approximately f/6 over scattering angles ranging from 80 degrees to 120 degrees. The laser energy and lens size, relative to an existing 1.2 J f/12 system, greatly increases the number of scattered photons collected per unit length of laser beam. This is the third generation of this polychromator to be built and a number of modifications have been made to facilitate mass production and to improve performance. Detected scattered signals will be digitized at a rate of 1 GS/s by 8 bit analog to digital converters (ADCs.) Data may be read out from the ADCs between laser pulses to allow for real-time analysis.

To examine the programming effects of maternal renal dysfunction (created by subtotal nephrectomy in ewes prior to mating; STNx), renal and cardiovascular function were studied in 6-month-old male and female offspring of STNx and control pregnancies. After studies were conducted on a low salt diet (LSD) some female offspring underwent salt loading (0.17 m NaCl in the drinking water for 5–7 days; HSD). On LSD both male and female offspring of STNx had similar mean arterial pressures (MAP), heart rates, cardiac outputs and renal function to those measured in offspring of control ewes. In female STNx offspring on a HSD, plasma sodium levels increased and haematocrits fell, indicating volume expansion (P < 0.05). Plasma renin levels were not suppressed despite the increases in plasma sodium concentrations, but aldosterone levels were reduced. In control animals plasma renin levels fell (P < 0.05) but there was no change in plasma aldosterone concentrations. There was a positive relationship between GFR and MAP which was present only in female STNx offspring. In conclusion, in STNx offspring there was an impaired ability to regulate glomerular filtration independent of arterial pressure, renin release was insensitive to a high salt intake and control of aldosterone secretion was abnormal. This study provides evidence of abnormal programming of the renin–angiotensin system and glomerular function in offspring of pregnancies in which there is impaired maternal renal function.

The chromanol derivative 293B was previously shown to inhibit a cAMP regulated K+ conductance in rat colon crypts. Subsequent studies on cloned K+ channels from the rat demonstrated that 293B blocks specifically IsK channels expressed in Xenopus oocytes, but does not affect the delayed and inward rectifier Kv1.1 and Kir2.1, respectively. In the present study, the specificity of 293B for the cardiac K+ conductances IKs and IKr, and for the cloned guinea pig IsK channel and the human HERG channel, which underly IKs and IKr, respectively, was analyzed. 293B inhibited both the slowly activating K+ conductance IKs in cardiac myocytes and guinea pig IsK channels expressed in Xenopus oocytes with a similar IC50 (2-6 micromol/l). In contrast, high concentrations of 293B had only a negligible effect on the more rapid activating IKr. Similarly, 293B exerted no effect on HERG channels expressed in Xenopus oocytes. In summary, 293B appears to be a rather specific inhibitor of IKs and the underlying IsK channels.

A new coherence imaging Doppler spectroscopy diagnostic has been deployed on the UK's Mega Amp Spherical Tokamak for scrape-off-layer and divertor impurity flow measurements. The system has successfully obtained 2D images of C III, C II, and He II line-of-sight flows, in both the lower divertor and main scrape-off-layer. Flow imaging has been obtained at frame rates up to 1 kHz, with flow resolution of around 1 km/s and spatial resolution better than 1 cm, over a 40° field of view. C III data have been tomographically inverted to obtain poloidal profiles of the parallel impurity flow in the divertor under various conditions. In this paper we present the details of the instrument design, operation, calibration, and data analysis as well as a selection of flow imaging results which demonstrate the diagnostic's capabilities.

Triggered vertical shifts of the MAST spherical tokamak plasma have been found to stabilize 2/1 neoclassical tearing modes (NTMs) for a number of MAST shots, without impacting on core confinement. This stabilization is a result of favourable modifications of the density, temperature and pressure profiles at the location of an NTM by means of a brief transition from high (H) to low (L) confinement mode. Using this method, the high confinement phase can typically be recovered, and the NTM removed, within 20 ms of onset.

Alkali-treated extracts of Rhodopseudomonas palustris growing photosynthetically on benzoate were examined by gas chromatography/mass spectrometry for partially reduced benzoate derivatives. Two cyclic dienes, cyclohexa-2,5-diene-1-carboxylate and cyclohexa-1,4-diene-1-carboxylate, were detected. Either compound supported cell growth as effectively as benzoate. These results suggest that these cyclohexadienecarboxylates, probably as their coenzyme A esters, are the initial reduction products formed during anaerobic benzoate metabolism by R. palustris.

Absolutely calibrated vacuum ultraviolet (VUV) spectroscopy has been used to determine the energy fluxes of VUV photons at an electrically floating substrate in a low-pressure 13.56-MHz radiofrequency plasma reactor used for polymer surface treatments. These fluxes have been compared with the positive ion flux that was reported in an earlier study. At the typical operating parameters of 10-mTorr pressure and 10-W power, the total VUV energy flux is 2.2 mW cm-2, compared with a value of 3.3 mW cm-2 from the ions. With increasing power (from 0.5 to 12 W), both the ion and VUV energy fluxes increase monotonically. However, as the pressure increases, (1−100 mTorr), the ion energy flux declines, while the VUV component increases. At discharge powers of 10 W, and pressures greater than 25 mTorr, the greater part of the energy flux to the surface is from the VUV photons. These measurements are used to determine which of the plasma components, VUV or ions, will be most effective in the treatment of polystyrene surfaces in a pure Ar plasma. Because of the low VUV absorption coefficient of polystyrene most of the photon flux penetrates through the outermost 2 nm of the polymer surface without attenuation. Consequently, we calculate that under typical plasma operating parameters, the ions play the major role in the modification of polystyrene surfaces because of their shorter stopping distances.

New diagnostic, modelling and plant capability on the Mega Ampère Spherical Tokamak (MAST) have delivered important results in key areas for ITER/DEMO and the upcoming MAST Upgrade, a step towards future ST devices on the path to fusion currently under procurement. Micro-stability analysis of the pedestal highlights the potential roles of micro-tearing modes and kinetic ballooning modes for the pedestal formation. Mitigation of edge localized modes (ELM) using resonant magnetic perturbation has been demonstrated for toroidal mode numbers n = 3, 4, 6 with an ELM frequency increase by up to a factor of 9, compatible with pellet fuelling. The peak heat flux of mitigated and natural ELMs follows the same linear trend with ELM energy loss and the first ELM-resolved Ti measurements in the divertor region are shown. Measurements of flow shear and turbulence dynamics during L-H transitions show filaments erupting from the plasma edge whilst the full flow shear is still present. Off-axis neutral beam injection helps to strongly reduce the redistribution of fast-ions due to fishbone modes when compared to on-axis injection. Low-k ion-scale turbulence has been measured in L-mode and compared to global gyro-kinetic simulations. A statistical analysis of principal turbulence time scales shows them to be of comparable magnitude and reasonably correlated with turbulence decorrelation time. Te inside the island of a neoclassical tearing mode allow the analysis of the island evolution without assuming specific models for the heat flux. Other results include the discrepancy of the current profile evolution during the current ramp-up with solutions of the poloidal field diffusion equation, studies of the anomalous Doppler resonance compressional Alfvén eigenmodes, disruption mitigation studies and modelling of the new divertor design for MAST Upgrade. The novel 3D electron Bernstein synthetic imaging shows promising first data sensitive to the edge current profile and flows. © 2013 IAEA, Vienna.

The aim was to determine the effect and mechanisms by which myocyte stretch interacts with the prolongation of action potential duration (APD) by the class III antiarrhythmic agent E-4031.Action potentials and whole-cell currents were measured in isolated guinea pig ventricular myocytes with a patch clamp procedure during perfusion of normotonic, normotonic with addition of E-4031, and hypotonic plus E-4031 solutions.Cell swelling leading to membrane stretch of myocytes in the whole-cell recording configuration occurred with hypotonic solution perfusion. APD, prolonged by E-4031, was reduced to less than control value with hypotonic-induced stretch. Evaluation of whole-cell currents after hypotonic-induced stretch revealed no significant changes in the L-type Ca2+ current, inward rectifier K+ current or the rapid component of the delayed rectifier K+ current. The slow component of the delayed rectifier K+ current (IKs) was upregulated and a stretch-induced CI- current was activated in hypotonic solutions. The hypotonic-induced modulation of these currents was not effected by protein kinase A or C inhibition.Hypotonic-induced stretch shortens APD and counteracts the effects of E-4031. This APD shortening is secondary to upregulation of IKs and activation of a stretch-induced Cl- current.

Disruptions are of significant concern to future devices, due to the large amount of energy released during the rapid quenching of the plasma. Disruption mitigation has been performed on MAST, to study the effect on heat loads and disruption time scales in a spherical tokamak. Massive gas injection is performed using a disruption mitigation valve capable of injecting between 0.6 and 1.5 × 1022 particles, corresponding to 10–150 times the plasma inventory. Noble gases are used for mitigation studies, specifically helium, argon, neon and a mixture of 10% argon and helium. The effect of mitigation is studied using a comparison between mitigated discharges and reference unmitigated disruptions.Mitigation has been seen to be effective at reducing peak divertor power loads, with reductions of up to 60% in the divertor power load being observed. The decrease in divertor power load is reflected by an increase in radiated power, mainly as a result of line radiation from the injected impurities. The largest reduction in the divertor power loads are seen for the impurities with Z > 4. The energy load to the divertor has been reduced by between 30–40% of the total stored plasma energy in ohmic, L mode and H mode discharges. Mitigation also affects the current quench times, accelerating the quench of the plasma with increasing injection quantity, with saturation observed at the highest injected quantities.

A real-time system has been developed to trigger both the MAST Thomson scattering (TS) system and the plasma control system on the phase and amplitude of neoclassical tearing modes (NTMs), extending the capabilities of the original system. This triggering system determines the phase and amplitude of a given NTM using magnetic coils at different toroidal locations. Real-time processing of the raw magnetic data occurs on a low cost field programmable gate array (FPGA) based unit which permits triggering of the TS lasers on specific amplitudes and phases of NTM evolution. The MAST plasma control system can receive a separate trigger from the FPGA unit that initiates a vertical shift of the MAST magnetic axis. Such shifts have fully removed m∕n = 2∕1 NTMs instabilities on a number of MAST discharges.

Massive gas injection (MGI) is one means of ameliorating disruptions in future devices such as ITER, where the stored energy in the plasma is an order of magnitude larger than in present-day devices. The penetration of the injected impurities during MGI in MAST is diagnosed using a combination of high-speed (20 kHz) visible imaging and high spatial (1 cm) and temporal (0.1 ms) resolution Thomson scattering (TS) measurements of the plasma temperature and density. It is seen that the rational surfaces, in particular q = 2, are the critical surfaces for disruption mitigation. The TS data shows the build-up of density on rational surfaces in the edge cooling period of the mitigation, leading to the collapse of the plasma in a thermal quench. The TS data are confirmed by the visible imaging, which shows filamentary structures present at the start of the thermal quench. The filamentary structures have a topology which matches that of a q = 2 field line in MAST, suggesting that they are located on the q = 2 surface. Linearized magnetohydrodynamic stability analysis using the TS profiles suggests that the large density build-up on the rational surfaces drives modes within the plasma which lead to the thermal quench. The presence of such modes is seen experimentally in the form of magnetic fluctuations on Mirnov coils and the growth of an n = 1 toroidal mode in the period prior to the thermal quench. These results support the observations of other machines that the 2/1 mode is the likely trigger for the thermal quench in a mitigated disruption and suggests that the mitigation process in spherical tokamaks is similar to that in conventional aspect ratio devices.

Saccharomyces cerevisiae was used as a model system to characterize the covalent attachment of palmitoleic acid to proteins. Chemically synthesized cis-[9,10-3H]hexadecenoic acid (palmitoleic acid) was used to demonstrate the attachment of this lipid species to at least six proteins (m = 122, 58, 45, 41, 31, and 17 kDa). The majority of the labeled proteins are distinct from those labeled with [3H]palmitic acid (16:0). Based on the lability of the bond in the presence of methanolic KOH or hydroxylamine (pH 8), we propose that [3H] palmitoleic acid is attached to proteins via a thioester linkage. The identity of the palmitoleic acid was established by C-18 reverse phase high performance thin layer chromatography and argentation thin layer chromatography analysis after the fatty acid was liberated from the proteins by either transesterification or saponification. Incorporation of [3H]palmitoleic acid into proteins was only slightly inhibited (relative to [3H] myristic acid) by the presence of cycloheximide, indicating that the attachment of [3H]palmitoleic acid occurs post-translationally. This report is the first description of multiprotein acylation by a long chain unsaturated fatty acid.

On MAST studies of the profile evolution of the electron temperature (Te), electron density (ne), radial electric field (Er) as well as novel measurements of the ion temperature (Ti) and toroidal current density (j) in the pedestal region allow further insight into the processes forming and defining the pedestal such as the H-mode access conditions and MHD stability. This includes studies of fast evolution of Te, ne and Er with ¿t = 0.2 ms time resolution and the evolution of pe and j through an edge-localized mode (ELM) cycle. Measurements of the H-mode power threshold, PL-H revealed that about 40% more power is required to access H-mode in 4He than in D and that a change in the Z-position of the X-point can change PL-H significantly in single and double null configurations. The profile measurements in the L-mode phase prior to H-mode suggest that neither the gradient nor the value of the mean Te or Er at the plasma edge play a major role in triggering the L–H transition. After the transitions, first the fluctuations are suppressed, then the Er shear layer and the ne pedestal develops followed by the Te pedestal. In the banana regime at low collisionality (¿) ¿Ti ˜ 0 leading to Ti > Te in the pedestal region with Ti ~ 0.3 keV close to the separatrix. A clear correlation of ¿Ti with ¿ is observed. The measured j (using the motional Stark effect) Te and ne are in broad agreement with the common peeling–ballooning stability picture for ELMs and neoclassical calculations of the bootstrap current. The j and ¿pe evolution ¿t ˜ 2 ms as well as profiles in discharges with counter current neutral beam injection raise questions with respect to this edge stability picture.

To see if arginine vasopressin (AVP) influenced fetal sodium balance, we infused AVP i.v. (45 mU h-1 kg-1) into two groups of chronically catheterized fetal sheep. One group had urinary osmolalities of 147 +/- 23 mosm kg-1 (SEM, n = 6) and the other group had higher urinary osmolalities (339 +/- 3 mosm kg-1, n = 4, P0.001). The group with high urinary osmolalities had higher systolic pressures (P0.05), higher glomerular filtration rates (GFR; P0.05), and higher urinary electrolyte excretion rates (P0.05), but lower membrane blood flows (P0.05) and lower fractional reabsorption of sodium by the proximal tubule (P0.01). In the group with low urinary osmolalities, AVP caused a rise in arterial blood pressure (P0.001), a fall in heart rate (P0.001), a fall in membrane blood flow (P0.02), but no change in placental or renal blood flow. Renal sodium excretion increased (P0.001) because GFR rose (P0.001) and proximal fractional sodium reabsorption fell (P0.001). Distal fractional sodium reabsorption increased (P0.001), but not enough to compensate for the fall in proximal fractional reabsorption. Lung liquid flow decreased (P = 0.006), as did lung liquid sodium excretion (P = 0.002). There were no changes in fetal plasma sodium, blood volume or haematocrit. The effects of AVP infusion were similar in the group with high urinary osmolalities. This study shows that high levels of AVP, such as occur in fetal "stress", have widespread effects on fetal cardiovascular, renal and lung functions. The characteristic profile of the fetuses with high urinary osmolalities prior to AVP infusion could be entirely explained by high endogenous AVP levels and AVP could possibly be a sole mediator of these widespread effects of fetal "stress". Furthermore, although during infusion of AVP sodium excretion increased, blood volumes did not change. Therefore, the fetuses must have accessed additional sodium from either their extracellular fluids, the amniotic and/or allantoic cavities or across the placenta.

The newly upgraded MAST Thomson scattering (TS) system provides excellent spatial resolution (~1?cm) at over 130 radial locations across a full plasma diameter, and utilizes eight individual Nd:?:YAG laser systems which can be fired sequentially, providing electron temperature and density profiles approximately every 4?ms throughout a plasma discharge. By operating the system in burst mode, whereby the laser separation can be adjusted to within a few microseconds of each other, it is possible to obtain detailed profiles of transient and periodic phenomena such as sawteeth crashes, massive gas injection for disruption mitigation and the temperature perturbations associated with neoclassical tearing mode (NTM) islands. Following Fitzpatrick et al (1995 Phys. Plasmas 2 825), we consider a simplified model in which finite parallel diffusive heat transport can provide a threshold for NTM island growth and demonstrate that the TS derived electron temperature profiles around an island can be used to obtain both the island width and the critical island width below which temperature gradients are maintained across the island, potentially removing the bootstrap current drive for the NTM. Initial results from high beta, neutral beam injection heated discharges on MAST show that the measured island width inferred from the TS data is in good agreement with magnetic estimates of the island width (considering both a cylindrical approximation and using a full field line tracing estimate). The temporal behaviour of the island width obtained from the magnetic diagnostics indicates that for the scenarios considered to date, finite parallel diffusion is likely to play an important role in NTM threshold physics in MAST.

The spheromak device SPHEX has been modified by adding a current-carrying rod along the geometric axis, providing a preexisting toroidal field. We show that plasma can be succesfully injected into such a field from a helicity source; the field assists plasma ejection from the gun and improves the coupling between gun and plasma, so that T e , T i , and the toroidal current all increase with rod current.

Here we describe a new experiment to test the shielding concept of a dipole-like magnetic field and plasma, surrounding a spacecraft forming a 'mini magnetosphere'. Initial laboratory experiments have been conducted to determine the effectiveness of a magnetized plasma barrier to be able to expel an impacting, low beta, supersonic flowing energetic plasma representing the solar wind. Optical and Langmuir probe data of the plasma density, the plasma flow velocity and the intensity of the dipole field clearly show the creation of a narrow transport barrier region and diamagnetic cavity virtually devoid of energetic plasma particles. This demonstrates the potential viability of being able to create a small 'hole' in a solar wind plasma, of the order of the ion Larmor orbit width, in which an inhabited spacecraft could reside in relative safety. The experimental results have been quantitatively compared with a 3D particle-in-cell 'hybrid' code simulation that uses kinetic ions and fluid electrons, showing good qualitative agreement and excellent quantitative agreement. Together the results demonstrate the pivotal role of particle kinetics in determining generic plasma transport barriers.

The University of Manchester Linear System (ULS) [M. G. Rusbridge et al., Plasma Phys. Controlled Fusion 42, 588 (2000)] is an experiment in which a steady-state plasma stream is confined along a longitudinal magnetic field. The plasma passes through a tapered orifice into a separate gas target chamber (GTC) where it interacts with neutral gas at pressures of up to 15mTorr. The upstream plasma beam is 6–14mm in diameter with electron temperatures between 2and15eV, and densities in the range of 1017–1019m−3. The primary aim of this study is to investigate physical processes relevant to gas target divertors in tokamaks. Upstream parameters, in terms of electron temperature and density, can be varied in the ULS enabling the investigation of electron-ion recombination (EIR) or molecular-activated recombination (MAR) processes. Conditions for assaying chemotaxis in Bacillus subtilis are described. The chemotaxis medium we used afforded excellent motility for hours. In it, chemotaxis measured by capillary assays was insensitive to pH between 5.5 and 9, and to temperature between 28 degrees C and 42 degrees C. Chemotaxis was observed toward all 20 common amino acids, with thresholds varying from 3nM for alanine to 0.1 mM for glutamate, in the capillary assay, and from 0.1 muM for alanine to 0.32 mM for glutamate in the microscope assay.

Specificities of chemoreceptors for the 20 common amino acids, toward which Bacillus subtilis shows chemotaxis, were assessed by competition ("jamming") experiments using a modification of the traditional capillary assay, called the "sensitivity capillary assay." Many amino acids were sensed by at least two chemoreceptors. All the highest affinity chemoreceptors for the amino acids were distinct, except glutamate and aspartate, which may share one chemoreceptor, and tyrosine, for which the data could not be collected due to low solubility. The data suggest the hypothesis that each amino acid-chemoreceptor complex binds to a different signaler (from each amino acid-chemoreceptor complex binds to a different signaler (from which signals travel to the flagella to modify behavior appropriately), and that many of the signalers can also bind other attractant-chemoreceptor complexes as antagonists (no signals to flagella).

The binding of the substrates, ATP and ribose-5-P, and the most effective inhibitor, ADP, to phosphoribosylpyrophosphate synthetase from Salmonella typhimurium was characterized using equilibrium dialysis of these compounds labeled with 32P. In the absence of ribose-5-P, ATP, ADP, and the ATP analogue alpha,beta-methylene ATP each bind cooperatively with half-saturation at 50 to 90 microM and Hill coefficients of 1.5 to 2. We propose that all three compounds bind at the same set of sites, which are presumably the active sites. When ribose-5-P was added, methylene ATP and ADP binding at these sites became tighter (Kd approximately 3 to 6 microM at 10 mM ribose-5-P) and lost its cooperativity. In the presence of ribose-5-P, ADP, but not methylene ATP, bound to a second site with half-saturation at approximately 150 microM and a Hill coefficient greater than 3. This result confirms the existence of an allosteric ADP site, which was previously postulated from kinetic studies (Switzer, R. L., and Sogin, D. C. (1973) J. Biol. Chem. 248, 1063-1073). Binding of ribose-5-P could not be detected in the absence of nucleotides, but it was readily measured in their presence. The apparent Kd of ribose-5-P varied from greater than 1 mM to approximately 5 microM as the concentration of either ADP or methylene ATP was increased from 0 to 2 mM. Inhibition of the enzyme by action of ADP at both active and allosteric sites could be observed kinetically.
