Search

The fragmentation reactions of neutron-rich unstable nuclei 81Ga and 82Ge at 250 MeV/nucleon have been studied in order to search the optimal mean for the production of very neutron-rich nuclei in the vicinity of 78Ni. The newly measured cross sections were compared with various calculations, showing a good agreement with EPAX3.01. The present work provides experimental insights into the two-step scheme—fragmentation following fission—as a method to produce very neutron-rich nuclei through a combination of ISOL and fragmentation of post-accelerated beams of unstable nuclei. Our results enable an evaluation of the potential of the two-step scheme in the production of very neutron-rich nuclei for the 78Ni region. The two-step scheme using fragmentation of a post-accelerated 81Ga beam could be an option to produce neutron-rich nuclei around 78Ni when the 81Ga beam intensity reaches 1.0×108 pps, compared to the one-step scheme with fission of 238U.

Background and Objective: In endometrial cytology, differentiating endometrial glandular stromal breakdown (EGBD) from endometrial endometrioid carcinoma (G1-EEC) is often difficult. In this study, we provided a new focus on chondroitin sulfate (CS), a major substrate component of the endometrial stroma, and assessed the diagnostic utility of Alcian Blue (AB) staining in the differential diagnosis in liquid-based cytological (LBC) samples. Materials and Methods: LBC specimens from 19 patients with a proliferative endometrium, 36 with EGBD, and 30 with G1-EEC who underwent endometrial cytology were stained with AB (pH 1.0), and their reactivity was observed. In addition, immunocytochemical staining of CS and CD31 was performed for five cases each to evaluate their interrelationship with blood vessels. Results: Regarding the 30 G1-EEC cases, at least one of the three representative staining patterns was observed by AB staining: dot-like, microtubular, and finely branched linear patterns. Moreover, the inner portion of the tubular material observed by AB staining expressed CD31. Conversely, in the 36 EGBD cases, only five metaplastic clusters with irregular protrusions and condensed stromal clusters (CSCs) showed a dot-like positive pattern, and background CSCs did not show reactivity to AB staining in any of the cases. Furthermore, the vascular structure expressing CD31 in cell clusters was also unclear. Conclusions: We demonstrated that AB staining shows different staining patterns in G1-EEC and EGBD, reflecting their different tissue structures. Our data provide new insights into endometrial cell diagnosis changes and demonstrate that AB staining is a potential new diagnostic aid tool for the differentiation of G1-EEC from EGBD.

Abstract The QCD phase diagram in the space of temperature and imaginary baryon chemical potential has been an interesting subject in numerical lattice QCD simulations because of the absence of the sign problem and its deep structure related to confinement/deconfinement. We study constraints on the phase diagram by using an ’t Hooft anomaly. The relevant anomaly is an anomaly in the space of imaginary chemical potential. We compute it in the UV, and discuss how it is matched by the pion effective field theory at low temperatures. Then we study implications of the anomaly to the phase diagram. There must be a line of phase transition studied in the past by Roberge and Weiss such that the expectation value of the Polyakov loop is not smooth when we cross the line. Moreover, if the greatest common divisor of the color and flavor numbers is greater than one, the phase transition across the Roberge-Weiss line must be either a first order phase transition, or a second order phase transition described by a nontrivial interacting three-dimensional CFT.

IntroductionThis study aimed to assess the localization of chondroitin sulfate (CS), a primary extracellular matrix component, in the stromal region of endometrial carcinoma (EC).MethodsImmunostaining was performed on 26 endometrial endometrioid carcinoma (EEC) samples of different grades and 10 endometrial serous carcinoma (ESC) samples to evaluate CS localization. This was further confirmed by Alcian Blue (AB) staining as well.ResultsIn the G1-EEC samples, CS showed reactivity with fibrovascular stroma, supporting closely packed glandular crowding and papillary structures. As the grade increased, the original interstitial structure was re-established, and the localization of CS in the perigulandular region decreased. In the ESC samples, the thick fibrous strands supporting the papillary architecture showed reactivity with CS; however, the delicate stromal region branching into the narrow region showed poor reactivity. The AB staining results showed similar characteristics to the immunostaining ones.ConclusionsThe characteristic localization of CS in various EC types was elucidated. The present study provides new information on endometrial stromal assessment.

Superconducting (SC) tokamak JT-60SA plays an essential role in fusion research and development by supporting and complementing the ITER project, providing directions to the DEMO design activity and fostering next generation scientists and engineers. Since the short circuit incident at the terminal joints of equilibrium field coil #1 during the integrated commissioning (IC) in March 2021, both EU and JA implementing agencies (IAs) have examined how to ensure safe operation of JT-60SA by mitigating the risk of possible discharge occurrence inside the cryostat. Based on the experience of the global Paschen tests, the IAs have established a strategy of risk mitigation measures, which is a combination of (i) reinforcement of insulation, (ii) avoiding unnecessary voltage application to the coil systems and (iii) immediate de-energization of the coils when deteriorated vacuum conditions are detected. Thanks to the considerable efforts of the Integrated Project Team members, the IC restarted in May 2023. After confirmation of the SC state of the coil systems (TF, EF and CS), the coil energization test and the plasma operation phase 1 (OP-1) started. The first plasma was successfully achieved on 23 October 2023 with a limited value of voltage and current applied to the coils. The plasma configuration control was also confirmed with low plasma current and low auxiliary heating power conditions. Based on the IO–F4E–QST collaboration, activities of JT-60SA have been shared with the IO and provided an important lesson for ITER assembly and commissioning, and will provide an outstanding contribution to fusion research at large. After OP-1, maintenance & enhancement phase 1 (M/E-1) starts from January 2024, in which in-vessel components are installed, and heating and diagnostic systems are extensively upgraded to allow a high power heating experiment planned in OP-2. In order to make the best use of JT-60SA, a newly organized JT-60SA experiment team will refine the research plan for the future high heating power operation phase.

ABSTRACT: To facilitate sustainable dairy farming, it is essential to assess and support the mental health of dairy farm workers, which is affected more than that of workers in other industries, as indicated by the relatively few studies to date. In addition, the limited investigations on mental health in dairy workers minimize the opportunities to suggest practical approaches of improvement of their mental health. Therefore, further data acquisition and analysis is required. In the present study, we undertook quantitative surveys on 17 management factors and administered a mental health questionnaire to 81 dairy farm managers (80 male, 1 female) in Hokkaido, northern Japan. The management factors were categorized into 3 groups: production input, production output, and facility indicator; mental health was evaluated based on the Center for Epidemiologic Studies Depression Scale (CES-D). Principal component analysis assigned the factors into 2 groups: intensiveness factors of dairy production systems (PC1: livestock care cost, fat- and protein-corrected milk, stocking density, medical consultation fee per unit time per animal unit, nonfamily wages, fertilizer and pesticide expenses, and net agricultural income ratio) and basic dairy management factors (PC2: net agricultural income ratio, quantity of concentrate feed, and milk quality variable). The depression symptoms of dairy farm managers were not significantly associated with PC1 and milking methods; however, they were significantly negatively associated with PC2, which integrated 3 management factors, including factors related to finances, feeding, and milk quality. According to the findings of the present study, the efforts needed for stable economic farm management, adequate feed supply, and milk quality maintenance may increase the depression levels of dairy farm managers and negatively affect their mental health. These findings could be the basis for future studies on the relationship between the mental health of farm managers and sustainable dairy farm management and production.

Background Adoptive cellular therapies with chimeric antigen receptor T cells have revolutionized the treatment of some malignancies but have shown limited efficacy in solid tumors such as glioblastoma and face a scarcity of safe therapeutic targets. As an alternative, T cell receptor (TCR)–engineered cellular therapy against tumor-specific neoantigens has generated significant excitement, but there exist no preclinical systems to rigorously model this approach in glioblastoma.Methods We employed single-cell PCR to isolate a TCR specific for the Imp3D81N neoantigen (mImp3) previously identified within the murine glioblastoma model GL261. This TCR was used to generate the Mutant Imp3-Specific TCR TransgenIC (MISTIC) mouse in which all CD8 T cells are specific for mImp3. The therapeutic efficacy of neoantigen-specific T cells was assessed through a model of cellular therapy consisting of the transfer of activated MISTIC T cells and interleukin 2 into lymphodepleted tumor-bearing mice. We employed flow cytometry, single-cell RNA sequencing, and whole-exome and RNA sequencing to examine the factors underlying treatment response.Results We isolated and characterized the 3×1.1C TCR that displayed a high affinity for mImp3 but no wild-type cross-reactivity. To provide a source of mImp3-specific T cells, we generated the MISTIC mouse. In a model of adoptive cellular therapy, the infusion of activated MISTIC T cells resulted in rapid intratumoral infiltration and profound antitumor effects with long-term cures in a majority of GL261-bearing mice. The subset of mice that did not respond to the adoptive cell therapy showed evidence of retained neoantigen expression but intratumoral MISTIC T cell dysfunction. The efficacy of MISTIC T cell therapy was lost in mice bearing a tumor with heterogeneous mImp3 expression, showcasing the barriers to targeted therapy in polyclonal human tumors.Conclusions We generated and characterized the first TCR transgenic against an endogenous neoantigen within a preclinical glioma model and demonstrated the therapeutic potential of adoptively transferred neoantigen-specific T cells. The MISTIC mouse provides a powerful novel platform for basic and translational studies of antitumor T-cell responses in glioblastoma.

A search for the neutrinoless quadruple beta decay of 136Xe was conducted with the liquid-xenon detector XMASS-I using 327 kg ×800.0 days of the exposure. The pulse shape discrimination based on the scintillation decay time constant which distinguishes γ-rays including the signal and β-rays was used to enhance the search sensitivity. No significant signal excess was observed from the energy spectrum fitting with precise background evaluation, and we set a lower limit of the half life of 3.7 × 1024 years at 90% confidence level. This is the first experimental constraint of the neutrinoless quadruple beta decay of 136Xe.

The CALorimetric Electron Telescope, CALET, has been measuring high-energy cosmic rays on the International Space Station since October 13, 2015. The scientific objectives addressed by the mission are to search for possible nearby sources of high-energy electrons and potential signatures of dark matter, and to investigate the details of galactic cosmic-ray acceleration and propagation. The calorimetric instrument, which is 30 radiation lengths or 1.3 proton interaction lengths thick with fine imaging capability, is optimized to measure cosmic-ray electrons by achieving large proton rejection and excellent energy resolution well into the TeV region. In addition, very wide dynamic range of energy measurement and individual charge identification capability enable us to measure proton and nuclei spectra from a few tens GeV to a PeV scale. Nearly 20 million cosmic-ray shower events over 10 GeV per month are triggered and the continuous observation has been kept without any major interruption since the start of operation. Using the data obtained over 6.5 years of operation, we will present a brief summary of the CALET observation including electron spectrum, and proton and nuclei spectra as well as the performance study on orbit with MC simulations.

Abstract Sex allocation is one of the most studied traits in evolutionary biology because its theoretical predictions match the empirical data. Here, using the Ryukyu dry-wood termite Neotermes sugioi, we investigated several factors that could bias the sex allocation in three populations (Okinawa, Ishigaki/Iriomote, and Yonaguni). Our survey showed that there were more queen-only colonies than king-only colonies in these populations, suggesting a longer lifespan of the queens than that of the kings. In this condition, sex-asymmetric reproductive value (SRV) theory predicts female bias, because even after the short-lived kings die, the long-lived queens can continue reproduction with their sons. However, sex allocation in this species seemed to be biased toward males. Furthermore, we examined the possibility of intrasexual competition among siblings (ICS). If ICS is the cause of the bias, the allocation is expected to change depending on the total investment in sexual offspring. However, the biomass of both male and female alates increased linearly with the increase in the total biomass of the alates in these populations. Thus, neither the SRV nor the ICS theory could explain the male-biased sex ratio of N. sugioi. On the basis of these results, we discuss the remaining possibilities in this species.

In this study, time-resolved ex situ Raman and Fourier transform infrared (FT-IR) spectroscopy were performed to monitor structural changes in a polyamide (PA) 66 gear during operation and reveal the failure mechanisms of the material. A gear test was performed using a lab-built gear tester, which is a power-absorption type. Raman and FT-IR analyses were performed on the side gear surfaces around the root of the gear (at the side of the active flank), where a bending stress is continuously loaded during operation. The results showed that the crystallinity changed, amide related bonds were broken, trans-gauche (TG) transformation occurred, and a change in the inner stress condition of the PA66 gear occurred over time. It was concluded that these changes interact with each other during operation and are related to the failure of the PA66 gear, such as the initiation of the plastic gear degradation.

Abstract N-cadherin is a homophilic cell adhesion molecule that stabilizes excitatory synapses, by connecting pre- and post-synaptic termini. Upon NMDA receptor (NMDAR) activation by glutamate, membrane-proximal domains of N-cadherin are cleaved serially by a-disintegrin-and-metalloprotease 10 (ADAM10) and then presenilin 1(PS1, catalytic subunit of the γ-secretase complex). To assess the physiological significance of the initial N-cadherin cleavage, we engineer the mouse genome to create a knock-in allele with tandem missense mutations in the mouse N-cadherin/Cadherin-2 gene (Cdh2 R714G, I715D, or GD) that confers resistance on proteolysis by ADAM10 (GD mice). GD mice showed a better performance in the radial maze test, with significantly less revisiting errors after intervals of 30 and 300 s than WT, and a tendency for enhanced freezing in fear conditioning. Interestingly, GD mice reveal higher complexity in the tufts of thorny excrescence in the CA3 region of the hippocampus. Fine morphometry with serial section transmission electron microscopy (ssTEM) and three-dimensional (3D) reconstruction reveals significantly higher synaptic density, significantly smaller PSD area, and normal dendritic spine volume in GD mice. This knock-in mouse has provided in vivo evidence that ADAM10-mediated cleavage is a critical step in N-cadherin shedding and degradation and involved in the structure and function of glutamatergic synapses, which affect the memory function.

E. coli is the main pathogen of UTI. It is important to be aware the local epidemiological data for an appropriate initial treatment. Resistance to antimicrobial agents has increased, especially to first-choice antibiotics in the treatment of cystitis. There are few studies on the sensivity profile of community uropathogen in our region. Objective: To characterize antimicrobials the sensitivity profile to E. coli isolated from urocultures of women treated at Basic Health Units and Emergency Care Units of Londrina- Paraná- Brazil during a period of 12 months (June 1, 2016 to June 1, 2017). Methodology: A cross-sectional study was carried out from June 2016 to June 2017. All urine samples collected in the Basic Health Units and Emergency Departments in the city of Londrina (Paraná State, Brazil) were sent to a Central Laboratory where the identification and antimicrobial susceptibility testing were performed. Clinical Laboratory Standards Institute (CLSI) breakpoints were used for the interpretation of susceptibility testing results. Results: 56,555 urine cultures were performed in the period, of which 8,832 were positive, of which 5,377 were women. Of these samples, 4.7% were enterobacteria producing extended-spectrum beta-lactamases (ESBL) and 15.5% resistant to quinolones. TMP- SMX was resistant in more than 30% of the samples in all age groups. Among quinolone-resistant isolates, resistance to cephalothin, ampicillin and sulfamethoxazole-trimethoprim was greater than 60%. Nitrofurantoin was the only antimicrobial that showed 90% of sensitivity. Conclusion: The antimicrobials sensitivity profile was similar to that reported in the literature, with TMP- SMX resistance greater than 30% in the studied samples. Nitrofurantoin maintains high sensitivity rates greater than 90%. Resistance to quinolones increases proportionally with age, as well ESBL.

This paper is a continuation of the authors' previous paper (Part 1) on the feasibility of ensemble flood forecasting for a small dam catchment (Kasahori dam; approx. 70 km2) in Niigata, Japan, using a distributed rainfall–runoff model and rainfall ensemble forecasts. The ensemble forecasts were given by an advanced four-dimensional, variational-ensemble assimilation system using the Japan Meteorological Agency nonhydrostatic model (4D-EnVar-NHM). A noteworthy feature of this system was the use of a very large number of ensemble members (1600), which yielded a significant improvement in the rainfall forecast compared to Part 1. The ensemble flood forecasting using the 1600 rainfalls succeeded in indicating the necessity of emergency flood operation with the occurrence probability and enough lead time (e.g., 12 h) with regard to an extreme event. A new method for dynamical selection of the best ensemble member based on the Bayesian reasoning with different evaluation periods is proposed. As the result, it is recognized that the selection based on Nash–Sutcliffe efficiency (NSE) does not provide an exact discharge forecast with several hours lead time, but it can provide some trend in the near future.

A search for dark matter (DM) with mass in the sub-GeV region (0.32–1 GeV) was conducted by looking for an annual modulation signal in XMASS, a single-phase liquid xenon detector. Inelastic nuclear scattering accompanied by bremsstrahlung emission was used to search down to an electron equivalent energy of 1 keV. The data used had a live time of 2.8 years (3.5 years in calendar time), resulting in a total exposure of 2.38 ton-years. No significant modulation signal was observed and 90% confidence level upper limits of 1.6×10−33 cm2 at 0.5 GeV was set for the DM-nucleon cross section. This is the first experimental result of a search for DM mediated by the bremsstrahlung effect. In addition, a search for DM with mass in the multi-GeV region (4–20 GeV) was conducted with a lower energy threshold than previous analysis of XMASS. Elastic nuclear scattering was used to search down to a nuclear recoil equivalent energy of 2.3 keV, and upper limits of 2.9 ×10−42 cm2 at 8 GeV was obtained. Abstract Previously, through a TILLING (Targeting Induced Local Lesions in Genomes) approach applied on barley chloroplast mutator (cpm) seedlings a high frequency of polymorphisms in the rpl23 gene was detected. All the polymorphisms corresponded to five differences already known to exist in nature between the rpl23 gene located in the inverted repeats (IRs) and the rpl23 pseudogene located in the large single copy region (LSC). In this investigation, polymorphisms in the rpl23 gene were verified and besides, a similar situation was found for the pseudogene in cpm seedlings. On the other hand, no polymorphisms were found in any of those loci in 40 wild type barley seedlings. Those facts and the independent occurrence of polymorphisms in the gene and pseudogene in individual seedlings suggest that the detected polymorphisms initially arose from gene conversion between gene and pseudogene. Moreover, an additional recombination process involving small recombinant segments seems to occur between the two gene copies as a consequence of their location in the IRs. These and previous results support the hypothesis that the CPM protein is a component of the plastome mismatch repair (MMR) system, whose failure of the anti-recombination activity results in increased illegitimate recombination between the rpl23 gene and pseudogene.

A search for dark matter using an underground single-phase liquid xenon detector was conducted at the Kamioka Observatory in Japan, particularly for Weakly Interacting Massive Particles (WIMPs). We have used 705.9 live days of data in a fiducial volume containing 97kg of liquid xenon at the center of the detector. The event rate in the fiducial volume after the data reduction was (4.2±0.2)×10−3day−1kg−1keVee−1 at 5keVee, with a signal efficiency of 20%. All the remaining events are consistent with our background evaluation, mostly of the “mis-reconstructed events” originated from 210Pb in the copper plates lining the detector's inner surface. The obtained upper limit on a spin-independent WIMP-nucleon cross section was 2.2×10−44cm2 for a WIMP mass of 60GeV/c2 at the 90% confidence level, which was the most stringent limit among results from single-phase liquid xenon detectors. We have searched for exotic neutrino-electron interactions that could be produced by a neutrino millicharge, by a neutrino magnetic moment, or by dark photons using solar neutrinos in the XMASS-I liquid xenon detector. We observed no significant signals in 711 days of data. We obtain an upper limit for neutrino millicharge of 5.4×10−12e at 90% confidence level assuming all three species of neutrino have common millicharge. We also set flavor-dependent limits assuming the respective neutrino flavor is the only one carrying a millicharge, 7.3×10−12e for νe, 1.1×10−11e for νμ, and 1.1×10−11e for ντ. These limits are the most stringent yet obtained from direct measurements. We also obtain an upper limit for the neutrino magnetic moment of 1.8×10−10 Bohr magnetons. In addition, we obtain upper limits for the coupling constant of dark photons in the U(1)B−L model of 1.3×10−6 if the dark photon mass is 1×10−3 MeV/c2, and 8.8×10−5 if it is 10 MeV/c2.

Hidden photons and axion-like particles are candidates for cold dark matter if they were produced non-thermally in the early universe. We conducted a search for both of these bosons using 800 live-days of data from the XMASS detector with 327 kg of liquid xenon in the fiducial volume. No significant signal was observed, and thus we set constraints on the α′/α parameter related to kinetic mixing of hidden photons and the coupling constant gAe of axion-like particles in the mass range from 40 to 120 keV/c2, resulting in α′/α

Excited states of 77Cu have been investigated by use of single proton knock-out reaction at the Radioactive Isotope Beam Factory in RIKEN in order to reveal the main components of the proton single-particle states. Three excited states were observed at 271, 902 and 2068 keV in 77Cu. The lowest-energy excited state follows the trend predicted for the crossing of the 3/21− and 5/21− states. Comparing the excitation energies of the 3/2−, 5/2− and 7/2− levels from 69Cu to 77Cu one can see that the Z=28 shell gap between the p3/2 and f7/2 states is rather stable, while the f5/2−f7/2 spin–orbit splitting decreases by ∼1.5 MeV in agreement with shell model calculations using the tensor force.

Neoantigens represent promising targets for personalized cancer vaccine strategies. However, the feasibility of this approach in lower mutational burden tumors like glioblastoma (GBM) remains unknown. We have previously reported the use of an immunogenomics pipeline to identify candidate neoantigens in preclinical models of GBM. Here, we report the application of the same immunogenomics pipeline to identify candidate neoantigens and guide screening for neoantigen-specific T cell responses in a patient with GBM treated with a personalized synthetic long peptide vaccine following autologous tumor lysate DC vaccination. Following vaccination, reactivity to three HLA class I- and five HLA class II-restricted candidate neoantigens were detected by IFN-γ ELISPOT in peripheral blood. A similar pattern of reactivity was observed among isolated post-treatment tumor-infiltrating lymphocytes. Genomic analysis of pre- and post-treatment GBM reflected clonal remodeling. These data demonstrate the feasibility and translational potential of a therapeutic neoantigen-based vaccine approach in patients with primary CNS tumors.

Glabridin is a prenylated isoflavonoid isolated from the roots of Glycyrrhiza glabra. We investigated the metabolic effects of glabridin, using healthy volunteers and patients with type 2 diabetes. Seven healthy volunteers and 10 patients with type 2 diabetes were recruited, and assigned to receive an oral dose of glabridin (300 mg) once daily for 12 weeks. Total cholesterol, low-density lipoprotein cholesterol, glycated hemoglobin, immunoreactive glucagon, and body fat composition significantly decreased in the healthy individuals, after 12 weeks of glabridin administration, while there were subtle effects in the patients with type 2 diabetes. Glabridin significantly improved flow-mediated vasodilatation, and pulse wavelength in the healthy volunteers. The results of our pilot study indicate that glabridin exerted several favorable metabolic effects and improved vascular functions, especially in healthy volunteers. Therefore, glabridin might be better to use in healthy subjects for preventing metabolic disorder rather than the patients with type 2 diabetes.

Double electron capture is a rare nuclear decay process in which two orbital electrons are captured simultaneously in the same nucleus. Measurement of its two-neutrino mode would provide a new reference for the calculation of nuclear matrix elements whereas observation of its neutrinoless mode would demonstrate lepton number violation. A search for two-neutrino double electron capture on 124Xe is performed using 165.9 days of data collected with the XMASS-I liquid xenon detector. No significant excess above background was observed and we set a lower limit on the half-life as 4.7×1021 years at 90% confidence level. The obtained limit has ruled out parts of some theoretical expectations. We obtain a lower limit on the 126Xe two-neutrino double electron capture half-life of 4.3×1021 years at 90% confidence level as well.
