The presence of luteinizing hormone receptors in human pineal glands from five females and three males, ranging in age from 61-89 yr, was examined by in situ hybridization and immunocytochemistry. The results demonstrated the presence of these receptors at the mRNA and protein levels in all the pineal glands examined. Pineal gland luteinizing hormone receptors could potentially be involved in the regulation of melatonin synthesis.

Specific gravity values for the pineal gland in any species are lacking. These data are necessary for calculating and interrelating pineal weight and volume. This report deals with the specific gravity of human pineal gland. The specific gravity of fresh, unfixed human pineal has been determined to be 1.197 +/- 0.036 SEM at 20 degrees C (N = 11) using water as the measuring medium. By contrast, human brain has been reported in the literature to have a specific gravity of 1.036. Specific gravities of human pineals fixed in Bouin's fluid, Bouin's fluid followed by 80% ethanol, 80% ethanol, and embalming fluid were found to be 1.113 +/- 0.025 (N = 5), 0.941 +/- 0.009 (N = 5), 0.990 +/- 0.022 (N = 3), and 1.149 +/- 0.021 (N = 7), respectively. These are corrected values accounting for the specific gravities of the respective measuring fluids. It is shown that the specific gravity of human pineal as derived in this study is directly applicable to calculations of pineal volumes and weights not only in humans, but in other species as well.

The vomeronasal organ (VNO) is a chemosensory structure of the vertebrate nasal septum that has been recently shown to exist in nearly all adult humans. Although its link to reproductive behaviors has been shown in some primates, its functionality in humans is still debated. Some authors have suggested that the human VNO has the capacity to detect pheromones, while others described it as little more than a glandular pit. However, no studies have utilized histochemical techniques that would reveal whether the human VNO functions as a generalized gland duct or a specialized chemosensory organ. Nasal septal tissue from 13 humans (2-86 years old) were compared to that of two adult lemurs (Microcebus murinus) and eight adult voles (four Microtus pennsylvanicus and four Microtus ochrogaster). Sections at selected intervals of the VNO were stained with periodic acid-Schiff (PAS), alcian blue (AB), AB-PAS, and PAS-hematoxylin procedures. Results revealed typical well-developed VNOs with tubuloacinar glands in Microtus and Microcebus. VNO glands were AB-negative and PAS-positive in voles and mouse lemurs. Homo differed from Microtus and Microcebus in having more branched, AB and PAS-positive glands that emptied into the VNO lumen. Furthermore, the human VNO epithelium had unicellular mucous glands (AB and PAS-positive) and cilia, similar to respiratory epithelia. These results demonstrate unique characteristics of the human VNO which at once differs from glandular ducts (e.g., cilia) and also from the VNOs of mammals possessing demonstrably functional VNO.

The accessory olfactory bulb (AOB) is the first neural integrative center for the olfactory-like vomeronasal sensory system. In this article, we first briefly present an overview of vomeronasal system organization and review the history of the discovery of mammalian AOB. Next, we briefly review the evolution of the vomeronasal system in vertebrates, in particular the reptiles. Following these introductory aspects, the structure of the rodent AOB, as typical of the well-developed mammalian AOB, is presented, detailing laminar organization and cell types as well as aspects of the homology with the main olfactory bulb. Then, the evolutionary origin and diversity of the AOB in mammalian orders and species is discussed, describing structural, phylogenetic, and species-specific variation in the AOB location, shape, and size and morphologic differentiation and development. The AOB is believed to be absent in fishes but present in terrestrial tetrapods including amphibians; among the reptiles AOB is absent in crocodiles, present in turtles, snakes, and some lizards where it may be as large or larger than the main bulb. The AOB is absent in bird and in the aquatic mammals (whales, porpoises, manatees). Among other mammals, AOB is present in the monotremes and marsupials, edentates, and in the majority of the placental mammals like carnivores, herbivores, as well as rodents and lagomorphs. Most bat species do not have an AOB and among those where one is found, it shows marked variation in size and morphologic development. Among insectivores and primates, AOB shows marked variation in occurrence, size, and morphologic development. It is small in shrews and moles, large in hedgehogs and prosimians; AOB continues to persist in New World monkeys but is not found in the adults of the higher primates such as the Old World monkeys, apes, and humans. In many species where AOB is absent in the adult, it often develops in the embryo and fetus but regresses in later stages of development. Finally, new areas in vomeronasal system research such as the diversity of receptor molecules and the regional variation in receptor neuron type as well as in the output neurons of the AOB and their projection pathways are briefly discussed. In view of the pronounced diversity of size, morphologic differentiation, and phylogenetic development, the need to explore new functions for the vomeronasal system in areas other than sexual and reproductive behaviors is emphasized.

The mere appearance of a tubular, epithelially-covered, bilateral structure, no matter how minuscule, on the anteroventral nasal septum of tetrapods, is generally called the vomeronasal organ (of Jacobson). However, considering the functionality of this chemosensory structure, the presence of a non-cilated (microvillar) neuroepithelium (and not just any odd type of epithelium) encased in a variously shaped vomeronasal cartilage, along with vomeronasal nerve bundles and above all an accessory olfactory bulb connected to the limbic vomeronasal amygdala, are the absolute essential neurostructural characteristics and anatomic requirement for a functional VNO and the accessory olfactory system in any tetrapod. The distribution of the vomeronasal organ is reported here in two mammalian orders: Chiroptera and Primates. An impressive data pool on the vomeronasal organ of bats is now available, pointing to the fact that at this time bats may be the only group in which this organ system is extremely variable, ranging from total absence (even in the embryo) to spectacular development with numerous intervening stages in different chiropteran species. Of the eighteen bat families, only one family of New World leaf-nosed bats, family Phyllostomidae, exhibits functional vomeronasal organs. The vespertilionid bat Miniopterus, and the mormoopid bat Pteronotus, present exceptions to this rule. Among Primates, very few species have been rigorously studied. As a result, developmental variability of the vomeronasal organ is almost unknown; either the vomeronasal organ is well developed (such as in New World monkeys) or absent (as in Old World monkeys and great apes) in the adult. The concept whether adult humans or embryonic and fetal forms are endowed with this so-called sixth sense, is a controversial one and is under intense study in our laboratory and by others. The general phylogenetic implications based on our cladistic analysis of bats are that the vomeronasal organ complex has evolved several times. Among the prosimians and platyrrhine primates, the organ is well developed, although to a varying degree. Among catarrhine primates, its loss has occurred only once, as it is generally absent in the adult forms.

Even though the chorioallantoic placenta of Suncus has been previously investigated with light and electron microscopy, controversies related to its structure still remain. To resolve these, Suncus murinus placentae from several (early and late limb bud, advanced pregnancy, and full term) were examined by electron microscopy. The interhemal membrane of Suncus comprises a maternal endothelium with basal lamina, syncytial trophoblast with its basal lamina, fetal mesenchyme, and fetal capillary endothelium. Layering of these components remain unchanged throughout gestation. The most striking feature of the interhemal membrane is the hypertrophied mesenchymal and maternal epithelial layers. The syncytiotrophoblast develops into a sieve-like transtrophoblastic spongy layer bearing numerous processes of the hypertrophied maternal endothelium. Mesenchymal cells intervene amidst the fetal endothelium displaying a high protein synthesizing activity. The interhemal membrane of Suncus is confirmed to be endotheliochorial.

Morphological variations of the thyroid gland are common and generally occur superior to the gland, reflecting its developmental origin. In this report we describe an accessory lobe located inferior to both lateral lobes and the isthmus. The accessory lobe was supplied by a branch of the right inferior thyroid artery and its vein drained via the plexus thyroideus impar. A discussion of thyroid anomalies is presented and a system for classifying variations of thyroid anatomy is proposed.

This paper presents the first description of a functional vomeronasal organ in the colugo or 'flying lemur' Cynocephalus, the sole living representative of the order Dermoptera (Mammalia). The vomeronasal organ complex comprises patent nasopalatine and vomeronasal ducts, a 10-mm-long epithelial tube consisting of an 8- to 10-cell thick, 65-microns-tall neurosensory epithelium, the vomeronasal nerve, and the accessory olfactory bulb. The vomeronasal glands are sparse. Among mammals, the vomeronasal organ of the colugo is one of the relatively longest, at nearly 48% of nasal cavity length. It is exceeded among archontans (colugos, bats, primates, and tree shrews) only by that of the strepsirhine primate Microcebus murinus.

This investigation is based upon the pineal organs of 92 specimens of 36 species of the family Pteropodidae (Mammalia, Chiroptera). The size of the megachiropteran pineal correlates well with body size (r = 0.864), confirming the former conclusions that generally larger bodied bats have larger pineals. The range of the pineal size index in 36 megachiropteran species is from 33 to 4393. In most species the pineal organs are small, deeply recessed under the cerebral hemispheres and of Type A (except in Dobsonia and Pteropus, where they are of Type alpha beta C and AB, respectively). Morphological and volumetric data gathered from serially sectioned brains include body and brain weights, pineal type, dimensions, volume and size index for each species. There are distinct dorsal and ventral subdivisions of the pineal in some species and a clear separation of pineal parenchyma into cortical and medullary regions in others. In several species where overlying ependyma is lacking pinealocyte clusters communicate freely with the CSF. Groups of intrapineal neurons are noted in the connective tissue beside blood vessels. The habenular commissure shows much interspecific variation in its course through the pineal. Detailed examination of pineal-brain relationships clearly suggests that, due to the generally deep location of the pineal in relation to cerebral hemispheres, pinealectomies in the species studied may be extremely difficult, it not entirely impossible. The absolutely and relatively largest pineal organ among bats, and relatively perhaps among all vertebrates, has been discovered in the New Guinean naked-backed bat, Dobsonia praedatrix, with pineal size index of 4393, and a volume of 16.3447 mm3, which is 0.56% of the brain. This alpha beta C-type, mushroom-shaped, solid and compact pineal organ measures 5.33 x 4.51 mm. The cortical and medullary parenchyma are divided into lobes by large calibre blood vessels along which numerous intrapineal neurons are observed. A smaller but similarly shaped pineal is noted in the other three Dobsonia. Data on the largest known pineals in ratitae birds, seals and walruses have been compared with that of D. praedatrix and the human pineal. This study supports the hypothesis that pineal development may reflect dependence on habitat and possibly other related factors.

The vomeronasal organ (YNO) receptor-free epithelium (RFE) of Diaemus youngii is compa red with that of Desmodus rotundus, Diphylla ecaudata, Artibeus jamaicensis, and Carollia perspicillata. The entire RFE of Diaemus bears tall cilia, whereas mixed populations of ciliated and non-ciliated cells occur in Desmodus and Diphylla, dropping to a few ciliated ones in the latter species. The RFE in Artibeus and Carollia is non-ciliated. It is suggested that the function of RFE cilia may have to be performed by the neuroepithelial microvilli in the non-ciliated VNO of Artibeus and Carollia. Since the Diaemus RFE is heavily ciliated, it is inferred that the overall chemoreceptive function of the VNO might be greatly enhanced by the cilia clearing the organ lumen of its fluids at a quicker pace, thus making the Diaemus VNO a highly efficient chemosensory organ compared to these other species.

The ultrastructure of the pineal organ was studied in the tropical megachiropteran Rousettus leschenaulti. The pineal lies deep beneath the hemispheres adjacent to the third ventricle and is traversed by the habenular commissure anteriorly. Its parenchyma consists of a uniform population of light and occasional dark pinealocytes which appear to differ only in the degree of cytoplasmic staining. Pinealocytes are characterized by well developed Golgi bodies associated with numerous small vesicles, many mitochondria and polyribosomes, and frequent subsurface cisternae. Lipid droplets and elements of smooth endoplasmic reticulum are scant. Cisternae of granular endoplasmic reticulum are occasionally dilated. A distinct feature is the abundance of clear vesicles in the pinealocyte pericapillary terminals, which also frequently contain granular vesicles and a very large vacuole. The pineal is further characterized by the presence of a small number of glial cells and myelinated nerve fibers. A broad perivascular space investing numerous capillaries contains glial-cell and pinealocyte processes, collagen fibrils and abundant unmyelinated nerve fibers. Tortuous extensions of the perivascular space enter the pineal parenchyma where they come in close proximity to branched intercellular channels or canaliculi characterized by specialized junctions and microvilli. Differences between the pineal of the non-hibernating megachiropteran Rousettus and that of the hibernating microchiropteran bats, and structural similarities to the pineal of tropical rodents are discussed.

This investigation describes the ultrastructure of the atrioventricular node of Eptesicus fuscus. Two conducting cells types (nodal and transitional) are indentified which differ in location, myofibrillar content, and types of intercellular junctions. Centrally located nodal cells display variable staining intensity and contain disorganized myofibrils which rarely form sarcomeres. Desmosomes and nexus-like junctions connect the nodal cells. Transitional cells, situated peripherally, exhibit distinct sarcomeres and are attached to the adjacent cells through desmosomes and underdeveloped intercalated discs. Longitudinal arrangement of the mitochondrial cristae is frequently seen in both cells types. In the connective tissue stroma, numerous capillaries (with micropinocytotic vesicles), axons, and possibly axonal terminals, some filled with vesicles, are observed. A large ganglionated nerve trunk is present on the nodal periphery. True nexuses and neuromuscular junctions are not observed. It is suggested that nodal cell types previously reported in different vertebrates under various names are merely simple variations of the two basic types of conducting cell--nodal and transitional. No interspecific differences are observed between these cells of the big brown bat compared with those in other vertebrates.

The morphology of the vomeronasal organ complex was histologically described in eight out of fourteen chiropteran species investigated. Of the six families examined, all except the family Pteropodidae (suborder Megachiroptera) were found to have at least one member possessing the organ. The organ is best developed in phyllostomatids. It is absent in vespertilionids (including a Myotis embryo) except in Miniopterus. An accessory olfactory bulb is reported for the first time in the latter. The organ is described for the first time in Rhinopoma, Megaderma, and Hipposideros. The organ in Rhinolophus is also described. Homologous anterior nasal cartilages and patent nasopalatine ducts are present in all species. The organ occupies the anterior ventral nasal septum region. In Megaderma and Hipposideros it is level with the nasal cavity floor. Areas of epithelium similar to olfactory epithelium have been observed in some organs. Epithelia, vascular sinuses, vomeronasal nerves, paravomeronasal ganglia, accessory olfactory bulbs, and vomeronasal glands have been investigated. In bats with regressed or rudimentary organs (Megaderma, Rhinopoma, Rhinolophus, Hipposideros) accessory olfactory bulbs could not be identified. Thus, presence of the organ does not necessarily indicate presence of the accessory olfactory bulb. Septal pockets located superior to the organ complex and lined with pseudostratified columnar epithelium are described in Hipposideros and may play a part in nasophonation. A unique role is proposed for the organ in the feeding behaviour of Desmodus. The desirability of extending the useful terms 'diosmatic' and 'monosmatic' to all vertebrates in reference to their respective possession or lack of the vomeronasal organ is suggested.

Volumes of the main olfactory bulb (MOB) in 76 species of Insectivora, Scandentia, Primates and Macroscelidea, and some of the laminar components in 34 species were measured. No statistically significant differences were found (1) between the two sides in the 162 individuals and (2) between males and females in the 19 species in which both sexes were examined. In interspecific comparisons the relative size (expressed by size indices) shows a definite tendency to decrease from Insectivora through prosimians and simians to man. The average indices were 100-64-10-3.6, respectively. Scandentia and Macroscelidea have the highest average MOB indices (135 and 166). The relative MOB size is discussed in relation to feeding and social behaviour. It was shown that MOB development is largely linked to dietary adaptations but that its importance in feeding behaviour is paralleled by a similar importance in social behaviour. Within each dietary type, MOB development is associated with the different characteristics of the occupied niche. Among Insectivora, ground-dwelling species occupy the upper, and semiaquatic species the lower positions in the size scale for the MOB. Among Primates, in closely related species, the nocturnal species have in general better developed MOBs than the diurnal species. The composition of the MOB is relatively stable, i.e. the laminar components (layers 1 + 2, 3, and 4-6) show no clear change in their percentage size from well developed to strongly reduced MOBs. Only in the diurnal simians are layers 4-6 relatively small. This corresponds with the general observation (obtained from light-microscopy) that the granular layer (layer 6) is reduced and decomposed in higher Primates, and especially in man.

The nasal epithelia of two species of bats were quantified with respect to relative surface areas and olfactory epithelial volumes. In the macrosmatic Aribeus jamaicensis 55.9% of the nasal cavity surface was covered by olfactory epithelium (232.4 mm2), in contrast to only 28.9% in the microsmatic Myotis lucifugus (36.4 mm2). The roles of the various nasal epithelia have been discussed as they may relate to olfaction, respiration and echolocation. In the olfactory bulbs of both species, the estimated concentration of mitral cells approximated at 2,500/mm2 compared to an olfactory nerve concentration of 5/mm2. In Artibeus, calculated total volume of olfactory epithelium was on the order of 16 times greater than in Myotis, and Artibeus' olfactory bulb diameter was twice as great. These findings, together with previously published surface, volume and physiological relationships, suggest a developmental design mechanism for an olfactory bulb in which the number of olfactory receptors increases some 450-fold above an initially established ratio of 2:1 between receptors and mitral cells. Key governing factors could be requisite mechanical rigidity of the cribriform plate of the ethmoid bone and response thresholds of higher brain centers.
