In plants, cytoplasmic mRNA decay is critical for posttranscriptionally controlling gene expression and for maintaining cellular RNA homeostasis. Arabidopsis DCP1-ASSOCIATED NYN ENDORIBONUCLEASE 1 (DNE1) is a cytoplasmic mRNA decay factor that interacts with proteins involved in mRNA decapping and nonsense-mediated mRNA decay (NMD). There is limited information on the functional role of DNE1 in RNA turnover, and the identities of its endogenous targets are unknown. In this study, we utilized RNA degradome approaches to globally investigate DNE1 substrates. Monophosphorylated 5′ ends, produced by DNE1, should accumulate in mutants lacking the cytoplasmic exoribonuclease XRN4, but be absent from DNE1 and XRN4 double mutants. In seedlings, we identified over 200 such transcripts, most of which reflect cleavage within coding regions. While most DNE1 targets were NMD-insensitive, some were upstream ORF (uORF)-containing and NMD-sensitive transcripts, indicating that this endoribonuclease is required for turnover of a diverse set of mRNAs. Transgenic plants expressing DNE1 cDNA with an active-site mutation in the endoribonuclease domain abolished the in planta cleavage of transcripts, demonstrating that DNE1 endoribonuclease activity is required for cleavage. Our work provides key insights into the identity of DNE1 substrates and enhances our understanding of DNE1-mediated mRNA decay.

Aims: To evaluate the effect of different colour mulches on Water use efficiency under protected cultivation a field experiment was carried out during April–June 2021 in Cucumber (Cucumis sativus). Study Design: Split plot design statistical design. Place and Duration of Study: Department of Soil and Water Conservation Engineering, Agricultural Engineering College and Research Institute, Tamil Nadu Agricultural University, Coimbatore, between April to June 2021. Methodology: Different coloured Plastic mulches (black-M1, silver-M2, yellow-M3, red-M4, white-M5) and no cover M6 (control) with different irrigation levels (I1-100% PE, I2-75% PE and I3-50% PE) was adopted with split plot design. Results: The observed data revealed that using mulches decreased irrigation requirements of cucumber crop with comparing to no cover (without mulch). The data shows that Black Mulch M1 with I3-50%PE recorded the highest values of WUE (31.30 kg/m3) followed by White Mulch M5 with I3-50% PE (30.03kg/m3). Conclusion: Increasing irrigation level up to I2-75% improved yield with different coloured plastic mulch treatments, while the yield decreased with increasing irrigation level to I1-100%PE. Though using 50% PE with black plastic mulch increased WUE compared to using 75% PE or 100% PE without plastic mulch (control).

Land use land cover (LULC) change detection is essential for sustainable development, planning and management. This study was an attempt to evaluate the LULC change in the lower bhavani basin from 2014 to 2019, using Landsat 8 data integrating Google Earth Engine (GEE) as a web-based platform and Geographic Information System. The CART and Random Forest classifiers in GEE were used for performing supervised classification. The classified map accuracy was assessed using high resolution imagery and evaluated using a confusion matrix implemented in GEE. Five major LULC classes, viz., agriculture, built up, current fallow, forest and waterbody, were identified, and the dominant land use in the study area was agriculture and current fallow, followed by dominant land use of forest. During the study period (2014–2019) the change inbuilt-up area 7.37% in 2019 and 5.45% in 2014, was noted due to urban sprawl. GEE showed significant versatility and proved to be an effective platform for LULC detection.

During a rainy season, dry and wet spells tend to persist and can be represented using a Markov process. Knowing the succession of dry and wet periods is necessary to plant crops and carry out agricultural operations. This study aimed to analyze the probability of dry/wet spell rainfall using the Markov chain model in the Dharmapuri district of Tamil Nadu, India. In estimating the chance of dry and wet spells, the model used rainfall of below 20 mm in a week as a dry calendar week and rainfall of 20 mm or more as a wet calendar week from the years 1980 to 2019. From the 1st through the 32nd Standard Meteorological Week (SMW), a continuous dry week probability was 75-100%. The likelihood of a dry week trailed by another dry week was more up to the 32nd standard week, while the chance of a dry week followed by a wet week was more up to the 31st standard week, ranging from 75 to 100%. During the 37th to 45th weeks, the conditional likelihood of a rainy week followed by another rainy week ranged from 43.8 to 68%. According to a review of consecutive dry and wet spells, two consecutive dry weeks had a 55 to 97.5% chance of occurring within the first 32 weeks of the year. In the first 32nd week of the year, the chance of three successive dry weeks ranged from 32.6 to 92.6%. Consecutive dry weeks suggest the need for additional irrigation and proper moisture management practices. In contrast, consecutive wet calendar weeks indicate an abundance of extra water available for rainwater collection and the necessity for proper soil erosion control measures.

Objective : The aim of the present study is to formulate a polyherbal formulation whose formula is given in AFI and evaluate its antidiabetic potential in animals. Method: The polyherbal formulation was prepared using the methanol extracts and aqueous extracts of the given parts of herbs in AFI. The quality of the finished product was evaluated as per the World Health Organization’s guidelines for the quality control of herbal materials. The quality testing parameters of the polyherbal formulation were within the limits. Results : The acute toxicity studies of the polyherbal formulation did not show any toxic symptoms in doses up to 2000 mg/kg over 14 days. The extracts of both the formulations were evaluated for In-vitro and In-vivo anti-diabetic study. The oral anti-diabetic activity of the polyherbal formulation (200 and 400 mg/kg, p.o) was screened against intraperitoneal (i.p.) STZ (35 mg/kg) 15 min after i.p. administration of nicotinamide (200 mg/kg) continuously for 2 days, to induced diabetes mellitus in Wistar rats. The investigational drug was administered for 28 consecutive days, and the effect of the polyherbal formulation on blood glucose levels was studied at regular intervals.

Sleep is commonly defined as a state in which physical activities and sensory perception are greatly reduced, and it is frequently associated with our body's recuperative period. However, research has shown that sleep is also required for other vital processes such as memory consolidation and normal physiological functioning. Extensive research has shown that the areas that control our sleeping behaviour are the hypothalamus, brain stem, midbrain, and amygdala. These areas coordinate events during the non-REM and REM phases of sleep-wake cycles. GABA and adenosine, two chemical neurotransmitters, are also involved and play an important role in our sleep cycle. Insomnia has caused a slew of psychological and physiological issues such as fatigue, decreased mental concentration, irritable nature and a higher risk of heart attacks and stroke. It has reduced the patient's quality of life of the patient and have a social impact on them. Primary insomnia is defined as the inability/absence of proper sleep and impaired daily life functioning. Secondary insomnia, on the other hand, is believed to result from pre-existing medical conditions, substance abuse, or as a side effect of certain drug therapy. Insomnia is diagnosed through physical examination and the use of electrical devices to monitor sleep behavior.

The proportion of agricultural water consumption is continuously decreasing due to increased competition for water resources by urban, industrial, and agricultural users. Drip irrigation is more efficient in terms of water and energy utilization. These considerations are critical in view of the ongoing struggle for water resources among various consumers due to water scarcity. Some of the most critical criteria in the effective design and maintenance of drip irrigation systems are the shape and size of the volume of wet soil beneath the emitter. Hence several statistical models were constructed in this research to estimate the dimensions of wetting patterns, which are critical for designing an optimal drip irrigation system. The Nash-Sutcliffe efficiency (NSE), coefficient of correlation (CC), and root mean square error (RMSE) criteria were used to assess the models' performance. The results showed that the Polynomial model was the most accurate for horizontal advance, with 0.94, 0.93, and 1.33 (cm) values for CC, NSE, and RMSE, respectively. For vertical advance, the logarithmic model showed 0.96, 0.96, and 0.72 (cm) values for CC, NSE, and RMSE. Thus, in the absence of a wetting pattern and under identical conditions, these models can be utilized to generate synthetic horizontal and vertical advances data.

Estimating evapotranspiration's spatiotemporal variance is critical for regional water resource management and allocation, including irrigation scheduling, drought monitoring, and forecasting. The Surface Energy Balance Algorithm for Land (SEBAL) method can be used to estimate spatio-temporal variations in evapotranspiration (ET) using remote sensing-based variables like Land Surface Temperature (LST), Normalized Difference Vegetation Index (NDVI), surface albedo, transmittance, and surface emissivity. The main aim of the study was to evaluate the actual evapotranspiration for the lower Bhavani basin, Tamil Nadu based on remote sensing methods using Landsat 8 data for the years 2018 to 2020. The actual evapotranspiration was estimated using SEBAL model and its spatial variation was compared over different land covers. The estimated values of daily actual evapotranspiration in the lower Bhavani basin ranged from 0 to 4.72 mm day-1. Thus it is evident that SEBAL model can be used to predict ET with limited ground base hydrological data. The spatially estimated ET values will help in managing the crop water requirement at each stage of crop and irrigation scheduling, which will ensure the efficient use of available water resources.

Watershed is a natural geohydrological unit that allows surface runoff to a particular point or outlet, Morphometric analysis plays a vital role in the proper planning and development of the watershed. Morphometric analysis of the Nanjannad watershed is carried out using integrated approach of remote sensing and GIS techniques. The present study attempted to determine the morphometric characteristics of Nanjannad watershed of Ooty, Tamil Nadu. Geographical information system (GIS) is used to evaluate linear, areal and relief aspects of morphometric parameters. The drainage area of the watershed is 434 ha, the watershed is a Fourth-order drainage basin, and the lower order streams are mostly dominating in the watershed. Physiography, rainfall and lithology of the location control the stream orders. The development of stream segments in the watershed area is affected by rain, slope of the watershed varies from 0 to 87%. The mean Rb of the entire watershed is 4.38, indicating that geological structures do not influence the drainage pattern much. The whole watershed elongation ratio is 0.67 showing the majority of the area has Moderate relief and a steep slope, Relief ratio values implies that the discharge of the Nanjannad watershed is high and groundwater potential is meager, the moderate value of drainage density, stream frequency and drainage intensity, indicates that runoff is more likely to cause water logging. This work can be used as input to evaluate hydrology and water resources and will be helpful for integrated watershed planning.

Background: Pearlmillet is conventionally cultivated by farmers either broadcasting or uniform row spacing of 45 cm where it is difficult to accommodate an intercrop. Modified planting pattern provides additional space for intercrop component as well as harvest better solar energy in the system. Methods: The field experiment was conducted in Department of Agronomy, Tamil Nadu Agricultural University, Coimbatore during Kharif (July-October) of 2019. The experiment was laid out in split plot design and were replicated thrice. Different crop geometry viz. 45 x 15 cm, 60 x 15 cm, 30/60 x 15 cm and 30/90 x 15 cm were adopted in main plots, whereas intercropping [S1- Greengram, S2-Sesame and S3- No intercrop] were allotted in subplots. Result: Pearlmillet grown under paired row sowing 30/90 x 15 cm recorded higher growth attributes and yield parameters. Grain and stover yields was also recorded higher in the above-mentioned crop geometry. In intercropping system higher growth and yield parameters was observed in sole crop treatment and it was on par with greengram intercropping system. Pearlmillet grown under paired row sowing 30/90 x 15 cm (M4) + greengram (S1) recorded higher equivalent yield.

A rapid, simple and highly sensitive stability-indicating reverse-phase high-performance liquid chromatographic technique, coupled with a photodiode array detector, was developed and validated for the estimation of Deferasirox (DFS). The chromatographic separation was achieved using a C-18 (250 × 4.6 mm, 5 μm) stationary phase and a mobile phase composed of 0.1% orthophosphoric acid and acetonitrile at a flow rate of 1 mL/min. The detection was carried out at a wavelength of 245 nm with a constant injection volume of 10 μL throughout the analysis. With an R2 value of 0.9996, the calibration curve was determined to be linear over an appropriate concentration range of 50–500 ng/mL. According to the International Conference on Harmonization (ICH) Q1 (R2) guideline, DFS was evaluated under stress conditions that included hydrolytic (acid, alkali and neutral), oxidative and thermal degradation. The findings demonstrated that significant degradation was observed in acidic degradation conditions, whereas drug substance was found to be stable when exposed to neutral, basic, oxidative and thermal degradation. The developed method was validated as per ICH guidelines. The developed method was employed successfully to estimate the amount of DFS in bulk and pharmaceutical formulation.

The different groups of MPXVs contain unique tandem repeats with different copy numbers in the ITR regions, and these repeats may be likely to play a role in the genetic diversity of the virus. Clade IIb (B) MPXV contains 38 and 32 repeats similar to the Tandem repeats reported in the human and rodent genome, respectively.

Present study involves the synthesis, characterization, antimalarial and anticancer activities of some novel substituted amino analogues of 1,4-naphthoquinone. The chloro group present in the key starting materials like 2,3-dichloro-1,4-naphthoquinone (1) and 2-chloro-3-[trans-4-(4-chlorophenyl)- cyclohexyl]-1,4-naphthoquinone (2) was replaced by substituted amines. These analogues were isolated, purified and screened for antimalarial and anticancer activities. A few of the novel compounds were found to possess substantial biological activity and are reasonably potent.

Background: In veterinary practice, cutaneous injuries are a very common occurrence. Various methods have been employed to close large skin defects like skin graft or flap often associated with relatively more complex surgical procedures. Use of tension relieving suture techniques like spider plasty and bow tie technique allow the mobilization of adjacent local tissue to cover large cutaneous defects which was thus utilised in the study. Methods: Twelve dogs brought to the Madras Veterinary College Teaching Hospital, Chennai,TANUVAS with large diameter open wounds were selected for the study. Irrespective of the stage of closure the two tension relieving suture techniques, i.e. Spiderplasty and Bow-tie, were performed for closure of the large wounds. The preoperative and postoperative wounds assessment were performed through subjective evaluation, planimetry, bacteriological examination and histopathological evaluation. Result: The Spiderplasty technique was more time consuming, required more surgical expertise and had slightly lower cost efficiency ratio. It could however be better utilised to close wounds located in confined anatomical areas, as compared to the Bow-tie procedure which was limited by its requirement to resect 36% of adjacent normal tissue.

Background: The present study was undertaken to evaluate the efficiency of collagen urethral stents on wound healing post-urethrotomy in dogs and to assess the uptake of collagen urethral stents through urethrography, ultrasonography and hematobiochemical analysis. Methods: A total of 12 dogs presented to Madras Veterinary College teaching hospital with the history of urinary obstruction were chosen for the study and were randomly divided into 2 groups of 6 dogs in each. In Group I, dogs were treated with conventional urethrotomy procedure and in group II, animals were treated with urethrotomy with collagen urethral stent placement. Hematobiochemical, plain and contrast radiography, ultrasonograhy were performed in both the groups. Result: The study revealed that in comparison with Group I, Group II animals had a faster wound healing rates; reduction in urethral lumen diameter and urethral leakage post-operatively were not observed in both the groups. In the present study, ultrasound was used to identify and locate the calculi but also to check the collapse, migration and absorption of the collagen stent placed inside the urethra wherein no stent collapse or migration noticed. Partial collagen stent absorption was noticed on day 7 and a complete stent absorption was noticed on day 14. There was no significant difference in the pain scores between the groups indicating that the collagen stent does not elicit any extra pain as compared to normal urethrotomy procedure.

Field experiment was conducted to evaluate the performance of different automated drip irrigation on tomato crop under sandy clay loam soil in Tamil Nadu Agricultural University during kharif 2019 and kharif 2020. Five treatments comprising 4 different automated drip irrigation systems are time based drip irrigation, volume based drip irrigation, soil moisture sensor based irrigation, switching tensiometer based irrigation and one is conventional method of irrigation were tested. The results revealed that tensiometer based drip irrigation recorded higher fruit yield of 95.11 and 96.21 t ha-1 and water use efficiency of 21.10 and 25.42 t ha-mm-1 resulting in increment over conventional method of irrigation. However, the above treatment was followed by soil moisture sensor based drip irrigation in tomato. Tensiometer based drip irrigation helps to save the water up to 54.91 and 60.55 % compared to conventional method of irrigation during kharif 2019 and 2020.
