Abstract

The intraerythrocytic stages of plasmodia are capable of synthesizing purine nucleotides and apparently deoxycytidylate by salvage syntheses. Data obtained by studying the incorporation of radioactive precursor molecules into intact cells and kinetic experiments on purified enzyme preparations suggest biosynthetic routes which, generally, are similar to those of the host's cell metabolism. However, details on the regulation of both the uptake of nucleosides and bases into the intraerythrocytic stages of plasmodia and of the metabolic routes involved in this incorporation are still lacking.

Abstract

The catabolism of adenosine 5'-monophosphate in promastigotes of L. tropica suggests the presence of a parasite specific pathway. In continuing the investigation on enzymes of this pathway two purine nucleoside cleaving enzymes have been found, adenosine phosphorylase and inosine nucleosidase. Various purine nucleoside analogues inhibited the activity of both enzymes. A mode of action of the growth inhibition of L. tropica promastigotes by allopurinol has been suggested on the basis of Michaelis and inhibitor constants.

Abstract

Ornithine decarboxylase of P. falciparum, the initial and rate-limiting enzyme of the synthesis of polyamines, was inhibited by chloroquine when P. falciparum-infected red cells in culture were incubated in the presence of chloroquine for two hours. The degree of these inhibition produced by mefloquine was less. On the basis of these findings a sequence of reactions is discussed to define the mode of action of chloroquine. Additionally a preliminary investigation of the ornithine analogue, difluoromethylornithine, showed inhibitory effects on ornithine decarboxylase activity of P. falciparum.

Abstract

A purine nucleoside hydrolase from Trypanosoma gambiense was purified 160-fold. Preferred substrates of the reaction were adenosine, inosine and guanosine with a maximum of activity at pH 5.4. Competitive inhibitors of the adenosine hydrolysis were dimethylallyl adenosine, 6-methylmercaptopurine riboside, tubercidin, formycin B, 6-mercaptopurine riboside and deoxyadenosine. A metabolic scheme of adenosine nomophosphate salvage synthesis is discussed.

Abstract

The specific activities of enzymes participating in AMP-catabolizing reactions in promastigotes of Leishmania tropica were determined. The following sequence of reactions is suggested: AMP leads to adenosine leads to adenine leads to hypoxanthine. The initial enzyme of this sequence, AMP nucleotidase, is apparently membrane-bound. The significance of AMP catabolism with respect to adenylate energy charge and a signal transfering mechanism is discussed. Adenine deaminase has been partially purified and characterized. Several purine analogues, inter alia allopurinol, proved to be inhibitors of the adenine deaminase catalyzed reaction.

Abstract

Thin-sectioning and freeze-etching electron microscopy were applied to explore the structure and the temperature- and Ca2+-response of the different host and parasite membranes during intraerythrocytic development of Plasmodium knowlesi in Macaca mulatta. The plasma membrane of uninfected erythrocytes is temperature- and Ca2+-responsive: chilling to 4 degrees C and exposure to 5 mM Ca2+ induces a slight decrease in IMP-frequency and the emergence of small IMP-devoid patches on P-faces. On parasite infection, the erythrocyte membrane becomes modified as indicated by an enhanced temperature-response and the appearance of caveolae, ca. 70-90 nm in diameter. The frequency of these caveolae is increased in schizont-infected erythrocytes. Moreover, electron dense plaques, ca. 40 nm in width, appear just beneath the erythrocyte membrane in late trophozoites and schizonts, thus indicating a further modification of the host cell membrane during parasite development. The membrane of the parasitophorous vacuole, derived from the host plasma membrane, dramatically reduces the IMP-frequency especially on the P-face upon parasite infection. This leads to an apparent reversal of the IMP-distribution persisting throughout the whole infection cycle. The parasite plasma membrane forms local compaction domains with the nuclear envelope in ca. 30% of the ring-stages and trophozoites, which disappear in late trophozoites and schizonts. Moreover, the IMP-frequency on plasma membrane fracture faces almost doubles during parasite development. Chilling induces a decrease in the IMP-frequency on P-faces of the plasma membrane. Surprisingly, however, the parasite plasma membrane and the vacuolar membrane respond to externally applied Ca2+ with almost a doubling of the IMP-frequency. Abstract

A novel compaction domain between the nuclear envelope and the plasma membrane is observed in intraerythrocytic Plasmodium knowlesi and Plasmodium chabaudi. The plasma membrane and the two nuclear membranes stick very closely together thus forming a heptalamina domain. This has an outer diameter of 14.3 +/- 1.1 nm and can be as long as 2/um. Fracture faces in this region appear to be devoid of the typical membrane-intercalating-particles. This compaction domain occurs only in ring-stages and early to mid trophozoites, but never in schizonts.

Abstract

Membrane preparations of erythrocytes from normal and P. chabaudi-infected mice and membrane preparations of P. chabaudi-infected and uninfected erythrocytes from infected mice and separated by zonal centrifugation were characterized by the pattern of proteins and extracted glycoproteins obtained by SDS-polyacrylamide gel electrophoresis and by the specific activities of membrane associated enzymes. The protein pattern of the membrane preparation of infected erythrocytes showed similar differences from membrane preparations of normal erythrocytes as those described by Weidekamm et al. for P. berghei. The pattern of glycoproteins extracted by the chloroform-methanol method showed characteristic differences as compared to the controls. A new band (PASi) with a molecular weight of about 165,000 corresponds with the protein band IIa. In membrane preparations of normal erythrocytes and of nonparasitized erythrocytes separated from parasitized erythrocytes by zonal centrifugation was no difference in specific activities of ATPase, adenylate kinase and acetylcholinesterase. Adenylate kinase activity was markedly increased and acetyl-cholinesterase activity was slightly increased in membrane preparations of infected cells. Specific activities of ATPase of membrane preparations of normal and parasitized erythrocytes did not show significant differences. There was a decrease in enzyme activity of ATPase and an increase of acetylcholinesterase in Triton X 100 containing samples. Specific activities of an acid phosphatase were lower in membrane preparations of parasitized cells than in the controls.

Abstract

There are stage-specific differences of adenosine 5'-monophosphate (AMP)-catabolizing reactions in amastigote and promastigote Leishmania donovani cells: 1. the pH-optimum of nucleotidase activity is 9.2 in amastigote cells and 5.4 in promastigote cells, 2. adenosine, product of the nucleotidase reaction, is metabolized in a different way: in amastigote cells it is deaminated to inosine while in promastigote cells it is cleaved to adenine. Consequently, hypoxanthine is formed by a cleavage of inosine in amastigote cells and by a deaminating reaction of adenine in promastigote cells. These findings indicate that the rate of the adenylate energy charge and the purine nucleotide synthesis could be regulated stage-specifically. The membrane-bound nucleotidase might serve as a receptor and as the initial link of this sequence of reactions which signalizes an environmental change into the cell.

Abstract

Ornithine decarboxylase activity was inhibited in Plasmodium chabaudi-infected red cells of mice treated with chloroquine, mefloquine, primaquine and floxacrine. In vitro experiments on membrane-bound enzymes performed with intact P. chabaudi-infected red cells and corresponding ghost preparations showed a stimulation of NA+K+-ATP'ase and Ca2+Mg2+-ATP'ase activity in the presence of chloroquine, mefloquine and floxacrine.

Abstract

Chloroquine in a concentration range from 5 ng to 2 mug per ml of biological fluid can be determined in plasma and urine by the spectrofluorometric methods. The spectrodensitometric method is less sensitive and should be used when other drugs are interfering with the chloroquine determination in solution. After extraction from urine and separation from interfering drugs and metabolites by TL-chromatography chloroquine can be determined by spectrodensitometry in a minimum concentration of 5 mug per ml of urine. Chloroquine should be determined in plasma and urine of patients suffering from malaria with an impaired renal function. Whether the fluorometric determination of chloroquine should be used in field investigations will depend on the problem to be investigated and the regional facilities.
