1. Perspectives of Kanaka Well-Being: The Stories of Native Hawaiian Doctoral Students

Abstract

This study examined Kanaka (Native Hawaiian) well-being and focused on two questions: (a) How do Kanaka doctoral students describe their well-being? and (b)What lived experiences contribute to the development of their well-being? Rooted in a multiple case study design, this exploratory qualitative inquiry tells the story of 11 Kanaka doctoral students at the University of Hawai'i at Manoa and their genealogy of well-being. Well-being is explored through hula as a research approach. This approach is informed by a combination of Kanaka 'Oiwi Critical Race Theory with aspects of embodied cognition theories. It also draws upon hula as an art form in the research design. The author's hula genealogy framed what she heard, smelled, tasted, touched, and felt and informed how she heard, understood, and re-told the stories of the Kanaka who participated. She developed a (k)new understanding of well-being as pono (balance within the mind, body, and spirit) and malama (care for the relationships with others and 'iaina, as land and ancestor), to make sense of how contemporary Kanaka well-being was rooted in a practice of care. Kanaka doctoral students experienced and embodied imbalance, and through the process, I discovered pono and malama routines and activities that maintained their well-being practice. While challenging, the COVID-19 pandemic provided the opportunity for these Kanaka to honor and nurture themselves and their needs in ways that may have had other benefits. Results highlight the potential for prejudice to influence self-stigma and negative attitudes about self-care. Results also suggest the privilege of having a relationship with 'aina, and the need for cultivating a culture of care through healing Hawaiian communities. Abstract

BackgroundTo understand age-related changes in sweet taste perception in daily life, it is important to understand taste intensity at the suprathreshold level. Previous studies have attempted to characterize the temporal aspects of human taste perception in terms of time–intensity evaluations. The perception of dynamic taste intensity in older adults increases slowly for salty taste; however, there have been no previous studies on time–intensity sensory evaluation of sweet taste in older adults. We hypothesized that older adults perceive sweet taste intensity more slowly than young adults.MethodsFifty young and 40 older adults participated in the study and glucose solutions of 0.6 M and 1.5 M were used as stimuli. The study comprised two experiments: (1) a cup tasting test (static taste perception in the mouth), and (2) a time–intensity sensory evaluation, in which the solutions were presented using a custom-made delivery system. The intra-oral device was made to fit each participant’s dentition. Further, the level of gag reflex was taken into consideration for each participant in the design of the intra-oral device. A suction tube was placed across the posterior tongue near the throat to remove solution and saliva. The solution delivery system was controlled by an original computer program.ResultsOlder adults presented significantly different maximum intensity timing and slope for both concentrations compared with young adults (slope for 1.5 M, p < 0.01; others, p < 0.05). No significant differences were found between the older and young adults for reaction timing and maximum intensity.ConclusionWe conclude that older adults perceived sweetness more slowly than young adults, and ultimately perceived almost the same intensity as young adults. This is the first reported characterization of the time–intensity profile of sweet taste intensity of glucose in older adults. Using a standardized system enabled us to assess and compare feedback on taste intensities among different age groups in real-time. Based on this, we recommend older adults “savor” to perceive sweet tastes at the same level experienced by young adults.

Abstract

Abstract Structural plasticity of enzymes dictates their function. Yet, our ability to rationally remodel enzyme conformational landscapes to tailor catalytic properties remains limited. Here, we report a computational procedure for tuning conformational landscapes that is based on multistate design of hinge-mediated domain motions. Using this method, we redesign the conformational landscape of a natural aminotransferase to preferentially stabilize a less populated but reactive conformation and thereby increase catalytic efficiency with a non-native substrate, resulting in altered substrate selectivity. Steady-state kinetics of designed variants reveals activity increases with the non-native substrate of approximately 100-fold and selectivity switches of up to 1900-fold. Structural analyses by room-temperature X-ray crystallography and multitemperature nuclear magnetic resonance spectroscopy confirm that conformational equilibria favor the target conformation. Our computational approach opens the door to targeted alterations of conformational states and equilibria, which should facilitate the design of biocatalysts with customized activity and selectivity.

Abstract

Marine diazotrophs convert dinitrogen (N2) gas into bioavailable nitrogen (N), supporting life in the global ocean. In 2012, the first version of the global oceanic diazotroph database (version 1) was published. Here, we present an updated version of the database (version 2), significantly increasing the number of in situ diazotrophic measurements from 13 565 to 55 286. Data points for N2 fixation rates, diazotrophic cell abundance, and nifH gene copy abundance have increased by 184 %, 86 %, and 809 %, respectively. Version 2 includes two new data sheets for the nifH gene copy abundance of non-cyanobacterial diazotrophs and cell-specific N2 fixation rates. The measurements of N2 fixation rates approximately follow a log-normal distribution in both version 1 and version 2. However, version 2 considerably extends both the left and right tails of the distribution. Consequently, when estimating global oceanic N2 fixation rates using the geometric means of different ocean basins, version 1 and version 2 yield similar rates (43–57 versus 45–63 Tg N yr−1; ranges based on one geometric standard error). In contrast, when using arithmetic means, version 2 suggests a significantly higher rate of 223±30 Tg N yr−1 (mean ± standard error; same hereafter) compared to version 1 (74±7 Tg N yr−1). Specifically, substantial rate increases are estimated for the South Pacific Ocean (88±23 versus 20±2 Tg N yr−1), primarily driven by measurements in the southwestern subtropics, and for the North Atlantic Ocean (40±9 versus 10±2 Tg N yr−1). Moreover, version 2 estimates the N2 fixation rate in the Indian Ocean to be 35±14 Tg N yr−1, which could not be estimated using version 1 due to limited data availability. Furthermore, a comparison of N2 fixation rates obtained through different measurement methods at the same months, locations, and depths reveals that the conventional 15N2 bubble method yields lower rates in 69 % cases compared to the new 15N2 dissolution method. This updated version of the database can facilitate future studies in marine ecology and biogeochemistry. 5. Development of crystal orientation fabric in the Dome Fuji ice core in East Antarctica: implications for the deformation regime in ice sheets

Abstract

The crystal orientation fabric (COF) of a polar ice sheet has a significant effect on the rheology of the ice sheet. With the aim of better understanding the deformation regime of ice sheets, the work presented here investigates the COF in the upper 80 % of the Dome Fuji Station ice core in East Antarctica. Dielectric anisotropy (Δε) data were acquired as a novel indicator of the vertical clustering of COF resulting from vertical compressional strain within the dome. The Δε values were found to exhibit a general increase with depth, but with fluctuations over distances in the order of 10–102 m. In addition, significant decreases in Δε were found to be associated with depths corresponding to three major glacial to interglacial transitions. These changes in Δε are ascribed to variations in the deformational history caused by dislocation motion occurring from near-surface depths to deeper layers. Fluctuations in Δε over distances of less than 0.5 m exhibited a strong inverse correlation with Δε at depths greater than approximately 1200 m, indicating that they were enhanced during the glacial-interglacial transitions. The Δε data also exhibited a positive correlation with the concentration of chloride ions and an inverse correlation with the amount of dust particles in the ice core at greater depths corresponding to decreases in the degree of c axis clustering. Finally, we found that fluctuations in Δε persisted to approximately 80 % of the total depth of the ice sheet. These data suggest that the factors determining the deformation of ice include the concentration of chloride ions and the amount of dust particles, and that the layered contrast associated with the COF is preserved all the way from the near-surface to a depth corresponding to approximately 80 % of the thickness of the ice sheet. These findings provide important implications regarding further development of the COF under the various stress-strain configurations that the ice will experience in the deepest region, approximately 20 % of the total depth from the ice/bed interface.

6. Contrasting source contributions of Arctic black carbon to atmospheric concentrations, deposition flux, and atmospheric and snow radiative effects

Abstract

Black carbon (BC) particles in the Arctic contribute to rapid warming of the Arctic by heating the atmosphere and snow and ice surfaces. Understanding the source contributions to Arctic BC is therefore important, but they are not well understood, especially those for atmospheric and snow radiative effects. Here we estimate simultaneously the source contributions of Arctic BC to near-surface and vertically integrated atmospheric BC mass concentrations (MBC_SRF and MBC_COL), BC deposition flux (MBC_DEP), and BC radiative effects at the top of the atmosphere and snow surface (REBC_TOA and REBC_SNOW) and show that the source contributions to these five variables are highly different. In our estimates, Siberia makes the largest contribution to MBC_SRF, MBC_DEP, and REBC_SNOW in the Arctic (defined as >70∘ N), accounting for 70 %, 53 %, and 41 %, respectively. In contrast, Asia's contributions to MBC_COL and REBC_TOA are largest, accounting for 37 % and 43 %, respectively. In addition, the contributions of biomass burning sources are larger (29 %–35 %) to MBC_DEP, REBC_TOA, and REBC_SNOW, which are highest from late spring to summer, and smaller (5.9 %–17 %) to MBC_SRF and MBC_COL, whose concentrations are highest from winter to spring. These differences in source contributions to these five variables are due to seasonal variations in BC emission, transport, and removal processes and solar radiation, as well as to differences in radiative effect efficiency (radiative effect per unit BC mass) among sources. Radiative effect efficiency varies by a factor of up to 4 among sources (1471–5326 W g−1) depending on lifetimes, mixing states, and heights of BC and seasonal variations of emissions and solar radiation. As a result, source contributions to radiative effects and mass concentrations (i.e., REBC_TOA and MBC_COL, respectively) are substantially different. The results of this study demonstrate the importance of considering differences in the source contributions of Arctic BC among mass concentrations, deposition, and atmospheric and snow radiative effects for accurate understanding of Arctic BC and its climate impacts.

7. Differences in dynamic perception of salty taste intensity between young and older adults

Abstract

Abstract In super-aged societies, high salt intake substantially increases the risk of stroke and cardiovascular disease. Perceived low salty taste often prompts the addition of table salt to food. However, it remains unclear how older adults perceive the nature and intensity of salty taste in the mouth and brain. We compared the perceptions of salty taste intensities of older adults with those of young adults. Participants were 74 healthy adults: 31 older (age, 60–81 years [65.0 ± 5.5 SD]) and 43 young (age, 21–39 years [25.0 ± 3.6 SD]). Our research project comprises three sequential experiments. This article reports on the first two, which were (1) static and (2) dynamic sensory evaluations of taste perceptions in the mouth. Participants assessed the taste of 0.3 M and 0.5 M sodium chloride solutions in two types of sensory evaluations: (1) a cup tasting test, in which they sipped the solution from cups, spat it out, and rated static salty taste intensity, and (2) a time-intensity sensory evaluation, in which the solutions were delivered to participants’ tongues through a custom-made delivery system while they recorded dynamic taste intensities on a hand-held meter. 8. TReSR: A PCR-compatible DNA sequence design method for engineering proteins containing tandem repeats.

Abstract

Protein tandem repeats (TRs) are motifs comprised of near-identical contiguous sequence duplications. They are found in approximately 14% of all proteins and are implicated in diverse biological functions facilitating both structured and disordered protein-protein and protein-DNA interactions. These functionalities make protein TR domains an attractive component for the modular design of protein constructs. However, the repetitive nature of DNA sequences encoding TR motifs complicates their synthesis and mutagenesis by traditional molecular biology workflows commonly employed by protein engineers and synthetic biologists. To address this challenge, we developed a computational protocol to significantly reduce the complementarity of DNA sequences encoding TRs called TReSR (for Tandem Repeat DNA Sequence Redesign). The utility of TReSR was demonstrated by constructing a novel constitutive repressor synthesized by duplicating the LacI DNA binding domain into a single-chain TR construct by assembly PCR. Repressor function was evaluated by expression of a fluorescent reporter delivered on a single plasmid encoding a three-component genetic circuit. The successful application of TReSR to construct a novel TR-containing repressor with a DNA sequence that is amenable to PCR-based construction and manipulation will enable the incorporation of a wide range of TR-containing proteins for protein engineering and synthetic biology applications.

Abstract

Records of chemical impurities from ice cores enable us to reconstruct the past deposition of aerosols onto polar ice sheets and alpine glaciers. Through this they allow us to gain insight into changes of the source, transport and deposition processes that ultimately determine the deposition flux at the coring location. However, the low concentrations of the aerosol species in the ice and the resulting high risk of contamination pose a formidable analytical challenge, especially if long, continuous and highly resolved records are needed. Continuous flow analysis, CFA, the continuous melting, decontamination and analysis of ice-core samples has mostly overcome this issue and has quickly become the de facto standard to obtain high-resolution aerosol records from ice cores after its inception at the University of Bern in the mid-1990s. Here, we present continuous records of calcium (Ca2+), sodium (Na+), ammonium (NH4+), nitrate (NO3-) and electrolytic conductivity at 1 mm depth resolution from the NGRIP (North Greenland Ice Core Project) and NEEM (North Greenland Eemian Ice Drilling) ice cores produced by the Bern Continuous Flow Analysis group in the years 2000 to 2011 (Erhardt et al., 2021). Both of the records were previously used in a number of studies but were never published in full 1 mm resolution. Alongside the 1 mm datasets we provide decadal averages, a detailed description of the methods, relevant references, an assessment of the quality of the data and its usable resolution. Along the way we will also give some historical context on the development of the Bern CFA system. Abstract

In a honeycomb-lattice spin-orbit assisted Mott insulator α-RuCl_{3}, ultrafast magnetization is induced by circularly polarized excitation below the Mott gap. Photocarriers play an important role, which are generated by turning down the synergy of the onsite Coulomb interaction and the spin-orbit interaction realizing the insulator state. An ultrafast 6 fs measurement of photocarrier dynamics and a quantum mechanical analysis clarify the mechanism, according to which the magnetization emerges from a coherent charge motion between different t_{2g} orbitals (d_{yz}-d_{xz}-d_{xy}) of Ru^{3+} ions. This ultrafast magnetization is weakened in the antiferromagnetic (AFM) phase, which is opposite to the general tendency that the inverse Faraday effect is larger in AFM compounds than in paramagnetic ones. This temperature dependence indicates that the interorbital charge motion is affected by pseudospin rotational symmetry breaking in the AFM phase.

11. Variations in mineralogy of dust in an ice core obtained from northwestern Greenland over the past 100 years

Abstract

Our study is the first to demonstrate a high-temporal-resolution record of mineral composition in a Greenland ice core over the past 100 years. To reconstruct past variations in the sources and transportation processes of mineral dust in northwestern Greenland, we analysed the morphology and mineralogical composition of dust in the SIGMA-D ice core from 1915 to 2013 using scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS). The results revealed that the ice core dust consisted mainly of silicate minerals and that the composition varied substantially on multi-decadal and inter-decadal scales, suggesting that the ice core minerals originated from different geological sources in different periods during the past 100 years. The multi-decadal variation trend differed among mineral types. Kaolinite, which generally formed in warm and humid climatic zones, was abundant in colder periods (1950–2004), whereas mica, chlorite, feldspars, mafic minerals, and quartz, which formed in arid, high-latitude, and local areas, were abundant in warmer periods (1915–1949 and 2005–2013). Comparison to Greenland surface temperature records indicates that multi-decadal variation in the relative abundance of these minerals was likely affected by local temperature changes in Greenland. Trajectory analysis shows that the minerals were transported mainly from the western coast of Greenland in the two warming periods, which was likely due to an increase in dust sourced from local ice-free areas as a result of shorter snow/ice cover duration in the Greenland coastal region during the melt season caused by recent warming. Meanwhile, ancient deposits in northern Canada, which were formed in past warmer climates, seem to be the best candidate during the colder period (1950–2004). Our results suggest that SEM–EDS analysis can detect variations in ice core dust sources during recent periods of low dust concentration.

Abstract

Air in polar ice cores provides unique information on past climatic and atmospheric changes. We developed a new method combining wet extraction, gas chromatography and mass spectrometry for high-precision, simultaneous measurements of eight air components (CH4, N2O and CO2 concentrations; δ15N, δ18O, δO2∕N2 and δAr∕N2; and total air content) from an ice-core sample of ∼ 60 g. The ice sample is evacuated for ∼ 2 h and melted under vacuum, and the released air is continuously transferred into a sample tube at 10 K within 10 min. The air is homogenized in the sample tube overnight at room temperature and split into two aliquots for mass spectrometric and gas chromatographic measurements. Care is taken to minimize (1) contamination of greenhouse gases by using a long evacuation time, (2) consumption of oxygen during sample storage by a passivation treatment on sample tubes, and (3) fractionation of isotopic ratios with a long homogenization time for splitting. Precision is assessed by analyzing standard gases with artificial ice and duplicate measurements of the Dome Fuji and NEEM ice cores. The overall reproducibility (1 SD) of duplicate ice-core analyses are 3.2 ppb, 2.2 ppb and 2.9 ppm for CH4, N2O and CO2 concentrations; 0.006 ‰, 0.011 ‰, 0.09 ‰ and 0.12 ‰ for δ15N, δ18O, δO2∕N2 and δAr∕N2; and 0.63 mLSTP kg−1 for total air content, respectively. Our new method successfully combines the high-precision, small-sample and multiple-species measurements, with a wide range of applications for ice-core paleoenvironmental studies.

Summary: Generally, CT and MR images of antiresorptive agent-related osteonecrosis of the jaw (ARONJ)/medication-related osteonecrosis of the jaw (MRONJ) show nonspecific findings as seen in active osteomyelitis．However, there are some characteristics as follows; unilateral maxillary sinusitis adjacent to ipsilateral maxillary ARONJ/MRONJ, DRONJ presenting larger sequestrum and periosteal reaction more frequently than BRONJ, BRONJ resulting from intravenous administration of the drug presenting larger and more frequent buccolingual cortical bone perforations than BRONJ from oral administration, and better diagnostic accuracy of extent of perilesional soft tissue inflammation on MR imaging than CT. The CT and MR imaging features of ARONJ/MRONJ are summarized in this report. Abstract

Ice core derived marine biogenic sulphate does not agree with marine sediment records. Here based on new ice core records spanning the past 720,000 years obtained from Dome Fuji the authors propose that dust contributed a higher percentage of sulphate aerosols than previously thought.

Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. 23. Developing odontoma with an atypical radiological appearance: A case report

Abstract

Developing odontoma, formerly named ameloblastic fibro-odontoma (AFO), shows similar histopathologic appearance to ameloblastic fibroma (AF) in that it is composed of epithelial and mesenchymal elements. However, unlike AF, developing odontoma contains dental hard tissues. Radiologically, developing odontoma shows well-defined, mainly unilocular radiolucency containing various sizes and shapes of radiopaque materials, whereas AF shows well-defined unilocular or multilocular radiolucency without opacities. As such, the definition between developing odontoma and AF is generally clear in intraoral or panoramic radiographs. We present a case of developing odontoma which showed atypical radiological appearance apparently similar to AF. A 9-year-old boy was referred with the chief complaints of delayed eruption of the upper left second deciduous molar and first molar. Panoramic radiography revealed the radiolucent lesion with a well-defined margin and irregular surface. Impacted teeth were involved, but radiopaque materials were not found inside the lesion. Computed tomography showed a multilocular low-density lesion with only a few punctate enamel-like high-density areas. Magnetic resonance imaging showed a heterogeneous internal structure with the same signal intensity as muscle in T1-weighted images and high signal intensity in T2-weighted images with fat suppression. Although these images were atypical, developing odontoma was suspected based on the existence of enamel-like areas. AF was diagnosed by biopsy; however, the histopathological examination revealed dental hard tissues. Subsequently, it was diagnosed as developing odontoma. Recurrence has not been seen in 23 months postoperatively. Further reading of radiographic images over comparison with histopathological specimen was suggested to distinguish between atypical developing odontoma and AF. 26. COMPARING QUESTIONNAIRE RESULTS AND LEARNING OUTCOMES IN CLINICAL LABORATORY TRAINING PROGRAMS TO SUPPORT LEARNING IN JAPAN

Abstract

In Japan, the Ministry of Education, Culture, Sports, Science, and Technology requires every university to complete a learning behavior survey, assess the results, and disclose this information as part of the university's evaluation. This study examines the correlation between the results of the analysis conducted in a university for medical technologists and their learning outcomes, including GPA, graduation exam scores, and national exam scores. The learning behavior survey conducted in March 2020 included 345 participants studying medical technology. In total, 20 items were categorized based on seven factors. The results for the senior students were collated with their GPA scores, graduation exam scores, and national examination scores through factor analysis and multiple regression analysis using their scores and the factor scores. The results revealed that as students advance to upper grades, their efforts toward studying, such as study time, increase; however, no simple correlation exists between the increased study time and the resulting achievement. The medical technologist national exam scores were found to have a strong, positive correlation to three factors: effective use of leisure time, living environment, and observance of deadlines. The learning behavior survey analysis offers suggestions for students on the actions they must take to pass the national medical technologist exam.

Abstract

The anterior insula and rolandic operculum are key regions for flavour perception in the human brain; however, it is unclear how taste and congruent retronasal smell are perceived as flavours. The multisensory integration required for sour flavour perception has rarely been studied; therefore, we investigated the brain responses to taste and smell in the sour flavour-processing network in 35 young healthy adults. We aimed to characterise the brain response to three stimulations applied in the oral cavity—sour taste, retronasal smell of mango, and combined flavour of both—using functional magnetic resonance imaging. Effective connectivity of the flavour-processing network and modulatory effect from taste and smell were analysed. Flavour stimulation activated middle insula and olfactory tubercle (primary taste and olfactory cortices, respectively); anterior insula and rolandic operculum, which are associated with multisensory integration; and ventrolateral prefrontal cortex, a secondary cortex for flavour perception. Dynamic causal modelling demonstrated that neural taste and smell signals were integrated at anterior insula and rolandic operculum. These findings elucidated how neural signals triggered by sour taste and smell presented in liquid form interact in the brain, which may underpin the neurobiology of food appreciation. Our study thus demonstrated the integration and synergy of taste and smell.

Background: Dental anxiety is a common reason for avoiding dental visits and is associated with poor dental status. The short version of Dental Anxiety Inventory (SDAxI) is an easy-to-use, multi-faceted questionnaire for assessing the level of trait dental anxiety. However, there was no neurophysiological data indicating if its score associates with the state anxiety when an individual is under real/mock dental environment. We hypothesized that there exists such an association.Materials and Methods: Twenty systemic healthy adults with dental attendance experience and self-claimed free of dental phobia were recruited in this cross-sectional study, with their dental anxiety level assessed by SDAxI. Functional magnetic resonance imaging recorded their brain signals in response to audiovisual footages resembling dental scaler or turbine in action. After the brain imaging, they gave fear ratings to the footages in visual analog scale (VAS).Results: Participants’ SDAxI scores positively correlated with their responses in the insular cortex (r2 = 0.388–0.445, P < 0.005). Their SDAxI scores also positively correlated with their fear ratings of the footages (r2 = 0.415–0.555, P < 0.005).Discussion: Our findings indicated a possible neurobiological relevance of SDAxI, and reinforced its neurobiological validity in assessing dental anxiety level of dental attenders.

Abstract

The influence of temperature and time of snow sample melting on the measurement of mass concentration and size distribution of black carbon (BC) in snow was evaluated experimentally. In the experiments, fresh (Shirouma) and aged (Hakusan) snow samples were melted at different temperatures or at different time lengths, and the BC mass concentration and size distribution in the melted snow samples were measured using a nebulizer and a single-particle soot photometer (SP2). In the experiment where melting temperature was varied, the BC mass concentration in the liquid decreased at a melting temperature of 70 °C. This decrease was 8.0 % for the Shirouma sample and 46.4 % for the Hakusan sample and depended on BC particle size, with a significant decrease found at BC diameters less than 350 nm. A similar decrease in BC mass concentration was found when the Hakusan snow sample that had been melted at 5 °C was heated to 70 °C. The experiment in which melting time was varied indicated that BC mass concentration in the liquid did not change for the Shirouma sample but decreased significantly with a longer melting time for the Hakusan sample (38.6 %). These results indicate that melting of snow samples at high temperatures or over long time periods can significantly affect the measurement of BC mass and its size distribution, especially for aged snow samples.

Aims: The research aimed to investigate the shade response of E. gracilis Klebs while making the irradiance a crucial factor for photosynthesis based physiological activities and its applicability for industrial level culture conditions. The light compensation point (LCP), light saturation point (LSP), and dark respiration rate (DRR) were obtained. Cell volume and cell number in each culture were measured simultaneously. Cells were collected and obtained dry mass (DM) after drying aliquots at 80˚C. Specific growth rate (SGR) and relative growth rate (RGR) were calculated. Tests for homogeneity of variance were performed on all parameters and LSDs were used for the mean separation. Results: In the TP, the lowest LCP was achieved in the higher light culture. The values of both the DRR and the LSP were the same as in EP. The DRR, LCP and LSP are lower in lower PFD cultures and decreased with increasing cell titers. The cellular growth levels were lower in lower light culture and decreased as each culture grew. Cellular DM was maintained constant in the EP, where SGR almost equaled RGR. In the EP, SGR was maintained constant in each culture, SGR displayed a saturation phenomenon. In the later TP, SGR became equal to RGR and all the cultures revealed constant DM. Conclusion: Euglena photoautotrophic cultures can tolerate low light intensities. With the SGR and RGR behavior under the shade conditions, they can maintain the constant photosynthesis rate and constant dry matter level.

41. Readability of the 100 Most-Cited Neuroimaging Papers Assessed by Common Readability Formulae

Abstract

Background: From time to time, neuroimaging research findings receive press coverage and attention by the general public. Scientific articles therefore should be written in a readable manner to facilitate knowledge translation and dissemination. However, no published readability report on neuroimaging articles like those published in education, medical and marketing journals is available. As a start, this study therefore aimed to evaluate the readability of the most-cited neuroimaging articles.Methods: The 100 most-cited articles in neuroimaging identified in a recent study by Kim et al. (2016) were evaluated. Headings, mathematical equations, tables, figures, footnotes, appendices, and reference lists were trimmed from the articles. The rest was processed for number of characters, words and sentences. Five readability indices that indicate the school grade appropriate for that reading difficulty (Automated Readability Index, Coleman-Liau Index, Flesch-Kincaid Grade Level, Gunning Fog index and Simple Measure of Gobbledygook index) were computed. An average reading grade level (AGL) was calculated by taking the mean of these five indices. The Flesch Reading Ease (FRE) score was also computed. The readability of the trimmed abstracts and full texts was evaluated against number of authors, country of corresponding author, total citation count, normalized citation count, article type, publication year, impact factor of the year published and type of journal.Results: Mean AGL ± standard deviation (SD) of the trimmed abstracts and full texts were 17.15 ± 2.81 (college graduate level) and 14.22 ± 1.66 (college level) respectively. Mean FRE score ± SD of the abstracts and full texts were 15.70 ± 14.11 (college graduate level) and 32.11 ± 8.56 (college level) respectively. Both items indicated that the full texts were significantly more readable than the abstracts (p < 0.001). Abstract readability was not associated with any factors under investigation. ANCOVAs showed that review/meta-analysis (mean AGL ± SD: 16.0 ± 1.4) and higher impact factor significantly associated with lower readability of the trimmed full texts surveyed.Conclusion: Concerning the 100 most-cited articles in neuroimaging, the full text appears to be more readable than the abstracts. Experimental articles and methodology papers were more readable than reviews/meta-analyses. Articles published in journals with higher impact factors were less readable.

Abstract

Whereas ice cores from high-accumulation sites in coastal Antarctica clearly demonstrate annual layering, it is debated whether a seasonal signal is also preserved in ice cores from lower-accumulation sites further inland and particularly on the East Antarctic Plateau. In this study, we examine 5 m of early Holocene ice from the Dome Fuji (DF) ice core at a high temporal resolution by continuous flow analysis. The ice was continuously analysed for concentrations of dust, sodium, ammonium, liquid conductivity, and water isotopic composition. Furthermore, a dielectric profiling was performed on the solid ice. In most of the analysed ice, the multi-parameter impurity data set appears to resolve the seasonal variability although the identification of annual layers is not always unambiguous. The study thus provides information on the snow accumulation process in central East Antarctica. A layer counting based on the same principles as those previously applied to the NGRIP (North Greenland Ice core Project) and the Antarctic EPICA (European Project for Ice Coring in Antarctica) Dronning Maud Land (EDML) ice cores leads to a mean annual layer thickness for the DF ice of 3.0 ± 0.3 cm that compares well to existing estimates. The measured DF section is linked to the EDML ice core through a characteristic pattern of three significant acidity peaks that are present in both cores. The corresponding section of the EDML ice core has recently been dated by annual layer counting and the number of years identified independently in the two cores agree within error estimates. We therefore conclude that, to first order, the annual signal is preserved in this section of the DF core. This case study demonstrates the feasibility of determining annually deposited strata on the central East Antarctic Plateau. It also opens the possibility of resolving annual layers in the Eemian section of Antarctic ice cores where the accumulation is estimated to have been greater than in the Holocene.

44. Computational remodeling of an enzyme conformational landscape for altered substrate selectivity

Abstract

Structural plasticity of enzymes dictates their function. Yet, our ability to rationally remodel enzyme conformational landscapes to tailor catalytic properties remains limited. Here, we report a computational procedure for tuning conformational landscapes that is based on multistate design. Using this method, we redesigned the conformational landscape of a natural aminotransferase to preferentially stabilize a less populated but reactive conformation, and thereby increase catalytic efficiency with a non-native substrate to alter substrate selectivity. Steady-state kinetics of designed variants revealed selectivity switches of up to 1900-fold, and structural analyses by room-temperature X-ray crystallography and multitemperature nuclear magnetic resonance spectroscopy confirmed that conformational equilibria favoured the target conformation. Our computational approach opens the door to the fine-tuning of enzyme conformational landscapes to create designer biocatalysts with tailored functionality.

Abstract

Background: It might be difficult for clinicians and scientists to identify comprehensively the major research topics given the large number of publications. A bibliometric report that identifies the most-cited articles within the body of the relevant literature may provide insight and guidance for readers toward scientific topics that are considered important for researchers and all relevant workers of academia. To our knowledge, there is a lack of an overall evaluation of the most-cited articles and hence of a comprehensive review of major research topics in neuroscience. The present study was therefore proposed to analyze and characterize the 100 most-cited articles in neuroscience.Methods: Based on data provided from Web of Science, the 100 most-cited articles relevant to neuroscience were identified and characterized. Information was extracted for each included article to assess for the publication year, journal published, impact factor, adjusted impact factor, citation count (total, normalized, and adjusted), reference list, authorship and article type.Results: The total citation count for the 100 most-cited articles ranged from 7,326 to 2,138 (mean 3087.0) and the normalized citation count ranged from 0.163 to 0.007 (mean 0.054). The majority of the 100 articles were research articles (67%) and published from 1996 to 2000 (30%). The author and journal with the largest share of these 100 articles were Stephen M. Smith (n = 6) and Science (n = 13) respectively. Among the 100 most-cited articles, 37 were interlinked via citations of one another, and they could be classified into five major topics, four of which were scientific topics, namely neurological disorders, prefrontal cortex/emotion/reward, brain network, and brain mapping. The remaining topic was methodology. Interestingly 41 out of 63 of the rest, non-interlinked articles could also be categorized under the above five topics. Adjusted journal impact factor among these 100 articles did not appear to be associated with the corresponding adjusted citation count.Conclusion: The current study compiles a comprehensive list and analysis of the 100 most-cited articles relevant to neuroscience that enables the comprehensive identification and recognition of the most important and relevant research topics concerned.

Abstract

Mature progression-free survival (PFS) data from the phase III J-ALEX study showed superiority for alectinib versus crizotinib [hazard ratio (HR) 0.37, 95% confidence interval (CI) 0.26-0.52; median PFS 34.1 versus 10.2 months, respectively] in advanced ALK (anaplastic lymphoma kinase)-positive non-small-cell lung cancer (NSCLC). Overall survival (OS) data were immature (HR 0.80, 99.8799% CI 0.35-1.82) at the time of data cut-off (30 June 2018). We report final OS data after ≥5 years of follow-up.ALK inhibitor naive Japanese patients who were chemotherapy naive or had received one prior chemotherapy regimen were enrolled. Patients were randomized to receive alectinib 300 mg (n = 103) or crizotinib 250 mg (n = 104) twice daily until progressive disease, unacceptable toxicity, death, or withdrawal. The primary endpoint was independent review facility-assessed PFS, with OS (not fully powered) as a secondary endpoint.Median duration of OS follow-up was 68.6 months with alectinib and 68.0 months with crizotinib. Treatment with alectinib did not prolong OS relative to crizotinib (HR 1.03, 95.0405% CI 0.67-1.58; P = 0.9105). Five-year OS rates were 60.9% (95% CI 51.4-70.3) with alectinib and 64.1% (95% CI 54.9-73.4) with crizotinib. In total, 91.3% (n = 95/104) of crizotinib-treated patients and 46.6% (n = 48/103) of alectinib-treated patients received at least one subsequent anticancer therapy. After study drug discontinuation, 78.8% of patients in the crizotinib arm switched to alectinib, while 10.7% of patients in the alectinib arm switched to crizotinib as a first subsequent anticancer therapy. Patients randomized to crizotinib tended to switch treatment earlier than those randomized to alectinib.Final OS analysis from J-ALEX did not show superiority of alectinib to crizotinib; this result was most likely confounded by treatment crossover. Alectinib remains a standard of care for the treatment of patients with advanced ALK-positive NSCLC.

Abstract

Background The purpose of this study was to compare the displacement of tooth arrangement in dentures fabricated by additive manufacturing (AM) and heat curing. Methods Three-dimensional (3D) scanning was performed for edentulous jaw models. After the teeth were arranged, 3D scanning for the wax denture was performed. Heat-cured dentures were fabricated with heat-cure polymer resin. Based on data obtained by subtracting the model data from wax denture data, AM dentures were fabricated from ultraviolet-cured acrylic resin. Accuracy was verified by superimposing heat-cured and AM dentures on the tooth region data from the wax dentures and measuring displacement of the tooth arrangement. Results In the maxillary dentures, the amount of tooth displacement for the heat-cured dentures and for the AM dentures ranged from -0.08 to +0.06 mm and from -0.25 to +0.06 mm respectively. A significant difference was observed between two dentures. In the mandibular dentures, the amount of tooth displacement for the heat-cured dentures and for the AM dentures ranged from -0.09 to +0.07 mm and from -0.03 to +0.07 mm respectively. No significant difference was observed between two dentures. Conclusions The artificial teeth of the maxillary dentures fabricated by AM showed a greater displacement compared to those by heat curing.

Abstract

Objectives The JO25567 randomized Phase II study demonstrated a statistically significant progression-free survival (PFS) benefit with erlotinib plus bevacizumab compared with erlotinib monotherapy in chemotherapy-naive Japanese patients with epidermal growth factor receptor mutation-positive (EGFR+) non-small-cell lung cancer (NSCLC). Here we present updated PFS and final overall survival (OS) data after a median follow-up of 34.7 months. Materials and methods Patients with stage IIIB/IV or postoperative recurrent NSCLC were randomized to receive oral erlotinib 150 mg once daily (n = 77) or erlotinib in combination with intravenous bevacizumab 15 mg/kg every 21 days (n = 75) until disease progression or unacceptable toxicity. OS was analyzed using an unstratified Cox proportional hazards model. Results Consistent with the primary analysis, addition of bevacizumab to erlotinib was associated with a significant improvement in PFS (hazard ratio [HR] 0.52; 95 % confidence interval [CI]: 0.35–0.76; log-rank two-sided P = 0.0005; median 16.4 months vs 9.8 months, respectively). In contrast, a significant improvement in OS was not seen (HR 0.81; 95 % CI, 0.53–1.23; P = 0.3267; median 47.0 months vs 47.4 months, respectively). Post-study therapy was similar between the treatment arms and EGFR mutation type did not affect OS outcomes. The 5-year OS rate was numerically higher with erlotinib plus bevacizumab vs erlotinib monotherapy (41 % vs 35 %). Updated safety analyses confirmed the previously reported manageable tolerability profile, with no new safety issues. Conclusion Addition of bevacizumab to first-line erlotinib did not show significant improvement in OS in Japanese patients with stage IIIB/IV or postoperative recurrent EGFR+ NSCLC. Both treatment arms showed a similar median OS benefit (as long as 4 years), irrespective of individual patient characteristics. Results from ongoing studies evaluating the combination of EGFR and VEGF signaling inhibitors are eagerly awaited. Trial registration JapicCTI-111390 and JapicCTI-142569.
