Currently, a change in the species composition of the Białowieża Forest, eastern Poland, is occurring. Parallel to the dying of spruce (Picea abies L. Karst.), caused by Ips typographus (L.), there is a spread of deciduous tree species, among which hornbeam (Carpinus betulus L.) is the most active. Against the background of climate change, it is of interest to consider the possibility of reintroducing relict species in areas occupied until recently by spruce. One of these is silver fir (Abies alba Mill.), which is biologically and ecologically very similar to spruce. The Tisovik tract (the Belarusian part of the Białowieża Forest) is the most northeastern and the only preserved refuge of autochthonous silver fir in the region. Therefore, it is the most suitable source of propagation of this species outside the mountainous part of its range. The target area of our study was a 26-year old artificial stand included in the information system of forest genetic resources conserved in Europe. It was created in 1996 in the Polish part of the Białowieża Forest in the Hajnówka Forest District (Forestry Wilczy Jar) from 10 half-sib families originating from seeds collected in the Tisovik tract. The goal of the study was to assess the influence of provenance factors and spatial structure on productivity and phenotypic variation as measured by diameter (DBH). The study’s initial results showed that the provenance factor’s influence at the pole wood stage manifests itself more clearly than spatial structure. Even within the framework of individual half-sibs, there was a clear differentiation of diameters between investigated trees. Interpretation of the obtained results allowed us to conclude that there is a “threshold value” of the distance between trees in a stand of a given age, which determines the point when its increase no longer contributes to an increase in DBH. The most promising phenotypes were selected for further research and practical actions to reproduce silver fir further.

This study validated Polish versions of the Coronavirus Stress Measure (CSM) and the COVID-19 Burnout Scale (COVID-19-BS) to measure stress and burnout associated with COVID-19. Participants were 431 Polish young adults (72.6% female; Meanage = 26.61 ± 12.63). Confirmatory factor analysis verified a one-factor solution for both the CSM and the COVID-19-BS. Both scales had high internal consistency reliability. Coronavirus stress and COVID-19 burnout were positively related to depression, anxiety, and stress and negatively related to resilience. The coronavirus stress and COVID-19 burnout were correlated with elevated levels of depression, anxiety, and stress over and beyond resilience, age, and gender. Findings suggest that the Polish versions of the CSM and the COVID-19-BS are valid scales to measure stress and burnout related to COVID-19. Findings also demonstrated that the coronavirus stress and COVID-19 burnout experienced during the later stages of the pandemic might be a permanent risk factor for mental health problems.

Producción Científica, Fusarium circinatum, the causal agent of pine pitch canker (PPC), is currently one of the most important threats of Pinus spp. globally. This pathogen is known in many pine-growing regions, including natural and planted forests, and can affect all life stages of trees, from emerging seedlings to mature trees. Despite the importance of PPC, the global distribution of F. circinatum is poorly documented, and this problem is also true of the hosts within countries that are affected. The aim of this study was to review the global distribution of F. circinatum, with a particular focus on Europe. We considered (1) the current and historical pathogen records, both positive and negative, based on confirmed reports from Europe and globally; (2) the genetic diversity and population structure of the pathogen; (3) the current distribution of PPC in Europe, comparing published models of predicted disease distribution; and (4) host susceptibility by reviewing literature and generating a comprehensive list of known hosts for the fungus. These data were collated from 41 countries and used to compile a specially constructed geo-database. A review of 6297 observation records showed that F. circinatum and the symptoms it causes on conifers occurred in 14 countries, including four in Europe, and is absent in 28 countries. Field observations and experimental data from 138 host species revealed 106 susceptible host species including 85 Pinus species, 6 non-pine tree species and 15 grass and herb species. Our data confirm that susceptibility to F. circinatum varies between different host species, tree ages and environmental characteristics. Knowledge on the geographic distribution, host range and the relative susceptibility of different hosts is essential for disease management, mitigation and containment strategies. The following paper presents the results on the determination of the diversity of species from the Phytophthora genus occurring in the declining oak stands in Krotoszyn Plateau in Poland. From the 50s of the last century, significant deterioration of oak health was observed in these stands, and Phytophthora species were suggested as one of the factors of the decline. In order to determine the presence of pathogenic organisms from the Phytophthora genus in these stands, 180 rhizosphere soil samples from three forest districts throughout the Krotoszyn Plateau were collected and subjected to the isolation method. Phytophthora species were consistently isolated from all the sampled stands, and 194 isolates from 111 positive samples were obtained. However, 150 (77%) and 44 (23%) isolates originated from the samples taken under the symptomatic and asymptomatic trees, respectively. All the obtained isolates were morphologically classified using the light and scanning electron microscopy and divided into morphological groups. Genomic DNA was isolated from selected isolates representing each group, ITS regions were amplified and sequence analyses were performed. In total, four different Phytophthora species were detected, including P. cactorum, P. plurivora, P. quercina and P. europaea. The most often isolated species were P. cactorum and P. plurivora. This is the first report of P. europaea in oak stands in Poland.

Background aims. Epstein-Barr virus (EBV)-associated post-transplant lymphoproliferative disorders (PTLD) belong to the most dreaded complications of immunosuppression. The efficacy of EBV-specific T-cell transfer for PTLD has been previously shown, yet the optimal choice of EBV-derived antigens inducing polyclonal CD4þ and CD8þ T cells that cover a wide range of human leukocyte antigen types and efficiently control PTLD remains unclear. Methods. A pool of 125 T-cell epitopes from seven latent and nine lytic EBV-derived proteins (EBVmix) and peptide pools of EBNA1, EBNA3c, LMP2a and BZLF1 were used to determine T-cell frequencies and to isolate T cells through the use of the interferon (IFN)-g cytokine capture system. We further evaluated the phenotype and functionality of the generated T-cell lines in vitro. Results. EBVmix induced significantly higher T-cell frequencies and allowed selecting more CD4þIFN-gþ and CD8þIFN-gþ cells than single peptide pools. T cells of all specificities expanded similarly in vitro, recognized cognate antigen, and, to a lower extent, EBV-infected cells, exerted moderate cytotoxicity and showed reduced alloreactivity. However, EBVmix-specific cells most efficiently controlled EBV-infected lymphoblastoid cell lines (LCLs). This control was mainly mediated by EBVspecific CD8þ cells with an oligoclonal epitope signature covering both latent and lytic viral proteins. Notably, EBVspecific CD4þ cells unable to control LCLs produced significantly less perforin and granzyme B, probably because of limited LCL epitope presentation. Conclusions. EBVmix induces a broader T-cell response, probably because of its coverage of latent and lytic EBV-derived proteins that may be important to control EBV-transformed B cells and might offer an improvement of T-cell therapies.

The paper presents the results of the study performed with aims to determine the presence and diversity of Phytophthora species on maple trees in Serbia. Due to high aggressiveness and their multicyclic nature, presence of these pathogens is posing significant threat to forestry and biodiversity. In total, 29 samples of water, soil and tissues were taken from 10 different localities, and six different maple hosts were tested. After the isolation tests, 17 samples from five different maple hosts were positive for the presence of Phytophthora spp., and 31 isolates were obtained. After the detailed morphological and physiological classification, four distinct groups of isolates were separated. DNA was extracted from selected representative isolates and molecular identification with sequencing of ITS region was performed. Used ITS4 and ITS6 primers successfully amplified the genomic DNA of chosen isolates and morphological identification of obtained isolates was confirmed after the sequencing. Four different Phytophthora species were detected, including P. cactorum, P. gonapodyides, P. plurivora and P. lacustris. The most common isolated species was homothallic, and with very variable and semipapillate sporangia, P. plurivora with 22 obtained isolates. This is the first report of P. plurivora and P. gonapodyides on A. campestre, P. plurivora and P. lacustris on Acer heldreichii and first report of P. lacustris on A. pseudoplatanus and A. tataricum in Serbia. Numerous studies have shown that several Phytophthora species are directly connected with the tree decline in various forests, and that these organisms are the main cause of decay and loss of fine roots, necrosis as well as of the wounds of mother roots. Due to the previously recorded symptoms and high risk to forestry and biodiversity posed by the presence of these species, a study was performed in the Forest Estate „Sumarstvo“ - Raska, in the forest management unit „Turjak-Vrsine“, Forest administration in Novi Pazar. The aims of the research were to isolate and identify the Phytophthora species and their associated hosts. The samples of rhizophere soil, water and necrotic tissues were taken, and the isolation was performed according to a known methodology, using baits and direct setting of tissue culture on the media. Eleven hosts were tested, five hosts were positive and a total of 29 isolates were obtained. The hosts with the most isolates obtained were Alnus glutinosa with 12 and Fagus sylvatica with 7 isolates. The most frequently isolated species was Phytophthora plurivora, which occurred on three hosts. P. cactorum was also obtained, and the identification of five other isolates is currently going on. This is the first record of P. plurivora, P. gonapodyides and Phytophthora spp. on alder trees in Serbia. [Projekat Ministarstva nauke Republike Srbije, br. 'The protection against discrmination practices in the field of employment in resp e c t o f P o l i sh a n d G e r m a n l a w i n t h e l i g h t o f E U d i r e c t i v e s ' The article is devoted to the subject of non-discrimination regulations during employment in respect of Polish and German law. The thesis examines the legal basis of non-discrimination and equality in employment and occupation with reference to European Union law. Regulations against workplace discrimination are included in primary and secondary sources of European Union legislation. The fundamental regulations for countering discrimination are European directives, which forbid discrimination based on race, ethnic origins, religion, beliefs, disability, age, sexual orientation and other listed grounds for hiring, firing, job advertisements and other terms and conditions of employment. Directives are implemented by the EU m ember states. According to the Treaty on the Functioning of the European Union they are free to choose the form and methods of implementation in a way that ensures the most effective protection against discrimination. Implementation of the EU anti-discrimination directives in Poland was executed by amending Polish labour law, while Germany decided to transpose them through the establishment of new regulations - the General Equal Treatment Act. The paper presents definitions of different forms of discrimination (e.g. direct, indirect), exceptions to anti-discrimination legislation and other issues related to protection against discrimination in German and Polish law.

Multidrug-resistant (MDR) cytomegalovirus (CMV) emerged after transient responses to ganciclovir, foscarnet, and cidofovir in a CMV-seropositive recipient who underwent allogeneic hematopoietic stem cell transplantation from a CMV-seronegative donor. Experimental treatments using leflunomide and artesunate failed. Re-transplantation from a CMV-seropositive donor supported by adoptive transfer of pp65-specific T cells and maribavir was followed by lasting suppression. This case illustrates that successful MDR CMV therapy may require individualized multidisciplinary approaches.

Wheat germin is a homopentameric 125 kD glycoprotein mainly localized in the cell wall of monocots, and is a specific marker of the onset of growth in germinating seeds. The major objective of this study was to examine the expression and oxalate oxidase activity of two wheat germin isoforms: gf-2.8 and gf-3.8 in transgenic tobacco plants. The transgenic tobacco plants were created with different constructs: 1) one entire excision of gf-2.8 germin promoter and two partially deleted promoter sequences were used to generate 3 independent GUS constructs; 2) the whole gf-2.8 gene construct and the fusion with CaMV 35S promoter; 3) one entire excision of gf-3.8 germin gene and one partially deleted gf-3.8 promoter sequences were used to generate 2 independent GUS constructs; 4) the whole gf-3.8 gene and the fusion with CaMV 35S promoter. Hormonal treatment (auxin and gibberellin), salt treatment, heavy metals (Mn, Fe, Co, Ni, Cu, Zn, Cd, Hg, As) and Al induced high GUS activity in tobacco transformed with entire and one partially deleted of the gf-2.8 gene. The immunoblotting confirmed induction of gf-2.8 gene and its product expressed oxalate oxidase activity in tobacco transformed with the entire gf-2.8 construct. Neither nicotinic acid, salicylic acid, heat shock, cold nor UV-C have enhanced significant GUS activity and germin gf-2.8 synhesis and activity.

[Abstract] One of the most compelling approaches in the discovery of novel antimicrobials is screening of natural sources. In our publication we report on the activity of a compound 8hydroxyserrulat-14-en-19-oic acid (EN4), a diterpene isolated from the Australian plant Eremophila neglecta. We evaluate its applicability for treatment of implant-associated infections. A comprehensive analysis of the mechanism of action of EN4 against Staphylococci revealed its membranolytic properties and a general inhibition of macromolecular biosynthesis, which was confirmed in a macromolecular biosynthesis assay and suggested a multitarget activity. The method used to investigate an influence of EN4 on the synthesis of peptidoglycan, RNA, DNA and proteins is based on precipitation of macromolecules with trichloroacetic acid. These macromolecules are synthesised from respective [ 3 H]-labelled precursors. The incorporated radioactivity with and without an antimicrobial is measured and it reflects the mode of action of the tested compound. Antibiotics with known mechanisms of action are used as controls.

Treatment options are limited for implant-associated infections (IAI) that are mainly caused by biofilm-forming staphylococci. We report here on the activity of the serrulatane compound 8-hydroxyserrulat-14-en-19-oic acid (EN4), a diterpene isolated from the Australian plant Eremophila neglecta . EN4 elicited antimicrobial activity toward various Gram-positive bacteria but not to Gram-negative bacteria. It showed a similar bactericidal effect against logarithmic-phase, stationary-phase, and adherent Staphylococcus epidermidis , as well as against methicillin-susceptible and methicillin-resistant S. aureus with MICs of 25 to 50 μg/ml and MBCs of 50 to 100 μg/ml. The bactericidal activity of EN4 was similar against S. epidermidis and its Δ ica mutant, which is unable to produce polysaccharide intercellular adhesin-mediated biofilm. In time-kill studies, EN4 exhibited a rapid and concentration-dependent killing of staphylococci, reducing bacterial counts by >3 log 10 CFU/ml within 5 min at concentrations of >50 μg/ml. Investigation of the mode of action of EN4 revealed membranolytic properties and a general inhibition of macromolecular biosynthesis, suggesting a multitarget activity. In vitro -tested cytotoxicity on eukaryotic cells was time and concentration dependent in the range of the MBCs. EN4 was then tested in a mouse tissue cage model, where it showed neither bactericidal nor cytotoxic effects, indicating an inhibition of its activity. Inhibition assays revealed that this was caused by interactions with albumin. Overall, these findings suggest that, upon structural changes, EN4 might be a promising pharmacophore for the development of new antimicrobials to treat IAI.

Thioredoxin (Trx)-1 is a small, ubiquitously expressed redox-active protein with known important cytosolic functions. However, Trx1 is also upregulated in response to various stress stimuli, is found both at the cell surface and secreted into plasma, and has known anti-inflammatory and antiapoptotic properties. Previous animal studies have demonstrated that exogenous Trx1 delivery can have therapeutic effects in a number of disease models and have implicated an interaction of Trx1 with the complement system. We found that Trx1 is expressed in a redox-active form at the surface of HUVEC and acts as an inhibitor of complement deposition in a manner dependent on its Cys-Gly-Pro-Cys active site. Inhibition occurred at the point of the C5 convertase of complement, regulating production of C5a and the membrane attack complex. A truncated form of Trx1 also exists in vivo, Trx80, which has separate nonoverlapping functions compared with the full-length Trx1. We found that Trx80 activates the classical and alternative pathways of complement activation, leading to C5a production, but the inflammatory potential of this was also limited by the binding of inhibitors C4b-binding protein and factor H. This study adds a further role to the known anti-inflammatory properties of Trx1 and highlights the difference in function between the full-length and truncated forms.

The aim of this article is to assess the impact of the tax… in the special economic zones on the state of finances of territorial council. The investigation was based on the example of first Polish special economic zone—the SSE Euro—Park Mielec. The period of functioning of the zone in Mielec was described from the point of its establishment in 1995. The analysis which was carried out showed that it is not essential for the local budget if the zone inventors have the tax exemption. What is more, it was noticed that there was a notable growth of budget's income from the income taxes and real estate taxes. It was the result of the dynamic development in enterprise in the region of collapsing industry. The inflowing investors to the zone of Mielec, who were encouraged by the given tax privileges, contributed the formation of next companies in the area. The SSE Euro—Park Mielec ended the monoculture of the industry through the diversification of the structure of industry. However its most important contribution was overcoming the crisis on the labour market—unemployment rate is nowadays less than the average for Poland and Podkarpackie province. The example of Mielec confirms the high effectiveness of the special economic zones as the stimulating instrument of economic development, particularly on the regional level. Preferential tax policy had consequences in the economic promotion of regions in Poland, not only in the country, but abroad as well.

A model for tree pathogen diagnosis – Prunus domestica L. has been studied against pathogenic bacteria. An orchard of 110 trees of P. domestica showed dying back symptoms in May 2009 and nineteen of these trees were eradicated and burnt for prophylaxis. No symptoms correlated with those caused by pathogens previously observed in stone fruit die back in Europe or elsewhere (Pseudomonas syringae pv syringae van Hall, Pseudomonas syringae pv morsprunorum Lazarowtz, Phytophthora sp., Diaporthe perniciosa Marchal., phytoplasma or viruses) were not found. Interestingly, cutting the trunk in transversal sections allowed the observation of stem heart necrosis which was mostly important at the grafting point. Isolations from necrotic stem heart allowed to identify anot yet described Pseudomonas species not related to P. syringae. The method described in the paper for isolation of pathogenic bacteria and their quick an reliable identification can be also applied for detection of pathogens in forest tree plantations. BACKGROUND: Aspergillus and Mucorales species cause severe infections in patients after hematopoietic stem cell transplantation (HSCT). Induction of antifungal CD4(+) T-helper type 1 (Th1) immunity is an appealing strategy to combat these infections. Immunotherapeutic approaches are so far limited because of a lack of antigens inducing protective T cells, their elaborate production, and the need of targeting a broad spectrum of pathogenic fungi. METHODS: We examined the response to different Aspergillus fumigatus proteins in healthy individuals and patients after HSCT and compared rapid selection protocols for fungus-specific T cells based on CD137 or CD154 expression. RESULTS: The A. fumigatus proteins Crf1, Gel1, and Pmp20 induced strong Th1 responses in healthy individuals. T cells specific for these antigens expanded in patients with active invasive aspergillosis, indicating their contribution to infection control. Th1 cells specific for the 3 proteins can be selected with similar specificity within 24 hours, based on CD137 or CD154 expression. These cells recognize naturally processed A. fumigatus and the multispecific T-cell lines, directed against all 3 proteins, especially those selected by CD154, additionally cross-react to different Aspergillus and Mucorales species. CONCLUSIONS: These findings may form the basis for adoptive T-cell transfer for prophylaxis or treatment in patients with these devastating infections.

Thin polymer films that prevent the adhesion of bacteria are of interest as coatings for the development of infection-resistant biomaterials. This study investigates the influence of grafting density and film thickness on the adhesion of Staphylococcus epidermidis to poly(poly(ethylene glycol) methacrylate) (PPEGMA) and poly(2-hydroxyethyl methacrylate) (PHEMA) brushes prepared via surface-initiated atom transfer radical polymerization (SI-ATRP). These brushes are compared with poly(ethylene glycol) (PEG) brushes, which are obtained by grafting PEG onto an epoxide-modified substrate. Except for very low grafting densities (rho = 1%), crystal violet staining experiments show that the PHEMA and PPEGMA brushes are equally effective as the PEG-modified surfaces in preventing S. epidermis adhesion and do not reveal any significant variations as a function of film thickness or grafting density. These results indicate that brushes generated by SI-ATRP are an attractive alternative to grafted-onto PEG films for the preparation of surface coatings that resist bacterial adhesion.
