Abstract Background CHRDL1 belongs to a novel class of mRNA molecules. Nonetheless, the specific biological functions and underlying mechanisms of CHRDL1 in oral squamous cell carcinoma (OSCC) remain largely unexplored. Methods RT-qPCR and immunohistochemical staining were employed to assess the mRNA and protein expression levels of the MED29 gene in clinical samples of OSCC. Additionally, RT-qPCR and Western Blot analyses were conducted to investigate the mRNA and protein expression levels of the MED29 gene specifically in OSCC. The impact of MED29 on epithelial-mesenchymal transition (EMT), invasion, and migration of OSCC was evaluated through scratch assay, transwell assay, and immunofluorescence staining. Furthermore, wound healing assay and Transwell assay were utilized to examine whether CHRDL1 influences the malignant behavior of OSCC by modulating MED29 in vitro. The regulatory role of CHRDL1 on MED29 was further elucidated in vivo through a tail vein lung metastasis model in nude mice. Results MED29 expression was elevated in tumor tissues of OSCC patients compared with adjacent cancer tissues. Moreover, in CAL27 and SCC25 cell lines, MED29 was upregulated and associated with increased cell migration and invasion abilities. Overexpression of MED29 facilitated EMT in OSCC cell lines, whereas knockdown of MED29 impeded EMT, resulting in diminished cell migration and invasion capacities. CHRDL1 exerted inhibitory effects on the expression of MED29, thereby suppressing EMT progression and consequently restraining the invasion and migration of OSCC cells. Furthermore, CHRDL1 mediated the inhibition of migration of OSCC cell lines to the OSCC through its regulation of MED29. Conclusions MED29 facilitated the epithelial-mesenchymal transition process in OSCC, thereby promoting migration and invasion. On the other hand, CHRDL1 exerted inhibitory effects on the invasion and metastasis of OSCC by suppressing MED29 through the inhibition of the MAPK signaling pathway.

In situ regeneration is a promising strategy for constructing tissue engineering heart valves (TEHVs). Currently, the decellularized heart valve (DHV) is extensively employed as a TEHV scaffold. Nevertheless, DHV exhibits limited blood compatibility and notable difficulties in endothelialization, resulting in thrombosis and graft failure. The red blood cell membrane (RBCM) exhibits excellent biocompatibility and prolonged circulation stability and is extensively applied in the camouflage of nanoparticles for drug delivery; however, there is no report on its application for large-scale modification of decellularized extracellular matrix (ECM). For the first time, we utilized a layer-by-layer assembling strategy to immobilize RBCM on the surface of DHV and construct an innovative TEHV scaffold. Our findings demonstrated that the scaffold significantly improved the hemocompatibility of DHV by effectively preventing plasma protein adsorption, activated platelet adhesion, and erythrocyte aggregation, and induced macrophage polarization toward the M2 phenotype in vitro. Moreover, RBCM modification significantly enhanced the mechanical properties and enzymatic stability of DHV. The rat models of subcutaneous embedding and abdominal aorta implantation showed that the scaffold regulated the polarization of macrophages into the anti-inflammatory and pro-modeling M2 phenotype and promoted endothelialization and ECM remodeling in the early stage without thrombosis and calcification. The novel TEHV exhibits excellent performance and can overcome the limitations of commonly used clinical prostheses.

Abstract The influence of liver fibrosis on the rate of liver regeneration and complications following ALPPS has yet to be fully understood. This study aimed to scrutinize the effects of liver fibrosis on the postoperative complications, and prognosis subsequent to ALPPS. Clinical data were collected from patients with primary liver cancer who underwent ALPPS at Peking Union Medical College Hospital between May 2014 and October 2022. The degree of liver fibrosis was assessed using haematoxylin–eosin staining and Sirius red staining. This study encompassed thirty patients who underwent ALPPS for primary liver cancer, and there were 23 patients with hepatocellular carcinoma, 5 with cholangiocarcinoma, and 2 with combined hepatocellular-cholangiocarcinoma. The impact of severe liver fibrosis on the rate of liver regeneration was not statistically significant (P = 0.892). All patients with severe complications belonged to the severe liver fibrosis group. Severe liver fibrosis exhibited a significant association with 90 days mortality (P = 0.014) and overall survival (P = 0.012). Severe liver fibrosis emerges as a crucial risk factor for liver failure and perioperative mortality following the second step of ALPPS. Preoperative liver function impairment is an important predictive factor for postoperative liver failure.

Abstract Diabetes often induces bone loss and the dysregulation of gut microbiota (GM) is an important cause. Punicalagin (PU) was reported to regulate GM. Therefore, we hypothesized that PU could alleviate diabetes‐induced bone loss through GM and their metabolites. In this study, high‐fat diet‐induced diabetic mice showed bone erosion and poor biomechanical properties, while PU intake significantly improved the bone condition of diabetic mice. Further investigation revealed that the abundance of some beneficial bacteria, such as Akkermansia and Ruminiclostridium_9, was higher after PU intake and highly positively correlated with the concentrations of short‐chain fatty acids (SCFAs) and serum vitamin K2, respectively. In addition, these bacteria were associated with the levels of bone metabolism‐related markers such as procollagen type I N‐terminal propeptide (P1NP) and runt‐related transcription factor‐2 (Runx2). Mechanistically, PU, on the one hand, promotes the metabolism of SCFAs, thereby increasing the levels of bone synthesis markers and inhibiting the secretion of bone absorption markers. On the other hand, the higher level of vitamin K2 greatly accelerated bone mineralization and enhanced bone strength. This work provides a new perspective to explore the mechanism by which PU intervention alleviates diabetes‐induced bone loss by regulating the GM and their metabolic products.

Abstract Skyrmions in existing 2D van der Waals (vdW) materials have primarily been limited to cryogenic temperatures, and the underlying physical mechanism of the Dzyaloshinskii–Moriya interaction (DMI), a crucial ingredient for stabilizing chiral skyrmions, remains inadequately explored. Here, we report the observation of Néel-type skyrmions in a vdW ferromagnet Fe3−x GaTe2 above room temperature. Contrary to previous assumptions of centrosymmetry in Fe3−x GaTe2, the atomic-resolution scanning transmission electron microscopy reveals that the off-centered FeΙΙ atoms break the spatial inversion symmetry, rendering it a polar metal. First-principles calculations further elucidate that the DMI primarily stems from the Te sublayers through the Fert–Lévy mechanism. Remarkably, the chiral skyrmion lattice in Fe3−x GaTe2 can persist up to 330 K at zero magnetic field, demonstrating superior thermal stability compared to other known skyrmion vdW magnets. This work provides valuable insights into skyrmionics and presents promising prospects for 2D material-based skyrmion devices operating beyond room temperature.

BackgroundThe ongoing global health crisis of COVID-19, and particularly the challenges posed by recurrent infections of the Omicron variant, have significantly strained healthcare systems worldwide. There is a growing body of evidence indicating an increased susceptibility to Omicron infection in patients suffering from Acute Kidney Injury (AKI). However, the intricate molecular interplay between AKI and Omicron variant of COVID-19 remains largely enigmatic.MethodsThis study employed a comprehensive analysis of human RNA sequencing (RNA-seq) and microarray datasets to identify differentially expressed genes (DEGs) associated with Omicron infection in the context of AKI. We engaged in functional enrichment assessments, an examination of Protein-Protein Interaction (PPI) networks, and advanced network analysis to elucidate the cellular signaling pathways involved, identify critical hub genes, and determine the relevant controlling transcription factors and microRNAs. Additionally, we explored protein-drug interactions to highlight potential pharmacological interventions.ResultsOur investigation revealed significant DEGs and cellular signaling pathways implicated in both Omicron infection and AKI. We identified pivotal hub genes, including EIF2AK2, PLSCR1, GBP1, TNFSF10, C1QB, and BST2, and their associated regulatory transcription factors and microRNAs. Notably, in the murine AKI model, there was a marked reduction in EIF2AK2 expression, in contrast to significant elevations in PLSCR1, C1QB, and BST2. EIF2AK2 exhibited an inverse relationship with the primary AKI mediator, Kim-1, whereas PLSCR1 and C1QB demonstrated strong positive correlations with it. Moreover, we identified potential therapeutic agents such as Suloctidil, Apocarotenal, 3′-Azido-3′-deoxythymidine, among others. Our findings also highlighted a correlation between the identified hub genes and diseases like myocardial ischemia, schizophrenia, and liver cirrhosis. To further validate the credibility of our data, we employed an independent validation dataset to verify the hub genes. Notably, the expression patterns of PLSCR1, GBP1, BST2, and C1QB were consistent with our research findings, reaffirming the reliability of our results.ConclusionOur bioinformatics analysis has provided initial insights into the shared genetic landscape between Omicron COVID-19 infections and AKI, identifying potential therapeutic targets and drugs. This preliminary investigation lays the foundation for further research, with the hope of contributing to the development of innovative treatment strategies for these complex medical conditions.

A high atmospheric vapour pressure deficit (VPD) hinders calcium absorption in tomatoes (Solanum lycopersicum L.), which severely reduces tomato yield, crop water productivity (WPcrop), and quality. Although reducing the VPD is effective for increasing tomato yield and water productivity, regulating the VPD is costly. Therefore, this study aimed to explore the effects of VPD regulation (high VPD=2.22 kPa, low VPD=0.95 kPa) during different growth stages on tomato WPcrop, yield, and quality to identify a VPD management method that saves costs and enhances the above parameters. The results showed that the yield was significantly positively correlated with fruit calcium absorption. Decreasing the VPD during the fruit expansion stage and increasing it during the flowering stage significantly increased calcium accumulation in the fruits, thereby improving tomato WPcrop, yield, and quality. Additionally, a high VPD during the flowering stage increased stomatal conductance and mesophyll conductance, thereby increasing intercellular and chloroplast CO2 concentrations and enhancing plant photosynthetic capacity, and reduced the stem water potential, leaf water potential, and leaf relative water content, thereby increasing the water potential difference, leaf hydraulic conductance, and calcium absorption. By integrating game theory and the technique for order of preference by similarity to ideal solution method, a comprehensive analysis of tomato appearance, nutrients, and flavour quality revealed that growing plants under a high VPD during the seedling, flowering, and fruit ripening stages and a low VPD during the fruit expansion stage not only enhanced the comprehensive tomato quality value but also maximally reduced the humidification cost. Conclusively, controlling the environmental VPD at 2.22 kPa during the seedling, flowering, and fruit ripening stages and at 0.95 kPa during the fruit expansion stage can effectively increase tomato WPcrop, yield, and quality. This study provides a theoretical basis for the environmental regulation of high-quality, efficient, and water-conserving cultivation of greenhouse tomatoes.

Abstract Aims Diabetic cardiomyopathy (DCM) is a major cause of mortality in patients with diabetes, and the potential strategies for treating DCM are insufficient. Melatonin (Mel) has been shown to attenuate DCM, however, the underlying mechanism remains unclear. The role of vascular endothelial growth factor-B (VEGF-B) in DCM is little known. In present study, we aimed to investigate whether Mel alleviated DCM via regulation of VEGF-B and explored its underlying mechanisms. Methods and results We found that Mel significantly alleviated cardiac dysfunction and improved autophagy of cardiomyocytes in type 1 diabetes mellitus (T1DM) induced cardiomyopathy mice. VEGF-B was highly expressed in DCM mice in comparison with normal mice, and its expression was markedly reduced after Mel treatment. Mel treatment diminished the interaction of VEGF-B and Glucose-regulated protein 78 (GRP78) and reduced the interaction of GRP78 and protein kinase RNA -like ER kinase (PERK). Furthermore, Mel increased phosphorylation of PERK and eIF2α, then up-regulated the expression of ATF4. VEGF-B−/− mice imitated the effect of Mel on wild type diabetic mice. Interestingly, injection with Recombinant adeno-associated virus serotype 9 (AAV9)-VEGF-B or administration of GSK2656157 (GSK), an inhibitor of phosphorylated PERK abolished the protective effect of Mel on DCM. Furthermore, rapamycin, an autophagy agonist displayed similar effect with Mel treatment; while 3-Methyladenine (3-MA), an autophagy inhibitor neutralized the effect of Mel on high glucose-treated neonatal rat ventricular myocytes. Conclusions These results demonstrated that Mel attenuated DCM via increasing autophagy of cardiomyocytes, and this cardio-protective effect of Mel was dependent on VEGF-B/GRP78/PERK signaling pathway.

In the context of climate change, water availability has emerged as a pressing concern in agriculture, indicating the urgency to enhance irrigation management. Nevertheless, accurately quantifying irrigation schedules in diverse climates and soil textures remains a complex challenge. Here, a three-year irrigation experiment (2020–2022) of greenhouse tomato was conducted at three typical locations (Yulin, Yangling and Hanzhong) in northwest China. The irrigation schedules were determined by the cumulative evaporation (CE) measured from a 20 cm pan. A total nine treatments were set by combining three irrigation frequencies (IF) with three single doses (ID). Twenty indices related to tomato growth, photosynthesis, yield and water use, fruit quality, and environmental benefit were evaluated comprehensively by using TOPSIS (technique for order preference by similarity to ideal solution) and GRA (grey relational analysis) methods of combining weights based on game theory. The results showed that the indicator with highest weight was yield, followed by crop water productivity and vitamin C. The optimal ranges of irrigation frequency (irrigation dose) were determined as follows: 10.00–11.08 mm (0.81–0.90 CE) in Yulin, 13.12–15.68 mm (0.77–0.89 CE) in Yangling, and 11.44–16.40 mm (0.67–0.77 CE) in Hanzhong, respectively. This study introduces a method applied to prioritize the key indicators for irrigation scheduling, providing a reference for optimizing irrigation practices in greenhouse tomato production and improving water management in agriculture.

ObjectiveThe objective was to analyze the impact of the uterine artery pulsatility index (PI) on pregnancy outcomes by measuring uterine artery blood flow on the day of endometrial transformation in patients undergoing frozen–thawed embryo transfer (FET).MethodsThis was a case-control study. In total, 2,036 patients who underwent FET at the Third Affiliated Hospital of Zhengzhou University from October 2019 to September 2020 were included. The patients were divided into a clinical pregnancy group and a nonclinical pregnancy group according to pregnancy outcome. A multivariate logistic regression model was used to analyze the factors affecting the clinical pregnancy rate. The receiver operating characteristic (ROC) curve was used to determine the optimal mean PI cutoff value of 1.75. After 1:1 propensity score matching (PSM), 562 patients were included. For statistical description and analysis, the patients were divided into two groups: a group with a mean PI > 1.75 and a group with a mean PI ≤ 1.75.ResultsThe clinical pregnancy group included 1,218 cycles, and the nonclinical pregnancy group included 818 cycles. There were significant differences in female age (P 0.05).ConclusionIn this study, we found that the uterine artery PI on the day of endometrial transformation in patients undergoing FET is not a good predictor of pregnancy outcomes.

The signaling environment, or niche, often governs the initial difference in behavior of an adult stem cell and a derivative that initiates a path towards differentiation. The transition between an instructive stem cell niche and differentiation niche must generally have single-cell resolution, suggesting that multiple mechanisms might be necessary to sharpen the transition. Here, we examined the Drosophila ovary and found that Cap cells, which are key constituents of the germline stem cell (GSC) niche, express a conserved microRNA (miR-124). Surprisingly, loss of miR-124 activity in Cap cells leads to a defect in differentiation of GSC derivatives. We present evidence that the direct functional target of miR-124 in Cap cells is the epidermal growth factor receptor (EGFR) and that failure to limit EGFR expression leads to the ectopic expression of a key anti-differentiation BMP signal in neighboring somatic escort cells (ECs), which constitute a differentiation niche. We further found that Notch signaling connects EFGR activity in Cap cells to BMP expression in ECs. We deduce that the stem cell niche communicates with the differentiation niche through a mechanism that begins with the selective expression of a specific microRNA and culminates in the suppression of the major anti-differentiation signal in neighboring cells, with the functionally important overall role of sharpening the spatial distinction between self-renewal and differentiation environments.

Morphological structure can greatly affect the magnetic domain of magnetic materials, which plays a significant role on regulating the magnetic characteristics and electromagnetic properties. Herein, Fe3O4 nanorings/nanotubes with adjustable aspect ratio were fabricated through a hydrothermal route followed by reduction annealing treatment. The magnetic flux distributions in radial and axial direction were characterized by electron holography, suggesting that the Fe3O4 nanorings/nanotubes show magnetic vortex structure. The magnetic vortex-domain structure is favored to break the Snoek’s limit of Fe3O4, improving the high-frequency permeability and boosting the natural resonance band to 1 ∼ 10 GHz. Meanwhile the complex permittivity is highly sensitive to the aspect ratio. The Fe3O4 nanorings/nanotubes perform excellent comprehensive microwave absorption properties, owing to the special magnetic vortex structure, tunable complex permittivity, big complex permeability, large magnetization and high natural resonance. The Fe3O4 nanorings with the smallest aspect ratio show a minimum reflection loss (RL) value of −47.9 dB at 2.6 mm and EAB of 4 GHz at 3 mm. Besides, rotational orientation by an external magnetic field can cause the parallel arrangement of nanorings/nanotubes, which can increase both the complex permittivity and broad the resonance band. Rotational orientation can realize effective absorption efficiency at a broader frequency range with a thinner thickness, further improving the comprehensive microwave absorption. These findings not only suggest that designing particular magnetic-domain structures by morphologies can break the Snoek’s limit of magnetic materials, but also demonstrate that aspect ratio and magnetic field-assisted rotational orientation is promising ways to regulate the electromagnetic parameters, resulting in high-performance microwave absorption.

Summary: Aiming at the current SPI (solder paste inspection) system for printing solder paste similar defects detection accuracy is not high, the system intelligence degree is low and so on, design a for the solder paste similar defects and combined with phase modulation profile measurement technique and improve the YOLOX intelligent detection system. The core of the system is the improved YOLOX depth model based on s-mosica and kt-iou algorithms proposed in this paper. The experimental results show that the proposed s-mosica and kt-iou algorithms can effectively improve the detection accuracy of printed solder paste, and when combined with the YOLOX model, the best 90.33% detection accuracy is obtained, which is better than the detection performance of the existing algorithms in the same scenario, and it provides an effective and feasible reference program for the design of the SPI high-precision intelligent detection system.

Hybrid electric vehicle (HEV) is one of the ideal transportation tools to face the challenge of ‘Carbon peak carbon neutralization’. The high complexity of the latest HEVs result to the difficulties in vehicle fault diagnostics, which is considered to be the main factors of vehicle durability. In this study, a multi-fault mode detection method of a P2 diesel HEV was investigated by using support vector machine(SVM). The HEV physical model was built and validated by using the experimental data under China typical urban driving cycle (CTUDC). The SVM algorithm was coded in Matlab/Simulink environment. The training data vectors for normal mode and fault mode were acquired from the HEV model. Through the analysis of failure mode, the OVO-SVM method was proved as the best accuracy, the success rate of diagnosis reaches 98%. Case studies of single fault mode and multi-fault mode fault detection were conducted. Offline and online level test were performed to show the effectiveness of the detection algorithm. The study may support the development of intelligent diagnostic methods for the different types of HEVs.

CFRP hybrid bonded–bolted (HBB) joints combine the advantages of traditional joining methods, namely adhesive bonding, and bolting, to achieve optimal connection performance, making them the most favored connection method. The structural parameters of CFRP HBB joints, including overlap length, bolt-hole spacing, and fit clearance relationships, have a complex impact on connection performance. To enhance the connectivity performance of joint structures, this paper develops a multiscale finite element analysis model to investigate the impact of structural parameters on the strength of CFRP HBB joint structures. Coupled with experimental validation, the study reveals how changes in structural parameters affect the unidirectional tensile failure force of the joints. Building on this, an analytical approach and inverse design methodology for the mechanical properties of CFRP HBB joints based on deep supervised learning algorithms are developed. Neural networks accurately and efficiently predict the performance of joints with unprecedented combinations of parameters, thus expediting the inverse design process. This research combines experimentation and multiscale finite element analysis to explore the unknown relationships between the mechanical properties of CFRP HBB joints and their structural parameters. Furthermore, leveraging DNN neural networks, a rapid calculation method for the mechanical properties of hybrid joints is proposed. The findings lay the groundwork for the broader application and more intricate design of composite materials and their connection structures.

Abstract Background Previous studies have reported that after laparoscopic cystectomy of ovarial endometrioma, the ovarian response to gonadotropin (Gn) significantly decreased. However, for patients with diminished ovarian reserve (DOR) after ovarian surgery, how to choose the most appropriate controlled ovarian hyperstimulation protocol has not been concluded. Compared with the traditional agonist regimen, the gonadotropin (Gn)-releasing hormone (GnRH) antagonist, microstimulation, and progestin-primed ovarian stimulation (PPOS) protocols are simple to operate and have a shorter cycle, which are often used in patients with DOR. So the purpose of our study is to compare the assisted reproductive outcomes of these three controlled ovarian hyperstimulation protocols in patients with DOR following laparoscopic cystectomy of ovarial endometrioma. Methods In this retrospective cohort study, 89 patients with DOR who had undergone in vitro fertilisation/intracytoplasmic sperm injection at the Department of Reproductive Medicine at the Third Affiliated Hospital of Zhengzhou University from 1 to 2018 to 31 December 2020 were included. According to the controlled ovarian hyperstimulation protocols employed, the patients were divided into GnRH antagonist (38 patients), PPOS (27 patients), and microstimulation (24 patients) groups. The basic data and clinical outcomes of the three groups were compared. The main outcome measure was the cumulative live birth rate. Results No significant differences in the age of the female patients and their spouses and female patients’ body mass index and basal endocrine levels (follicle-stimulating hormone and oestradiol) were noted among the three groups (P > 0.05). The GnRH antagonist group had higher antral follicle counts, greater endometrial thickness on the human chorionic Gn injection day, greater number of oocytes retrieved, and higher two pronuclear embryo counts than did the other two groups. However, the starting dosage of Gn was lower in the GnRH antagonist group than in the other two groups. The microstimulation group had a significantly higher oocyte output rate and high-quality embryo rate than did the other two groups (P 0.05). Conclusion In conclusion, for patients aged under 40 years who experienced DOR after laparoscopic cystectomy of ovarial endometrioma, GnRH antagonist protocol and PPOS protocol can obtain better ovulation induction outcomes and cumulative live birth rate than microstimulation protocol, and are more suitable ovulation induction protocols.

Aiming at the communication bottleneck problem when the current federated learning security aggregation algorithm was applied in a complex network environment, an adaptive federated learning secure aggregation scheme based on threshold homomorphic encryption was proposed.While protecting gradient privacy, users adaptively compress gradients based on the current available bandwidth, greatly reduced communication overhead for federated users.Furthermore, the new dynamic decryption task distribution algorithm and gradient combination algorithm were designed in the phase of aggregation gradient decryption, which relieved the user’s uplink communication pressure.The experimental results show that the proposed scheme can sharply reduce the amount of communication to 4% compared with the existing federated learning scheme with a trivial model accuracy loss of 1%.

Myocardial injury as one of the severe complications leads to the increasing morbidity and mortality in patients with sepsis. Recent studies reported that reactive oxygen species (ROS)-mediated ferroptosis plays a critical role in the development of heart diseases. Therefore, we hypothesized that anti-ferroptosis agent might be a novel potential therapeutic strategy for sepsis-induced cardiac injury. Herein, we demonstrated that a small biocompatible and MRI-visible melanin nanoparticles (MMPP) improves myocardial function by inhibiting ROS-related ferroptosis signaling pathway. In LPS-induced murine sepsis model, after a single dose intravenously injection of MMPP treatment, MMPP markedly alleviated the myocardial injury including cardiac function and heart structure disorder through suppressing iron-accumulation induced ferroptosis. In vitro, MMPP inhibited cardiomyocyte death by attenuating oxidative stress, inflammation and maintaining mitochondrial homeostasis. Collectively, our findings demonstrated that MMPP protected heart against sepsis-induced myocardial injury via inhibiting ferroptosis and inflammation, which might be a novel therapeutic approach in future.

Summary: MicroRNA (miRNA) plays physiologically and pathologically important roles in post-transcriptional regulation. Although miRNA has been suggested to dynamically interact with cellular organelles, the dynamicity of intracellular miRNA behavior has remained unclear. Here, by introducing fluorescently labeled pre-miRNA into living cells, we improved the miRNA visualization method using exogenous miRNA precursors. Through the combination of our miRNA visualization method and single-molecule sensitive fluorescence microscopy, we quantitatively analyzed the process of miRNA maturation. Furthermore, single-particle tracking of fluorescent miRNA in cells revealed the directed movements of miRNA on cytoskeletal components (i.e., microtubules and actin filaments). Our results also suggest that cytoskeleton-dependent miRNA trafficking is associated with the interaction of miRNAs with the nucleus and the endoplasmic reticulum/Golgi apparatus. Our method should facilitate the elucidation of the mechanism and physiological significance of the subcellular localization and organelle interaction of miRNA.

Low carbon operation of power systems is a key way to achieve the goal of energy power carbon peaking and carbon neutrality. In order to promote the low carbon transition of energy and power and the coordinated and optimized operation of distributed energy sources in virtual power plants (VPP), this paper proposes a framework for collaborative utilization of pumped storage–carbon capture–power-to-gas (P2G) technologies. It also constructs a multi-time scale low carbon economic dispatch model for VPP to minimize the internal resource operation cost of VPP in each time period. During the intraday scheduling stage, the day-ahead scheduling results as the planned output and the energy flow is then dynamically corrected at a short-term resolution in the framework. This allows for the exploration of the low-carbon potential of each aggregation unit within the virtual power plant. The results of the simulation indicate that the strategy and model proposed in this paper can effectively encourage the consumption of renewable energy sources, promote the low-carbon operation of power system power, and serve as a valuable reference for the low-carbon economic operation of the power system.

Parasitoids commonly manipulate their host’s metabolism and immunity to facilitate their offspring survival, but the mechanisms remain poorly understood. Here, we deconstructed the manipulation strategy of a newly discovered parasitoid wasp, L. myrica, which parasitizes D. melanogaster. Using RNA-seq, we analyzed transcriptomes of L. myrica-parasitized and non-parasitized Drosophila host larvae. A total of 22.29 Gb and 23.85 Gb of clean reads were obtained from the two samples, respectively, and differential expression analysis identified 445 DEGs. Of them, 304 genes were upregulated and 141 genes were downregulated in parasitized hosts compared with non-parasitized larvae. Based on the functional annotations in the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases, we found that the genes involved in host nutrition metabolism were significantly upregulated, particularly in carbohydrate, amino acid, and lipid metabolism. We also identified 30 other metabolism-related DEGs, including hexokinase, fatty acid synthase, and UDP-glycosyltransferase (Ugt) genes. We observed that five Bomanin genes (Boms) and six antimicrobial peptides (AMPs) were upregulated. Moreover, a qRT-PCR analysis of 12 randomly selected DEGs confirmed the reproducibility and accuracy of the RNA-seq data. Our results provide a comprehensive transcriptomic analysis of how L. myrica manipulates its host, laying a solid foundation for studies on the regulatory mechanisms employed by parasitoid wasps in their hosts.

To address the inherent challenges of deep space exploration, such as communication delays and the unpredictability of spacecraft environments, this study focuses on enhancing spacecraft adaptability and autonomy, which are essential for Autonomous Space Scientific Exploration. A pivotal aspect of this endeavor is the advancement of spacecraft task scheduling, which is integral to increasing spacecraft autonomy. Current research in this domain predominantly revolves around mission timing planning and is primarily executed from ground stations. However, these plans often lack the granularity required for direct implementation by spacecraft. In response, our study proposes an innovative approach to augment spacecraft autonomy, introducing a method that articulately describes mission objectives and resource information. We designed a novel hierarchical task network-timeline (HTN-T) algorithm, an amalgamation of the HTN scheduling method and the distinctive elements of existing research. This algorithm addresses time constraints through horizontal and vertical expansions, building upon the resolution of logical constraints found in conventional planning methods. Furthermore, it introduces a priority-based strategy for resolving resource conflicts in spacecraft tasks. This algorithm is substantiated through validation, including proof-of-principle demonstrations and assessments within a Space–ground Collaborative Management and Control System encompassing both ground and spacecraft operations. The findings indicate that our proposed algorithm achieves high rates of scheduling success and operational efficiency within a feasible timeframe, thus effectively navigating the complexities of autonomous spacecraft task scheduling.

This study adopted the method of “exchanging space for time” and set up three experimental groups based on the shape, degree of damage, and degree of humification of the litter, namely the undecomposed layer, the semi-decomposed layer, and the decomposed layer. Using typical slopes of arbor and bamboo forests in the Pi River Basin as the research object, from October 2021 to December 2022, the soil carbon release flux was measured by using a closed static chamber gas chromatography method to reveal the carbon release law at the soil–air interface during the decomposition process of litter and quantitatively characterize the dynamic impact of the litter decomposition process on soil carbon release flux. Results showed that soil methane flux remained negative (sink) while soil carbon dioxide flux was positive (source) in both litter-covered and bare soil conditions. The methane and carbon dioxide release from soil was positively correlated with and significantly influenced by environmental factors such as soil moisture content and temperature. The methane release flux from soil showed a linear fitting relationship with soil moisture content and temperature, while the carbon dioxide release flux from soil was more in line with the exponential fitting relationship with soil moisture content and temperature. However, there were significant differences in the roles of various factors under different types of litter.

BackgroundIn critically ill patients, transcutaneous oxygen saturation (SpO2) upon admission is typically associated with in-hospital mortality. Nevertheless, the available information for patients with non-traumatic subarachnoid hemorrhage (SAH) is limited. In our study, our objective was to assess the correlation between SpO2 levels and mortality among patients diagnosed with severe SAH.MethodsIn this study, we extracted data from the Medical Information Marketplace in Intensive Care (MIMIC-IV) database, which comprises information on critically ill patients. By employing matching ICD-9 and ICD-10 codes, we identified 3,328 patients diagnosed with SAH. Every individual who was admitted to the intensive care unit (ICU) had their SpO2 data and various covariates, including age, sex, diagnosis, and duration of stay, recorded upon admission. Subsequently, the patients were categorized into three distinct groups according to their SpO2 levels: low (≤95%), moderate (95–98%), and high (≥98%). To investigate the association between percutaneous oxygen saturation and mortality in patients with severe SAH, logistic regression, and cubic spline models were utilized. The main outcomes of interest were 28- and 90-day mortality rates. ObjectiveThe purpose of this study was to evaluate the cumulative live birth rate (CLBR) of mild stimulation and conventional stimulation for the low-prognosis population undergoing PPOS protocols.MethodsThis was a retrospective cohort study. We included women with a low prognosis. All women underwent PPOS protocols, and the starting gonadotropin (Gn) dose was 150 IU or 300 IU. The primary outcome measure was CLBR. The secondary outcome measures were the number of oocytes retrieved, number of 2PN oocytes and number of available embryos.ResultsIn total, 171 women with mild stimulation and 1810 women with conventional stimulation met the criteria. ObjectiveThe study aimed to evaluate the relationship between serum sodium and mortality in critically ill patients with non-traumatic subarachnoid hemorrhage.MethodsThis is a retrospective investigation of critically ill non-traumatic patients with subarachnoid hemorrhage (SAH) utilizing the MIMIC-IV database. We collected the serum sodium levels at admission and determined the all-cause death rates for the ICU and hospital. We employed a multivariate Cox proportional hazard regression model and Kaplan–Meier survival curve analysis to ascertain the relationship between serum sodium and all-cause mortality. In order to evaluate the consistency of correlations, interaction and subgroup analyses were also conducted.ResultsA total of 864 patients with non-traumatic SAH were included in this study. All-cause mortality in the ICU and hospital was 32.6% (282/864) and 19.2% (166/864), respectively. Sodium levels at ICU admission showed a statistically significant J-shaped non-linear relationship with ICU and hospital mortality (non-linear P-value < 0.05, total P-value < 0.001) with an inflection point of ~141 mmol/L, suggesting that mortality was higher than normal serum sodium levels in hypernatremic patients. Multivariate analysis after adjusting for potential confounders showed that high serum sodium levels (≥145 mmol/L) were associated with an increased risk of all-cause mortality in the ICU and hospital compared with normal serum sodium levels (135–145 mmol/L), [hazard ratio (HR) = 1.47, 95% CI: 1.07–2.01, P = 0.017] and (HR = 2.26, 95% CI:1.54–3.32, P < 0.001). Similarly, Kaplan–Meier (K-M) survival curves showed lower survival in patients with high serum sodium levels. E3 ubiquitin ligases (E3s) play a pivotal role in regulating the specificity of protein ubiquitination, and their significant functions as regulators of immune responses against tumors are attracting considerable interest. RBCK1—an RBR E3 ligase—is involved in immune regulation and tumor development. However, the potential effect of RBCK1 on glioma remains enigmatic. In the present study, we performed comprehensive analyses of multilevel data, which disclosed distribution characteristics of RBCK1 in pan-cancer, especially in glioma. Functional roles of RBCK1 were further confirmed using immunohistochemistry, cell biological assays, and xenograft experiments. Aberrant ascending of RBCK1 in multiple types of cancer was found to remodel the immunosuppressive microenvironment of glioma by regulating immunomodulators, cancer immunity cycles, and immune cell infiltration. Notably, the MES-like/RBCK1High cell population, a unique subset of cells in the microenvironment, suppressed T cell-mediated cell killing in glioma. Elevated expression levels of RBCK1 suggested a glioma subtype characterized by immunosuppression and hypo-responsiveness to immunotherapy but manifesting surprisingly increased responses to anti-angiogenic therapy. In conclusion, anti-RBCK1 target therapy might be beneficial for glioma treatment. Moreover, RBCK1 assisted in predicting molecular subtypes of glioma and response rates of patients to different clinical treatments, which could guide personalized therapy.

Abstract A new trapezoid-shaped Frisch-grid ionization chamber (TFG-IC) has been built as a part of a $$\varDelta {E}-E$$ Δ E - E telescope system for the detection and identification of charged particles at energies down to a few MeV. To study the effect of the drift electric field uniformity, two types of sealed windows, namely a pair of SSA (split-strip aluminized mylar film) and a pair of DSA (double-sided aluminized mylar film) sealed windows have been investigated. The detector’s performances were studied using a standard $$^{241}$$ 241 Am source at different gas pressures, and the total energy-deposit resolution achieved is about 1.1%(FWHM). The $$\varDelta {E}-E$$ Δ E - E telescope, which was composed of TFG-IC and a DSSSD (double-sided silicon strip detector), has been tested using a three-component $$\alpha $$ α source and the $$^{241}$$ 241 Am source under laboratory conditions. The results show that the energy resolution with the SSA sealed windows which provide uniform drift electric field has a smaller fluctuation than that with the DSA ones; the fluctuations are about 1% and 4% for the former and the latter, respectively. Simulations using the COMSOL software also confirmed the electric-field distortion at the edge of the detector with the DSA windows. A correlation curve between energy resolution and energy deposit of charged particles at various gas pressures and for two gas species is derived for TFG-IC with the SSA sealed windows using the measurement with the $$^{241}$$ 241 Am source. Incorporating the above results, we performed Monte Carlo simulations to evaluate the particle-identification capability of the telescope. The results show that the telescope can be extended to the identification of low-energy particles.

Abstract Background Prunus mume is an early spring flower of Rosaceae, which owns high application value in gardens. Being an excellent ornamental trait, the double flower trait has always been one of the important breeding goals of plant breeders. However, the key regulatory genes of double flower traits of P. mume are still unclear at present. Results The floral organs’ morphological differences of 20 single and 20 double flower cultivars of P. mume were compared firstly. And it was found that double flower trait of P. mume were often accompanied by petaloid stamen, multiple carpels and an increase in the total number of floral organs. Then, transcriptome sequencing of two representative cultivars P. mume ‘Danban Lve’ and P. mume ‘Xiao Lve’ were conducted at 3 Stage of flower bud development with distinct morphological differentiation. 3256 differentially expression genes (DEGs) were detected, and 20 candidate genes for double flower trait of P. mume were screened out including hub genes PmAP1–1 and PmAG-2 based on DEGs function analysis and WGCNA analysis. And it was found that epigenetic and hormone related genes may also play an important role in the process of double flower. Conclusions This study suggested that the double flower trait of P.mume is more like accumulation origin based on morphological observation. 20 genes and co-expression network related to the formation of double flower P. mume were preliminarily screened through transcriptomics analysis. The results provided a reference for further understanding of the molecular mechanism of double flower trait in P. mume.

Abstract The prognosis of non-metastatic gallbladder adenocarcinoma (NM-GBA) patients is affected by the status of metastatic lymph nodes. The purpose of this study was to explore the prognostic value of the log odds of positive lymph nodes (LODDS) and develop a novel nomogram to predict the overall survival in NM-GBA patients. A total of 1035 patients confirmed to have NM-GBA were selected from the Surveillance, Epidemiology, and End Results (SEER) database and further divided into training and validation cohorts. The discrimination and calibration of the nomogram were evaluated using the concordance index (C-index), the area under the time-dependent receiver operating characteristic curve (time-dependent AUC), and calibration plots. The net benefits and clinical utility of the nomogram were quantified and compared with those of the 8th edition American Joint Committee on Cancer (AJCC) Tumor-Node-Metastasis (TNM) staging system using decision curve analysis (DCA), net reclassification index (NRI), and integrated discrimination improvement (IDI). The risk stratifications of the nomogram and the TNM-staging system were compared. LODDS showed the highest accuracy in predicting OS for NM-GBA. The C-index (0.730 for the training cohort and 0.746 for the validation cohort) and the time-dependent AUC (> 0.7) indicated the satisfactory discriminative ability of the nomogram. Electrical switching of topological antiferromagnetic states in Mn3Sn thin films has been a subject of active investigation. However, dependences of switching behaviors on the film thickness, external field, and crystal orientation remain to be fully explored, which motivate the present study. In this work, (112̄0)-orientated Mn3Sn thin films are fabricated on sapphire substrates, in which a large anomalous Hall effect over a wide temperature range (270–400 K) can be identified. The current-induced spin–orbit torques (SOTs) are utilized to electrically manipulate the topological antiferromagnetic states in Mn3Sn/Pt bilayers. The robust SOT switching can be realized in Mn3Sn films with thicknesses up to 100 nm and with in-plane fields up to 1200 mT. Furthermore, SOT switching behaviors that are independent of the choice of crystal orientations are clearly revealed. Our results could be useful for implementing Mn3Sn films for efficient and stable antiferromagnetic spintronics.

ObjectiveTo explore whether the duration of estrogen treatment before progesterone application affects neonatal and perinatal outcomes in artificial frozen embryo transfer (FET) cycles.MethodsThis was a retrospective cohort study. Patients who underwent FET via artificial cycles and delivered a singleton live birth between January 2015 and August 2019 were included in the analysis. According to the duration of estrogen treatment before progesterone application, we divided the cycles into four groups: ①≤12 days, ②13-15 days, ③16-19 days, and ④≥20 days. The ‘≤12 days group’ was considered the reference group. The main outcome measures were preterm birth (PTB), small-for-gestational age (SGA), low birth weight (LBW), macrosomia, large-for-gestational age (LGA), gestational diabetes mellitus (GDM), gestational hypertension, premature rupture and placenta previa.ResultsOverall, 2010 FET cycles with singleton live births were included for analysis. Cycles were allocated to four groups according to the duration of estrogen treatment before progesterone application: ①≤12 days (n=372), ②13-15 days (n=745), ③16-19 days (n=654), ④≥20 days (n=239). The neonatal outcomes, including PTB, SGA, LBW, macrosomia and LGA, were comparable among the groups (P=0.328, P=0.390, P=0.551, P=0.565, P=0.358). The rates of gestational hypertension, premature rupture and placenta previa (P=0.676, P=0.662, P=0.211) were similar among the groups. The rates of GDM among the four groups were 4.0% (15/372), 6.7% (50/745), 6.4% (42/654), and 11.3% (27/239), with statistical significance (P=0.006). After multiple logistic regression analysis, the duration of estrogen treatment did not affect the rate of GDM or other outcomes.ConclusionThe estrogen treatment duration before progesterone application does not affect neonatal and perinatal outcomes in single frozen blastocyst transfer cycles.

The basic helix-loop-helix (bHLH) transcription factor family is the second-largest family in plants, where it plays essential roles in development, and the responses to multiple abiotic and biotic stressors. However, little information is available about this gene family in Prunus mume, which is widely cultivated in East Asia as an ornamental fruit tree. Here, 100 PmbHLH genes were identified, and their evolution and functions were explored in P. mume for the first time. The PmbHLH genes were classified into 21 subfamilies. The chromosomal distribution, physicochemical properties, bHLH domain, conserved motif, and intron/exon compositions were also analyzed. Furthermore, the evolutionary pattern, divergence time of the PmbHLH family, and genetic relationships among P. mume, Arabidopsis thaliana, and Prunus persica and Fragaria vesca of Rosaceae were explored. The functional prediction analysis of these PmbHLHs indicated that their functions varied, and included participating in the formation of organs and tissues, responding to stress, and the biosynthesis and metabolism of hormones and other secondary metabolites. Interestingly, expression analyses of PmbHLHs also revealed diverse expression patterns. Most of the PmbHLH genes were highly expressed in roots and stems, and a few were highly expressed in leaves, buds, and fruits, indicating tissue expression specificity. Eight PmbHLH genes, which were upregulated during low-temperature stress, may have critical roles in the response to cold stress. Ten PmbHLHs were differentially expressed between weeping and upright branches in a P. mume F1 population. These results shed light on the structure and evolution of the PmbHLH gene family, and lay a foundation for further functional studies of the bHLH genes.

Hepatocellular carcinoma (HCC) is often diagnosed at an unresectable stage without opportunities for curative therapy. Future liver remnant (FLR) insufficiency limits the range of patients who can undergo radical resection. Associating liver partition and portal vein ligation for staged hepatectomy (ALPPS) can ultimately achieve short-term hypertrophy of the FLR in patients with viral hepatitis-related fibrosis/cirrhosis and R0 resection. However, the influence of immune checkpoint inhibitors (ICIs) on liver regeneration remains unknown. We report two patients diagnosed with Barcelona Clinic Liver Cancer (BCLC)-B stage hepatitis B virus (HBV)-related HCC who underwent pioneering ALPPS after immunotherapy without posthepatectomy liver failure (PHLF). ALPPS has been shown to be safe and feasible in patients with HCC who underwent immunotherapy previously for the first time and might provide an alternative salvage option for future conversion therapy of HCC.

Abstract Magnetic skyrmions are topologically protected noncollinear spin textures, which are regarded as promising information carriers for next‐generation spintronic devices due to their small size and the low current density needed to drive their motion. Stability of skyrmions in zero external magnetic field is important for promoting fundamental studies and device applications. A few zero‐field skyrmion‐hosting materials have been developed, but none of them have been successfully integrated into a magnetic tunnel junction (MTJ), a crucial device for converting skyrmion information into an electrical signal. Here, a zero‐field exchange‐biased skyrmion material is developed and incorporated into an MTJ device. An Ir layer is inserted between the antiferromagnetic and ferromagnetic layers, which plays a crucial role in prohibiting interlayer diffusion under thermal annealing, resulting in simultaneous enhancement of exchange bias and thermal stability. The smallest zero‐field skyrmions have a size of 100 nm at room temperature. The zero‐field skyrmion material is then integrated into a perpendicularly magnetized MTJ, leading to the first demonstration of zero‐field skyrmions in an MTJ, which is an important step toward developing skyrmion‐based spintronic devices.

Summary: Vitellogenin receptor (VgR) is essential to vitellogenin uptaking and dominates ovary maturation in insects. However, the function of VgR in parasitoid wasps is largely unknown. Here, we applied the Drosophila parasitoid Leptopilina boulardi as a study model to investigate the function of VgR in parasitoids. Despite the conserved sequence characteristics with other insect VgRs, we found L. boulardi VgR (LbVgR) gene was highly expressed in head but lower in ovary. In addition, we found that LbVgR had no effects on ovary development, but participated in host-searching behavior of female L. boulardi and mating behavior of male L. boulardi. Comparative transcriptome analysis further revealed LbVgR might play crucial roles in regulating the expression of some important chemoreception genes to adjust the parasitoid behaviors. These results will broaden our knowledge of the function of VgR in insects, and contribute to develop advanced pest management strategies using parasitoids as biocontrol agents.

Abstract Background Drought stress is the most limiting factor for plant growth and crop production worldwide. As a major cereal crop, wheat is susceptible to drought. Thus, discovering and utilizing drought-tolerant gene resources from related species are highly important for improving wheat drought resistance. In this study, the drought tolerance of wheat Zhongmai 8601-Thinopyrum intermedium 7XL/7DS translocation line YW642 was estimated under drought stress, and then two-dimensional difference gel electrophoresis (2D-DIGE) based proteome analysis of the developing grains was performed to uncover the drought-resistant proteins. Results The results showed that 7XL/7DS translocation possessed a better drought-tolerance compared to Zhongmai 8601. 2D-DIGE identified 146 differential accumulation protein (DAP) spots corresponding to 113 unique proteins during five grain developmental stages of YW642 under drought stress. Among them, 55 DAP spots corresponding to 48 unique proteins displayed an upregulated expression, which were mainly involved in stress/defense, energy metabolism, starch metabolism, protein metabolism/folding and transport. The cis-acting element analysis revealed that abundant stress-related elements were present in the promoter regions of the drought-responsive protein genes, which could play important roles in drought defense. RNA-seq and RT-qPCR analyses revealed that some regulated DAP genes also showed a high expression level in response to drought stress. Conclusions Our results indicated that Wheat-Th. intermedium 7XL/7DS translocation line carried abundant drought-resistant proteins that had potential application values for wheat drought tolerance improvement.

The content and composition of wheat storage proteins are the major determinants of dough rheological properties and breadmaking quality and are influenced by cultivation conditions. This study aimed to investigate the effects of water deficit and high N-fertilizer application on wheat storage protein synthesis, gluten secondary structure, and breadmaking quality. Reverse-phase ultrahigh-performance liquid chromatography analysis showed that storage protein and gluten macropolymer accumulation was promoted under both independent applications and a combination of water-deficit and high N-fertilizer treatments. Fourier-transform infrared spectroscopy showed that water deficit and high N-fertilizer treatments generally improved protein secondary structure formation and lipid accumulation, and reduced flour moisture. In particular, high N-fertilizer application increased β-sheet content by 10.4% and the combination of water-deficit and high N-fertilizer treatments increased random coil content by 7.6%. These changes in gluten content and secondary structure led to improved dough rheological properties and breadmaking quality, including superior loaf internal structure, volume, and score. Our results demonstrate that moderately high N-fertilizer application under drought conditions can improve gluten accumulation, gluten secondary structure formation, and baking quality.

Underwater wireless optical communication (UWOC) is considered as an enabling technology with a mass of potential applications. The silicon photomultiplier (SiPM) exhibits a bright prospect for UWOC thanks to the traits of low-light detection capability, low-voltage operation, and superior operability. However, the performance of the SiPM-based UWOC system is severely degraded by the dead-time caused nonlinear response. In this paper, to mitigate the dead-time induced nonlinear distortion and explore the achievable capacity of the newly developed SiPM, we propose and experimentally demonstrate a 55-m/2-Gbps UWOC system by virtue of SiPM diversity reception and nonlinear decision-feedback equalizer (NDFE). The performance of NDFE is superior to that of the conventional decision-feedback equalizer (DFE), and NDFE with a pruning factor of 5 declares similar performance as that without pruning strategy, while the number of the nonlinear equalizer can be reduced by ~31.8%. Significant performance improvement is also obtained by the proposed scheme under different turbidity waters. The measured data rate is pushed from 1 Gbps to 2 Gbps with a receiver sensitivity as low as −41.96 dBm, which to the best of our knowledge is the largest data rate ever achieved using the off-the-shelf SiPM among the reported UWOC works. In accordance with the receiver sensitivity and the model of optical propagation in the water channel, the maximum attainable distance/data rate is predicted to be 147 m/1 Gbps and 128 m/2 Gbps with the proposed scheme. The research results are promising for long-reach and high-speed UWOC.

Relationship between urban diversity and urban vitality is imperative for guiding better design in urban development, although existing frameworks are not able to efficiently examine the relationship at multiple scales. In this article, we propose a new framework to integrate nighttime light (NTL) imagery and multisource urban data into multiscale geographically weighted regression (MGWR) models to examine the varying relationship between diversity and vitality across space and time. NTL is used as a proxy for urban nighttime vitality. Public transport, taxi transit, and points of interest data are used to derive three aspects of urban diversity indices: ridership diversity, spatial interaction diversity, and built environment diversity. By comparing the models in holiday and nonholiday weeks in Shenzhen, China, the NTL-based vitality proxy was found to be strongly correlated with the urban diversity indices, given by the satisfactory goodness of fit (r-squared = 0.9) of the MGWR models. The spatially varying relationships between diversity indices and nighttime vitality were observed and patterns discussed. The analysis of the coefficients revealed the importance of stable public transport and fluctuating taxi trips for nighttime vitality. The new index proposed for the diversity of spatial interaction (DSI) is a strong indicator for nighttime vitality, adding to existing vitality indicators. Furthermore, this study found that DSI and density of catering have less temporal variation, indicating their robustness in measuring nighttime vitality. This study provided empirical insights into how nighttime vitality is related to urban diversity, demonstrating new applications of NTL for intracity studies.

Abstract Magnetic skyrmions are promising information carriers for dense and energy‐efficient information storage owing to their small size, low driving‐current density, and topological stability. Electrical detection of skyrmions is a crucial requirement to drive skyrmion devices toward applications. The use of a magnetic tunnel junction (MTJ) is commonly suggested for this purpose as MTJs are key spintronic devices for large‐scale commercialization that can convert magnetic textures into electrical signals. To date, however, it has been challenging to realize skyrmions in MTJs due to incompatibility between standard skyrmion materials and highly efficient MTJ electrodes. Here, a material stack combining magnetic multilayers, which host 100 nm scale skyrmions, with a perpendicularly magnetized MTJ, is reported. The devices are designed so that the skyrmions in the multilayer are imprinted into the MTJ's free layer via magnetostatic interactions. The electrical response of a single skyrmion is successfully identified by employing simultaneous imaging of the magnetic texture and the electrical measurement of the MTJ resistance. The results are an important step toward all‐electrical detection of skyrmions.

Abstract A new Frisch-grid ionization chamber has been built to explore the appropriate choice of Frisch-grid. Detailed studies of the relationship between grid geometries and detector performance have been performed with an $$^{241}$$ 241 Am source. This paper describes and compares the energy resolution of ionization chambers with parallel-wire and mesh grids of different grid parameters. Some specific recommendations for grid selection are provided based on the data currently available. To obtain optimal energy resolution, the operating voltage of the chamber must satisfy the condition of minimum electron collection on the grid with distinct geometries and parameters, respectively. Since there is no established theory applicable to both types of grids, we have devised a careful simulation procedure incorporating the COMSOL and Garfield++ codes to search for the conditions of the minimum electron collection on the grid. The simulation results fit the experimental data well, suggesting that this simulation method successfully predicts the suitable voltage setting when using a mesh grid or parallel wires grid as the shielding electrode.

ObjectiveTo investigate the clinical outcomes of Day 7 (D7) frozen-thawed embryo transfer (FET) and to provide a reference value for clinical work.MethodsThis was a retrospective cohort study. Patients undergoing FET cycles in the Reproductive Medicine Center of the Third Affiliated Hospital of Zhengzhou University between December 2015 and January 2021 were included. According to the developmental stage of the embryos at transfer, the embryos were divided into three groups: Day (D) 5, D6 and D7 blastocysts. Group D7 was compared with Groups D5 and D6. Simultaneously, the preimplantation genetic testing (PGT) and non-PGT cycles in Group D7 were analyzed and compared. The main outcomes were the clinical pregnancy, live birth and miscarriage rates. The secondary outcomes were the implantation and euploidy rates.ResultsIn total, 5945, 4094 and 137 FET cycles were included in the D5, D6 and D7 groups, respectively. Rail Conveyor is a highly energy-efficient and cost-effective bulk material transportation system, which merges the benefits of both belt conveyor technology and railway. Reducing vibration and noise is a key problem in the design of rail conveyor system, which has great social and economic significance for the realization of green environmental protection and sustainable development of bulk materials transportation. This article describes the field experimental test of vibration and noise. The vibration acceleration of rail conveyor was tested under different belt speed and different carriage spacing and different wheel materials of support carriage, and the noise of rail conveyor was tested under different belt speed and different test distance. Experimental results and analysis show that: With the increase of belt speed, the vibration and noise of wheel and rail of rail conveyor increase greatly, and the wheel rail vibration and noise decrease with the increase of the distance between the support carriages, and the wheel material has a great influence on the vibration.

IntroductionParathyroid carcinoma (PC) is a very rare endocrine malignancy occurring in less than 1% of all cases of primary hyperparathyroidism (pHPT). The liver is the second most common target organ for distant metastases of PC, but no guidelines are available for the diagnosis and treatment of liver metastases. In this study, we attempted to summarize the characteristics of the diagnosis and treatment of liver metastases based on our patients and other cases reported in the literature.Materials and methodsThe files of all patients diagnosed with PC with liver metastases summarized at our center between 2000 and 2022 were reviewed, and three datasets from Medline, Web of Science, and Embase were systematically searched to identify relevant articles.ResultsThree patients with liver metastases from our center and 11 patients from the literature were included in the study. All patients had pHPT with borderline remission of hypercalcemia after each operation. A total of 71.4% of the patients’ liver lesions were found by abdominal CT scans, while 35.7% were found by MRI, PET-CT, and fine-needle aspiration biopsy (FNAB), which were also helpful for diagnosis. Eight of nine patients (88.9%) who underwent surgery, radiofrequency ablation (RFA), or transcatheter arterial embolization (TAE) were alive, and only one postoperative patient died after a follow-up of 60 months.ConclusionsPC is a rare malignant tumor prone to recurrence and metastasis, and postoperative reviews should be carried out routinely. Abnormally elevated parathyroid hormone (PTH) and serum calcium can indicate recurrence or metastasis. Enhanced CT and MRI can provide valuable support for the diagnosis of liver metastases, but whether [18F]FDG-PET-CT, [18F]FCH-PET-CT, or [11C]choline-PET-CT can be used as a diagnostic basis requires further study. Resection of liver metastases, segmental hepatectomy, or RFA can significantly improve patients’ symptoms.

As a novel type of non-coding RNAs, covalently closed circular RNAs (circRNAs) are ubiquitously expressed in eukaryotes. Emerging studies have indicated that dysregulation of circRNAs was related to neurological diseases. However, the biogenesis, regulation, function, and mechanism of circRNAs in Parkinson's disease (PD) remain largely unclear. In this study, thirty-three differentially expressed circRNAs (DECs) were detected by RNA-sequencing between the MPTP-induced PD mice model and the wild-type mice. Quantitative real-time PCR was used to determine the RNA level of DECs in the striatum (STR), substantia nigra pars compacta (SNpc), and serum exosomes, and it was found that circSV2b was downregulated in PD mice. Then, functional experiments in vivo were employed to explore the effect of circSV2b in PD. For the mechanism study, dual-luciferase reporter, fluorescence in situ hybridization (FISH), RNA immunoprecipitation (RIP), RNA pull-down, gene editing, and CUT & Tag were performed in vitro to confirm that circSV2b directly sponged miR-5107-5p and alleviated the suppression of the expression of the target gene Foxk1, and then positively regulated Akt1 transcription. In vivo, the mechanistic analysis demonstrated that circSV2b overexpression resisted oxidative stress damage through the ceRNA-Akt1 axis in PD models. Taken together, these findings suggested that the miR-5107-5p-Foxk1-Akt1 axis might serve as a key target of circSV2b overexpression in PD treatment, and highlighted the significant change of circSV2b in serum exosomes. Therefore, circSV2b might be a novel biomarker for the diagnosis and treatment of PD.
