ObjectiveTo analyze the whole-genome characteristics of four coxsackievirus B5 (CVB5) strains isolated in Yunnan province, China in 2022. MethodsPrimers were designed to amplify and sequence the whole genome of CVB5 isolates. The whole genome was amplified in segments, and sequenced using the "primer walking" method. Sequence splicing and assembly were performed. Genetic characteristics of the CVB5 strains were analyzed using software such as MEGA 7.0, Simplot 3.5.1, and DNAStar 7.1. ResultsBased on VP1 and whole-genome phylogenetic analysis, all four CVB5 isolates belonged to genotype C. Compared with the prototype strain, multiple amino acid mutations occurred in the VP1 region of the four strains. Phylogenetic analysis of P1, P2, and P3 regions and Simplot recombination analysis showed that the CVB5 isolates in this study may have undergone recombination with multiple enterovirus B strains. ConclusionThe four CVB5 strains isolated from clinical samples of hand, foot and mouth disease (HFMD) patients in Kunming, Yunnan province in 2022 all belonged to genotype C.

Abstract MoS2 as a typical layered transition metal dichalcogenide (LTMD) has attracted considerable attention to work as sodium host materials for sodium‐ion batteries (SIBs). However, it suffers from low semiconducting behavior and high Na+ diffusion barriers. Herein, intercalation of N‐doped amorphous carbon (NAC) into each interlayer of the tiny MoS2 nanosheets embedded on rGO conductive network is achieved, resulting in formation of rGO@MoS2/NAC hierarchy with interoverlapped MoS2/NAC superlattices for high‐performance SIBs. Attributed to intercalation of NAC, the resulting MoS2/NAC superlattices with wide MoS2 interlayer of 1.02 nm facilitates rapid Na+ insertion/extraction and accelerates reaction kinetics. Theoretical calculations uncover that the MoS2/NAC superlattices are beneficial for enhanced electron transport, decreased Na+ diffusion barrier and improved Na+ adsorption energy. The rGO@MoS2/NAC anode presents significantly improved high‐rate capabilities of 228, 207, and 166 mAh g−1 at 20, 30, and 50 A g−1, respectively, compared with two control samples of pristine MoS2 and MoS2/NAC counterparts. Excellent long‐term cyclability over 10 000 cycles with extremely low capacity decay is demonstrated at high current densities of 20 and 50 A g−1. Sodium‐ion full cells based on the rGO@MoS2/NAC anode are also demonstrated, yielding decent cycling stability of 200 cycles at 5C. Our work provides a novel interlayer strategy to regulate electron/Na+ transport for fast‐charging SIBs.

Abstract Background In recent years, natural bone extracellular matrix (ECM)-inspired materials have found widespread application as scaffolds for bone tissue engineering. However, the challenge of creating scaffolds that mimic natural bone ECM’s mechanical strength and hierarchical nano-micro-macro structures remains. The purposes of this study were to introduce an innovative bone ECM-inspired scaffold that integrates a 3D-printed framework with hydroxyapatite (HAp) mineralized graphene oxide-collagen (GO-Col) microscaffolds and find its application in the repair of mandibular bone defects. Methods Initially, a 3D-printed polycaprolactone (PCL) scaffold was designed with cubic disks and square pores to mimic the macrostructure of bone ECM. Subsequently, we developed multi-layer mineralized GO-Col-HAp microscaffolds (MLM GCH) to simulate natural bone ECM's nano- and microstructural features. Systematic in vitro and in vivo experiments were introduced to evaluate the ECM-inspired structure of the scaffold and to explore its effect on cell proliferation and its ability to repair rat bone defects. Results The resultant MLM GCH/PCL composite scaffolds exhibited robust mechanical strength and ample assembly space. Moreover, the ECM-inspired MLM GCH microscaffolds displayed favorable attributes such as water absorption and retention and demonstrated promising cell adsorption, proliferation, and osteogenic differentiation in vitro. The MLM GCH/PCL composite scaffolds exhibited successful bone regeneration within mandibular bone defects in vivo. Conclusions This study presents a well-conceived strategy for fabricating ECM-inspired scaffolds by integrating 3D-printed PCL frameworks with multilayer mineralized porous microscaffolds, enhancing cell proliferation, osteogenic differentiation, and bone regeneration. Loquat (Eriobotrya japonica) is a subtropical evergreen fruit tree with high economic benefits and nutritional value. AP2/ERF (APETALA2/ethylene responsive factor) are a wide range of transcription factors found in plants, which play an important role in regulating plant development and biological processes. However, the systematic identification and characterization of AP2/ERF genes in loquat remains unclear. In the present research, a total of 189 loquat AP2/ERF members were identified. According to the phylogenetic tree, these members were divided into three subfamilies, including AP2 (42), ERF (96), and DREB (51). Except for three genes located on the skeleton, most of the loquat AP2/ERF genes were unevenly distributed across all chromosomes. The collinearity results showed that 175 duplicate gene pairs were found in the AP2/ERF gene family of loquat. Ka/Ks results indicated that these genes underwent purification selection pressure. The syntenic analysis of AP2/ERF genes between Arabidopsis, pomegranate, peach, and loquat provided valuable clues for the potential evolution of AP2/ERF family in loquat. Cis-element analysis showed that most of the AP2/ERF genes in loquat had multiple cis-elements related to stress response, plant hormone signal transduction, and plant growth and development. Transcriptomic data indicated that the expression patterns of AP2/ERF genes exhibited diversity in different loquat cultivars during fruit development and ripening. The results of this study displayed the first comprehensive analysis of AP2/ERF genes in loquat, which provides a solid foundation for future research on the function of AP2/ERF genes in loquat.

BackgroundEGFR kinase domain duplication (EGFR-KDD) is an infrequent oncogenic driver mutation in lung adenocarcinoma. It may be a potential target benefit from EGFR-tyrosine kinase inhibitors (TKIs) treatment.Case presentationA 66-year-old Chinese male was diagnosed with lung adenocarcinoma in stage IVb with brain metastases. Next-generation sequencing revealed EGFR-KDD mutation. The patient received furmonertinib 160mg daily for anti-cancer treatment and obtained therapeutic efficacy with partial response (PR). Progression-free survival (PFS) duration from monotherapy was 16 months. With slow progressions, combined radiotherapy and anti-vascular targeted therapy also brought a continuous decrease in the tumors. The patient has an overall survival (OS) duration of more than 22 months and still benefits from double-dose furmonertinib.ConclusionsThis report provided direct evidence for the treatment of EGFR-KDD to use furmonertinib. A Large-scale study is needed to confirm this preliminary finding.

Polystyrene microplastics (PS-MPs) are new types of environmental pollutant that have garnered significant attention in recent years since they were found to cause damage to the human respiratory system when they are inhaled. The pulmonary fibrosis is one of the serious consequences of PS-MPs inhalation. However, the impact and underlying mechanisms of PS-MPs on pulmonary fibrosis are not clear. In this study, we studied the potential lung toxicity and PS-MPs-developed pulmonary fibrosis by long-term intranasal inhalation of PS-MPs. The results showed that after exposing to the PS-MPs, the lungs of model mouse had different levels of damage and fibrosis. Meanwhile, exposing to the PS-MPs resulted in a markedly decrease in glutathione (GSH), an increase in malondialdehyde (MDA), and iron overload in the lung tissue of mice and alveolar epithelial cells (AECs). These findings suggested the occurrence of PS-MP-induced ferroptosis. Inhibitor of ferroptosis (Fer-1) had alleviated the PS-MPs-induced ferroptosis. Mechanically, PS-MPs triggered cell ferroptosis and promoted the development of pulmonary fibrosis via activating the cGAS/STING signaling pathway. Inhibition of cGAS/STING with G150/H151 attenuated pulmonary fibrosis after PS-MPs exposure. Together, these data provided novel mechanistic insights of PS-MPs-induced pulmonary fibrosis and a potential therapeutic paradigm.

Layered transition metal dichalcogenides are promising candidates for sodium storage but suffering from low intrinsic electronic conductivity and limited interlayer spacing for fast electron/ion transport, which restricts their high-rate capability and cycling stability. In this work, rGO@MoSe2/NAC hierarchical architectures, consisting of conductive reduced graphene oxide (rGO) supported by hollow nanospheres that are rolled from superlattices of alternatively overlapped MoSe2 and N-doped amorphous carbon (NAC) monolayers, are synthesized as a high-performance sodium storage anode. Theoretical calculations reveal the intercalation of NAC monolayer between two adjacent MoSe2 monolayers improving electronic conductivity of MoSe2 in both surface and internal bulk to fully accelerate electron transport and enhance Na+ ​adsorption. The interoverlapped MoSe2/NAC superlattice featuring a wide interlayer expansion (72.3 ​%) of MoSe2 dramatically decreases Na+ ​diffusion barriers for fast insertion/extraction. Moreover, the hollow nanospheres and the rGO conductive network contribute to a robust hiberarchy that can well release internal stress and buffer the volume expansion, thereby enabling outstanding structural stability. Consequently, the rGO@MoSe2/NAC anode exhibits excellent high-rate capability of 194 mAh g−1 and ultralong cyclability of 12 ​000 cycles with a low capacity fading rate of 0.0038 ​% per cycle at an ultra-high current of 50 ​A ​g−1, delivering the best high-rate cycling performance to date. Remarkably, the Na3V2(PO4)3‖rGO@MoSe2/NAC full cells also present outstanding cycling stability (600 cycles) at 10C rate, which proves the great potential in fast-charging applications.

The stability of scaffolding structures is crucial for quality management in construction. Currently, scaffolding assembly quality monitoring relies on visual inspections performed by designated on-site personnel, which are highly subjective, inaccurate, and inefficient, hindering the advancement of intelligent construction practices. This study proposes an automated method for scaffolding assembly quality inspection using point cloud data and feature selection algorithms. High-precision point cloud data of the scaffolding are captured by a Trimble X7 3D laser scanner. After registration with the forward design model, a 2D slicing comparison method is developed to measure geometric dimensions with an accuracy controlled within 0.1 mm. The collected data are used to build an SVM model for automated assembly quality inspection. To combat the curse of dimensionality associated with high-dimensional data, an optimized genetic algorithm is employed for the dimensionality reduction in the raw sample data, effectively eliminating data redundancy and significantly enhancing convergence speed and classification accuracy of the detection model. Case studies indicate that the proposed method can reduce feature dimensionality by 70% while simultaneously improving classification accuracy by 13.9%. The proposed method enables high-precision automated inspection of scaffolding assembly quality. By identifying the optimal feature subset, the method differentiates the priority of various structural parameters during inspection, providing insights for optimizing the quality inspection process.

Abstract Background The incidence of rebleeding in patients with upper gastrointestinal bleeding (UGIB) remains despite advances in intervention approaches. Therefore, early prediction of the risk of rebleeding could help to greatly reduce the mortality rate in these patients. We aim to develop and validate a new prediction model to predict the probability of rebleeding in patients with AUGIB. Methods A total of 1170 AUGIB patients who completed the procedure of emergency gastroscopy within 48 h of admission were included. Logistic regression analyses were performed to construct a new prediction model. A receiver operating characteristic curve, a line graph, and a calibration and decision curve were used to assess the predictive performance of our new prediction model and compare its performance with that of the AIMS65 scoring system to determine the predictive value of our prediction model. Results A new prediction model was constructed based on Lactic acid (LAC), neutrophil percentage (NEUTP), platelet (PLT), albumin (ALB), and D-DIMER. The AUC values and their 95% confidence interval (CI) for the new prediction model and the AIMS65 score were 0.746 and 0.619, respectively, and 0.697–0.795 and 0.567–0.670, respectively. In the training group, the C index values based on the prediction model and the AIMS65 scoring system were 0.720 and 0.610, respectively. In the validation group, the C index values based on the prediction model and the AIMS65 scoring system were 0.828 and 0.667, respectively. The decision and calibration curve analysis also showed that the prediction model was superior to the AIMS65 scoring system in terms of accuracy of prediction, consistency, and net clinical benefit. Conclusion The prediction model can predict the probability of rebleeding in AUGIB patients after endoscopic hemostasis therapy.

Abstract Objectives To compare the efficacy, overall survival (OS) and safety of drug-eluting beads-TACE (DEB-TACE) and C-TACE as initial treatment in advanced hepatocellular carcinoma (HCC) patients with main portal vein tumor thrombus (mPVTT). Methods The medical records of consecutive advanced HCC patients with mPVTT who underwent initial DEB-TACE or C-TACE from September 2015 to October 2021 were retrospectively evaluated. Treatment crossover was allowed in this retrospective research. The adverse events, disease control rate (DCR), time to tumor progression (TTP) and OS of patients who underwent DEB-TACE were compared with those of patients who underwent C-TACE. Results Eighty-three patients were included: 42 patients in DEB-TACE group and 41 patients in C-TACE group. DEB-TACE could be safely performed in HCC patients with mPVTT, and they gained a better DCR than those submitted to the C-TACE (76.2% vs. 53.7%, P = 0.031), which might have resulted in longer TTP (median TTP: 9.0 months vs. 3.0 months, P

Abstract Background As an invasive technique, selective venous sampling (SVS) is considered a useful method to identify a lesion’s location to increase the success rate of secondary surgery in patients with primary hyperparathyroidism (pHPT) caused by ectopic parathyroid adenomas. Case presentation We present a case of post-surgical persistent hypercalcemia and elevated parathyroid hormone (PTH) levels in a 44-year-old woman with previously undetected parathyroid adenoma. An SVS was then performed for further localization of the adenoma, as other non-invasive methods showed negative results. After SVS, an ectopic adenoma was suspected in the sheath of the left carotid artery, previously considered as a schwannoma, and was pathologically confirmed after the second operation. Postoperatively, the patient’s symptoms disappeared and serum levels of PTH and calcium normalized. Conclusions SVS can provide precise diagnosis and accurate positioning before re-operation in patients with pHPT.

Abstract Purpose To investigate the clinical efficacy and safety of combination therapy of hepatic arterial infusion chemotherapy (HAIC) and anti‐programmed cell death protein‐1 (PD‐1) therapy in the treatment of advanced hepatocellular carcinoma (HCC). Methods In this retrospective clinical research, from March 2018 to December 2019, 1158 HCC patients categorized as BCLC stage C were reviewed for eligibility. We utilized propensity score matching (PSM) to mitigate initial disparities between the groups. The evaluation of the best tumor response was conducted in accordance with mRECIST 1.1 criteria. The difference in survival outcomes including overall survival (OS), progression‐free survival (PFS), and objective response rate (ORR) between groups were compared. Results Following the eligibility review, 453 patients underwent a combined treatment of HAIC with PD1 inhibitors (HAIC‐PD1 group), while 221 patients received HAIC monotherapy (HAIC group) met the inclusion criteria and were finally enrolled in this study. Background Lack of tumor-infiltrating T lymphocytes and concurrent T-cell dysfunction have been identified as major contributors to glioblastoma (GBM) immunotherapy resistance. Upregulating CXCL10 in the tumor microenvironment (TME) is a promising immunotherapeutic approach that potentially increases tumor-infiltrating T cells and boosts T-cell activity but is lacking effective delivery methods.Methods In this study, mesenchymal stem cells (MSCs) were transduced with a recombinant lentivirus encoding Cxcl10, Nrf2 (an anti-apoptosis gene), and a ferritin heavy chain (Fth) reporter gene in order to increase their CXCL10 secretion, TME survival, and MRI visibility. Using FTH-MRI guidance, these cells were injected into the tumor periphery of orthotopic GL261 and CT2A GBMs in mice. Combination therapy consisting of CXCL10-Nrf2-FTH-MSC transplantation together with immune checkpoint blockade (ICB) was also performed for CT2A GBMs. Thereafter, in vivo and serial MRI, survival analysis, and histology examinations were conducted to assess the treatments’ efficacy and mechanism.Results CXCL10-Nrf2-FTH-MSCs exhibit enhanced T lymphocyte recruitment, oxidative stress tolerance, and iron accumulation. Under in vivo FTH-MRI guidance and monitoring, peritumoral transplantation of CXCL10-Nrf2-FTH-MSCs remarkably inhibited orthotopic GL261 and CT2A tumor growth in C57BL6 mice and prolonged animal survival. While ICB alone demonstrated no therapeutic impact, CXCL10-Nrf2-FTH-MSC transplantation combined with ICB demonstrated an enhanced anticancer effect for CT2A GBMs compared with transplanting it alone. Histology revealed that peritumorally injected CXCL10-Nrf2-FTH-MSCs survived longer in the TME, increased CXCL10 production, and ultimately remodeled the TME by increasing CD8+ T cells, interferon-γ+ cytotoxic T lymphocytes (CTLs), GzmB+ CTLs, and Th1 cells while reducing regulatory T cells (Tregs), exhausted CD8+ and exhausted CD4+ T cells.Conclusions MRI-guided peritumoral administration of CXCL10 and Nrf2-overexpressed MSCs can significantly limit GBM growth by revitalizing T lymphocytes within TME. The combination application of CXCL10-Nrf2-FTH-MSC transplantation and ICB therapy presents a potentially effective approach to treating GBM.

Abstract Background Conventional-transarterial chemoembolization (C-TACE) was proven to improve overall survival (OS) in hepatocellular carcinoma (HCC) patients with portal vein tumor thrombus (PVTT), drug-eluting microsphere-TACE (DEM-TACE) was supposed to provide more benefit than C-TACE in this respect. Purpose To compare the safety and efficacy between DEM-TACE and C-TACE as the initial treatment in HCC patients with PVTT and to identify prognostic factors of OS. Methods The medical records of advanced HCC patients with PVTT who underwent DEM-TACE or C-TACE as the initial thearpy from September 2015 with mean follow-up time 14.9 ± 1.2 (95% CI 12.6–17.2) months were retrospectively evaluated. A total of 97 patients were included, 49 patients in the DEM-TACE group and 48 in the C-TACE group. Adverse events (AEs) related to TACE were compared. Tumor and PVTT radiologic response, time to tumor progression (TTP) and OS were calculated and compared in both groups. Results Patients in DEM-TACE group had a better radiologic response (Tumr response: 89.8% vs. 75.0%; PVTT response: 85.7% vs. 70.8%; overall response: 79.6% vs. 58.3%, P = 0.024) and longer TTP (7.0 months vs. 4.0 months, P = 0.040) than patients in C-TACE group. A lower incidence of abdominal pain was found in the DEM-TACE group than in C-TACE group (21 vs. 31, P = 0.032), but there were no significant differences between DEM-TACE and C-TACE patients in any other AEs reported. When compared to C-TACE, DEM-TACE also showed significant OS benefits (12.0 months vs. 9.0 months, P = 0.027). DEM-TACE treatment, the absence of arterioportal shunt (APS), lower AFP value and better PVTT radiologic response were the independent prognostic factors for OS in univariate/multivariate analyses, which provided us with a guide for better patient selection. Conclusions Based on our retrospective study, DEM-TACE can be performed safely and might be superior to C-TACE as the initial treatment for HCC patients with PVTT. Since 2019, the number of journal articles on virtual reality (VR), augmented reality (AR) and mixed reality (MR) has increased dramatically, but existing virtual technology literature reviews have mainly been based on earlier articles. A comprehensive literature review is thus necessary to gain an accurate understanding of the progress and trends in research in this field. Based on the Scopus database, this study systematically reviewed the research on virtual technology in the field of hospitality and tourism in the past 10 years; a total of 274 journal papers were reviewed. The results of bibliometric analysis using VOSviewer identified the research progress and influential literature. Four research clusters were identified based on the bibliometric map: VR research, AR research, VR experiences and virtual technology applications. Thirteen research themes under different clusters were discussed based on content analysis, and research agendas in seven areas were proposed to guide future research. This study contributes to the research on virtual technology in hospitality and tourism by identifying existing research themes and proposing further research directions. It also provides implications for hospitality and tourism marketers to develop marketing strategies using virtual technology.

IntroductionLignification of fruit flesh is a common physiological disorder that occurs during post-harvest storage, resulting in the deterioration of fruit quality. Lignin deposition in loquat fruit flesh occurs due to chilling injury or senescence, at temperatures around 0°C or 20°C, respectively. Despite extensive research on the molecular mechanisms underlying chilling-induced lignification, the key genes responsible for the lignification process during senescence in loquat fruit remain unknown. MADS-box genes, an evolutionarily conserved transcription factor family, have been suggested to play a role in regulating senescence. However, it is still unclear whether MADS-box genes can regulate the lignin deposition that arises from fruit senescence.MethodsBoth senescence- and chilling-induced flesh lignification were simulated by applying temperature treatments on loquat fruits. The flesh lignin content during the storage was measured. Transcriptomic, quantitative reverse transcription PCR and correlation analysis were employed to identify key MADS-box genes that may be involved in flesh lignification. The Dual-luciferase assay was utilized to identify the potential interactions between MADS-box members and genes in phenylpropanoid pathway. Results and DiscussionThe lignin content of the flesh samples treated at 20°C or 0°C increased during storage, but at different rates. Results from transcriptome analysis, quantitative reverse transcription PCR, and correlation analysis led us to identify a senescence-specific MADS-box gene, EjAGL15, which correlated positively with the variation in lignin content of loquat fruit. Luciferase assay results confirmed that EjAGL15 activated multiple lignin biosynthesis-related genes. Our findings suggest that EjAGL15 functions as a positive regulator of senescence-induced flesh lignification in loquat fruit.

Abstract An advanced electrocatalyst to accelerate the sluggish kinetics of multistep redox reactions and suppress the severe shuttle effects is desirable in Li‐S batteries (LSBs). Phase engineering can provide a fascinating way to modulate electronic structures and boost catalytic activity of electrocatalysts. In this study, few‐layered 1T‐MoSe2 nanosheets grown on carbon cloth (1T‐MoSe2/CC) are synthesized and employed as a multifunctional interlayer as well as a catalytic 3D current collector in LSBs to promote both physiochemical confinement and catalytic conversion toward lithium polysulfides (LiPSs). Density functional theory (DFT) calculations reveal that 1T‐MoSe2 has metallic properties beneficial for rapid electronic transport and exhibits a superior catalytic activity to reduce the Gibbs free energy barriers toward LiPS conversion. Significant improvements in chemisorption toward LiPSs, diffusion coefficients of Li ions, and Li2S deposition/decomposition reaction kinetics are realized by the 1T‐MoSe2/CC film. Consequently, the Al@S/AB@1T‐MoSe2/CC LSB, where 1T‐MoSe2/CC is used an interlayer, presents high rate capability of 1253 (1C), 1052 (2C) and 882 (4C) mAh g−1 and excellent long‐term cycling stability at a high rate (2000 cycles at 4C) with a low capacity fading rate (0.017% per cycle). Moreover, with a sandwiched cathode of 1T‐MoSe2/CC@S/AB@1T‐MoSe2/CC, where 1T‐MoSe2/CC works as both a catalytic 3D current collector and a multifunctional interlayer, the LSB at high S loading of 5.7 mg cm−2 and low electrolyte/sulfur ratio of 7.8 μL mg−1 exhibits a high initial areal capacity of 5.43 mAh cm−2 and remarkable rate‐cycling performance (200 cycles).

Loquat is a high-value fruit tree with medicine and fruit homology. Loquat flowers with special fragrance, strong cold resistance and rich in various bioactive components, are valuable agricultural auxiliary products and have been widely used for making floral teas and beverages in recent years. In this study, we found the concentration of active components increased from the floral buds to initial flowers along with flower development, the bioactives of initial flowers were the richest in four flowering stages, and loquat flowers contained major volatile components such as alcohols, aldehydes and esters, which are the source of fragrance. When extract with hot water, the most efficient method was 80 °C for 30 min or boiling water within 2 h. For Baijiu (56% Vol), the best solid-to-liquid ratio was 3:100 (Dry flower: Baijiu) in 6–12 h. Baijiu achieved higher bioactive content than water extraction, the amygdalin concentration in Baijiu reached 0.3 mg/mL.

Building information models (BIMs) offer advantages, such as visualization and collaboration, making them widely used in the management of existing buildings. Currently, most BIMs for existing indoor spaces are manually created, consuming a significant amount of manpower and time, severely impacting the efficiency of building operations and maintenance management. To address this issue, this study proposes an automated reconstruction method for an indoor scene BIM based on a feature-enhanced point transformer and an octree. This method enhances the semantic segmentation performance of point clouds by using feature position encoding to strengthen the point transformer network. Subsequently, the data are partitioned into multiple segments using an octree, collecting the geometric and spatial information of individual objects in the indoor scene. Finally, the BIM is automatically reconstructed using Dynamo in Revit. The research results indicate that the proposed feature-enhanced point transformer algorithm achieves a high segmentation accuracy of 71.3% mIoU on the S3DIS dataset. The BIM automatically generated from the field point cloud data, when compared to the original data, has an average error of ±1.276 mm, demonstrating a good reconstruction quality. This method achieves the high-precision, automated reconstruction of the indoor BIM for existing buildings, avoiding extensive manual operations and promoting the application of BIMs for the maintenance processes of existing buildings.

Abstract A recently developed synthetic retinoid abrogates proliferation and induces apoptosis of drug‐resistant malignant‐cancer‐stem‐cell‐like cells. However, the underlying mechanisms of how the synthetic retinoid induces cancer‐stem‐cell‐like cell tumor‐repopulating cell (TRC) apoptosis are elusive. Here, it is shown that although the retinoid and conventional anticancer drugs cisplatin, all‐trans retinoic acid, and tazarotene all inhibit cytoskeletal tension and decondense chromatin prior to inducing TRC apoptosis, half‐maximal inhibitory concentration of the retinoid is 20‐fold lower than those anticancer drugs. The synthetic retinoid induces retinoic acid receptor gamma (RARγ) translocation from the nucleus to the cytoplasm, leading to reduced RARγ binding to Cdc42 promoter and Cdc42 downregulation, which decreases filamentous‐actin (F‐actin) and inhibits cytoskeletal tension. Elevating F‐actin or upregulating histone 3 lysine 9 trimethylation decreases retinoid‐induced DNA damage and apoptosis of TRCs. The combinatorial treatment with a chromatin decondensation molecule and the retinoid inhibits tumor metastasis in mice more effectively than the synthetic retinoid alone. These findings suggest a strategy of lowering cell tension and decondensing chromatin to enhance DNA damage to abrogate metastasis of cancer‐stem‐cell‐like cells with high efficacy.

The optimization of irrigation and fertilization indexes for peanuts with drip irrigation is urgently needed in Xinjiang. A field experiment was conducted during the 2021 peanut growing season at Urumqi, Xinjiang, in Northwestern China, to evaluate the effects of different water and nitrogen treatments on the growth, yield, and water and nitrogen utilization of peanuts. In field experiments, we set up three irrigation levels (irrigation water quotas of 22.5, 30, and 37.5 mm, respectively, for W1, W2, and W3), two nitrogen application levels (77.5 and 110 kg·ha−1, recorded as N1 and N2), and a control treatment (W2N0) that did not include the application of nitrogen. The results showed that nitrogen application enhanced the growth, physiological indexes, yield, and water use efficiency of the W1, W2, and W3 treatments when the irrigation volume remained the same. In comparison with no nitrogen application (W2N0), the peanut growth, physiological indexes, yield, and water use efficiency improved with increasing irrigation amounts in the N1 and N2 treatments. With an increase in the irrigation volume, the water use efficiency grew; the W3N2 treatment had the highest water use efficiency, which was 1.32 kg·m−3. The total water consumption and reproductive-stage water consumption of the peanuts in all treatments increased with the irrigation volume, and a high yield was achieved at 402.57 mm, which was 5.2974 Mg·ha−1. In the W1, W2, and W3 treatments, the nitrogen partial factor productivity significantly decreased as the nitrogen application increased, with the nitrogen partial factor productivity in the W3N1 treatment being the highest, at 60.61 kg·kg−1. A comprehensive evaluation based on principal component analysis assigned W3N2 the higher score. These findings suggest that irrigation water quotas of 37.5 mm should be coupled with 110 kg·ha−1 nitrogen applications for peanuts using drip irrigation in mulch film in Xinjiang.

To improve the quality of reclaimed asphalt pavement materials (RAPs) and reduce the variability of RAPs, a refined decomposition process was applied to treat RAPs. The change rule and variability of aggregate gradation, asphalt content, aged asphalt properties and aggregate (extracted) properties of RAPs were analyzed with different frequency parameters. In addition, the gradation variability control method and the asphalt content variability control method were used to calculate the maximum addition proportion of RAPs according to the quality control requirements of hot-mixing asphalt mixtures. The results indicate that the variability of aggregate gradation, asphalt content and asphalt properties significantly reduced for the refined decomposition RAP. Compared with the original RAP (0 Hz) of 4.75–9.5 mm and 9.5–19 mm, the “false particle” content of the refined decomposition RAP (50 Hz) reduced by 75.6% and 64.3%, respectively. The refined decomposition process is conducive to the road performance of recycled asphalt mixtures, especially the dynamic stability and the maximum bending tensile strain. Comparing the recycled asphalt mixture with the original RAP (0 Hz), the maximum bending strain of the recycled asphalt mixture with the refined decomposition RAP (50 Hz) increased by 69%, and the immersion residual stability increased to 87.9%. The refined decomposition process improves the quality of the RAP and provides a reference for recycled asphalt mixtures with a high proportion of RAPs. This study contributes to RAP high-level recycling and carbon reduction in the highway maintenance industry.

Abstract Background Recurrent hepatocellular carcinoma (HCC) shows strong resistance to sorafenib, and the tumor-repopulating cells (TRCs) with cancer stem cell-like properties are considered a driver for its high recurrent rate and drug resistance. Methods Suppression of TRCs may thus be an effective therapeutic strategy for treating this fatal disease. We evaluated the pharmacology and mechanism of sulfarotene, a new type of synthetic retinoid, on the cancer stem cell-like properties of HCC TRCs, and assessed its preclinical efficacy in models of HCC patient-derived xenografts (PDXs). Results Sulfarotene selectively inhibited the growth of HCC TRCs in vitro and significantly deterred TRC-mediated tumor formation and lung metastasis in vivo without apparent toxicity, with an IC50 superior to that of acyclic retinoid and sorafenib, to which the recurrent HCC exhibits significant resistance at advanced stage. Sulfarotene promoted the expression and activation of RARα, which down-regulated SOS2, a key signal mediator associated with RAS activation and signal transduction involved in multiple downstream pathways. Moreover, sulfarotene selectively inhibited tumorigenesis of HCC PDXs with high expression for SOS2. Conclusions Our study identified sulfarotene as a selective inhibitor for the TRCs of HCC, which targets a novel RARα-SOS2-RAS signal nexus, shedding light on a new, promising strategy of target therapy for advanced liver cancer.

Autoimmune diseases are a series of diseases involving multiple tissues and organs, characterized by the over production of abnormal multiple antibodies. Although most studies support that the impaired immune balance participates in the development of autoimmune diseases, the specific pathogenesis of it is not fully understood. Intestinal immunity, especially the intestinal mucosal barrier has become a research hotspot, which is considered to be an upstream mechanism leading to the impaired immune balance. As an important defense barrier, the intestinal mucosal barrier regulates and maintains the homeostasis of internal environment. Once the intestinal barrier function is impaired under the effect of multiple factors, it will destroy the immune homeostasis, trigger inflammatory response, and participate in the development of autoimmune diseases in the final. However, the mechanism of the intestinal mucosal barrier how to regulate the homeostasis and inflammation is not clear. Some studies suggest that it maintains the balance of immune homeostasis through the zonulin pathway, intestinal microbiome, and Toll-like receptor signaling pathway. Our review focused on the composition and the function of the intestinal mucosal barrier to describe the research progress of it in regulating the immune homeostasis and inflammation, and also pointed that the intestinal mucosal barrier was the potential targets in the treatment of autoimmune diseases.

Abstract In recent years, halide perovskite solar cells (HPSCs) have attracted a great attention due to their superior photoelectric performance and the low‐cost of processing their quality films. In order to commercialize HPSCs, the researchers are focusing on developing high‐performance HPSCs. Many strategies have been reported to increase the power conversion efficiency and the long‐term stability of HPSCs over the past decade. Herein, we review the latest efforts and the chemical‐physical principles for preparing high‐efficiency and long‐term stability HPSCs in particular, concentrating on the perovskite materials, technologies for perovskite films, charge transport materials and ferroelectric effect to reduce the carrier loss, and photon management via plasmonic and upconversion effects. Finally, the key issues for future researches of HPSCs are also discussed with regard to the requirements in practical application.

PURPOSEWe aimed to evaluate the feasibility and safety of a modified technique for portal vein recanalization, percutaneous transluminal sharp recanalization (PTSR), when performing transjugular intrahepatic portosystemic shunt (TIPS) for the treatment of chronic portal vein occlusion (CPVO) and portal hypertension.METHODSNine consecutive patients with CPVO and portal hypertension had undergone TIPS and PTSR procedure after failing in conventional percutaneous catheterization from March 2017 to July 2019. Technical success rates, effectiveness, and complications were evaluated. Follow-up of patients’ clinical outcomes and shunt patency were performed periodically. Primary and secondary shunt patency were analyzed by Kaplan-Meier method.RESULTSThe occluded portal veins were successfully recanalized after failing in conventional percutaneous catheterization, and TIPS procedures were completed in all 9 patients. By 4000 BP, trans-Eurasian agricultural exchanges increased across the Hexi Corridor. However, the nature and timing of many early prehistoric agricultural exchanges remain unclear. We present systematically collected archaeobotanical data from the ancient Haizang site (3899–3601 cal a BP) within the Hexi Corridor. Adding to previous archaeobotanical studies of the Hexi Corridor, we find that agricultural production transformed from purely millet-based agriculture during the Machang Period (4300–4000) to predominantly millet-based agriculture increasingly supplemented with wheat and barley during the Xichengyi and Qijia periods (4000–3600 BP). These transformations are likely due to adaption to a cooler and drier climate through cultural exchange. A warm and humid climate during 4300–4000 BP likely promoted millet agriculture, Machang cultural expansion westward, and occupation across the Hexi corridor. However, after the “4.2 ka BP cold event” people adopted wheat and barley from the West to make up for declining millet agricultural productivity. This adoption began first with the Xichengyi culture, and soon spread further eastward within the Hexi Corridor to the Qijia culture.

Demulsifiers are considered the key materials for oil/water separation. Various works in recent years have shown that demulsifiers with polyoxypropylen epolyoxyethylene branched structures possess better demulsification effects. In this work, inspired by the chemical structure of demulsifiers, a novel superhydrophilic/underwater superoleophobic membrane modified with a polyoxypropylene polyoxyethylene block polymer was fabricated for enhanced separation of O/W emulsion. First, a typical polyoxypropylene polyoxyethylene triblock polymer (Pluronic F127) was grafted onto the poly styrene-maleic anhydride (SMA). Then, the Pluronic F127-grafted SMA (abbreviated as F127@SMA) was blended with polyvinylidene fluoride (PVDF) for the preparation of the F127@SMA/PVDF ultrafiltration membrane. The obtained F127@SMA/PVDF ultrafiltration membrane displayed superhydrophilic/underwater superoleophobic properties, with a water contact angle of 0° and an underwater oil contact angle (UOCA) higher than 150° for various oils. Moreover, it had excellent separation efficiency for SDS-stabilized emulsions, even when the oil being emulsified was crude oil. The oil removal efficiency was greater than 99.1%, and the flux was up to 272.4 L·m−2·h−1. Most importantly, the proposed F127@SMA/PVDF membrane also exhibited outstanding reusability and long-term stability. Its UOCA remained higher than 150° in harsh acidic, alkaline, and high-salt circumstances. Overall, the present work proposed an environmentally friendly and convenient approach for the development of practical oil/water separation membranes.

Chromatin is a unique structure of DNA and histone proteins in the cell nucleus and the site of dynamic regulation of gene expression. Soluble factors are known to affect the chromatin structure and function via activating or inhibiting specific transcription factors. Forces on chromatin come from exogenous stresses on the cell surface and/or endogenous stresses, which are regulated by substrate mechanics, geometry, and topology. Forces on chromatin involve direct (via adhesion molecules, cytoskeleton, and the linker of nucleoskeleton and cytoskeleton complexes) and indirect (via diffusion and/or translocation processes) signaling pathways to modulate levels of chromatin folding and deformation to regulate transcription, which is controlled by histone modifications and depends on magnitude, direction, rate/frequency, duration, and modes of stresses. The rapid force transmission pathway activates multiple genes simultaneously, and the force may act like a “supertranscription factor.” The indirect mechanotransduction pathways and the rapid force transmission pathway together exert sustained impacts on the chromatin, the nucleus, and cell functions.

In the process of waterflooding development of heavy oil, W/O emulsion has a strong ability to improve the mobility ratio and block the high-permeability layer, which can effectively improve the sweep coefficient and enhance oil recovery. In this paper, the stability and droplet size distribution of emulsions under different conditions were studied by taking heavy oil and formation water from Jimusar Oilfield in Xinjiang as samples. On this basis, double-pipe core flooding experiments were carried out to study the shut-off ability and oil displacement efficiency of W/O emulsion, and then a numerical simulation was carried out. The results show that oil and water can be completely emulsified when the stirring speed is higher than 4000 r/min. A stable emulsion can be formed when the experimental temperature is lower than 60 °C. A lower water cut results in a more stable emulsion. The emulsion is difficult to stabilize after the salinity exceeds 10,000 mg/L. When the pH value is about 7, the stability of the emulsion is the worst. With the increase in stirring speed, the increase in temperature, and the decrease in water content and salinity, the emulsion droplet size range is relatively concentrated, and the average particle size is smaller. In heterogeneous reservoirs, the permeability of different percolation channels is quite different, such that the displacement fluid only percolates along the high-permeability channel and cannot drive oil effectively. The results of displacement experiments show that the emulsion with a water cut of 60% has high viscosity and obvious sweep ability, but its stability is very poor; the effect is opposite when the water cut is less than 40%. The shut-off ability of W/O emulsion disappears gradually when the permeability contrast is more than 5.92. The research results are of great significance for improving oil recovery in heterogeneous heavy oil reservoirs.

Study design: A systematic review and meta-analysis. Objective: The objective of this meta-analysis was to evaluate the association between calcium-phosphorus balance and adolescent idiopathic scoliosis (AIS). Methods: Databases, including PubMed, OVID database, Web of Science, CBM database and CNKI database were searched for the relevant case control studies and cross-sectional studies. Two authors selected studies and extracted data independently. Data analysis was performed by Review Manager Software 5.0. Subgroup analysis was performed on the serum level of vitamin D according to gender and menstruation. Results: Five studies were included, with a total of 646 cases of AIS and 791 controls. AIS group had a lower serum level of vitamin D compared to control group [MD = −6.74, 95% CI (−9.47, −4.00)]. Gender and menstruation condition were thought to have no effect on the primary outcome of vitamin D level by subgroup analysis [MD = −5.97, 95% CI (7.61, −4.34)]. The AIS group had a lower calcium level [SMD= −0.77, 95% CI (−1.51, −0.02)] and calcitonin level compared to control group. There was no statistical difference in phosphorus level [SMD=0.5, 95% CI (−0.46, 0.57)] and parathyroid hormone level [SMD = −0.11, 95% CI (−0.54, −0.31)]. Meanwhile, the observational indexes, including serum levels of calcium, phosphorus, parathyroid hormone and calcitonin were within normal limits. Conclusion: Vitamin D deficiency may be involved in the pathogenesis of AIS by influencing the regulation of calcium-phosphors metabolism on human bone. Abstract Background The development of Systemic lupus erythematosus (SLE) has been associated with the balance of Th17 and Treg cells. IL-2 and rapamycin can influence the populations of both Th17 and Treg cells. However, it is unclear whether low dose of IL-2 and rapamycin can relieve the symptoms of SLE patients and what is the mechanisms. In this study, we aim to analyze the effect of low dose of IL-2 plus rapamycin on the number of Tregs, Th17 cells and the ratio of Th17/Treg cells, as well as to evaluate its therapeutic efficacy in refractory SLE patients. Result Fifty refractory SLE patients and 70 healthy controls were enrolled and followed up for 24 weeks. We found that compared with HC, the refractory SLE patients had a lower number of Tregs, a similar number of Th17 cells, but an increased ratio of Th17/Treg. After the treatment, the number of Tregs of the patients at 12th and 24th week was significantly increased. While the number of Th17 cells was unchanged, the ratio of Th17/Treg was significantly decreased at both 6 weeks and 24 weeks. After 6, 12 and 24 weeks of treatment, the SLEDAI score was significantly reduced. The prednison dosage at 6th,12th and 24th week post treatment was significantly decreased. Conclusion Our results support that the reduction of Tregs and the imbalance of Th17/Treg cells were correlated with the occurrence and development of refractory SLE. Low dose of IL-2 combined with rapamycin was able to restore the number of Tregs and the balance of Th17/Treg cells. As a result, this approach was able to induce immune tolerance and promote disease remission, allowing for the reduction in prednisone dosage. Purpose: To present the early results of pirarubicin-eluting microsphere transarterial chemoembolization (PE-TACE) for patients with unresectable hepatocellular carcinoma (HCC). Materials and methods: We retrospectively analyzed 55 consecutive patients with HCC who received PE-TACE between April 1, 2015 and August 30, 2016. The complication rate, tumor response rate, progression-free survival (PFS), and overall survival (OS) were analyzed. Results: Adverse events were generally mild and included abdominal pain and fever, although a major complication was reported in 1 patient (1.8%). During a median follow-up of 10.0 months (range, 3.0–24.0 months), 14 patients (25.5%) achieved a complete tumor response, 25 (45.5%) had a partial response, 9 (16.4%) showed stable disease, and 7 (12.7%) had disease progression. The 1-month overall response rate was 70.9%, and the local tumor response rate was 89.0%. The 1-month tumor response rate was 100% for Barcelona Clinic Liver Cancer (BCLC) stage A or B disease and 62.8% for BCLC stage C disease. The median PFS was 6.1 months (95% confidence interval [95%CI], 3.4–8.8 months; range, 1.0–24.0 months). The median OS was 11.0 months (95%CI, 7.1–14.9 months; range, 2.0–24.0 months). Kaplan-Meier analysis (log-rank test) found significant differences in OS between patients grouped by tumor number (P = 0.006), tumor size (P = 0.035), and Eastern Cooperative Oncology Group (ECOG) score (P = 0.005). The tumor number (1 vs. ≥2) was the only factor independently associated with OS (hazard ratio [HR], 2.867; 95%CI, 1.330–6.181; P = 0.007). Conclusions: PE-TACE for unresectable HCC may be safe, with favorable tumor response rates and survival time, especially in patients with a single large tumor. Longer follow-up using a larger series is necessary to confirm these preliminary results. Helicobacter pylori harbors a dipeptide (Dpp) transporter consisting of a substrate-binding protein (DppA), two permeases (DppB and C), and two ATPases (DppD and F). The Dpp transporter is responsible for the transportation of dipeptides and short peptides. We found that its expression is important for the growth of H. pylori. To understand the role of the Dpp transporter in the pathogenesis of H. pylori, the expression of virulence factors and H. pylori-induced IL-8 production were investigated in H. pylori wild-type and isogenic H. pylori Dpp transporter mutants. We found that expression of CagA was downregulated, while expression of type 4 secretion system (T4SS) components was upregulated in Dpp transporter mutants. The DppA mutant strain expressed higher levels of outer membrane proteins (OMPs), including BabA, HopZ, OipA, and SabA, and showed a higher adhesion level to gastric epithelial AGS cells compared with the H. pylori 26695 wild-type strain. After infection of AGS cells, H. pylori ΔdppA induced a higher level of NF-κB activation and IL-8 production compared with wild-type. These results suggested that in addition to supporting the growth of H. pylori, the Dpp transporter causes bacteria to alter the expression of virulence factors and reduces H. pylori-induced NF-κB activation and IL-8 production in gastric epithelial cells.

Abstract The rational design of electrode structure with catalysts adequately utilized is of vital importance for future fuel cells. Herein, a novel 3D oriented wholly integrated electrode comprising core–shell Fe3O4@N‐doped‐C (Fe3O4@NC) nanoparticles embedded into N‐doped ordered interconnected hierarchical porous carbon (denoted as Fe3O4@NC/NHPC) is developed for the oxygen reduction reaction (ORR). The as‐prepared catalyst possesses novel structure and efficient active sites. In rotating disk electrode measurements, the Fe3O4@NC/NHPC exhibits almost identical ORR electrocatalytic activity, superior durability, and much better methanol tolerance compared with the commercial Pt/C in acidic media. To the authors’ knowledge, this is among the best non‐precious‐metal ORR catalysts reported so far. Importantly, the Fe3O4@NC/NHPC is successfully in situ assembled onto carbon paper by the electrophoresis method to obtain a well‐designed 3D‐ordered electrode. With improved mass transfer and maximized active sites for ORR, the 3D‐oriented wholly integrated electrode shows superior performance to the one fabricated by the traditional method.

Increasing importance is placed upon the effect of mechanical forces on cell regulation, fate and disease states. Here, the authors describe a deformable fluorescent nanoparticle labeled elastic microsphere which can be used to calculate strain and traction forces in vitro and in vivo.

Multimodality optical imaging probes have emerged as powerful tools that improve detection sensitivity and accuracy, important in disease diagnosis and treatment. In this review, we focus on recent developments of optical fluorescence imaging (OFI) probe integration with other imaging modalities such as X-ray computed tomography (CT), magnetic resonance imaging (MRI), positron emission tomography (PET), single-photon emission computed tomography (SPECT), and photoacoustic imaging (PAI). The imaging technologies are briefly described in order to introduce the strengths and limitations of each techniques and the need for further multimodality optical imaging probe development. The emphasis of this account is placed on how design strategies are currently implemented to afford physicochemically and biologically compatible multimodality optical fluorescence imaging probes. We also present studies that overcame intrinsic disadvantages of each imaging technique by multimodality approach with improved detection sensitivity and accuracy. The gasification kinetics of bituminous coal char was investigated in a mixture of CO2, H2O, CO, H2, and N2 under isothermal conditions. In addition, the impacts of gasification temperature, gasification time, and gas composition on the gasification process were analyzed. As the experimental results suggest, there is a significant increase of the carbon conversion degree of bituminous coal char not just when gasification temperature and time increase, but also when H2 and CO concentration decreases. The kinetics of bituminous coal char in the gasification process was successfully modeled as a shrinking unreacted core. It is concluded that the gasification of bituminous coal char is controlled by an internal chemical reaction in the early stage and diffusion in the later stage. The activation energies of bituminous coal char gasification for different stages were studied. Moreover, it is proposed for the first time, to our knowledge, that the diffusion-control step is significantly shortened with the decrease of the CO2/H2O ratio. As scanning-electron-microscopy results suggest, bituminous coal char gasified in CO2/H2O = 1/3 atmosphere has numerous inner pores (0⁻5 m). Therefore, in the process of gasification, the inner pores provide a gas channel that reduces the gas diffusion resistance and thus shortens the diffusion-control step. These results can serve as a reference for industrialized application of the technology of coal gasification direct reduced iron.

Vanadium titano-magnetite (VTM) is a significant resource in China—analysis shows that China possesses approximately 10 billion tons of VTM. In this study, we characterize VTM’s isothermal reduction mechanisms in the mixture of H2, CO, and N2 where the variables considered include reduction time, reduction temperature, gas composition, and pellet size. The kinetics of the reduction process were studied following a shrinking core model. The results indicate that the reduction degree of oxidized VTM pellets increases with increases of reduction time and reduction temperature but decreases with increasing pellet size. Moreover, we found that an increase of H2/(H2 + CO) ratio induced an increase of the reduction degree. We discuss the transformation of main Ti-bearing mineral phases, and we consider the most probable reaction mechanism. For the entire reduction process, the kinetic results confirm the existence of an early and later stages that are controlled by interface chemical reaction and diffusion, respectively. Furthermore, the results show that the diffusion-control step can be observably shortened via decreased pellet size because a thinner product layer is formed during the reduction process. Our study thus provides a valuable technical basis for industrial applications of VTM.

Systemic lupus erythematosus (SLE) is an autoimmune disease that is characterized by the overproduction of autoantibodies against an array of nuclear and cytoplasmic antigens and affects multiple organs, such as the skin, joints, kidneys, and neuronal tissues. T cells have been recognized as important players in the development of SLE due to their functions in cytokine secretion, antigen presentation, and supporting B cells for antibody production. γδ T cells are a minor population of T cells that play important roles in infection and tumor-associated disease. In recent years, the role of γδ T cells in autoimmune diseases has been investigated. In this review, we discussed the role of γδ T cells in the pathogenesis of SLE.

Systemic lupus erythematosus (SLE) is a multisystem autoimmune disease with different variety of clinical manifestations. Natural killer T (NKT) cells are innate lymphocytes that play a regulatory role during broad range of immune responses. A number of studies demonstrated that the quantity and quality of invariant NKT (iNKT) cells showed marked defects in SLE patients in comparison to healthy controls. This finding suggests that iNKT cells may play a regulatory role in the occurrence and development of this disease. In this review, we mainly summarized the most recent findings about the behavior of NKT cells in SLE patients and mouse models, as well as how NKT cells affect the proportion of T helper cells and the production of autoreactive antibodies in the progress of SLE. This will help people better understand the role of NKT cells in the development of SLE and improve the therapy strategy.

Dehydrins (DHNs) are a family of plant proteins typically induced in response to stress conditions that cause cellular dehydration, such as low temperatures, high salinity, and drought. Loquat (Eriobotrya japonica) is a perennial fruit crop that blossoms during winter. Loquat fruitlets are frequently injured by freezing. To evaluate the role of the EjDHNs in freezing resistance in loquat fruitlets, two cultivars of loquat, the freezing-sensitive 'Ninghaibai' (FS-NHB) and the freezing-tolerant 'Jiajiao' (FT-JJ), were analyzed under induced freezing stress. Freezing stress led to obvious accumulation of reactive oxygen species and considerable lipid peroxidation in membranes during the treatment period. Both these phenomena were more pronounced in 'FS-NHB' than in 'FS-JJ.' Immunogold labeling of dehydrin protein was performed. DHN proteins were found to be concentrated mainly in the vicinity of the plasma membrane, and the density of the immunogold labeling was significantly higher after freezing treatment, especially in the more freezing-tolerant cultivar 'FT-JJ.' Seven DHNs, showing four different structure types, were obtained from loquat fruitlets and used to study the characteristics of different EjDHN proteins. These DHN proteins are all highly hydrophilic, but they differ significantly in size, ranging from 188 to 475 amino acids, and in biochemical properties, such as theoretical pI, aliphatic index, and instability index. Freezing treatment resulted in up-regulation of the expression levels of all seven EjDHNs, regardless of structure type. The accumulation of the transcripts of these EjDHN genes was much more pronounced in 'FT-JJ' than in 'FS-NHB.' Altogether, this study provides evidence that EjDHNs are involved in the cryoprotection of the plasma membrane during freeze-induced dehydration in loquat fruitlets.
