Abstract In this study, changes in bioactive compound contents and the in vitro biological activity of mixed grains, including oats, sorghum, finger millet, adzuki bean, and proso millet, with eight different blending ratios were investigated. The total phenolic compounds and flavonoid contents ranged from 14.43–16.53 mg gallic acid equivalent/g extract and 1.22–5.37 mg catechin equivalent/g extract, respectively, depending on the blending ratio. The DI-8 blend (30% oats, 30% sorghum, 15% finger millet, 15% adzuki bean, and 10% proso millet) exhibited relatively higher antioxidant and anti-diabetic effects than other blending samples. The levels of twelve amino acids and eight organic acids in the grain mixes were measured. Among the twenty metabolites, malonic acid, asparagine, oxalic acid, tartaric acid, and proline were identified as key metabolites across the blending samples. Moreover, the levels of lactic acid, oxalic acid, and malonic acid, which are positively correlated with α-glucosidase inhibition activity, were considerably higher in the DI-blending samples. The results of this study suggest that the DI-8 blend could be used as a functional ingredient as it has several bioactive compounds and biological activities, including anti-diabetic activity.

We comparatively evaluated the antioxidant properties of key triterpenes from Centella asiatica, including asiatic acid (AA), asiaticoside, madecassic acid, and madecassoside, in several cell types, including skin fibroblasts, macrophages, hepatocytes, and endothelial cells, under conditions promoting oxidative stress. AA conferred the highest viability on Hs68 cells exposed to ultraviolet B (UVB) irradiation. Triterpene pretreatment attenuated the UVB-induced generation of reactive oxygen species (ROS) and malondialdehyde (MDA), as well as the UVB-induced depletion of glutathione (GSH) in skin fibroblasts. AA most potently inhibited UVB-induced MMP generation, resulting in increased intracellular collagen levels. Pretreatment with triterpenes, particularly AA, significantly improved cell viability and attenuated TBHP-induced levels of ROS, alanine aminotransferase, and aspartate aminotransferase in HepG2 cells. Triterpenes attenuated ROS levels and reduced MDA and GSH expression in EA.hy926 cells. In RAW264.7 macrophages, production of nitric oxide, tumor necrosis factor-α, and interleukin-6 (indicators of LPS-induced oxidative damage) was significantly reduced by treatment with any of the triterpenes. Statistical analyses of triterpene biological activities using principal component analysis and hierarchical clustering revealed that AA exerted the greatest overall influence and showed remarkable activity in Hs68 and HepG2 cells.

Purpose To develop and validate a deep learning-based screening tool for the early diagnosis of scoliosis using chest radiographs with a semi-supervised generative adversarial network (GAN). Materials and Methods Using a semi-supervised learning framework with a GAN, a screening tool for diagnosing scoliosis was developed and validated through the chest PA radiographs of patients at two different tertiary hospitals. Our proposed method used training GAN with mild to severe scoliosis only in a semi-supervised manner, as an upstream task to learn scoliosis representations and a downstream task to perform simple classification for differentiating between normal and scoliosis states sensitively. Results The area under the receiver operating characteristic curve, negative predictive value (NPV), positive predictive value, sensitivity, and specificity were 0.856, 0.950, 0.579, 0.985, and 0.285, respectively. Conclusion Our deep learning-based artificial intelligence software in a semi-supervised manner achieved excellent performance in diagnosing scoliosis using the chest PA radiographs of young individuals; thus, it could be used as a screening tool with high NPV and sensitivity and reduce the burden on radiologists for diagnosing scoliosis through health screening chest radiographs.

Centella asiatica possess various health-promoting activities owing to its bioactive compounds such as triterpenes, flavonoids, and vitamins. Ultrasound treatment during the post-harvest process is a good strategy for eliciting secondary metabolite in plants. The present study investigated the effect of ultrasound treatment for different time durations on the bioactive compounds and biological activities of C. asiatica leaves. The leaves were treated with ultrasound for 5, 10, and 20 min. Ultrasound elicitation (especially for 10 min) markedly elevated the accumulation of stress markers, leading to enhanced phenolic-triggering enzyme activities. The accumulation of secondary metabolites and antioxidant activities were also significantly improved compared with that in untreated leaves. In addition, ultrasound-treated C. asiatica leaves protected myoblasts against H2O2-induced oxidative stress by regulating reactive oxygen species production, glutathione depletion, and lipid peroxidation. These findings indicate that elicitation using ultrasound can be a simple method for increasing functional compound production and enhancing biological activities in C. asiatica leaves.

Unsaponifiable matter (USM) from wheat bran, a by-product obtained from wheat milling, is abundant in health-promoting compounds such as phytosterols, tocopherols, policosanols, and alkylresorcinols. This study aimed to examine the effects of USM from the wheat bran of normal and waxy type wheat, Saekeumkang (SKK) and Shinmichal (SMC), on hepatic lipid accumulation in free fatty acid (FFA)-induced hepatocytes and to investigate the cellular mechanism. The total phytochemical contents were 46.562 g/100 g USM and 38.130 g/100 g USM from SKK and SMC, respectively. FFA treatment increased intracellular lipid accumulation by approximately 260% compared to the control group; however, treatment with USM from SKK and SMC significantly attenuated lipid accumulation in the hepatocytes in a dose-dependent manner. Moreover, USM downregulated the expression of lipogenic factors such as fatty acid synthase and sterol regulatory-element-binding protein 1c by approximately 40% compared to the FFA treatment group. Treatment with USM promoted lipolysis and positively regulated the expression of the proteins involved in β-oxidation, including peroxisome proliferator-activated receptor α and its downstream protein, carnitine palmitoyltransferase 1A. Moreover, the blockade of AMPK activation significantly abolished the inhibitory effects of USM on hepatic lipid accumulation. These results indicated that the USM from both SKK and SMC can alleviate lipid accumulation in hepatocytes in an AMPK-dependent manner. Therefore, USM from wheat bran may be useful as a therapeutic intervention for treating metabolic-dysfunction-associated fatty liver disease.

This paper examines the convergence of carbon dioxide (CO2) emissions in OECD countries. Specifically, we are interested in examining to what extent the per capita emissions of different countries tend to draw closer over time. This issue is important since this question is associated with fairness regarding the distribution of per capita emissions. In this context, we examine the stochastic convergence of per capita carbon dioxide emissions for 30 OECD countries from 1960 to 2018. We adopt a newly developed test by Payne et al. (2022), which allows for two structural breaks and cross-correlations in a factor structure. Our results accounting for breaks and factors jointly show that per capita CO2 emissions do not converge, implying that the differences in CO2 emissions exist and are persistent.

This paper proposes new cointegration tests based on instrumental variable (IV) estimation. An important property of our tests is that the asymptotic distribution remains standard normal (or Chi-square) regardless of the number of regressors, differing deterministic terms, structural dummies, and inclusion of stationary covariates. Thus, our IV cointegration tests have the operational advantage that they do not depend on nuisance parameters. As such, we can incorporate stationary covariates into a model to enhance power without affecting the asymptotic distribution of the test. This is important because it alleviates the need to tabulate the critical values for every possible case or to bootstrap the critical values.

Finite time stability involving dynamical systems whose trajectories converge to a Lyapunov stable equilibrium state in finite time have been studied for both continuous-time and discrete-time systems. For continuous-time systems, finite time stability is defined for equilibria of continuous but non-Lipschitzian nonlinear dynamics, whereas discrete-time systems can exhibit finite time stability even when the system dynamics are linear, and hence, Lipschitz continuous. Alternatively, for impulsive dynamical systems it may be possible to reset the system states to an equilibrium state achieving finite time stability without requiring a non-Lipschitz condition for the continuous-time part of the hybrid system dynamics. In this paper, we develop sufficient Lyapunov conditions for finite time stability of impulsive dynamical systems using both a scalar differential Lyapunov inequality on the continuous-time dynamics as well as a scalar difference Lyapunov inequality on the discrete-time resetting dynamics. Furthermore, using our proposed finite time stability results, we design universal hybrid finite time stabilizing control laws for impulsive dynamical systems. Finally, we present several numerical examples for finite time stabilization of network impulsive dynamical systems.

The accurate screening of otitis media (OM) lies in clarifying the numerous confounding and quantitative factors that are discovered during primary inspections. Increased awareness about bacterial biofilms and inflammation has allowed researchers to develop a better understanding of the bacterial infections that occur in the middle ear. In this study, four live guinea pigs were inoculated with Streptococcus pneumonia to induce OM-related inflammatory changes. Since optical techniques have been effectively used for diagnosis in medicine, low-coherence interferometry-based optical coherence tomography (OCT) was employed for depth-resolved high-resolution data screening. Multiple locations of the tympanic membrane (TM), mastoid mucosa, and round window membrane were examined to assess inflammatory changes. We performed qualitative assessments and thickness quantifications and investigated the variations due to adverse inflammatory affects. The findings of the present study provide a better understanding of S. pneumonia bacteria caused inflammatory changes in middle ear and it further provides a fundamental platform for future clinical utility.

Optical fiber is widely used in optical coherence tomography (OCT) to propagate light precisely with low attenuation and low dispersion. However, the total optical path length within the optical fiber varies in accordance with changes of the temperature. This leads changes in the total optical travel path of the interfering signals and results in shifting of OCT image position to an unintended depth pixel value. In this paper, we presented the temperature-based automatic path length compensating method in OCT to limit the external temperature effect and control the image position in micro-scale without manual movement of optical components. By utilizing developed hardware and software of automatic temperature control system, the external temperature of optical fiber is precisely regulated that evokes thermal expansion and finally changes the physical length of fiber, which is main mechanism of temperature-based path length compensating method. The effectiveness of the presented method was verified by two-dimensional OCT images of mirror and in vivo retina. The obtained results confirmed the path length variance due to temperature change is computable and can be regulated in real-time for whole pixel range of OCT image. Therefore, the proposed temperature-based path length compensating method can be used as an alternative method to precisely control the position of OCT image, while eliminating the effect of external temperature and apply to effectively configuring compact optical systems.

This study investigated the effects of high hydrostatic pressure (HHP) treatment on the physicochemical properties of rice flour according to its moisture levels in order to develop new materials for processed rice foods. The rice varieties used were the Samkwang variety (normal and hard type) and the Shingil variety (processing and soft type). The moisture content of the rice flour was adjusted to 35–55% and it was treated with the HHP treatment at 400–600 MPa. The water absorption capacity, solubility, and swelling power of the rice flour increased as the moisture levels and pressure increased. The 600 MPa enzymatic hydrolysis-treated rice flour showed similar results to the heat-treated rice flour. Scanning electron microscopy showed few cavities, resulting in a dense structure. X-ray diffraction confirmed that the 23° peak, which indicates the degree of gelatinization, decreased with increasing moisture levels and pressure. The HHP treatment of the rice flour changed its physicochemical properties according to the moisture levels and pressure applied. These results can provide important information on the development and production of various foods.

We examined the effect of chrysoeriol on adipogenesis and lipolysis and elucidated the underlying molecular mechanisms. Chrysoeriol inhibited fat deposition in adipocytes. Treatment with chrysoeriol suppressed the expression of peroxisome proliferator-activated receptor γ, fatty acid synthase, fatty acid-binding protein, CCAAT/enhancer-binding proteins (C/EBP) α, C/EBPβ, and sterol regulatory element-binding protein-1. In addition, chrysoeriol significantly elevated the activation of 5′-adenosine monophosphate-activated protein kinase. Moreover, chrysoeriol increased free glycerol and fatty acid levels and promoted lipolysis in adipocytes. Overexpression of adipose triglyceride lipase and hormone-sensitive lipase by chrysoeriol led to increased lipolysis in 3T3-L1 adipocytes. Taken together, chrysoeriol showed anti-adipogenic and lipolytic properties in adipocytes.

Phenolic acids, found in cereals, legumes, vegetables, and fruits, have various biological functions. We aimed to compare the antihypertensive potential of different phenolic acids by evaluating their ACE inhibitory activity and cytoprotective capacity in EA.hy 926 endothelial cells. In addition, we explored the mechanism underlying the antihypertensive activity of sinapic acid. Of all the phenolic acids studied, sinapic acid, caffeic acid, coumaric acid, and ferulic acid significantly inhibited ACE activity. Moreover, gallic acid, sinapic acid, and ferulic acid significantly enhanced intracellular NO production. Based on the results of GSH depletion, ROS production, and MDA level analyses, sinapic acid was selected to study the mechanism underlying the antihypertensive effect. Sinapic acid decreases endothelial dysfunction by enhancing the expression of antioxidant-related proteins. Sinapic acid increased phosphorylation of eNOS and Akt in a dose-dependent manner. These findings indicate the potential of sinapic acid as a treatment for hypertension.

Background: To overcome disadvantages of germinated brown rice, we germinated rough rice and tested effects of its useful ingredients on prevention of diabetes. Objective: This study investigated the in vitro antioxidant and in vivo antidiabetic effects of rough rice (Oryza sativa L.) with hulls, before and after germination. Rough rice was germinated for 4 days and extracted with water at 120°C. Design: This study measured antioxidants and antioxidative effects and inhibitory activities against α-amylase and α-glucosidase of rough rice before and after germination and investigated antidiabetic effects of rough rice through animal experiments. Results: All these factors increased after germination. Also, α-amylase and α-glucosidase inhibition and glucose uptake by 3T3-L1 adipocytes were significantly increased after germination. Oral administration of the germinated rough rice extract for 8 weeks significantly increased insulin levels and decreased blood glucose levels in a C57BLKS/J-db/db mice model. Immunohistochemical analysis showed that germinated rough rice effectively protected against liver, kidney, and pancreatic tissue damage. Discussion: Useful ingredients in germinated rough rice could be used to prevent diabetes. Conclusions: These results suggest that germinated rough rice extract had a beneficial effect on diabetes by increasing the antioxidant activity and further purification studies are necessary to elucidate the mechanism of the extract’s antidiabetic activity.

On-field in situ inspection of Marssonina coronaria infected apple blotch for apple leaf specimens was demonstrated using compact backpack-type optical coherence tomography (OCT). Simultaneously, the acquired OCT results were precisely matched with a widely used agricultural plant pathogen inspection technique called loop-mediated isothermal amplification (LAMP) to confirm on-field applicability of the compact backpack-type OCT. Specimens were examined for 8 weeks. Automated Amplitude-scan (A-scan) depth profiling and post-processed infected tissue boundary detection from acquired OCT images were performed to investigate the increase in the internal layer gaps of the leaf specimens resulting from the disease. Clearly identifiable morphological difference between healthy and infected-suspected specimens was observed through the OCT images, which were well correlated with LAMP results. OCT-LAMP correlations and confirmed feasibility study results conclude that the compact backpack-type OCT diagnosing modality can be effective and extensively applicable for various novel agricultural discoveries.

Whole wheat (Triticum aestivum L.) is a rich source of bioactive compounds with health benefits such as antioxidant, anti-inflammatory, and anti-proliferative activities. In this study, we evaluated the antioxidant and cytoprotective capacities of 41 wheat cultivars. The antioxidant capacities of whole wheat grain extracts (WWE) from various wheat cultivars were evaluated using total phenolic content and ABTS and DPPH radical-scavenging activities. The relative antioxidant capacity index (RACI) was calculated to integrate different chemical methods of antioxidant capacity. The cytoprotective capacity of the WWE was investigated using hepatocytes (HepG2), myoblasts (C2C12), and endothelial (EA.hy926) cells. The RACI was the highest and lowest for Dajoong (1.71) and Topdong (−1.96), respectively. Pearson correlation tests were conducted to determine the correlation between the antioxidant and cytoprotective activities. However, no significant correlations between RACI and the cytoprotective capacities were ascertained. Principal component analysis revealed that the first two components represented 68.93% of the total variability. Hierarchical cluster analysis classified WWE into three groups based on measured parameters. The results of this study highlight the variation in the antioxidant and cytoprotective capacities of 41 different wheat cultivars. This study provides basic information that can guide decisions in wheat breeding programs for the development of functional food crops and new dietary ingredients.

We focused on the functional components, antioxidant activity, skin-whitening, and anti-wrinkle properties of subcritical and supercritical water (SCW)-treated rutin. Rutin treatments were performed at the following temperature and pressure conditions: 200 °C/15 bar, 300 °C/100 bar, and 400 °C/250 bar. ABTS and DPPH radical scavenging activities and reducing power presented their highest values (1193.72 mg AAE/g, 728.73 mg AAE/g, and 0.65, respectively) at 300 °C/100 bar. The tyrosinase inhibitory activity of SCW-treated rutin was 21.72–60.05% at 1 mg/mL. The ethyl acetate fraction showed 14.91% melanin inhibitory activity at a concentration of 10 µg/mL compared to the α-MSH treatment group. The protein expression inhibition rates of MITF, tyrosinase, TRP-1, and TRP-2 in the ethyl acetate fractions were 14.05%, 72%, 93.05%, and 53.44%, respectively, at a concentration of 10 µg/mL, compared to the control. These results indicate that SCW treatment could be used to develop cosmetic materials and functional food with physiological activity, and that SCW-treated rutin can be used as a skin-whitening cosmetic material.

This study investigated the physicochemical properties of rice flour suspensions under ultrahigh hydrostatic pressure (UHP) treatment. Rice flour suspensions were subjected to 200, 400, and 600 MPa of pressure for 10 min, and heat treatment was used as a control. Proximate characteristics of different rice cultivar were analyzed to amylose, damage starch content, and particle size. Changes in physicochemical properties of rice flour suspensions according to UHP treatment were analyzed to microscopic structure, iodine reaction, α-amylase hydrolysis rate, and resistant starch content. Microscopic structural analyses showed that the structures of the rice flours were altered under both heat and 600 MPa treatment conditions. Water absorption rates were highest under heat treatment (467.53–554.85%), followed by 600 MPa treatment (269.55–334.57%). Iodine reaction values increased with increasing applied pressure. α-Amylase hydrolysis rates and resistant starch contents were highest under heat treatment and increased with increasing applied pressure. Based on these results, 600 MPa treatment of rice flour suspensions was shown to be comparable to heat treatment; as a result, the development of the new rice processing method with different physicochemical properties is expected from rice cultivars treated under UHP processing methods.

Background: Heat treatments are applied to ginseng products in order to improve physiological activities through the conversion of ginsenosides, which are key bioactive components. During heat treatment, organic acids can affect ginsenoside conversion. Therefore, the influence of organic acids during heat treatment should be considered. Methods: Raw ginseng, crude saponin, and ginsenoside Rb1 standard with different organic acids were treated at 130°C, and the chemical components, including ginsenosides and organic acids, were analyzed. Results: The organic acid content in raw ginseng was 5.55%. Organic acids were not detected in crude saponin that was not subjected to heat treatment, whereas organic acids were found in crude saponin subjected to heat treatment. Major ginsenosides (Rb1, Re, and Rg1) in ginseng and crude saponin were converted to minor ginsenosides at 130°C; the ginsenoside Rb1 standard was very stable in the absence of organic acids and was converted into minor ginsenosides in the presence of organic acids at high temperatures. Conclusion: The major factor affecting ginsenoside conversion was organic acids in ginseng. Therefore, the organic acid content as well as ginsenoside content and processing conditions should be considered important factors affecting the quality of ginseng products. In this study, the effects of sucrose on nutritional and functional composition as well as antioxidant activities in buckwheat sprouts during germination were investigated. Treatment with 1%, 2%, or 3% sucrose resulted in an increase in the amount of vitamins, four C-glycosylflavones, and rutin. Tocopherols and β-carotene in sprouts treated with 1%, 2%, and 3% sucrose were increased in a dose-dependent manner. Similarly, the sum of individual flavonoid was 1.35, 1.29, and 1.60 times higher than that of control sprouts. Sucrose treatment increased tyrosine and phenylalanine ammonia-lyase activities resulting in an increase in the flavonoid content. Moreover, treatment of HepG2 cells with buckwheat sprouts ameliorated oxidative damages by modulating reactive oxygen species production, malondialdehyde generation, and antioxidant enzyme activities. These results suggest that sucrose treatment could improve the nutritional composition and antioxidant effects of germinating buckwheat sprouts.

The objective of this study was to investigate the protective effect of esculetin against ethanol-induced liver injury in both HepG2 cells and mice. Treatment with esculetin significantly prevented ethanol-induced cytotoxicity in HepG2 cells by reducing the levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST). This protective effect was likely associated with antioxidant potential of esculetin, as evidenced by the attenuation of reactive oxygen species (ROS) and malondialdehyde (MDA) production and restoration of the depleted glutathione (GSH) levels in ethanol-induced HepG2 cells. Esculetin also dramatically increased the nuclear translocation of NF-E2-related factor 2 (Nrf2) and antioxidant response element (ARE) reporter gene activity, which led to the upregulation of ARE-responsive enzymes. In animal studies, esculetin markedly prevented ethanol-induced liver injury by reducing GSH depletion, MDA production, and the lipid contents in liver. Moreover, esculetin lowered the levels of serum ALT, serum AST, and the content of serum lipids.

Mice and rats are rodent specimens commonly used in multidisciplinary research. Specifically, vasculature imaging of rodents has been widely performed in preclinical studies using various techniques, such as computed tomography, magnetic resonance imaging, and ultrasound imaging. Photoacoustic CT (PACT) is a noninvasive, nonionizing optical imaging technique derived from photoacoustic tomography and benefits from using intrinsic endogenous contrast agents to produce three-dimensional volumetric data from images. In this study, a commercial PACT device was employed to assess the cervicothoracic vasculature of mouse and rat specimens, which has rarely been examined using PACT, under two conditions with depilation and skin incision. Various blood vessels, including the common carotid artery, internal/external jugular veins, cranial vena cava, internal thoracic vein, and mammary, were identified in the acquired PACT images. The difference between the depilated and skin-incised specimens also revealed the presence of branches from certain blood vessels and specific anatomical features such as the manubrium of the sternum. This study presents detailed PACT images observing the cervicothoracic vasculature of rodent specimens and is expected to be used as a reference for various preclinical experiments on mice and rats.

This study aimed to evaluate the effects of taxifolin and sorghum ethanol extract on free fatty acid (FFA)-induced hepatic insulin resistance. FFA treatment decreased glucose uptake by 16.2% compared with that in the control, whereas taxifolin and sorghum ethanol extract increased the glucose uptake. Additionally, taxifolin and sorghum ethanol extract increased the expression of p-PI3K, p-IRS1, p-AKT, p-AMPK, and p-ACC in FFA-induced hepatocytes. Furthermore, FFA treatment increased the expression of miR-195. However, compared with the FFA treatment, treatment with taxifolin and sorghum ethanol extract decreased miR-195 expression in a dose-dependent manner. Taxifolin and sorghum ethanol extract enhanced p-IRS1, p-PI3K, p-AMPK, p-AKT, and p-ACC expression by suppressing miR-195 levels in miR-195 mimic- or inhibitor-transfected cells. These results indicate that taxifolin and sorghum ethanol extract attenuate insulin resistance by regulating miR-195 expression, which suggests that taxifolin and sorghum ethanol extract may be useful antidiabetic agents.

Photoacoustic imaging (PAI) is a hybrid non-invasive imaging technique used to merge high optical contrast and high acoustic resolution in deep tissue. PAI has been extensively developed by utilizing its advantages that include deep imaging depth, high resolution, and label-free imaging. As a representative implementation of PAI, photoacoustic microscopy (PAM) has been used in preclinical and clinical studies for its micron-scale spatial resolution capability with high optical absorption contrast. Several handheld and portable PAM systems have been developed that improve its applicability to several fields, making it versatile. In this study, we developed a laboratory-customized, two-axis, waterproof, galvanometer scanner-based handheld PAM (WP-GVS-HH-PAM), which provides an extended field of view (14.5 × 9 mm2) for wide-range imaging. The fully waterproof handheld probe enables free movement for imaging regardless of sample shape, and volume rate and scanning region are adjustable per experimental conditions. Results of WP-GVS-HH-PAM-based phantom and in vivo imaging of mouse tissues (ear, iris, and brain) confirm the feasibility and applicability of our system as an imaging modality for various biomedical applications.

Amaranthus species are widely used as grain and leaf vegetables around the world and are potential dietary sources of antioxidants and phenolic compounds. In this study, we examined the variation in total flavonoid contents, total polyphenol contents, and antioxidant activities among 120 accessions of nine Amaranthus species. The antioxidant activity of DPPH (2,2-diphenyl-1-picrylhydrazyl) of the 120 amaranth accessions ranged from 1.1 (A. tricolor) to 75.2 (A. tricolor mg ascorbic acid equivalents (AAE)/g in 2018, and 8.5 (A. tricolor) to 68.8 (A. dubius) mg AAE/g in 2019. ABTS (2,2′-azinobis (3-ethylbenzothiazoline 6-sulfonate)) antioxidant activity ranged from 16.7 (A. tricolor) to 78.3 (A. hypochondriacus) mg AAE/g in 2018, and 36.6 (A. tricolor) to 79.2 (A. dubius) mg AAE/g in 2019. Total flavonoid content (TFC) of 2018 and 2019 ranged from 21.7 (A. caudatus) to 52.7 (A. hybridus) and from 22.3 (A. viridis) to 54.7 (A. tricolor), respectively Antioxidant activities were compared using two methods and all components were measured in plants grown both in 2018 and 2019. We identified wide variation among the accessions and between plants grown in the two years. Antioxidant activities and phytochemical contents were consistently negatively correlated. The nine species and 120 accessions clustered into three groups according to their antioxidant activities, total flavonoid contents, and total polyphenol contents in each year. These results provide information about the nutritional profiles of different Amaranthus species.

Ginseng seeds are rich in phytosterols, ginsenosides, and fatty acids, and can therefore be used in skincare to delay the aging process. Ginseng seed embryo (GSE) and ginseng seed coat (GSC) were separated from ginseng seeds (Panax ginseng Meyer). This study evaluated the protective activity and underlying mechanism of GSE and GSC on UVB irradiation-induced skin photoaging using Hs68 cells. Their bioactive compounds, including phytosterols, ginsenosides, tocopherols, tocotrienols, and fatty acids were determined by HPLC and GC. The levels of reactive oxygen species, matrix metalloproteinases (MMPs), and collagen levels were measured in human dermal fibroblast cell line, Hs68 cells. The antioxidant capacity and contents of total polyphenols and flavonoids were higher in GSC than those in GSE. Linoleic acid was the major fatty acid in both GSE and GSC. GSE and GSC treatment alleviated UVB-induced increase of reactive oxygen species (ROS), matrix metalloproteinase (MMP)-1, and MMP-3, resulting in reduced collagen degradation. Increased UVB-mediated phosphorylation of mitogen activated protein kinase (MAPK) and activator protein-1 (AP-1) was inhibited by GSE and GSC treatment. Moreover, GSE and GSC effectively upregulated transforming growth factor-β (TGF-β) 1 levels. It was found that ginseng seeds regulate the expression of TGF-β/Smad and MAPK/AP-1 pathways. Ginseng seeds contain various bioactive compounds and have protective activity against UVB-induced skin photoaging. Therefore, ginseng seeds have the potential for use in cosmeceutical preparations.

The anti-adipogenic mechanism of capsicoside G in 3T3-L1 adipocytes was elucidated for the first time. Capsicoside G inhibited in a dose-dependent manner (1–20 µg/mL) the accumulation of lipid droplets in adipocytes without cytotoxicity. The major effects of capsicoside G on the inhibition of adipocyte differentiation were achieved by the early stage of adipogenesis. Treatment with capsicoside G inhibited the expression of the major adipogenic transcription factors and their target genes during differentiation of preadipocytes into adipocytes. Moreover, capsicoside G treatment significantly increased phosphorylation of 5′-adenosine monophosphate-activated protein kinase (AMPK) α1, thereby modulating fatty acid metabolism. In addition, the anti-adipogenic activity of capsicoside G was mediated by the activation of AMPK, as confirmed by using compound C, a highly selective inhibitor of AMPK, and the AMPKα1 small interfering RNA system. Capsicoside G from pepper seeds may have potential in the treatment of obesity.

Depth-visualizing sensitivity can be degraded due to imperfect optical alignment and non-equidistant distribution of optical signals in the pixel array, which requires a measurement of the re-sampling process. To enhance this depth-visualizing sensitivity, reference and sample arm-channeled spectra corresponding to different depths using mirrors were obtained to calibrate the spectrum sampling prior to Fourier transformation. During the process, eight interferogram patterns corresponding to point spread function (PSF) signals at eight optical path length differences were acquired. To calibrate the spectrum, generated intensity points of the original interferogram were re-indexed towards a maximum intensity range, and these interferogram re-indexing points were employed to generate a new lookup table. The entire software-based process consists of eight consecutive steps. Experimental results revealed that the proposed method can achieve images with a high depth-visualizing sensitivity. Furthermore, the results validate the proposed method as a rapidly performable spectral calibration technique, and the real-time images acquired using our technique confirm the simplicity and applicability of the method to existing optical coherence tomography (OCT) systems. The sensitivity roll-off prior to the spectral calibration was measured as 28 dB and it was halved after the calibration process.

Background: The current typical drying methods for red ginseng are sun drying and hot-air drying. The purpose of this study was to investigate drying characteristics of red ginseng by using far-infrared drying. Methods: The far-infrared drying tests on red ginseng were conducted at two drying stages: (1) high temperature for 24 h drying and (2) low temperature drying until the final moisture content was 13 ± 0.5% (wet basis). The high temperature drying stage included three drying chamber temperature conditions of 60°C, 65°C, and 70°C. The low temperature drying stage was conducted at temperatures of 45°C and 50°C. Drying characteristics were analyzed based on factors such as drying rate, color changes, energy consumption, and saponin content. The results were compared with those of the hot-air and sun drying methods. Results: The results revealed that increases in drying temperature caused a decrease in drying time and energy consumption for far-infrared drying. The saponin content decreased under all drying conditions after drying, the highest value (11.34 mg/g) was observed at drying conditions of 60∼50°C. The sun drying condition showed the lowest color difference value when compared with far-infrared and hot-air drying. Conclusion: The far-infrared drying showed a faster drying rate, higher saponin content, lower color difference value, and a decrease in energy consumption than seen in hot-air drying.

The extracts of pepper (Capsicum annuum L.) seeds are known to inhibit differentiation and adipogenesis of 3T3-L1 preadipocytes. In this study, bioassay-guided fractionation of a crude ethanolic extract from defatted pepper seeds led to the isolation of two anti-adipogenic furostanol saponins, capsicoside A and capsicoside G. The anti-adipogenic activity of these compounds was evaluated by measuring their inhibitory effect on lipid accumulation in 3T3-L1 adipocytes. The chemical structures of the pure compounds were elucidated using spectroscopic methods (1H, 13C NMR, and ESI-MS) and compared with the available data in the literature. The results of activity screening showed that capsicosides A and G (1–20 µg/mL) reduced intracellular lipid accumulation in 3T3-L1 adipocytes in a dose-dependent manner without causing cytotoxicity. The inhibitory effect of capsicoside G at 20 µg/mL on the lipid accumulation during 3T3-L1 adipocyte differentiation was significantly greater than that of capsicoside A at the same concentration. These results suggest that capsicoside G isolated from pepper seeds may prevent obesity.

Obesity is one of the most serious health problems in both Westernized and developing countries. AMP-activated protein kinase (AMPK) has emerged as a major regulator of appetite, body weight, and cellular energy balance. In this study, we investigated the effect of esculetin (ECT) on adipogenesis via activation of AMPK in 3T3-L1 cells. ECT markedly inhibited lipid accumulation and suppressed the expression of adipogenic specific proteins including peroxisome proliferator-activated receptors (PPARγ), CCAAT/enhancer binding protein a (C/EBPα), and adipocyte fatty acid binding protein (aP2). Moreover, ECT significantly increased phosphorylation of AMPK and acetyl-CoA carboxylase (ACC) and intracellular reactive oxygen species (ROS) production. However, pretreatment with compound C, a specific AMPK inhibitor, abolished the inhibitory effects of ECT on PPARγ and C/EBPα expression. Therefore, these results indicate that ECT has anti-adipogenic effects through modulation of PPARγ and C/EBPα via the AMPK signaling pathway.

The present study was conducted to evaluate the antioxidant and anti-inflammatory activities of crude methanolic extract of Abeliophyllum distichum Nakai, and those of its partitioned fractions, including hexane, ethyl acetate, n-butanol, and aqueous. The antioxidant activities were analyzed by DPPH free radical scavenging and oxygen radical antioxidant capacity assay. Results showed that the BuOH fraction possessed a strong antioxidant activity through a hydrogen atom transfer reaction-based mechanism and a single electron transfer reaction-based mechanism. In lipopolysaccharide (LPS)-stimulated RAW 264.7 cells, the BuOH fraction of A. distichum methanol extract exhibited a strong inhibitory effect on the nitric oxide production and inhibited the expression of pro-inflammatory mediators, including COX-2, TNF-α, and IL-6, through the inhibition of the MEK/ERK signaling pathway. In addition, the BuOH fraction inhibited the LPS-induced ROS level through the NADPH oxidase-independent mechanism. Furthermore, HPLC analysis identified chlorogenic acid, caffeic acid, gentisic acid, rutin, ferulic acid, and quercetin, and suggested that the antioxidant and anti-inflammatory activities of the BuOH fraction should be mediated by the presence of higher amounts of caffeic acid, rutin, and ferulic acid than other fractions. Taken together, these results suggest that A. distichum extract is a source of antioxidant and anti-inflammatory compounds, and could be developed as a potential source for functional food and dietary health supplement.

Background: This study evaluated changes in ginsenoside compositions and antioxidant activities in hydroponic-cultured ginseng roots (HGR) and leaves (HGL) with heating temperature. Methods: Heat treatment was performed at temperatures of 90°C, 110°C, 130°C, and 150°C for 2 hours. Results: The ginsenoside content varied significantly with heating temperature. The levels of ginsenosides Rg1 and Re in HGR decreased with increasing heating temperature. Ginsenosides F2, F4, Rk3, Rh4, Rg3 (S form), Rg3 (R form), Rk1, and Rg5, which were absent in the raw ginseng, were formed after heat treatment. The levels of ginsenosides Rg1, Re, Rf, and Rb1 in HGL decreased with increasing heating temperature. Conversely, ginsenosides Rk3, Rh4, Rg3 (R form), Rk1, and Rg5 increased with increasing heating temperature. In addition, ginsenoside contents of heated HGL were slightly higher than those of HGR. The highest extraction yield was 14.39% at 130°C, whereas the lowest value was 10.30% at 150°C. After heating, polyphenol contents of HGR and HGL increased from 0.43 mg gallic acid equivalent/g (mg GAE eq/g) and 0.74 mg GAE eq/g to 6.16 mg GAE eq/g and 2.86 mg GAE eq/g, respectively. Conclusion: Antioxidant activities of HGR and HGL, measured by 1,1-diphenyl-2-picrylhydrazyl and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging ability, increased with increasing heating temperature. These results may aid in improving the biological activity and quality of ginseng subjected to heat treatments.

Chrysanthemum zawadskii var. latilobum (CZ) has been widely used as a tea in Korea. In this study, we investigated the involvement of anti-inflammatory haem oxygenase-1 (HO-1) in the inhibitory activity of a CZ leaf extracts on nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. At the highest concentration of CZ leaf extract (50 μg/mL), NO production was 10% of that in LPS-stimulated macrophages, whereas iNOS protein expression was 5% of that in LPS-stimulated macrophages. CZ leaf extract also inhibited iNOS mRNA expression in a concentration-dependant manner, and at 50 μg/mL, iNOS mRNA expression was approximately 15% of that in LPS-stimulated cells. The CZ leaf extract induced HO-1 expression in a dose-dependent manner, and blocking HO-1 activity abolished the anti-inflammatory effect of CZ leaf extract. These results suggest that CZ leaf extract has a potent anti-inflammatory activity in RAW264.7 cells through the induction of HO-1.
