The aim of this study was to investigate the biological function and molecular mechanism of ARPC1A (actin related protein 2/3 complex subunit 1A) in prostate cancer progression. RT-qPCR and IHC results showed that the level of ARPC1A in prostate cancer tissues was significantly higher than that in adjacent tissues. The results of TCGA (the cancer genome atlas) database analysis showed that high expression of ARPC1A indicates poor prognosis in prostate cancer patients. In vitro functional experiments confirmed that downregulation of ARPC1A expression resulted in decreased cell viability and invasive ability of prostate cancer cells, as ARPC1A knockdown promoted ferroptosis. The transcriptional regulation mechanism of STAT3 (signal transduction and activators of transcription 3) on ARPC1A was elucidated by Co-IP, ChIP and luciferase reporter assays. In vivo experiments also supported the in vitro results. We propose that reduced ARPC1A expression inhibits prostate cancer cell viability and invasion in a ferroptotic manner. The ARPC1A level may serve as an independent predictor of prognosis in prostate cancer patients.

This study sought to provide contemporary data from a multi-institution with respect to DNA-repair genes (DRGs) status and its impact on effects of platinum-based chemotherapy in treatment-emergent neuroendocrine prostate cancer (t-NEPC), for which little data exist.All patients were retrospectively collected with eligible biopsied tissues for targeted next generation sequencing (NGS). The main outcomes were radiologic progression-free survival and overall survival according to Response Evaluation Criteria in Solid Tumors, version 1.1.Among the 43 NEPC patients, 13/43 (30%) harbored homozygous deletions, deleterious mutations, or both in DRGs. Eleven patients (11/13, 85%) with DRGs aberrations had effective response, including 7 patients with BRCA1/2 defects and 2 with mismatch repair-deficient caused by MSH2 alterations. While significantly fewer responders (30%) were detected in patients without DRGs aberrations (odds ratio = 12.83, p = 0.003). Compared with patients without genomic DRGs aberrations, the hazard ratio (HR) for radiologic progression in those with DRGs defects was 0.42 (95% confidence interval [CI]: 0.19-0.93), and the HR for death was 0.65 (95% CI: 0.24-1.72). The most common adverse event of Grade 3 or 4 was anemia, as noted in 7 patients (16%).The DRGs status is therapeutically meaningful in t-NEPC. Given the potential responses to platinum-based chemotherapy, our findings support the clinical use of NGS in t-NEPC patients to identify DRGs aberrations.

Bladder cancer is the most common malignant tumor of the urinary system, and in China it is first among urogenital system tumors. More therapeutic targets are still urgently required to combat this disease. Lamin B2 (LMNB2) is a type of nuclear lamina filament protein, which is involved in multiple cellular processes, and known as an oncogene affecting the progression of multiple types of cancers. Although the multiple effects of LMNB2 on cancer progression have been elucidated, its possible role in bladder cancer remains unclear. In the present study, it was determined that LMNB2 expression was upregulated in human bladder cancer tissues, and its expression was correlated with the prognosis and the clinical features, including tumor stage (P=0.001) and recurrence (P=0.006) of patients with bladder cancer. In addition, it was further revealed that LMNB2 depletion inhibited bladder cancer cell proliferation, stimulated cell cycle arrest and apoptosis

Cell division cycle-associated 5 (CDCA5) can regulate cell cycle-related proteins to promote the proliferation of cancer cells. The purpose of the present study was to investigate the expression level of CDCA5 in prostate cancer (PCa) and its effect on PCa progression. The signalling pathway by which CDCA5 functions through was also attempted to elucidate. Clinical specimens of PCa patients were collected from the Second Hospital of Tianjin Medical University. The expression level of CDCA5 in cancer tissues and paracancerous tissues from PCa patients was detected by RT-qPCR analysis and IHC. The relationship between the expression level of CDCA5 and the survival rate of PCa patients was analysed using TCGA database. Two stable cell lines (C4-2 and PC-3) with CDCA5 knockdown were established, and the effects of CDCA5 on PCa cell proliferation were detected by MTT and colony formation assays. Flow cytometry was performed to detect the effect of CDCA5 on the PCa cell division cycle, and western blot analysis was used to determine changes in ERK phosphorylation levels after CDCA5 knockdown. The effect of CDCA5 expression on prostate tumour growth was assessed using a mouse xenograft model. The results revealed that the mRNA and protein expression levels of CDCA5 were significantly higher in PCa tissues than in paracancerous tissues. High CDCA5 expression was associated with the prognosis of patients with PCa. CDCA5 expression knockdown significantly reduced the number of PCa cells in mitoses and inhibited their proliferation in vitro and in vivo. When CDCA5 was knocked down, the phosphorylation level of ERK was also reduced. Collectively, CDCA5 was upregulated and affected the prognosis of patients with PCa. Decreased CDCA5 expression inhibited PCa cell proliferation by inhibiting the ERK signalling pathway. Thus, CDCA5 may be a potential therapeutic target for PCa.

It is important to acquire the impedance of switched-mode power supply (SMPS) under its operating condition for designing electromagnetic interference (EMI) filter. The online impedance extraction technique has become a research hot topic. However, the result of current online measurement technique is inaccurate due to ignorance of the different impedance of two power lines to ground of SMPS. To cope with this, an online impedance extraction technique is proposed in this paper. Considering the practical characteristics of impedance of two power lines to ground, the extraction principle and method is developed based on the established measurement models. Finally, the measurement platform with SMPS and measurement device under operating condition is built. Experimental results validate that the proposed technique can extract the differential mode (DM) and the impedance of two power lines to ground.

Smaller size and lower loss are always the target of EMI filter which suppresses EMI emitted by power electronics converter. Digital EMI filter (DEF) is the best solution to suppress the conducted EMI in size and loss, especially for high-power converters. However, for current DEF model, it is not enough to describe the filtering ability. The model which can describe DEF stability is absent. To cope with these, this paper proposes a modeling technique about filter and stable behavior of DEF system. In proposed technique, the insertion gain model can accurately describe the filtering behavior, and the loop gain model can predict the stability of DEF system. Experimental testbed based on a Boost converter as EMI interference is built. Experimental results validate that the proposed model can successfully predict the stability and performance of DEF system.

Developing progress of virtual simulation experiment (VSE) driven by project data for the course of Comprehensive Practice of Electric Power System (CPEPS) of Electrical Engineering and the impacts of the experiment on motivation and innovation of students at Xi’an University of Technology are described. The participants were not just teachers, but also students and engineers in companies. The algorithms, data and project cases came from research accumulated by teachers and real engineering cases supplied by companies. Comparing the online teaching efficiency between CPEPS with online experiment and other courses without any experiment during 2019-nConV, it indicted that online lectures with VSE can decrease challenges for students and teachers, and can stimulate students’ study interests and motivation. Meanwhile, adopted encouraging and inductive teaching method, undergraduates’ self-learning abilities and capacity of innovation were improved.

Traditional alternating-current-driven electroluminescent (AC-EL) devices adopting a sandwich structure are commonly used in solid-state lighting and displays, while the emerging coplanar-electrode alternating-current-driven light-emitting variants manifest excellent application prospects in intelligent, multifunctional, and full-color displays, and sensing purposes. In this work, an asymmetrically enhanced coplanar-electrode AC-EL device with a universal and straightforward architecture is designed based on the impedance adjustment strategy. This newly devised asymmetric structure extends the functionalities of the coplanar-electrode AC-EL devices by overcoming the bottlenecks of complicated patterning procedures and high driving voltages of symmetric configuration. The developed device design enables a new type of information encryption and ultrahighly stretchable patterned displays. Notably, the novel encryption appliances demonstrate feasible encryption/decryption features, multiple encryptions, and practical applicability; the biaxially stretchable display devices achieve the highest tensile performance in the field of stretchable electroluminescent pattern displays, and outperform the ultrahighly stretchable sandwich devices in terms of simple patterning process, higher brightness, wider color gamut, and long-term stability. The proposed configuration opens up new avenues for AC-EL devices toward a plethora of smart applications in wearable electronics with intelligent displays, dynamic interaction of human-machine interface, and soft robotics.

In order to investigate the hypolipidaemic and antioxidant effects of various dark teas produced from different post-fermentation using the same raw material, a hyperlipidaemia zebrafish model combined with binding bile salts assay and antioxidant assays were performed in this study. Results showed that the hypolipidaemic effect of dark tea extracts increased significantly (p 0.05) while the antioxidant ability decreased sharply compared with raw material. Particularly, Liupao tea (50%) and Pu-erh tea (48%) showed promising hypolipidaemic potential; however, the antioxidant capacity of Pu-erh tea decreased (31-49%) most dramatically. Besides, the levels of total polyphenols and catechins decreased sharply, but theabrownin, gallic acid, and caffeine increased significantly after post-fermentation. Moreover, the potential mechanisms of regulating hyperlipidaemia by dark tea extracts were discussed. These results suggest that microbial fermentation significantly affects the bioactivity of dark teas, and provide theoretical basis for processing and improving of dark tea products for hyperlipidaemia therapy.

Current power supply networks across the world are mostly based on three-phase electrical systems as an efficient and economical way for generation, transmission and distribution of electricity. Now, many electrically driven devices are relying on direct current or single-phase alternating current power supply that complicates utilization of three-phase power supply by requiring additional elements and costly switching mechanisms in the circuits. For example, light-emitting devices, which are now widely used for displays, solid-state lighting etc. typically operate with direct current power sources, although single-phase alternating current driven light-emitting devices have also gained significant attention in the recent years. Yet, light-emitting devices directly driven by a three-phase electric power has never been reported before. Benefiting from our precious work on coplanar electrodes structured light-emitting devices, in this article we demonstrate proof of a concept that light-emitting components can be driven by three-phase electric power without utilizing intricate back-end circuits and can compose state detection sensors and pixel units in a single device inspiring from three primary colors. Here we report a three-phase electric power driven electroluminescent devices fabricated featuring of flexibility and multi-functions. The design consists of three coplanar electrodes with dielectric layer(s) and light emission layer(s) coated on a top of input electrodes. It does not require transparent electrodes for electrical input and the light emission occurs when the top light-emitting layers are connected through a polar bridge. We demonstrate some applications of our three-phase electric power driven electroluminescent devices to realize pixel units, interactive rewritable displays and optical-output sensors. Sliding mode observer (SMO) which has a strong robustness is used in the sensorless control system of permanent magnet synchronous motor (PMSM). The method is usually used to estimate the rotor position although it has the chattering problem. According to the causes of the chattering in traditional SMO, the method of improved sliding mode observer (ISMO) is proposed in this paper. Sign function is replaced by the saturation function to suppress the chattering problem, and the speed of PMSM is estimate when combined with field oriented control (FOC) method. A hardware platform of 2.7 kW, 10 000 r/min PMSM is built to verify the control effectiveness of ISMO. Simulation analysis and experimental result show that ISMO can estimate the rotor speed accuracy and reduce the chatting at the same time.

Arc welding inverter supply with switch and digital control is widely used, due to the advantages of high controllability, small size, and high efficiency. With the constant increasing of switching frequency and decreasing of arc welding machine size, the conducted electromagnetic interference (EMI) becomes serious, especially for common mode (CM)-conducted EMI. Meanwhile, as the development of digital processing technique, digital active EMI filter (DAEF) technology, which uses digital control and embedded mode, has become a hot research topic. However, the conventional DAEF fails to consider time delay effect. Its model cannot accurately describe DAEF filtering behavior. Moreover, decoupling circuit lacks scientific design method. To cope with these, this paper proposes a precise DAEF model which considers both the time delay of digital processing portion and the parasitic parameters of passive components. Moreover, based on impedance matching principle, a detailed and scientific design method of decoupling circuit is put forward. Meanwhile, the arc welding inverter system with embedded DAEF is designed, modeled, and compensated. Finally, experiment results show that the proposed precise model can predict the system filtering performance accurately. And the proposed design method of decoupling circuit is very effective to suppress the CM-conducted EMI. The embedded DAEF can improve system stability and dynamic performance.

Purpose. KIF20A is essential in the process of spindle assembly and cytokinesis regulation. The role of KIF20A during tumorigenesis and tumor development has been well studied in several cancers. But the association between the KIF20A clinical role and prostate cancer (PCa) has not been reported yet. In this study, we investigated its potential prognostic effect and its role in progression of prostate cancer. Methods. Real-time quantitative polymerase chain reaction and Western blots were used to investigate the KIF20A transcription and translation levels in 7 pairs of fresh PCa tissue and adjacent normal prostate tissue. Immunohistochemistry (IHC) was used to investigate the KIF20A protein level in 114 PCa tissue samples. Bioinformatics analysis was performed to analyze the effect of KIF20A in oncologic prognosis in PCa patients. MTT assay, transwell assay, and colony formation assay in vitro and tumor formation assay in vivo were performed to evaluate the biological behavior of KIF20A in prostate cancer. Results. KIF20A was significantly elevated in tumor tissue compared with normal prostate tissue at both the mRNA and the protein level. High expression of KIF20A at the protein level was correlated with adverse clinicopathological features. Bioinformatics analysis showed that the high KIF20A expression group has a poor biochemical recurrence- (BCR-) free survival. Knocking down KIF20A suppressed the proliferation, migration, and invasion of the prostate cancer cell both in vitro and in vivo. Conclusions. Our data demonstrated that the high expression of KIF20A was associated with poor clinical outcome and targeting KIF20A could reduce proliferation, migration, and invasion of the prostate cancer cell, indicating that KIF20A might be a potential prognostic and therapeutic target for PCa patients.

A circuit topology of high voltage DC pulse dust removal power supply which can remove wide-diameter dust is proposed to solve the problem of the present high voltage dust removal power supply to fine particle dust removal effect is not great, and can't overcome the current situation of anti-corona problem under the condition of high resistivity power. The circuit topology adopts double voltage rectifier to realize DC high frequency base voltage, and pulse inversion to realize high voltage superimposed pulse. The circuit topology of the power supply can not only improve the dust removal effect of fine particle dust, but also suppress the phenomenon of anti-corona problem under the condition of high resistivity power. Meanwhile, the simulation model of the proposed circuit topology of the power supply is established, and the waveform control strategy is verified. Finally, the prototype of the power supply system is built to realize the topology of the proposed power supply circuit, and the dust removal effect of the proposed topology is verified by experiments.

Development of alternating current (AC)-driven devices with versatile architecture is a potential approach to realize multifunctional light-emitting sources. Planar AC-powered electroluminescent (pAC-EL) devices using lateral placement of electrodes, instead of conventional stacking, are an emerging design that manifests promising applications beyond displays. While phosphors and organic light-emitting materials have been applied in pAC-EL devices, further enhancing the color purity and brightness remains a daunting challenge. In this Letter, we explore the utilization of quantum dots as the emitting layer for pAC-EL single-insulation devices without external injection. In such architecture, light is produced by the recombination of internally generated holes and field-induced electrons in the emissive layer of two in-plane light-emitting units alternately. The developed pAC-QLEDs exhibited a maximum brightness of 2023, 6327, and 613 cd/m2 for red, green, and blue (RGB) emissions at 150 kHz, respectively. Furthermore, a white-emitting pAC-QLED and a bi-color pAC-QLED were also constructed by stacking the R/G/B QD layers in serial and putting the R/G QD layers in parallel, respectively. In addition, compatibility of the proposed device configuration with flexible substrates is also manifested. The development of pAC-QLEDs provides an effective route to achieve high brightness without external injection, indicating diverse applications of these light sources for health monitoring sensors and phototherapy.

With the development of China high-speed train technology, high-speed train has entered the mass production stage nowadays. But the EMI control technology for the assembled overall-train is immature, and the EMI mechanism of the overall-train is not clear enough as well. In this paper, the EMI control is not ideal after assembly of China standardized high-speed train, and this paper studies the EMI mechanism of the train. An EMI model for high-speed train has been established. The model which have direct meaning to the EMI problem in the present stage of the standardized train group can be predict the EMI characteristics of the overall-train accurately.

The high frequency high voltage (HFHV) transformer is one of the biggest contributors for conducted electromagnetic interference (EMI) emission of switching power supply. The transformer technologies for reducing conducted EMI emission focus on the shielding and changing the winding structures. However, for suppressing conducted EMI, the suppression capability of shielding is limited; current methods of changing the winding structures don't fully reduce all the distributed capacitance, especially in the HFHV switching power supply. This paper proposes a novel winding method to suppress the conducted EMI. The proposed method not only considers the distributed capacitance between the primary winding and secondary winding, the interlayer distributed capacitance of the primary or secondary winding, but also considers the interturn distributed capacitance of the winding. Experimental testbed based on 1kV DC switching power supply is built. Simulation and experimental results show the proposed winding method is very effective in reducing conducted EMI emission.

For suppressing of the conducted electromagnetic interference (EMI), passive EMI filter (PEF) is the most widely applied technique at present. The near-field coupling due to PEF passive components layout has great influence on filtering performance. Currently, the layout of PEF components completely dependents on the experience of EMC engineer. However, there is a lack in systematic design method and scientific design basis. For this reason, in this paper, an optimization design method of PEF layout is presented. The method is based on distribution rule and mechanism of near-field coupling of PEF components, which can decrease volume and improve the filtering performance of PEF. Finally, theoretical analysis and experimental results show that the presented method of PEF layout can effectively improves the filtering performance of PEF.

As the switching frequency of switching power supply increases, its size become smaller and smaller. This phenomenon results in serious electromagnetic interference (EMI) problem. General passive EMI filter cannot meet the demand of size, weight, power losses and filter performance at the same time. Digital Active EMI Filters (DAEF) is a good choice, for it implement EMI suppression using digital and active. However, the cost-effectiveness of current DAEF is low. This paper proposes a novel switching period control method for the DAEF to reduce the cost and increase the efficiency of the whole EMI filtering system, which, in turn, improves the filtering efficiency of the DAEF. This paper gives detailed design procedure and parameter calculation of DAEF. Experimental testbed based on boost DC/DC converter is built. Simulation and experimental results of DAEF show the proposed switching period control is very effective in reducing cost and improving filtering performance of the DAEF system.

Digital high-frequency arc welding inverter is widely used in welding areas because of its many advantages such as high efficiency, small size, easy to control etc. However, with the development of high frequency of the switching, the conducted EMI generated by arc welding inverter becomes increasingly serious, especially common mode conducted EMI. Based on Digital Active EMI Filter (DAEF) controller, this paper proposes a new method which can reduce common mode conduction EMI for arc welding inverter. It is a closed EMI control loop by sensing EMI signal, sampling, digital compensation control, DAC output and injection. The EMI control can be embedded in original controller of arc welding inverter to reduce costs. Moreover, since there is no component in series connect with the power circuit, DAEF solves the problem of traditional EMI filter in filter size and power consumption fundamentally. In this paper, the arc welding inverter system with DAEF is designed, molded and compensated. Meanwhile, by simulation and analyses, the advantages of arc welding inverter with DAEF in system stability and dynamic performance are showed. Finally, experiment results showed that the proposed method has advantages in reducing conducted EMI emission and improving dynamic performance of arc welding inverter.

Digital Active EMI Filtering (DAEF) technique is a new direction of conductive EMI suppression technologies for switched-mode power converters. However, the conventional DAEF topology only suppresses EMI to ground on the AC (or DC) ports. It doesn't definitely aim to Common Mode (CM) or Differential Mode (DM) EMI. In this paper, a novel DAEF topology is proposed. It can flexibly suppress CM and DM EMI, and also effectively adjust EMI suppression ability as well as prevent interconversion between CM and DM EMI. On the other hand, because of the parasitic parameters of circuit and the time delay of controlling system, the traditional model of DAEF cannot describe the system behavior accurately. This paper proposes a precise model of DAEF system. The model not only considers the passive device parasitic parameters of sense circuit, injection circuit and decoupling circuit, but also considers the time delay of the digital control system. This model can accurately describe filter performance of DAEF. Finally, simulation and experimental results show that the proposed DAEF effectively suppress CM EMI. Moreover, the precise model can predict the system filter performance accurately.

Passive Power Filter (PPF) is widely used in power system because of its advantages in performance-price ratio. For PPF, there are several topologies, each of which has different frequency characteristic. In power system, in order to achieve the certain filtering effect, it is necessary to combine different filter topologies. Currently, the selection of PPF topologies is based on the engineering practical experience of PPF designer. However, there is a lack in systematic design method and scientific design basis. For this reason, this paper, taking the filtering effect and filters cost as the design indices, deeply investigates the selection problem of PPF topologies, and proposes a selection method of PPF topologies. The research results show that the most reasonable combination of different filter topologies can be obtained by this method for different situation needed to mitigate harmonic and compensate reactive power.

The Passive Power Filter (PPF) is widely used in power system because of its high performance-price ratio. Firstly, the design of PPF is to meet the requirements of filtering effect. Meanwhile it is restricted by the power factor, economic indicators and other conditions. Traditional design method, which is usually based on one economic and technical indexes or engineering experience, is too dependent on experience and unable to reach the optimum design. This paper present a design method of PPF based on Particle Swarm Optimization (PSO) algorithm, the initial investment cost of PPF, reactive power compensation capacity and the filtering effect are taken as three optimization goals. This method is validated by a simulation power system experiments. The experimental results show that the optimization design method of PPF is correct and effective. And the optimized PPF has good comprehensive performance.

Because the design process of TCR+FC type SVC device which is now widely used in power system, involves a lot of factors and a wide range of the relevant device parameters, and a lot of checking standards of device parameter, it is very big for the amount of design work which usually is repetitive, and its design cycle is very long, and its design cost is very high. The general parameters of SVC system, especially the FC filter network parameters, which strongly depend on power grid system parameter, need to be analyzed and verified, therefore, the parameters of SVC required to be optimized. As to the case of SVC, in order to solve the problem of the complicated design process, this paper develops a software platform of SVC system parameter optimization design and analysis. The interface of the software platform is concise and friendly, and involves an area widely. The overall design of software platform is well modular. Each module is centered on database. The input and output of data are very convenience. Not only all parameters of SVC system, including the rated voltage, rated current, rated power, and error range etc., can be worked out quickly by the platform, even including the models of some commonly used devices, but also it can simulate operation effect of SVC in alliance with MATLAB/SIMULINK. Through simulation analysis the optimal parameters can be obtained. By this software platform in design process of SVC, the amount of design work is largely reduced, the design cycle is largely shortened, and all this are benefit to the industrialization of SVC.

In this paper, the cusp electromagnetic field is researched to improve the quality of single crystal. The cusp electromagnetic field in 8 inch single crystal furnace is modeled with ANSYS software, the electromagnetic field distribution, intensity and the factors that influence electromagnetic field intensity, such as current of the coil, distance between two coils and etc., of the cusp electromagnetic field are simulated and analyzed and optimized using finite element method. Finally, the optimum design of cusp electromagnetic field is validated, analyzing the key parameters of the electromagnetic field whether can meet the demands offered by the crystal growth or not.

Switching power supply of low voltage and high current based on power electronic technology is widely applied in some industry fields, especially in electroplating, welding and heating. Harmonics in high-power switching supply pollute power system heavily, which must be suppressed effectively. On basis of analyzing harmonic current of switching supply that the output technical specification is 60V/500A, this paper designs active power filter for switching power supply with high power and frequency, which adopts ip-iq current detecting method and hysteresis current control method. Based on simulation platform of Matlab/Simulink, the filtering effect is simulated and analyzed. Simulation results demonstrate the active power filter has expected performance, which can not inhibit harmonic current instantaneously, but also compensate reactive power. The designed active power filter can effectively improve the power quality. In order to meet the requirements of the magnetron sputtering, an asymmetric bipolar pulsed power supply based on DSP is presented in this paper. It can freely adjust the pulse parameters including pulse amplitude, frequency, duty cycle, and even the number and transition time of the positive and negative pulse according to the sputtering demands. Finally, a 5kW/ 100kHz prototype is designed. It adopts two isolated DC/DC converters as the current source, and an asymmetric pulses generator. The digital signal processor (DSP) fulfills the digital closed loop control of current sources. Meanwhile, it achieves the adjustment of pulse segment. The experimental results verify the designed.
