Biodegradable bone grafts have gained considerable attention in the field of clinical research as they can be used to avoid re-surgery to remove scaffolds. We fabricated biodegradable composite ceramic scaffolds using magnesium oxide and calcium phosphate following the vat photopolymerization technology. We studied the effects of different magnesium oxide content on the process of in vitro degradation of the composite scaffolds and discussed the degradation process by comparing the extent of degradation realized in Tris-HCl buffer and simulated body fluids. We also explored the variations in the mechanical properties of the scaffolds during degradation. The results revealed that the composite scaffolds maintained the strength standard for spongy bone (compressive strength σ: 2–20 MPa) even after 28 d of degradation. Additionally, the strength of the composite scaffold containing 80 wt% magnesium oxide was comparable to the strength standard recommended for cortical bones (σ: 100–200 MPa). Controllable degradation rates were recorded, and acceptable mechanical properties were observed for these scaffolds during degradation. The results reported herein can be potentially used to expand the application prospects of biodegradable scaffolds.

Abstract Background Total cholesterol is inversely associated with mortality in dialysis patients, which seems implausible in real-world clinical practice. May there be an optimal range of total cholesterol associated with a lower mortality risk? We aimed to evaluate the optimal range for peritoneal dialysis (PD) patients. Methods We conducted a retrospective real-world cohort study of 3565 incident PD patients from five PD centers between January 1, 2005, and May 31, 2020. Baseline variables were collected within one week before the start of PD. The associations between total cholesterol and mortality were examined using cause-specific hazard models. Results 820 (23.0%) patients died, including 415 cardiovascular deaths, during the follow-up period. Restricted spline plots showed a U-curved association of total cholesterol with mortality. Compared with the reference range (4.10–4.50 mmol/L), high levels of total cholesterol (> 4.50 mmol/L) were associated with increased risks of all-cause (hazard ratio [HR] 1.35, 95% confidence index [CI] 1.08–1.67) and cardiovascular mortality (HR 1.38, 95% CI 1.09–1.87). BackgroundAlzheimer’s disease (AD) is characterized by the presence of gray matter lesions and alterations in white matter. This study aims to investigate the research related to white matter in the context of AD from a Bibliometric standpoint.MethodsRegular and review articles focusing on the research pertaining to Alzheimer’s disease (AD) and white matter were extracted from the Web of Science Core Collection (WOSCC) database, covering the period from its inception to 10th July 2023. The “Bibliometrix” R package was employed to summarize key findings, to quantify the occurrence of top keywords, and to visualize the collaborative network among countries. Furthermore, VOSviewer software was utilized to conduct co-authorship and co-occurrence analyses. CiteSpace was employed to identify the most influential references and keywords based on their citation bursts. The retrieval of AD- and white matter-related publications was conducted by the Web of Science Core Collection. Bibliometric analysis and visualization, including the examination of annual publication distribution, prominent countries, active institutions and authors, core journals, co-cited references, and keywords, were carried out by using VOSviewer, CiteSpace, the Bibliometrix Package, and the ggplot2 Package. The quality and impact of publications were assessed using the total global citation score and total local citation score.ResultsA total of 5,714 publications addressing the intersection of Alzheimer’s disease (AD) and white matter were included in the analysis. The majority of publications originated from the United States, China, and the United Kingdom. Prominent journals were heavily featured in the publication output. In addition to “Alzheimer’s disease” and “white matter,” “mild cognitive impairment,” “MRI” and “atrophy” had been frequently utilized as “keywords.”ConclusionThis Bibliometric investigation delineated a foundational knowledge framework that encompasses countries, institutions, authors, journals, and articles within the AD and white matter research domain spanning from 1981 to 2023. The outcomes provide a comprehensive perspective on the broader landscape of this research field.

Abstract Background New lipid-lowering therapy at the start of dialysis and measurement of lipid parameters over the follow-up period is not recommended in dialysis patients, which seems unappropriated in clinical practice. We aimed to examine the effect of hyperlipidemia on mortality in patients undergoing continuous ambulatory peritoneal dialysis (CAPD). Methods A retrospective cohort study was performed, including 2939 incident CAPD patients from five dialysis facilities between January 1, 2005, and December 31, 2018. The primary outcome was all-cause mortality. The association between hyperlipidemia at the start of CAPD and all-cause mortality was evaluated using Cox proportional hazards regression. Results Of 2939 with a median age of 50.0 (interquartile range, 39.0–61.0), 1697 (57.7%) were men, 533 (18.1%) had hyperlipidemia, 549 (18.7%) had diabetes mellitus, 1915 (65.2%) had hypertension, and 410 (14.0%) had a history of CVD. During the median follow-up period of 35.1 months, 519 (17.7%) died, including 402 (16.7%, 47.4/1000 patient-years) in the non-hyperlipidemia group and 117 (22.0%, 71.1/1000 patient-years) in the hyperlipidemia group. Over the overall follow-up period, patients with hyperlipidemia had an equally high risk of all-cause mortality throughout follow-up as those without hyperlipidemia ([HR] 1.04, 95% confidence interval [CI] 0.83 to 1.31). However, from the 48-month follow-up onwards, hyperlipidemia was associated with a 2.26 (95% CI 1.49 to 3.43)-time higher risk of all-cause mortality than non-hyperlipidemia. This study aimed to evaluate the suppressive potential of different soils on soybean cyst nematodes (SCN) and to estimate the suppressive mechanism. Fifteen soils (designated as soil A to O) from different agricultural fields with varying organic inputs were added with SCN-infested soil and grown with a green soybean variety. The SCN density in the soil at 6 weeks of soybean growth was markedly different depending on the soils used, indicating a different level of disease suppressiveness. No significant correlation was observed between the SCN density and any of the soil physicochemical and biological characteristics tested. Then, to estimate a suppression mechanism, F-soil that showed the lowest density of SCN was added to the SCN-infested soil with or without streptomycin to kill bacteria and grown with soybean. SCN density was not increased by the addition of streptomycin, indicating that soil bacteria may not be involved in the suppressiveness of F-soil. In total, 128 fungal strains were isolated from the rhizosphere of F-soil and inoculated in a combination or singly in the SCN-infested soil. After repeated screenings, five strains were selected since the SCN density was consistently decreased by them. Sequence analysis showed that they were closest to Clonostachys rosea, Aspergillus niger, Aspergillus fumigatus, Fusarium oxysporum, and Cylindrodendrum alicantinum. All five strains significantly reduced the mobility of second-stage juveniles (J2). Further, C. rosea a2, A. niger a8, and F. oxysporum a25 significantly decreased hatching. Overall, the present study demonstrated that soil fungi played an important role in SCN suppression in F-soil.

Long-term fertilization can result in changes in the nitrogen (N) cycle in maize rhizosphere soil. However, there have been few reports on the impacts of plant–soil–microbe regulatory mechanisms on the N cycle in soil. In this study, soil samples were collected from a long-term experimental site located at Jilin Agricultural University, Changchun City, Jilin Province, Northeast China. We then analyzed the changes in the functional genes related to the N cycle, soil enzyme activity, and maize root exudates under long-term fertilizer application using metagenomics and liquid chromatography analysis. We aimed to investigate the response of the N cycle to long-term fertilizers, the interaction among plant, soil, and microbes, and the effect of the plant–soil–microbe system on the N cycle. Long-term fertilization had a significant effect on soil N contents, N2O emissions, and enzyme activity related to the N cycle in maize rhizosphere soil. The functional genes of the N cycle were mainly enriched in the N degradation pathway in maize rhizosphere soil. N fertilizer application decreased the abundance of functional genes related to N fixation and degradation, denitrification, and assimilatory nitrate reduction (ANRA) and increased the abundance of functional genes participating in dissimilatory nitrate reduction (DNRA) and anaerobic ammonia oxidation/hydroxylamine oxidation. The soil environment was positively related to soil enzyme activity and negatively related to the microbial community composition and amino acids in root exudates. The contribution rate of microorganisms to the N cycle was the highest (r2 = 0.900), followed by amino acids (r2 = 0.836) and the soil environment (r2 = 0.832). Therefore, we concluded that N fertilizer is the main factor limiting the soil N cycle and that microorganisms are the main factor regulating the N cycle in the plant–soil–microorganism system.

MicroRNAs (miRNAs) are emerging as effective therapeutic agents. When testing whether miR-145-5p could alleviate kidney injury, we unexpectedly found that extracellular vesicles loaded with miR-145-5p induced proteinuria and podocyte foot process effacement in normal control mice. To explore the mechanism of miR-145-5p’s toxicity to podocytes, we hypothesized that miR-145-5p could enter podocytes and inhibit genes essential for podocytes. We demonstrated that systemically administered miRNA can enter podocytes. Next, we predicted 611 podocyte essential genes based on single-cell RNA sequencing (RNA-seq) and found that 32 of them are predicted to be targeted by miR-145-5p. Functional annotation of the 32 podocyte essential genes revealed small GTPase-mediated signal transduction as the top pathway. We experimentally validated that miR-145-5p targeted Arhgap24 and Srgap1, the essential regulators of the Rho family of small GTPases, increased the activity of Rac1 and Cdc42, and reduced RhoA activity, accompanied by cellular injury, in podocytes. These results explain how miR-145-5p has deleterious effect on podocytes. Most importantly, our study provides a novel approach to investigate how a miRNA affects a given cell type, allowing not only identification of the molecular mechanism underlying an observed side effect of a miRNA drug but also prediction of miRNA drug toxicity on various cell types.

Chronic internal carotid artery occlusion (CICAO) has high prevalence and incidence rates, and patients with CICAO can be completely asymptomatic, experience a devastating stroke or die. It is important to note that CICAO causes cerebrovascular accidents. Currently, the external carotid-internal carotid (EC-IC) bypass technique is used to treat CICAO. However, many clinical studies showed that EC-IC bypass was not beneficial for many patients with CICAO. Meanwhile, endovascular intervention treatment options for CICAO are evolving, and an increasing number of patients are undergoing endovascular intervention therapy. Accordingly, a review comparing both techniques is warranted. For this review, we searched PubMed and collected relevant case study reports comparing endovascular interventional therapy and internal and external cervical bypass surgeries to provide strategies for clinical treatment.

Abstract Kidney function is affected in COVID-19, while kidney itself modulates the immune response. Here, hypothesize if COVID-19 urine biomarkers level can assess immune activation vs. clinical trajectory. Considering the kidney’s critical role in modulating the immune response, we sought to analyze activation markers in patients with pre-existing dysfunction. This was a cross-sectional study of 68 patients. Blood and urine were collected within 48 h of hospital admission (H1), followed by 96 h (H2), seven days (H3), and up to 25 days (H4) from admission. Serum level ferritin, procalcitonin, IL-6 assessed immune activation overall, while the response to viral burden was gauged with serum level of spike protein and αspike IgM and IgG. 39 markers correlated highly between urine and blood. Age and race, and to a lesser extend gender, differentiated several urine markers. The burden of pre-existing conditions correlated with urine DCN, CAIX and PTN, but inversely with IL-5 or MCP-4. Higher urinary IL-12 and lower CAIX, CCL23, IL-15, IL-18, MCP-1, MCP-3, MUC-16, PD-L1, TNFRS12A, and TNFRS21 signified non-survivors. APACHE correlated with urine TNFRS12, PGF, CAIX, DCN, CXCL6, and EGF. Admission urine LAG-3 and IL-2 predicted death. Pre-existing kidney disease had a unique pattern of urinary inflammatory markers. Acute kidney injury was associated, and to a certain degree, predicted by IFNg, TWEAK, MMP7, and MUC-16. Remdesavir had a more profound effect on the urine biomarkers than steroids. Urinary biomarkers correlated with clinical status, kidney function, markers of the immune system activation, and probability of demise in COVID-19.

BackgroundLong-term alcohol exposure is associated with oxidative stress, endoplasmic reticulum (ER) stress, and neuroinflammation, which may impair cognitive function. Probiotics supplements can significantly improve cognitive function in neurodegenerative diseases such as Alzheimer’s disease. Nevertheless, the effect of Lactobacillus plantarum ST-III culture supernatant (LP-cs) on alcohol-induced cognitive dysfunction remains unclear.MethodsA mouse model of cognitive dysfunction was established by intraperitoneal injection of alcohol (2 g/kg body weight) for 28 days. Mice were pre-treated with LP-cs, and cognitive function was evaluated using the Morris water maze test. Hippocampal tissues were collected for biochemical and molecular analysis.ResultsLP-cs significantly ameliorated alcohol-induced decline in learning and memory function and hippocampal morphology changes, neuronal apoptosis, and synaptic dysfunction. A mechanistic study showed that alcohol activated protein kinase R-like endoplasmic reticulum kinase (PERK) signaling and suppressed brain derived neurotrophic factor (BDNF) levels via ER stress in the hippocampus, which LP-cs reversed. Alcohol activated oxidative stress and inflammation responses in the hippocampus, which LP-cs reversed.ConclusionLP-cs significantly ameliorated alcohol-induced cognitive dysfunction and cellular stress. LP-cs might serve as an effective treatment for alcohol-induced cognitive dysfunction.

BackgroundIn dialysis patients, lowering low-density lipoprotein cholesterol (LDL-C) did not provide benefits, which seemed implausible in clinical practice. We hypothesized a U-shaped association between LDL-C and mortality in dialysis patients.MethodsIn this multi-center retrospective real-world cohort study, 3,565 incident Chinese peritoneal dialysis (PD) patients between January 1, 2005, and May 31, 2020, were included. The associations between baseline LDL-C and mortality were examined using cause-specific hazard models.ResultsOf 3,565 patients, 820 died, including 415 cardiovascular deaths. As compared with the reference range (2.26-2.60 mmol/L), both higher levels of LDL-C (> 2.60 mmol/L) and lower levels of LDL-C (< 2.26 mmol/L) were associated with increased risks of all-cause mortality (hazard ratio [HR],1.35, 95% confidence index [CI], 1.09-1.66; HR 1.36, 95%CI, 1.13-1.64) and cardiovascular mortality (HR, 1.31, 95% CI, 1.10-1.72; HR, 1.64; 95% CI, 1.22-2.19). Malnutrition (albumin < 36.0 g/L) modified the association between LDL-C and cardiovascular mortality (P for interaction = 0.01). A significantly increased risk of cardiovascular mortality was observed among patients with malnutrition and lower levels of LDL-C (HR 2.96, 95%CI 1.43-6.12) or higher levels of LDL-C (HR 2.81, 95%CI 1.38-5.72).ConclusionLow and high levels of LDL-C at the start of PD procedure were associated with increased all-cause and cardiovascular mortality risks. Malnutrition may modify the association of LDL-C with cardiovascular mortality.

Long-term intake of high-energy diet can lead to decreased insulin sensitivity and even insulin resistance, eventually leading to diabetes. Diabetes often occurs in middle-aged and elderly people. However, there is growing evidence that the incidence rate of young body is increasing over the years. This means that insulin resistance can be caused by excessive energy intake in both young and old people. In this study, high-fat diet (HFD) and normal diet were fed to rats of elderly experimental group (EE), elderly control group (EC), young experimental group (YE), and young control group (YC), respectively, for 8 weeks, by which insulin resistance model was obtained. Insulin sensitivity was measured, histopathology changes in liver and skeletal muscle tissues were observed, and mitochondrial fusion and division and cell senescence were detected in four groups of rats. The results showed that both young and elderly rats developed significant insulin resistance, fat deposition, decline of mitochondrial function and mitochondrial biosynthesis in liver and skeletal muscle, and cell aging after HFD feeding. In addition, the degree of mitochondrial dysfunction and aging in young rats was similar to that of aged rats fed a normal diet after HFD. This experiment provides a reference for an in-depth study of the regulatory mechanisms of cellular energy metabolism in this state.

BackgroundThe association between serum creatine kinase and mortality in patients with peritoneal dialysis (PD) remained unknown.MethodsWe retrospectively collected data on 3,446 incident patients with from five PD centers in China between 1 January 2005 and 31 May 2020. Creatine kinase was collected 1 week before the start of PD. We examined the association between creatine kinase and mortality using Cox proportional hazards model.ResultsThe median creatine kinase was 113 (range, 1.22–4,574) IU/L. With a median follow-up of 39.5 (range, 3.1–181.5) months, 763 (22.1%) all-cause deaths occurred, including 384 (11.1%) cardiovascular deaths. Changbai Mountain is an important part of the development and opening pilot area of Changjitu. It is the birthplace of Songhua River, Yalu River, and Tumen River, and is known as the source of the three rivers. Millions of people live in the basin. A volcanic eruption accompanied by earthquakes would lead to a large number of landslides, debris flows, and show a chain effect, the formation of a secondary geological disaster chain, which is a serious threat to people’s lives and property safety. This paper selected indexes from three aspects: the hazard of earthquake-induced geological disaster chain, the exposure and vulnerability of disaster-bearing bodies, and the risk assessment of earthquake-induced geological disaster chain. The sensitivity values of each influence factor were calculated by the certainty factor (CF) using the support vector machine, and then, the susceptibility assessment was obtained. The cumulative displacement calculated by the Newmark model represented the potential risk intensity. We considered the Changbai Mountain volcanic earthquake–landslide disaster chain as an example. The results of risk assessment showed that the extremely high and high risk areas were mainly located within the 12 km radius of Tianchi Lake, and the other areas in the study area were mainly associated with very low to low risk values. The verification results showed that the receiver operating characteristic (ROC) curve area was 0.8373, indicating that the method was very effective in the identification and assessment of seismic hazard chain risk. In these high-risk areas, relevant countermeasures should be formulated to prevent the risk of geological disasters, strengthen the implementation of regional disaster prevention and reduction work, and ensure the safety of residents’ lives and property.

Background: Preexisting cardiovascular disease (CVD) and hypertension are each associated with poor prognosis in peritoneal dialysis (PD) patients. Joint associations of preexisting CVD and hypertension have not been comprehensively evaluated in this population. Methods: We conducted a retrospective cohort study of 3073 Chinese incident PD patients from five dialysis centres between January 1, 2005, and December 31, 2018. The joint associations between preexisting CVD, hypertension, and mortality were analysed using Cox regression models. Results: Over a median of 33.7 months of follow-up, 581 (18.6%) patients died, with 286 (9.3%) deaths due to CVD. After adjusting for confounding factors, the preexisting CVD coexisting with hypertension, preexisting CVD, and hypertension groups had higher risks of all-cause mortality (hazard ratio [HR]: 3.97, 95% confidence interval [CI]: 3.06 to 5.15; HR: 2.21, 95% CI: 1.29 to 3.79; and HR: 1.83, 95% CI: 1.47 to 2.29, respectively) and CVD mortality (HR: 4.68, 95% CI: 3.27 to 6.69; HR: 2.10, 95% CI: 0.95 to 4.62; and HR: 1.86, 95% CI: 1.36 to 2.54, respectively) than the control group without preexisting CVD or hypertension (p for trend 0.05). The joint associations showed similar patterns using the Fine–Gray competing risk models. Conclusions: Preexisting CVD and hypertension at the start of PD were additive prognostic utilities for mortality, and preexisting CVD was more strongly associated with mortality than hypertension.

Identification of novel immune biomarkers to gauge the underlying pathology and severity of COVID-19 has been difficult due to the lack of longitudinal studies. Here, we analyzed serum collected upon COVID-19 admission (t1), 48 hours (t2), and seven days later (t3) using Olink proteomics and correlated to clinical, demographics, and therapeutic data. Older age positively correlated with decorin, pleiotrophin, and TNFRS21 but inversely correlated with chemokine (both C-C and C-X-C type) ligands, monocyte attractant proteins (MCP) and TNFRS14. The burden of pre-existing conditions was positively correlated with MCP-4, CAIX, TWEAK, TNFRS12A, and PD-L2 levels. Individuals with COVID-19 demonstrated increased expression of several chemokines, most notably from the C-C and C-X-C family, as well as MCP-1 and MCP-3 early in the course of the disease. Similarly, deceased individuals had elevated MCP-1 and MCP-3 as well as Gal-9 serum levels. LAMP3, GZMB, and LAG3 at admission correlated with mortality. Only CX3CL13 and MCP-4 correlated positively with APACHE score and length of stay, while decorin, MUC-16 and TNFRSF21 with being admitted to the ICU. We also identified several organ-failure-specific immunological markers, including those for respiratory (IL-18, IL-15, Gal-9) or kidney failure (CD28, VEGF). Treatment with hydroxychloroquine, remdesivir, convalescent plasma, and steroids had a very limited effect on the serum variation of biomarkers. Our study identified several potential targets related to COVID-19 heterogeneity (MCP-1, MCP-3, MCP-4, TNFR superfamily members, and programmed death-ligand), suggesting a potential role of these molecules in the pathology of COVID-19.

BackgroundType 2 diabetes (T2D) is a metabolic dysfunction disease that causes several complications. Liver injury is one of these that severely affects patients with diabetes. Fibroblast growth factor 1 (FGF1) has glucose-lowering activity and plays a role in modulation of several liver injuries. Nevertheless, the effects and potential mechanisms of FGF1 against diabetes-induced liver injury are unknown.MethodsTo further investigate the effect of FGF1 on diabetic liver injury, we divided db/db mice into two groups and intraperitoneally (i.p.) injected either with FGF1 at 0.5 mg/kg body weight or saline every other day for 4 weeks. Then body weights were measured. Serum and liver tissues were collected for biochemical and molecular analyses.ResultsFGF1 significantly reduced blood glucose and ameliorated diabetes-induced liver steatosis, fibrosis, and apoptosis. FGF1 also restored defective hepatic autophagy in db/db mice. Mechanistic investigations showed that diabetes markedly induced oxidative stress and endoplasmic reticulum stress and that FGF1 treatment significantly attenuated these effects.ConclusionsFGF1-associated glucose level reduction and amelioration of cellular stress are potential protective effects of FGF1 against diabetes-induced liver injury.

Abstract:: Three wheat cultivars—Zhengmai 366 (ZM366), Zhoumai 27 (ZM27) and Yangmai 15 (YM15)—were used in the present study. After storage at 25 ℃ for 15, 30, 45, 60, 90, 120 and 150 d, the crude starch, amylose, amylopectin, damaged starch and reducing sugar contents in wheat during the after-ripening period were measured. The results revealed (1) a marginal decrease in the total starch content; (2) no significant change in the amylopectin content; and (3) significant change in the amylose and reducing sugar contents during the after-ripening period in all the three wheat cultivars. Further, the amylose content in the three cultivars exhibited an increasing trend during the initial phase of the after-ripening period. Among the cultivars, ZM366 exhibited the highest increase, which stabilized after 90 d. The reducing sugar content showed a significant decrease during the initial phase of the after-ripening period with ZM366 showing the highest decrease, which attained stability after 90 d. The damaged starch content in all the three wheat cultivars exhibited a decreasing trend with ZM27 showing the highest decrease, which stabilized after 90 d. There were no changes in the forms of starch crystal among the three wheat cultivars after the after-ripening period, with only ZM366 showing a significant increase in starch crystallinity.
