In the course of her career, German ornithologist Emilie Snethlage (1868-1929), who worked in Brazil in the early twentieth century, was involved in all the steps that characterize the “production” of a specimen for scientific collection: from fieldwork, with the collection and preparation of materials, to their description and publication of results. Each of these stages mobilizes different material practices and sociability networks. During fieldwork or in her museum activities, the fact of being a woman demanded from Snethlage specific strategies for establishing her scientific legitimacy, analyzed in this article, especially her activities related to collecting practices.

In environmental risk assessment either for registration purposes or for retrospective assessments of monitoring data, the hazard assessment is predominantly based on effect data from ecotoxicity studies. Most regulatory frameworks require studies used for risk assessment to be evaluated for reliability and relevance. Historically, the Klimisch methodology was used in many regulatory procedures where reliability needed to be evaluated. More recently, the Criteria for Reporting and Evaluating Ecotoxicity Data (CRED) have been developed for aquatic ecotoxicity studies, providing more detailed guidance on the evaluation and reporting of not only the reliability but also the relevance of a scientific study. Here, we discuss the application of the CRED methodology for assessing sediment and soil ecotoxicity studies, addressing important sediment- and soil-specific criteria that should be included as part of the CRED evaluation system. We also provide detailed recommendations for the design and reporting of sediment and soil toxicity studies that can be used by scientists and researchers wishing to contribute ecotoxicological data for effect assessments carried out within regulatory frameworks. The progress in chemical analytics and understanding of pesticide dynamics in surface waters allows establishing robust data on compounds with frequent exceedances of quality standards. The current chemical, temporal, and spatial coverage of the pesticide monitoring campaigns differs strongly between European countries. A questionnaire revealed differences in monitoring strategies in seven selected European countries; Nordic countries prioritize temporal coverage, while others focus on spatial coverage. Chemical coverage has increased, especially for non-polar classes like synthetic pyrethroids. Sweden combines monitoring data with agricultural practices for derived quantities, while the Netherlands emphasizes spatial coverage to trace contamination sources. None of the EU member states currently has established a process for linking chemical surface water monitoring data with regulatory risk assessment, while Switzerland has recently established a legally defined feedback loop. Due to their design and objectives, most strategies do not capture concentration peaks, especially 2-week composite samples, but also grab samples. Nevertheless, for substances that appear problematic in many data sets, the need for action is evident even without harmonization of monitoring programs. Diclofenac, a widely used nonsteroidal anti-inflammatory drug (NSAID), enters the aquatic environment worldwide. The effect values available for the derivation of an environmental quality standard (EQS) are markedly heterogeneous, even within the same species. This heterogeneity could partially be attributed to inter-laboratory variation, but is also observed in repeated tests within the same facility. Diclofenac is ionisable; its speciation and potential for uptake and thus toxicity is influenced by pH. A high correlation has previously been observed between effects in zebrafish embryos and the pH-specific partitioning coefficient logD for diclofenac. We hypothesized that the observed heterogeneity could also be attributed to differences in study pH. To test this hypothesis, we reviewed physicochemical data and selected ecotoxicity data that were considered to be reliable and relevant in the latest EU EQS Dossier for which a study pH was reported for further analysis and EQS derivation. We adjusted the reported effect concentrations for differences in uptake using the delta logD value for the worst case pH value of 6.5. pH adjustment of effect values resulted in decreased heterogeneity of the acute effect data and a better fit of the chronic species sensitivity distribution. Both, the MAC-EQS and the AA-EQS were derived using the deterministic approach as data requirements for deriving EQS based on the SSD were not fulfilled. Many studies had to be discarded because test pH was not reported or exposure concentrations had not been analytically verified. Physico-chemical data had to be discarded due to non-relevant experimental conditions or missing information. We strongly encourage scientists publishing ecotoxicity data for ionisable substances to report the test pH together with the effect values and encourage measurement of physico-chemical parameters at environmentally relevant conditions. Background: Amyotrophic lateral sclerosis (ALS) is an incurable neurodegenerative disease characterized by the loss of motor neurons in cerebral cortex, brainstem and spinal cord. Numerous studies have demonstrated signs of oxidative stress in postmortem neuronal tissue, cerebrospinal fluid, plasma and urine of ALS patients, without focusing on the specific processes within motor neurons. Thus, we aimed to investigate the relevance of reactive oxygen species (ROS) detoxification mechanisms and its consequences on the formation of toxic/lethal DNA double strand breaks (DSBs) in the ALS model of the Wobbler mouse. Methods: Live cell imaging in dissociated motor neuronal cultures was used to investigate the production of ROS using Dihydroethidium (DHE). The expression levels of ROS detoxifying molecules were investigated by qPCR as well as Western blots. Furthermore, the expression levels of DNA damage response proteins p53bp1 and H2ax were investigated using qPCR and immunofluorescence staining. Proof-of-principle experiments using ROS scavengers were performed in vitro to decipher the influence of ROS on the formation of DNA double strand breaks quantifying the γH2ax spots formation. Results: Here, we verified an elevated ROS-level in spinal motor neurons of symptomatic Wobbler mice in vitro . As a result, an increased number of DNA damage response proteins p53bp1 and γH2ax in dissociated motor neurons of the spinal cord of Wobbler mice was observed. Furthermore, we found a significantly altered expression of several antioxidant molecules in the spinal cord of Wobbler mice, suggesting a deficit in ROS detoxification mechanisms. This hypothesis could be verified by using ROS scavenger molecules in vitro to reduce the number of γH2ax foci in dissociated motor neurons and thus counteract the harmful effects of ROS. Conclusion: Our data indicate that maintenance of redox homeostasis may play a key role in the therapy of the neurodegenerative disease ALS. One of the most critical situations in urban areas is when motorists turn right in an intersection and cyclists cross the road. Many of those crashes result in severe consequences for cyclists. In order to increase the safety of cyclists, especially in the case of conflicts with right-turning vehicles, an online infrastructure-based assistance system may be a promising solution warning drivers and cyclists when a conflict or crash is predicted. By means of automated video traffic detection, the resulting trajectories of road users can be analysed and a warning can be sent to vehicles and cyclists equipped with vehicle-to-anything communication (V2X) when a high risk is estimated. An approach for online risk estimation was developed combining the surrogate measure of safety (SMoS) gap time (GT) with trajectory prediction-based estimates of the time-to-arrival (TTA) or distance to conflict point (DCp) and velocity (v). A decision tree as classifier of risk levels based on the previous named risk features was trained to model the risks perceived by humans. Expert ratings of traffic conflict scenes were used to build a model, apply the model, and improve it in the field. The warning system was evaluated by test drives in real traffic at the urban AIM Research Intersection in Braunschweig, Germany. In general, the system warned reliably. This study assessed the acute and chronic risk of pesticides, singly and as mixtures, for fish using comprehensive chemical data of four monitoring studies conducted in small- and medium-sized streams of Switzerland between 2012 and 2018. Pesticides were ranked based on single substance risk quotients and relative contribution to mixture risk. Concentrations of the pyrethroid insecticides, λ-cyhalothrin, cypermethrin and deltamethrin, and the fungicides, carbendazim and fenpropimorph, posed acute or chronic single substance risks. Risk quotients of eighteen additional pesticides were equal or greater than 0.1, and thirteen of those contributed ≥30% to mixture risk. Relatively few substances dominated the mixture risk in most water samples, with chronic and acute maximum cumulative ratios never exceeding 5 and 7, respectively. A literature review of toxicity data showed that concentrations of several pesticides detected in Swiss streams were sufficient to cause direct sublethal effects on fish in laboratory studies. Based on the results of our study, we conclude that pesticides detected in Swiss streams, especially pyrethroid insecticides, fungicides and pesticide mixtures, pose a risk to fish health and can cause direct sublethal effects at environmental concentrations. Sensitive life stages of species with highly specialized life history traits may be particularly vulnerable; however, the lack of toxicity data for non-model species currently prevents a conclusive assessment across species.

Synthetic chemicals are frequently detected in water bodies, and their concentrations vary over time. Water monitoring programs typically employ either a sequence of grab samples or continuous sampling, followed by chemical analysis. Continuous time-proportional sampling yields the time-weighted average concentration, which is taken as proxy for the real, time-variable exposure. However, we do not know how much the toxicity of the average concentration differs from the toxicity of the corresponding fluctuating exposure profile. We used toxicokinetic-toxicodynamic models (invertebrates, fish) and population growth models (algae, duckweed) to calculate the margin of safety in moving time windows across measured aquatic concentration time series (7 pesticides) in 5 streams. A longer sampling period (14 d) for time-proportional sampling leads to more deviations from the real chemical stress than shorter sampling durations (3 d). The associated error is a factor of 4 or less in the margin of safety value toward underestimating and an error of factor 9 toward overestimating chemical stress in the most toxic time windows. Under- and overestimations occur with approximate equal frequency and are very small compared with the overall variation, which ranged from 0.027 to 2.4 × 10 10 (margin of safety values). We conclude that continuous, time-proportional sampling for a period of 3 and 14 d for acute and chronic assessment, respectively, yields sufficiently accurate average concentrations to assess ecotoxicological effects.
