Canopy coverage calculated from 2D images effectively estimates several parameters, such as dry weight above the ground, stem number, and risk of lodging, without destroying the crops. This study aimed to clarify how to accurately calculate canopy coverage, regardless of environmental conditions, using various colour models and a relative depth estimation model, MiDaS. For wheat cultivated in 2022/23, the canopy coverage at the initiation of the stem elongation stage (CC30), calculated using only the HSV colour model, was overestimated because of the presence of weeds. However, when the heatmap generated using MiDaS was combined with the results extracted from the HSV colour model, it showed a CC30 similar to the accurate CC30 calculated by hand drawing, regardless of the wheat variety, sowing pattern, and location of the image taken, with an RMSE as low as 3.48. The CC30 calculated by combining the HSV colour model and MiDaS was significantly positively correlated with the number of stems, dry weight above ground, and nitrogen content above ground at the initiation of the stem elongation stage of ‘Sanukinoyume 2009’ grown in 2021/22 and 2022/23 (n = 72, r > 0.798). The combination of the HSV colour model and MiDaS effectively and accurately calculates canopy coverage regardless of environmental conditions and can be used for growth diagnosis and variable nitrogen fertiliser application after the initiation of the stem elongation stage.

Abstract Periodontitis is a prevalent oral inflammatory disease that leads to alveolar bone loss and may exert an adverse impact on systemic health. Periodontal disease may be associated with hepatocellular carcinoma (HCC); however, the mechanism of such an association is unknown. In this study, Stelic Animal model (STAM) mice, a model of nonalcoholic steatohepatitis (NASH)-HCC, were induced to develop periodontitis and subjected to histopathological and immunological analyses. HCC progression was greater in STAM mice with experimental periodontitis compared with that in STAM mice without experimental periodontitis. Tumor necrosis factor-α (TNFα), matrix metalloproteinase-9 (MMP9), collagen 1, and angiopoietin-like protein 2 (ANGPTL2) gene expression was significantly increased in the liver of the periodontitis group. ANGPTL2 was previously reported to be involved in the pathogenesis of periodontitis, and HCC and ANGPTL2 protein tended to be more abundant in the pocket epithelium of STAM mice with experimental periodontitis than in control STAM mice. ANGPTL2 levels in the serum of STAM mice with experimental periodontitis tended to be higher than in control STAM mice. Our results indicate that ANGPTL2 is produced in chronically inflamed periodontal tissue and then travels to the liver via the bloodstream where it accumulates to promote the progression of hepatocellular carcinoma.

The pyrolysis characteristics of Spirulina platensis residue (SPR) with silica–alumina catalysts were investigated using thermogravimetric analysis (TGA). The effects of differing amounts of catalysts on thermochemical behavior and kinetics parameters (pre-exponential factor in Arrhenius equation [A] and activation energy [Ea]) were studied. The experiment was carried out from 30 to 1000? at a heating rate of 20?/min for the case of non-catalytic and catalytic pyrolysis (silica–alumina). For the catalytic pyrolysis, also of interest were the catalyst-to-SPR weight ratios of 1:1 and 1:2. The TGA curve and differential thermogravimetric peak analysis results suggest that the use of catalysts in pyrolysis (particularly at a catalyst-to-SPR weight ratio of 1:1) reduces both pyrolysis time and temperature range to 14.68 min and 230–555?, respectively. The kinetic parameters were then calculated in a one-step global non-isothermal model and solved using a least squares method in MATLAB. The presence of catalyst was able to reduce Ea to the lowest value from 41.10 kJ/mol (without catalyst) to 40.77 kJ/mol (weight ratio of 1:2) and 39.46 kJ/mol (weight ratio of 1:1) in Zone 1. However, the increase of catalyst quantity was not in line with the increase of reaction rate constant (k) and resulted in reasonably low A of, respectively, 593.30, 406.31, and 266.37.

The effect of Mfa1 fimbriae of Porphyromonas gingivalis on the progression of bone resorption remains unclear, especially compared with another fimbriae, FimA. We investigated the effect of Mfa1 on osteoclastogenesis together with FimA. We also investigated the role of Toll-like receptors (TLRs) in Mfa1 recognition during osteoclast differentiation. Receptor activator of nuclear factor κβ ligand (RANKL)-prestimulated RAW264 cells were used to examine the effects of purified Mfa1 fimbriae. The number of osteoclasts was examined by tartrate-resistant acid phosphate (TRAP) staining, osteoclast activation was investigated by bone resorption assays, and gene expression of differentiation markers was examined by quantitative real-time PCR. Transfection of Tlr2 and Tlr4 siRNAs into RAW264 cells was also employed and their role in Mfa1 recognition was investigated. Mfa1 effectively induced the formation of TRAP-positive multinucleated cells and activated osteoclasts. Mfa1 also increased gene expression of Acp5, Mmp9, and Ctsk in RANKL-prestimulated RAW264 cells compared with the control. The osteoclastogenesis induced by Mfa1 was significantly decreased in cells transfected with Tlr2 or Tlr4 siRNAs compared with control siRNA. Our results revealed the role of Mfa1 fimbriae in osteoclastogenesis that may contribute to the partial elucidation of the mechanisms of periodontal disease progression and the development of new therapeutic strategies.

Periodontal disease, a major cause of tooth loss, is an infectious disease caused by bacteria with the additional aspect of being a noncommunicable disease closely related to lifestyle. Tissue destruction based on chronic inflammation is influenced by host and environmental factors. The treatment of periodontal disease varies according to the condition of each individual patient. Although guidelines provide standardized treatment, optimization is difficult because of the wide range of treatment options and variations in the ideas and skills of the treating practitioner. The new medical concepts of “precision medicine” and “personalized medicine” can provide more predictive treatment than conventional methods by stratifying patients in detail and prescribing treatment methods accordingly. This requires a new diagnostic system that integrates information on individual patient backgrounds (biomarkers, genetics, environment, and lifestyle) with conventional medical examination information. Currently, various biomarkers and other new examination indices are being investigated, and studies on periodontal disease-related genes and the complexity of oral bacteria are underway. This review discusses the possibilities and future challenges of precision periodontics and describes the new generation of laboratory methods and advanced periodontal disease treatment approaches as the basis for this new field.

Inherent metallic species retained by coal char or coke, such as Na and Ca, behave as catalysts in gasification. The char/coke normally contains inherent SiO2, which can react with the inherent catalysts to form silicates, resulting in catalyst deactivation over the range of pyrolysis, carbonization and gasification, and thereby reducing the char/coke reactivity. The present authors simulated the inherent catalyst deactivation experimentally by blending a Victorian lignite with SiO2, briquetting the SiO2/lignite blend, carbonizing the briquette, and then gasifying the coke with CO2. The kinetic analysis of the gasification employed a comprehensive model, which assumed progress in parallel of non-catalytic and catalytic gasification. The model quantitatively described the measured kinetics of the coke gasification with different SiO2 contents over a range of coke conversion up to 99.9%. The kinetic analysis revealed that the SiO2 deactivated substantial and entire portions of the most active catalyst and its precursor, respectively, before the gasification (i.e., during the carbonization). The catalyst deactivation also occurred during the gasification, but mainly following a self-deactivation mechanism that involved no silicates formation. Kinetics of thermal decomposition of benzene on lignite-derived char was investigated at 900 °C by applying a new method to continuously monitor the char surface activity. Benzene vapor was continuously forced to pass through a micro fixed bed of char with residence time as short as 7.6 ms, and then detected continuously by a flame-ionization detector. Results showed the presence of two different types of char surfaces; consumptive Type I surface and non-consumptive (sustainable) Type II surface. Type I surface of a partially CO2-gasified char had an capacity of carbon deposit from benzene over 20 wt%-char and an initial activity (represented by a first-order rate constant) as high as 160 s−1. Both of them decreased with increasing carbon deposit due to consumption of micropores accessible to benzene, and finally became zero leaving Type II surface that had a very stable activity with rate constant of 4 s−1. The chars without gasification had capacities of Type I surfaces smaller by two orders of magnitude than the partially gasified char, while the Type II surfaces had activities similar to that of the partially gasified char. It was found that Type II surface converted benzene into not only carbon deposit but also diaromatics and even greater aromatics. Composition of the greater aromatics was unknown because they were deposited onto the reactor wall immediately after passing through the char bed. Streptococcus pneumoniae is an important causative organism of respiratory tract infections. Although periodontal bacteria have been shown to influence respiratory infections such as aspiration pneumonia, the synergistic effect of S. pneumoniae and Porphyromonas gingivalis, a periodontopathic bacterium, on pneumococcal infections is unclear. To investigate whether P. gingivalis accelerates pneumococcal infections, we tested the effects of inoculating P. gingivalis culture supernatant (PgSup) into S. pneumoniae-infected mice. Mice were intratracheally injected with S. pneumoniae and PgSup to induce pneumonia, and lung histopathological sections and the absolute number and frequency of neutrophils and macrophages in the lung were analyzed. Proinflammatory cytokine/chemokine expression was examined by qPCR and ELISA. Inflammatory cell infiltration was observed in S. pneumoniae-infected mice and S. pnemoniae and PgSup mixed-infected mice, and mixed-infected mice showed more pronounced inflammation in lung. The ratios of monocytes/macrophages and neutrophils were not significantly different between the lungs of S. pneumoniae-infected mice and those of mixed-infected mice. PgSup synergistically increased TNF-α expression/production and IL-17 production compared with S. pneumoniae infection alone. We demonstrated that PgSup enhanced inflammation in pneumonia caused by S. pneumoniae, suggesting that virulence factors produced by P. gingivalis are involved in the exacerbation of respiratory tract infections such as aspiration pneumonia.

Apical periodontitis, an inflammatory lesion causing bone resorption around the apex of teeth, is treated by eradicating infectious bacteria from the root canal. However, it has a high recurrence rate and often requires retreatment. We investigated the bactericidal effect of antimicrobial photodynamic therapy (aPDT)/photodynamic antimicrobial chemotherapy (PACT) using indocyanine green (ICG)-loaded nanospheres coated with chitosan and a diode laser on a biofilm of Enterococcus faecalis, a pathogen of refractory apical periodontitis. Biofilm of E. faecalis was cultured in a porcine infected root canal model. ICG solution was injected into the root canal, which was then irradiated with a laser (810 nm wavelength) from outside the root canal. The bactericidal effect was evaluated by colony counts and scanning electron microscopy. The result of the colony counts showed a maximum 1.89 log reduction after irradiation at 2.1 W for 5 min. The temperature rise during aPDT/PACT was confirmed to be within a safe range. Furthermore, the light energy transmittance through the root was at a peak approximately 1 min after the start of irradiation, indicating that most of the ICG in the root canal was consumed. This study shows that aPDT/PACT can suppress E. faecalis in infected root canals with high efficiency.

We propose an evaluation methodology to analyze the safety level of advanced driver assistance systems (ADAS) as a human–machine systems in terms of comparing the increase in the safety level during normal system operation and the decrease in the safety level during a system malfunction. We propose a concept of combined error for the human–machine system and quantify this combined error by driving simulator investigations and simulation studies. First, we investigated the drivers’ behavior when avoiding rear-end collisions with a preceding vehicle when equipped with ADASs-like adaptive cruise control systems (ACC) and lane keeping assistance systems (LKA). Then, we confirmed that the risk of collision induced by overdependence on the systems was not increased when the ACC and LKA were mounted on the vehicle using simulation studies based on the concept of combined error for the human–machine system. We also confirmed that the decrease in collisions when the ADASs operated appropriately was much larger than the increase in collisions during a system malfunction.

Gelatin methacryloyl (GelMA) is a versatile biomaterial that has been used in various biomedical fields. UV light is commonly used to photocrosslink such materials; however, its use has raised several biosafety concerns. We investigated the mechanical and biological properties of a visible-wavelength (VW)-light-crosslinked gelatin-based hydrogel to evaluate its viability as a scaffold for bone regeneration in bone-destructive disease treatment. Irgacure2959 or riboflavin was added as a photoinitiator to create GelMA solutions. GelMA solutions were poured into a mold and exposed to either UV or VW light. KUSA-A1 cell-laden GelMA hydrogels were crosslinked and then cultured. Mechanical characterization revealed that the stiffness range of GelMA–RF hydrogel was suitable for osteoblast differentiation. KUSA-A1 cells encapsulated in GelMA hydrogels photopolymerized with VW light displayed significantly higher cell viability than cells encapsulated in hydrogels photopolymerized with UV light. We also show that the expression of osteogenesis-related genes at a late stage of osteoblast differentiation in osteoblasts encapsulated in GelMA–RF hydrogel was markedly increased under osteoblast differentiation-inducing conditions. The GelMA–RF hydrogel served as an excellent scaffold for the encapsulation of osteoblasts. GelMA–RF hydrogel-encapsulated osteoblasts have the potential not only to help regenerate bone mass but also to treat complex bone defects associated with bone-destructive diseases such as periodontitis.

Porphyromonas gingivalis Mfa1 fimbriae are thought to act as adhesion factors and to direct periodontal tissue destruction but their immunomodulatory actions are poorly understood. Here, we investigated the effect of Mfa1 stimulation on the immune and metabolic mechanisms of gingival fibroblasts from periodontal connective tissue. We also determined the role of Toll-like receptor (TLR) 2 and TLR4 in Mfa1 recognition. Mfa1 increased the expression of genes encoding chemokine (C-X-C motif) ligand (CXCL) 1, CXCL3, intercellular adhesion molecule (ICAM) 1 and Selectin endothelium (E) in gingival fibroblasts, but did not have a significant effect on genes that regulate metabolism. Mfa1-stimulated up-regulation of genes was significantly suppressed in Tlr4 siRNA-transfected cells compared with that in control siRNA-transfected cells, which indicates that recognition by TLR4 is essential for immunomodulation by Mfa1. Additionally, suppression of Tlr2 expression partially attenuated the stimulatory effect of Mfa1. Overall, these results help explain the involvement of P. gingivalis Mfa1 fimbriae in the progression of periodontal disease.

Interleukin (IL)-35 is an immunosuppressive cytokine mainly produced by regulatory T cells. IL-35 mediates immunological functions by suppressing the inflammatory immune response. However, the role of IL-35 in bone-destructive diseases remains unclear, especially in terms of osteoclastogenesis. Therefore, the current study investigated the synergistic effect of IL-35 on osteoclastogenesis that is involved the pathogeneses of periodontitis and rheumatoid arthritis. Osteoclastic differentiation and osteoclastogenesis of RAW264 (RAW) cells induced by receptor activator of nuclear factor (NF)-κB ligand (RANKL) and IL-35 were evaluated by tartrate-resistant acid phosphate staining, hydroxyapatite resorption assays, and quantitative polymerase chain reaction. The effect of IL-35 on RANKL-stimulated signaling pathways was assessed by Western blot analysis. Costimulation of RAW cells by RANKL and IL-35 induced osteoclastogenesis significantly compared with stimulation by RANKL alone. Phosphorylations of extracellular signal-regulated kinase (ERK) and p38 mitogen-activated protein kinase tended to be increased by RANKL and IL-35 compared with RANKL or IL-35 alone. Additionally, the osteoclastogenesis induced by RANKL and IL-35 was suppressed by inhibition of ERK. In this study, IL-35 and RANKL induced osteoclastogenesis synergistically. Previous reports have shown that IL-35 suppresses the differentiation of osteoclasts. Therefore, IL-35 might play dual roles of destruction and protection in osteoclastogenesis.

This study aimed to investigate the effect of carbon derived from glucose (C) on the physicochemical characteristics and catalytic activity of Ni, supported over SiO2, ZSM-5, and TiO2 in methane dry reforming. Among the Ni catalysts without C, Ni/SiO2 exhibited the highest CH4-CO2 conversion and stability at all experimented temperatures. On the other hand, the C-incorporated catalysts prepared by glucose impregnation, followed by pyrolysis, showed dissimilar performances. C improved the stability of Ni/SiO2 in the reforming at 650 °C and 750 °C and increased the CH4 and CO2 conversion to the level close to the thermodynamic equilibrium at 850 °C. However, this element did not substantially affect the activity of Ni/ZSM-5 and exerted a retarding effect on Ni/TiO2. Characterizations with H2-TPD, XRD, EXAFS, and STEM-EDS revealed that the different influences of C by the supports were attributed to the extent of metal dispersion and metal-support interaction.

Angiopoietin-like protein 2 (ANGPTL2) maintains tissue homeostasis by inducing inflammation and angiogenesis. It is produced in infiltrating immune cells or resident cells, such as adipocytes, vascular endothelial cells, and tumor cells. We hypothesized that ANGPTL2 might play an important role as a unique mediator in both systemic and periodontal disease. We demonstrated an increased ANGPTL2 concentration in gingival crevicular fluid from chronic periodontitis patients. Porphyromonas gingivalis lipopolysaccharide (LPS) treatment strongly induced ANGPTL2 mRNA and protein levels in Ca9-22 human gingival epithelial cells. Recombinant human ANGPTL2 increased interleukin 1β (IL-1β), IL-8, and tumor necrosis factor-α (TNF-α) mRNA and protein levels in Ca9-22 cells. In this study, various types of ionic liquids (ILs) were examined for catalytic activity in the pyrolysis of cellulose for the production of levoglucosenone, which is a valuable and versatile compound for the synthesis of a variety of novel compounds. Cellulose was simply mixed with the ILs and subjected for the pyrolysis, typically at 300 °C, to produce volatile products, including levoglucosenone, separated from the ILs phase. The type of IL anion significantly affected the catalysis, and the use of ILs bearing sulfonate anion resulted in distinguished yields of levoglucosenone and IL recoveries for the reutilization. Detailed thermogravimetric analysis and discussion on properties of ILs revealed the active and thermally stable nature of the sulfonate ILs. Catalytic pyrolysis with those ILs was applied to the conversion of other saccharides composed of glucose molecules, resulting in a preferential formation of levoglucosenone but at low yields as compared to that from cellulose.

Periodontitis is a chronic inflammatory disease that affects the periodontium. Recent studies suggest an association between periodontal and cardiovascular diseases. However, the detailed molecular mechanism is unknown. A previous study has demonstrated that experimental periodontitis induces serum amyloid A (SAA) in the liver and peripheral blood of ApoE-deficient mice as an atherosclerosis model. SAA is an acute-phase protein that affects systemic inflammation. The aim of this study is to investigate the atherosclerosis-onset mechanism using human aortic endothelial cells (HAECs) stimulated by SAA in vitro. Atherosclerosis PCR array and qPCR analyses showed upregulation of adhesion molecules such as intercellular adhesion molecule-1, vascular cell adhesion molecule-1, and E-selectin in HAECs upon SAA stimulation. In addition, the results demonstrated that Toll-like receptor, TLR2, could serve as an important receptor of SAA in HAECs. Furthermore, small interfering RNA (siRNA) against TLR2 inhibited the upregulation of adhesion molecules in HAECs stimulated by SAA. Our results suggest that SAA stimulates the expression of adhesion molecules via TLR2. SAA could be an important molecule for atherosclerosis induced by periodontal disease.

Antimicrobial photodynamic therapy (aPDT) has been proposed as an adjunctive strategy for periodontitis treatments. However, use of aPDT for periodontal treatment is complicated by the difficulty in accessing morphologically complex lesions such as furcation involvement, which the irradiation beam (which is targeted parallel to the tooth axis into the periodontal pocket) cannot access directly. The aim of this study was to validate a modified aPDT method that photosensitizes indocyanine green-loaded nanospheres through the gingivae from outside the pocket using a diode laser. To establish this trans-gingival irradiation method, we built an in vitro aPDT model using a substitution for gingivae. Irradiation conditions and the cooling method were optimized before the bactericidal effects on Porphyromonas gingivalis were investigated. The permeable energy through the gingival model at irradiation conditions of 2 W output power in a 50% duty cycle was comparable with the transmitted energy of conventional irradiation. Intermittent irradiation with air cooling limited the temperature increase in the gingival model to 2.75 °C. The aPDT group showed significant bactericidal effects, with reductions in colony-forming units of 99.99% after 5 min of irradiation. This effect of aPDT against a periodontal pathogen demonstrates the validity of trans-gingival irradiation for periodontal treatment.
