Prostate cancer is the most common cancer among men and has a high incidence and associated mortality worldwide. It is an androgen-driven disease in which tumor growth is triggered via ligand-mediated signaling through the androgen receptor (AR). Recent evidence suggests that the widespread use of effective AR pathway inhibitors may increase the occurrence of neuroendocrine prostate cancer (NEPC), an aggressive and treatment-resistant AR-negative variant; however, mechanisms controlling NEPC development remain to be elucidated. Various preclinical models have recently been developed to investigate the mechanisms driving the NEPC differentiation. In the present study, we summarized strategies for the development of NEPC models and proposed a novel method for model evaluation, which will help in the timely and accurate identification of NEPC by virtue of its ability to recapitulate the heterogeneity of prostate cancer. Moreover, we discuss the origin and the mechanism of NEPC. The understanding of the regulatory network mediating neuroendocrine differentiation presented in this review could provide valuable insights into the identification of novel drug targets for NEPC as well as into the causes of antiandrogenic drug resistance.

Yarn linear density and linear density unevenness between fragments involve the mass and mass variation of yarn per unit length, which are important indices to reflect the uniformity of yarn thickness. Aiming at the shortcomings of the traditional testing method, which uses a yarn length tester to test these indices by counting length first and then cutting and measuring weight, a testing device that measures weight and counts length synchronously is designed and developed, so that the yarn can be continuous, recyclable, and reusable. Length counting is conducted by a length-counting disk connected to a photoelectric coded disk, and the result of length counting can be accurate to 0.01 m. The original skein frame with a perimeter of 1 m is replaced, so that the error caused by yarn overlapping is avoided. Through program control, the testing of a plurality of groups of linear density values can be completed at one time to calculate the linear density unevenness of different fragments of yarn, and the yarn can be led to a recovery spool through a yarn guide cylinder to form a new package. Polyester/viscose 65/35 blended yarn was taken as a test sample and subjected to statistical analysis using SPSS software. It is found that the results of the newly developed device are closer to the arbitration value; the whole experiment is completed at one time, which avoids the secondary error and reduces the labor intensity; and the raw materials can be recycled, which saves labor and raw material costs. The device has high value for industrialization and popularization.

In this work, a polypropylene (PP) porous film was made-up by melt electrospinning. The fibrous film was coated by a superthin layer of silicon dioxide (SiO2) gel. The surface morphology of the SiO2 gel was decoded by scanning electron microscopy and atomic force microscopy. The SiO2 gel interface with film, as well as its crystallinity, were characterized by Fourier transform infrared spectroscopy, energy dispersive spectroscopy, and X-ray diffraction. The PP composite film's thermomechanical properties were studied through thermal shrinkage, thermal gravimetric analysis, dynamic mechanical analysis, and an Instron tensile machine with a heating chamber. The results showed that the coated SiO2 gel network could effectively reduce the PP film's thermal shrinkage by 48.5% without change of crystallinity. The coated SiO2 gel is capable of enlarging the decomposition temperature range and the storage modulus of the PP film. Meanwhile, it was discovered that, along with the increase of measured temperature, the loads on both pure PP film and PP/SiO2 gel film decreased under a constant strain, or the tensile strain of both of them was enhanced under the same load. The solid gel network endows the PP electrospun film with relatively higher thermal safety.

Neuroendocrine transdifferentiation (NED) of prostate cancer (PCa) is the main cause of failure of androgen receptor inhibitor treatment. However, the molecular mechanisms underlying the development of NEPC, especially treatment-induced NEPC, remain unclear. Emerging evidence indicates that elevated monoamine oxidase A (MAOA) contribute to the proliferation, cell stemness, and bone metastasis in PCa. Here, we generated an enzalutamide-induced NED cell model to assess the role of MAOA during NED. Overall, MAOA expression was significantly increased upon Enz long-term exposure and was required for neuroendocrine marker expression. In particular, Enz was found to induce NED via the MAOA/mTOR/HIF-1α signaling axis. Further analyses revealed that the MAOA inhibitor clorgyline(CLG) may bring multiple benefits to CRPC patients, including better therapeutic effect and delays NED. These findings suggest that MAOA may be an important target for the development of anti-NED therapies, thereby providing a novel strategy for the combined application of CLG and AR inhibitors in the clinic.

Through biogenetic engineering, the posterior silk glands of Bombyx mori larvae are degenerated, and only the middle silk glands are retained to secrete sericin to produce sericin fibers, so as to obtain sericin cocoons with high sericin content (>98.5%). For this paper, sericin cocoons were studied, and the morphological structure characteristics and properties of sericin cocoons and common Bombyx mori cocoons were compared and analyzed through such modern test techniques as SEM, FTIR, and XRD. Results show that sericin cocoons have the basic morphological structure of cocoons, but because of the brittleness and hardness of sericin, the ssericin cocoons have poor integrity with a thermal decomposition rate that is higher than that of cocoons; the two cocoons are of a similar protein structure, with both α-helix and β-sheet structures, and dominantly the β-sheet structure; and the crystallinity of sericin cocoons (10.48%) is lower than that of the common Bombyx mori cocoons (27.10%). Sericin was extracted from both cocoons to obtain a mixed sericin solution respectively, which were coated on base fabrics of polyester taffeta (210T) for coating finish and subjected to qualitative research for their anti-UV properties. The base fabrics coated with mixed sericin solutions of sericin cocoons and Bombyx mori cocoons have an anti-UV performance index (UPF) of 23.9% and 22.5% respectively, which are higher than that of the base fabric (8.9%).

In this work, the effects of degumming treatment on the structure and properties of OS-BADF were discussed. Alkali degumming and biological enzyme degumming were used to obtain AD-BADF and BED-BADF respectively. Spinnability index, thermal property, macromolecular structure, bending property and antibacterial property of OS-BADF (Original Sample of Bast Fiber of Alchornea Davidii Franch), AD-BADF (Alkali Degummed Bast Fiber of Alchornea Davidii Franch), and BED-BADF (Biological Enzyme degummed Bast Fiber of Alchornea Davidii Franch) were tested. The surface morphology of the above three kinds of fiber was analyzed by SEM. The results showed that the main component of degummed OS-BADF was cellulose, and the fiber had stripes in longitudinal direction, and kidney ellipsoid, irregular polygon in the cross section. After degumming treatment, the fineness of fiber decreased obviously and reached the spinnability index. Non-cellulose substances in OS-BADF can be removed by degumming. SEM observation showed that AD-BADF and BED-BADF became smoother after degumming. Compared with AD-BADF, BED-BADF had finer linear density, better thermal stability and higher antibacterial property. The equivalent flexural modulus and flexural rigidity of AD-BADF and BED-BADF were decreased compared with OS-BADF. In particular, the equivalent flexural modulus and the flexural rigidity of BED-BADF were mostly reduced, which indicated that BED-BADF became thinner and softer. The initial thermal decomposition temperature and the highest thermal decomposition temperature of AD-BADF and BED-BADF had been significantly increased. And -OH (free group) decreased significantly after degumming treatment. The testing results of the antibacterial properties of the three blended fabrics showed that the antibacterial properties of AD-BADF and BED-BADF were improved. Compared with AD-BADF, the antibacterial properties of BED-BADF were improved significantly. On the one hand, this research can provide a basis for technological research for the development of ADF (Alchornea Davidii Franch) bast fiber blended yarn and its textile product development. On the other hand, because of the high mechanical properties of ADF bast fiber, it also provides new development idea for the development of natural high-performance fiber reinforcement composites.

The aim of the present study was to investigate the effects and mechanisms of 17‑AAG combined with salinomycin treatment on proliferation and apoptosis of the SGC‑7901 gastric cancer cell line. An MTT assay was used to detect the proliferation of SGC‑7901 cells. Morphological alterations of cells were observed under inverted phase‑contrast and fluorescence microscopes. Cell cycle and apoptosis were assessed by flow cytometry analysis. The protein expression of nuclear factor (NF)‑κB p65 and Fas‑ligand (L) were evaluated by immunocytochemistry. Salinomycin with a concentration range of 1‑32 µmol/l was demonstrated to inhibit growth of SGC‑7901 cells effectively, affect the morphology and apoptosis rate of cells, and arrest SGC‑7901 cells in S phase. Furthermore, salinomycin significantly increased the protein expression of Fas‑L and decreased the protein expression of NF‑κB p65. The alterations in SGC‑7901 cells co‑treated with salinomycin and 17‑AAG were more significant compared with cells treated with one drug only. In conclusion, the individual use of salinomycin and combined use with 17‑AAG may significantly inhibit SGC‑7901 gastric cancer cell proliferation and induce cell apoptosis. The potential mechanisms may be associated with upregulation of Fas‑L and downregulation of NF‑κB. These results provide a basis for the potential use of salinomycin in gastric cancer treatment.

The phase, luminescence spectrum, chromaticity and afterglow properties of rare-earth strontium aluminate luminescent fibers, which appear the primary colors of red, yellow and blue in the sun, were measured by XRD, fluorescence spectrometer, spectral scanning-meter and illuminometer. With the analysis, the experiment results indicated that the emission peak of yellow and blue luminescent fibers were located at 520 nm, which were caused by transition of Eu2+. The crystal phase structure of red luminescent fiber was destructed, the yellow light emission being created at 580 nm. This may be the transition of Dy3+ of rare-earth strontium aluminate. The three kinds of chromatic light of fiber are yellow, yellow-green and green light successively (The illumination strength is declining with the order of rare-earth strontium aluminate, yellow, blue and red). Comparing to rare-earth strontium aluminate, the afterglow duration of luminescent fibers were extended.

This study is to investigate the therapeutic effects of bone marrow mesenchymal stem cells (BMSCs) expressing interleukin (IL)-12 on malignant ascites tumor-bearing mice and the related mechanisms. Malignant ascites tumor mouse model was established by the intraperitoneal inoculation with MethA or H22 tumor cells. Mouse BMSCs were transfected with lentiviral vector containing IL-12, and then transplanted into these mouse models via intraperitoneal injection. The peritoneal permeability in these mice was evaluated and compared. The contents of INF-γ and VEGF in ascites were determined by ELISA. Mouse models receiving IL-12-expressing BMSCs were rechallenged with tumor cells, and the animal survival was observed and analyzed. In both MethA and H22 tumor cell-induced malignant ascites tumor mouse models, there were no significant differences in the peritoneal permeability between the normal saline (NS), BMSC-control, and BMSC-null groups. However, compared with NS control group, the peritoneal permeability was significantly decreased by IL-12-expressing BMSCs. Moreover, ELISA showed that, in both the MethA and H22 tumor cell-induced mouse models, compared with the NS control group, the contents of INF-γ in ascites were significantly elevated, while the contents of VEGF in ascites were significantly decreased, in the BMSC-IL-12 groups. In addition, IL-12-expressing BMSCs significantly elongated the survival of mouse models after rechallenging with tumor cells. IL-12-expressing BMSCs exert protective effects against malignant ascites tumor, and the anti-tumor effects might be associated with the enhanced anti-tumor immunity. Our findings might bring new insights into the treatment of tumors with immunotherapy.

To explore the association between LMNA gene mutation and familiar dilated cardiomyopathy (DCM) (FDCM) and idiopathic DCM (IDCM) in Uygurs and Hans people in Xinjiang area.Peripheral blood samples were collected from 28 family member with FDCM and 123 sporadic patients with IDCM(56 Uygur patients and 67 Han patients), 80 Uygur and 80 Han people were chosen as normal controls. PCR was used to amplify the 12 exons of LMNA gene. The amplified products were sequenced and compared with the standard sequence in the NCBI to determine the mutation sites.Transmission of the allele C and T of rs4641 was similar in Han FDCM patients. One new variation(c.1714CT) located at exon 10 of LMNA gene was identified in 1 Han patient with IDCM, this mutation caused an amino acid substitution (R572C). In Uygurs people, rs553016 polymorphism was significant different between IDCM and control groups (P0.05). Logistic regression revealed that rs553016 was an independent risk factor for Uygurs patients with IDCM (OR = 3.178, P = 0.035).LMNA rs4641 is not associated with FDCM of Hans people in Xinjiang while LMNA mutation is associated with IDCM and rs533106 polymorphism is an independent risk factor for Uygurs patients with IDCM.

To study the effect of the HSP90 inhibitor, 17-allylamino-17-demethoxygeldanamycin (17-AAG), on cell proliferation and apoptosis of human cancer SGC-7901 cells and explore the mechanisms.The inhibitory effect of 17-AAG on the proliferation and morphology of SGC-7901 cells was assessed with MTT assay and DNA-PI staining, respectively. Flow cytometry was employed to analyze the changes in cell cycle and apoptosis of the cells following 17-AAG exposure. The cellular expression of Fas protein was detected by immunohistochemistry.17-AAG significantly suppressed the proliferation of SGC-7901 cells in a time- and dose-dependent manner. After treatment with 17-AAG for 48 h, SGC-7901 cells showed cell cycle arrested at G(2)/M stage, and the cell apoptosis rate increased with the 17-AAG concentration. The expression of Fas protein in the cytoplasm of SGC-7901 cells increased gradually with the increase of 17-AAG concentration.17-AAG can induce apoptosis, alters the cell cycle distribution and up-regulates the expression of Fas protein in SGC-7901 cells to suppress the cell proliferation.

The purpose of this study was to clone human HSP90beta cDNA and construct its eukaryote expression vector . The total RNA was isolated by TRIzol Reagent (Invitrogen) from human NPC and its cDNA was gained by RT-PCR. The purified RT-PCR products and PGEM-T Easy Vector were ligated and transformed into XL1-blue E. coli bacteria. The white clones were selected and the plasmid was purified, which was further identified by double enzyme digestion and sequenced. PGEM-hHSP90beta and pcDNA3.1(+) DNA were digested by AflII and Xbal respectively. After purification, the two fragments obtained were ligased by using T4 DNA ligase (Fermentas). This recombinant DNA was then transformed into E. coli Competent Cells XL1-blue and positive clones were selected on the LB agarose plate containing Ampr (100 microg/ml). Single clones were identified by double digestion with AflII and Xbal, and two fragments with the size 5.4 kb and 2.1 kb were produced as expected. The hHSP90beta gene was successfully inserted into the eukaryote expression vector pcDNA3.1(+) by the recombination technique in vitro.

To study the expression of HSP70 and HSP90beta and its clinical significance in human nasopharyngeal carcinoma.Immunohistochemical method SP was used to detect the expression of HSP70 and HSP90beta in 50 cases and its clinical significance were studied.The positive rate of HSP70 and HSP90beta were 72% and 56% respectively. Analysis of patients, survival demonstrated that the prognosis of NPC with HSP70 (-) and HSP90beta (-) expression were significantly better than the others.The overexpression of HSP70 and HSP90beta were probably concerned with the occurrence, development and prognosis of nasopharyngeal carcinoma.
