Corallium species have ecological, cultural and commercial importance and thus require tools to assist with their identification for both management and trade. The organic matrix (OM) of the skeletons of four Corallium species (C. rubrum, C. konojoi, C. secundum and C. elatius) was examined to provide insight into the biomineralization process and to develop a new tool of identification. The pattern of OM and the set of soluble organic matrix proteins (SOM) in the skeletons were examined by gel electrophoresis. Staining of cross-sections of skeletons showed a common cyclic, concentric pattern of OM during growth. Differences in molecular weight and isoelectric point were observed for proteins in the SOM in different Corallium species but not among different populations of Corallium rubrum. Immunolabeling with antibodies against the SOM of C. rubrum showed labeling of the OM of the three other Corallium species suggesting the presence of common epitopes.

International audience; Regeneration of artificially induced lesions was monitored in nubbins of the branching coral Acropora muricata at two reef-flat sites representing contrasting environments at Réunion Island (21u079S, 55u329E). Growth of these injured nubbins was examined in parallel, and compared to controls. Biochemical compositions of the holobiont and the zooxanthellae density were determined at the onset of the experiment, and the photosynthetic efficiency (F v /F m) of zooxanthellae was monitored during the experiment. Acropora muricata rapidly regenerated small lesions, but regeneration rates significantly differed between sites. At the sheltered site characterized by high temperatures, temperature variations, and irradiance levels, regeneration took 192 days on average. At the exposed site, characterized by steadier temperatures and lower irradiation, nubbins demonstrated fast lesion repair (81 days), slower growth, lower zooxanthellae density, chlorophyll a concentration and lipid content than at the former site. A trade-off between growth and regeneration rates was evident here. High growth rates seem to impair regeneration capacity. We show that environmental conditions conducive to high zooxanthellae densities in corals are related to fast skeletal growth but also to reduced lesion regeneration rates. We hypothesize that a lowered regenerative capacity may be related to limited availability of energetic and cellular resources, consequences of coral holobionts operating at high levels of photosynthesis and associated growth.

Background: Sclerites of alcyonarian species are biominerals formed of organic matrix molecules trapped inside a mineral inorganic fraction. Results: Scleritin is a small basic organic matrix protein, synthetized by scleroblasts and incorporated into sclerites. Conclusion: Scleritin is the first organic matrix protein fully characterized and localized in the sclerites of C. rubrum. Significance: Scleritin provides information on the biomineralization pathway in C. rubrum., We report here for the first time the isolation and characterization of a protein from the organic matrix (OM) of the sclerites of the alcyonarian, Corallium rubrum. This protein named scleritin is one of the predominant proteins extracted from the EDTA-soluble fraction of the OM. The entire open reading frame (ORF) was obtained by comparing amino acid sequences from de novo mass spectrometry and Edman degradation with an expressed sequence tag library dataset of C. rubrum. Scleritin is a secreted basic phosphorylated protein which exhibits a short amino acid sequence of 135 amino acids and a signal peptide of 20 amino acids. From specific antibodies raised against peptide sequences of scleritin, we obtained immunolabeling of scleroblasts and OM of the sclerites which provides information on the biomineralization pathway in C. rubrum.

The capacity of corals for repairing partial mortality is a fundamental determinant of reef resilience. This capacity was assessed in the major reef-building coral Porites lutea by monitoring the regeneration of artificially induced lesions of standard size (330 ± 50 mm2, 3 mm deep) in 4 shallow reef flat populations at Réunion Island, under different environmental conditions related to sites and seasons, during a period without positive temperature anomalies. An exponential decay model with an asymptote described the lesion regeneration through 14 experiments. In spite of fast initial lesion regeneration, limited capacity for repair in this massive coral was indicated, as only 18% of the inflicted lesions healed completely within 6 to 9 mo. Lesion regeneration was fastest and most complete in the cooling and cool seasons, and may be impaired during the warming and hot seasons. Both solar radiation and seawater temperature contributed to seasonal changes in regeneration capacity, although they had opposite effects. While high radiation during the warming season decreased lesion regeneration rate, potentially through reduction of the photosynthetic efficiency of zooxanthellae, high temperature boosted it but also increased the amount of lesion area that could not be regenerated. Study sites were characterised by different temperature and radiation regimes, but these parameters alone could not explain all site effects on lesion regeneration capacity. Additional factors, most probably chronic stress caused by inputs of organic matter and run-off from land, may further compromise the regeneration capacity of corals and the resilience of coral populations.

We analysed the soluble organic matrix (SOM) of two biominerals formed by the same organism but differing by their morphological characteristics: the axial skeleton and the sclerites of Corallium rubrum. The results of 1D SDS-PAGE electrophoresis show for the two biominerals that SOM proteins bands have similar apparent molecular weight but differ in quantity. Further analysis by 2D electrophoresis reveals each protein band as a line of spots with different isoelectric points. Our results suggest that each SOM protein band consists of a mix of proteins and/or one unique protein with post-translational modifications. By immunohistochemistry, we show that antibodies raised against the SOM of axial skeleton and sclerites label the SOM of the two biominerals but also label the insoluble organic matrix suggesting the presence of common epitopes between the two biominerals and the two organic fractions.
