Abstract

Abstract Background Dysphonia is a frequent comorbidity of asthma and has been suggested to be a local side effect of inhaled corticosteroids due to laryngeal candidiasis. We hypothesized that dysphonia in asthmatics was not due to laryngeal organic lesions but to laryngeal dysfunction during phonation (LDP). Objective We compared the frequency of LDP in female asthmatic patients treated with inhaled corticosteroids to female controls. Methods We compared 68 asthmatic female patients to 53 female control subjects. Pulmonary function tests were performed and the asthmatic patients classified according to the level of inhaled corticosteroids. Dysphonia was defined as a Vocal Handicap Index ≥18 or GRBAS score ≥2. Abstract

International audience; The paradigm for all toxicological bioassays in the risk assessment of pesticide registration reflects the principle that experimental conditions should be controlled to avoid any other factors that may affect the endpoint measures. As honeybee colonies can be frequently exposed to bio-aggressors in real conditions, often concomitantly with pesticides, co-exposure to pesticide/bio-aggressors is becoming a concern for regulatory authorities. We investigated the effects of the neonicotinoid insecticide thiamethoxam on the homing performances of foragers emerging from colonies differentiated by health status (infestation with Varroa destructor mites, microsporidian parasite Nosema spp. and Deformed Wing Virus). We designed a homing test that has been recently identified to fill a regulatory gap in the field evaluations of sublethal doses of pesticides before their registration. We also assessed the effect of temperature as an environmental factor. Our results showed that the Varroa mite exacerbates homing failure (HF) caused by the insecticide, whereas high temperatures reduce insecticide-induced HF. Through an analytical Effective Dose (ED) approach, predictive modeling results showed that, for instance, ED level of an uninfested colony, can be divided by 3.3 when the colony is infested by 5 Varroa mites per 100 bees and at a temperature of 24°C. Our results suggest that the health status of honeybee colonies and climatic context should be targeted for a thorough risk assessment.

Abstract

Autism spectrum disorder (ASD) is a developmental disorder underdiagnosed in adults. To date, no consistent evidence of alterations in brain structure has been reported in adults with ASD and few studies were conducted at that age. We analyzed structural magnetic resonance imaging data from 167 high functioning adults with ASD and 195 controls. We ran our analyses on a discovery (n = 301) and a replication sample (n = 61). The right caudal anterior cingulate cortical thickness was significantly thinner in adults with ASD compared to controls in both the discovery and the replication sample. Our work underlines the relevance of studying the brain anatomy of an adult ASD population.

Abstract

International audience; Purpose: Head and neck paragangliomas (HNPGLs) can relapse after primary treatment. Optimal imaging protocols have not yet been established for posttreatment evaluation. The aim of the present study was to assess the diagnostic value of 18F-FDOPA PET/CT and MR/CT angiography (MRA/CTA) in HNPGL patients with clinical relapse during their follow-up.Methods: Sixteen consecutive patients presenting with local pain, tinnitus, dysphagia, hoarse voice, cranial nerve involvement, deafness, or retrotympanic mass appearing during follow-up after the initial treatment of HNPGLs were retrospectively evaluated. Patients underwent both 18F-FDOPA PET/CT and MRA (15 patents) or CTA (1 patent). Both methods were first assessed under blinded conditions and afterwards correlated. Head and neck imaging abnormalities without histological confirmation were considered true-positive results based on a consensus between radiologists and nuclear physicians and on further 18F-FDOPA PET/CT and/or MRA.Results: 18F-FDOPA PET/CT and MRA/CTA were concordant in 14 patients and in disagreement in 2 patients. 18F-FDOPA PET/CT and MRA/CTA identified, respectively, 12 and 10 presumed recurrent HNPGLs in 12 patients. The two lesions diagnosed by PET/CT only were confirmed during follow-up by otoscopic examination and MRA performed 29 and 17 months later. 18F-FDOPA PET/CT images were only slightly influenced by the posttreatment sequelae, showing a better interobserver reproducibility than MRA/CTA. Finally, in 2 of the 16 studied patients, 18F-FDOPA PET/CT detected two additional synchronous primary HNPGLs.Conclusion: 18F-FDOPA PET/CT is highly sensitive in posttreatment evaluation of patients with HNPGLs, and also offers better interobserver reproducibility than MRA/CTA and whole-body examination. We therefore suggest that 18F-FDOPA PET/CT is performed as the first diagnostic imaging modality in symptomatic patients with suspicion of HNPGL relapse after primary treatment when 68Ga-labeled somatostatin analogues are not available.

Abstract

Depuis l’obtention de la premiere plante transgenique de riz en 1988, de larges progres ont ete accomplis pour manipuler les caracteres d’interet agronomique par ingenierie genetique chez cette plante d’importance alimentaire, economique et culturelle majeure. Les caracteres concernes vont de la resistance aux insectes ravageurs et aux pathogenes fongiques, bacteriens et viraux a la tolerance vis-a-vis des contraintes abiotiques (toxicites et carences minerales, temperature excessive ou basse, exces ou deficit en eau) en passant par les qualites nutritives ou industrielles du grain. Des portions entieres de voies metaboliques peuvent a present etre finement manipulees, permettant par exemple la production de β-carotene (provitamine A) dans l’albumen du grain, qui en est naturellement depourvu, ou l’accumulation des enzymes de la photosynthese C4 dans des tissus foliaires precis. La production de molecules d’interet therapeutique ou industriel dans le grain a aussi ete realisee. Bien que certaines de ces innovations soient pretes a etre transferees dans le champ du riziculteur depuis plusieurs annees, les politiques publiques sont encore hesitantes a franchir le pas. Ce retard relatif a ete mis a profit pour conduire de nombreuses etudes sur les impacts alimentaires et environnementaux lies a la consommation et au deploiement de ces nouvelles cultures.

Abstract

Le riz ( Oryza sativa L.) a le double statut de cereale la plus consommee par l’homme et d’espece modele pour les monocotyledones. Elucider la fonction des genes sous-tendant les principaux caracteres d’interet agronomique chez cette plante contribuerait donc a accelerer non seulement l’amelioration varietale du riz mais aussi celle des autres cereales. La transformation genetique est un outil permettant d’accelerer la decouverte de la fonction des genes et l’innovation varietale. La premiere plante transgenique de riz a ete obtenue des 1988, et, depuis, les technologies ont regulierement evolue vers plus d’efficacite et de precision. Nous presentons ici les grandes etapes de l’evolution de ces techniques, ainsi que leurs applications pour l’analyse fonctionnelle des genes. En particulier, les perspectives ouvertes par de nouveaux sauts technologiques en cours, qui permettent a present une modification ciblee des genomes sans introduction d’acide desoxyribonucleique (ADN) superflu, seront illustrees et discutees.

Abstract

Background: It is usual to incriminate inhaled corticosteroids in the occurrence of dysphonia in asthmatics. For a long time, local adverse effects of inhaled corticosteroid therapy have been supposed to be the cause of dysphonia. We hypothesized that dysphonia in asthmatics were mostly functional and not based on organic lesions. Aims: To investigate, prospectively, what could be the cause of dysphonia occurring in women with a persistent asthma treated with inhaled corticosteroids. Methods: Women9s outpatients with persistent asthma, aged 18-65 years, treated with inhaled corticosteroids were recruited at the chest diseases department, and compared with controls without asthma. Subjects with higher smoking 5 pack-years, symptomatic gastroesophageal reflux, obesity, laryngeal surgery history and an organic abnormal vocal cord unattributed to a vocal forcing were excluded. Tests to assess a vocal dysfunction were performed : Vocal Handicap Index, GRBAS scale, phonation quotient, fundamental laryngeal tests and laryngeal stroboscopic video. A functional dysphonia was defined as having one of these tests anormal. Results: 68 asthmatics, 51 controls were selected. In the asthmatic group 87.3% had dysphonia vs 12.7% (p=10 -4 ). Among the dysphonic asthmatic patients 80% had a functional dysphonia (IC : 67-90). Laryngeal organic lesions were not seen in the asthmatic group and only one in the control group. No asthmatic patient had a laryngeal mycosis. Conclusion: For the 1st time we have demonstrated that dysphonia occurring in asthmatic women who used inhaled corticosteroids was mostly due to a functional mechanism and not as a consequence of organic lesions (like inflammation or mycosis).

Abstract

DNA replication is a key determinant of chromosome segregation and stability in eukaryotes. The yeast Saccharomyces cerevisiae has been extensively used for cell cycle studies, yet simple but key parameters such as the fraction of cells in S phase in a population or the subnuclear localization of DNA synthesis have been difficult to gather for this organism. 5-ethynyl-2'-deoxyuridine (EdU) is a thymidine analogue that can be incorporated in vivo and later detected using copper-catalyzed azide alkyne cycloaddition (Click reaction) without prior DNA denaturation. This chapter describes a budding yeast strain and conditions that allow rapid EdU incorporation at moderate extracellular concentrations, followed by its efficient detection for the analysis of DNA replication in single cells by flow cytometry and fluorescence microscopy.

Abstract

Correspondance: e-mail: guiderdoni@cirad.fr; International audience; Retrotransposons are mobile genetic elements, ubiquitous in Eukaryotic genomes, which have proven to be major genetic tools in determining phylogeny and structuring genetic diversity, notably in plants. We investigate here the diversity of the Ty1-copia retrotransposon Tos17 in the cultivated rice of Asian origin (Oryza sativa L.) and related AA genome species of the Oryza genus, to contribute understanding of the complex evolutionary history in this group of species through that of the element in the lineages. In that aim, we used a combination of Southern hybridization with a reverse transcriptase (RT) probe and an adapter-PCR mediated amplification, which allowed the sequencing of the genomic regions flanking Tos17 insertions. This analysis was carried out in a collection of 47 A-genome Oryza species accessions and 202 accessions of a core collection of Oryza sativa L. representative of the diversity of the species. Our Southern hybridization results show that Tos17 is present in all the accessions of the A-genome Oryza species, except for the South American species O. glumaepatula and the African species O. glaberrima and O. breviligulata. In O. sativa, the number of putative copies of Tos17 per accession ranged from 1 to 11 and multivariate analysis based on presence/absence of putative copies yielded a varietal clustering which is consistent with the isozyme classification of rice. Adapter PCR amplification and sequencing of flanking regions of Tos17 insertions in A-genome species other than O. sativa, followed by anchoring on the Nipponbare genome sequence, revealed 13 insertion sites of Tos17 in the surveyed O. rufipogon and O. longistaminata accessions, including one shared by both species. In O. sativa, the same approach revealed 25 insertions in the 6 varietal groups. Four insertion sites located on chromosomes 1, 2, 10, and 11 were found orthologous in O. rufipogon and O. sativa. The chromosome 1 insertion was also shared between O. rufipogon and O. longistaminata. The presence of Tos17 at three insertion sites was confirmed by retrotransposon-based insertion polymorphism (RBIP) in a sample of O. sativa accessions. The first insertion, located on chromosome 3 was only found in two japonica accessions from the Bhutan region while the second insertion, located on chromosome 10 was specific to the varietal groups 1, 2, and 5. The third insertion located on chromosome 7 corresponds to the only insertion shown active in rice so far, notably in cv. Nipponbare, where it has been extensively used for insertion mutagenesis. This copy was only found in a few varieties of the japonica group 6 and in one group 5 accession. Taken together, these results confirm that Tos17 was probably present in the ancestor of A-genome species and that some copies of the element remained active in some Oryza lineages--notably in O. rufipogon and O. longistaminata--as well as in the indica and japonica O. sativa L. lineages.
