Genes of tolerance for herbicide has become one of the options more used in world level for the weed control. This study had as objective to evaluated crosses between tolerant irrigated rice plants and sensitive to herbicides of the imidazolinone chemical group, through the evaluation of F2 populations. Five genotypes of irrigated rice were crossed. Three of these parents are sensitive to the herbicide: BRS Querencia, BRS Fronteira and BRS Pampa and two genotypes, BRS Sinuelo CL and Puita INTA CL, tolerant to the herbicide. F1 seeds were obtained in greenhouse, and generation advance was performed in Embrapa Rice and Beans, resulting in the F2 population. F2 plants in the stage V4 received herbicide applications of (Only®) at a dose of 75 and 25 g a.i. L−1, and Dash® in dose of 0.5% v/v. A second application was performed after one week. The degree of phytotoxicity was scored as dead leaves and phytotoxicity intensity, percent survival of leaves and the relative growth. The results showed that the F2 hybrids, resulting of the crosses between the cultivars carriers of the gene of herbicide tolerance of the imidazolinone (Only®) class and conventional irrigated rice cultivars, are viable, thus enabling the establishment of herbicide-tolerant irrigated rice populations, with greater capacity to fight the red rice and with characteristics of agronomic interest. F2 progenies descending from crosses with the cultivar Puita INTA CL presented more tolerance when compared with progenies descending from crosses with the cultivar BRS Sinuelo CL.

Several plant species have development limitations on saline soils; however under water stress situations salicylic acid (SA) may have defense function by acting as antioxidant. Thereby, this study was aimed at analyzing physiological changes occurring in maize seeds treated with SA and germinated on sodium chloride moistened substrate; as well as assessing the effect of antioxidant enzymes produced by the seedlings. To this, seeds were soaked into zero and 1 mM SA solutions, for 60 min., and germinated on different NaCl concentrations (0, 30, 90, and 180 mM), and afterwards evaluated for germination, germination speed index, initial growth, and effect of antioxidant enzymes on seedlings. For electrical conductivity test, the seeds were previously soaked in NaCl solutions with and without SA. The SA application has minimized damages induced by salt stress to maize seed germination by favoring their germination percent; however without SA was not able on maintaining seed viability over 90 mM. Seedling initial growth was impaired by increasing NaCl concentrations in substrate; and electric conductivity has shown low values for release of solutes in the presence of SA. Activity of antioxidant enzymes has increased until 30 mM NaCl concentration; however above such concentration activity of these enzymes was reduced. The SA maintains germination of corn seeds above 90 mM NaCl; however, generally it is not efficient to ensure vigor of seedlings. Diversas espécies apresentam limitações no desenvolvimento em solos salinos, entretanto, em situações de estresse, o ácido salicílico (AS) pode apresentar função antioxidante. Portanto, objetivou-se analisar modificações fisiológicas em sementes e plântulas de milho tratadas com AS e germinadas em cloreto de sódio, além das respostas do sistema antioxidante em plântulas. As sementes foram embebidas durante 60 minutos em soluções de AS (zero e 1 mM) e colocadas para germinar nas concentrações de NaCl (zero; 30; 90 e 180 mM), sendo avaliadas quanto a germinação, índice de velocidade de germinação, crescimento inicial e atividade de enzimas antioxidantes das plântulas. Para a condutividade elétrica, as sementes foram embebidas em mistura das soluções de NaCl e AS. O AS minimizou os danos causados pela salinização na germinação, que foi prejudicada a partir de 90 mM de NaCl, na ausência de AS. O crescimento inicial foi prejudicado com o aumento das concentrações salinas, enquanto a condutividade elétrica apresentou menor lixiviação de solutos na presença do AS. Houve aumento na atividade das enzimas antioxidantes até 30 mM de NaCl com posterior redução. O AS mantêm a germinação das sementes de milho acima de 90 mM de NaCl, porém, de maneira geral, não é eficiente para garantir o vigor das plântulas.

Rooting and acclimatization are limiting steps in plant micropropagation, especially in woody plant species. This study aimed to evaluate the IAA and IBA effect on the in vitro rooting and acclimatization of micropropagated shoots of Japanese plum (Prunus salicina Lindl.) cv. América. Shoots from 3 to 4 cm long were inoculated in MS medium with half salt and vitamin concentrations (MS/2) added with IAA and IBA (0, 0.25, 0.5, 0.75 and 1 mg L-1). After a 20-day period in in vitro cultivation, the shoots were evaluated, and then transferred to a greenhouse, and evaluated after 30 days. At the end of the in vitro cultivation period, no significant interactions were observed for number of roots per shoot and rooting percentage, but a significant effect was recorded for auxin type only, for which shoots grown in media added with IBA showed high values - 0.87 and 41.95%, respectively. A linear increase response from 1.45 to 5.75 cm was verified for root length of shoots cultivated in IBA medium; however, no significant effect was observed, and a 0.86 cm average root length per shoot grown in medium added with IAA was found. After 30 days of acclimatization period, the largest survival percentage was obtained from shoots cultivated in medium with 1 mg L-1 of IBA and IAA (88% and 92%, respectively). Although, IBA provided the highest in vitro rooting, most of the surviving shoots were those originated in IAA-added medium, probably because IBA promoted longer fibrous roots, less appropriate for transplant and soil fixation, as they are easily damaged. It was concluded that in vitro rooting with the addition of the highest IAA concentration (1 mg L-1) provided the greatest plant survival during the acclimatization period of the Japanese plum cv. América.

O gênero Plectranthus é considerado um dos mais ricos em óleos essenciais dentro da família Lamiaceae, compreendendo muitas espécies com propriedades medicinais. Algumas destas são conhecidas popularmente como boldo, as quais possuem semelhanças taxonômicas e diversas sinonímias, possuindo ações anti-dispépticas, analgésicas e digestivas. O objetivo deste estudo foi avaliar quantitativa e qualitativamente os óleos essenciais presentes nas folhas das espécies P. amboinicus, P. barbatus, P. grandis e P. neochilus. A extração do óleo foi realizada por hidrodestilação, utilizando pentano como solvente extrator, repetida por três vezes para cada uma das espécies. A análise dos componentes dos óleos essenciais das quatro espécies de Plectranthus, através da CG/EM, permitiu identificar 14 componentes químicos, sendo a maioria sesquiterpenos. O trans-cariofileno se apresentou em elevada concentração nos óleos estudados. Alguns componentes químicos demonstraram ser específicos para cada espécie e outros apresentaram ocorrência comum a todas as quatro, possibilitando a diferenciação das mesmas em dois grupos, um formado por P. amboinicus e P. neochilus e o outro por P. grandis e P. barbatus. A shoot regeneration system for Plectranthus neochilus was studied from leaf explants. Leaves developed under in vitro conditions were cultured on Wood Plant Medium supplemented with 0.2mg dm-3 α-naphthaleneacetic acid (NAA) and different 6-benzilaminopurine (BAP) or thidiazuron (TDZ) concentrations (0, 1.5, 3.0, 4.5 and 6.0mg dm-3). An increase in percentage of responsive explants (85.3%) and in the number of shoots developed per explant (3.2) was observed when the explants were treated with 5.3 and 4.7mg dm-3 BAP, respectively. The leaf explants cultured on media supplemented with TDZ became vitreous and did not form buds. The regeneration system used is efficient for boldo bud induction and shoot development, showing high potential for advanced cellular and molecular studies. O objetivo do presente trabalho foi adequar um protocolo de regeneração a partir de explantes foliares de Plectranthus neochilus. Folhas desenvolvidas sob condições in vitro foram cultivadas em meio WPM suplementado com 0,2mg L-1 de ácido α-naftaleno acético (ANA) e diferentes concentrações (0; 1,5; 3,0; 4,5 e 6,0mg L-1) de 6-benzilaminopurina (BAP) ou thidiazuron (TDZ). Observou-se o aumento na percentagem de explantes regenerantes (85,3%) e no número de brotações por explante (3,2), quando os explantes foliares foram cultivados em meio contendo 5,3 e 4,7mg L-1 de BAP, respectivamente. Os explantes foliares cultivados em TDZ tornaram-se vítreos e não formaram gemas. O protocolo utilizado é eficiente para a indução de gemas e brotações, demonstrando a potencialidade de seu uso para estudos mais avançados de biologia celular e molecular.

As formigas cortadeiras mostram preferência por algumas espécies vegetais, as quais são constantemente desfolhadas, enquanto outras não são atacadas, embora sejam abundantes e localizadas próximas ao ninho. As formigas dos gêneros Atta e Acromyrmex cortam plantas e transportam pedaços para os formigueiros. O material vegetal é usado para o cultivo do fungo Leucoagaricus gongylophorus, do qual se alimentam. Vários trabalhos utilizando extratos de plantas têm sido realizados para avaliar o efeito sobre o desenvolvimento do fungo simbionte. O objetivo deste trabalho foi avaliar a influência de meios de cultura acrescidos de diferentes extratos sobre o crescimento do fungo L. gongylophorus. Fungos das espécies Acromyrmex ambiguus, Acromyrmex crassispinus, Acromyrmex heyeri e Acromyrmex lundi foram inoculados nos meios Ágar Nutritivo e BDA, ambos acrescidos de melaço e de extratos de azevém (Lolium multiflorum Lam.), tifa (Typha angustifolia L.) e formiga (Atta sexdens piriventris). O meio Pagnocca foi utilizado como padrão. As placas foram mantidas em câmaras climatizadas à temperatura de 25 + 1°C e escotofase de 24 hs, por um período de 49 dias. O crescimento fúngico foi avaliado com base no diâmetro da colônia, sendo este avaliado em intervalos semanais, totalizando sete avaliações. Também foi avaliado o peso seco do fungo aos 53 dias. Os resultados mostraram que o fungo cultivado por diferentes espécies de formigas tem crescimento diferenciado em meios de cultura com diferentes extratos. The cutter ants show preference for some vegetable species, which are constantly defoliated, while others are not attacked, although they are abundant and located close to the nest. The ants of the goods Atta and Acromyrmex cut plants and they transport pieces to the anthills. The vegetable material is used for the cultivation of the mushroom Leucoagaricus gongylophorus, mushroom from of which theyfeed. Several works using extracts of plants have been accomplished to evaluate the effect on the development of the mushroom symbiont. The objective of this work was to evaluate the influence of added culture means of different extracts on the growth of the L. gongylophorus mushroom. Where mushrooms of the species Acromyrmex ambiguus, Acromyrmex crassispinus, Acromyrmex heyeri and Acromyrmex lundi were inoculated in the means Nutritious Ágar and BDA, both added of molasses and extracts of Lolium multiflorum Lam., Typha angustifolia L. and ant (Atta sexdens piriventris). The Pagnocca medium was used as pattern. The plates were maintained in cameras acclimatized to the temperature of 25 + 1°C and 24 hours of darkness, for a period of 49 days. The fungal growth was evaluated with base in the diameter of the colony, which was accomplished with weekly intervals, totalizing seven evaluations. Also the dry weight was evaluated from the mushroom to the 53 days. The results showed that the mushroom cultivated by different species of ants has differentiated growth in culture means with different extracts.

O objetivo deste trabalho foi avaliar o efeito da temperatura sobre a biologia dos biótipos "milho" e "arroz" de Spodoptera frugiperda (J.E. Smith, 1797) (Lepidoptera: Noctuidae) e estimar o número de gerações por ano em laboratório e campo. Foram coletadas lagartas de quatro populações de S. frugiperda no Estado do Rio Grande do Sul, identificadas eletroforeticamente como os biótipos "milho" e "arroz", em áreas isoladas, distanciadas entre si em mais de 300 km, produtoras de milho e arroz irrigado e em áreas adjacentes, que produzem milho e arroz irrigado lado a lado. A temperatura mais adequada para o desenvolvimento dos dois biótipos foi 25ºC. Em laboratório, podem ser obtidas 11,0 e 11,3 gerações ano-1 do biótipo "milho" proveniente de áreas isoladas e adjacentes, respectivamente. Foram estimadas 12,1 gerações ano-1 do biótipo "arroz" quando proveniente de áreas isoladas e 12,2 gerações ano-1 quando proveniente de áreas adjacentes. Em campo, estimou-se a ocorrência de 8,3 e 6,1 gerações ano-1 do biótipo "milho", respectivamente, em áreas isoladas e áreas adjacentes e 8,4 e 7,0 gerações ano-1 do biótipo "arroz", respectivamente, em áreas isoladas e áreas adjacentes. Em áreas adjacentes, o biótipo "arroz" apresenta uma geração a mais ao longo de um ano em relação ao biótipo "milho". The objective of this work was to evaluate the effect of the temperature on the biology of the biotypes "corn" and "rice" of Spodoptera frugiperda (J.E. Smith, 1797) (Lepidoptera: Noctuidae) and to estimate the number of generations per year in laboratory and field. Caterpillars of four populations of S. frugiperda were collected in Rio Grande do Sul State, Brazil, identified by electrophoresis as the biotypes "corn" and "rice" in isolated areas (spaced for more than 300 km), areas of corn and irrigated rice production, as well as in adjacent areas that produces corn and rice irrigated side by side. The most appropriate temperature for the development of the two biotypes was 25ºC. In laboratory, a number between 11.0 and 11.3 generations year-1 for the biotype "corn" can be obtained, originating from isolated and adjacent areas, respectively, and, for the biotype "rice", 12.1 and 12.2 generations year-1 when originating from isolated and adjacent areas, respectively. In the field, the occurrence of 8.3 and 6.1 generations year-1 for the biotype "corn" was estimated, respectively, in isolated and adjacent areas and 8.4 and 7.0 generations year-1 for the "rice" biotype, respectively, in isolated and adjacent areas. In adjacent areas, the "rice" biotype presents an extra generation per year in relation to the "corn" biotype.

O objetivo do trabalho foi analisar a estrutura e a diversidade molecular de quatro populações de Spodoptera frugiperda (J.E. Smith) associadas às culturas de milho e arroz no Rio Grande do Sul. Foram coletadas lagartas de quatro populações em áreas isoladas (distanciadas em mais de 300 km) nos municípios de Santa Rosa (milho) e Uruguaiana (arroz irrigado) e em áreas adjacentes, no município de Pelotas (milho e arroz irrigado). A análise de 40 lagartas (10 de cada população), com cinco combinações de oligonucleotídios iniciadores geraram o total de 241 locos, dentre os quais 229 (95%) foram polimórficos. O dendrograma gerado pelo método de agrupamento UPGMA, baseado na matriz de similaridade de Dice, mostrou sobreposições entre as lagartas das populações estudadas. Os índices de diversidade evidenciaram que 88% da diversidade genética foi observada dentro das populações e 12% entre as populações de S. frugiperda. O dendrograma gerado pelo método de agrupamento UPGMA, baseado na matriz de dissimilaridade das distâncias euclidianas, evidenciou a existência de uma diferenciação entre as populações de acordo com a planta hospedeira, sendo os locos 42 e 47 (M-CTG/E-ACC) capazes de detectar 100% das diferenças genéticas. Esses resultados sugerem que a variação genética das populações de S. frugiperda está associada às plantas hospedeiras, confirmando a presença dos biótipos "milho" e "arroz" de S. frugiperda no Rio Grande do Sul. The goal of this work was to analyze the molecular structure and diversity of four Spodoptera frugiperda (J.E. Smith) populations associated to the corn and rice crops in Rio Grande do Sul State. Four populations of caterpillars were collected from distinct areas (more than 300 km apart), from the counties of Santa Rosa (corn) and Uruguaiana (irrigated rice), and in adjacent areas in Pelotas county (corn and irrigated rice). The analysis of 40 caterpillars (10 from each population), with five combinations of primers generated a total of 241 loci, among then 229 (95%) were polymorphic. The dendrogram generated by the clustering method UPGMA, based on the Dice similarity matrix, showed overlappings among the studied populations. The diversity indexes evidenced that 88% of the genetic diversity was observed within populations and 12% between S. frugiperda populations. The dendrogram generated by clustering UPGMA method, based on the dissimilarity matrix from the euclydean distances, evidenced the existence of a differentiation among populations according to the host plant, being the loci 42 and 47 (M-CTG/E-ACC) capable of detecting 100% of the genetic differences. These results showed that the genetic variation of S. frugiperda populations is associated to host plants, confirming the presence of the "corn" and "rice" biotypes of S. frugiperda in Rio Grande do Sul State.

The germination and development of several plant species can be limited when those plants are grown in soils with high salinity, which reduces seedling viability and vigor, as well as activating the antioxidant defense system. The aim of this study was to evaluate germination, initial growth and activity of antioxidant enzymes (superoxide dismutase, catalase and ascorbate peroxidase) in seedlings of three onion cultivars (Madrugada, Fepagro 27 and Petroline) exposed to different concentrations of NaCl (0, 40, 80, 120 and 160 mM). Seedlings were evaluated for viability, vigor and antioxidant enzyme activity. The experimental procedures were completely randomized in 3 × 5 factorial design, with each treatment performed in triplicate, at a significance level of 5%. For all cultivars, viability and vigor decreased in parallel with increasing NaCl concentrations, whereas antioxidant enzyme activity increased, and one cultivar (Madrugada) showed less salt tolerance than did the others. We conclude that high NaCl concentrations have a negative effect in the physiological quality of onion seeds, resulting in lower seedling growth rates and increased antioxidant enzyme activity, where Fepagro 27 and Petroline cultivars were more tolerant to salt stress than 'Madrugada'.

The aim of this research was to analyze the effect of different temperatures on physiological quality and relate the responses with the antioxidant activity of three lots of corn seeds. The seeds from three lots were exposed for 24h at temperatures of 15, 25, 35 and 40°C and then evaluated for percentage, speed index and first germination count, as well as early growth and activity of antioxidant enzymes in the seedlings. The experimental design was completely randomized with four replications, with subsequent polynomial regression analysis. The results showed that the seeds from the three lots of corn exposed to temperatures of 15 and 40°C, were of better high physiological quality (vigor), which explains the low activity of enzymes of the antioxidant defense system in these same temperatures. The antioxidant defense system is related to the physiological quality of corn seeds exposed to thermal stress. O objetivo neste trabalho foi analisar o efeito de diferentes temperaturas na qualidade fisiológica e relacionar com a atividade antioxidante de três lotes de sementes de milho, as quais foram expostas por 24h às temperaturas de 15, 25, 35 e 40°C e, após, avaliadas quanto à porcentagem, índice de velocidade e primeira contagem de germinação, como também crescimento inicial e atividade de enzimas antioxidantes nas plântulas. O delineamento experimental foi inteiramente casualizado, com quatro repetições, com posterior análise de regressão polinomial. Os resultados evidenciaram que as sementes dos três lotes de milho, expostas às temperaturas de 15 e 40°C, foram de melhor qualidade fisiológica (vigor), o que explica a baixa atividade das enzimas do sistema de defesa antioxidante nessas mesmas temperaturas. O sistema de defesa antioxidante tem relação com a qualidade fisiológica de sementes de milho expostas ao estresse térmico.

The Plectranthus genus comprises several species generally referred to as boldo, which are highly used in popular medicine due to their anti-dyspeptic, analgesic and digestion-stimulating properties. The amount of natural active principles can vary greatly in the related genotypes, which may lead to the inappropriate use of these plants. This work aimed to analyze the interspecific diversity among four species of the Plectranthus genus (P. grandis, P. barbatus, P. neochilus and P. amboinicus) and the intraspecific diversity of P. barbatus collected from different places in southern Brazil, by means of the RAPD technique. A higher genetic similarity was observed between the P. neochilus and P. amboinicus species (80%), followed by P. grandis and P. barbatus (77%). P. barbatus genotypes from Passo Fundo and Porto Alegre showed a genetic similarity which was close to 100%, while the genetic similarity for P. barbatus genotypes from other locations was higher than 96%. Although a low variability among genotypes of this species was found in this study, RAPD markers allowed a clear differentiation among the analyzed genotypes, showing a 53% mean genetic similarity, with a high correlation value (r = 0.99), which proves a high agreement between the genetic similarity and clustering data.

A aveia branca é uma cultura de inverno utilizada em sucessão ao cultivo do arroz irrigado, estando sujeita ao estresse salino, devido ao acúmulo de sais no solo, principalmente na região sul do Rio Grande do Sul. O objetivo deste trabalho foi avaliar a germinação de sementes, o crescimento inicial das plântulas de nove genótipos de aveia branca, bem como identificar os genótipos contrastantes para o caráter tolerância à salinidade. Para isso, os genótipos foram submetidos a diferentes concentrações salinas (0, 25, 50, 75 e 100 mM de NaCl por litro de água), sendo analisadas as seguintes variáveis: porcentagem de germinação, primeira contagem de germinação, índice de velocidade de germinação, comprimento e massa seca da parte aérea e raízes das plântulas. As variáveis analisadas foram afetadas negativamente com o aumento da concentração de NaCl. O comprimento de raiz e o comprimento de parte aérea constitui em variáveis promissoras na separação dos genótipos em relação às diferentes concentrações salinas, permitindo selecionar dois genótipos contrastantes à salinidade, o URS 22 caracterizado como tolerante e o UPF 16 como sensível.

Aiming at verifying effects of priming on germination and emergence of seeds subjected to low temperatures, rice seeds (cv. IRGA 418) were primed into a KNO3 solution during 24, 48, and 72 h, at 20 °C; and into distilled water, for 24 h (control). Germination (G), first count of germination (FCG), and germination speed index (GSI) were the parameters assessed under temperatures of 17 °C, 20 °C and 25 °C. Besides, field emergence (FE), and emergence speed index (ESI) were also assessed under field conditions. A completely randomized design was used with treatments arranged in 4 x 3 factorial (treatments x temperatures) in all experiments. None statistically significant difference was found for seed germination within the 24 h imbibition period, under any of temperatures assessed; although after 48 and 72 h, at 17 °C, this variable has decreased. When seeds were imbibed in KNO3, during 48 and 72 h, the FCG and GSI were decreased, as compared to 24 h imbibition period, and for control treatment, at 17 °C and 25 ºC. After 24 h imbibition, the FE has not differed statistically from control treatment; as occurred for periods of 24 and 48 h imbibition for the ESI parameter. Priming in KNO3 does not cause beneficial effect on seed germination or on seedling emergence of rice. Com o objetivo de verificar o efeito do condicionamento osmótico na germinação e emergência de sementes de arroz submetidas a baixas temperaturas, sementes de arroz cv. IRGA 418 foram condicionadas em solução de nitrato de potássio (KNO3) por 24, 48 e 72 h, a 20 ºC. Como controle as sementes foram embebidas em água destilada por 24 h. Foram realizados os testes: germinação (G), primeira contagem (PCG) e índice de velocidade de germinação (IVG), nas temperaturas de 17 °C, 20 °C e 25 °C, com fatorial 4x3 (tratamentos x temperaturas); além da emergência (EC) e do índice de velocidade de emergência (IVE), com delineamento inteiramente casualizado. Não houve diferença significativa na germinação das sementes em 24 h de embebição em nenhuma temperatura, enquanto que nos períodos de embebição de 48 h e 72 h aos 17 °C houve redução desta variável. A PCG e o IVG diminuíram quando as sementes foram embebidas em KNO3 por 48 h e 72 h comparados às 24 h de embebição e controle a 17 ºC e 25 ºC. Em 24 h de embebição a EC não diferiu do controle, assim como os tratamentos de 24 h e 48 h para o IVE. O condicionamento osmótico em KNO3 não promove efeito benéfico sobre a germinação e a emergência de plântulas de arroz.
