Background Fluoride (F) is one of the main environmental pollutants, and high concentrations are commonly detected in the air and in both surface and groundwater. However, the effects of this pollutant on seed germination and on the initial growth of crop seedlings are still poorly understood. In this context, the aim of this study was to assess morphoanatomical, physiological and biochemical fluoride effect indicators in Phaseolus vulgaris L. seeds and seedlings. Methods P. vulgaris seeds were exposed to a liquid potassium fluoride solution (KF, pH 6.0) at concentrations of 0 (control), 10, 20, 30 mg L−1 for 7 days. A completely randomized experimental design was applied, consisting of four treatments with four replications each. During the experimental period, physiological (7 days) anatomical and histochemical (2 days), biochemical and chemical (4 days) assessments. An analysis of variance was performed followed by Dunnett’s test. to determine significant differences between the KF-exposed groups and control seeds; and a multivariate analysis was performed. Results The germination parameters, and anatomical, morphological, physiological, biochemical and nutritional characteristics of the seedlings did not show negative effects from exposure to KF at the lowest doses evaluated. On the other hand, treatment with the highest dose of KF (30 mg L−1) resulted in a lower germination rate index and increase in abnormal seedlings, and higher electrical conductivity. A lower root length, magnesium content and photochemical efficiency were also observed. The exposure of P. vulgaris to KF, regardless the dose did not affect seeds anatomy and the accumulation of starch and proteins, in relation to the control group. Conclusions Our findings demonstrated that P. vulgaris seedlings were tolerant to KF solutions up to 20 mg L−1, and sensitive when exposed to 30 mg KF L−1.

Fluoride is one of the main phytotoxic environmental pollutants, and high concentrations (10–30 mg L−1) are commonly detected in surface and groundwater. Little, however, is known about the effects of this pollutant on crops that require irrigation during their development, which, in addition to phytotoxicity, may cause negative human health effects. Thus, the aim of this study was to characterize the effects of potassium fluoride (KF) on the germination of lettuce seeds and identify the physiological and anatomical markers of this pollutant’s action on plants exposed to it during growth. Initially, lettuce seeds were sown in gerboxes and soaked in solutions containing 0 mg L−1, 10 mg L−1, 20 mg L−1, and 30 mg L−1 KF. Plants grown in a greenhouse were treated daily with KF irrigation at the same KF concentrations for 40 days. KF exposure reduced the germination rate and germination speed index of lettuce seeds at 20 mg L−1 and 30 mg L−1, resulting in compromised root development at the highest KF concentration. Lettuce plants displayed a slight photosynthesis reduction and a significant photochemical efficiency decrease after exposures to all KF concentrations. Lower chlorophyll contents and nitrogen balance indices were observed in plants exposed to 30 mg L−1 KF. On the other hand, increases in phenolic compounds and malondialdehyde were noted with increasing KF concentrations. Lettuce plants can, therefore, accumulate fluoride in leaves when irrigated with KF-rich water. The investigated physiological and biochemical variables were proven to be adequate fluoride action biomarkers in lettuce plants and may become an important tool in the study of olericulture contaminants.

Abstract The baru (Dipteryx alata Vogel), is fruit tree displaying economic, environmental, industrial and food importance. The objective of this study was to adapt to the tetrazolium test methodology to baru seeds and evaluate the physiological quality of seeds stored for 12 months at 10 and 20 ºC, using X-ray images as a toll for detection damage throughout the storage period while also carrying out germination, emergence and electrical conductivity tests. Baru seeds remained viable for 12 months of storage, albeit with a small decrease in vigor after nine months. The applied tetrazolium test methodology was efficient in determining internal seed damage. The pre-soak time for the tetrazolium test and solution concentration was 24 hours and 0.05%. No changes in seed morphology and radiographic images were observed during the storage periods, regardless of the assessed temperature. The X-ray test enabled a precise visualization of internal baru seed structures.

Pterodon pubescens is a native Brazilian species typical of the cerrado biome, belonging to the Leguminosae-Papilionoideae (Fabaceae) family and popularly known as sucupira branca or faveiro. Its seeds exhibit different integument colors, which may exhibit a direct association to physiological quality related to higher germination percentage rates, vigor, and germination speed index (GSI). Therefore, selection and evaluation methods concerning P. pubescens seed quality are required for the preservation of this species. In this context, the aim of the present study was to determine the relationship between P. pubescens seed integument color and seed quality, through a combination of radiographic imaging and physiological, histochemical and biochemical assays. P. pubescens seeds were obtained from five matrices, and visually classified into four color classes, yellow, light brown, dark brown and black. The coordinates “L”, “a” and “b”, indicated by the colorimeter and the calculations of the chroma and hue angle values, verified that there was a difference in the color of the seeds, eliminating the subjectivity of the visual classification. Thus, the clearer the integuments, the greater their density, filling and α-amylase and β-amylase hydrolytic enzyme activities, the latter comprising important germination power indicators, in addition to high ascorbate peroxidase (APX) enzyme activities, responsible for hydrogen peroxide (H2O2) elimination. Because of this, lighter colored seeds led to higher vigor, germination and GSI rates. The removal of darker seeds from P. pubescens seed lots guarantees higher germination rates and vigor of new plants in nurseries destined to recompose P. pubescens populations.

Abstract The species Campomanesia adamantium and C. pubescens present a great morphological variability, with the existence of morphotypes and the indication of a possible hybridization process occurring between them. Thus, the aim of this study was to characterize the Campomanesia adamantium species and morphotypes of species Campomanesia pubescens through palynological reviews and with the use of molecular markers to assist the taxonomic description of the species. The plant material was collected in the experimental field of Federal University of Jataí, in bloom of 2014, and analyzes were performed in the Morphology Laboratory of Plant Taxonomy and in the Laboratory of Genetics and Plant Breeding belonging to the same institution. Analysis of scanning electron microscopy was performed at the Federal University of Lavras. Important information was found that reinforce the possibility of cross between Campomanesia adamantium and Campomanesia pubescens, generating morphotypes intermediaries, indicating a possible hybridisation process. It can be concluded that Campomanesia adamantium is farthest from the morphotype 2 from Campomanesia pubescens, and morphotypes 1 and 3 are intermediate between them, possibly being hybrids.

Background Fluoride pollution is a global problem because of its high phytotoxicity. Fluoride is released in air, water and soil through industrial processes, where it damages various plant species. Ricinus communis is widely distributed in Brazil, India and China and has been extensively used as a phytoremediation species in heavy metal-contaminated soils. However, few studies regarding the effect of air pollutants on R. communis have been published, and no information about the exposure of this species to fluoride is available. Therefore, the aim of the present study was to investigate the effects of fluoride on R. communis morphoanatomical and physiological responses using simulated rainfall containing potassium fluoride (KF). Methods Young plants at approximately 10 days after emergence were treated daily with KF using simulated rainfall at 0, 1.5, 3.0 and 4.5 mg L−1, for 37 consecutive days. Chlorophyll a fluorescence, gas exchange, anatomical characteristics and fluoride accumulation in the roots and leaves were evaluated after this period. Results No visual or anatomical symptoms were observed for the first three treatments. Necrosis and chlorosis were visually evident after the 37th day of KF application at 4.5 mg L−1, followed by changes in parenchyma tissues, cell collapse and phenolic compound accumulation at the end of the experiment. No damage was observed in terms of photosynthetic photochemical and biochemical stages. Maintenance of physiological characteristics in the presence of fluoride accumulation in roots and leaves were shown to be important fluoride biomarkers. These characteristics suggest that R. communis is tolerant to 1.5 and 3.0 mg L−1 KF, and is anatomically sensitive at 4.5 mg L−1 KF.

Efficient methodologies for automated seed quality evaluations are important for the seed industry. Advanced seed technology research requires the use of adequate methods to ensure good seed performance under adverse environmental conditions; thus, providing producers with detailed, quick, and accurate information on structural seed integrity and ensuring vigorous production. To address this problem, this study aimed to determine Brachiaria brizantha (Marandu cv., Piatã cv. and Xaraés cv.) seed quality through radiographic imaging analyses associated with vigor tests and anatomical characterizations. Brachiaria seed cultivars displaying different physical and physiological attributes were selected and subjected to the 1000-seed weight test, water content determinations, X-ray analyses, germination tests, and anatomical characterizations. The X-ray analyses made it possible to establish a relationship between the X-ray images and other determined variables. Furthermore, the X-ray images can indicate evidence of internal and external damage that could later compromise germination. The Marandu and Piatã cultivars presented the highest germination percentages, germination speed indices, normal seedling development, and cellular structure preservation compared to the Xaraés cultivar. To summarize, X-ray analyses are efficient methods used for the selection of higher physical quality cultivars and can aid in the decision-making processes of companies and seed producers worldwide.

The cajuzinho-do-cerrado is a native species of the cerrado of great potential of use and is threatened of extinction. The aim of this study was to determine and evaluate the thermodynamic properties of Anacardium humile St. Hil. achenes from the different hygroscopic equilibrium points according to the moisture content. To obtain the isotherms, the indirect static method was used at temperatures of 10, 20, 30 and 40 °C and for moisture contents of 13.4, 11.1, 8.7 and 5.3% dry basis (db). The Copace model was used to describe the hygroscopic equilibrium of the achenes. The thermodynamic properties are influenced by the moisture content because the latent heat of vaporization of the water of the achenes increases with the decrease in the equilibrium moisture content, varying between 3,035.63 and 2,510.80 kJ kg-1 for moisture contents of 4.51 to 13.4% db, respectively. The differential enthalpy and entropy are closely related to the moisture content of the achenes. The isokinetic temperature of 304.67 K denotes the linear chemical compensation between the differential enthalpy and entropy of the Anacardium humile St. Hil. achenes, and the desorption process is controlled by enthalpy.

Sementes de maior tamanho apresentam embrião melhor formado e com maior quantidade de reserva, o que pode conferir maior vigor. O objetivo deste trabalho foi avaliar a emergência e o crescimento de plantas de baru, oriundas de sementes com diferentes espessuras e comprimentos. Sementes de baru foram coletadas em Santa Helena de Goiás, Goiás, e levadas ao Laboratório de Sementes do Instituto Federal Goiano, Campus Rio Verde, Rio Verde, para extração, beneficiamento, separação manual, em três classes de comprimento, curta, intermediária e longa, e separação mecânica, em um conjunto de três peneiras com orifício circular (13 mm) e orifícios oblongos (9 mm x 15 mm e 8 mm x 15 mm). O experimento foi montado em delineamento inteiramente casualizado, em esquema fatorial 3 x 2 (comprimento x espessura), em quatro repetições de 20 sementes, que foram semeadas em tubetes plásticos, totalizando 24 unidades experimentais (parcelas). As mudas permaneceram em casa de vegetação, dotada de sistema de irrigação automatizado, por aspersão. Realizaram-se a determinação do percentual de emergência, índice de velocidade de emergência e de parâmetros morfológicos e de crescimento das mudas. O tamanho da semente influenciou a maior parte das variáveis avaliadas e as sementes de baru com maior comprimento e espessura proporcionam maior velocidade de emergência e crescimento de plantas.

The present study aimed to determine the imbibition curve and evaluate the effects of ‘reducing the water content of cagaita seeds (Eugenia dysenterica DC.), using the radiographic image analysis technique. To this end, three assays were carried out: seed imbibition curve and root protrusion were determined in assay 1. Seeds in silica gel were oven dried for to 72 h and were then analyzed for water content and vigor in assay 2. Seeds were dried up to certain levels of the water content and were evaluated by radiographic imaging analysis in assay 3. A rapid mass gain was observed in the first 60 imbibition hours and the root protrusion reached 94.4% after 192 imbibition hours. Cagaita seeds did not tolerate drying periods longer than 24 h, which corroborates the recalcitrant behavior of the seeds of these species. The analysis of the radiographic image obtained by an X-ray test allowed the establishment of a direct relation between the increased free space area within the seed and the decrease in the seedling emergence. The drying process altered the internal morphology of the seed and such changes directly influenced the development of the seedlings.

The objective of this study was to evaluate different pre-germination treatments, histochemical characteristics and physiological performance of Sapindus saponaria L. seeds as a function of temperature and the germination substrate. The S. saponaria seeds were collected in the municipality of Rio Verde, GO, and analysed through the execution of 2 tests (dormancy breakage and germination tests). Test 1 consisted of 10 treatments for overcoming dormancy, whereas test 2 consisted of a 35-day evaluation of germination on several substrates (paper and sand) under the following 4 temperature conditions: 20, 25, 30°C and alternating 20-30°C (± 0.5°C). The results of the dormancy breakage and germination tests were submitted to analysis of variance (ANOVA), and the means were compared by the Tukey test. Scarifying S. saponaria seeds with concentrated sulfuric acid for 90 min provided a relatively high speed and percentage of emergence, without causing anatomical damage. Pre-germination treatments using high- (42 °C) or low-(10 °C) temperature stress caused cellular damage in the endosperm region of S. saponaria seeds. The greatest percentage of S. saponaria seeds germinated on a paper substrate at 30°C.

ABSTRACT Sorghum arundinaceum (Desv.) Stapf is a weed that belongs to the Poaceae family and is widespread throughout Brazil. Despite the frequent occurrence, infesting cultivated areas, there is little research concerning the biology and physiology of this species. The objective of this research was to evaluate the growth, carbon partitioning and physiological characteristics of the weed Sorghum arundinaceum in greenhouse. Plants were collected at regular intervals of seven days, from 22 to 113 days after transplanting (DAT). In each sample, we determined plant height, root volume, leaf area and dry matter, and subsequently we perfomed the growth analysis, we have determined the dry matter partitioning among organs, the accumulation of dry matter, the specific leaf area, the relative growth rate and leaf weight ratio. At 36, 78 and 113 DAT, the photosynthetic and transpiration rates, stomatal conductance, CO2 concentration and chlorophyll fluorescence were evaluated. The Sorghum arundinaceum reached 1.91 in height, with slow initial growth and allocated much of the biomass in the roots. The photosynthetic rate and the maximum quantum yield of FSII are similar throughout the growth cycle. At maturity the Sorghum arundinaceum presents higher values of transpiration rate, stomatal conductance and non-photochemical quenching coefficient (NPQ).

The objective of this research was to evaluate the effects of using different herbicides as desiccants in pre - harvest and the effects of storage on the physiological and sanitary quality of azuki bean seeds ( Vigna angularis Willd). The experiment was arranged in a randomized complete block design in a split plot scheme, with four replications. Four herbicides were tested: paraquat (400 g a.i. ha - 1 ), glufosinate ammonium (400 g a.i. ha - 1 ), glyphosate (720 g a.i. ha - 1 ), flumioxazin (30 g a.i. ha - 1 ) and a control without herbicide application. In the subplots seed quality was tested in two evaluation periods: at harvest and six months after harvest. Desiccant was applied when the azuki beans were physiologically mature. We assessed the physiological and sanitary quality of the seeds using a vigour and seed health test. The use of glyphosate resulted in a higher incidence of abnormal seedlings and reduced size and weight of the seedlings. With paraquat and flumioxazin the physiological quality was maintained and there was reduced pathogen infestation in the seeds six months after harvest. Storage affected the physiological quality of the azuki bean seeds.

Rhamnidium elaeocarpum Reissek is a species found in forests, especially in riparian forests. This medicinal species is commonly used to reduce itching of the gums in children at the onset of teething and contains several substances responsible for antiulcer activity. The aim of the present study was to describe the external and anatomical features of the seeds, fruits, and developmental stages of seedlings of R. elaeocarpum Reissek. The processing consisted of maceration in sieves to separate the seeds from the waste. To describe the fruit, external details of the pericarp were observed and for the seeds, the external structures of the integument and internal structures of the endosperm and embryonic axis were analyzed. The morphological aspects of the seeds, germination, and seedlings of Rhamnidium elaeocarpum Reissek are homogenous in all phases. The fruit is simple, fleshy, indehiscent, and berry monospermic. The seed is exalbuminous and cream colored, with a thin and rough integument, the presence of tannin content, and an endosperm rich in starch, calcium oxalate crystals, and lipid content. Germination is hypogeal, and the seedling is cryptocotyledonary.

This study aimed to investigate the desiccation tolerance of Rhamnidium elaeocarpum Reissek seeds through physiological and biochemical alterations. Fruit were collected from the municipality of Jaupaci, Goias State, Brazil, when the water content of their seeds had a 37% wet basis (w.b.) and the seeds were subsequently kept in a drying oven with air circulation at a temperature of 35ºC until a water content was reached of 20 to 12% (w.b.). The following parameters were evaluated: germination, germination speed index, emergence, emergence speed index, electrical conductivity and respiration rate. Furthermore, the electrophoretic profile of the isoenzymes: catalase, peroxidase and esterase, the enzymatic activities of endo-β-mannanase and α-amylase and the electrophoretic analysis of heat-resistant proteins were performed. A higher germination speed and respiratory rate was obtained for seeds with a water content of 12% w.b., and the activity of catalase isoforms was more pronounced in seeds with a higher degree of dehydration, which was in contrast to peroxidase, showed decreased activity. The seeds contained heat-resistant proteins of low molecular weight that ranged from 48.7 to 13.2 kDa. We found that the acquisition of drying tolerance in R. Elaeocarpum Reissek seeds is associated more with catalase than with esterase and peroxidase.

Studies evaluating the internal morphology and seed quality of native species are essential for successful conservation programs. Our aim was to verify the efficiency of X-ray imagery in evaluating cashew-tree-of-the-cerrado (Anacardium othonianum Rizz.) achene viability after desiccation. The achenes were collected at 12% water content (w.b.) and dried in silica gel until they reached 10, 8, 6, and 4% (w.b.). The fruit morphology and the quality of the seeds were evaluated by X-ray test together with vigor, electrical conductivity and emergence tests. Achenes with different water contents were exposed to an X-ray machine at 18 kV for 11 s and were thereafter submitted to emergence tests. The images were analyzed, and the achenes were classified based on internal morphology as completely full, malformed, or empty. These results were compared to those from the emergence tests. The statistical design was a complete randomized factorial (5 x 3). Desiccation to 4% (w.b.) did not damage or modify the internal structures. X-ray was efficient in evaluating the internal morphology and detecting achene quality, making it possible to remove empty and abnormal fruit and form vigorous seed lots, reducing the cost of storage and bedding plant production for this native species.

The use of low quality seeds with a lower physiological reflects one of the major causes of low productivity. Thus the storage conditions of seed must be taken into consideration. This research aimed to evaluate the influence of natural and synthetic insecticides on emergence and seed storage of crambe, as these substances are essential to prevent infestation of seeds of other species by harmful organisms. The experimental design was a randomized block in factorial 3 x 8 ( 8 substances and 3 storage times ) with 4 replications. We assessed the following natural insecticides: saffron, lime, ash, neem, diatomaceous earth, and synthetic: chlorpyrifos and deltamethrin; besides the control consisting of seeds without any treatment. We evaluated the percentage of emergence, speed of emergence index and time to reach 50 % of emergency. In all characteristics, it was found that no influence of neem on seed vigor. There are disadvantages in the application of the insecticides chlorpyrifos and diatomaceous earth, which interfered with the emergence rate of seeds of crambe. The seeds treated with other insecticides had different behavior of untreated seeds after 120 days of storage to assess the time that they take to reach 50 % germination.

The objective of this study was to evaluate the effect of immersion for varying periods in different concentrations of gibberellic acid and separate methods of scarification on the germination of seeds tucum. In the first trial, testing different soaking periods (24 and 48 hours), different forms of soaking (fast and slow) and different concentrations of gibberellic acid (0 , 100 and 200 mg L-¹ ) compared to the control (water) under completely randomized design in a factorial 2x2x3. The second experiment evaluated the different scarification treatments being: physical (seed coat removal in the hilar region with the aid of a scalpel), chemical (sulfuric acid 98 PA for 2:04 minutes) and thermal (hot water at approximately 98 ° C and cold water at about 2°C for 4 minutes). In the first trial were evaluated % of contaminated seeds (seeds infected by microorganisms) and hard seeds ( who did not start the germination process , but not soiled), and in the second we assessed germination percentage (%) every two days for three months; germination Speed Index (GSI), time to occurrence of 50 % germination (T50) and Emergency Speed Index (ESI). The use of gibberellic acid was ineffective in promoting germination of Tucum (Astrocaryum Huaimi Mart.). Scarification treatments were effective in promoting germination and emergence of seedlings in the nursery and the most efficient physical removal scarification of the seed coat in the hilar region with the highest percentage of germination.

Native Cerrado plants are exposed to soils with low pH and high availability of Al. In this study, we measured the Al content in adult plants, and investigated the effects of various Al doses on germination and early development of Eugenia dysenterica plants. For germination tests, the seeds were soaked in Al solution and evaluated for twenty days in growth chambers. In a second experiment, young plants were cultivated in hydroponic systems with various Al concentrations to evaluate the morphological, anatomical and physiological characteristics of E. dysenterica. Anatomical changes and low germinative vigor were observed in seeds germinated in 600 and 800 μmol Al3+ L−1. In the hydroponic system, 200 μmol Al3+ L−1 stimulated root growth in young plants. The activity of antioxidant enzymes and the accumulation of phenolic compounds were greatest at the highest Al doses, preventing changes in gas exchange and chlorophyll a fluorescence. Starch grain accumulation was noted in plant cells exposed to 200 and 400 μmol Al3+ L−1. Adult E. dysenterica trees also accumulated Al in leaves, bark and seeds. These data suggest that E. dysenterica is tolerant to Al.

This study was performed to improve the speed of and standardize the process for the germination of macaw palm seeds. The effect of mechanical scarification (removal of the tegument in the hilum region), thermal scarification (imbibition of seeds in water at approximately 100°C for two or four minutes) and chemical scarification (98% sulfuric acid treatment for two or four minutes) were compared to a control (no scarification) in both a germination chamber and a greenhouse, representing trials one and two, respectively. The effect of imbibition speed (fast or slow) and different concentrations of gibberellic acid (0, 100 or 200 mg L-1) on the germination percentage and index of germination velocity were evaluated during a third trial. Mechanical scarification to remove the seed tegument in the hilum region enhanced the germination of the seeds in the germination chamber and seedling emergence in the greenhouse, and this type of scarification should be recommended for breaking dormancy in this species. Not obtained increase in seed germination with gibberellic acid, regardless of the application form in the seeds.

Studies on conservation and exploitation of the Brazilian Savannah native species are essential to technologies that lead to the rational exploitation of these products. This study aimed at determining the desorption isotherms of Anacardium othonianum seeds for different temperature and water activity conditions, as well as adjusting different mathematical models to the experimental data, by selecting the one that best represents the phenomenon. The hygroscopicity was determined with the aid of the gravity static method, for temperatures of 25ºC, 30ºC, 35ºC and 40ºC and water activity between 0.12 and 0.89 (decimal). It was observed that the equilibrium moisture content decreased while the temperature increased, for the same water activity, similarly to what happens with hygroscopic products. The Chung-Pfost model reached the highest determination coefficient and the lowest values for relative average error, estimated medium error and chi-square test, being the one selected to predict the hygroscopic equilibrium of Anacardium othonianum seeds. The isosteric heat increased, while the moisture content decreased, i.e., it increased the energy required to remove water, varying from 4,586.35 kJ kg-1 to 2,572.7 kJ kg-1, for the moisture content of 1.76-6.56 (d.b. %).

The objective of this study was to evaluate the physiological quality of babassu (Orbignya phalerata Mart.) embryos as a function of desiccation. The fruits were placed in a convection oven at 37±2°C for 0, 3, 6, 9, or 12 days. After these periods, the moisture contents of the fruits and seeds were determined. A reduction in the water content occurred more rapidly in the fruits than in the seeds. The decrease in seed moisture content from 12.59% (on the basis of fresh weight) to 4.56% did not affect the percentage of germinated zygotic embryos; however, we did observe a decline in the vigor as the seed moisture content decreased. Embryos with a 4.56% moisture content developed into seedlings of shorter lengths after 30, 60, and 90 days of in vitro growth. The longest seedlings originated from embryos with a 10.42% moisture content after 90 days. Thus, it was evident that the babassu embryos were tolerant to desiccation, whereas the seedling length was negatively affected by a reduction in the seed moisture content. Therefore, babassu fruits may be dehydrated and stored without losing their viability, which thereby guarantees the conservation of their germplasm.O objetivo deste estudo foi avaliar a qualidade fisiológica de embriões de babaçu (Orbignya phalerata Mart.) em função da dessecação. Os frutos foram mantidos em estufa de circulação forçada a 37±2°C por 0, 3, 6, 9 e 12 dias. Após esses períodos, foi determinado o teor de água dos frutos e sementes, sendo também avaliado a percentagem de embriões zigóticos germinados in vitro, GRI (definir) e desenvolvimento das plântulas. A redução no teor de água ocorre mais rapidamente em frutos que em sementes. A diminuição no conteúdo de água da semente a partir de 12,59% (umidade inicial) para 4,56% não afeta a porcentagem de germinação in vitro dos embriões zigóticos, no entanto foi observado diminuição no vigor dos embriões à medida que decresce o teor de água da semente. Embriões com 4,56% de umidade desenvolveram as menores plântulas após 30, 60 e 90 dias de cultivo in vitro. As maiores plântulas foram obtidas de embriões com 10,42% de umidade, após 90 dias. Diante disso, fica evidente que os embriões de babaçu toleram a dessecação, mas que a altura das plântulas foi afetada negativamente pela redução do teor de água das sementes.

Knowing the importance of preventive use of insecticides for seed treatment, this study aimed to identify potential insect pests of storage for seeds of radish by means of traps set on the field for six months and to evaluate the effect of natural and synthetic substances insecticides on the emergence of radish seedlings stored for 120 days. The bioassay, field-level, was conducted over a period of six months in the following environments: savannah, area of cultivation of crops, pasture and forest at the Federal Institute Goiano. For each of these environments were placed five traps made of PET bottles, containing respectively in maize, millet, soybean, crambe and wild radish. The traps were inspected every seven days to verify the the occurrence of insects plague. The substances were: 1) saffron, 2) limestone, 3) Gray, 4) neem, 5) diatomaceous earth, 6) chlorpyrifos, 7) deltamethrin and 8) control treatment. The experimental design was randomized blocks in factorial scheme 8 x 2 (8 substances and two storage periods, is not stored seeds and seeds stored for 120 days) with four replications. Were evaluated the percentage of emergence, hold rate of emergence, time to reach 50% emergence and the record of the dry mass of seedlings. The natural and synthetic substances do not interfere with seedling emergence in seeds of wild radish, even after storage for 120 days in a room. In the traps in the field, containing seeds of wild radish noted the presence of the insect Carpophilus sp. Sabendo da importância do uso preventivo de inseticidas no tratamento de sementes, esta pesquisa teve por objetivos identificar possíveis insetos praga de armazenamento para sementes de nabo forrageiro por meio de armadilhas montadas em campo durante seis meses e avaliar o efeito de substâncias inseticidas, naturais e sintéticas na emergência de plântulas em sementes de nabo forrageiro armazenadas por 120 dias. O bioensaio, em nível de campo, foi conduzido por um período de seis meses nos seguintes ambientes: cerrado, área de cultivo de grandes culturas, pastagem e mata do Instituto Federal Goiano. Para cada um desses ambientes foram colocadas cinco armadilhas confeccionadas em garrafas PET, contendo respectivamente sementes de milho, milheto, soja, crambe e nabo forrageiro. As armadilhas foram inspecionadas a cada sete dias para a verificação da ocorrência de insetos praga. As substâncias consistiram de: 1) açafrão, 2) calcário, 3) cinza, 4) nim, 5) terra de diatomáceas, 6) clorpirifós, 7) deltametrina e 8) tratamento controle. O delineamento experimental foi em blocos ao acaso, no esquema fatorial 8 x 2, sendo oito substâncias e armazenamento por zero e 120 dias, com 4 repetições. Avaliaram-se porcentagem de emergência, índice de velocidade de emergência, tempo para atingir 50% da emergência e massa seca das plântulas emergidas. As substâncias naturais e sintéticas não interferem na emergência de plântulas em sementes de nabo forrageiro, mesmo após o armazenamento de 120 dias em sala climatizada. Nas armadilhas em campo contendo sementes de nabo forrageiro, foi observado a presença do inseto Carpophilus sp.

This study was performed to evaluate the physiological quality of Crambe abyssinica Hochst fruits subjected to drying under different treatments with air. The crambe fruits, with moisture content of 21.0 ± 1.3 (% w.b.), were manually collected and dried until their moisture content reached 7.0 ± 1.4 (% w.b.). The crambe fruits were dried with natural air (23°C mean temperature and 60.6% average relative humidity) and seven days after harvest in an experimental dryer with forced ventilation at temperatures of 30, 40, 50, 60 and 70°C and relative humidities of 37.4, 22.3, 14.0, 8.3 and 5.1%, respectively. The evaluation was the percent germination, the germination speed index (GSI), the amount of water absorption and the electrical conductivity. The results of the study showed that increases in the drying temperature decreased the drying time and negatively affected the quality of the fruits. The study’s findings for the GSI and for the percent germination showed that the drying temperature of 30°C yielded superior results. The moisture content of the crambe fruits after absorption was higher at the drying temperatures of 23, 40 and 70ºC. In contrast, the electrical conductivity measurement was unaffected by drying at high temperatures.Objetivou-se avaliar a qualidade fisiológica dos frutos de Crambe abissynica, submetidas à secagem em diversas condições de ar. Frutos de crambe foram colhidos manualmente com teor de água de 21,0 ± 1,3 (% b.u) e secas até o teor de 7,0 ± 1,4 (% b.u.). A secagem foi realizada com ar natural (temperatura média de 23ºC e umidade relativa média de 60,6%) e sete dias após a colheita em secador experimental nas temperaturas de 30, 40, 50, 60 e 70°C e umidades relativas de 37,4; 22,3; 14,0; 8,3 e 5,1% respectivamente. A qualidade dos frutos foi avaliada pela porcentagem de germinação, índice de velocidade de germinação (IVG), absorção de água e condutividade elétrica. Com o aumento da temperatura o tempo de secagem reduziu e a qualidade dos frutos foi influenciada negativamente. Para a temperatura de 30ºC o IVG e a porcentagem de germinação foram superiores às demais temperaturas avaliadas. O teor de água dos frutos de crambe depois da absorção de água foi mais elevado para as temperaturas de secagem de 23, 40 e 70ºC. A secagem em temperaturas elevadas não influenciou a condutividade elétrica.

This study evaluated mechanisms of the dehydration and rehydration of fruits and seeds on the resulting embryo viability of the macaw palm, Acrocomia aculeata (Jacq.) Loddiges ex Mart. Dehydration was performed in a forced-air oven at 37 ± 2°C for 0, 3, 6, 9, 12 or 15 days, and after these periods, the embryo viability was evaluated by the tetrazolium test. The effect of hydration on the germination percentage and velocity index was evaluated by the imbibition of seeds with distilled water in a germinator at 30 ± 2°C for 0, 2, 4, 6, 8 or 10 days. The seeds were then treated with fungicide and maintained in a Mangelsdorf germinator at 30 ± 2°C. Drying caused a progressive water loss in the fruits and facilitated the extraction of the seeds. Fruits subjected to drying for up to nine days had no loss of vigor; however, after dehydration for 15 days, a loss in seed viability was observed. Simple linear regression estimated the seed water content based on the fruit water content. Seed imbibition in distilled water favored an increase in dead by fungally contaminated seeds.Objetivou-se com esse estudo elucidar os mecanismos de desidratação e hidratação de frutos e sementes na viabilidade de embriões de macaúba Acrocomia aculeata (Jacq.) Loddiges ex Mart. A desidratação foi realizada em estufa de circulação de ar, ajustada a 37 ± 2°C por 0, 3, 6, 9, 12 e 15 dias, após esses períodos, foi avaliada a viabilidade dos embriões pelo teste de tetrazólio. O efeito da hidratação na porcentagem e velocidade de germinação foi avaliado pela embebição das sementes em água destilada em germinador com temperatura a 30 ± 2°C por 0, 2, 4, 6, 8 e 10 dias. As sementes foram tratadas com fungicida e mantidas em germinador do tipo Mangelsdorf. A secagem promoveu perda pregressiva no conteúdo de água dos frutos e facilitou a extração das sementes. Quando os frutos foram secos por nove dias não há perda de vigor, no entanto, depois de 15 dias de desidratação, foi observado perda na viabilidade das sementes. Com a regressão linear simples é possível estimar o conteúdo de água nas sementes baseando-se no teor de água dos frutos. A embebição das sementes em água destilada favoreceu e aumentou o número de sementes mortas pela contaminação fúngica.

This study evaluated the effect of light, temperature, physiologic stage at harvest and length of time on the germination of Hyptis marrubioides seeds. Two trials were conducted. The first experiment was performed immediately after the seed harvest and consisted of a 2 x 3 x 3 factorial design with 2 environmental conditions (light and dark), 3 temperatures (20, 30 and 20/30ºC) and 3 seed harvest times (green colored seeds, light brown seeds and dark brown seeds). The second experiment was conducted in the presence of light at a temperature of 30ºC and consisted of a 4 x 3 factorial design, with 4 storage times (0, 6, 12 and 18 months) and the 3 three harvest physiologic stages used in the previous experiment. Both of the experiments were conducted in randomized blocks, with 4 replications of 100 seeds. Light did not affect germination. By contrast, a temperature of 20ºC retarded the germination process, although the percentage of germinating seeds was not affected. Seed storage and the different harvest physiologic stages affected the Speed of germination index (SGI) and the germination percentage. Seeds that were harvested at the more mature stage (dark brown color) could be stored for up to 18 months.O presente trabalho teve como objetivos avaliar a influência da luz, da temperatura, do estádio fisiológico na colheita e do armazenamento na germinação das sementes de Hyptis marrubioides. Foram realizados dois experimentos, sendo o primeiro implantado logo após a colheita das sementes, constituído por um fatorial 2 x 3 x 3, com 2 ambientes (luz e escuro) x 3 temperaturas (20, 30 e 20/30ºC) x 3 épocas de colheita das sementes (sementes com coloração verde, coloração marrom claro e coloração marrom escuro). O segundo experimento, em presença de luz e à temperatura de 30ºC, foi constituído por um esquema fatorial 4 x 3, sendo 4 tempos de armazenamento (0, 6, 12 e 18 meses) e 3 três estádios fisiológicos de coleta mencionados no experimento anterior. Ambos os experimentos foram implantados no delineamento em blocos ao acaso, com quatro repetições de 100 sementes. Foi verificado que o fator luz não influenciou a germinação. Por outro lado, a temperatura de 20ºC retardou a germinação, apesar de não influenciar a sua porcentagem. O armazenamento e os diferentes estádios fisiológicos de colheita das sementes influenciaram o IVG e a porcentagem de germinação. Sementes coletadas no estádio mais maduro (coloração marron escuro) demonstraram ser apropriadas para o armazenamento por até 18 meses.

The great agricultural development in the Brazilian savannah brought with it the reduction of its natural vegetation, which has been quickly lost, especially due to extractive exploitation by man. Murici (Byrsonima verbascifolia Rich) is a fruit bearing plant of great medicinal importance in the savannah; however, its propagation is hindered by the fact that the seeds have low germination and seedling emergence in the field is slow. This study evaluated the effect of different dormancy breakage methods in murici seeds. Therefore, were evaluated effect of different temperatures; soaking in gibberellic acid and water; chemical and mechanical scarification and two types of substrate (distilled water and KNO3). Treatments were arranged in a randomized block design. Records made daily on the number of germinated seeds were used to determine the germination percentage and germination speed index. Among the methods evaluated, giberellic acid at 2,309.46 ?M gave the greatest seed germination percentage, especially when the germination paper was moistened with KNO3. O grande desenvolvimento agrícola na região do Cerrado foi acompanhado pela redução da vegetação atual e essa rica formação vegetal vem sofrendo uma rápida depredação, principalmente devido à exploração extrativista por parte do homem. O murici (Byrsonima verbascifolia Rich) destaca-se por ser uma planta de grande importância medicinal e frutífera do cerrado, entretanto, sua propagação é dificultada pelo fato de suas sementes terem baixa taxa de germinação e a emergência das plântulas em campo serem lentas. Objetivou-se com este trabalho avaliar o efeito de diferentes métodos de superação de dormência em sementes de murici. Para tanto, foram avaliados o efeito de diferentes temperaturas; imersão em ácido giberélico; imersão em água e escarificação química e mecânica do endocarpo e dois tipos de substrato umedecidos (água destilada e KNO3), dispostos em delineamento de blocos ao acaso. Foram realizadas contagens diárias para determinar a porcentagem de germinação e índice de velocidade de germinação. Entre os diversos métodos avaliados, o ácido giberélico a 2.309,46?M proporcionou maior porcentagem de germinação das sementes, sobretudo quando o papel de germinação foi umedecido com KNO3.

This work aimed to evaluate methods to breaking seed dormancy of Brachiaria brizantha cv. Marandu. An experiment was carried out at the Laboratory of Seeds from Instituto Federal Goiano – Campus Rio Verde. A completely randomized design with eight treatments to breaking seed dormancy was used. The treatments were: 1) control; 2) immersion in gibberellic acid (100 mg L-1) for 48 h; 3) immersion in gibberellic acid (200 mg L-1) for 48 h; 4) immersion in concentrated sulphuric acid for 5 min; 5) immersion in concentrated sulphuric acid for 10 min; 6) immersion in hot water (100 ºC) for 1 min; 7) immersion in hot water (100 ºC) for 2 min and 8) immersion in hot water (100 ºC) for 3 min with four replicates. Daily, germination was evaluated to determine the germination speed index. After the complete germination (25 days), the seed germination percentage was quantified. Hot water treatments, independently of the seed immersion time, were the most effective to breaking the seed dormancy of B. brizantha cv. Marandu.O objetivo deste trabalho foi avaliar métodos de superação da dormência de sementes de Brachiaria brizantha cv. Marandu. O experimento foi conduzido no Laboratório de Sementes do IFGoiano – Campus Rio Verde. Foi utilizado o delineamento inteiramente casualizado, com oito tratamentos: 1) testemunha; 2) imersão em ácido giberélico (100mgL-1) por 48h; 3) imersão em ácido giberélico (200mgL-1) por 48h; 4) imersão em ácido sulfúrico concentrado por 5 min.; 5) imersão em ácido sulfúrico concentrado por 10 min.; 6) imersão em água fervente por 1 min.; 7) imersão em água fervente por 2 min.; 8) imersão em água fervente por 3 min, com 4 repetições de 50 sementes. Diariamente, foram realizadas as avaliações de germinação, para determinar o Índice de Velocidade da Germinação e, após a completa estabilização da germinação (25 dias), foi quantificada a porcentagem de germinação. Os tratamentos com água fervente, independentemente do tempo de imersão das sementes, foram os mais efetivos para superar a a dormência de sementes de Brachiaria brizantha cv. Marandu.

Sementes de croada (Mouriri elliptica Mart.) Melastomataceae, espécie frutífera nativa do cerrado, foram submetidas aos seguintes tratamentos, visando a superação de dormência: pré-resfriamento a 5° C por 7 dias; pré-aquecimento em estufa com circulação de ar a 40º C por 7 dias; escarificação química em ácido sulfúrico concentrado por 5 e 15 minutos; imersão em água fervente por 5 e 15 minutos; imersão em ácido giberélico a 100 e 200 mg L-1 por 48 horas; imersão em água destilada por 24 e 48 horas; escarificação mecânica com lixa nº 80 na parte superior e oposta ao eixo embrionário e testemunha (sementes sem tratamento prévio). Quatro repetições de quinze sementes foram colocadas para germinar sobre papel umedecido com água destilada ou nitrato de potássio a 0,2%, em temperatura de 30º C em presença de luz. Foram realizadas contagens diárias do 2º dia após a implantação do experimento até a estabilização da porcentagem de germinação, que acorreu no 60º dia. A germinação foi mais rápida em sementes pré-embebidas em ácido giberélico (em substrato umedecido com água ou com solução de nitrato de potássio 0,2%), pré-embebidas em água por 24 horas, em substrato umedecido com água, e pré-embebidas em água por 48 horas ou escarificadas mecanicamente e colocadas para germinar em substrato umedecido com água. Taxas de germinação mais elevadas ocorreram em sementes pré-embebidas em ácido giberélico a 100 ou 200 mg L-1, em substrato umedecido com solução de nitrato de potássio 0,2%.Croada seeds (Mouriri elliptica Mart.) Melastomataceae, a fruit-bearing species native of the cerrado (savannah-like vegetation), were submitted to the following dormancy breaking methods: pre-cooling at 5° C for 7 days; pre-heating in a oven air circualtion at 40º C for 7 days; chemical escarification in concentrated sulfuric acid for 5 and 15 min; soaking in boiling water for 5 and 15 min; soaking in gibbereli acid at 100 and 200 mg L-1 for 48 hours; soaking into distilled water for 24 and 48 hours; mechanical escarification with a sandpaper number 80 at teh upper aprt and opposed to the embryonary axis and control (seeds without any previous treatment). The seeds were put to germinate on a "germitest" type paper moistened with destilled water or 0.2% potassiun nitrate. Daily counts between the 2nd day after the establishment of the experiment till the stabilization, which took place on the 60th day, were performed. Seeds germinated faster when they were pre-soaked in gibberelic acid, independent of if placed to germinate on a water-moistened substrate or with a soultion of 0.2% potassiun nitrate, pre-soaked into water for 24 hours, on a water-moistened substrate, or independent of the substrate, and, pre-soaked into water for 48 hours or mechanical escarification and put to germinate on a water-containing substrate. Higher germination rates occurred in seeds pre-soaked into 100 or 200 mg L-1 gibberelic acid, on a substrate moistend with a 0.2% potassiun nitrate containing solution.

Na fase de pós-colheita do pinhão manso a secagem é o processo mais utilizado para assegurar sua qualidade e estabilidade. Assim, objetivou-se no presente trabalho analisar a secagem das sementes de pinhão manso, bem como verificar o efeito das diversas condições de ar na qualidade do produto, por meio das análises de condutividade elétrica, absorção de água, porcentagem de germinação e índice de velocidade de germinação (IVG). A secagem foi realizada em secador experimental testando as temperaturas de 30; 40; 50; 60 e 70 ºC, em delineamento inteiramente casualizado, com quatro repetições. Verificou-se que o aumento da temperatura do ar de secagem aumenta os danos nas sementes de pinhão manso, porém não interfere na absorção de água do produto. Já a germinação se mateve elevada, mesmo na temperatura de secagem mais alta, apresentando valores acima de 91% e o IVG não apresentou diferenças estatísticas. Portanto, conclui-se que a qualidade fisiológica das sementes de pinhão manso é afetada pela temperatura de secagem.The post-harvest drying of jatropha is the most frequently used process to ensure quality and stability. The aim of this work was to analyze the seed drying of jatropha and check the effect of air conditions on seed quality, through electrical conductivity, water absorption germination test and index of germination velocity (IGV). Drying was done in an experimental drier, testing controlled temperatures of 30; 40; 50; 60 and 70 ºC, in a randomized design with four replications. It was found that increasing the temperature of the drying air increases the damage on the seeds of jatropha, but does not interfere with water absorption of the product. The germination is high even at the highest drying temperature, with values above 91% and IVG did not show statistical differences. Therefore, it was concluded that the physiological quality of seeds of Jatropha curcas is affected by drying temperature.

In the present work, the level of irradiation of 20, 60 and 100% of natural light on the growth, content and composition of the essnetial oil of “hortelã do campo” (Hyptis marrubioides) was evaluated. The experiment was conducted in the completely randomized design with twenty-five replicates per treatment. After 132 days of cultivation, plant growth and the content and compositoin of the essntial oil were investigated. At 100% of irradiation, the plants presented less height as compared with the shaded treatments. The ratio branch/leaf was higher at the level of irradiation of 20%. It was found that in the full sunshine, the plants showed lower height as compared with the shaded treatments. The leaf/ branch ratio was higher at the lowest level of irradition of 20%. The number of branches and the plant mass accumulation were greatest at the level of irradition of 100%. The content of essential oil was not influenced by the levels of irradiation, but the yield was greater when grown at the level of 100% of irradiation. The chemical composition of the essential oil of Hyptis marrubioides did not present any alterations with the different levels of irradiation, except for iso-3-tujanol and ?-cadineno, which presented highest concentrations in plants grown, respectively at 100 and 60% of irradiation. The major component of teh essential oil of H. marrubioides, independent of the levels of irradiation was cistujona oxygenated monoterpene (37.78%), followed by sequiterpenes hiydrocarbonetes (E)-cariofileno (14.93%), ?-copaeno (11.03%) and ?-muuroleno (9.60%).No presente trabalho avaliou-se o nível de irradiância de 20, 60 e 100% de luz natural no crescimento, teor e a composição do óleo essencial de hortelã-do-campo (Hyptis marrubioides). O experimento foi conduzido no delineamento inteiramente casualizado, com vinte e cinco repetições por tratamento. Após 132 dias de cultivo analisou-se o crescimento da planta, o teor e a composição do óleo essencial. Em 100% de irradiância as plantas apresentaram menor altura, quando comparado aos tratamentos sombreados. A relação ramo/folha foi maior no nível de irradiância de 20%. O número de ramos e o acúmulo de fitomassa foram maiores no nível de irradiância de 100%. O teor de óleo essencial não foi influenciado pelos níveis de irradiância, mas o rendimento foi maior quando cultivadas a 100% de irradiância. A composição química do óleo essencial não apresentou alterações com os diferentes níveis de irradiância, exceto para o iso-3-tujanol e ?-cadineno, que apresentaram maiores concentrações em plantas cultivadas respectivamente a 100 e 60% de irradiância. O componente majoritário do óleo essencial de H. marrubioides, independente dos níveis de irradiância foi o monoterpeno oxigenado cistujona (37,78%), seguido pelos hidrocarbonetos sesquiterpênicos (E)-cariofileno (14,93%), ?-copaeno (11,03%) e ?-muuroleno (9,60%).

In this research it was studied the influence of the radiation intensity on growth and on the essential oil yield of Baccharis trimera. The plants growth was studied under four levels of radiation: 100, 60, 50 and 20% during a 259-day long period. Plants were then evaluated for height, number of nodes, branches, stem diameter (at the height of the colon), biomass formation, and content and composition of the oil. The highest level of radiation caused increases in all of the variables evaluated, except for height, which was reduced with increasing radiation. Essential oil content also increased with increasing level of radiation.A influência do nível de radiância no crescimento da planta, rendimento e qualidade do óleo essencial de Baccharis trimera foi estudado neste trabalho. As plantas foram cultivadas sob 4 níveis de radiância: 100, 60, 50 e 20% durante um período de 259 dias. Foram avaliadas, quanto ao comprimento, número de nós, ramos, diâmetro do caule (altura do colo), acúmulo de fitomassa, conteúdo e composição do óleo essencial. O nível mais elevado de radiação causou aumento em todas as características avaliadas, exceto para o comprimento, que foi reduzido com o aumento na radiação. O rendimento de óleo essencial também aumentou com a elevação do nível de radiância.

Com este trabalho, objetivou-se verificar a ação da celulase sobre a decomposição do pergaminho das sementes de cafeeiro. Foram implantados dois experimentos, sendo o primeiro, realizado em condições in vitro. Constituído por três ensaios para avaliar os efeitos das concentrações da enzima, do substrato e sacarificação enzimática do pergaminho. O segundo, foi realizado em condições in vivo, com imersão das sementes de cafeeiro em solução de celulase na concentração de 1,6 g.L-1 em tampão citrato de potássio 0,05 mol.L-1. Observou-se que a quantidade de açúcar liberado in vitro foi diretamente relacionada com a concentração de enzima, e que houve aumento expressivo e linear na quantidade de açúcares redutores liberados nas duas concentrações (1,6 e 3,2 g.L-1) à medida que se aumentou o tempo de incubação das amostras de substrato (pergaminho) com a solução enzimática. Foi verificado também, nos testes de germinação e emergência, que as sementes sem pergaminho apresentam maiores índices de velocidade e porcentagem de germinação e emergência comparado às sementes com pergaminho e que a celulase nesta concentração ainda não proporciona aumento na velocidade de germinação das sementes, necessitando-se de concentrações mais elevadas.This paper aimed to verify the action of cellulase on the decomposition of parchment of coffee seeds. For this purpose two experiments were conducted and the first, was realized in vitro conditions, containing of three assays (effect of the concentration of the enzyme, effect of the concentration of the substrate and enzyme scarification of the parchment). The second was made in vitro conditions soaking of coffee seeds in a cellulase solution at the concentration of 1.6 g.L-1 in 0.05 M potassium citrate buffer. It was found that the amount of sugar released was directly related with the enzyme concentration and that there was a linear and marked increase in the amount of reducing sugars released at the concentrations (1.6 and 3.2 g.L-1) well as incubation time of the substrate samples (parchment) with the increase of enzyme solution. It was also found that that the parchmentless seeds presented increase in the germination and emergence velocity index, germination and emergence percentage as compared with the seeds with a parchment and that cellulase at this concentration do not provide any increase in germination velocity of seeds, being needed higher concentrations.

Realizou-se este trabalho com o objetivo de estudar condições nutricionais e hormonais para maximizar a produção de calos friáveis de carqueja [Baccharis trimera (Less.) DC]. Foi verificado que a iniciação de calo é dependente de fitorreguladores e da concentração do meio. A melhor indução de calo ocorreu em meio MS contendo 50% da concentração de sais, inositol e vitaminas, suplementado com 15,0 mM ANA. Proliferação de brotos foi obtida pelo uso de TDZ.The influence of various growth regulators and medium concentrations, in different quantities, on the in vitro callus induction of carqueja [Baccharis trimera (Less.) DC.] was evaluated. It was found that the callus initiation was dependent on both, the growth regulator and medium concentration. The highest callus induction and development were obtained by using 15.0 mM 1-napthaleneacetic acid (NAA) as growth regulators and half strength of salts, vitamins, and myo-inositol of Murashige and Skoog medium. In vitro shoot proliferation was obtained by using thidiazuron.

As sementes de café germinam lentamente em razão de a semeadura ser feita logo após a colheita, o que coincide com o período frio do ano, e também por causa de o pergaminho constituir uma barreira que dificulta a absorção de água pela semente. Objetivando avaliar o efeito da aplicação exógena da enzima celulase sobre a germinação de sementes de café, foi realizado o experimento com sementes de café, cultivar Acaiá do Cerrado, imersas em solução de celulase em diferentes concentrações (0; 1,6; 3,2 e 6,4 g.L-1 em tampão citrato de potássio 0,05 M e pH 4,8), por 0, 72 e 144 horas. Observou-se que a enzima celulase na concentração de 6,4 g.L-1 proporcionou maior índice de velocidade de germinação (IVG) e porcentagem de germinação, quando as sementes ficam em contato com a solução por 144 horas.The seeds of coffee trees germinate slowly, because the seeding is done soon after the crop is harvested, what coincides with the cold period of the year, and also due to the parchment, which constitutes a barrier that hinders the absorption of water for the seed. Aiming to evaluate the effect of exogen application of cellulase enzyme to coffee seed germination, an experiment was performed soaking coffee seeds, cultivar “Acaiá do Cerrado”, in cellulase solution at different concentrations (0, 1.6, 3.2, and 6.4, g L-1) in 0.05 M K-citrate buffer at pH 4.8 for 0, 72, and 144 hours. It was found that the cellulase enzyme at the concentration of 6.4 g L-1 provided the greatest index of speed germination and the greatest germination percentage when the seeds remained in contact with the solution for 144 hours.

Strychnos pseudoquina, popularmente conhecida por quina-de-cerrado, atinge em média 4 metros de altura e pode ser encontrada no cerrado brasileiro. Possui folhas opostas, simples, pecioladas, ápice agudo a obtuso ou ligeiramente acuminado. A floração ocorre de janeiro a abril, e suas flores são pequenas, seu fruto possui baga com cerca de 2 cm, amarela; pericarpo coriáceo; possui de 1 a 4 sementes. O tronco possui casca espessa e rimosa e possui finalidade medicinal (Almeida et al, 1998). O uso de fitoterápicos no mundo é cada vez maior e o Brasil possui um extensa flora utilizada na medicina. A quina já foi uma das plantas mais utilizadas no país por ser considerada tônica e febrífuga de alto valor, útil no combate as inflamações dos gânglios mesentérios e as moléstias do fígado, baço e estômago (Almeida et al, 1998). Reconhecendo a importância regional para a população da região do cerrado da referida espécie na medicina popular e alimentação, torna se necessário o estabelecimento de técnicas que tornem viáveis a multiplicação em larga escala desta espécie. Este trabalho teve como objetivo estudar a germinação das sementes de quina em diferentes concentrações de meio de cultura e luz (claro e escuro), visando o estabelecimento in vitro desta espécie para futuros trabalhos sobre a multiplicação em larga escala.

Abstract: Castor bean seeds are the raw material used for extracting oil destined to pharmaceutical and industrial ends. The appropriate application of post-harvest technologies, such as processing, is required to ensure the production of top quality seeds. Thus, this study aimed at assessing the physiological quality of seeds from two castor bean genotypes, classified according to their specific mass and size. Castor seeds of the genotypes EVF102 and EVF103 were mechanically harvested and then classified with the aid of cylindrical sieves and a densimetric table. This procedure was conducted at the processing unit of the company Sementes Goiás LTDA [Goiás Seeds Co.]. Evaluations of seed physiological quality were carried out in the Seed Laboratory of Instituto Federal Goiano, following a completely randomized design. The testing was conducted in four replicates, in a 2 x 2 x 5 factorial scheme (two genotypes x two sizes x five specific masses). The following parameters were appraised: (a) mass of one thousand seeds; (b) germination; (c) germination speed index; (d) emergence test; (e) emergence speed index; (f) electrical conductivity; and (g) accelerated aging. Genotype EVF102 seeds sized 8 mm, as classified by the densimetric table, showed the highest physiological quality, considering the germination, germination speed index, and emergence. Seeds of the genotype EVF103, sorted by size, exhibited no difference in physiological quality.
