The objective of this study is to document the pathological manifestations of the Casarão dos Fabricantes, one of the oldest buildings in Fortaleza, which was severely damaged by a major fire in 2020. Despite its historical significance, there are no records detailing the condition of the building either before or after the fire. The internal walls and two facades were the only construction elements that were not destroyed by the fire. The survey of the pathological manifestations was conducted by visual assessment and infrared thermography, revealing different degradation processes across the evaluated structures and providing a systematic view of the extent of the damage to the building. Mapping this damage provided an integrated approach, offering a scientific basis to support the restoration process.

Abstract The study aimed to develop a mango ice cream (Mangifera indica L. cv. Bourbon) added with whey protein and evaluate its acceptability, using a 9 points hedonic scale. The participants were 157 tasters not affected by cancer and 75 patients undergoing chemotherapy at the Cancer Hospital in Cuiabá, in the state of Mato Grosso. The last group also evaluated the ice cream through sensory and well-being perceptions using the “Check all that apply” - CATA method. The ingredients for the ice cream formulation were mango, passion fruit, whey protein, and unflavored coconut oil. The attributes for the overall assessment established by tasters not affected by cancer were as follows: aroma, appearance, flavor, texture, and overall impression. The mango ice cream was free of contamination, with 4.6% of protein. In fact, 96.8% of participants reported liking the taste of ice cream. Even though the ice cream did not have added sugar, 87.26% of the participants rated the ideal sweetness level. Furthermore, more than 95% of them reported that they would buy the product. Regarding the tests carried out with patients undergoing chemotherapy treatment, 32% said the ice cream relieved their symptoms of nausea and 90% responded that they liked it in the affective test, validating the acceptability of the ice cream. The study proved the healthiness and sanity of consuming this ice cream, which is a well-accepted food, bringing benefits to patients undergoing the chemotherapy treatment. Still, above all, it used a fruit that has a regional and comfort food appeal, since for many patients, consuming mango ice cream brought back memories of their childhood.

Endemic and pandemic viruses represent significant public health challenges, leading to substantial morbidity and mortality over time. The COVID-19 pandemic has underscored the urgent need for the development and discovery of new, potent antiviral agents. In this study, we present the synthesis and anti-SARS-CoV-2 activity of a series of benzocarbazoledinones, assessed using cell-based screening assays. Our results indicate that four compounds (4a, 4b, 4d, and 4i) exhibit EC50 values below 4 μM without cytotoxic effects in Calu-3 cells. Mechanistic investigations focused on the inhibition of the SARS-CoV-2 main protease (Mpro) and papain-like protease (PLpro) have used enzymatic assays. Notably, compounds 4a and 4b showed Mpro inhibition activity with IC50 values of 0.11 ± 0.05 and 0.37 ± 0.05 µM, respectively. Furthermore, in silico molecular docking, physicochemical, and pharmacokinetic studies were conducted to validate the mechanism and assess bioavailability. Compound 4a was selected for preliminary drug-likeness analysis and in vivo pharmacokinetics investigations, which yielded promising results and corroborated the in vitro and in silico findings, reinforcing its potential as an anti-SARS-CoV-2 lead compound.

Abstract Zika virus (ZIKV) infection is a global public health concern linked to adult neurological disorders and congenital diseases in newborns. Host lipid metabolism, including lipid droplet (LD) biogenesis, has been associated with viral replication and pathogenesis of different viruses. However, the mechanisms of LD formation and their roles in ZIKV infection in neural cells are still unclear. Here, we demonstrate that ZIKV regulates the expression of pathways associated with lipid metabolism, including the upregulation and activation of lipogenesis-associated transcription factors and decreased expression of lipolysis-associated proteins, leading to significant LD accumulation in human neuroblastoma SH-SY5Y cells and in neural stem cells (NSCs). Pharmacological inhibition of DGAT-1 decreased LD accumulation and ZIKV replication in vitro in human cells and in an in vivo mouse model of infection. In accordance with the role of LDs in the regulation of inflammation and innate immunity, we show that blocking LD formation has major roles in inflammatory cytokine production in the brain. Moreover, we observed that inhibition of DGAT-1 inhibited the weight loss and mortality induced by ZIKV infection in vivo. Our results reveal that LD biogenesis triggered by ZIKV infection is a crucial step for ZIKV replication and pathogenesis in neural cells. Therefore, targeting lipid metabolism and LD biogenesis may represent potential strategies for anti-ZIKV treatment development.

Esta pesquisa assumiu como objetivo discutir e analisar as principais questões que devem consideradas ao planejar aulas de educação física na educação infantil e se propõe a responder à seguinte pergunta: quais questões envolvem o planejamento das aulas de educação física para a educação infantil? Para isso utilizou-se como metodologia a narrativa autobiográfica no formato de relato de experiência. Os instrumentos de coleta de dados foram subsidiados pelo resgate dos portfólios elaborados durante o período de estágio, bem como a literatura científica acerca do tema. A pesquisa foi dividia em dois eixos de análise: saberes experienciais no/do/com o currículo da educação infantil e educação física e saberes experienciais sobre o planejamento didático-pedagógico das aulas. Os resultados apontam o planejamento de aula baseado nas concepções dos currículos, da escola, do professor, da aula e da criança.

RESUMO Objetivo: descrever as características de clustering e switching da prova de fluência verbal semântica e fonológica de deficientes auditivos adultos - usuários do português brasileiro - e verificar sua relação com o total de palavras evocadas e fatores biossociais. Métodos: 42 indivíduos deficientes auditivos adultos, usuários do português brasileiro oral participaram desta pesquisa. Realizaram as provas de fluência verbal semântica ("animais") e fonológica ("palavras com 'F'") e foram contabilizados: (a) número total de palavras, (b) número de categorias evocadas, (c) tamanho médio do cluster e (d) número de switches. Comparou-se aos dados biossociais e às características da deficiência auditiva e do dispositivo auditivo eletrônico. Resultados: o total de palavras evocadas foi de 16,38±6,18 para prova semântica e 10,88±6,1 para fonológica, sendo acessadas todas as categorias previamente estabelecidas. A média do tamanho do cluster foi de 1,41±0,8 para fluência verbal semântica e 0,71±0,73 para fonológica. O número médio de switches foi de 7,14±3,69 para fluência semântica e 6,36±4,17 para fonológica. This paper aims to present an approach that was developed as part of the Community-University Alliance Research (CURA) entitled “Coastal Communities Challenges of the Estuary and the Gulf of St. Lawrence under a Changing Climate” (CURA-CCC). We describe the process of capacity building we used in several coastal communities in Eastern Canada to enhance their resilience. We use the examples of the municipalities of Maria and Bonaventure in Quebec to illustrate this approach, which is based on participatory action research. The co-construction of adaptation strategies that emerged from this work is based on an exploratory model examining the hazards and their effects on these communities, considering the uncertainty of knowledge and capacities of the actors to act. These coastal habitats are subject to climate change hazards including storm surges causing coastal erosion, floods and river overflows. It is important for these communities to develop a better understanding of these issues when developing long-term collective solutions that can go beyond their local territories. In fact, we believe that such an approach could potentially be used in several other coastal rural areas of eastern Canada.

RESUMO Objetivo: descrever proposta metodológica para a dinâmica de atuação do comitê de juízes por questionário eletrônico implementada e testada na adaptação cultural do instrumento Diabetes Empowerment Scale-Short Form. Método: estudo metodológico realizado com 38 especialistas das áreas de Letras e Ciências da Saúde que avaliaram o instrumento traduzido via questionário eletrônico, desenvolvido e aplicado por meio da ferramenta web e-Surv. Utilizando as respostas ao questionário, o nível de aceitação dos juízes foi avaliado por meio de frequências absolutas e relativas e o nível de concordância entre os juízes foi avaliado por meio do cálculo do coeficiente de Kappa de Cohen-Fleisss. As análises foram realizadas no ambiente de programação estatística R. Resultados: a colaboração interdisciplinar aliada a uma metodologia de coleta de dados online resultou em uma heterogeneidade de opiniões, o que gerou baixa concordância entre os juízes na adaptação do instrumento traduzido. No entanto, acredita-se que essa diversidade de opiniões tenha gerado sugestões que enriqueceram o processo tradutório. Conclusão: a ferramenta online se mostrou confiável, segura e efetiva na dinâmica do comitê de juízes para avaliar instrumentos em saúde.

This study aimed to check for any significant differences in perceived quality of life, specifically aspects of a physical nature, among volunteers who are more physically active and those less physically active in a university community. The sample consisted of 1,966 volunteers in a university community in Brazil. To assess physical activity levels, volunteers responded to the International Physical Activity Questionnaire (IPAQ), and to analyse the perception of quality of life they responded to WHOQOL-bref, which is classified into three groups according to level of physical activity, taking into account the metabolic equivalent index (MET) over a full week. For comparison, consideration was given to the first and third tertiles, respectively, namely groups of more and less active students. The results indicated that individuals who engaged in more physical activity had a more positive perception of quality of life compared to those who were less active in physical aspects related to the ability to work, energy for day-to-day activities and locomotion.

La eficiencia de remoción de materia particulada y nutrientes disueltos de efluentes de estanques de reproductores de Litopenaeus vannamei fue evaluada a escala de laboratorio, empleando tratamientos de sedimentación, filtración por Crassostrea rhizophorae y absorción por Ulva fasciata. En cada tratamiento se empleó un tiempo de residencia hidráulica del efluente de seis horas. En la etapa de sedimentación se utilizaron estanques con 90 L del efluente. Para la etapa de filtración, se emplearon estanques con 20 L del efluente sobrenadante de la etapa de sedimentación. En la absorción se utilizaron estanques con 15 L del efluente sobrenadante de la etapa de filtración. Al final de cada etapa se colectaron muestras de 500 mL para los análisis de calidad de agua. Los resultados indicaron que en la sedimentación, la turbidez, clorofila-a y sólidos suspendidos totales presentaron una remoción de 93,8%, 94,5% y 65,9%, respectivamente. La filtración alcanzó eficiencias de 89,3% y 100% para bacterias totales y clorofila a, respectivamente. En la absorción, fosfato (PO4-3 ), amonio (N-NH4-) y nitrato (N-NO3) fueron reducidos en 53,6%, 49,6% y 70,2%, respectivamente, en relación a los valores del estanque control. Los tratamientos combinados de sedimentación, filtración y absorción, bacterias totales (95,1%), turbidez (97,1%) sólidos suspendidos totales (81,3%), clorofila-a (99,1%), amonio (54,1%), nitrito (58,0%), nitrato (69,2%) y fosfato (52,9%), presentaron una alta eficiencia de remoción, en relación a los valores del efluente bruto. Se concluye que la combinación de tratamientos mejora significativamente la calidad de los efluentes del cultivo del camarón Litopenaeus vannamei.

This article describes a conceptual framework in order to explore drivers of vulnerability to climate-related hazards as well as options to adapt to climate change. The framework helps to guide reflections in a simple and non-technical language while being compatible with different conceptualisations of vulnerability. Il was developed through the elaboration of an action plan for adaptation with the municipal administration of Rivière-au-Tonnerre on the northern coast of the Gulf of Saint Lawrence, in the province of Québec in Canada, in collaboration with the planning department of the regional municipality (MRC) of Minganie. The framework consists in separating the causal chain for the development of consequences from climate-related hazards into three phases : The development of secondary hazards in the natural environment ; biophysical damages ; and the disturbance of human activities. It is implemented through two tables guiding a) factors favouring negative consequences, therefore contributing to vulnerability and risk, and b) factors and practices reducing negative consequences, contributing to resilience. The latter table allows exploring practices that can be improved or new actions to implement, pointing to opportunities for adaptation. The example developed in Rivière-au-Tonnerre helped situating the role of different actors at the local, regional, provincial and federal levels. It also allowed indentifying factors that could be quantified to monitor the effects of certain adaptation initiatives.

This study aimed to evaluate the quality of life (QoL) of patients with cervicofacial nonmelanoma skin cancer (NMSC) using the Brazilian Portuguese‐adapted and validated version of the skin cancer index (SCI). After collecting demographic and clinical data from 182 patients with cervicofacial NMSC, the Brazilian versions of the SCI and the Dermatological Life Quality Index (DLQI) were applied preoperatively (T0) and 4 months postoperatively (T1). Assessments were carried out using the Shapiro–Wilk test, Student's t‐test, Mann–Whitney test, Kruskal–Wallis test, and Spearman's correlation. The QoL was evaluated using the Student's t‐test in paired samples at T0 and T1. Significant results were observed, with an increase in scores on the SCI scale in all its dimensions and a decrease in scores on the DLQI scale, demonstrating better postoperative QoL. The variables that presented significant results on the total scale, which indicated better QoL, were men, without children, income above four minimum wages, from the private sector, did not report pruritus, and scalp lesions. The QoL measurement indicated a change from the baseline and improvement after 4 months postoperatively in all subscales, indicating that surgical treatment increased the QoL of these patients from an emotional, social, and physical appearance point of view. The multivariate analysis produced several statistically significant findings in relation to emotional, social, appearance, and total scores. [ABSTRACT FROM AUTHOR]
