Abstract While the development of weakly coordinating anions (WCAs) received much attention, the progress on weakly coordinating and inert solvents almost stagnated. Here we study the effect of strategic F-substitution on the solvent properties of fluorobenzenes C6FxH6−x (xFB, x = 1–5). Asymmetric fluorination leads to dielectric constants as high as 22.1 for 3FB that exceeds acetone (20.7). Combined with the WCAs [Al(ORF)4]− or [(FRO)3Al-F-Al(ORF)3]− (RF = C(CF3)3), the xFB solvents push the potentials of Ag+ and NO+ ions to +1.50/+1.52 V vs. Fc+/Fc. The xFB/WCA-system has electrochemical xFB stability windows that exceed 5 V for all xFBs with positive upper limits between +1.82 V (1FB) and +2.67 V (5FB) vs. Fc+/Fc. High-level ab initio calculations with inclusion of solvation energies show that these high potentials result from weak interactions of the ions with solvent and counterion. To access the available positive xFB potential range with stable reagents, the innocent deelectronator salts [anthraceneF]+∙[WCA]− and [phenanthreneF]+∙[WCA]− with potentials of +1.47 and +1.89 V vs. Fc+/Fc are introduced.

BackgroundExisting literature on moral conflicts that healthcare professionals encounter in dementia care has explored, amongst others, issues related to autonomy, decision-making capacity, privacy, and more. Notably, conflicts related to healthcare professionals who support informal dementia caregiving and who are confronted with family members being overburdened with their care responsibly remains an underexplored topic in the current literature, particularly in the context of Low-and Middle-Income Countries. The present paper introduces such an encounter, presenting an ethical case analysis of a conflict that occurred during a larger research project conducted in North Macedonia.Case to be studiedDue to the absence of formal care services that could have relieved an overburdened family caregiver, healthcare professionals felt compelled to reach out to the uninvolved adult daughters, requesting them to participate in their parents’ care. Wondering about whether their reaching out to the daughters might count as an attempt of pressure and undue interference, professionals conflicted over the appropriateness of their action. This paper follows up on their concern, ethically assessing the professionals’ action. To answer the question on whether the healthcare professionals acted appropriately or not, and to what extent, theories of filial duties are applied, embedding their action in the larger context of dementia care in North Macedonia.Results and conclusionIt is argued that the lack of formal care services in North Macedonia is of utmost relevance to the conflict. Thus, the conclusion is that the ethical inappropriateness of the case is to be located not so much with the action of the healthcare professionals but with the state because of its failure to provide professional care services that allow healthcare professionals to take ethically sound actions to counteract overarching burdens that family members face when providing informal dementia care

Background: Medical tape is one of the most ubiquitous resources in the hospital. Although tape is advertised by manufacturers as a single patient-use item, half-used rolls are a common sight in hospitals. Tape is often manipulated by un-sanitized and ungloved hands and comes in close contact with patient skin. Medical tape has the potential to be a source of hospital-acquired infection as it has been documented to be colonized with pathogens ranging from MRSA to Rhizopus. Despite infection risk, currently the only clinical guidelines of tape use are outlined in the Centers for Medicare & Medicaid Services guidance for Hemodialysis patients issued in 2008 that requires “tape should be dedicated to a single patient and discarded after use” as hemodialysis patients are at higher risk of infection. However, there is a lack of standards in the practice of tape use across hospital systems. Methods: To understand the current practices and beliefs of tape use at our institution, we developed a standardized survey to query individuals from various roles (RN, Physician, Patient Care Technicians, respiratory therapists, phlebotomists) across all patient care areas at a 746-bed academic, tertiary care center. Results: 52 units were surveyed, including 225 employees. Qualitative analysis revealed a wide variety of uses for medical tape for patient care, with venipuncture, securing IVs, and wound dressings being the most common. Only 1.4% of individuals reported single use of tape rolls. 54% of individuals reported tape use behaviors that carry an elevated risk for inoculation of pathogens. 70% of individuals reported that tape was discarded after the patient was discharged from their respective area. These practices did not change across procedure-heavy areas such as the Emergency Department or the Operating Rooms, in fact only 22% of individuals surveyed reported single use of tape in these areas. Beliefs about tape use varied: 95% of individuals agreed that a roll of tape could be used multiple times on a single patient, and 52% of individuals agreed that a roll of tape could be used on multiple patients. Conclusions: Tape use practices varied across hospital units, indicating the need for standardized policies for tape use and storage. Beliefs about tape not being a single-use item were consistent across the hospital and suggest that education and culture change efforts are needed to decrease the risk for hospital-acquired infections from improper medical tape use.

Geospatial data are fundamental in most global-change and sustainability-related domains. However, readily accessible information on data quality and provenance is often missing or hardly accessible for users due to technical or perceptual barriers, for example, due to unstructured metadata information or missing references. Within an interdisciplinary process encompassing perspectives of data users, data producers, and software developers, we identified major needs to facilitate effective fitness-for-use assessments by data users and developed approaches to address these. We provided a stylized analysis of large-scale land use data to showcase selected approaches. To support data users, interoperable quality and provenance information need to be meaningfully represented. Data producers need efficient workflows and tools supporting them in creating high-quality, structured and detailed quality and provenance information. Our newly developed approaches to increase the availability of structured metadata synthesize new and existing tools to extract metadata or to generate provenance data during processing. Within our approaches to improve interoperability and accessibility we present novel tools to support (i) the creation of curated and linked registers of data quality indicators and thematic terms, and (ii) linked visualization of data quality and provenance information. Following our approaches increases transparency, facilitates fitness-for-use assessments, and ultimately improves research quality.

In the spring of 2020, the SARS-CoV-2 pandemic presented a formidable challenge to national and global healthcare systems. Immunocompromised individuals or those with relevant pre-existing conditions were particularly at risk of severe coronavirus disease 2019 (COVID-19). Thus, understanding the immunological processes in these patient groups is crucial for current research. This study aimed to investigate humoral immunity following vaccination and infection in liver transplant recipients. Humoral immunity analysis involved measuring IgG against the SARS-CoV-2 spike protein (anti-S IgG) and employing a surrogate virus neutralization test (sVNT) for assessing the hACE2 receptor-binding inhibitory capacity of antibodies. The study revealed that humoral immunity post-vaccination is well established, with positive results for anti-S IgG in 92.9% of the total study cohort. Vaccinated and SARS-CoV-2-infected patients exhibited significantly higher anti-S IgG levels compared to vaccinated, non-infected patients (18,590 AU/mL vs. 2320 AU/mL, p < 0.001). Additionally, a significantly elevated receptor-binding inhibitory capacity was observed in the cPassTMTM sVNT (96.4% vs. 91.8%, p = 0.004). Furthermore, a substantial enhancement of anti-S IgG levels (p = 0.034) and receptor-binding inhibition capacity (p < 0.001) was observed with an increasing interval post-transplantation (up to 30 years), calculated by generalized linear model analysis. In summary, fully vaccinated liver transplant recipients exhibit robust humoral immunity against SARS-CoV-2, which significantly intensifies following infection and with increasing time after transplantation. These findings should be considered for booster vaccination schemes for liver transplant recipients.

IntroductionThe increasing number of people living with dementia and its burden on families and systems particularly in low- and middle-income countries require comprehensive and efficient post-diagnostic management. This study aimed to explore the acceptability and efficacy of a multi-professional case management and psychoeducation model (North Macedonia Interprofessional Dementia Care, or NOMAD) delivered by mobile teams for people with dementia and their caregivers in North Macedonia.MethodWe conducted a two-arm randomized controlled trial comparing the intervention with treatment as usual. Participants were recruited from 12 general practitioner (GP) offices in the Skopje region. The NOMAD intervention included the delivery of a personalized care plan over four home visits to dyads of people with dementia and their caregivers by a team including a dementia nurse and a social worker, in collaboration with GPs and dementia experts, and the introduction of a caregiver manual. We assessed caregivers' depressive symptoms, burden, and quality of life and the neuropsychiatric symptoms, daily living activities, and service utilization of people with dementia at baseline and follow-up; we also assessed the acceptability of the intervention by analyzing case notes and attendance rates.ResultsOne hundred and twenty dyads were recruited and randomized to either the control (n = 60) or the intervention group (n = 60). At follow-up, caregivers in the intervention group had, on average, scores that were 2.69 lower for depressive symptoms (95% CI [−4.75, −0.62], p = 0.012), and people with dementia had, on average, 11.32 fewer neuropsychiatric symptoms (95% CI [−19.74, −2.90], p = 0.009) and used, on average, 1.81 fewer healthcare services (95% CI [−2.61, −1.00], p < 0.001) compared to the control group. The completion of the home visits was 100%, but the intervention's acceptability was underpinned by relationship building, GP competencies, and resources to support families with dementia. There were no differences in the caregivers' quality of life and burden levels or daily living activities in people with dementia. NOMAD is the first case management, non-pharmacological, and multi-professional intervention tested in North Macedonia.DiscussionThe trial showed that it is effective in reducing caregivers' depressive symptoms and neuropsychiatric symptoms in people with dementia and the burden on health and social care services, and it is acceptable for families. Implementing NOMAD in practice will require building primary care capacity and recognizing dementia as a national priority.

Abstract Conservation funding is currently limited; cost‐effective conservation solutions are essential. We suggest that the thousands of field stations worldwide can play key roles at the frontline of biodiversity conservation and have high intrinsic value. We assessed field stations’ conservation return on investment and explored the impact of COVID‐19. We surveyed leaders of field stations across tropical regions that host primate research; 157 field stations in 56 countries responded. Respondents reported improved habitat quality and reduced hunting rates at over 80% of field stations and lower operational costs per km2 than protected areas, yet half of those surveyed have less funding now than in 2019. Spatial analyses support field station presence as reducing deforestation. These “earth observatories” provide a high return on investment; we advocate for increased support of field station programs and for governments to support their vital conservation efforts by investing accordingly.

Abstract Using Industrial Revolution 4.0 (IR4.0) technology, companies can upscale their innovation in organizing, managing, and controlling the value chain life cycle. IR4.0 is anticipated to bring challenges and opportunities to developing economies such as Malaysia, but due to its novelty to the Malaysian business community, the concept still needs clarity in its definition for proper understanding and business practice. This paper aims to examine the impact of IR4.0 adoption on Malaysian small and medium enterprises (SMEs) by analyzing the organizational readiness, relative advantage, compatibility, top management support, government regulation, and competitive pressure, and its relationship with the adoption of IR4.0 to Malaysian SMEs. This study framework is based on the diffusion of innovation theory (DOI) and technology–organization–environment (TOE) framework. The study results verified the importance of relative advantage, compatibility, competitive pressure, and top management support as significant predictors of IR4.0 adoption. The study is expected to benefit regulators and business ecosystems in understanding the challenges in implementing IR4.0 in Malaysia and formulating intervention processes and programs for successful IR4.0 adoption.

Abstract In the last decade, drones have become an affordable technology offering highly mobile aerial platforms that can carry a range of sensory equipment into hitherto uncharted areas. Drones have thus become a widely applicable tool for surveying animal populations and habitats to assist conservation efforts or to study the behavioural ecology of species by monitoring individual and group behaviour. Here, we review current applications for drone surveys and the potential of recently developed computer algorithms for automatic species detection and individual tracking in drone footage. We further review which factors are reportedly associated with animal disturbance during drone presentations and how drones may be used to study anti‐predator behaviour. Drone surveys of species and their environments allow scientists to create digital terrain models of habitats, estimate species abundance, monitor individual behaviour and study the composition, spatial organization and movement of groups. As drones can influence the behaviour of many bird and mammal species directly, they also provide an experimental tool to study animal responses to novel situations, including the drone itself. We conclude that the combined use of drones and automated detection software can assist population estimates and opens new possibilities to study individual and collective behaviour. With regard to drone‐related disturbance and their potential use as predator models, we recommend to interpret results against the background of population‐specific predation pressure and sources of anthropogenic disturbance.

Assessing immune responses to cytomegalovirus (CMV) after liver transplant in patients on immunosuppressive therapy remains challenging. In this study, employing ELISPOT assays, 52 liver-transplant recipients were evaluated for antiviral T-cell activity in peripheral blood mononuclear cells (PBMCs), measuring interferon-γ (IFN-γ) secretion upon stimulation with CMV-specific peptides (CMV peptide pool, CMV IE-1, and pp65 antigens). Parameters such as stimulation index, mean spot size, and mean spot count were measured. The study found that heightened immunosuppression, especially with prednisolone in triple therapy, significantly dampened CMV-specific immune responses. This was demonstrated by decreased IFN-γ production by CMV-specific T-cells (CMV peptide pool: p = 0.036; OR = 0.065 [95% CI: 0.005–0.840], pp65 antigen: p = 0.026; OR = 0.048 [95% CI: 0.003–0.699]). Increased immunosuppression correlated with reduced IFN-γ secretion per cell, reflected in smaller mean spot sizes for the CMV peptide pool (p = 0.019). Notably, shorter post-transplant intervals correlated with diminished antiviral T-cell IFN-γ release at two years (CMV peptide pool: p = 0.019; IE antigen: p = 0.010) and five years (CMV peptide pool: p = 0.0001; IE antigen: p = 0.002; pp65 antigen: p = 0.047), as did advancing age (pp65 antigen: p = 0.016, OR = 0.932, 95% CI: 0.881–0.987). Patients with undetectable CMV antigens had a notably higher risk of CMV reactivation within six months from blood collection, closely linked with triple immunosuppression and prednisolone use. These findings highlight the intricate interplay between immunosuppression, immune response dynamics, and CMV reactivation risk, emphasizing the necessity for tailored immunosuppressive strategies to mitigate CMV reactivation in liver-transplant recipients. It can be concluded that, particularly in the early months post-transplantation, the use of prednisolone as a third immunosuppressant should be critically reconsidered. Additionally, the use of prophylactic antiviral therapy effective against CMV in this context holds significant importance.

Abstract The steepness of dominance hierarchies provides information about the degree of competition within animal social groups and is thus an important concept in socioecology. The currently most widely used metrics to quantify steepness are based on David's scores (DS) derived from dominance interaction networks. One serious drawback of these DS‐based metrics is that they are biased, that is, network density systematically decreases steepness values. We provide a novel approach to estimate steepness based on Elo‐ratings, implemented in a Bayesian framework (STEER: Steepness estimation with Elo‐rating). We evaluate and validate its performance by means of experimentation on empirical and artificial datasets and compare its performance to that of several other steepness estimators. STEER has two key advantages. First, it is unbiased, precise and more robust to data density than DS‐based steepness. Second, it provides explicit probability distributions of the estimated steepness coefficient, which allows uncertainty assessment. In addition, it relies on the same underlying concept and is on the same scale as the original measure, and thus allows comparison to existing published results. STEER provides a considerable improvement over existing methods to estimate dominance hierarchy steepness. We demonstrate its application with an example comparing within‐ and between species variation in steepness in a comparative analysis and present guidelines on how to use it. The R package EloSteepness allows convenient numeric and graphical assessment of the new steepness measure.

Short-term memory is implicated in a range of cognitive abilities and is critical for understanding primate cognitive evolution. To investigate the effects of phylogeny, ecology and sociality on short-term memory, we tested the largest and most diverse primate sample to date (421 non-human primates across 41 species) in an experimental delayed-response task. Our results confirm previous findings that longer delays decrease memory performance across species and taxa. Our analyses demonstrate a considerable contribution of phylogeny over ecological and social factors on the distribution of short-term memory performance in primates; closely related species had more similar short-term memory abilities. Overall, individuals in the branch of Hominoidea performed better compared to Cercopithecoidea, who in turn performed above Platyrrhini and Strepsirrhini. Interdependencies between phylogeny and socioecology of a given species presented an obstacle to disentangling the effects of each of these factors on the evolution of short-term memory capacity. However, this study offers an important step forward in understanding the interspecies and individual variation in short-term memory ability by providing the first phylogenetic reconstruction of this trait’s evolutionary history. The dataset constitutes a unique resource for studying the evolution of primate cognition and the role of short-term memory in other cognitive abilities.

Abstract Background Multiple myeloma (MM) is the third most common hematologic malignancy with increasing importance due to improving treatment strategies and long-term outcomes in an aging population. This study aims to analyse influencing factors on health-related quality of life (HRQoL), such as treatment strategies, participation in a clinical trial and patient characteristics like anxiety, depression, gender, and age. A better understanding of the individual factors in context with HRQoL could provide a helpful instrument for clinical decisions. Methods In this prospective observational study, the HRQoL of MM patients with different therapies (first-line and relapse) was quantified by standardized questionnaires (EORTC QLQ-C30 and -MY20) in the context of sociodemographic data, individual anxiety and depressiveness (PHQ-4), and a selected number of clinical parameters and symptoms at defined time-points before, during, and after therapy. Results In total, 70 patients were included in the study. The median age of the study cohort was 62 years. 44% were female and 56% were male patients. More than half of the patients were fully active with an ECOG 0. Global health status was significantly higher in patients with first-line treatment and even increased after start of therapy, while the pain level decreased. In contrast, patients with relapsed MM reported a decreasing global health status and increasing pain. Additionally, there was a higher global health status in less anxious/depressive patients. HRQoL decreased significantly after start of chemotherapy in the parameters body image, side effects of treatment, and cognitive functioning. Tandem stem-cell transplantation was not found to be a risk factor for higher impairment of HRQoL. Participation in a clinical study led to an improvement of most aspects of HRQoL. Among others, increased anxiety and depression, female gender, older age, impaired performance status, and recurrent disease can be early indicators for a reduced HRQoL. Conclusion This study showed the importance of regular longitudinal assessments of patient reported outcomes (PROs) in routine clinical care. For the first time, to our knowledge, we were able to demonstrate a potential impact between participation in clinical trials and HRQoL. However, due to frequently restrictive inclusion criteria for clinical trials, these MM patients might not be directly comparable with patients treated within standard therapy concepts. Further studies are needed to clarify the relevance of this preliminary data in order to develop an individualized, patient-centred, therapy concept.

Unlike in humans, older Barbary macaques do not specifically ignore negative social information or avoid stressful situations, but rather rely on general avoidance strategies to decrease the risk of hazardous social interactions.

Protest in response to unequal reward distribution is thought to have played a central role in the evolution of human cooperation. Some animals refuse food and become demotivated when rewarded more poorly than a conspecific, and this has been taken as evidence that non-human animals, like humans, protest in the face of inequity. An alternative explanation—social disappointment—shifts the cause of this discontent away from the unequal reward, to the human experimenter who could—but elects not to—treat the subject well. This study investigates whether social disappointment could explain frustration behaviour in long-tailed macaques, Macaca fascicularis. We tested 12 monkeys in a novel ‘inequity aversion’ paradigm. Subjects had to pull a lever and were rewarded with low-value food; in half of the trials, a partner worked alongside the subjects receiving high-value food. Rewards were distributed either by a human or a machine. In line with the social disappointment hypothesis, monkeys rewarded by the human refused food more often than monkeys rewarded by the machine. Our study extends previous findings in chimpanzees and suggests that social disappointment plus social facilitation or food competition effects drive food refusal patterns.

Abstract Background Polypharmacy is common in people with dementia. The use of psychotropic drugs (PDs) and other, potentially inappropriate medications is high. The aims of this cross-sectional study were 1) to investigate the use of drugs in people with advanced dementia (PWAD), living at home or in long term care (LTC); 2) to focus on PD use; and 3) to identify determinants of PD use. Methods The study was performed in the context of EPYLOGE (IssuEs in Palliative care for people in advanced and terminal stages of YOD and LOD in Germany). 191 PWAD were included. All drugs that were administered at the date of the examination were recorded. Multiple logistic regression analysis identified determinants of PD use. Results 96% of PWAD received medication with a median number of four drugs. 49.7% received five or more drugs. According to the Beers Criteria 39% of PWAD ≥ 65 years received at least one potentially inappropriate medication. 79% of PWAD were treated with PDs. Older PWAD and PWAD living in LTC facilities received significantly more drugs than younger PWAD, and PWAD living at home, respectively. Dementia etiology was significantly associated with the use of antipsychotics, antidepressants and sedative substances. Place of living was associated with the use of pain medication. Behavioral disturbances were associated with the use of antipsychotics and sedative substances. Conclusions To mitigate the dangers of polypharmacy and medication related harm, critical examination is required, whether a drug is indicated or not. Also, the deprescribing of drugs should be considered on a regular basis. Trial registration Clinicaltrial.gov, NCT03364179 . Registered 6 December 2017.

Many real-world decisions in social contexts are made while observing a partner’s actions. To study dynamic interactions during such decisions, we developed a setup where two agents seated face-to-face to engage in game-theoretical tasks on a shared transparent touchscreen display (‘transparent games’). We compared human and macaque pairs in a transparent version of the coordination game ‘Bach-or-Stravinsky’, which entails a conflict about which of two individually-preferred opposing options to choose to achieve coordination. Most human pairs developed coordinated behavior and adopted dynamic turn-taking to equalize the payoffs. All macaque pairs converged on simpler, static coordination. Remarkably, two animals learned to coordinate dynamically after training with a human confederate. This pair selected the faster agent’s preferred option, exhibiting turn-taking behavior that was captured by modeling the visibility of the partner’s action before one’s own movement. Such competitive turn-taking was unlike the prosocial turn-taking in humans, who equally often initiated switches to and from their preferred option. Thus, the dynamic coordination is not restricted to humans but can occur on the background of different social attitudes and cognitive capacities in rhesus monkeys. Overall, our results illustrate how action visibility promotes the emergence and maintenance of coordination when agents can observe and time their mutual actions.

Abstract Collective movement of social groups requires coordination between individuals. When cohesion is imperative, consensus must be reached, and specific individuals may exert disproportionate influence during decision-making. Animals living in multi-level societies, however, often split into consistent social subunits during travel, which may impact group coordination processes. We studied collective movement in the socially tolerant multi-level society of Guinea baboons (Papio papio). Using 146 group departures and 100 group progressions from 131 Guinea baboons ranging in Senegal’s Niokolo-Koba National Park, we examined individual success at initiating group departures and position within progressions. Two-thirds of attempted departures were initiated by adult males and one third by adult females. Both sexes were equally successful at initiating departures (> 80% of initiations). During group progressions, bachelor males were predominantly found in front, while reproductively active ‘primary’ males and females were observed with similar frequency across the whole group. The pattern of collective movement in Guinea baboons was more similar to those described for baboons living in uni-level societies than to hamadryas baboons, the only other multi-level baboon species, where males initiate and decide almost all group departures. Social organization alone therefore does not determine which category of individuals influence group coordination.

Social comparisons are a fundamental feature of human thinking and affect self-evaluations and task performance. Little is known about the evolutionary origins of social comparison processes, however. Previous studies that investigated performance-based social comparisons in nonhuman primates yielded mixed results. We report three experiments that aimed (a) to explore how the task type may contribute to performance in monkeys, and (b) how a competitive set-up affects monkeys compared to humans. In a co-action touchscreen task, monkeys were neither influenced by nor interested in the performance of the partner. This may indicate that the experimental set-up was not sufficiently relevant to trigger social comparisons. In a novel co-action foraging task, monkeys increased their feeding speed in competitive and co-active conditions, but not in relation to the degree of competition. In an analogue of the foraging task, human participants were affected by partner performance and experimental context, indicating that the task is suitable to elicit social comparisons in humans. Our studies indicate that specifics of task and experimental setting are relevant to draw the monkeys’ attention to a co-actor and that, in line with previous research, a competitive element was crucial. We highlight the need to explore what constitutes “relevant” social comparison situations for monkeys as well as nonhuman animals in general, and point out factors that we think are crucial in this respect (e.g., task type, physical closeness, and the species’ ecology). We discuss that early forms of social comparisons evolved in purely competitive environments with increasing social tolerance and cooperative motivations allowing for more fine-grained processing of social information. Competition driven effects on task performance might constitute the foundation for the more elaborate social comparison processes found in humans, which may involve context-dependent information processing and metacognitive monitoring.

Visual bias in social cognition studies is often interpreted to indicate preference, yet it is difficult to elucidate whether this translates to social preference. Moreover, visual bias is often framed in terms of surprise or recognition. It is thus important to examine whether an interpretation of preference is warranted in looking time studies. Here, using touchscreen training, we examined (1) looking time to non-social images in an image viewing task, and (2) preference of non-social images in a paired choice task, in captive long-tailed macaques, Macaca fascicularis. In a touchscreen test phase, we examined (3) looking time to social images in a viewing task, and (4) preference of social images in a paired choice task. Finally, we examined (5) looking time to social images in a non-test environment. For social content, the monkeys did not exhibit clear preferences for any category (conspecific/heterospecific, in-group/outgroup, kin/non-kin, young/old) in the explicit choice paradigm, nor did they differentiate between images in the viewing tasks, thus hampering our interpretation of the data. Post-hoc analysis of the training data however revealed a visual bias towards images of food and objects over landscapes in the viewing task. Similarly, across choice-task training sessions, food and object images were chosen more frequently than landscapes. This suggests that the monkeys’ gaze may indeed indicate preference, but this only became apparent for non-social stimuli. Why these monkeys had no biases in the social domain remains enigmatic. To better answer questions about attention to social stimuli, we encourage future research to examine behavioral measures alongside looking time.

Abstract Innate immunity triggers responsible for viral control or hyperinflammation in COVID‐19 are largely unknown. Here we show that the SARS‐CoV‐2 spike protein (S‐protein) primes inflammasome formation and release of mature interleukin‐1β (IL‐1β) in macrophages derived from COVID‐19 patients but not in macrophages from healthy SARS‐CoV‐2 naïve individuals. Furthermore, longitudinal analyses reveal robust S‐protein‐driven inflammasome activation in macrophages isolated from convalescent COVID‐19 patients, which correlates with distinct epigenetic and gene expression signatures suggesting innate immune memory after recovery from COVID‐19. Importantly, we show that S‐protein‐driven IL‐1β secretion from patient‐derived macrophages requires non‐specific monocyte pre‐activation in vivo to trigger NLRP3‐inflammasome signaling. Our findings reveal that SARS‐CoV‐2 infection causes profound and long‐lived reprogramming of macrophages resulting in augmented immunogenicity of the SARS‐CoV‐2 S‐protein, a major vaccine antigen and potent driver of adaptive and innate immune signaling.

Echinococcus multilocularis is endemic in Germany. However, alveolar echinococcosis is a rare disease. Most commonly the parasite affects the liver, behaving like a malignant tumour. Bones are affected in less than 2% of cases. We report a case of vertebral osteomyelitis accompanied by recurrent cutaneous fistula formation.

Abstract Microsatellite genotyping is an important genetic method for a number of research questions in biology. Given that the traditional fragment length analysis using polyacrylamide gel or capillary electrophoresis has several drawbacks, microsatellite genotyping‐by‐sequencing (GBS) has arisen as a promising alternative. Although GBS mitigates many of the problems of fragment length analysis, issues with allelic dropout and null alleles often remain due to mismatches in primer binding sites and unnecessarily long PCR products. This is also true for GBS in catarrhine primates where cross‐species amplification of loci (often human derived) is common. We therefore redesigned primers for 45 microsatellite loci based on 17 available catarrhine reference genomes. Next, we tested them in singleplex and different multiplex settings in a panel of species representing all major lineages of Catarrhini and further validated them in wild Guinea baboons (Papio papio) using fecal samples. The final panel of 42 microsatellite loci can efficiently be amplified with primers distributed into three amplification pools. With our microsatellite panel, we provide a tool to universally genotype catarrhine primates via GBS from different sample sources in a cost‐ and time‐efficient way, with higher resolution, and comparability among laboratories and species.

Biosphere reserves (BR) balance biodiversity protection and sustainable use through different management restrictions in three zones: core areas, buffer zones, and transition areas. Information about the links between zoning and ecosystem services (ES) is lacking, particularly in terms of the relative roles of natural contributions (ecosystem properties and functions) and anthropogenic contributions (human inputs such as technology and infrastructure) in coproducing ES. This study aimed to: (1) analyse how coproduction of four ES (crop production, grazing, timber production, recreation) differs across the three zones of BRs; and (2) understand which predictors (zoning, natural and anthropogenic contributions, other environmental characteristics) best explain ES provision within BRs. To do this, we collected spatial data on 137 terrestrial BRs in the European Union and on 16 indicators of ES coproduction. We used non-parametric pairwise Wilcoxon rank sum tests to calculate differences in indicators between zones. We used model selection and multiple linear regression to identify predictors of ES provision patterns. Anthropogenic contributions showed most differences between zones, with contributions generally increasing from buffer zones to transition areas. Natural contributions did not, on average, differ between zones, however, for recreation and crop production they decreased from buffer zones to transition areas. ES provision differed between zones only for crop production and grazing, which increased from buffer zones to transition areas. Regression analysis showed that natural contributions are the best predictors of ES provision for all four services. Our results indicate that zoning of BRs has implications for ES coproduction.

Scientific disciplines face concerns about replicability and statistical inference, and these concerns are also relevant in animal cognition research. This paper presents a first attempt to assess how researchers make and publish claims about animal physical cognition, and the statistical inferences they use to support them. We surveyed 116 published experiments from 63 papers on physical cognition, covering 43 different species. The most common tasks in our sample were trap-tube tasks (14 papers), other tool use tasks (13 papers), means-end understanding and string-pulling tasks (11 papers), object choice and object permanence tasks (9 papers) and access tasks (5 papers). This sample is not representative of the full scope of physical cognition research; however, it does provide data on the types of statistical design and publication decisions researchers have adopted. Across the 116 experiments, the median sample size was 7. Depending on the definitions we used, we estimated that between 44% and 59% of our sample of papers made positive claims about animals’ physical cognitive abilities, between 24% and 46% made inconclusive claims, and between 10% and 17% made negative claims. Several failures of animals to pass physical cognition tasks were reported. Although our measures had low inter-observer reliability, these findings show that negative results can and have been published in the field. However, publication bias is still present, and consistent with this, we observed a drop in the frequency of p-values above .05. This suggests that some non-significant results have not been published. More promisingly, we found that researchers are likely making many correct statistical inferences at the individual-level. The strength of evidence of statistical effects at the group-level was weaker, and its p-value distribution was consistent with some effect sizes being overestimated. Studies such as ours can form part of a wider investigation into statistical reliability in comparative cognition. However, future work should focus on developing the validity and reliability of the measurements they use, and we offer some starting points.

Against the background of the seminal papers on the vervet monkey alarm call system by Seyfarth, Cheney and Marler (1980a. 1980b), I provide an overview of context specificity in calling and call comprehension learning in the genus Chlorocebus, and to which degree these findings inform the reconstruction of the evolution of speech and language. The alarm calls of vervet monkeys (Chlorocebus pygerythrus) and West African green monkeys (Chlorocebus sabaeus) are astoundingly similar in terms of the structure of calls given in response to their main predator types. Green monkeys also spontaneously produced calls that sounded like ‘eagle alarms’ in response to a novel aerial threat (a drone), supporting the view that both the link between the predator category and the call, as well as the call structure itself are largely hard-wired. In contrast, learning shapes the responses to sounds. As recent field experiments showed, monkeys are able to rapidly attach meaning to a novel sound, and they are also able to factor in contextual information when choosing their responses. These findings corroborate the view of a fundamental dichotomy in the degree of flexibility in terms of the production of calls with a specific structure vs. the comprehension of calls and suggest that the emergence of auditory learning abilities preceded the evolution of flexible vocal production. In summary, nonhuman primate communication shares fewer similarities with human speech than many scholars may have hoped for, but we would never have learned so much about primate vocal communication were it not for the breakthrough study by Seyfarth, Cheney, and Marler.

The number of publications applying nonlinear mixed‐effect (NLME) modeling has increased yearly since its first appearance in 1979. Here, we evaluated articles that have used NLME modeling, were published in journals that attract a broader audience, and we discussed the standard of presentation of these to stimulate target audience‐specific improvements for increased impact in the future.

Abstract Background Since people with advanced dementia are usually not able to make complex decisions, it is usually the family caregivers, as proxies, who have to decide on treatments and their termination. However, these decisions are difficult for the caregivers to make, as they are often inadequately informed and cannot properly assess the consequences; moreover, they are concerned about harming the sick person. We aimed to first develop an informative booklet about palliative care issues for caregivers of people with advanced dementia. Secondly, we aimed to investigate a change in family caregivers’ knowledge regarding palliative care issues and caregivers’ involvement in medical and care decisions before and after studying this booklet. Methods A first version of the booklet was drafted by an experienced psychiatrist and palliative care specialist based on existing booklets and guidelines; necessary cultural adaptions were taken into consideration. A nominal group process was conducted to develop the informative guide. In order to investigate the acceptance of the booklet and the possibility to implement it, 38 patient-caregiver dyads were recruited, and caregivers were interviewed both before receiving the booklet and after 3 months of receiving the booklet. Results Experts from various disciplines collaborated on a German booklet for family caregivers of people with advanced dementia as an information aid regarding issues of palliative care. The subsequent test showed that all caregivers had experienced a personal benefit from the booklet. Caregivers had a significant gain of knowledge after provision of the booklet. A large proportion of caregivers who had not previously considered and/or discussed medical topics reported that they had done so within 3 months after obtaining the booklet, or planned to do so in the near future. Conclusions The caregivers valued the comprehensible, concise and well-structured information guide on palliative care issues in advanced dementia. They agreed it increases knowledge and prompts decision making and therefore should be developed in many languages and disseminated among family caregivers of people with dementia. Regulation of cellular metabolism is now recognized as a crucial mechanism for the activation of innate and adaptive immune cells upon diverse extracellular stimuli. Macrophages, for instance, increase glycolysis upon stimulation with pathogen-associated molecular patterns (PAMPs). Conceivably, pathogens also counteract these metabolic changes for their own survival in the host. Despite this dynamic interplay in host-pathogen interactions, the role of immunometabolism in the context of intracellular bacterial infections is still unclear. Here, employing unbiased metabolomic and transcriptomic approaches, we investigated the role of metabolic adaptations of macrophages upon Salmonella enterica serovar Typhimurium (S. Typhimurium) infections. Importantly, our results suggest that S. Typhimurium abrogates glycolysis and its modulators such as insulin-signaling to impair macrophage defense. Mechanistically, glycolysis facilitates glycolytic enzyme aldolase A mediated v-ATPase assembly and the acidification of phagosomes which is critical for lysosomal degradation. Thus, impairment in the glycolytic machinery eventually leads to decreased bacterial clearance and antigen presentation in murine macrophages (BMDM). Collectively, our results highlight a vital molecular link between metabolic adaptation and phagosome maturation in macrophages, which is targeted by S. Typhimurium to evade cell-autonomous defense.

Background and Objectives: Data on suicidal ideation, behavior and the risk factors in patients with dementia is scarce. To evaluate the prevalence of death wishes, suicidal ideation, and suicidal behavior of young (YOD) and late onset dementia (LOD) and to identify risk factors for suicidal ideation and behavior.Methods: We interviewed 157 family caregivers of patients with advanced dementia using questions from the Columbia-Suicide Severity Rating Scale to gather information about suicidal ideation and behavior before the onset of symptoms of dementia, after the onset of dementia and within 30 days prior to the interview. At the time of the interview, we also assessed disease severity, cognitive function, and other psychological, behavioral and physical symptoms of the patients as well as the caregivers' psychological well-being.Results: Forty four (28%) of the patients expressed suicidal ideation or behavior at some time after the onset of symptoms, and 14 (9%) of these within the month prior to the assessment. Two patients had attempted suicide after the onset of dementia. There were no statistically significant differences between patients with and without suicidal ideations or behavior with regards to demographics or age at onset of dementia. In patients with advanced dementia, Alzheimer's disease (rather than frontotemporal lobar degeneration), better cognitive function, more severe psychological, behavioral, and physical symptoms, and a reduced quality of life were associated with the expression of suicidal ideation.Conclusions: According to caregivers' reports, majority of patients with dementia did not express suicidal ideation or show suicidal behavior. Patients who expressed suicidal ideation during early stages of dementia often stopped expressing them in advanced stages. It remains unclear if this was due to reduced communication abilities, a reduction of disease awareness, and/ or an adjustment to their situation.

Most statistical problems encountered throughout life require the ability to quantify probabilities based on proportions. Recent findings on the early ontogeny of this ability have been mixed: For example, when presented with jars containing preferred and less preferred items, 12-month-olds, but not 3- and 4-years-olds, seem to rely on the proportions of objects in the jars to predict the content of samples randomly drawn out of them. Given these contrasting findings, it remains unclear what the probabilistic reasoning abilities of young children are and how they develop. In our study, we addressed this question and tested, with identical methods across age groups and similar methods to previous studies, whether 12-month-olds and 3- and 4-years-olds rely on proportions of objects to estimate probabilities of random sampling events. Results revealed that neither infants nor preschoolers do. While preschoolers' performance is in line with previous findings, infants' performance is difficult to interpret given their failure in a control condition in which the outcomes happened with certainty rather than a graded probability. More systematic studies are needed to explain why infants succeeded in a previous study but failed in our study.

Following the expanding use and applications of virtual reality in everyday life, realistic virtual stimuli are of increasing interest in cognitive studies. They allow for control of features such as gaze, expression, appearance, and movement, which may help to overcome limitations of using photographs or video recordings to study social responses. In using virtual stimuli however, one must be careful to avoid the uncanny valley effect, where realistic stimuli can be perceived as eerie, and induce an aversion response. At the same time, it is important to establish whether responses to virtual stimuli mirror responses to depictions of a real conspecific. In the current study, we describe the development of a new virtual monkey head with realistic facial features for experiments with nonhuman primates, the “Primatar.” As a first step toward validation, we assessed how monkeys respond to facial images of a prototype of this Primatar compared to images of real monkeys (RMs), and an unrealistic model. We also compared gaze responses between original images and scrambled as well as obfuscated versions of these images. We measured looking time to images in six freely moving long-tailed macaques (Macaca fascicularis) and gaze exploration behavior in three rhesus macaques (Macaca mulatta). Both groups showed more signs of overt attention to original images than scrambled or obfuscated images. In addition, we found no evidence for an uncanny valley effect; since for both groups, looking times did not differ between real, realistic, or unrealistic images. These results provide important data for further development of our Primatar for use in social cognition studies and more generally for cognitive research with virtual stimuli in nonhuman primates. Future research on the absence of an uncanny valley effect in macaques is needed, to elucidate the roots of this mechanism in humans.

Splicing is a vital cellular process that modulates important aspects of animal physiology, yet roles in regulating innate immunity are relatively unexplored. From genetic screens in C. elegans, we identified splicing factor RNP-6/PUF60 whose activity suppresses immunity, but promotes longevity, suggesting a tradeoff between these processes. Bacterial pathogen exposure affects gene expression and splicing in a rnp-6 dependent manner, and rnp-6 gain and loss-of-function activities reveal an active role in immune regulation. Another longevity promoting splicing factor, SFA-1, similarly exerts an immuno-suppressive effect, working downstream or parallel to RNP-6. RNP-6 acts through TIR-1/PMK-1/MAPK signaling to modulate immunity. The mammalian homolog, PUF60, also displays anti-inflammatory properties, and its levels swiftly decrease after bacterial infection in mammalian cells, implying a role in the host response. Altogether our findings demonstrate an evolutionarily conserved modulation of immunity by specific components of the splicing machinery.

Induced neurons, but not induced pluripotent stem cell (iPSC)-derived neurons, preserve age-related traits. Here, the authors demonstrate that blood-derived induced neural stem cells (iNSCs), despite lacking a pluripotency transit, lose age-related signatures.

Baboons, members of the genus Papio, comprise six closely related species distributed throughout sub-Saharan Africa and southwest Arabia. The species exhibit more ecological flexibility and a wider range of social systems than many other primates. This article summarizes our current knowledge of the natural history of baboons and highlights directions for future research. We suggest that baboons can serve as a valuable model for complex evolutionary processes, such as speciation and hybridization. The evolution of baboons has been heavily shaped by climatic changes and population expansion and fragmentation in the African savanna environment, similar to the processes that acted during human evolution. With accumulating long-term data, and new data from previously understudied species, baboons are ideally suited for investigating the links between sociality, health, longevity and reproductive success. To achieve these aims, we propose a closer integration of studies at the proximate level, including functional genomics, with behavioral and ecological studies.

Inferring the evolutionary history of cognitive abilities requires large and diverse samples. However, such samples are often beyond the reach of individual researchers or institutions, and studies are often limited to small numbers of species. Consequently, methodological and site-specific-differences across studies can limit comparisons between species. Here we introduce the ManyPrimates project, which addresses these challenges by providing a large-scale collaborative framework for comparative studies in primate cognition. To demonstrate the viability of the project we conducted a case study of short-term memory. In this initial study, we were able to include 176 individuals from 12 primate species housed at 11 sites across Africa, Asia, North America and Europe. All subjects were tested in a delayed-response task using consistent methodology across sites. Individuals could access food rewards by remembering the position of the hidden reward after a 0, 15, or 30-second delay. Overall, individuals performed better with shorter delays, as predicted by previous studies. Phylogenetic analysis revealed a strong phylogenetic signal for short-term memory. Although, with only 12 species, the validity of this analysis is limited, our initial results demonstrate the feasibility of a large, collaborative open-science project. We present the ManyPrimates project as an exciting opportunity to address open questions in primate cognition and behaviour with large, diverse datasets.

BackgroundHypercholesterolaemia is common in patients after cardiac transplantation. Monoclonal antibodies that inhibit proprotein convertase subtilisin-kexin type 9 (PCSK9) reduce low-density lipoprotein (LDL) cholesterol levels and subsequently the risk of cardiovascular events in patients with dyslipidaemia. There are no published data on the effect of this medication class on cholesterol levels in patients after cardiac transplantation.MethodsIn this retrospective study we investigated patients who were treated with PCSK9 inhibitors either because of intolerance of statins or residual hypercholesterolaemia with evidence of cardiac allograft vasculopathy. We compared the data of patients prior to the start with these medications with their most recent dataset.ResultsTen patients (nine men; mean age 58±6 years) underwent cardiac transplantation 8.3±4.5 (range 3-15) years ago. The treatment duration of Evolocumab or Alirocumab was on average 296±125 days and lead to a reduction of total Cholesterol (281±52 mg/dl to 197±36 mg/dl; p = 0.002) and LDL Cholesterol (170±22 mg/dl to 101±39 mg/dl; p = 0.001). No significant effects on HDL Cholesterol, BNP, Creatin Kinase or hepatic enzymes were noticed. There were no unplanned hospitalisations, episodes of rejections, change of ejection fraction or opportunistic infections. Both patients on Alirocumab developed liver pathologies: One patient died of hepatocellular carcinoma and the other developed hepatitis E.ConclusionsOur study demonstrates that the PCSK9 inhibitors Evolocumab and Alirocumab lead to a significant reduction of LDL Cholesterol in heart transplantation recipients. No effect on cardiac function or episodes of rejections were noticed. Larger and long-term studies are needed to establish safety and efficacy of PCSK9 inhibitors after cardiac transplantation.

The prevalence of bacterial pathogens being resistant to antibiotic treatment is increasing worldwide, leading to a severe global health challenge. Simultaneously, the development and approval of new antibiotics stagnated in the past decades, leading to an urgent need for novel approaches to avoid the spread of untreatable bacterial infections in the future. We developed a highly comprehensive screening platform based on quantification of pathogen driven host-cell death to detect new anti-virulence drugs targeting Pseudomonas aeruginosa (Pa) and Salmonella enterica serovar Typhimurium (ST), both known for their emerging antibiotic resistance. By screening over 10,000 small molecules we could identify several substances showing promising effects on Pa and ST pathogenicity in our in vitro infection model. Importantly, we could detect compounds potently inhibiting bacteria induced killing of host cells and one novel comipound with impact on the function of the type 3 secretion system (T3SS) of ST. Thus, we provide proof of concept data of rapid and feasible medium- to high-throughput drug screening assays targeting virulence mechanisms of two major Gram-negative pathogens.

Emotional expressions provide strong signals in social interactions and can function as emotion inducers in a perceiver. Although speech provides one of the most important channels for human communication, its physiological correlates, such as activations of the autonomous nervous system (ANS) while listening to spoken utterances, have received far less attention than in other domains of emotion processing. Our study aimed at filling this gap by investigating autonomic activation in response to spoken utterances that were embedded into larger semantic contexts. Emotional salience was manipulated by providing information on alleged speaker similarity. We compared these autonomic responses to activations triggered by affective sounds, such as exploding bombs, and applause. These sounds had been rated and validated as being either positive, negative, or neutral. As physiological markers of ANS activity, we recorded skin conductance responses (SCRs) and changes of pupil size while participants classified both prosodic and sound stimuli according to their hedonic valence. As expected, affective sounds elicited increased arousal in the receiver, as reflected in increased SCR and pupil size. In contrast, SCRs to angry and joyful prosodic expressions did not differ from responses to neutral ones. Pupil size, however, was modulated by affective prosodic utterances, with increased dilations for angry and joyful compared to neutral prosody, although the similarity manipulation had no effect. These results indicate that cues provided by emotional prosody in spoken semantically neutral utterances might be too subtle to trigger SCR, although variation in pupil size indicated the salience of stimulus variation. Our findings further demonstrate a functional dissociation between pupil dilation and skin conductance that presumably origins from their differential innervation.

Different hypotheses have been put forward to explain the interaction between size perception and spatial position. To explore the evolutionary roots of these phenomena, we tested long-tailed macaques' performance in a two-choice discrimination task on a touchscreen and contrasted two hypotheses. First, a hierarchy association in which large objects are associated with top positions, due to a link between power, dominance and importance with top position. Second, a naive Aristotelian association in which large objects are associated with bottom positions, due to the experience that larger objects are heavier and thus more likely to be found at the bottom. Irrespective of training regime (positively reinforcing the small (Touch-Small) or large (Touch-Large) stimulus), the monkeys had a bias to touch the bottom compared to the top location. Individuals in the Touch-Small group took significantly longer to acquire the task, but subsequently made fewer mistakes. When presented with two stimuli of equal medium size, the Touch-Large group had a clear bias to touch the lower stimulus, while the Touch-Small group touched both locations at equal rates. Our findings point to an innate bias towards larger stimuli and a natural preference for the lower position, while the extent of interaction between size and position depends on executive control requirements of a task.

Human infants, apes and capuchin monkeys engage in intuitive statistics: they generate predictions from populations of objects to samples based on proportional information. This suggests that statistical reasoning might depend on some core knowledge that humans share with other primate species. To aid the reconstruction of the evolution of this capacity, we investigated whether intuitive statistical reasoning is also present in a species of Old World monkey. In a series of four experiments, 11 long-tailed macaques were offered different pairs of populations containing varying proportions of preferred versus neutral food items. One population always contained a higher proportion of preferred items than the other. An experimenter simultaneously drew one item out of each population, hid them in her fists and presented them to the monkeys to choose. Although some individuals performed well across most experiments, our results imply that long-tailed macaques as a group did not make statistical inferences from populations of food items to samples but rather relied on heuristics. These findings suggest that there may have been convergent evolution of this ability in New World monkeys and apes (including humans).

During intracellular infections, autophagy significantly contributes to the elimination of pathogens, regulation of pro-inflammatory signaling, secretion of immune mediators and in coordinating the adaptive immune system. Intracellular pathogens such as S. Typhimurium have evolved mechanisms to circumvent autophagy. However, the regulatory mechanisms targeted by S. Typhimurium to modulate autophagy have not been fully resolved. Here we report that cytosolic energy loss during S. Typhimurium infection triggers transient activation of AMPK, an important checkpoint of mTOR activity and autophagy. The activation of AMPK is regulated by LKB1 in a cytosolic complex containing Sirt1 and LKB1, where Sirt1 is required for deacetylation and subsequent activation of LKB1. S. Typhimurium infection targets Sirt1, LKB1 and AMPK to lysosomes for rapid degradation resulting in the disruption of the AMPK-mediated regulation of mTOR and autophagy. The degradation of cytosolic Sirt1/LKB1/AMPK complex was not observed with two mutant strains of S. Typhimurium, ΔssrB and ΔssaV, both compromising the pathogenicity island 2 (SPI2). The results highlight virulence factor-dependent degradation of host cell proteins as a previously unrecognized strategy of S. Typhimurium to evade autophagy.

To manoeuvre in complex societies, it is beneficial to acquire knowledge about the social relationships existing among group mates, so as to better predict their behaviour. Although such knowledge has been firmly established in a variety of animal taxa, how animals acquire such knowledge, as well as its functional significance, remains poorly understood. In order to understand how primates acquire and use their social knowledge, we studied kin-biased redirected aggression in Japanese macaques (Macaca fuscata) relying on a large database of over 15 000 aggressive episodes. Confirming previous research, macaques redirected aggression preferentially to the kin of their aggressor. An analysis that controlled for the rate of affiliation between aggressors and targets of redirection showed that macaques identified the relatives of group mates on the basis of the frequency of their ongoing associations. By contrast, having observed group mates interact with their mother as infants did not increase the monkeys' success in correctly identifying kin relationships among third parties. Inter-individual variation in the successful identification of the kin of aggressors and in redirecting aggression accordingly translated into differences in the amount of aggression received, highlighting a selective advantage for those individuals that were better able to acquire and use social knowledge.

In social animals, kin relations strongly shape the social structure of a group. In female-bonded species, maternal relatedness is likely to be mediated via familiarity, but evidence is accumulating that non-human primates are able to recognize kin that they are not familiar with and adjust their behavior accordingly. In playback experiments, female rhesus macaques showed increased interest in 'coo' calls produced by unfamiliar paternal half-sisters compared to 'coo' calls produced by unfamiliar unrelated females, suggesting that these calls should have some common structural characteristics that facilitate the discrimination of kin from non-kin. Here we analyzed 'coo' calls of 67 adult female rhesus macaques from four groups and seven matrilines living on the island of Cayo Santiago (Puerto Rico). We tested whether the call structure of closely maternal and/or paternal related females, as determined from extensive pedigree data, differed from the call structure of unrelated females, while controlling for familiarity (i.e., group-matrilineal membership and age difference) of subjects. In contrast to our expectation, kinship did not predict similarities in 'coo' call structure, whereas 'coo' structure was more similar when produced by females of similar age as well as by females with higher familiarity, suggesting that experience is more decisive than genetic background. The high number of individuals in the analysis and the high accuracy of the assignment of calls to individuals render a lack of power as an unlikely explanation. Thus, based on the results of this study, kin recognition in rhesus monkeys does neither appear to be based on the assessment of self-similarity, nor on the comparison among related subjects (i.e., acoustic phenotype matching), but appears to be mediated by different or multiple cues. Furthermore, the results support the notion that frequent social interactions result in increasing acoustic similarity within largely innate call types ('vocal accommodation').

A 50-year-old man with no previous history of cardiovascular disease or risk factors was admitted for syncope and orthopnea. Importantly, he underwent recent chemotherapy with 5-fluorouracil (5-FU) until 1 day before his acute presentation. In the emergency room, patient developed asystole and was successfully resuscitated for 2 min. At coronary angiography, no signs of coronary artery disease were detectable, but transthoracic echocardiography showed a severely decreased left-ventricular systolic function. Due to the progressive cardiogenic shock, an extracorporeal membrane oxygenation (ECMO) support was used as bridge-to-recovery and to avoid the use of sympathomimetics with their known disadvantages. On ECMO support, hemodynamic stabilization was evident and medical heart failure treatment was commenced. Left-ventricular function recovered to normal values within a short period of time. Cardiac complications after chemotherapy with 5-FU are not rare and should be taken into consideration even in acute heart failure with cardiogenic shock. ECMO as the most potent form of acute cardiorespiratory support enables complete relief of cardiac workload and therefore recovery of cardiac function.

To understand the proximate and ultimate causes that shape acoustic communication in animals, objective characterizations of the vocal repertoire of a given species are critical, as they provide the foundation for comparative analyses among individuals, populations and taxa. Progress in this field has been hampered by a lack of standard in methodology, however. One problem is that researchers may settle on different variables to characterize the calls, which may impact on the classification of calls. More important, there is no agreement how to best characterize the overall structure of the repertoire in terms of the amount of gradation within and between call types. Here, we address these challenges by examining 912 calls recorded from wild chacma baboons (Papio ursinus). We extracted 118 acoustic variables from spectrograms, from which we constructed different sets of acoustic features, containing 9, 38, and 118 variables; as well 19 factors derived from principal component analysis. We compared and validated the resulting classifications of k-means and hierarchical clustering. Datasets with a higher number of acoustic features lead to better clustering results than datasets with only a few features. The use of factors in the cluster analysis resulted in an extremely poor resolution of emerging call types. Another important finding is that none of the applied clustering methods gave strong support to a specific cluster solution. Instead, the cluster analysis revealed that within distinct call types, subtypes may exist. Because hard clustering methods are not well suited to capture such gradation within call types, we applied a fuzzy clustering algorithm. We found that this algorithm provides a detailed and quantitative description of the gradation within and between chacma baboon call types. In conclusion, we suggest that fuzzy clustering should be used in future studies to analyze the graded structure of vocal repertoires. Moreover, the use of factor analyses to reduce the number of acoustic variables should be discouraged.

The bacterium Treponema pallidum is known to cause syphilis (ssp. pallidum), yaws (ssp. pertenue), and endemic syphilis (ssp. endemicum) in humans. Nonhuman primates have also been reported to be infected with the bacterium with equally versatile clinical manifestations, from severe skin ulcerations to asymptomatic. At present all simian strains are closely related to human yaws-causing strains, an important consideration for yaws eradication. We tested clinically healthy Guinea baboons (Papio papio) at Parc National Niokolo Koba in south eastern Senegal for the presence of anti-T. pallidum antibodies. Since T. pallidum infection in this species was identified 50 years ago, and there has been no attempt to treat non-human primates for infection, it was hypothesized that a large number of West African baboons are still infected with simian strains of the yaws-bacterium. All animals were without clinical signs of treponematoses, but 18 of 20 (90%) baboons tested positive for antibodies against T. pallidum based on treponemal tests. Yet, Guinea baboons seem to develop no clinical symptoms, though it must be assumed that infection is chronic or comparable to the latent stage in human yaws infection. The non-active character is supported by the low anti-T. pallidum serum titers in Guinea baboons (median = 1:2,560) versus serum titers that are found in genital-ulcerated olive baboons with active infection in Tanzania (range of medians among the groups of initial, moderate, and severe infected animals = 1:15,360 to 1:2.097e+7). Our findings provide evidence for simian infection with T. pallidum in wild Senegalese baboons. Potentially, Guinea baboons in West Africa serve as a natural reservoir for human infection, as the West African simian strain has been shown to cause sustainable yaws infection when inoculated into humans. The present study pinpoints an area where further research is needed to support the currently on-going second WHO led yaws eradication campaign with its goal to eradicate yaws by 2020.

The metabolic syndrome is a worldwide problem mainly caused by obesity. FTO was found to be a obesity-risk gene in humans and FTO deficiency in mice led to reduction in adipose tissue. Thus, FTO is an important factor for the development of obesity. Leptin-deficient mice are a well characterized model for analysing the metabolic syndrome. To determine the relevance of FTO for the development of the metabolic syndrome we analysed different parameters in combined homozygous deficient mice (Lep(ob/ob);Fto(-/-)). Lep(ob/ob);Fto(-/-) mice showed an improvement in analysed hallmarks of the metabolic syndrome in comparison to leptin-deficient mice wild type or heterozygous for Fto. Lep(ob/ob);Fto(-/-) mice did not develop hyperglycaemia and showed an improved glucose tolerance. Furthermore, extension of beta-cell mass was prevented in Lep(ob/ob);Fto(-/-)mice and accumulation of ectopic fat in the liver was reduced. In conclusion this study demonstrates that FTO deficiency has a protective effect not only on the development of obesity but also on the metabolic syndrome. Thus, FTO plays an important role in the development of metabolic disorders and is an interesting target for therapeutic agents.

Methodological variations in experimental conditions can strongly influence animals' performances in cognitive tests. Specifically, the procedure of the so-called object-choice task has been controversially discussed; here, a human experimenter indicates the location of hidden food by pointing or gazing at one of two or more containers. Whereas dogs usually succeed, results for nonhuman primates are ambiguous. In the standard version of the task the majority of subjects do not respond appropriately to human pointing. However, modifying the task setup, such as placing the containers further apart, seems to improve subjects' performances, suggesting that cue salience may be an important variable. Here we investigated whether the visibility of the experimenter inhibits long-tailed macaques' (Macaca fascicularis) usage of the pointing cue. In our baseline condition, with the experimenter fully visible, the monkeys chose the correct container in 61% of the trials. The performance increased significantly, however, when the experimenter was hidden behind a curtain and only the arm of the experimenter, a doll's arm, or a stick was visible. Furthermore, the monkeys performed significantly better when the tip of the pointing finger or device was close to the target compared to the more distant condition. Intriguingly, after these experiments the monkeys' performance was also significantly improved in the baseline condition (70%). Apparently, the monkeys were first distracted by the presence of the experimenter, but then learned to use the cue. These findings highlight the importance of the test conditions, and question some of the assumptions about species-specific differences in the object-choice task.
