The dynamic of rangelands results from complex interactions between vegetation, soil, climate, and human activity. In arid and semiarid areas, rainfall coefficients of variability are over 30%. This scenario makes rangeland's condition challenging to monitor, and degradation assessment should be carefully considered to study grazing pressures. In the present work, we study the interaction of vegetation and soil moisture in arid rangelands, using vegetation and soil moisture indexes. We aim to study the feasibility of using water soil moisture (soil drought) as a warning index for vegetation drought. An arid agricultural region in the southeast of Spain, in the province of Almeria (Los Velez), was selected for this study. MODIS images, with 250 and 500 m spatial resolution, from 2002 to 2019, were acquired to calculate the anomaly (Z-score) for the Vegetation Condition Index (ZVCI) and the Water Condition Index (ZWCI). ZVCI was calculated using the Normalised Difference Vegetation Index (NDVI). Soil moisture component (W) was obtained using the Optical Trapezoid Model (OPTRAM). The probability of coincidence of their negative anomalies between ZVCI and ZWCI, with lags between them, was calculated. The results show that for specific seasons, the anomaly of the water content index had a strong probability of informing in advance where the negative anomaly of VCI will decrease. Soil water content and vegetation indices show more similar dynamics in the months with lower temperatures (from autumn to spring). In these months, given the low temperatures, precipitation leads the vegetation growth. In the following months, water availability depends on evapotranspiration and vegetation type as the temperature rises and the precipitation falls. The stronger relationship between precipitation and vegetation from autumn to the beginning of spring is reflected in the feasibility of ZWCI to aid the prediction of vegetation index anomalies. During these months, using ZWCI and ZVCI as warning indices are possible for two Spanish semiarid rangeland areas: Los Velez (Almeria) and Bajo Aragon (Teruel). Particularly, November to January showed an average increase of 20-30% in the predictability of vegetation index anomalies. Water sustainability involves several natural and human spheres conditioning the availability and quality of water resources and life conservation. Regarding water quality, currently emerging pollutants (EPs) are a key topic at global scale since they are difficult to remove by traditional water treatment systems. In this context, this work aims to evaluate experts’ perception of several EPs issues with a special focus on semi-arid Mediterranean areas where EPs are negatively impacting water, environmental, and agricultural systems as, in these areas, effluents from water treatment plants are widely reused in the irrigation of crops, urban gardens, and golf courses or directly discharged on natural streams. Particularly, a detailed survey composed of questions about EPs regulation, risk insight, equipment, social and economic impacts, was performed collecting 437 responses. Main results suggested that EPs existence may pose a significant risk and a destabilizing factor in wastewater reuse, with negative impacts to crop irrigation, being managers the class with more concern followed by scientifics and administratives. New EPs regulations raises uncertainty amongst experts since 29 % considered positive its creation, while 20 % estimated this fact as regular and 14 % as negative. As well, although the combination of technologies to improve EPs removal generated agreement, aspects like the treatment charges at water treatment plants or the price/bill of EPs-free water were features of ambiguity. Within EPs elimination, the three expert groups highlighted that technologies impacts in the sustainability spheres will be positive on public health (87 % of responses), social trust (75 %), and environmental sustainability (76 %). Likewise, 88 % of experts concurred that the future of wastewater reuse relies on a combination of technologies. These findings offer valuable information to water legislators and policymakers to manage water resources, especially in semi-arid areas due to the final use of treated effluents and, therefore, the great implications for agriculture, environment and human health.

Two semiarid agricultural regions were selected in Spain for this study: Los Vélez (Almería) and Bajo Aragón (Teruel). MODIS images, with 250 m and 500 m spatial resolution, from 2002 to 2019, were acquired to calculate the Vegetation Condition Index (VCI) and the Water Condition Index (WCI) based on the Normalised Difference Vegetation Index (NDVI) and soil moisture component (W), respectively. The Optical Trapezoid Model (OPTRAM) estimated this latter W index. From them, the anomaly (Z-score) for each index was calculated, being ZVCI and ZWCI, respectively. The probability of coincidence of their negative anomalies was calculated every 10 days (10-day periods). The results show that for specific months, the ZWCI had a strong probability of informing in advance, where the negative ZVCI will decrease. The cactus moth, Cactoblastis cactorum (Berg) (Lepidoptera: Pyralidae) is native to South America and has been used as a biocontrol agent of Opuntia (Cactaceae) in Australia and South Africa. Its invasion in North America has raised concerns for the native Opuntia in the USA and Mexico. We investigated the reproductive biology and rearing procedures of a host-specific potential biocontrol agent, Apanteles opuntiarum Martínez and Berta (Hymenoptera: Braconidae). Given the gregarious nature of the parasitoid larvae, we studied the morphology of the immature stages and evaluated evidence of polyembryony and superparasitism. We also investigated the effects of host exposure arena and host density on parasitism rates and wasp production. The morphological descriptions provide a basis for comparison with other species. Early larval instars of A. opuntiarum are similar to other microgastrine immature stages. However, the mature larva exhibits placoid sensilla in the epistomal region, a unique character not previously reported. We provide evidence that A. opuntiarum eggs are not polyembryonic; females frequently superparasitize and have an oviposition preference for larvae parasitized 1–2 d previously. Exposing larvae of C. cactorum to wasps while inside the cactus resulted in lower parasitism and fewer offspring from each host than exposing larvae in the arena without the cactus. Parasitism and mortality rates were higher at lower host densities, possibly due to reduced host group defensive behavior. These results suggest that preference for superparasitism, host defensive behavior, and interactions with the cactus may play an important role in per-host wasp production under laboratory conditions.

BackgroundOperational tolerance in liver transplantation (OT-LT), defined as the graft survival with normal function in absence of immunosuppression, has been a field of intense research since the 1980s. Thereafter, tens of clinical trials and hundreds of articles have been published, making it challenging for researchers to assimilate all the information, more so outside of their disciplines. The aim of the present study was to analyze the research in OT-LT through a new web tool (https://tolerance.imib.es).MethodsWe have developed a web resource that allowed the identification of the present trends and potential research avenues in OL-LT, an overview biomedical terms that were most often cited, including which journals published the most articles, and an advanced search engine that exploited all the information in these publications.ResultsA total of 734 studies were analyzed until November 2023, with a mean of 15 articles published per year, a total sum of 3,751 impact factor points and a total of 26,542 citations. The analysis of citations allowed us to establish a ranking of the most prolific countries, authors, journals and institutions, in addition to the most influential publications in OT-LT. Likewise, keyword and co-occurrence analyses answered which themes involving OT-LT are the most popular, whereas cooperation analysis showed that principal authors in OT-LT form a network, although the lack of international cooperation, especially with regard to clinical trials, appears to be one of the main challenges.ConclusionDespite its limitations, our web tool will allow both OT-LT expert and novel researchers to be able to draw a comprehensive picture of the past, present and future of OT-LT research.

Abstract Background Whole-exome sequencing (WES) and whole-genome sequencing (WGS) have become indispensable tools to solve rare Mendelian genetic conditions. Nevertheless, there is still an urgent need for sensitive, fast algorithms to maximise WES/WGS diagnostic yield in rare disease patients. Most tools devoted to this aim take advantage of patient phenotype information for prioritization of genomic data, although are often limited by incomplete gene-phenotype knowledge stored in biomedical databases and a lack of proper benchmarking on real-world patient cohorts. Methods We developed ClinPrior, a novel method for the analysis of WES/WGS data that ranks candidate causal variants based on the patient’s standardized phenotypic features (in Human Phenotype Ontology (HPO) terms). The algorithm propagates the data through an interactome network-based prioritization approach. This algorithm was thoroughly benchmarked using a synthetic patient cohort and was subsequently tested on a heterogeneous prospective, real-world series of 135 families affected by hereditary spastic paraplegia (HSP) and/or cerebellar ataxia (CA). Results ClinPrior successfully identified causative variants achieving a final positive diagnostic yield of 70% in our real-world cohort. This includes 10 novel candidate genes not previously associated with disease, 7 of which were functionally validated within this project. We used the knowledge generated by ClinPrior to create a specific interactome for HSP/CA disorders thus enabling future diagnoses as well as the discovery of novel disease genes. Conclusions ClinPrior is an algorithm that uses standardized phenotype information and interactome data to improve clinical genomic diagnosis. It helps in identifying atypical cases and efficiently predicts novel disease-causing genes. This leads to increasing diagnostic yield, shortening of the diagnostic Odysseys and advancing our understanding of human illnesses.

Abstract Background The soil microbiota has a direct impact on plant development and other metabolic systems, such as the degradation of organic matter and the availability of microelements and metabolites. In the context of agricultural soils, microbial activity is crucial for maintaining soil health and productivity. Thus, the present study aimed to identify, characterize, and quantify the microbial communities of four types of substrates with varying proportions of marine port sediment used for cultivating lemons. By investigating microbial diversity and relative abundance, the work aimed to highlight the importance of soil microbial communities in agriculture when alternative culture media was used. Results The composition and structure of the sampled microbial communities were assessed through the amplification and sequencing of the V3-V4 variable regions of the 16 S rRNA gene The results revealed a diverse microbial community composition in all substrate samples, with a total of 41 phyla, 113 classes, 266 orders, 405 families, 715 genera, and 1513 species identified. Among these, Proteobacteria, Bacteroidota, Planctomycetota, Patescibacteria, Chloroflexi, Actinobacteriota, Acidobacteriota, Verrucomicrobiota, and Gemmatimonadota accounted for over 90% of the bacterial reads, indicating their dominance in the substrates. Conclusions The impact of the substrate origin on the diversity and relative abundace of the microbiota was confirmed. The higher content of beneficial bacterial communities for plant development identified in peat could explain why is considered an ideal agricultural substrate. Development of “beneficial for plants” bacterial communities in alternative agricultural substrates, regardless of the edaphic characteristics, opens the possibility of studying the forced and specific inoculation of these culture media aiming to be agriculturally ideals.

Hybrid scaffolds that are based on PLA and PLA/PMMA with 75/25, 50/50, and 25/75 weight ratios and functionalized with 10 wt.% of bioglass nanoparticles (n-BG) were developed using an electrospinning technique with a chloroform/dimethylformamide mixture in a 9:1 ratio for bone tissue engineering applications. Neat PLA and PLA/PMMA hybrid scaffolds were developed successfully through a (CF/DMF) solvent system, obtaining a random fiber deposition that generated a porous structure with pore interconnectivity. However, with the solvent system used, it was not possible to generate fibers in the case of the neat PMMA sample. With the increase in the amount of PMMA in PLA/PMMA ratios, the fiber diameter of hybrid scaffolds decreases, and the defects (beads) in the fiber structure increase; these beads are associated with a nanoparticle agglomeration, that could be related to a low interaction between n-BG and the polymer matrix. The Young’s modulus of PLA/PMMA/n-BG decreases by 34 and 80%, indicating more flexible behavior compared to neat PLA. The PLA/PMMA/n-BG scaffolds showed a bioactive property related to the presence of hydroxyapatite crystals in the fiber surface after 28 days of immersion in a Simulated Body Fluids solution (SBF). In addition, the hydrolytic degradation process of PLA/PMMA/n-BG, analyzed after 35 days of immersion in a phosphate-buffered saline solution (PBS), was less than that of the pure PLA. The in vitro analysis using an HBOF-1.19 cell line indicated that the PLA/PMMA/n-BG scaffold showed good cell viability and was able to promote cell proliferation after 7 days. On the other hand, the in vivo biocompatibility evaluated via a subdermal model in BALC male mice corroborated the good behavior of the scaffolds in avoiding the generation of a cytotoxic effect and being able to enhance the healing process, suggesting that the materials are suitable for potential applications in tissue engineering.

Efficient boron removal from irrigation waters is crucial for sustainable agriculture, as elevated levels of boron can be toxic to many plants, limiting growth and crop productivity. In this context, the present study investigated the sorption equilibrium of boron using zeolites in two types of aqueous matrices: a synthetic solution containing only boron and natural irrigation waters. Through the application of various isothermal sorption models (Langmuir, Freundlich, Sips, Toth, Jovanovic, Temkin, Dubinin–Radushkevich, and Redlich–Peterson), the efficacy of zeolite for boron removal under controlled and real conditions was evaluated. The results indicated a notable difference in sorption behavior between the two matrices, reflecting the complexity and heterogeneity of interactions in the boron–zeolite system. In the synthetic solution, the Freundlich model provided the best fit (R2 = 0.9917), suggesting heterogeneous and multilayer sorption, while the Sips model showed high efficacy in describing the sorption in both matrices, evidencing its capability to capture the complex nature of the interaction between boron and zeolite under different environmental conditions. However, in natural irrigation waters, the Jovanovic model demonstrated the most accurate fit (R2 = 0.999), highlighting the importance of physical interactions in boron sorption. These findings underscore the significant influence of the water matrix on the efficacy of zeolite as a boron removal agent, emphasizing the need to consider the specific composition of irrigation water in the design of removal treatments. Additionally, the results stress the importance of selecting the appropriate isothermal model to predict boron sorption behavior, which is crucial for developing effective and sustainable treatment strategies. This study provides a basis for optimizing boron removal in various agricultural and industrial applications, contributing to the design of more efficient and specific water treatment processes.

The global cultivation of blood oranges is experiencing an increase due to their remarkable nutritional properties. Blood orange by-products, especially the peel, have a high concentration of bioactive compounds with exceptional antioxidant potential, making them an ideal choice for incorporation into various food products. This study aimed to determine the morphological parameters and primary and secondary metabolite content of peel of eight blood orange varieties using 1H NMR and HPLC-ESI-DAD-MSn. “Tarocco Meli” had the highest weight (367.83 g), caliber (94.13 mm and 88.87 mm), peel thickness (6.73 mm), and peel weight (155.0 g). “Tarocco Rosso”, “Sanguinelli”, and “Tarocco Gallo” had the highest levels of total amino acids (25.57 g kg−1 DW), total organic acids (29.99 g kg−1 DW), and total sugars (68.56 g 100 g−1 DW), respectively. The peel of “Moro” had significantly higher concentrations of total anthocyanins, hydroxycinnamic acids, and flavones (650.67, 263.33, and 449.85 mg kg−1, respectively) compared to the other varieties. In conclusion, “Tarocco Meli” had the most interesting values for morphological parameters, “Tarocco Rosso”, “Sanguinelli”, and “Tarocco Gallo” for primary metabolites, and “Moro” for secondary metabolites. With the increasing interest in utilizing co-products, these findings could be useful in developing functional food products that meet consumer demands for healthier and more sustainable food choices.

The koinobiont endoparasitoid genus Triraphis Ruthe, 1855 (Rogadinae Foerster, 1863) is a group of braconid wasps that contains 74 species distributed along the Nearctic, Neotropical, Oriental and Palearctic regions. We amplified a fragment of the cytochrome oxidase subunit I (COI) for 19 specimens of Triraphis from the Chamela Biological Station (CBS), a region mainly composed of tropical dry forest near the Pacific coast of Jalisco, Mexico. Based on genetic distances among specimens of Triraphis from the CBS and all COI sequences of BINs assigned to Triraphis and Rogas Nees, 1819 available in the BOLDSYSTEMS database, we identified three clusters in the CBS that correspond with T. bradzlotnicki Sharkey, 2021, T. davidwahli Sharkey, 2021 and T. defectus Valerio, 2015, which were previously described from Costa Rica. Based on morphology, we identified individuals of T. fusciceps Cresson, 1869 and provided COI sequences of this species for the first time. Four genetic clusters of Triraphis correspond to four new species that are described here: T. kardia sp. nov., T. ocellatus sp. nov., T. divergens sp. nov. and T. luzabrilae sp. nov. Since T. bradzlotnicki and T. davidwahli were exclusively described with molecular data (COI), we morphologically described them based on Mexican specimens.

Inflammation is a tightly coordinated response of the host immune system to bacterial and viral infections, triggered by the production of inflammatory cytokines. Sepsis is defined as a systemic inflammatory response followed by immunosuppression of the host and organ dysfunction. This imbalance of the immune response increases the risk of mortality of patients with sepsis, making it a major problem for critical care units worldwide. The P2X7 receptor plays a crucial role in activating the immune system by inducing the activation of peripheral blood mononuclear cells. In this study, we analyzed a cohort of abdominal origin septic patients and found that the expression of the P2X7 receptor in the plasma membrane is elevated in the different subsets of lymphocytes. We observed a direct relationship between the percentage of P2X7-expressing lymphocytes and the early inflammatory response in sepsis. Additionally, in patients whose lymphocytes presented a higher percentage of P2X7 surface expression, the total lymphocytes populations proportionally decreased. Furthermore, we found a correlation between elevated soluble P2X7 receptors in plasma and inflammasome-dependent cytokine IL-18. In summary, our work demonstrates that P2X7 expression is highly induced in lymphocytes during sepsis, and this correlates with IL-18, along with other inflammatory mediators such as IL-6, IL-8, and procalcitonin.

Late-season varieties of mandarin (Citrus reticulata Blanco) have a high economic value, so their study, characterization, and comparison among different commercial varieties is of great interest for agriculture. Detailed metabolomic analysis of mandarin leaves can provide valuable information on agronomic characteristics, vegetative development, and tree response to abiotic and biotic stresses. In this study, an analysis of the main metabolites presents in the leaves of three late-season mandarin orange varieties (‘Afourer’, ‘Orri’ and ‘Tango’), cultivated under homogeneous conditions, was carried out using nuclear magnetic resonance (1H NMR) and multivariate statistical analysis techniques. The results show that organic acids and sugars are the metabolites with the highest presence in mandarin leaves, especially malate and sucrose. Ten amino acids and other metabolites such as choline and trigonelline were also detected. Metabolites such as asparagine and isoleucine were widely implicated in the metabolic pathways of the detected compounds. The ‘Orri’ variety showed significantly more differences in metabolite concentrations compared to the other two varieties studied. Malate and sucrose were shown to be the metabolites with the greatest significant differences between the varieties compared. From an agronomic point of view, the ‘Orri’ variety differs from the other two varieties because it has concentrations of metabolites that provide good resistance to abiotic and biotic stresses and fruits of higher quality and sweetness.

El caldén es una especie arbórea de ambientes templado-semiáridos y endémica de la Argentina. El objetivo fue estudiar la morfología, desarrollo y biología de la agalla inducida por un cecidómido en el pecíolo y pulvino del caldén. Fueron colectadas hojas con y sin agallas en fragmentos de bosques periurbanos de Santa Rosa, La Pampa. Los cortes histológicos temporarios fueron realizados a mano alzada y los permanentes con micrótomo rotatorio. Fueron examinadas larvas, pupas y adultos del inductor. La formación de la agalla altera la zona de la base foliar y principalmente el pulvino primario. La agalla es globoide, cerrada, castaño amarillo cuando madura, con 1-3 cámaras larvales y constituida por dos partes (superior e inferior). La organización interna en compartimentos de tejidos es acorde a la estructura general de las agallas de cecidómidos. Entre los rasgos distintivos se destacan la formación de súber, la modificación de las células epidérmicas en esclereidas en la zona de contacto entre las dos partes y la dehiscencia. El inductor es un cecidómido univoltino, cumple el ciclo de vida de la primavera de un año a la primavera siguiente, sobrevive en invierno en el interior de la agalla como larva madura, sobre la planta.

A pesar de situarse en puestos sobresalientes a nivel nacional, la educación primaria en Castilla y León a mediados del siglo xix adoleció de evidentes taras cuantitativas y cualitativas: escasez de escuelas, principalmente en las zonas rurales; escolarización limitada, con fuertes contrastes interprovinciales, escorada por sexos y poco atractiva para gran parte de familias; deficiente y parcial formación del profesorado, con una dotación económica totalmente insuficiente; y metodologías anticuadas que consolidaron posturas retardatarias. A pesar de los años transcurridos desde los ideales postulados educativos del primer liberalismo, mediante el Diccionario de Madoz visualizaremos un cuadro decepcionante que tardará décadas en modernizarse y convertirse en pilar básico del desarrollo socioeconómico regional.

A nivel mundial hay 537 millones de pacientes diabéticos y 14.1 millones en México. Se ha reportado que el Aloe barbadensis Miller, comúnmente conocido como Aloe vera (AV), presenta propiedades anti-hiperglucemiantes y éste contiene compuestos fenólicos. En el presente estudio se evaluó el contenido fenólico (FC), la capacidad antioxidante (AC) y el efecto anti-hiperglucémico (HE) del AV en ratas diabéticas, utilizando muestras con diferentes tratamientos de pasteurización (65, 75 y 85 °C por 15 y 25 min). En los resultados de AC, en dos de los métodos (ABTS y SDS-MFA), los tratamientos con las tres temperaturas y 25 min fueron los menos afectados por la pasteurización. En una curva de tolerancia a la glucosa con ratas sanas, el Aloe vera pasteurizado (PA) a 75 °C durante 25 min, produjo el mayor HE en comparación con el gel de Aloe vera fresco (FAG). Por otro lado, un tratamiento de 21 días con ratas diabéticas, tratadas con FAG y PA a 75 °C durante 25 min; los niveles de glucosa con tratamiento de PA disminuyó por debajo del grupo control diabético y del tratado con FAG. Por lo tanto, el PA mostró un mayor HE que el FAG y dicho efecto pudiera estar relacionado con una mayor biodisponibilidad de los compuestos bioactivos producida por la temperatura de 75 °C. With a progressively decreasing availability of water for irrigation, the utilization of lower agronomic quality water sources is becoming more prevalent. Compounds such as sodium and boron, due to their impact on crop development and production, are gaining significance in these water sources. Finding novel methods to immobilize these compounds in irrigation water is a top priority in the global agricultural sector. This study focused on exploring the potential of natural zeolite, commonly used as a soil improver and as a sorbent for sodium and boron in natural agricultural waters. The zeolite exhibited favorable properties, including a surface area of 40 m2/g and a cation-exchange capacity of 1.8 mg/g. Using a central composite factorial design, the zeolite’s capacity to remove sodium and boron from irrigation water was investigated. The results demonstrated significant efficiency in boron removal, while sodium removal was limited, with occasional desorption episodes. Response surface analysis revealed optimal conditions for the removal of each cation. Additionally, adsorption kinetics and pH effects were explored, emphasizing the influence on sodium sorption. Kinetic models were applied, and the pseudo-first-order model proved suitable for describing the sorption kinetics. These findings enhance our understanding of zeolite efficacy in irrigation water purification, emphasizing the complexity of cation interactions in “complex” solutions.

The pomegranate (Punica granatum L.) attracts attention in studies for its nutritional and medicinal properties. However, a recurring issue in the literature arises due to the multidisciplinary nature of these studies, leading to a mistaken repetition of basic botanical terms. The problem stems from the misapplication of the term “aril” to the pomegranate seed, despite the fruit being exariled, signifying the absence of an aril. This confusion may be attributed to the sarcotesta’s appearance, resembling a complete aril, coupled with a lack of awareness in fields such as medicine, pharmacy, and cosmetics. This study specifically examines the Kingdom-variety pomegranate, due its economic importance in the fruit market. The fruits were evaluated at different developmental stages—initial, intermediate, and commercial. Magnification photography techniques were used to study the development state of the pomegranate fruits. The physiological studies confirm that the pomegranate seed constitutes the complete grain, with the juicy, sweet part surrounding it identified as the testa, not an aril. The findings underscore a persistent error in the existing literature, emphasizing the necessity for dissemination and education in future studies. A thorough grasp of pomegranate anatomy and precise use of terminology are indispensable for ensuring accuracy and rigor in scientific communication.

The significance of Linked Open Data datasets for traffic information extends beyond just including open traffic data. It incorporates links to other relevant thematic datasets available on the web. This enables federated queries across different data platforms from various countries and sectors, such as transport, geospatial, environmental, weather, and more. Businesses, researchers, national operators, administrators, and citizens at large can benefit from having dynamic traffic open data connected to heterogeneous datasets across Member States. This paper focuses on the development of a semantic model that enhances the basic service to access open traffic data through a LOD-enhanced Traffic Information System in alignment with the ITS Directive (2010/40/EU). The objective is not limited to just viewing or downloading data but also to improve the extraction of meaningful information and enable other types of services that are only achievable through LOD. By structuring the information using the RDF format meant for machines and employing SPARQL for querying, LOD allows for comprehensive and unified access to all datasets. Considering that the European standard DATEX II is widely used in many priority areas and services mentioned in the ITS Directive, LOD DATEX II was developed as a complementary approach to DATEX II XML. This facilitates the accessibility and comprehensibility of European traffic data and services. As part of this development, an ontological model called dtx_srti, based on the DATEX II Ontology, was created to support these efforts.

Molting is an essential developmental process in Caenorhabditis elegans. However, the study of molting in the worm has been limited by the lack of automated techniques that allow monitoring the process in a simple way. In 2015, Olmedo et al. published an automated method to monitor the timing of each larval stage and molt in C. elegans using bioluminescence. This new method has greatly contributed to the study of molting in this organism but requires the use of a high-sensitivity luminometer, which many laboratories do not have. We have adapted the method to a conventional luminometer, so that it can be used by most laboratories that work with C. elegans and do not have high-sensitivity equipment. • A customization of a method to study molting in C. elegans using a conventional luminometer instead of a high-sensitivity one. • This adaptation allows most laboratories to use their routine luminometers to study molting in C. elegans. • Although the use of a high-sensitivity luminometer, as proposed by Olmedo et al., remains the gold standard for studying molting, this adaptation is suitable for studying significant differences in molting and the duration of larval stages between different strains of C. elegans.

Medical cannabis (Cannabis sativa L.) is a source of bioactive phytochemicals with promising pharmacological and therapeutic applications. Enhancing the accumulation of valuable bioactive compounds is potentially a way of increasing the economic importance of this crop. Signaling molecules like salicylic acid (SA), jasmonic acid (JA), and γ-aminobutyric acid (GABA) are involved in the regulation of plant development and responses to biotic and abiotic stresses. Moreover, several phytohormones regulate plant trichome formation and elicit the synthesis of secondary metabolites in many plant species in both in vitro and in vivo systems. Therefore, exogenously delivered plant signaling molecules have the potential to modify the chemical profiles of medical cannabis. In this study, we found that the foliar application of SA, methyl jasmonate (MeJA), and GABA produces changes in the accumulation of the two major cannabinoids, cannabidiolic acid (CBDA) and Δ9- tetrahydrocannabinolic acid (THCA), in leaves and inflorescences of a medical cannabis variety. MeJA at 0.1 mM increased the CBDA content in inflorescences by 15.6%, while SA and MeJA at 0.1 mM increased CBDA and THCA accumulation in leaves by up to 57.3%. Treatments did not change the expression of genes participating in the final steps of the biosynthetic pathway of cannabinoids: olivetolic acid cyclase (CsOAC-1 and CsOAC-2), 2-acylphloroglucinol 4-prenyltransferase (CsPT4), cannabidiolic acid synthase (CsCBDAS), and tetrahydrocannabinolic acid synthase (CsTHCAS). Trichome density was not significantly different from the control plants in any treatment. Besides, we found strong correlations between several plant growth parameters and cannabinoid yields, showing a direct link between plant fitness and the production of cannabinoids.

Mandarins are one of the most important citrus fruits in the world in terms of tons produced. The late-season varieties of mandarin have a great economic value due to their high production in a season with few mandarin varieties. The objective is to carry out a preliminary study of characterization and comparison of the morphological and biochemical properties of the late varieties ‘Afourer’, ‘Tango’, and ‘Orri’. The characterization consisted of physicochemical parameters related to the quality of the fruits, highlighting the total antioxidant activity using ABTS and DPPH, the organic acids and sugars using HPLC and the metabolomics of the juice by 1H-NMR. Afourer’ mandarins were heavier and larger (120.75 g, 67.60 mm) than the other two varieties studied. Mandarins of the ‘Orri’ variety showed a different organic acid profile compared to the other varieties studied, and a higher amount of sugars (13.49 g/100 mL). ‘Tango’ variety mandarins grown on the Forner-Alcaide rootstock stood out for having a larger weight (113.52 g), a more intense color, and a greater amount of phenolic compounds (966.85 mg AGE/L Forner) than the fruits grown on Citrus macrophylla. The metabolomics analysis showed that these mandarin varieties had mainly non-essential amino acids.

Blood oranges have high concentrations of bioactive compounds that are beneficial to health. In Europe, the cultivation of blood oranges is increasing due to their excellent nutritional properties. In Citrus crops, rootstocks play an important role in juice and can increase the content of bioactive compounds. The morphological, qualitative and nutritional parameters were analyzed in cultivars ‘Tarocco Ippolito’, ‘Tarocco Lempso’, ‘Tarocco Tapi’ and ‘Tarocco Fondaconuovo’ grafted onto Citrus macrophylla and Citrus reshni. ‘Tarocco Lempso’ grafted onto Citrus macrophylla obtained the highest values of weight (275.78 g), caliber (81.37 mm and 76.79 mm) and juice content (162.11 g). ‘Tarocco Tapi’ grafted onto Citrus reshni obtained the most interesting qualitative parameters (15.40 °Brix; 12.0 MI). ‘Tarocco Lempso’ grafted onto Citrus reshni obtained the most intense red juice (a* = 9.61). Overall, the highest concentrations of primary metabolites were in proline, aspartate, citric acid, and sucrose. The results showed that ‘Tarocco Ippolito’ juice grafted onto Citrus reshni had the highest levels of total hydroxycinnamic acids (263.33 mg L−1), total flavones (449.74 mg L−1) and total anthocyanins (650.42 mg L−1). To conclude, ‘Tarocco Lempso’ grafted onto Citrus macrophylla obtained the best values of agronomic parameters, and the cultivars grafted onto Citrus reshni obtained significantly higher concentrations in primary and secondary metabolites.

The pomegranate tree is cultivated and its fruits consumed since ancient times. This tree is typical of the Mediterranean climate, with high thermal demands to mature properly. The main objective of this work was to study the influence of the fruit bagging technique on the morphometric and biochemical characteristics of the pomegranate fruits of two new varieties that are currently cultivated in the Southeast of Spain. The results indicated that the fruit bagging presented a significant effect on the weight, equatorial diameter, height and shape of the fruit, however, it did not show any influence on the peel thickness. No significant differences have been observed in the number of healthy fruits with and without bagging, however, the number of cracked fruits with Cryptoblabes gnidiella damage was higher for the non-bagged fruits. The fruit bagging presented a significant effect on the total soluble solids, maturity index, glucose, α-punicalagin, α + β-punicalagin and ellagic acid, but it did not show influence on pH, acidity, ABTS, DPPH, FRAP, total phenols, fructose, citric, malic, and quinic acid, β-punicalagin and anthocyanins. The internal fruit color was not affected by the bagging, although it did affect the external color of the fruit, and unevenly depending on the variety. Based on the results, it can be said that bagging can improve the quality of the fruit by reducing damage from pests and pathophysiology, and this benefit compensates or even exceeds the negative effects of bagging on peel color.

Lemon is one of the most significant crops globally, with annual production exceeding 20.8 million tons in 2021. Spain leads the production in Europe with over 62% of lemon production (1.17 million tons in 2021). This study evaluated the real impact of cultivation conditions (rootstock and culture medium) on the compositional characteristics of ‘Verna’ lemons (peel and juice) using 1H-MNR metabolomic identification techniques and multivariate analyses. Twenty metabolites were identified in both the peel and juice samples. Arginine, phenylalanine, ethanol, and trigonelline were absent in the peel samples but present in all the juice. On the other hand, the metabolites asparagine, glutamate, formate, and malate were present in the peel samples but absent in the juice. The analysis of the results indicates that the rootstock had a significant impact on the metabolites related to the energy metabolism of the plant, which directly affects the development of fruits and the influence of the culture conditions (rootstock and culture medium) on the plant’s adaptive response and modification of metabolic pathways.

In Europe, the cultivation of blood oranges for fresh consumption is increasing due to their excellent organoleptic and nutraceutical properties, which give this fruit the status of functional food. Blood orange has a higher content of bioactive compounds, which confer additional benefits for human health. The main morphological and qualitative parameters were studied together with the content of primary metabolites by 1H NMR and secondary metabolites by HPLC-ESI-DAD-MSn from eight varieties of blood orange grafted on Citrus macrophylla. Tarocco Dalmuso was the variety with the highest values of weight (350.6 g), caliber (86.4 mm and 88.6 mm) and juice content (214.2 g). Tarocco Gallo obtained the most interesting qualitative parameters (13.95 °Brix; 22.75 MI). The most intense red juice was in Sanguinelli (a* = 9.45) and, in crust, it was in Tarocco Scirè (a* = 40.13). The most abundant primary metabolites were proline, aspartate and asparagine, citric acid and sucrose. The results showed that the juice of the Moro had the highest levels of total flavones and flavanones (90.07 and 592.88 mg L−1, respectively), and Sanguinelli in total anthocyanins (101.06 mg L−1). To conclude, Tarocco Dalmuso obtained the best values of agronomic parameters, and Moro and Sanguinelli in the content of phenolic compounds.

Much attention has been paid to the health benefits of including fruits and vegetables in the diet. However, for the compounds responsible for this beneficial effect to be effective at the level of the human organism, they must be available for absorption after digestion. In this sense, in vivo studies are needed to demonstrate the bioavailability of these compounds and their physiological activity. In order to provide information in this regard, this study collects data on the levels of vitamin C (VC) and naringenin (NAG) in the blood serum of the 11 volunteer participants in this trial, before and after consuming two different grapefruit juices. The juices were prepared by rehydrating the grapefruit powder obtained by freeze-drying (FD) the fruit puree or by spray-drying (SD) the liquefied grapefruit. No significant differences (p > 0.05) neither by juice nor by participant were observed in any case. The mean relative increase of VC, NAG and the radical scavenging ability (RSA) in blood serum due to grapefruit juices intake was 12%, 28% and 26%, respectively. Just VC showed a positive and significant Pearson’s correlation with RSA. The mean bioavailability of VC was quantified as 1.529 ± 0.002 mg VC/L serum per 100 mg of VC ingested.

The reuse of food by-products is crucial for the well-being of the planet. Considering the high content of nutrients and other bioactive compounds in many of them, investigating their suitability for use as human food ingredients is an interesting challenge. In this study, in addition to the proximate composition, phenol content and antioxidant activity (AOA = 3.2 mmol Trolox equivalent (TE)/100 g, db) of orange juice powder by-product (CoP), different in vitro properties related to carbohydrate metabolism have been characterised. Specifically, the glycaemic index (GI), the glycaemic load (GL), the glucose dialysis retardation index (GDRI = 13.6%), the glucose adsorption capacity (GAC = 22.5 mM) and the inhibition capacity of α-amylase (α-A = 46.9%) and α-glucosidase (α-G = 93.3%) of powdered orange juice waste have been determined and related to fibre and phenolics composition. Taking advantage of the high fibre content of the by-product (36.67%), its GL was calculated for a CoP dose that allows labelling the food to which it is added as a source of fibre. The low GI value (24.4%) and the low GL (0.918 g available carbohydrates per serving) allowed us to conclude that the product studied could be an interesting opportunity for the food industry to offer it as a healthy food ingredient to be included in the diet, especially for those suffering from type 2 diabetes mellitus. Of the total phenolic compounds (TP = 509 mg equivalent of gallic acid (GAE)/100 g, db), 68% were found in free fraction (FP), and their contribution to the total AOA was 40.6%, while this was 54.9% for the 32% of phenols bound to plant tissues (BP).
