5571 Background: In the Phase III PAOLA-1/ENGOT-ov25 trial, the addition of maintenance olaparib (ola) to bevacizumab (bev) in patients (pts) with newly diagnosed advanced high-grade ovarian cancer (HGOC) resulted in prolonged progression-free survival (PFS), particularly for pts with HRD-positive tumors including a BRCA1/ BRCA2 mutation (BRCAm; Ray-Coquard et al. NEJM 2019). Preclinical data suggest that pts with mutations (mut) in the RING domain of BRCA1 are less sensitive to ola. The magnitude of benefit from ola + bev, according to the location of mut in the functional domains (FD) of BRCA , remains to be explored. Methods: Pts with newly diagnosed advanced HGOC in response after platinum-based chemotherapy + bev received maintenance bev (15 mg/kg q3w for 15 months [mo]) + either ola (300 mg bid for 24 mo) or placebo (pbo). In this post hoc exploratory analysis, PFS was analyzed in pts with BRCAm according to mut location in the FDs of BRCA1 (RING, DNA-binding domain [DNA-BD], or BRCA1 C terminus [BRCT]) and BRCA2 (RAD51-BD; DNA-BD). Results: Among the 806 randomized pts, 235 (29.2%) harbored a BRCAm: 160 (19.9%) with a BRCA1 mut and 76 (9.4%) with a BRCA2 mut. BRCA1 mut in FDs of RING, DNA-BD and BRCT were detected in 19 (11.8%), 41 (25.6%) and 34 (21.2%) pts, respectively. BRCA2 mut were detected in FDs of RAD51-BD and DNA-BD in 37 (48.7%) and 14 (18.4%) pts, respectively. With a median follow-up of 25.5 mo, 24-mo PFS rates and hazard ratios (HRs) according to mut locations are reported in the Table. In pts with a BRCA1 DNA-BD mut, 24-mo PFS was 89% and 14% (ola + bev vs pbo + bev; HR 0.08, 95% confidence interval [CI] 0.02–0.26) compared with 64% and 24% for pts with mut in RING + BRCT + other domains (HR 0.33, 95% CI 0.19–0.57). In pts with BRCA2 mut, 24-mo PFS for pts with mut in the DNA-BD was 91% vs 100% (ola + bev vs pbo + bev) compared with 82% and 44% for pts with mut in RAD51-BD + other domains (HR 0.21, 95% CI 0.08–0.54). Conclusions: In this exploratory analysis, pts with newly diagnosed advanced HGOC and a BRCAm had a PFS benefit from maintenance ola + bev regardless of mut locations in BRCA1/BRCA2. Sensitivity to ola + bev maintenance was particularly high for pts with mut in the DNA-BD of BRCA1. Pts with a mut in the DNA-BD of BRCA2 commonly had excellent outcomes. e13606 Background: SARS-CoV-2 vaccines have changed the course of the current global pandemic. Cancer patients were identified as highrisk of adverse infection outcomes. We have previously characterised the serological response to SARS-CoV-2 vaccines in 220 cancer patients treated at our institution. In addition these patients were given the possibility to report their symptoms (patient-reported outcomes, PROs) weekly using a digital platform (ePROs). We sought to determine if, in cancer patients, the prospectively recorded post-vaccination ePROs could predict the serological response to SARS-CoV-2 vaccines. Methods: We used a pre-existing digital platform that allows monitoring of PROs using weekly questionnaires sent to patients and available on their desktop computers, tablets or smartphones. Serial serologies were performed at 28, 50 and 115 days after vaccination. Results: We observed that at day 50 after the first vaccination dose, coinciding with three weeks after the second dose, patients could be divided into two groups according to their serological response (low – below 1500 U/ml and high – above or equal 1500 U/ml). A peak in symptom burden could be observed after the second dose, as previously described. Omitting ePRO features decreased prediction performance of all models, whereas omitting baseline symptom scores had inconsistent effects. Among all models and feature constructions, the top performance metrics were given by the nearest centroid model7 with baseline symptoms omitted and 20 features chosen with the aforementioned procedure. The model achieved an accuracy of 0.704, an F1-score of 0.759 and an MCC of 0.398. Conclusions: we were able to identify the patients who achieved higher antibody levels against SARS-COV-2 based on the symptom burden reported through ePROs. This represents the first model showing that symptoms, assessed through ePRO can be predictive of response to vaccines. Our results could also be useful information for patients, as they could assuage their fears about adverse -effects, through the knowledge that toxicity could predict better protection against SARS-COV-2. The same toxicity-based prediction of efficacy has been identified with immunotherapy in cancer and is now a routine part of clinical discussions with patients.

NodalBc: A Unique Program for Optimum Production of Sucker Rod Pumping Oil Wells Abstract Sucker rod pumping is the oldest artificial lift method used in production of oil wells. This method is usually designed and analyzed with the assistance of programs based on simplified models, such as the API RP 11L, or with a wave equation simulator. In many cases, however, a technique that permits consideration of the inflow and outflow performance of the well is a more efficient way to design and analyze an oil well. This technique, referred to as Total Analysis or System Analysis (SA), has been successfully applied to flowing and artificially lifted wells. This paper presents a computer program developed by Intevep S. A., called NodalB, created to perform the SA of sucker rod pumping wells. NodalB 's unique characteristics make it a competitive program in today's market of design and optimization software. This program allows consideration of the inflow performance of the reservoir depending on the drilling scheme, either vertical or horizontal, and determination of the sensitivity of the rod pumping system to its most relevant parameters, such as: pumping speed and stroke, pump diameter, gas oil ratio, gas separation efficiency, tubing diameter, pump depth, rod string design, oil viscosity, injected diluent (diesel, kerosene, etc.) ratio, water cut, reservoir pressure, productivity index, and different production schemes (i.e. production through the tubing or through the casing-tubing annulus). These features show the power of this program as a predictive tool. NodalB is specially applicable to design, optimization and troubleshooting of heavy and viscous oil wells, taking into account the viscosity effects in the rod pumping system. Introduction The current total oil production of Venezuela is approximately three (3) millions of barrels per day. One million of it is produced from almost 7000 sucker rod pumping wells, and consists, in its majority, of heavy oil and bitumen, which typical downhole viscosity ranges from 500 cp up to 7000 cp. The corresponding API gravity ranges between 8 and 12. Due to the high viscosity of the oil, cyclic steam injection has become a typical production scheme for those wells in which static reservoir pressure is below 1000 psi (up to 2500 ft of well depth). Also, other alternative production schemes have been developed to reduce 1) the drag forces between the rod string and the fluid and 2) the pressure drops in the tubing string and flowline. Such schemes involve the injection of a low viscosity fluid (diesel, kerosene) in a continuos basis into the wellbore and flowline. Furthermore, the oil being produced usually flows through the tubing-casing annulus instead of the tubing. Recently, a new completion scheme, called the Downhole Stuffing Box Production Scheme, has been successfully tested in the field, revealing several advantages over the other heavy oil production schemes currently applied in Venezuela.
