Objective: To determine whether administration of progesterone on the day of oocyte retrieval may reverse accumulation of fluid in the endometrial cavity. Methods: A total of 50 patients who underwent assisted reproductive technology (ART) cycles with endometrial cavity fluid (ECF) observed by ultrasound at the time of oocyte retrieval were included. Upon the identification of ECF, vaginal administration of natural progesterone was started. Two days later, the endometrial cavity was re-evaluated, and embryo transfer was performed in the absence of ECF. Results: ECF was absent two days after administration of vaginal progesterone in 47 of the 50 patients (94%). ECF persisted in 3 of the 50 patients (6%). The clinical pregnancy rate per transfer was 34.0%, and the implantation rate was 21.6%. Conclusion: Our data suggest that, in the presence of ECF, administration of intravaginal progesterone in ART cycles must be initiated on the day of follicle aspiration to reverse ECF and to avoid the deleterious effects of fluid on the blastocyst-endometrial interaction.

Agricultural intensification and landscape simplification have reduced flowering plant abundance and diversity worldwide, with subsequent negative effects on pollinators. To mitigate these effects, research and conservation have focused on incorporating flowering plants into agricultural landscapes through on-farm natural areas or flowering crops. However, while cover crops provide numerous agroecosystem services, they have received little attention for their role in providing floral resources for pollinators. In this study, we examined the value of cover crops for providing supplemental summer floral resources for both managed and wild bees in the Northern Great Plains of the United States, where a large proportion of honey bee colonies spend the summer. Specifically, using an experimental randomized plot design, we compared floral resources and bee visitation rates across 1, 2, 3, and 6-species cover crop mixes as well a 6-species annual native wildflower mix. We found that cover crop treatments including buckwheat provided the highest summer-long floral cover, higher than the annual native wildflower mix in which only two of six species bloomed, and that total bee visitation rates increased with higher floral cover. However, different bees were attracted to different treatments; honey bees and bumble bees were most attracted to mixes containing phacelia, while large-bodied solitary bees were more attracted to mixes including cultivated sunflower and to the wildflower mix, illustrating that the optimal plantings for attracting social or managed bees may differ from those attracting solitary wild bees. Additionally, while similar numbers of bee species visited the three main plants in our treatments (buckwheat, phacelia, and cultivated sunflower) more rare species visited cultivated sunflower while more common bees visited buckwheat and phacelia. Our results highlight the value of annual cover crops for providing pollinator resources in agricultural landscapes, particularly on rotated land where perennial flower plantings are less feasible, but illustrate trade-offs between attracting a large number of common generalist or managed bees versus attracting fewer individuals but species of potential conservation concern.

Objective It is generally accepted that the incidence of birth defects in spontaneously conceived children ranges between 2.0-4.0%. However, several studies have shown that babies born after assisted reproductive technology (ART) procedures tend to present more congenital malformations than naturally conceived children, with 6.5% of the children born after intracytoplasmic sperm injection (ICSI) presenting birth defects. The use of high magnification sperm selection before ICSI was introduced in the early 2000s to allow the identification of spermatozoa with low risk of sperm DNA damage. Intracytoplasmic morphologically selected sperm injection (IMSI) is expected to change the incidence of congenital malformations, although data on the incidence of birth defects in children conceived after IMSI are still scarce. Methods A systematic review based on searches performed in electronic databases (PubMed, EMBASE, Web of Science, SCOPUS, and Cochrane Central Register of Controlled Trials) including articles published by February 2021 was conducted to identify trials comparing the neonatal outcomes of ICSI and IMSI. The outcome measured was the rate of birth defects in children born after ICSI or IMSI. Three trials were included as targets for data extraction and meta-analysis. Results Our meta-analysis included 3907 children conceived after IMSI (1280) or ICSI (2627). The incidence of birth defects was statistically different, with 2.5% (32/1280) in IMSI and 4.5% (119/2627) in ICSI (RR=0.59; 95% CI=0.40-0.87; p=0.007). The results demonstrated that IMSI decreased the incidence of structural defects compared to ICSI - 2.2% (18/830) vs. 3.8% (78/2049) - in a statistically significant manner (RR=0.58; 95%CI=0.35-0.96; p=0.04). No significant difference was observed in chromosomal abnormalities (Trisomy 13; 18; 21 and Triple X) between children conceived after IMSI (8/830) or ICSI (19/2049) (RR=1.07; 95%CI=0.47-2.43; p=0.87). Conclusions IMSI seems to be an effective tool at reducing the incidence of structural defects compared to ICSI. However, IMSI does not change the incidence of chromosomal abnormalities.

Plant–pollinator interactions are mediated by floral signals and by the quantity and quality of floral rewards. Biotic and abiotic disturbances can influence plant reproductive success through both direct effects on plant performance and indirect effects on pollinator attraction. In this study, we examined the effects of drought on buckwheat (Fagopyrum esculentum Moensch), a globally cultivated plant that is prone to drought stress, dependent on insect pollinators for reproduction, and increasingly utilized in on‐farm conservation. Between drought‐stressed and control plants, we compared: nectar quantity and chemical composition, pollen quantity, floral volatile emissions, visits by both managed and wild pollinators, and plant reproductive success. Drought‐stressed plants produced significantly fewer flowers and less nectar per flower, though pollen quantity per flower was unaffected. Nectar from drought‐stressed plants had a lower proportion of sucrose relative to total sugars, though overall sugar concentration was unaffected. Significantly fewer bumble bees, honey bees, and flies were recorded on drought‐stressed plants. While there was no significant difference in the quantity of total floral volatile emissions, volatile compositions differed, with drought‐stressed plants having higher emissions of (Z)‐3‐hexenol, isobutyraldehyde, 2‐methylbutanal, and 3‐methylbutanal. Finally, drought stress had negative effects on seed set and total seed mass per plant. Our results show that drought stress can have significant effects on floral traits and pollinator attraction, reducing plant reproductive success, and the nectar resources available to pollinators. Thus, the potential value of this plant in pollinator conservation and as a honey plant may be reduced under drought stress.

Objective To assess the relationship between human blastocyst chromosomal ploidy established by niPGT-A and increasing age. Methods This is a prospective multicenter study carried out by ten assisted reproduction centers after their embryologists acquired training and validated their results with the previous use of niPGT-A. A total of 94 couples with indication for niPGT-A due to increase maternal age, male factor, repeated implantation failures, recurrent abortion or because they requested niPGT-A were included in this study. The couples had no karyotype abnormalities. After ICSI, the embryos were cultured until blastocyst stage using one or two step culture systems, single or sequential media respectively, at 37°C in an atmosphere of 6-7% CO2 and 5-20% O2 incubators. On day 3, we re-evaluated cleavage embryos to complete cumulus cells removal. The embryos were then cultured in individual well, with 20µl of medium under oil until they reached blastocyst stage. The blastocysts were vitrified and stored in liquid nitrogen. After that, the spent blastocyst culture medium (20µl) was transferred to a PCR tube and sent for analysis in the genetic laboratory, where it was stored at -80°C until sequencing. A total of 243 samples of spent blastocyst culture medium were collected on the 5th/6th day. Cell-free DNA secreted on culture medium was amplified using NICS Sample Preparation Kit (Yikon Genomics), based on the MALBAC technology. After whole genome amplification, the DNA was measured using a Qubit 2.0 fluorometer and subjected to next generation sequencing (NGS) using Illumina MiSeq® platform. The data were analyzed using the ChromGo® software (Yikon Genomics). Results The mean age of the patients was 38±4.08 years with an interval of 20-44 years. The euploid was diagnosed in 36.4% (80/220) of cases, aneuploidy in 31.3% (69/220), and mosaicism in 32.3% (71/220; with ≥60% aneuploidy) of blastocysts. Mosaic values ranged from 29.8% to 33.8% in different age groups. Individually, the most frequent chromosomal abnormality was XXY (Klinefelter Syndrome) occurring in 18 cases, followed by chromosome 21 (trisomy/monosomy) in 8 cases. The niPGT-A data showed a ≥60% incidence of aneuploid cells in all cases of chromosomal mosaicism (n=71). Conclusion A high degree of mosaicism with aneuploidy cells was detected, and some hypotheses were suggested for this data (niPGT-A sensitivity in detecting the self-correction of chromosomal abnormalities phenomenon). However, it did not vary remarkably with age. On the other hand, euploidy levels had a negative correlation with age and aneuploidy levels had a positive relationship. This is the first report in the literature to relate chromosomal ploidy in blastocysts using niPGT-A and increasing patient age.

Recently, a new technology known as the Noninvasive Preimplantation Genetic Testing for Aneuploidy (niPGT-A) emerged, using cell-free DNA present in the spent culture media of human blastocysts. Unlike PGT-A, in which only trophectoderm cells are used, niPGT-A reflects the ploidy state of these cells and internal cell mass, suggesting that this new technology may be less prone to error, being more reliable than the invasive test. The aim of the present study was to report the first occurrence of childbirth following niPGT-A in Brazil.

The inclusion of a smother crop used as a cash crop in an intercropping system may be an effective cultural control strategy for the management of weeds in organic production systems. In addition, a multilayered canopy created when intercropping species with different growth forms may limit germination cues for weed seeds and can allow for a more efficient utilization of resources that reduce competition to target crops from weeds. Watermelon (Citrullus lanatus) was evaluated for its ability to reduce weeds in a low-input organic system in Texas when planted alone or in various intercropping combinations that also included peanut (Arachis hypogaea), okra (Abelmoschus esculentus), cowpea (Vigna unguiculata), and hot pepper (Capsicum annuum). Watermelon significantly reduced total weed biomass when planted in monoculture and in all intercropping combinations compared with peanut, okra, cowpea, and pepper monocultures in year 1 of the 2-year study. Total weed biomass was reduced by 81%, 83%, 88%, and 92% in treatments containing watermelon on average as compared with pepper, peanut, okra, and cowpea grown in monoculture, respectively. Less effective weed suppression was obtained with watermelon in year 2. Pepper grown in monoculture had significantly higher weed biomass than all other treatments in year 2. Broadleaf weeds were effectively suppressed across all intercropping treatments in year 1, but nutsedges (Cyperus sp.) were consistently reduced both years, particularly when compared with monocrops with small leaf area such as pepper. The three and four species intercropping combinations consistently had high leaf area index (LAI) values, whereas pepper monoculture had significantly lower LAI values than all other treatments except for cowpea monoculture. There was a significant negative relationship between LAI and total weed biomass 33 d after last planting (r = −0.51, P < 0.01). There was a significant negative relationship between total weed biomass and total fruit yield in year 1 (r = −0.64, P < 0.01) but no significant relationship in year 2. Although findings were inconsistent in year 2 because of changes in precipitation amounts and in relative planting dates, these findings suggest that incorporating a multifunctional intercropping system that includes a low-growing vining crop such as watermelon or at least an architecturally complex mixture can optimize canopy density to reduce weed pressure from resilient perennial weeds such as nutsedge. This may offer organic producers another management tool for the control of perennial weeds.

Interspecies specific interactions are generally regarded as drivers of plant productivity in multispecies agroecosystems. Complementary use of resource in diverse communities can enhance community productivity through optimal use of plant-available resources and positive interactions such as facilitation can ameliorate high abiotic stress conditions. We studied the effects on physiological response, leaf traits and water use efficiency of a multifunctional species intercropping system consisting of peanut (Arachis hypogaea L.), watermelon [Citrullus lanatus (Thunb.) Matsum. & Nakai], okra [Abelmoschus esculentus (L.) Moench], cowpea [Vigna unguiculata (L.) Walp.], and pepper (Capsicum annuum L.) planted alone or in various intercropping combinations in a low fertilizer input system in the peak of summer heat in Texas. Differences in gas exchange measurements were detected only in watermelon in year 2 of the study when okra was the dominant crop. This same year watermelon specific leaf area (SLA) was significantly higher when okra was present in a treatment and particularly in the three and four species combinations, Wpwo and Wpwoc, 27.5 and 31.0 m2 kg−1, respectively, as compared to watermelon grown in monoculture, strip intercropped with peanut (Spw) and within row intercropped with peanut (Wpw), 20.4, 20.1, and 19.8 m2 kg−1, respectively. This corresponds with an increase watermelon leaf N concentration and a decrease in leaf C:N ratio in Wpwo and Wpwoc treatments. No differences in d13C composition, a measure of water use efficiency over the leaf lifespan, were detected across cropping system for each species. Water use efficiency based on per plant production (WUEyield) indicated an increase in water use efficiency in dominant crops such as watermelon in 2011 and okra in 2012, but a reduction in WUEyield subordinate crops such as cowpea and pepper both years of the study. Peanut grown in monoculture and strip intercropped with watermelon had significantly lower leaf water potential values in 2012, −2.2 and −2.1 MPa, respectively, as compared to intercropping systems increasing in level of integration (Wpw = − 1.1, Wpwo = − 0.6, Wpwoc = − 1.3, Wall = −1.1 MPa), indicating peanut benefited from alterations to microclimate and facilitative interactions with companion crops in some intercropping systems through a reduction in plant water stress. The results from this study suggest there may be a benefit to a multifunctional intercropping system in the form of increased food production per unit of water input in dominant crops and reduced water stress for some component species. This is important to producers; showing a method to increase overall crop production without increasing water inputs.

Objective The present meta-analysis aimed to evaluate whether the freeze-all strategy (Freeze/All-ET) could bring about improvements in the clinical assisted reproductive technique (ART) outcomes when compared with the fresh embryo transfer strategy (Fresh-ET) in patients undergoing an ART cycle in accordance with the mean number of oocytes collected. Methods A systematic review based on electronic searches in databases (PubMed, EMBASE, Web of Science, SCOPUS, and Cochrane Central Register of Controlled Trials) was carried out to identify randomized controlled trails (RCTs) comparing ART outcomes between fresh-embryo transfers versus elective frozen-embryo transfers up to February of 2017. Four reviewers independently evaluated abstracts, validity assessment and data extraction. Odds Ratio (OR) values with a 95% confidence interval (CI), and heterogeneity were evaluated. Results Five RCTs were included as targets for data extraction and meta-analysis purposes. Objective The aim of this study was to evaluate the influence of different periods of abstinence on conventional semen parameters as well as functional parameters in human semen, including mitochondrial function, chromatin packing and sperm DNA fragmentation. Methods We recruited a cohort of 2,458 men undergoing infertility investigation. Semen analyses were performed according to WHO guidelines/morphology-motile sperm organelle morphology examination/MSOME. For DNA integrity analysis, the percentages of DNA fragmentation (TUNEL), abnormal chromatin packaging/underprotamination (chromomycin A3/CMA3), abnormal mitochondrial membrane potential (MMP/MitoTracker Green), and apoptosis (annexin-V) were recorded. Associations between the sexual abstinence period and sperm parameters were assessed using Spearman correlation. For group comparisons, the subjects were categorized according to the sexual abstinence period (SAP) into three groups: SAP 5 days. Results The duration of abstinence had a statistically significant positive influence on sperm concentration and volume, the number of leukocytes and a statistically significant negative influence on sperm motility and vitality. The percentages of DNA fragmentation and MMP (mitochondrial damage) worsened with the increased duration of abstinence. The percentage of sperm protamination was statistically significantly increased with abstinence. Conclusion Increase in the sexual abstinence period influences sperm quality. This study reinforces the importance of the duration of ejaculatory abstinence on semen parameter variation. It highlights the deleterious effect of increased abstinence on DNA damage, which is most likely associated with ROS (mitochondrial damage/number of leukocytes). The increase in chromatin packaging can represent a protective feature for DNA.

This study aimed to evaluate if single medium is better than sequential medium at improving ongoing pregnancy rates in patients undergoing assisted reproductive technology (ART) procedures. The data featured in this meta-analysis were extracted from four randomized controlled trials yielded from a systematic search carried out on electronic databases. The primary endpoint was ongoing pregnancy rate. Secondary endpoints included clinical pregnancy and miscarriage rates. The endpoints for ongoing pregnancy rate were also analyzed based on the time at which the embryo transfers were performed: cleavage stage (day 2/3) and/or blastocyst stage (day 5/6). There were no significant differences between single and sequential medium for clinical pregnancy (RR=1.09; 95%CI=0.83-1.44; p=0.53), ongoing pregnancy (RR=1.11; 95%CI=0.87-1.40; p=0.39), or miscarriage rates (RR=0.89; 95%CI=0.44-1.81; p=0.74). No significant difference was found for ongoing pregnancy rate (RR=1.29; 95%CI=0.93-1.78; p=0.12) between single and sequential medium when only trials in which embryos were transferred at the blastocyst stage were included. In conclusion, the choice of embryo culture approach - single or sequential medium - did not affect the ongoing pregnancy rates of patients undergoing ART cycles.

Objective To evaluate whether or not one should use a new Protocol for Endometrial Receptivity Improvement (PRIMER) based on platelet-rich plasma (PRP) and granulocyte colony-stimulation factor (G-CSF) to enhance ongoing pregnancy rates in patients with recurrent implantation failure (RIF). Methods Women undergoing IVF/ICSI were prospectively divided into two groups: - PRIMER/RIF group (n:33): patients with RIF (defined as ≥2embryo transfers (ETs) and at least 5 morphologically good embryos transferred) in which intrauterine PRP injection and subcutaneous G-CSF-injection were performed. - Control group (n:33): patients in their first IVF/ICSI attempt/cycle (without PRP or G-CSF injection). The PRP was prepared using autologous fresh-whole blood processed to increase platelet-concentration in 2 to 4 fold. All patients undergoing the PRP-treatment received 0.7ml of it through intrauterine-injection 48 hours before the ET. G-CSF (300mg/0.5ml) started simultaneously to PRP and was administered subcutaneously every week. Results Regarding implantation, clinical pregnancy and miscarriage rates, we found no statistically significant difference (18.2% versus 17.6%, p=0.90; 36.4% versus 30.3%, p=0.61 and 25.0% versus 9.0%, p=0.43, respectively). The use of PRIMER enabled RIF patients (previous ET µ: 4.0±1.5) to reach similar ongoing pregnancy and live birth rates like those patients who had their first IVF/ICSI cycle attempt (27.3% versus 27.3%, p=0.99). Conclusions Our results showed, for the first time, evidence that this therapeutic protocol (PRIMER) could be used as a feasible treatment based on biological rationale for patients with RIF, considering its promising outcomes, it is a simple procedure and not associated with patient complications.

After more than 20 years of use of preimplantation genetic tests for aneuploidies (PGS/PGT-A) there are still many problems related to the efficiency of this technique, most of them still without an adequate solution (Gleicher et al., 2018; Homer, 2019). From the clinical point of view, the benefits attributed to invasive PGT-A in the selection of euploid embryos remain controversial, especially due to the lack of scientific proof of its effectiveness in increasing live birth rates in various clinical situations, such as patients with advanced age, repeated implantation failures or recurrent miscarriages. In addition, evidence-based medicine also severely criticizes the rare randomized trials analyzing the clinical use of invasive PGT-A (Orvieto, 2016). If these criticisms were not enough, and undoubtedly one of the most important, it would be difficult to accurately assess the presence of embryonic mosaicism creating significant levels of false positive results, and worse, causing a real possibility of discarding healthy embryos. This makes the clinical application of PGT-A as a risky approach (Munne et al., 2017; Spinella et al., 2018). Another problem, not less important, would be the obligation to perform PGT-A by experienced embryologists, since otherwise the embryonic loss due to biopsy would be a frequent fact, something usually estimated below 10% but in some laboratories it may reach up to 30% of biopsied embryos (Munne, 2018). On the other hand, there are doubts about the future risks of invasive action of the usually 5-10 cell removed during biopsy for genetic diagnosis. Would there be repercussions for the health of these children? In animals, there are data suggesting that embryonic biopsies could be linked to changes in fetal neural tube or adrenal development (Wu et al., 2014; Zeng et al., 2013). Recently, Xu et al. (2016) described noninvasive chromosomal screening (NICS) by obtaining and sequencing free DNA dripped by embryos in the culture medium (without the need of embryo biopsy) creating a new non-aggressive and elegant perspective to preimplantation genetic diagnosis.

Integrated farming systems are agricultural systems with multiple enterprises that interact in space and time. Key aspects of these systems are inter-dependence among enterprises within the system, synergistic transfer of resources among enterprises, and flexibility in the system to be sustainable in the long-run. Integration can be described along the three dimensions of organization, space, and time. Systems that are closely integrated in all three dimensions are those with many enterprises that interact at a common scale of space and time. Integrated farming often includes both crop and livestock enterprises. While integrated systems were common historically, the trend has been toward more specialized systems focused on productivity. Several types of drivers influence the adoption of integrated farming, including economic, social, and environmental drivers. Integrated systems enjoy economies of scope, but these benefits tend to decrease with larger enterprises that benefit from economies of scale. There are many challenges inherent in integrated farming, including the complexity of managing multiple enterprises. Future developments in technology, regulations, labor availability, and demands for food and ecosystem services will influence the adoption of these systems. Technologies that help managers deal with complexity may facilitate greater adoption of these systems, and integrated farming systems may play a key role in meeting future food needs while maintaining or enhancing ecosystem services.

Objective A variety of studies randomizing women/cycles or oocytes/embryos has been carried out to compare different culture media for culturing embryos up to cleavage or blastocyst stages showing controversial results. A recent systematic review suggested that data in the literature are insufficient to conclude the best culture medium for embryo quality, pregnancy and implantation. The objective of this study was to evaluate whether there is any difference between two commercial culture media regarding clinical outcomes after IMSI cycles. Methods A total of 120 patients, ≤39 years of age, undergoing ART treatment submitted to the IMSI program were prospectively broken down and randomized into two groups: Group I (Cook media) and Group II (Vitrolife media). Results Our data demonstrated that there was no difference using all the media from Cook or all the media from Vitrolife, for culturing embryos till day 2, in the bench incubator at low O2 concentration, in relation to fertilization, embryo quality, pregnancy and implantation rates (p>0.05). Conclusion Both culture media used, Cook medium and Vitrolife medium, for the IMSI procedure and for later embryo culture with transfer on the second day, are equally effective and can be used depending on the ease and availability of acquisition.

Certain gene polymorphisms are associated with implantation failure and pregnancy loss. Studies of leukaemia inhibitory factor (LIF) gene polymorphisms are scarce. The LIF single nucleotide polymorphism (SNP) thymine (T)/guanine (G) (rs929271) was studied in women to determine whether an association existed with pregnancy outcomes after intracytoplasmic sperm injection (ICSI); 411 women who underwent ICSI were recruited. DNA was extracted from the peripheral blood, and the LIF gene SNP T/G (rs929271) was genotyped using real-time polymerase chain reaction. Participants were divided into three groups according to their LIF genotype: T/T (n = 168), T/G (n = 202) and G/G (n = 41). All IVF and ICSI procedures were carried out under the same clinical and laboratory conditions. The ICSI cumulative results (from fresh plus frozen cycles) of each genotype group were analysed. The G/G genotype in women was associated with a higher implantation rate (T/T: 15.9%, T/G: 16.2%, G/G: 27.0%; P < 0.05), ongoing pregnancy rate/patient (T/T: 31.5%, T/G: 36.1%, G/G: 53.7%; P < 0.05) and ongoing pregnancy rate/transfer (T/T: 18.5%, T/G: 20.2%, G/G: 36.7%; P < 0.05). LIF SNP T/G (rs929271) seems to be a susceptibility biomarker capable of predicting implantation efficiency and pregnancy outcomes.

I agree with Agarwal et al. (1) that sperm DNA fragmentation (SDF) should be included in the evaluation of male factor infertility, along with semen analysis.

Perhaps with the intention of obtaining larger amounts of free-DNA, some groups are routinely postponing and establishing free-DNA collection in culture medium for Noninvasive preimplantation genetic testing for aneuploidies (niPGT-A) to day 6 for all blastocysts. A meta-analysis served as the basis for such decision, since statistically similar live birth rates were observed when the transfers of euploid blastocysts were performed on day 5 versus day 6 However, the euploidy analysis was conducted in only two studies However, after including the two more studies we performed a new meta-analysis that clearly showed the risks of losing live births with the decision of adopting the 6th day as the endpoint for gathering free-DNA. We would be losing 1.71x more live births.

Objective To investigate an association between polymorphisms related to the implantation process that together could help in the prediction of recurrent implantation failure (RIF). Design Cohort study. Setting Private fertility center and reproductive genetics laboratory. Patient(s) Forty-four women presenting RIF, who were included in study group (RIF group), and two control groups, one with 63 women who were attended at our service and became pregnant after the first IVF/intracytoplasmic sperm injection attempt (control group I) and other with 65 fertile women who had at least two children without any treatment and no history of miscarriage (control group II). Intervention(s) None. Main Outcome Measure(s) Genotyping was performed in the intron region of TP63, VEGFA, MMP2, ESR1, and ESR2 genes and in the 3′ untranslated region of the LIF gene on genomic DNA using real-time polymerase chain reaction. Result(s) The presence of ESR1/AA (rs12199722) and LIF/GT (rs929271) genotypes was more frequent in the RIF group, leading to a 7.9-fold increase in the chance of women presenting with RIF when compared with women who became pregnant on their first cycle of IVF/intracytoplasmic sperm injection and a 2.8-fold increase when compared with women who became pregnant without treatment. Conclusion(s) The association between ESR1 and LIF polymorphisms can help in the prediction of RIF.

Saline irrigation water can lead to salt buildup and reduced crop yields. Halophytic plants are known to accumulate excess salts in tissues, removing them from the immediate environment. This two-phase experiment explored the feasibility of intercropping watermelon (Citrullus lanatus (Thunb.) Matsum. and Nakai var. lanatus) with halophytic species to mitigate the negative effects of saline irrigation water while providing a value-added crop. In the first experiment, six greenhouse-grown species were irrigated with water that was either deionized (0 dS m−1) or contained 3 or 6 dS m−1 of salts for 41 days and screened for growth and salt removal. Two halophytes were selected to be additively intercropped with watermelon under field conditions and irrigated with the same saline irrigation levels as the first experiment. Results indicated that garden orache (Atriplex hortensis L.) exhibited the highest growth rates and purslane (Portulaca oleracea L.) accumulated high amounts of sodium in plant tissues under saline irrigation. The field experiment showed that watermelon yields, stem water potential, and fruit quality were not affected by saline irrigation; however, the watermelon/orache intercropping treatment had significantly higher yields. These results suggest intercropping with halophytes has the potential to contribute a value-added crop without reducing watermelon yields.

In natural ecosystems, plant communities composed of functionally diverse species produce more biomass overall than less diverse communities. This increased biomass production is thought to occur due to complementary use of resources such as nutrients and water, and facilitation during sub-optimal environmental conditions. Using the same concept in a crop setting may lead to increased yield (overyielding) in diverse cropping systems when compared to monocultures. Different combinations of peanut, watermelon, okra, cowpea, and pepper planted alone or in various intercropping combinations were investigated over two growing seasons in a low-input system in the peak of summer heat in Texas. Each species was selected to perform a specific function within the system. Results from land equivalent ratio (LER) indicate that the within-row combination with peanut, watermelon and okra (Wpwo) and peanut, watermelon, okra and cowpea (Wpwoc) consistently overyielded in 2011 and 2012. LER values were 1.17 each for Wpwo and Wpwoc in 2011 and 1.17 and 1.20 in 2012, respectively. In 2011, watermelon was the dominant crop and was up-regulated in all intercropping combinations while all other component crops were down-regulated. Watermelon per plant production was significantly higher in the combination containing all species (Wall) when compared to its monoculture, 5.50 and 2.09 kgfruit plant−1, respectively. In 2012, okra was the dominant crop and was up-regulated in all intercropping combinations while watermelon, cowpea, and pepper were down-regulated. Okra per plant production was significantly higher in Wpwoc and Wall than in monoculture, 2.28, 2.46, and 1.13 kgfruit plant−1, respectively. These findings suggest that three and four species intercropping combinations, whereby each crop is selected to perform a specific function within the system, may provide small-scale sustainably-minded producers a model system that can be utilized in suboptimal conditions and allow them to reduce inputs while increasing overall yields.

Objective: The aim of this study was to investigate the relationship between herpesvirus-associated ubiquitin-specific protease (HAUSP A/G, rs1529916), tumor protein p53 (TP53 Arg/Pro, rs1042522), leukemia inhibitory factor (LIF G/T, rs929271), glycoprotein 130 (gp130 A/T, rs1900173) and vascular endothelial growth factor (VEGF G/A, rs1570360) polymorphisms and recurrent implantation failure (RIF) in Brazilian women. Subjects and Methods: A total of 120 women with RIF (i.e. those with ≥5 cleaved embryos transferred and a minimum of 2 failed in vitro fertilization/intracytoplasmic sperm injection attempts) were included. The control group involved 89 women who had experienced at least 1 live birth (without any infertility treatment). DNA was extracted from the peripheral blood of all participants, and the abovementioned single-nucleotide polymorphisms (SNPs) were genotyped by real-time polymerase chain reaction. The data were evaluated using Fisher's test. Results: A significant difference between the RIF and control groups was found in the VEGF gene where the GG genotype showed a 2.1-fold increased chance of not being included in the RIF group, while the presence of an A allele increased this risk 1.6-fold. No significant differences were found for the other polymorphisms. Conclusion: This study showed an association between the VEGF -1154G/A polymorphism and RIF in Brazilian women.

OBJECTIVE KPIs have been employed for internal quality control (IQC) in ART. However, clinical KPIs (C-KPIs) such as age, AMH and number of oocytes collected are never added to laboratory KPIs (L-KPIs), such as fertilization rate and morphological quality of the embryos for analysis, even though the final endpoint is the evaluation of clinical pregnancy rates. This paper analyzed if a KPIs-score strategy with clinical and laboratorial parameters could be used to establish benchmarks for IQC in ART cycles. METHODS In this prospective cohort study, 280 patients (36.4±4.3years) underwent ART. The total KPIs-score was obtained by the analysis of age, AMH (AMH Gen II ELISA/pre-mixing modified, Beckman Coulter Inc.), number of metaphase-II oocytes, fertilization rates and morphological quality of the embryonic lot. RESULTS The total KPIs-score (C-KPIs+L-KPIs) was correlated with the presence or absence of clinical pregnancy. The relationship between the C-KPIs and L-KPIs scores was analyzed to establish quality standards, to increase the performance of clinical and laboratorial processes in ART. Objective This study aimed to determine what Brazilian childless women of reproductive age think about oocyte cryopreservation to postpone pregnancy and their reasons for performing or not performing this procedure. Methods Women of reproductive age were randomly selected from the general population using different e-mail lists and were invited to participate in the study by completing an online web survey regarding social oocyte cryopreservation. The survey was also distributed through social media to women of reproductive age. Results Although most of the responders had a partner (86.9%) and had already planned the pregnancy of their first child (69.6%), 85.4% (379) considered the potential of social oocyte freezing to improve their chances of giving birth later in life. Those that had already planned pregnancy were two times more likely to intend to freeze their oocytes (p=0.03). The most important barrier for not undergoing oocyte cryopreservation was cost. The women who indicated that they could not currently undergo the procedure now because of cost were two times (p=0.03) more likely to intend to cryopreserve their oocytes than women who thought that they would not need to delay pregnancy. Conclusion Brazilian women who think that they are not ready to have a family are discovering the option of oocyte cryopreservation. Most participants considered safeguarding their reproductive potential. Making the procedure more accessible could give women the opportunity to make proactive decisions about the future of their fertility.

Objective To evaluate varicocele effects on conventional semen parameters: sperm DNA fragmentation, chromatin packaging, mitochondrial membrane potential (MMP) and sperm apoptosis. Methods A cross-sectional study was carried out with semen samples from 2,399 men of couples who attended an infertility clinic. A total of 16.3% (391/2399) of the men were diagnosed with varicocele by a urologist. Results A regression analysis revealed that the percentages of sperm with DNA fragmentation, abnormal chromatin packaging, and abnormal MMP were significantly increased in individuals with varicocele, when compared to men without varicocele. Apoptosis was not influenced by varicocele. Conventional semen parameters were significantly worse in individuals with the disease. On the other hand, in men with varicocele, Spearman's correlation demonstrated that early apoptosis and abnormal MMP showed a positive and significant correlation with sperm DNA fragmentation. Conclusion Men with varicocele had worse semen parameters, including increased levels of sperm DNA fragmentation, inactive mitochondria, and abnormal chromatin packaging. These changes are possible causes of infertility in individuals with varicocele.

Methods Cross-sectional study of 400 elderly patients’ admission to a general hospital or nursing home diagnosed with delirium, SSD, dementia, or no-delirium/no-dementia (NDND). Symptom profiles were assessed using the Delirium Rating Scale-Revised-98 (DRS-R98). Results Twenty percent patients had delirium, 19.3% had SSD, 29.8% had dementia-only, and 31% had NDND. Eighty-one percent of subsyndromal and 76% of delirium groups had comorbid dementia. DRS-R98 scores showed ascending severity from NDND < dementia-only < SSD < delirium. DRS-R98 scores for items evaluating the three core symptom domains (cognitive, higher-order thinking, and circadian) distinguished SSD from delirium and both from nondelirium groups. DRS-R98 profiles were essentially the same in delirium and SSD subgroups with or without dementia, although total scale scores were generally higher when in comorbid subgroups. Discussion SSD shared characteristic core domain symptoms with delirium, which distinguished each from nondelirium groups, although severity was intermediate in the subsyndromal group. Delirium core symptoms overshadowed the dementia phenotype when comorbid. Milder disturbances of delirium core domain symptoms are highly suggestive of SSD.

MÉTODOS: Estudo transversal retrospectivo no qual foram incluídas 60 mulheres obesas com fenótipo clássico da SOP - Consenso de Rotterdam - e 70 obesas sem SOP. A SM foi diagnosticada pelos critérios do NCEP-ATP III. A obesidade foi definida pelo índice de massa corpórea e o hirsutismo, pelo Índice de Ferriman-Gallwey (IFG). As dosagens realizadas foram: testosterona total, sulfato de dehidroepiandrosterona (SDHEA), insulina e glicose, colesterol total, HDL e triglicerídios. A resistência insulínica (RI) foi avaliada pelo HOMA-IR e pelo índice de sensibilidade à insulina de Matsuda e De Fronzo. A analise estatística foi realizada com o teste t de Student, teste do χ² e análise de regressão logística multivariada (p0,05). CONCLUSÃO: Mulheres obesas com SOP apresentam maior frequência de SM quando comparadas às obesas não SOP. O hiperandrogenismo não mostrou influência nesse grupo de mulheres estudadas. PURPOSE: To assess the contribution of hyperandrogenism to the development of metabolic syndrome (MetS) in obese women with polycystic ovary syndrome (PCOS). METHODS: Retrospective cross-sectional study conducted on 60 obese women with classic PCOS phenotype - Rotterdam Consensus - and 70 non-PCOS obese women. MetS was diagnosed by the NCEP-ATP III criteria and obesity was defined by body mass index. The Ferriman-Gallwey score (mFG) was used to evaluate hirsutism. The following measurements were performed: total testosterone, dehydroepiandrosterone sulfate (DHEA-S), glucose and insulin, total cholesterol, HDL, and triglycerides. Insulin resistance was measured using the HOMA-IR and insulin sensitivity index of Matsuda and De Fronzo (ISI). Statistical analysis was performed using the Student's t-test, χ² test and multivariate logistic regression analysis (p0.05). CONCLUSION: Obese women with PCOS have a higher frequency of metabolic syndrome than non-PCOS obese women, and hyperandrogenism does not contribute to the development of metabolic syndrome in this group of women.
