Prebiotics are non-digestible molecules produced by probiotic microorganisms [1]. Probiotic microrganisms are generally bacteria or fungi recognized as safe, with their properties based on the production of organic acids, reduction of biogenic amines, digestion/breakdown of carbohydrates and proteins, immunomodulatory and anti-inflammatory responses, reduction of carcinogenic amines, and production of antimicrobial peptides, among others [2]. These days probiotics are mostly consumed as probiotic yogurts and other probiotic dairy products, dietary supplements, spoonable forms, and probiotic cultured drinks for daily dosage packaging, among others. Prebiotics are also claimed to enhance wellbeing through immunomodulatory and metabolic activities, and act as a natural barrier against pathological processes [1]. These molecules are considered to be a targeted for human and animal production and health, and represents a multimillionaire market of the functional foods. Furthermore, the increasing market of prebiotics counts today with a thousands of patented invention, related to isolation, production, preparation, methods of use, or application of newly health enhancing molecules. The global production and consumption of functional foods is a multi-billion industry, with an estimated market size around US$ 60 billion in 2008-9, several times greater than the health treatment costs only in USA in that years, in the order of US$ 832 million (Figure 1). As a comparison, the global market of probiotic products was US$ 15.9 billion in 2008 and US$ 19 billion in 2009, with a compound annual growth rate (CAGR) of 11.7 % (2009-2014). Furthermore, the probiotic market predicted by 2014 for Europe and Asia comprises, respectively, US$ 12.9 billion (11.1 % CAGR), and US$ 8.7 billion. Japan, a global leader of functional foods, devoted US$ 4.5 billion to the study and commercialization of prebiotics, with US$ 1.5 billion verted exclusively for the oligosaccharide commerce in 2009 [3]. The USA have occupied the second position in the last decade, with a commercialization of US$ 110 million for functional oligosaccharides (35 % inulin, 20 % mannan oligosaccharides, and 10 % fructan), and with a CAGR rate of 20 % The European and the U.S. market for prebiotics is projected to reach nearly US$ 1.2 billion and US$225 million, respectively, by the year 2015 [3]. This has reached nearly US$ 21.6 billion in 2010 and is expected to reach US$ 31.1 billion in 2015, and at a CAGR of 7.6 % for the 5-year period.

This work describes the design of a simple and easy-to-perform laboratory class in Biochemistry course. This practice consists of utilizing a spectrophotometer working in the range of visible light to obtain the association constant of the complex, obtained by the equilibrium of the ligand interaction (phenol red) with a protein (Bovine Serum Albumin, BSA). The parameter allows the calculation of the Gibbs energy variation of this interaction, and its comparison to the literature. This is an opportunity for the students to experience, in practice, abstract concepts that are difficult to understand, such as the listed thermodynamics quantities. Including the data acquisition and parameters quantification, this class can be performed in 60 minutes, the same duration as a usual class.

Este trabalho descreve a elaboração de uma aula prática simples e de fácil realização para uso em aulas de Bioquímica. A prática consiste na utilização de um espectrofotômetro na faixa de luz visível para obtenção da constante de associação do complexo formado pelo equilíbrio da interação de um ligante (vermelho de fenol) com uma proteína (albumina sérica bovina, BSA). O parâmetro permite o cálculo da variação de energia de Gibbs da interação, e sua comparação com a literatura, permitindo ao aluno vivenciar na prática conceitos abstratos de difícil apropriação, tais como as quantidades termodinâmicas elencadas. A prática pode ser aplicada no período convencional de uma aula prática (60 min), incluindo a coleta de dados e quantificação dos parâmetros.

International authorities classify the ricin toxin, present in castor seeds, as a potential agent for use in bioterrorism. Therefore, the detection, identification, and characterization of ricin are considered the first actions for its risk assessment during a suspected exposure, parallel to the development of therapeutic and medical countermeasures. In this study, we report the kinetic analysis of electro-oxidation of adenine released from hsDNA by the catalytic action of ricin by square wave voltammetry. The results suggest that ricin-mediated adenine release exhibited an unusual kinetic profile, with a progress curve controlled by the accumulation of the product and the values of the kinetic constants of 46.6 µM for Km and 2000 min−1 for kcat, leading to a catalytic efficiency of 7.1 × 105 s−1 M−1.

International authorities classify ricin toxin present in castor seed as a potential agent for use in bioterrorism. Therefore, the detection, identification, and characterization of ricin in various sample matrices are considered necessary actions for risk assessment during a suspected exposure. This study reports a portable electrochemical assay for detecting active ricin based on the adenine electro-oxidation released from herring sperm DNA substrate by its catalytic action. Also, kinetic parameters were calculated, and the values were Km of 3.14 µM and Kcat 2107 min−1. A linear response was found in optimized experimental conditions for ricin concentrations ranging from 8 to 120 ng/mL, and with a detection limit of 5.14 ng/mL. This proposed detection strategy emphasizes the possibility of field detection of active ricin in food matrices and can be applied to other endonucleolytic activities.

Kefir, a combined consortium of bacteria and yeast encapsulated by a polymeric matrix of exopolysaccharides, was used as anodic biocatalyst in a two-chamber microbial fuel cell (MFC). Fermentation was followed during 72 h and polarization curves were obtained from linear sweep voltammetry. The effect of methylene blue as charge-transfer mediator in the kefir metabolism was evaluated. UV/Vis spectrophotometry and cyclic voltammetry were applied to evaluate the redox state of the mediator and to characterize the electrochemical activity, whereas current interruption was used for internal resistance determination. Aiming to establish a relationship between the microbial development inside the anodic chamber with the generated power in the MFC, total titratable acidity, pH, viscosity, carbohydrate assimilation, and microbial counting were assayed. The kefir-based MFC demonstrated a maximum power density of 54 mW m−2 after 24 h fermentation, revealing the potential use of kefir as a biocatalyst for microbial fuel cells.

The development of a low-cost and single chamber solid-state cathode microbial fuel cell is the main objective of this work. The architecture was built using hydraulic connections, an agar-KCl salt bridge, and Saccharomyces cerevisiae as the anodic bioelectrocatalyst. A Prussian blue film electrodeposited on carbon mesh support was employed as a cost-effective, solid-state cathode. Scanning electron microscopy images and energy dispersive spectra confirm the deposition of the Prussian blue on the cathode material. Cyclic voltammetry measurements of the modified electrode indicate a diffusion-limited process due to the intercalation of potassium ions in the Prussian blue structure. Oxi-reduction reactions leading by hydrogen peroxide allow the reactivation and renewal of the electrode after the operation of the microbial fuel cell. Polarization curves exhibit maximum power densities of 136 mW m−2 using lyophilized yeast or 67 mW m−2 using a brewery effluent, which demonstrates the feasibility for bioelectricity generation from this kind of wastewater. The single chamber microbial fuel cell under US$ 1 and constructed with hydraulic couplers can be an alternative for fuel cells of similar architecture.

A α-amilase e uma enzima chave na producao das cervejas por ser responsavel pela clivagem do amido em acucares fermentaveis. Durante o processo de preparacao do mosto que ocorre de 40 a 80°C, essa enzima pode ser afetada por polifenois presentes na mistura. O objetivo foi avaliar a cinetica e o equilibrio da interacao de α-amilase com polifenois usualmente presentes em cerveja e nas faixas de temperatura empregadas no processo. Para tanto, polifenois comumente encontrados em cerveja (acido clorogenico, acido cafeico, acido ferulico e quercetina) foram incubados com α-amilase em 298, 313 e 323K, a atividade da enzima foi determinada por absorcao molecular pelo metodo de triiodina, e os resultados tratados pela equacao integrada de Michaelis-Menten. O equilibrio quimico da interacao foi abordado por titulacao espectrofluorimetrica em 293, 303, 313, 323 e 333 K. Os resultados apresentaram valores dos parâmetros cineticos de α-amilase a 323K de Km 1,37 ±0,19 mg.mL-¹ e Vmax 0,66 ±0,08 mg.mL-¹.min-¹, e um modelo de inibicao competitiva apresentando Ki entre 2,00x10-5 ±4,50x10-7 mol.L-¹ e 4,52x10-5 ±2,24x10-6 mol.L-¹. Foram encontrados valores de energia de ativacao da enzima para acido clorogenico > acido ferulico > acido cafeico > quercetina. O acido clorogenico foi o que apresentou maior estabilidade na formacao dos complexos, com menor valor de Ki, maior sensibilidade com a temperatura e maior inibicao na temperatura otima da enzima (323K), antagonicamente aos valores apresentados para a quercetina. Os resultados da interacao ligante-enzima revelaram valores de Kb crescente com a temperatura, sugerindo maior estabilidade na formacao do complexo proximo a 323 K, e coincidentes com os resultados de inibicao enzimatica. Os valores para ∆H e ∆S sugerem um mecanismo entropicamente dirigido da associacao. Estudo da relacao estrutura-atividade demonstrou uma correlacao positiva entre a atividade biologica (ligacao e catalise) da α-amilase com os descritores de natureza polar dos polifenois (numero total de doadores e aceptores de ligacoes de hidrogenio, polarizabilidade, TPSA, Pka). Ensaios de interacao da enzima com acido clorogenico em diferentes valores de pH e forca ionica apoiam os dados de estrutura-atividade, e sugerem a participacao do ion carboxilato do ligante na interacao com grupos basicos na vizinhanca do sitio catalitico da enzima.

α-amylase is a key enzyme in the production of beer due to the breakdown of starch into fermentable sugars. During the preparation of the brewing mash, the enzyme can be affected by polyphenols present in the mixture. Our aim was to evaluate the kinetics and equilibrium of the interaction of α-amylase with some polyphenols (chlorogenic, caffeic, ferulic acids and quercetin) present in beer in the usual range of mashing temperatures (40 - 80 °C), and to treat the results with integrated Michaelis-Menten and Stern-Volmer equations. The results showed a competitive inhibition model for all compounds with Ki values around 30 umol.L-1. The binding constants (Kb) revealed increasing values with temperature up to 323 K, corroborating the enzymatic data. Values for ΔH and ΔS revealed an entropy-driven association mechanism. Structure-activity relationships demonstrated a positive correlation between the biological activity of α-amylase and the polar nature descriptors of the polyphenols. Binding assays of the enzyme with chlorogenic acid at different pH and ionic strength support the structure-activity and thermodynamic data and suggest a plausible interaction of the carboxylate ion of the ligand with basic groups in the vicinity of the catalytic site of the enzyme.

This paper describes the construction and characterization of a biofuel cell with low-cost materials. The system uses the baker’s yeast (Saccharomyces cerevisiae) as the microorganism responsible for the generation of voltage, whose interaction with the electrode is mediated by methylene blue. Simple experiments are proposed to evaluate the effects of different substrates, inhibitors and cell viability, improving both the knowledge of the system as well as metabolic pathway concepts to the student. The proposed device was able to generate a power density of 41 ± 0.3 mW m−2, similar to those obtained with yeast-based biofuel cells. The low cost and easily acquisition of materials described allow the introduction of biofuel cell theme in different teaching levels, from high school to the college level.

Fructooligosaccharides from chicory (FOSC) are functional prebiotic foods recognized to exert several well-being effects in human health and animal production, as decreasing blood lipids, modulating the gut immune system, enhancing mineral bioavailability, and inhibiting microbial growth, among others. Mechanisms of actions directly on cell metabolism and structure are however little known. In this sense this work was targeted to investigate the interaction of FOSC with biomimetic membranes (liposomes and supported bilayer membrane; s-BLM) through cyclic voltammetry, impedance spectroscopy, spectrofluorimetry, and microscopy. FOSC was able to disrupt the membrane structure of liposomes and s-BLM from the onset of molecular pores induced on it. The mechanism of interaction of fructans with biomimetic membranes suggests hydrogen bonding between the polyhydroxylated structure of the oligosaccharides and the negative polar group of L-α-phosphatidylcholine (PC) present in both liposomes and s-BLM.

Após a água, o café é a bebida mais consumida no planeta, existindo uma demanda crescente por tipos especiais em alguns países importadores, como o café despolpado. Este tipo é obtido pela remoção da mucilagem, ao se submeter o café à fermentação, cujo processo varia com o clima e a microbiota associada, entre outros fatores, podendo levar ao detrimento da qualidade do produto e da saúde do consumidor. Assim, faz-se necessário investigar novas tecnologias que possam auxiliar na obtenção de produtos com melhor padronização, qualidade e que ofereçam maior segurança alimentar. O ozônio e o ultra-som são tecnologias usadas em diversas áreas, com grande flexibilidade e resultados promissores na obtenção e no tratamento de uma infinidade de alimentos. São conhecidas as potentes características oxidantes e microbicidas de ambos, mas seus mecanismos ainda não são bem esclarecidos quando utilizados em tecidos vivos. Seus efeitos sobre os vários parâmetros de qualidade devem ser estudados, compreendidos e ajustados, antes de serem utilizados em larga escala na indústria. No presente trabalho, foi aplicado o ozônio ou o ultra-som como tratamentos prévios à fermentação do café, demonstrando sua eficácia na melhoria da segurança alimentar do café despolpado, sem alteração perceptível da qualidade da bebida.There is a growing interest in technologies that improve food quality, as well as food safety aspects. After water, coffee is the most consumed beverage in the planet, with a rising demand for special types, such as fermented coffee, in some importing countries. This type of coffee is obtained by removing the mucilage, submitting the coffee to fermentation, a process that varies with the climate and the associated natural micro-organisms, among other factors, and that can lead to detrimental effects to the quality of the product and to the health of consumers. Thus, it is necessary to investigate new technologies that can help obtain better standardized products, of higher quality and that offer greater food safety. Ozone and ultrasound are technologies used in various areas, with great flexibility and promising results in the production and treatment of an infinity of food products. The potent oxidant and antimicrobial characteristics of both are well-known, however, their mechanisms are still not clearly understood when used in live tissues. Their effects on the various quality parameters must be studied, understood and adjusted, before widespread use in the food industry. In the present paper, ozone or ultrasound were applied as pre-treatments before coffee fermentation, demonstrating their efficacy in improving food safety of the fermented coffee, without perceptible changes in the quality of the beverage.

Kefir is an association of microrganisms generally grown in milk, with known probiotic activities identified from its soured suspensions. Aqueous media are also able to grow kefir, but little is known about the probiotic properties of its fermented products. This work aimed to evaluate some probiotic properties of a carbohydrate fraction isolated from sugary kefir (sugary kefir carbohydrate [SKC]). Anti-inflammatory activity of the isolated fraction of carbohydrate was tested both in vitro (cellular respirometry and macrophage culture) and in vivo (50% effective dose, rat paw edema, vascular permeability, and cicatrizing test). The results indicated no significant difference for oxygen uptake or macrophage culture between control and test groups. Rat paw edema, however, showed a significant inhibitory activity by 30 +/- 4% and 54 +/- 8% (P.001) for carrageenan and dextran, respectively. In the cicatrizing test, animals treated with SKC cream also presented less trauma after treatments as compared to the negative control group (P.05). The overall data suggested the SKC as a natural product that could be used as a constituent of an anti-inflammatory compound.

No presente trabalho foi avaliado o efeito dos agentes sanificantes dicloroisocianurato de sódio na concentração de 3 a 3,5 mg.L-1 (T1), dicloroisocianurato de sódio associado ao ultra-som (T2), ozônio na concentração de 3 a 3,5 mg.L-1 (T3), ozônio associado ao ultra-som (T4) e controle (T5) sobre a composição centesimal, perfil de ácidos graxos (AG), teor de colesterol e índice de peróxido de filés de tilápia. Os tratamentos afetaram (p < 0,05) a composição centesimal. Os filés foram tratados por imersão, durante 20 minutos, em soluções que apresentavam pH 6,0 e temperatura de 5 °C. Os sanificantes causaram uma perda de água e aumentaram a concentração de proteínas no filé, tendo esse fato sido notado mais acentuadamente no T3 (77,16% de umidade e 19,73% de proteína) quando comparado ao T5 (78,60% de umidade e 18,71% de proteína). Foi demonstrada diferença significativa (p < 0,05) na fração lipídica para ácidos graxos saturados (AGS), ácidos graxos monoinsaturados (AGM) e ácidos graxos polinsaturados (AGP) entre os tratamentos propostos. Os AGS apresentaram diferença entre os T3, T4 e T5, com as respectivas médias 37, 33,50 e 36,30%. Os T1 e T2 (34,90 e 34,80%) não diferiram entre si, mas foram diferentes dos T3, T4 e T5 (39,30, 36,30 e 39,10%) para AGM. O T3 foi o que mais preservou a fração AGM. Os AGP não apresentaram diferença entre os T1 e T2 (23 e 22,50%) e os grupos T3, T4 e T5 (18,30, 20,40 e 17,70%) diferiram entre si, tendo o T1 apresentado as maiores médias. Obteve-se uma redução mais acentuada do nível de colesterol no grupo T2 (43,94 mg.100 g-1) quando comparado com T5 (54,77 mg.100 g-1). Os teores médios de peróxido encontrados nos filés submetidos aos tratamentos não revelaram diferença estatística segundo análise de variância (p > 0,05), apesar dos valores serem bastante diferentes, apresentando uma variação de 6,07 a 39,57 meq.kg-1. The present research had as objectives to study the effects of chlorinated water (sodium dichloroisocyanurate - 3 to 3.5 mg.L-1) (T1); chlorinated water associated to ultrasound (T2); ozonated water (3 to 3.5 mg.L-1) (T3); ozonated water associated to ultrasound (T4), and control (T5) on the centesimal composition, fatty acid profile, cholesterol content and peroxide value in "tilápia" fillets. The fillets were treated by immersion in the solutions, at pH 6.0, 5 °C, for 20 minutes. The treatments affected the centesimal composition. Sanitizers caused a loss of water and increased the concentration of protein in the fillets, more strongly in T3 (77.16% moisture and 19.73% protein), when compared to T5 (78.60% moisture and 18.71% protein). Significant differences were observed for saturated fatty acids (SFA), monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (PUFA). SFA presented differences in T3, T4 and T5, with the respective averages of 37, 33.50 and 36.30%. T1 and T2 (34.90 and 34.80%) did not present differences, but were different from T3, T4 and T5 (39.30, 36.30 and 39.10%) for MUFA. T3 preserved more fractions of MUFA. PUFA did not differ in T1 and T2 (23 and 22.50%) and T3, T4 and T5 (18.30, 20.40 and 17.70%) differed from eachother, with T1 presenting the highest averages. There was a marked reduction in the level of cholesterol in T2 (43.94 mg.100 g-1), when compared to T5 (54.77 mg.100 g-1). There was no statistical difference in peroxide level among the treatments.

Kefir is a fermented beverage originating form the Caucasian regions composed of a number of bacteria and yeasts living together in polysaccharide grains secreted by them. Kefir can be considered a probiotic source as it presents anti-bacterial, anti-mycotic, anti-neoplasic and immunomodulatory properties. Aiming to appraise a possible anti-inflammatory effect of kefir we conducted cotton-induced granuloma and paw oedema assays in rats, the latter using carrageenan, dextran and histamine as stimuli. Kefir samples were thawed and continuously cultured during 15 days both in a molasses solution (50 g/l) and in cow's milk. A polysaccharide extract isolated from the grains (kefiran) was also tested in cotton-pellet experiments. The results showed significant inhibition in the formation of granuloma tissue for all the test groups, as compared to the blank group. Kefir suspensions in molasses presented an inhibition of 41 +/- 3% for the inflammatory process, fermented milk prepared from kefir showed 44 +/- 6% inhibition and kefiran extract 34 +/- 15%. Rat paw oedema also showed significant decreases with the mediators. Dextran-induced oedema was completely inhibited at 1 h after input, with a 76% inhibition after 2 h. Carrageenan stimulus was inhibited 62% after the 3rd hour, and histamine by 52% after the 2nd hour. These results points to the existence of anti-inflammatory prebiotic compounds present in symbiotic cultures of kefir growing in both aqueous and milky suspensions.

The ozonised sunflower oil, Bioperoxoil®, was tested for its antimicrobial activity against some pathological strains in vitro together with its healing potential against Staphylococcus aureus in vivo. Bioperoxoil® was tested against S. aureus, Pseudomonas aeroginosa, Candida albicans, S. typhimurium and Escherichia coli suspensions using the agar diffusion method. Healing experiments were carried out with Wistar rats through topical application of 3.5 mg/ml of the ozonised oil up to the 7th day after inoculation with S. aureus. Bioperoxoil® showed anti-inflammatory effects against all strains tested, with MIC values ranging from 2.0 to 3.5 mg/ml. Bioperoxoil® also demonstrated protective effects on skin connective tissue and to enhance wound healing during the treatment, as compared to a neomycin–clostebol association used as a positive control. The overall results indicated a significant antimicrobial activity, anti-inflammatory and wound-healing properties for Bioperoxoil®, as compared to other antimicrobial agents commercially available.

Esse trabalho descreve a construção de uma biocelula a combustível a partir de materiais de baixo custo e seu uso em aulas práticas de graduação. O sistema emprega a levedura de padaria (Saccharomyces cerevisiae) como micro-organismo responsável pela geração de potencial, cuja interação com o eletrodo é mediada por azul de metileno. Foram propostos experimentos simples para avaliar os efeitos de diferentes substratos, inibidores e viabilidade celular, que permitiram maior conhecimento do sistema bem como conceitos relacionados a vias metabólicas. Conceitos relacionados às biocélulas a combustível tais como aplicações, mecanismos e limitações foram trabalhados em duas aulas práticas de bioquímica com estudantes do Curso de Graduação em Química da Universidade Federal de Alfenas (UNIFAL-MG).

Myoglobin was immobilized with poly(methyl methacrylate)-block-poly[(2-dimethylamino)ethyl methacrylate]PMMA-block-PDMAEMA polymer synthesized by reversible addition-fragmentation chain transfer technique (RAFT). Cyclic voltammograms gave direct and slow quasireversible heterogeneous electron transfer kinetics between Mb-PMMA-block-PDMAEMA modified electrode and the redox center of the protein. The values for electron rate constant (Ks) and transfer coefficient (α) were0.055±0.01·s−1and0.81±0.08, respectively. The reduction potential determined as a function of temperature (293–328 K) revealed a value of reaction center entropy ofΔS0of351.3±0.0002 J·mol−1·K−1and enthalpy change of-76.8±0.1 kJ·mol−1, suggesting solvent effects and charge ionization atmosphere involved in the reaction parallel to hydrophobic interactions with the copolymer. The immobilized protein also exhibits an electrocatalytical response to reduction of hydrogen peroxide, with an apparentKmof114.7±58.7 μM. The overall results substantiate the design and use of RAFT polymers towards the development of third-generation biosensors.

O cogumelo tibetano (CT) é uma bebida probiótica fermentada por um grande conjunto de bactérias e leveduras, algumas das quais de conhecida atividade antimicrobiana, imunomodulatória, pró-digestiva e auxiliar da peristalse intestinal. Neste sentido, este trabalho teve como objetivo avaliar a possível ação da suspensão produzida pelo CT sobre a motilidade intestinal de ratos. Foram utilizados grupos de animais (ratos Wistar albinos machos, n = 6/grupo) pesando 200 ± 20 g. Os diferentes grupos foram tratados v.o. durante 15 dias (1,5 mL/animal de fermentado de 24 h de CT em cultivo contínuo em 50 g/l de açúcar mascavo comercial em água destilada), e os demais grupos tratados com água destilada (1,5 mL/animal). Após 15 dias os animais foram mantidos em jejum por 24 hs, e tratados com suspensão de CT (1,5 mL/animal/v.o), água destilada (controle, 1,5mL/animal/v.o), acetilcolina (1mg/kg/s.c.) e atropina (1mg/kg/i.m.). Após 30 min, os animais receberam uma suspensão de carvão ativo 10 % (0,1 mL/10 g, v.o), seguindo-se eutanásia após 45 min, excisão da porção do piloro até o início do ceco, e determinação da distância percorrida pelo carvão. A administração da suspensão de CT aumentou o trânsito intestinal em 68,7 % e os demais tratamentos apresentaram as seguintes percentagens: atropina 20,2 %, acetilcolina 78,7 % e controle 42,9 % (p < 0,05, ANOVA seguido de teste de Tukey-Kramer). Esses resultados mostraram que a suspensão de CT estimulou o peristaltismo nos animais, o que sugere um efeito positivo na digestão e absorção alimentar para o consórcio microbiano. The Tibetan mushroom (CT) is a probiotic beverage fermented by a big group of bacteria and yeasts, some of which of well-known anti-bacterial activity, immunomodulatory, pro-digestive and to aid of the intestinal peristalsis. In this sense, this work had the aim of evaluating the possible action of the suspension produced by CT on the peristalsis intestinal of rats. Animal groups weighing 200 ± 20 g were used (Wistar rats (male, n = 6/group)). The different groups were treated v.o. for 15 days (1,5 mL/animal of 24h fermented of CT in continuously cultured into molasses solutions (50 g/l)), and the other groups treated with distilled water (1,5 mL/animal). After 15 days the animals were maintained in fast for 24h, and treated with suspension CT (1,5 mL/animal/v.o), distilled water (control, 1,5mL/animal/v.o), acetylcholine (1mg/kg/s.c) and atropine (1mg/kg/i.m). After 30 min, the animals received a suspension of active coal 10% (0,1 mL/10g, v.o), after the euthanasia, excision of the portion of the pylorus to the beginning of the cecum, and determination of the distance traveled by the coal. The administration of the CT suspension increased the intestinal peristalsis in 68,7% and the other treatments presented the following percentages: atropine 20,2%, acetylcholine 78,7% and control 42,9% (p < 0,05, ANOVA followed by Tukey-Kramer test). Those results showed that the CT suspension stimulated the peristalsis in the animals, what suggests a positive effect in the digestion and alimentary absorption for the microbial consortium.

This study evaluated the inhibitory activity of copaiba oil (Copaifera officinalis against the cariogenic microorganism, Streptococcus mutans. For such purpose, a minimum inhibition concentration test of copaiba oil against S. mutans was performed, using the serial dilution in broth technique, with a negative control, a positive control (0.12% chlorhexidine) and a 10% copaíba oil solution as a test. A minimum bactericidal concentration test with tubes presenting microbial inhibition was also conduced. In the minimum inhibitory concentration test, copaiba oil showed inhibition of bacterial growth at all concentrations tested up to 0.78 µL/mL of the 10% copaiba oil solution in the broth. In addition, the negative control had no inhibition, and the 0.12% chlorhexidine solution was effective up to 6.25 µL/mL in the broth. Copaiba oil showed a bacteriostatic activity against S. mutans at low concentrations, and could be a an option of phytotherapic agent to be used against cariogenic bacteria in the prevention of caries disease. Este estudo avaliou a atividade inibitória do óleo de copaíba (Copaifera officinalis contra o microrganismo cariogênico, Streptococcus mutans. Para isso, foi realizado um teste de concentração mínima inibitória do óleo de copaíba contra S. mutans, utilizando a técnica de diluição seriada em caldo, com um controle negativo, um controle positivo (clorexidina a 0,12%) e uma solução de óleo de copaíba 10% como teste. Também foi conduzido um teste de concentração mínima bactericida com os tubos que apresentaram inibição microbiana. No teste de concentração inibitória mínima, o óleo de copaíba mostrou inibição do crescimento bacteriano em todas as concentrações testadas até 0,78 µL/mL da solução a 10% do óleo de copaíba no caldo. Além disso, o controle negativo não teve nenhuma inibição, e a solução de clorexidina 0,12% foi eficaz até 6,25 µL/mL no caldo. O óleo de copaíba mostrou uma atividade bacteriostática contra S. mutans em baixas concentrações, apresentando-se assim como uma opção de fitoterápico a ser utilizado contra bactérias cariogênicas na prevenção de cáries.

Probiotics are supplementary foods developed by microbial strains that improve animal health beyond basic nutrition. Probiotics are consumed orally, regardless of being considered as normal inhabitants of the intestines, able to survive in enzimatic and biliary secretions. Kefir is a probiotic originated from the old continent, fermented by several bacteria and yeasts, encapsulated in a polyssacharide matrix, and resembles jelly grains. Kefir is also presented as its sourish product both in sugary or milky suspensions containing vitamins, aminoacids, peptides, carbohydrates, ethanol, and volatile compounds. Kefir is known to have a diverse microbial content depending on the country and fermentative substrates, which cause distinct probiotic effects. In this sense, the purpose of this work was to isolate, identify, and quantify the microbial content of a native sugary kefir sample (fermented suspension and lyophilized natural grains). Serial dilutions were plated on Rogosa agar (AR) and De Man, Rogosa and Sharpe (MRS), for Lactobacillus; Brain Heart Infusion (BHI), for total bacteria; Sabouraud-Dextrose-Agar (SDA), for yeasts and filamentous fungi; Thioglycolate Agar (TA), for Streptococcus, Acetobacteria and Leuconostoc; and Coconut Water Agar (CWA), and CWA supplemented with yeast extract (CWAY), for various genera. Genera and species for all strains were identified through biochemical reactions and specific API systems. The microbial profile of kefir was different from other sources of grains despite the presence of similar microorganisms and others which have not been reported yet. The data obtained with the CWA and CWAE media suggest that both substrates are alternative and salutary media for culture of kefir strains.

O potencial de uso do óleo de copaíba (Copaifera officinalis) na prevenção da doença periodontal, eliminando seu agente etiológico, foi avaliado em 18 cães sem raça definida, distribuídos homogeneamente em três grupos: teste, (contendo óleo de copaíba) controle positivo e controle negativo. Os tratamentos ocorreram três vezes ao dia, durante oito dias. Ao nono dia, os animais receberam aplicação tópica de fucsina básica 0,5% para evidenciação do biofilme. Mudanças na halitose e gengivite foram avaliadas diariamente por inspeção visual. Adicionalmente, foram realizados testes laboratoriais de inibição de aderência de Streptococcus mutans e ensaio antimicrobiano de difusão em ágar, sobre bactérias formadoras de placa dental. Os resultados da placa evidenciada apontaram áreas de cobertura microbiana nos dentes de 53,4±8,8%, 28,5±5,4%, e 22,3±5,3% para os grupos negativo, positivo e teste, respectivamente, indicando diferença entre o controle negativo e os demais grupos (P

Há um crescente interesse em tecnologias que melhorem a qualidade dos alimentos, bem como o aspecto segurança alimentar. Após a água, o café é a bebida mais consumida no planeta, existindo uma demanda crescente por tipos especiais em alguns países importadores, como o café despolpado. Este tipo é obtido pela remoção da mucilagem, ao se submeter o café à fermentação, cujo processo varia com o clima e a microbiota associada, entre outros fatores, podendo levar ao detrimento da qualidade do produto e da saúde do consumidor. Assim, faz-se necessário investigar novas tecnologias que possam auxiliar na obtenção de produtos com melhor padronização, qualidade e que ofereçam maior segurança alimentar. O ozônio e o ultra-som são tecnologias usadas em diversas áreas, com grande flexibilidade e resultados promissores na obtenção e no tratamento de uma infinidade de alimentos. São conhecidas as potentes características oxidantes e microbicidas de ambos, mas seus mecanismos ainda não são bem esclarecidos quando utilizados em tecidos vivos. Seus efeitos sobre os vários parâmetros de qualidade devem ser estudados, compreendidos e ajustados, antes de serem utilizados em larga escala na indústria. No presente trabalho, foi aplicado o ozônio ou o ultra-som como tratamentos prévios à fermentação do café, demonstrando sua eficácia na melhoria da segurança alimentar do café despolpado, sem alteração perceptível da qualidade da bebida. There is a growing interest in technologies that improve food quality, as well as food safety aspects. After water, coffee is the most consumed beverage in the planet, with a rising demand for special types, such as fermented coffee, in some importing countries. This type of coffee is obtained by removing the mucilage, submitting the coffee to fermentation, a process that varies with the climate and the associated natural micro-organisms, among other factors, and that can lead to detrimental effects to the quality of the product and to the health of consumers. Thus, it is necessary to investigate new technologies that can help obtain better standardized products, of higher quality and that offer greater food safety. Ozone and ultrasound are technologies used in various areas, with great flexibility and promising results in the production and treatment of an infinity of food products. The potent oxidant and antimicrobial characteristics of both are well-known, however, their mechanisms are still not clearly understood when used in live tissues. Their effects on the various quality parameters must be studied, understood and adjusted, before widespread use in the food industry. In the present paper, ozone or ultrasound were applied as pre-treatments before coffee fermentation, demonstrating their efficacy in improving food safety of the fermented coffee, without perceptible changes in the quality of the beverage.

Kefir, a symbiont microorganism suspension, presents benefic effects to health. Some kefir grains were cultivated in brown sugar, allowing to isolate a substance named CSQ. This was evaluated on a biologic essay of mouse foot edema, presenting an inhibitory activity of 30+4 % against carrageenan after the stimulus. It was observed that a cultivation mean containing sucrose, and not the milky mean, lead to the production of different sugar polymeric chains of kefir. The results in vivo suggest that the CSQ exerted an anti-inflammatory activity.

The crystal structure of '2,2',4-trihydroxybenzophenone' (=(2,4-dihydroxyphenyl)(2-hydroxyphenyl)methanone; 1) was determined, and its molecular structure, along with intra- and intermolecular H-bonds, was analyzed. The anti-inflammatory potential of 1, evaluated by means of the rat-paw-edema assay, with carrageenan as inflammation stimulus, was found to be similar high as that of indomethacin. In contrast, benzophenone proper (2) was hardly active in this assay. Our results indicate that these anti-inflammatory effects are related to the action of kinins and prostaglandins. The radical-scavenging properties of 1 towards DPPH were found to be similar as those of typical phenolics, but somewhat lower than that of ascorbic acid. The structure-activity relationship (SAR) of 1 is discussed.

Kefir and its insoluble polysaccharide, kefiran, were both tested for antimicrobial and cicatrizing activities against several bacterial species and Candida albicans using an agar diffusion method. Comparator antimicrobials were also tested. Cicatrizing experiments were carried out on Wistar rats with induced skin lesions and Staphylococcus aureus inoculation, using a topical application of a 70% kefir gel. Both kefir and kefiran showed some activity against all organisms tested; the highest activity was against Streptococcus pyogenes. Cicatrizing experiments using 70% kefir gel had a protective effect on skin connective tissue and 7 days treatment enhanced wound healing compared with 5 mg/kg of neomycin-clostebol emulsion.

To investigate the antiulcer and antiinflammatory activities of the essential oil from Casearia sylvestris leaves (EOCS) the following tests were used: rat paw edema, granulomatous tissue test, vascular permeability, writhing test, gastric ulcer stress-induced and evaluation of gastric secretion (pylorus ligation test). The total yield of EOCS was 2.5% with LD50 of 1100 mg/kg in mouse. The major compounds identified using gas chromatography were caryophyllene, thujopsene, alfa-humulene, beta-acoradiene, germacrene-d, bicyclogermacrene, calamenene, germacrene B, spathulenol and globulol. The EOCS orally administered to the rats at 125 mg/kg resulted 36% of inhibition in carrageenan-induced edema in the rat paw assay (p0.05, Student's t-test). However, both rat paw edema dextran-induced and vascular permeability assay using histamine showed no significant inhibition. Mice submitted to the writhing test using acetic acid presented 58% and 56% of inhibition in writhes with EOCS and indomethacin, respectively. Furthermore, EOCS inhibited 90% of stress-induced gastric ulcer, while cimetidine inhibited 70% (p0.05, Student's t-test). The volume of gastric secretion in the group treated with EOCS was greater than the group treated with cimetidine. The EOCS also showed no changes related to H+ levels in the gastric secretion. With the overall results obtained in this study the authors suggest an anti-inflammatory activity for the EOCS of Casearia sylvestris beyond its anti-ulcer activity.

Tibetan mushroom (TM) is a fermented beverage composed by a dozen of bacteria and yeasts living together into polysaccharide grains secreted by them. TM is similar to kefir, a probiotic beverage originated in the Caucasian mountains exhibiting some anti-bacterial, anti-mycotic, anti-neoplastic and immunomodulatory effects. Aiming to evaluate a plausible anti-inflammatory property of TM we conducted cotton-induced granuloma and paw edema assays in rats, the latter using carrageenin, dextran and histamine as stimuli. TM samples were thawed and continuously cultured during 15 days into molasses solutions (50 g/l). The experiments used TM suspensions after 24h fermentation and TM grains mechanically disintegrated. The results showed a significant inhibition on the formation of granuloma tissue for the test group as compared to the negative control group. TM suspensions presented an inhibition of 43% for the inflammatory process. Rat paw edema also showed significant decreases with the mediators. The edema induced by carrageenin was inhibited 62% at the 3rd hour. The edema dextran-induced was completely inhibited at 1h and antagonized the histamine edema 52% at 1h.

A proposta central desse trabalho foi a construção e caracterização de biocélulas a combustível microbianas (MFC) empregando materiais de baixo custo, novo biocatalisador e novo modelo de arquitetura de uma câmara. Foram desenvolvidos três sistemas distintos, alterando-se o biocatalisador anódico (Saccharomyces cerevisiae ou microbiota de kefir), a arquitetura (uma ou duas câmaras) e o tipo de aceptor de elétrons no cátodo (ferricianeto de potássio ou o composto de coordenação azul da prússia). Todas as MFCs usaram azul de metileno como mediador e foram testadas/caracterizadas por meio de curvas de potência e polarização. Na primeira parte do trabalho foi feita uma abordagem do uso didático de MFCs com o uso da levedura Saccharomyces cerevisiae e arquitetura de duas câmaras, e propostos experimentos para avaliar o efeito da presença/ausência de mediador, diferentes substratos, inibidores, viabilidade celular e oxigênio sobre a resposta da MFC. Na segunda parte foi empregada a mesma arquitetura para a avaliação do consórcio microbiano kefir como biocatalisador anódico. A fermentação foi monitorada em intervalos de 24h até o tempo de 72h com as técnicas de voltametria linear (para construção das curvas de potência e polarização), voltametria cíclica, interrupção de corrente, espectrofotometria, contagem total de micro-organismos, e medidas dos parâmetros físico-químicos de pH, acidez total titulável, viscosidade e consumo de carboidratos. A última parte do trabalho foi voltada para a arquitetura e o cátodo, na qual foi desenvolvida uma MFC de uma câmara utilizando conexões hidráulicas e um filme de azul da prússia eletrodepositado sobre malha de carbono como aceptor de elétrons. Imagens obtidas por microscopia eletrônica de varredura e espectroscopia de energia dispersiva confirmaram a formação do filme eletroativo. Foi demonstrada a possibilidade de obtenção de energia elétrica a partir de resíduo de cervejaria empregando-se esse terceiro dispositivo. A potência gerada foi usada como parâmetro comum de comparação do desempenho de cada um dos sistemas, obtendo-se os valores máximos de, 41 mWm-2 (duas câmaras, S. cerevisiae), 54 mW m-2 (duas câmaras, cultura de kefir), 136 mW m-2 (uma câmara, S. cerevisiae) e 68 mW m-2 (uma câmara, resíduo de cervejaria). The central proposal of this work was the construction and characterization of Microbial Fuel Cells (MFC) using low cost materials, new biocatalyst and new model of single chamber architecture. Three different systems were developed by altering the anodic biocatalyst (Saccharomyces cerevisiae or kefir microbiota), the architecture (one or two chambers) and the type of electron acceptor at the cathode (potassium ferricyanide or the prussian blue coordination compound). All MFCs used methylene blue as a mediator and were tested / characterized by means of power and polarization curves. In the first part of the work an approach was taken on the didactic use of MFCs with the use of Saccharomyces cerevisiae and two chamber architecture, and proposed experiments to evaluate the effect of presence / absence of mediator, different substrates, inhibitors, cell viability and oxygen over the MFC response. In the second part, the same architecture was used for the evaluation of the microbial consortium kefir as an anodic biocatalyst. The fermentation was monitored at intervals of 24h to 72h with the techniques of linear voltammetry (for the construction of power and polarization curves), cyclic voltammetry, current interruption, spectrophotometry, total microorganism counts, and measurements of the parameters physical-chemical pH, titratable total acidity, viscosity and carbohydrate consumption. The last part of the work was focused on the architecture and the cathode, in which a one chamber MFC was developed using hydraulic connections and a prussian blue film electrodeposited on carbon cloth as an electron acceptor. Images obtained by scanning electron microscopy and dispersive energy spectroscopy confirmed the formation of the electroactive film. It was demonstrated the possibility of obtaining electric energy from the brewery residue using this third device. The power generated was used as a common parameter to compare the performance of each of the systems, obtaining maximum values of 41 mW m-2 (two chambers, S. cerevisiae), 54 mW m-2 (two chambers, kefir culture), 136 mW m-2 (one chamber, S. cerevisiae) and 68 mW m-2 (one chamber, brewery residue).

A α-amilase é uma enzima chave na produção das cervejas por ser responsável pela clivagem do amido em açúcares fermentáveis. Durante o processo de preparação do mosto que ocorre de 40 a 80°C, essa enzima pode ser afetada por polifenóis presentes na mistura. O objetivo foi avaliar a cinética e o equilíbrio da interação de α-amilase com polifenóis usualmente presentes em cerveja e nas faixas de temperatura empregadas no processo. Para tanto, polifenóis comumente encontrados em cerveja (ácido clorogênico, ácido cafeico, ácido ferúlico e quercetina) foram incubados com α-amilase em 298, 313 e 323K, a atividade da enzima foi determinada por absorção molecular pelo método de triiodina, e os resultados tratados pela equação integrada de Michaelis-Menten. O equilíbrio químico da interação foi abordado por titulação espectrofluorimétrica em 293, 303, 313, 323 e 333 K. Os resultados apresentaram valores dos parâmetros cinéticos de α-amilase a 323K de Km 1,37 ±0,19 mg.mL-¹ e Vmax 0,66 ±0,08 mg.mL-¹.min-¹, e um modelo de inibição competitiva apresentando Ki entre 2,00x10-5 ±4,50x10-7 mol.L-¹ e 4,52x10-5 ±2,24x10-6 mol.L-¹. Foram encontrados valores de energia de ativação da enzima para ácido clorogênico > ácido ferúlico > ácido cafeico > quercetina. O ácido clorogênico foi o que apresentou maior estabilidade na formação dos complexos, com menor valor de Ki, maior sensibilidade com a temperatura e maior inibição na temperatura ótima da enzima (323K), antagonicamente aos valores apresentados para a quercetina. Os resultados da interação ligante-enzima revelaram valores de Kb crescente com a temperatura, sugerindo maior estabilidade na formação do complexo próximo a 323 K, e coincidentes com os resultados de inibição enzimática. Os valores para ∆H e ∆S sugerem um mecanismo entropicamente dirigido da associação. Estudo da relação estrutura-atividade demonstrou uma correlação positiva entre a atividade biológica (ligação e catálise) da α-amilase com os descritores de natureza polar dos polifenóis (número total de doadores e aceptores de ligações de hidrogênio, polarizabilidade, TPSA, Pka). Ensaios de interação da enzima com ácido clorogênico em diferentes valores de pH e força iônica apoiam os dados de estrutura-atividade, e sugerem a participação do íon carboxilato do ligante na interação com grupos básicos na vizinhança do sítio catalítico da enzima. α-amylase is a key enzyme in the production of beer due to the hydrolysis of starch into fermentable sugars. During the preparation of its brewing mash (40 to 80 °C), this enzyme can be affected by polyphenols present in the mixture. Our aim was to evaluate the kinetics and equilibrium of the interaction of α-amylase with polyphenols usually present in beer at common temperature range employed in the process. Hence, polyphenols commonly found in beer (chlorogenic acid, caffeic acid, ferulic acid and quercetin) were incubated with α-amylase at 298, 313, 323K, enzyme activities were followed by molecular absorption using the triiodine method, and the results treated by the integrated Michaelis-Menten equation. The chemical equilibrium of the interaction was addressed by spectrofluorometric titration at 298, 303, 313, 323, 333K. The results showed values of the kinetic parameters of α-amylase at 323 K of Km 1.37 ±0.19 mg.mL-¹ and Vmax 0.66 ± 0.08 mg.mL-¹.min-¹, and a competitive inhibition model exhibiting inhibition constants (Ki) between 2.00 x 10 -5 ±4.50 x 10-7 mol.L-¹ and 4.52 x 10-5 ±2.24x10-6 mol.L-¹. Enzyme activation energy values were in order of chlorogenic acid > ferulic acid > caffeic acid > quercetin. Chlorogenic acid showed the highest stability in the complex formation, exhibiting the lowest Ki value, the more pronounced temperature sensitivity and the highest inhibition at optimum temperature (323K), in contrast to the values presented for quercetin. The results of the ligand-enzyme interaction revealed increasing values of the binding constant of the protein with the ligands (Kb) with temperature, suggesting a greater complex stability near 323 K, similar to that found from the enzyme inhibition data. The values for ∆H and ∆S suggest an entropy-driven force present in the association mechanism. The results for structure-activity relationship demonstrated a positive correlation between the biological activity (both binding and catalysis) of the α-amylase with the polar nature descriptors of the polyphenols (total number of hydrogen donors and acceptors of, polarizability, TPSA, pKa). Binding assays of the enzyme with chlorogenic acid at different pH values and ionic strength support the structure-activity results and suggest a plausible interaction of the carboxylate ion of the ligand with basic groups in the vicinity of the catalytic site of the enzyme.

O presente trabalho teve por objetivo a obtenção e caracterização de novos SAB através do levantamento de dados termodinâmicos de diferentes sistemas formados por soluções de sulfato de Níquel, Zinco e/ou Cobre com líquido iônico (LI) tetrafluorborato de 1-butil-3-metilimidazolio ([Bmim]BF4) e água. Foram obtidos nove diagramas de fases nas temperaturas de 283,15, 298,15 e 313,15 K. Avaliou-se o efeito das mesmas e como a natureza do sal influencia no equilíbrio líquido-líquido destes SAB. Para o SAB formado por sulfato de cobre observou-se a precipitação do sal com o aumento do comprimento da linha de amarração. Os dados experimentais das curvas binodais foram ajustados pela equação de Merchuk e os dados de equilíbrio líquido-líquido foram correlacionados pelas equações de Othmer-Tobias e Bancroft apresentando valores de R² superiores a 0,9. O efeito salting out no processo de segregação de fases foi avaliado quantitativamente por meio da equação de Setschenow. This study aimed at obtaining and characterization of new SAB by surveying thermodynamic data from different systems formed by nickel sulfate solution, zinc and / or copper with ionic liquid (IL) tetrafluoroborate 1-butyl-3-methylimidazolium ([Bmim] BF4) and water. nine phase diagrams were obtained at temperatures of 283.15, 298.15 and 313.15 K. We evaluated the effect thereof and the nature of the salt influences the liquid-liquid equilibrium of these SAB. For the SAB formed by copper sulfate was observed the precipitation of salt with increasing length of the mooring line. The experimental data of the binodal curves were fitted by equation Merchuk and liquid-liquid equilibrium data were correlated by the equation Othmer-Tobias and Bancroft presenting R2 values ​​higher than 0.9. Eletrodos quimicamente modificados têm sido alvo de estudo na busca por novos e melhores sensores eletroquímicos. Os óxidos condutores transparentes, como o óxido de estanho dopado com flúor (FTO), apresentam condutividade elétrica comparável à dos metais e tem sido estudados como candidatos a novos sensores. Neste trabalho, o FTO foi modificado por eletrodeposição de ouro por meio da técnica de voltametria cíclica, a partir de solução de ácido cloro-áurico. Diferentes eletrodos foram produzidos variando-se o número de ciclos de eletrodeposição. Foram feitas microscopias eletrônicas de varredura, difratometria de raios X e espectroscopias de impedância eletroquímica para se caracterizar os eletrodos quimicamente modificados. A partir dessas análises, foi possível caracterizar os comportamentos resistivos, capacitivos e difusionais dos eletrodos e relacionar essas características com o número de ciclos de eletrodeposição. Dentre os materiais produzidos, aquele com 100 ciclos de eletrodeposição apresentou a maior eficiência como sensor eletroquímico, pois apresentou, entre outras características, a menor impedância global, padrões cristalinos de difração de raios X, aglomerados de tamanhos nanométricos e a maior capacidade de adsorção (rugosidade). Esse eletrodo foi testado quanto à capacidade de detecção dos pares paracetamol/cafeína e ácido úrico/dopamina; os resultados apontam seletividade, diminuição dos potenciais de oxidação das espécies (0,58V e 0,97V vs. ECS para paracetamol e cafeína e 0,22 V e 0,47 V vs. ECS para dopamina e ácido úrico, respectivamente) e baixos limites de detecção (0,82 e 0,052 mol.L-¹ para paracetamol e cafeína e 0,27 e 1,01 mol.L-¹para ácido úrico e dopamina, respectivamente). Chemically modified electrodes have been explored to obtain new and better electrochemical sensors. Transparent conductive oxides, such as fluorine-doped tin-oxide (FTO), show electrical conductivity comparable to metals and are being studied as candidates for new sensors. In this work, FTO was modified by gold electrodeposition using cyclic voltammetry (CV) and a chlorine-auric acid solution. A set of different materials were produced, varying the number of cycles on CV. The chemically modified electrodes were characterized by: scanning electron microscopy, X-ray diffraction and electrochemical impedance spectroscopy. From these results, it was possible to evaluate the resistive, capacitive and diffusional aspects and to make a relationship between these parameters and the number of electrodeposition cycles. Among the produced electrodes, the one with 100 cycles presented the best behavior, the lowest global impedance, crystal patterns from DRX, nanometric clusters and the greater adsorption capacity, showing better quality for an electrochemical sensor. The electrode was tested for detection of paracetamol/caffeine and uric acid/dopamine; the results are good selectivity, decreased oxidation potential (0.58 V and 0.97 V vs. SCE, for paracetamol and caffeine; 0.22 V and 0.47 V vs. SCE, for dopamine and uric acid, respectively) and low detection limits (0.82 and 0.052 mol.L-¹ for paracetamol and caffeine; 0.27 and 1.01 mol.L-¹ for uric acid and dopamine, respectively).

Em várias décadas os Sistemas Aquosos Bifásicos (SAB) tem sido largamente estudados como promissores sistemas para separação, extração, purificação e pré-concentração de analitos diversos, em especial, os de interesse biotecnológicos. Embora esses sistemas sejam fáceis de serem preparados o controle de suas propriedades físico-químicas é dependente de inúmeros fatores e por isso, é essencial o conhecimento do comportamento de fase do SAB, expresso como um diagrama de fase, para sua aplicação. O presente trabalho teve por objetivo o estudo experimental do equilíbrio líquido-líquido em sistemas aquosos bifásicos (SAB) formados por mistura de PEG (4.000, 6.000 e 10.000g mol-¹) + bissulfito de sódio + água em quatro temperaturas variando de 288,15 a 318,15 K. Cada diagrama foi caracterizado por 5 linhas de amarração em cada temperatura. Nos diagramas obtidos foi observado um deslocamento significativo do locus das curvas binodais com a elevação da temperatura, levando a um aumento da região bifásica. Essa característica evidencia um processo de separação de fases endotérmico. O aumento da massa molar do polímero também favorece a formação do SAB. Estudos cinéticos com monitoramento do pH e composição das fases no tempo mostraram que o equilíbrio nos SAB estudados é alcançado com cerca de 3h em repouso no banho termostatizado. No intervalo de 72h do estudo cinético os SAB se mostraram estáveis. Os novos SAB obtidos ainda não foram descritos na literatura e podem apresentar potencial aplicação para processos de separação usando a propriedade de conservante químico do bissulfito de sódio. In several decades the Aqueous Two-Phase Systems (ATPS) have been widely studied as promising systems for separation, extraction, purification and pre-concentration of various analytes, particularly the biotechnological interest. While these systems are easy to prepared control of their physicochemical properties is dependent on many factors and so it is essential to know the phase behavior of the ATPS, expressed as a phase diagram for your application. This study aimed to the experimental study of liquid-liquid equilibrium in aqueous two-phase systems (ATPS) formed by mixing PEG (4,000, 6,000 and 10,000 g mol-¹) + sodium bisulfite + water at four temperatures of 288.15 to 318.15 K. each diagram was characterized by 5 mooring lines at each temperature. In the diagrams obtained was a significant shift in the locus of the binodal curves, leading to an increase of the biphasic region with increasing temperature. This characteristic shows a process of endothermic phase separation. Increasing the molecular weight of the polymer also promotes the formation of SAB. Kinetic studies with pH monitoring and composition of the phases in time showed that the balance in the ATPS studied is reached with around 3 pm at rest in the thermostatic bath. In the 72 hours interval of the kinetic study the ATPS were stable. The new ATPS were not obtained in the literature and may have potential application for separation processes with increased property as a chemical preservative sodium bisulfite

A poluição de corpos d´água por metais pesados é um problema mundial, mas particularmente importante em países que apresentam atividades intensas de mineração, como o Brasil. Os contaminantes, sobretudo aqueles conhecidos como metais pesados, podem apresentar toxicidade altíssima, como é o caso do cromo hexavalente, do cádmio, chumbo e outros. Diante desse quadro, esforços para o aprimoramento de técnicas de remoção de cátions de água são necessárias e podem ter impacto positivo na Saúde Pública e em outras áreas. Diante do exposto o projeto se propõe a produzir materiais poliméricos (orgânicos) sob medida e otimizados para a remoção de cátions de metais de transição de soluções aquosas. Para a síntese desses materiais foram usadas técnicas de polimerização via radical livre e controlada, com o intuito de otimizar a capacidade de remoção. Os polímeros tiveram como base o ácido poli(metacrílico) que tem alto poder de adsorção de cátions. Paralelamente foi criado um sistema de remoção de cátions de solução aquosa usando polímeros confinados em membranas de diálise. A influência da massa molar média, do tipo de síntese e da presença do comonômero metacrilamida foi estudada. Para o estudo da capacidade máxima de remoção de Cu(II) e Cd(II), os parâmetros temperatura, pH da solução, velocidade e tempo de agitação foram fixados. A capacidade máxima de remoção dos íons de Cu(II) variou entre 1,36 mmol.g-1 a 1,65 mmol.g-1, e para os íons de Cd(II) variou entre 1,13 mmol.g-1 a 1,55 mmol.g-1. A partir dos resultados foi possível observar uma relação positiva entre a massa molar média das cadeias e a capacidade de ligação aos cátions metálicos. A presença de metacrilamida na cadeia polimérica teve uma influência negativa na ligação com cádmio, mas pouca influência na ligação com o cobre. A partir desses estudos foi possível concluir que os polímeros sintetizados e imobilizados em sacos de diálise são eficientes na remoção de íons de Cu(II) e de Cd(II) de soluções aquosas. Destaca-se, além da eficiência, a praticidade do uso do método desenvolvido. New methods for heavy metal removal from aqueous solutions are necessary, particularly for regions where mining and other activities involving such toxicants are typical. The use of membrane-based methods in combination with polymers able to bind ions is a very efficient and easy way to remove toxic and undesirable ions from aqueous solutions. Herein, a new method for ion removal is presented. Solutions of different polymers based on polymethacrylic acid (PMAA) were entrapped in a dialysis sack, so that only the small ions can flow through the membrane and bind to the polymer inside. The polymers were synthesized by free radical polymerization (FRP) and the controlled radical polymerization method, RAFT. FRP and RAFT PMAA homopolymers were tested in removal assays as well as a PMAAM-methacrylamide copolymer. All materials were able to remove Cu(II) and Cd(II) from buffered aqueous solution (pH=5.7). The best result, obtained with PMAA-CRP, reached 94.8 mg of Cu(II) and 134.1mg of Cd(II) bound per gram of polymer. The better result for the FRP polymer may be due to its higher molar mass, although more studies are necessary to confirm that. The copolymer showed the lowest binding due most probably to its reduced negative density on the chain. The binding process is described as due to ion exchange, since the PMAA is considered a condensed polymer at the conditions studied. Because is a passive method, the removal system developed is, in principle, more convenient and energy efficient than other alternatives to be used in real conditions. A presente dissertação apresenta uma alternativa ao uso de espectrofotômetros aplicados a determinação do comprimento de onda de absorção máxima e concentração de substâncias que absorvem radiação eletromagnética na região do visível (400-800 nm), empregando-se imagens digitais para tal fim. Câmeras digitais foram usadas para obter todas as imagens digitais, sendo utilizada em um primeiro momento uma câmera e como segunda abordagem, visando diminuir os custos e aumentar a portabilidade, uma web cam. As variáveis fotográficas foram obtidas utilizando-se um software desenvolvido utilizando a linguagem JAVA®. Ferramentas matemáticas foram desenvolvidas utilizando-se o Método dos Mínimos Quadrados (MMQ), considerando as variáveis fotográficas: R (Vermelho), G (Verde), B (Azul), H (Matiz), S (Saturação) e Br (Brilho). Utilizando-se ANOVA verificou-se que todos os modelos foram viáveis em um nível de confiança de 95%. Para verificar a exatidão, em um primeiro momento o método foi aplicado em três amostras comerciais: Isotônico (Amarelo Tartrazina), Antiácido (Amarelo Crepúsculo) e Suplemeto de Ferro (Ferro total). Também foram coletadas amostras de água de mina, represas e corregos da região focando a determinação dos seguintes analitos: Alumínio, Ferro, Fósforo e Nitrito. Os limites de detecção (LOD) e de quantificação (LOQ) obtidos foram semelhantes aos resultados obtidos pelos métodos de referência. Foram efetuados testes de adição e recuperação, sendo as taxas de recuperação obtidas entre 93,50 e 108,30%. A precisão foi avaliada com o coeficiente de variação (n=3), sendo menor que 1,53%. As determinações com o método proposto e o método espectrofotométrico foram concordantes (teste-t pareado com 95% de confiabilidade). Em conclusão, este é um método simples e versátil, e mostra que imagens digitais podem ser aplicadas em determinações de concentração e ou comprimento de onda de máximo de absorção de analitos coloridos em solução aquosa. This dissertation presents an alternative to the use of spectrophotometers applied to determine the wavelength of maximum absorbance and concentration of substances that absorb electromagnetic radiation in the visible region (400-800 nm), using for this purpose digital images. Digital cameras were used to obtain all the digital images being used at first a camera and, as second approach in order to reduce costs and increase portability, a web cam. The photographic variables were obtained using software developed using JAVA®. Mathematical tools were developed using the least squares method (LSM), considering the photographic variables: R (Red), G (Green), B (blue), H (Hue), S (Saturation) and Br (Brightness). Thru the use of ANOVA all models were feasible on a confidence level of 95%. To verify accuracy, at first, the method was applied to three commercial samples: Isotonic (Yellow Tartrazine), Antacid (Sunset Yellow) and Suplemeto Iron (Iron total). Also, samples of mine water, dams and streams of the region focusing on the determination of the following analytes: Aluminum, Iron, Phosphorus and Nitrite. The limits of detection (LOD) and quantitation (LOQ) were similar to results obtained by standard methods. Tests of addition and recovery, were performed and the recovery rates obtained between 93.50 and 108.30%. Precision was evaluated as the relative standard deviation (n = 3), being less than 1.53%. Determinations with the proposed method and the spectrophotometric method were concordant (paired t-test with 95% confidence). In conclusion, this is a simple and versatile method and shows that the use of digital images may be applied to determinate the concentration and or the wavelength of maximum absorption of analytes in colored aqueous solution.
