Experiments and observations suggest that they contribute to terrestrial soil biodiversity and function, yet their ecological contribution to soil has never been assessed globally under natural conditions. Here we conducted the most comprehensive global standardized field study to quantify how soil mosses influence 8 ecosystem services associated with 24 soil biodiversity and functional attributes across wide environmental gradients from all continents. We found that soil mosses are associated with greater carbon sequestration, pool sizes for key nutrients and organic matter decomposition rates but a lower proportion of soil-borne plant pathogens than unvegetated soils. Mosses are especially important for supporting multiple ecosystem services where vascular-plant cover is low. Globally, soil mosses potentially support 6.43 Gt more carbon in the soil layer than do bare soils. The amount of soil carbon associated with mosses is up to six times the annual global carbon emissions from any altered land use globally. The largest positive contribution of mosses to soils occurs under a high cover of mat and turf mosses, in less-productive ecosystems and on sandy and salty soils. Applications of RNA-based molecular logic have been hampered by sequence constraints imposed on the input and output of the circuits. Here we show that the sequence constraints can be substantially reduced by appropriately encoded multi-arm junctions of single-stranded RNA structures. To conditionally activate RNA translation, we integrated multi-arm junctions, self-assembled upstream of a regulated gene and designed to unfold sequentially in response to different RNA inputs, with motifs of loop-initiated RNA activators that function independently of the sequence of the input RNAs and that reduce interference with the output gene. We used the integrated RNA system and sequence-independent input RNAs to execute two-input and three-input OR and AND logic in Escherichia coli, and designed paper-based cell-free colourimetric assays that accurately identified two human immunodeficiency virus (HIV) subtypes (by executing OR logic) in amplified synthetic HIV RNA as well as severe acute respiratory syndrome coronavirus-2 (via two-input AND logic) in amplified RNA from saliva samples. The sequence-independent molecular logic enabled by the integration of multi-arm junction RNAs with motifs for loop-initiated RNA activators may be broadly applicable in biotechnology.

Previous studies demonstrate an association between activation of the maternal immune system during pregnancy and increased risk of neurodevelopmental psychiatric conditions, such as schizophrenia and autism, in the offspring. Relatively recent findings also suggest that the gut microbiota plays an important role in shaping brain development and behavior. Here we show that maternal immune activation (MIA) accomplished by infection with a mouse-adapted influenza virus during pregnancy induced up-regulation of frontal cortex serotonin 5-HT2A receptor (5-HT2AR) density in the adult offspring, a phenotype previously observed in postmortem frontal cortex of schizophrenic subjects. 5-HT2AR agonist-induced head-twitch behavior was also augmented in this preclinical mouse model. Using the novel object recognition (NOR) test to evaluate cognitive performance, we demonstrate that MIA induced NOR deficits in adult offspring. Oral antibiotic treatment of prepubertal mice prevented this cognitive impairment, but not increased frontal cortex 5-HT2AR density or psychedelic-induced head-twitch behavior in adult MIA offspring. Additionally, gut microbiota transplantation from MIA mice produced behavioral deficits in antibiotic-treated mock mice. Adult MIA offspring displayed altered gut microbiota, and relative abundance of specific components of the gut microbiota, including Ruminococcaceae, correlated with frontal cortex 5-HT2AR density. Together, these findings provide a better understanding of basic mechanisms by which prenatal insults impact offspring brain function, and suggest gut-brain axis manipulation as a potential therapeutic approach for neurodevelopmental psychiatric conditions.

Using prenatal (E12.5) injection with poly-(I:C) as a mouse MIA model, we show that upregulation of the serotonin 5-HT2A receptor (5-HT2AR) is at least in part responsible for some of the effects of prenatal insults on frontal cortex dendritic spine structure and sensorimotor gating processes. Mechanistically, we report that this upregulation of frontal cortex 5-HT2AR expression is associated with MIA-induced reduction of nuclear translocation of the glucocorticoid receptor (GR) and, consequently, a decrease in the enrichment of GR at the 5-HT2AR promoter. The translational significance of these preclinical findings is supported by data in postmortem human brain samples suggesting dysregulated nuclear GR translocation in frontal cortex of schizophrenia subjects. Repeated (twice a day for 4 days) corticosterone administration augmented frontal cortex 5-HT2AR expression and reduced GR binding to the 5-HT2AR promoter. However, virally (AAV)-mediated augmentation of GR function reduced frontal cortex 5-HT2AR expression and improved sensorimotor gating processes via 5-HT2AR. Together, these data support a negative regulatory relationship between GR signaling and 5-HT2AR expression in mouse frontal cortex that may carry implications for the pathophysiology underlying 5-HT2AR dysregulation in neurodevelopmental psychiatric disorders.

However, unlike for plants and animals, a global assessment of hotspots for soil nature conservation is still lacking2. This hampers our ability to establish nature conservation priorities for the multiple dimensions that support the soil system: from soil biodiversity to ecosystem services. Here, to identify global hotspots for soil nature conservation, we performed a global field survey that includes observations of biodiversity (archaea, bacteria, fungi, protists and invertebrates) and functions (critical for six ecosystem services) in 615 composite samples of topsoil from a standardized survey in all continents. We found that each of the different ecological dimensions of soils—that is, species richness (alpha diversity, measured as amplicon sequence variants), community dissimilarity and ecosystem services—peaked in contrasting regions of the planet, and were associated with different environmental factors. Temperate ecosystems showed the highest species richness, whereas community dissimilarity peaked in the tropics, and colder high-latitudinal ecosystems were identified as hotspots of ecosystem services. These findings highlight the complexities that are involved in simultaneously protecting multiple ecological dimensions of soil. We further show that most of these hotspots are not adequately covered by protected areas (more than 70%), and are vulnerable in the context of several scenarios of global change. The SARS-CoV-2 pandemic is one of the most concerning health problems around the globe. We reported the emergence of SARS-CoV-2 variant B.1.1.519 in Mexico City. We reported the effective reproduction number (Rt) of B.1.1.519 and presented evidence of its geographical origin based on phylogenetic analysis. We also studied its evolution via haplotype analysis and identified the most recurrent haplotypes. Finally, we studied the clinical impact of B.1.1.519. The B.1.1.519 variant was predominant between November 2020 and May 2021, reaching 90% of all cases sequenced in February 2021. It is characterized by three amino acid changes in the spike protein: T478K, P681H, and T732A. Its Rt varies between 0.5 and 2.9. Its geographical origin remain to be investigated. Patients infected with variant B.1.1.519 showed a highly significant adjusted odds ratio (aOR) increase of 1.85 over non-B.1.1.519 patients for developing a severe/critical outcome (p = 0.000296, 1.33–2.6 95% CI) and a 2.35-fold increase for hospitalization (p = 0.005, 1.32–4.34 95% CI). The continuous monitoring of this and other variants will be required to control the ongoing pandemic as it evolves.

In Mexico, HPV vaccines available immunize against genotypes 16/18 and 16/18/6/11; however, there is limited surveillance about carcinogenic subtypes in different states of the country that allow evaluating the effectiveness of vaccination and cervical cancer screening programs. Here, we report the regional and age-specific prevalence of 14 hr-HPV genotypes as well as their prevalence in abnormal cytology (from ASCUS to cervical cancer) among Mexican women which were undergoing from cervical cancer screening in the Salud Digna clinics in 20 states of the country. This study includes women with social security from the majority of public health institutions (IMSS, ISSSTE, SEMAR, and PEMEX), and women without social security. For cervical cancer screening, we used the SurePath liquid-based cytology and the BD Onclarity HPV Assay. From December 1, 2016, to August 2, 2018, the hr-HPV prevalence among 60,135 women was 24.78%, the most prevalent types were HPV 16 (4.13%), HPV 31 (4.12%) and HPV 51 (3.39%), while HPV 18 (1.70%) was less prevalent among infected women. Interestingly, the genotypes not covered by current vaccines in Mexico were commonly found in precancerous lesions, evidencing their carcinogenic potential, so it is necessary to increase their surveillance and inclusion in cervical cancer screening triage., We gratefully acknowledge to Iromy Meza, Jessica Avitia, and Oswaldo Carrillo for their technical support in obtaining databases during this project. Serosurveillance helps establish reopening guidelines and determine the immunity levels in different populations to reach herd immunity. Then, there is an urgent need to estimate seroprevalence population wide. In Mexico, information about COVID-19 cases and related deaths is scarce. Also, there is no official serosurveillance, limiting our knowledge of the impact of the SARS-CoV-2 pandemic. Here, we report the prevalence of anti-SARS-CoV-2 antibodies in 522,690 unvaccinated people from July 5th to December 31st, 2020. The overall seroprevalence was 32.8% and highest in adults aged 30–39 years (38.5%) than people under 20 years (33.0%) or older (28.9%). Moreover, in a cohort of 1655 individuals confirmed COVID-19 by PCR, we found that symptomatic people (HR = 2.56) increased seroconversion than presymptomatic. Also, we identified that the most discriminative symptoms for COVID-19 that could predict seroconversion were anosmia and ageusia (HR = 1.70), fever, myalgia/arthralgia, and cough (HR = 1.75). Finally, we found that obese people had lower seroconversion (HR = 0.53) than healthy people, but the opposite happens in diabetic people (HR = 1.39). These findings reveal that around one-third of Mexican outpatients had anti-SARS-CoV-2 antibodies before vaccination. Also, some symptoms improve empirically COVID-19 diagnosis and seroconversion. This information could help fine-tune vaccination schemes and the reopening and back-to-work algorithms.

The structure and function of the soil microbiome of urban greenspaces remain largely undetermined. We conducted a global field survey in urban greenspaces and neighboring natural ecosystems across 56 cities from six continents, and found that urban soils are important hotspots for soil bacterial, protist and functional gene diversity, but support highly homogenized microbial communities worldwide. Urban greenspaces had a greater proportion of fast-growing bacteria, algae, amoebae, and fungal pathogens, but a lower proportion of ectomycorrhizal fungi than natural ecosystems. These urban ecosystems also showed higher proportions of genes associated with human pathogens, greenhouse gas emissions, faster nutrient cycling, and more intense abiotic stress than natural environments. City affluence, management practices, and climate were fundamental drivers of urban soil communities. Pharmacophore models for 5-HT2A receptor antagonists consist of two aromatic/hydrophobic regions at a given distance from a basic amine. We have previously shown that both aromatic/hydrophobic moieties are unnecessary for binding or antagonist action. Here, we deconstructed the 5-HT2A receptor antagonist/serotonin-dopamine antipsychotic agent risperidone into smaller structural segments that were tested for 5-HT2A receptor affinity and function. We show, again, that the entire risperidone structure is unnecessary for retention of affinity or antagonist action. Replacement of the 6-fluoro-3-(4-piperidinyl)-1,2-benz[ d]isoxazole moiety by isosteric tryptamines resulted in retention of affinity and antagonist action. Additionally, 3-(4-piperidinyl)-1,2-benz[ d]isoxazole (10), which represents less than half the structural features of risperidone, retains both affinity and antagonist actions. 5-HT2A receptor homology modeling/docking studies suggest that 10 binds in a manner similar to risperidone and that there is a large cavity to accept various N4-substituted analogues of 10 such as risperidone and related agents. Alterations of this "extended" moiety improve receptor binding and functional potency. We propose a new risperidone-based pharmacophore for 5-HT2A receptor antagonist action.

The aim of this study was to understand the responses of the microbial community of soil under different land uses to drought in a semi-arid Mediterranean area. In a laboratory incubation, soil samples from different land uses (natural forest, drip-irrigated orchard, rain-fed almond tree cultivation and abandoned area) were maintained at 20% and 60% of the WHC. The microbial biomass and potential enzyme activities were determined after four and fifty days of soil incubation. The diversity and composition of the microbial community were studied after 50 days of incubation. The total mineralisation of soil organic C (SOC), as well as, the mineralisation of fresh organic matter (FOM) and the “priming effect” were analysed after addition of 13C-enriched plant tissue. Both land use and drought had significant effects in the soil microbial community, but the effect of land use was stronger than that of drought. The PLFA content (microbial biomass) of the forests soil was greater under drought. After 50 days of soil incubation, the microbial biomass and most of potential enzyme activities of the almond tree and abandoned soil samples were not significantly affected by drought contrary to those in orchard soil. The total and FOM mineralisation were on average lower in soil under drought than under optimal moisture for all land uses. However, the responses of the priming effect to drought were dependent on the land use. Overall, we conclude that the resistance to drought of the soil microbial community from an agroecosystem having a semi-arid climate is strongly influenced by the previous land use.

Increased utilization of silver nanoparticles (AgNPs) can result in an accumulation of these particles in the environment. The potential detrimental effects of AgNPs in soil may be associated with the low fertility of soils in semiarid regions that are usually subjected to restoration through the application of organic amendments. Microbial communities are responsible for fundamental processes related to soil fertility, yet the potential impacts of low and realistic AgNPs concentrations on soil microorganisms are still unknown. We studied the effects of realistic citrate-stabilized AgNPs concentrations (0.015 and 1.5 μg kg−1 ) at two exposure times (7 and 30 days) on a sandy clay loam Mediterranean soil unamended (SU) and amended with compost (SA). We assessed soil microbial biomass (microbial fatty acids), soil enzyme activities (urease, β-glucosidase, and alkaline phosphatase), and composition of the microbial community (bacterial 16S rRNA gene and fungal ITS2 sequencing) in a microcosm experiment. In the SA, the two concentrations of AgNPs significantly decreased the bacterial biomass after 7 days of incubation. At 30 days of incubation, only a significant decrease in the Gram+ was observed at the highest AgNPs concentration. In contrast, in the SU, there was a significant increase in bacterial biomass after 30 days of incubation at the lowest AgNPs concentration. Overall, we found that fungal biomass was more resistant to AgNPs than bacterial biomass, in both SA and SU. Further, the AgNPs changed the composition of the soil bacterial community in SA, the relative abundance of some bacterial taxa in SA and SU, and fungal richness in SU at 30 days of incubation. However, AgNPs did not affect the activity of extracellular enzymes. This study demonstrates that the exposure time and organic amendments modulate the effects of realistic concentrations of AgNPs in the biomass and composition of the microbial community of a Mediterranean soil. La mayor utilización de nanopartículas de plata (AgNP) puede resultar en una acumulación de estas partículas en el medio ambiente. Los posibles efectos perjudiciales de los AgNP en el suelo pueden estar asociados con la baja fertilidad de los suelos en las regiones semiáridas que suelen estar sometidas a restauración mediante la aplicación de enmiendas orgánicas. Las comunidades microbianas son responsables de procesos fundamentales relacionados con la fertilidad del suelo, sin embargo, aún se desconocen los impactos potenciales de concentraciones bajas y realistas de AgNPs en los microorganismos del suelo. Estudiamos los efectos de concentraciones realistas de AgNP estabilizados con citrato (0,015 y 1,5 μg kg − 1) en dos tiempos de exposición (7 y 30 días) en un suelo mediterráneo franco arcilloso arenoso sin enmendar (SU) y enmendado con compost (SA). Evaluamos la biomasa microbiana del suelo (ácidos grasos microbianos), las actividades de las enzimas del suelo (ureasa, β-glucosidasa y fosfatasa alcalina) y la composición de la comunidad microbiana (gen bacteriano del ARNr 16S y secuenciación de ITS2 fúngico) en un experimento de microcosmos. En SA, las dos concentraciones de AgNP disminuyeron significativamente la biomasa bacteriana después de 7 días de incubación. A los 30 días de incubación, solo se observó una disminución significativa en el Gram + a la concentración más alta de AgNPs. En contraste, en la UB, hubo un aumento significativo en la biomasa bacteriana después de 30 días de incubación a la concentración más baja de AgNPs. En general, encontramos que la biomasa fúngica era más resistente a los AgNP que la biomasa bacteriana, tanto en SA como en SU. Además, los AgNP cambiaron la composición de la comunidad bacteriana del suelo en SA, la abundancia relativa de algunos taxones bacterianos en SA y SU, y la riqueza fúngica en SU ​​a los 30 días de incubación. Changing climatic conditions (warming and decreasing precipitation) have been found to be a threat to the agricultural sustainability of Mediterranean croplands. From the climate change perspective, biochar amendment may interact with the effects of warming and drought stresses on soil ecosystems. However, the responses of soil microbial communities to the joint effects of climate change and biochar in Mediterranean croplands are not sufficiently known. To help fill this knowledge gap, in this work we used a field experiment to determine the effects of partial rain exclusion alone or combined with a soil temperature increase in biochar-amended (20 t ha) and unamended plots under crop rotation on soil chemical properties, enzyme activities, and the microbial community activity, structure, composition, abundance, and functions. The biomass, composition, and activity of the soil bacterial and fungal communities were more responsive to biochar addition than to climate manipulation. Thus, soil chemical parameters, enzyme activities and the relative abundances of bacterial populations were not responsive to the interaction of biochar and climate manipulation, while the predicted functionality of the bacterial community was modified by both factors. Soil β-glucosidase activity significantly decreased in response to biochar addition and climate manipulation, while urease activity was significantly increased by biochar, and protease activity was significantly decreased by climate manipulation. Gram negative and fungal biomasses were significantly affected by the interaction of biochar with climate manipulation. Climate manipulation produced changes in the composition of the soil fungal community without loss of diversity. Background To validate the performance, reproducibility, and reliability of BD automated instruments in order to establish a fully automated clinical microbiology laboratory. Methods We used control strains and clinical samples to assess the accuracy, reproducibility, and reliability of the BD Kiestra WCA, the BD Phoenix, and BD Bruker MALDI-Biotyper instruments and compared them to previously established conventional methods. The following processes were evaluated: sample inoculation and spreading, colony counts, sorting of cultures, antibiotic susceptibility test, and microbial identification. Results The BD Kiestra recovered single colonies in less time than conventional methods (e.g. E. coli, 7h vs 10h, respectively) and agreement between both methodologies was excellent for colony counts (κ=0.824) and sorting cultures (κ=0.821). Antibiotic susceptibility tests performed with BD Phoenix and disk diffusion demonstrated 96.3% agreement with both methods. Finally, we compared microbial identification in BD Phoenix and Bruker MALDI-Biotyper and observed perfect agreement (κ=1) and identification at a species level for control strains. Together these instruments allow us to process clinical urine samples in 36h (effective time). Conclusion The BD automated technologies have improved performance compared with conventional methods, and are suitable for its implementation in very busy microbiology laboratories.

Short-rotation plantations for biomass production are wood systems in which fast growing tree species are grown under intensive agricultural practices to achieve high biomass yields. Paulownia sp. is an extremely fast-growing tree species which is commonly used to produce biomass for energetic purposes. This tree species has a great nutrient and water demand, and the establishment of its plantations may affect soil fertility and quality. For this fact, this study is focused on the evaluation of the responses of the soil microbial community – in terms of biomass, community structure, and activity – in a Paulownia plantation submitted to a semiarid climate. Particular attention was paid to the impact of the soil fertilization (mineral fertilizer or an organic-mineral amendment, BAN, obtained from the forestry industry) and level of irrigation on the soil microbial community. For both irrigation regimes, a significant increase in TOC was observed with the addition of the BAN to the soil. The content of water soluble C in the soil was affected by both factors (fertilization and irrigation) and their interaction. Both enzyme activities related with the C and P cycles in soil and dehydrogenase activity were significantly affected by the fertilization, irrigation, and their interaction. Regardless of the irrigation level, the microbial respiration rate was highest with the BAN treatment. The content of total PLFAs and PLFA biomarkers of microbial groups showed no significant differences between the fertilization treatments. However, the total PLFAs and the Gram-positive, fungal, and saturated PLFA concentrations differed significantly between the irrigation levels. In general, the use of residues generated in the forest biomass industry (pine bark and biomass ash) as a soil amendment had a positive effect on the soil microbial activity without altering the structure of the soil microbial community. Considering the water deficit of semiarid areas, such as SE Spain, the low level of irrigation tested in this study would be enough to maintain both the soil microbial activity and suitable production of biomass by Paulownia trees.

The increasing use of silver nanoparticles (AgNPs) due to their well-known antimicrobial activity, has led to their accumulation in soil ecosystems. However, the impact of environmental realistic concentrations of AgNPs on the soil microbial community has been scarcely studied. In this work, we have assessed the impact of AgNPs, that mimic real concentrations in nature, on tropical soils cultivated with Coffea arabica under conventional and organic management systems. We evaluated the biomass, extracellular enzyme activities, and diversity of the soil microbial community, in a microcosm experiment as a function of time. After seven days of incubation, we found an increase in microbial biomass in an AgNPs-concentration-independent manner. In contrast, after 60-day-incubation, there was a decrease in Gram+ and actinobacterial biomass, in both soils and all AgNPs concentrations. Soil physico-chemical properties and enzyme activities were not affected overall by AgNPs. Regarding the microbial community composition, only some differences in the relative abundance at phylum and genus level in the fungal community were observed. Our results suggest that environmental concentrations of AgNPs affected microbial biomass but had little impact on microbial diversity and may have little effects on the soil biogeochemical cycles mediated by extracellular enzyme activities. El uso cada vez mayor de nanopartículas de plata (AgNP) debido a su conocida actividad antimicrobiana, ha provocado su acumulación en los ecosistemas del suelo. Sin embargo, el impacto de las concentraciones ambientales realistas de AgNP en la comunidad microbiana del suelo ha sido escasamente estudiado. En este trabajo, hemos evaluado el impacto de los AgNP, que imitan concentraciones reales en la naturaleza, en suelos tropicales cultivados con Coffea arabica bajo sistemas de manejo convencionales y orgánicos. Evaluamos la biomasa, las actividades de las enzimas extracelulares y la diversidad de la comunidad microbiana del suelo, en un experimento de microcosmos en función del tiempo. Después de siete días de incubación, encontramos un aumento en la biomasa microbiana de una manera independiente de la concentración de AgNP. Por el contrario, después de 60 días de incubación, hubo una disminución en la biomasa Gram + y actinobacteriana, tanto en los suelos como en todas las concentraciones de AgNP. Las propiedades físico-químicas del suelo y las actividades enzimáticas no se vieron afectadas en general por los AgNP. Con respecto a la composición de la comunidad microbiana, solo se observaron algunas diferencias en la abundancia relativa a nivel de filo y género en la comunidad fúngica. Nuestros resultados sugieren que las concentraciones ambientales de AgNP afectaron la biomasa microbiana, pero tuvieron poco impacto en la diversidad microbiana y pueden tener pocos efectos en los ciclos biogeoquímicos del suelo mediados por actividades de enzimas extracelulares. Identifying the global drivers of soil priming is essential to understanding C cycling in terrestrial ecosystems. We conducted a survey of soils across 86 globally-distributed locations, spanning a wide range of climates, biotic communities, and soil conditions, and evaluated the apparent soil priming effect using 13C-glucose labeling. Here we show that the magnitude of the positive apparent priming effect (increase in CO2 release through accelerated microbial biomass turnover) was negatively associated with SOC content and microbial respiration rates. Our statistical modeling suggests that apparent priming effects tend to be negative in more mesic sites associated with higher SOC contents. In contrast, a single-input of labile C causes positive apparent priming effects in more arid locations with low SOC contents. Our results provide solid evidence that SOC content plays a critical role in regulating apparent priming effects, with important implications for the improvement of C cycling models under global change scenarios., The global ecological predictors of soil priming remain unclear. Here the authors conducted a global survey of soils from 86 global locations using an isotopic approach and find that in more mesic sites with high SOC concentrations, soil priming effects are more likely to be negative.

Despite the promise of research conducted at the intersection of computer-assisted language learning (CALL), natural language processing, and second language acquisition, few studies have explored the potential benefits of using intelligent CALL systems to deepen our understanding of the process and products of second language (L2) learning. The strategic use of technology offers researchers novel methodological opportunities to examine how incremental changes in L2 development occur during treatment as well as how the longitudinal impacts of experimental interventions on L2 learning outcomes occur on a case-by-case basis. Drawing on the pilot results from a project examining the effects of automatic input enhancement on L2 learners’ development, this article explores how the use of technology offers additional methodological and analytical choices for the investigation of the process and outcomes of L2 development, illustrating the opportunities to study what learners do during visually enhanced instructional activities.

Several pharmacophore models have been proposed for 5-HT2A serotonin receptor antagonists. These typically consist of two aromatic/hydrophobic moieties separated by a given distance from each other, and from a basic amine. Although specified distances might vary, the models are relatively similar in their general construction. Because our preliminary data indicated that two aromatic (hydrophobic) moieties might not be required for such action, we deconstructed the serotonin-dopamine antipsychotic agent risperidone (1) into four smaller structural fragments that were thoroughly examined in 5-HT2A receptor binding and functional (i.e., two-electrode voltage clamp (TEVC) and intracellular calcium release) assays. It was apparent that truncated risperidone analogues behaved as antagonists. In particular, 6-fluoro-3-(1-methylpiperidin-4-yl)benzisoxazole (4) displayed high affinity for 5-HT2A receptors (Ki of ca. 12 nM) relative to risperidone (Ki of ca. 5 nM) and behaved as a potent 5-HT2A serotonin receptor antagonist. These results suggest that multiple aromatic (hydrophobic) moieties are not essential for high-affinity 5-HT2A receptor binding and antagonist activity and that current pharmacophore models for such agents are very much in need of revision.

Pharmacophore models for 5-HT

We previously reported that co-expression of the Gi-coupled metabotropic glutamate receptor 2 (mGlu2R) and the Gq-coupled serotonin (5-HT) 2A receptor (2AR) in Xenopus oocytes (Fribourg et al. Cell 147:1011-1023, 2011) results in inverse cross-signaling, where for either receptor, strong agonists suppress and inverse agonists potentiate the signaling of the partner receptor. Importantly, through this cross-signaling, the mGlu2R/2AR heteromer integrates the actions of psychedelic and antipsychotic drugs. To investigate whether mGlu2R and 2AR can cross-signal in mammalian cells, we stably co-expressed them in HEK293 cells along with the GIRK1/GIRK4 channel, a reporter of Gi and Gq signaling activity. Crosstalk-positive clones were identified by Fura-2 calcium imaging, based on potentiation of 5-HT-induced Ca(2+) responses by the inverse mGlu2/3R agonist LY341495. Cross-signaling from both sides of the complex was confirmed in representative clones by using the GIRK channel reporter, both in whole-cell patch-clamp and in fluorescence assays using potentiometric dyes, and further established by competition binding assays. Notably, only 25-30 % of the clones were crosstalk-positive. The crosstalk-positive phenotype correlated with (a) increased colocalization of the two receptors at the cell surface, (b) lower density of mGlu2R binding sites and higher density of 2AR binding sites in total membrane preparations, and (c) higher ratios of mGlu2R/2AR normalized surface protein expression. Consistent with our results in Xenopus oocytes, a combination of ligands targeting both receptors could elicit functional crosstalk in a crosstalk-negative clone. Crosstalk-positive clones can be used in high-throughput assays for identification of antipsychotic drugs targeting this receptor heterocomplex.

Patches where shrubs have either positive or negative effects on their understory plant community are common in arid ecosystems. The intensity and balance of these effects change along environmental severity gradients but, despite the major role of soil microbes in plant interactions, little is known about the differences among soil microbial communities under these species and their possible influence on such contrasting shrub effects. We hypothesized that microbial communities associated to benefactor and allelopathic shrubs would differ among them and that differences would increase with environmental severity. To test these hypotheses we characterized soil microbial biomass, activity and community composition under a benefactor shrub species, Retama sphaerocarpa, an allelopathic shrub species, Thymus hyemalis, and in bare soil among plants (gaps) at three sites along an environmental severity gradient. Shrubs promoted an increase in soil bacterial diversity, being bacterial communities associated to benefactor shrubs, allelopathic shrubs and gaps different in composition. Microbial enzymatic activity and biomass increased under shrubs and under more mesic conditions; nonetheless, they were highest under benefactor shrubs at the most arid site and under allelopathic shrubs at the less severe site. Compared to gaps, the presence of shrubs induced changes in microbial activity and community composition that were larger at the most severe site than at the less severe site. Along the gradient, benefactor shrubs enhanced the abundance of bacterial groups involved in organic matter decomposition and N fixation as well as plant pathogens, which could contribute to Retama's outstanding positive effects on understory plant biomass and diversity. Plant patches mitigate the effects of extreme conditions on associated plant and soil microbial communities and promote soil biodiversity and ecosystem functioning in arid ecosystems, with shrubs actively selecting for specific microbial groups in their understory.

Schizophrenia is a psychiatric disorder that affects 1% of the population worldwide. The serotonin and glutamate receptor systems have been implicated in schizophrenia and its treatment. Serotonin 5-HT2A receptor is target of hallucinogens such as lysergic acid diethylamide (LSD) and psilocin, as well as involved in the mechanism of action of atypical antipsychotic drugs such as clozapine and risperidone. The metabotropic glutamate 2 (mGlu2) receptor modulates the physiological responses induced by the 5-HT2A receptor, and preclinical and clinical work suggests that this glutamate receptor may represent a new approach to treat schizophrenia. Here we review recent advances in our understanding of the crosstalk between these two receptors, as well as their implication in schizophrenia and antipsychotic drug action.

Green roofs have been considered as a promising approach for greening cities where green areas are scarce. Apart from aesthetic and ecological values, environmental benefits are well recognized: reducing the urban heat island effect, promoting CO2 sequestration, and increasing urban wildlife habitats. However, there are few studies conducted that assess the CO2 sequestration and the N fixation carried out by these novel ecosystems. In this sense, this work focuses on the study of C and N dynamics in eight different green roofs during 2 years of experimentation. For this purpose, we tested two different substrates: a compost-soil-bricks (1:1:3; v:v:v) and a compost-bricks (1:4; v:v) with two depths (5 and 10 cm). Each substrate was disposed in cultivation tables composed by eight independent experimental plots. Silene vulgaris (Moench) Garcke and Lagurus ovatus L. were the tested plant species in monoculture. Physicochemical and biochemical substrate characterizations were conducted both initially (T0: Nov. 2012) and at the end of the experiment (TF: Aug. 2014), along with plant development measurements (plant cover and water use efficiency), in order to find the most appropriate plant-substrate combination in terms of C and N sequestration. These results showed that the soil-containing mixture at a higher depth highly promotes plant growth, especially for S. vulgaris in this susbtrate (CSB10-S), being related to intense biochemical processes and also to the formation of humic substances in this substrate, which all contribute to the great amounts of C and N (1702.19 g C and 183.21 g N m− 2 during 2 years, respectively) achieved by this combination.

The serotonin 2A (5-HT(2A)) and metabotropic glutamate 2 (mGlu2) receptors regulate each other and are associated with schizophrenia. The Roman high- (RHA-I) and the Roman low- (RLA-I) avoidance rat strains present well-differentiated behavioral profiles, with the RHA-I strain emerging as a putative genetic rat model of schizophrenia-related features. The RHA-I strain shows increased 5-HT(2A) and decreased mGlu2 receptor binding levels in prefrontal cortex (PFC). Here, we looked for differences in gene expression and transcriptional regulation of these receptors. The striatum (STR) was included in the analysis. 5-HT(2A), 5-HT(1A), and mGlu2 mRNA and [(3)H]ketanserin binding levels were measured in brain homogenates. As expected, 5-HT(2A) binding was significantly increased in PFC in the RHA-I rats, while no difference in binding was observed in STR. Surprisingly, 5-HT(2A) gene expression was unchanged in PFC but significantly decreased in STR. mGlu2 receptor gene expression was significantly decreased in both PFC and STR. No differences were observed for the 5-HT(1A) receptor. Chromatin immunoprecipitation assay revealed increased trimethylation of histone 3 at lysine 27 (H3K27me3) at the promoter region of the HTR2A gene in the STR. We further looked at the Akt/GSK3 signaling pathway, a downstream point of convergence of the serotonin and glutamate system, and found increased phosphorylation levels of GSK3β at tyrosine 216 and increased β-catenin levels in the PFC of the RHA-I rats. These results reveal region-specific regulation of the 5-HT(2A) receptor in the RHA-I rats probably due to absence of mGlu2 receptor that may result in differential regulation of downstream pathways.

The long-term effects of deforestation on the soil microbial community and its functionality are largely unknown. In order to assess simultaneously the phylogeny and functionality, we applied phospholipid fatty acid analysis (PLFAs), metaproteomics and enzyme activities in soil samples from a natural area located in Southeast Spain (Caravaca de la Cruz), dominated by Pinus halepensis (F), and an adjacent area deforested 15-years ago (DF). Deforestation induced a long-term loss of bacterial biomass and enzyme activity, but an increase in the bacterial diversity as estimated by metaproteomics. Protein abundances analysis revealed that Proteobacteria was higher in F than DF. In addition, the abundance of cyanobacterial proteins was significantly higher in DF (7.3%) when compared to F (0.9%). Interestingly, cyanobacterial proteins involved in carbon fixation (Ribulose 1,5-bisphosphate carboxylase, phycocyanins and photosystem proteins) were only identified in DF. The data suggest that Cyanobacteria play a critical role in the ecosystem functioning and biotic carbon fixation when soil is deforested in semiarid areas.

Shrubs in semiarid ecosystems facilitate the establishment of other plant species under their canopies and promote changes in the understory plant communities as they grow. To better understand whether plant community dynamics are linked to changes in soil microbes, we characterized soil microbial communities in gaps (open spaces) and under Retama sphaerocarpa shrubs of three different size classes. Different methodological approaches including pyrosequencing of 16S rDNA, phospholipid-fatty acid (PLFA) analysis and microbial activity indicators were combined to characterize both the structure and function of soil microbial communities in the different treatments. Soil microbial communities under small shrubs showed higher microbial biomass and activity, as well as different relative abundance of several bacterial groups, than communities in gaps. Shrubs were associated with a higher relative abundance of Bacteroidetes , Betaproteobacteria and Gammaproteobacteria in detriment of Actinobacteria and Firmicutes without changes in overall bacterial diversity. Soil microbial community changed with shrub size, being most different in gaps and under the canopy of large shrubs. We suggest that changes in composition and function of soil microbial communities may promote the increase in the understory plant growth and species richness previously reported for this shrub species. Our data emphasize the importance of plant–soil interactions in defining the structure and composition of both plant and soil microbial communities and their impact on ecosystem functioning.

Serotonin 5-HT(2A) and metabotropic glutamate 2 (mGlu2) are G protein-coupled receptors suspected in the pathophysiology of psychiatric disorders, such as schizophrenia, depression, and suicide. Previous findings demonstrate that mGlu2 mRNA expression is down-regulated in brain cortical regions of 5-HT2A knockout (KO) mice. However, the molecular mechanism responsible for this alteration remains unknown. We show here repressive epigenetic changes at the promoter region of the mGlu2 gene in frontal cortex of 5-HT(2A)-KO mice. Disruption of 5-HT(2A) receptor-dependent signaling in mice was associated with decreased acetylation of histone H3 (H3ac) and H4 (H4ac) and increased tri-methylation of histone H3 at lysine 27 (H3K27me3) at the mGlu2 promoter, epigenetic changes that correlate with transcriptional repression. Neither methylation of histone H3 at lysine 4 (H3K4me1/2/3) nor tri-methylation of histone H3 at lysine 9 (H3K9me3) was affected. We found that Egr1, a transcription factor in which promoter activity was positively regulated by the 5-HT(2A) receptor agonist 4-bromo-3,6-dimethoxybenzocyclobuten-1-yl)methylamine hydrobromide, binds less to the mGlu2 promoter in frontal cortex of 5-HT(2A)-KO, compared with wild-type mice. Furthermore, expression of mGlu2 was increased by viral-mediated gene transfer of FLAG-tagged Egr1 in mouse frontal cortex. Together, these observations suggest that 5-HT(2A) receptor-dependent signaling epigenetically affects mGlu2 transcription in mouse frontal cortex.

Hallucinogenic drugs, such as lysergic acid diethylamide (LSD), mescaline and psilocybin, alter perception and cognitive processes. All hallucinogenic drugs have in common a high affinity for the serotonin 5-HT(2A) receptor. Metabotropic glutamate 2/3 (mGlu2/3) receptor ligands show efficacy in modulating the cellular and behavioral responses induced by hallucinogenic drugs. Here, we explored the effect of chronic treatment with the mGlu2/3 receptor antagonist 2S-2-amino-2-(1S,2S-2-carboxycyclopropan-1-yl)-3-(xanth-9-yl)-propionic acid (LY341495) on the hallucinogenic-like effects induced by LSD (0.24mg/kg). Mice were chronically (21 days) treated with LY341495 (1.5mg/kg), or vehicle, and experiments were carried out one day after the last injection. Chronic treatment with LY341495 down-regulated [(3)H]ketanserin binding in somatosensory cortex of wild-type, but not mGlu2 knockout (KO), mice. Head-twitch behavior, and expression of c-fos, egr-1 and egr-2, which are responses induced by hallucinogenic 5-HT(2A) agonists, were found to be significantly decreased by chronic treatment with LY341495. These findings suggest that repeated blockade of the mGlu2 receptor by LY341495 results in reduced 5-HT(2A) receptor-dependent hallucinogenic effects of LSD.

Ibi, Daisuke et al., Antipsychotic drugs remain the standard for schizophrenia treatment. Despite their effectiveness in treating hallucinations and delusions, prolonged exposure to antipsychotic medications leads to cognitive deficits in both schizophrenia patients and animal models. The molecular mechanisms underlying these negative effects on cognition remain to be elucidated. Here we demonstrate that chronic antipsychotic drug exposure increases nuclear translocation of NF-κB in both mouse and human frontal cortex, a trafficking event triggered via 5-HT-receptor-dependent downregulation of the NF-κB repressor IκBα. This upregulation of NF-κB activity led to its increased binding at the Hdac2 promoter, thereby augmenting Hdac2 transcription. Deletion of HDAC2 in forebrain pyramidal neurons prevented the negative effects of antipsychotic treatment on synaptic remodeling and cognition. Conversely, virally mediated activation of NF-κB signaling decreased cortical synaptic plasticity via HDAC2. There is little information about the potential effects of compost and zeolite or zeolite with leonardite as soil amendments in barley cultivation. Thus in this study, the following objectives were proposed: i) to compare the effects of the addition of compost, alone or simultaneously with zeolite, and of the addition of leonarditeenriched zeolite with those of the conventional NPK fertilization used in barley cultivation, on the soil nutritional status, microbial community structure, and enzyme activity in different stages of barley cultivation; and ii) to establish relationships between the different soil parameter trends, soil microbial community structure, and barley crop yield. In the field experiment carried out with a barley crop, the alternative fertilization treatments tested had an overall positive effect, in comparison with conventional fertilization with a mineral NPK fertilizer, when soil quality parameters, the nutritional level and yield of the barley crop were analyzed. Zeolite with leonardite increased barley yield in comparison with the compost treatments, either with zeolite or without, but had an excessive contribution to the water soluble contents of Na and N in soil. So, using environmental and agronomic criteria, the most rational action would be the use of compost in agriculture. It has been suggested that severe adverse life events during pregnancy increase the risk of schizophrenia in the offspring. The serotonin 5-HT2Aand the metabotropic glutamate 2 (mGlu2) receptors both have been the target of considerable attention regarding schizophrenia and antipsychotic drug development. We tested the effects of maternal variable stress during pregnancy on expression and behavioral function of these two receptors in mice. Prenatal stress increased 5-HT2Aand decreased mGlu2 expression in frontal cortex, a brain region involved in perception, cognition, and mood. This pattern of expression of 5-HT2Aand mGlu2 receptors was consistent with behavioral alterations, including increased head-twitch response to the hallucinogenic 5-HT2Aagonist DOI [1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane] and decreased mGlu2-dependent antipsychotic-like effect of the mGlu2/3 agonist LY379268 (1R,4R,5S,6R-2-oxa-4-aminobicyclo[3.1.0]hexane-4,6-dicarboxylate) in adult, but not prepubertal, mice born to stressed mothers during pregnancy. Cross-fostering studies determined that these alterations were not attributable to effects of prenatal stress on maternal care. Additionally, a similar pattern of biochemical and behavioral changes were observed in mice born to mothers injected with polyinosinic:polycytidylic acid [poly(I:C)] during pregnancy as a model of prenatal immune activation. These data strengthen pathophysiological hypotheses that propose an early neurodevelopmental origin for schizophrenia and other psychiatric disorders.
